    Plasma proteome signature of canine acute haemorrhagic diarrhoea syndrome (AHDS)

    Acute haemorrhagic diarrhoea is a common complaint in dogs. In addition to causes like intestinal parasites, dietary indiscretion, intestinal foreign bodies, canine parvovirus infection, or hypoadrenocorticism, acute haemorrhagic diarrhoea syndrome (AHDS) is an important and sometimes life-threatening differential diagnosis. There is some evidence supporting the link between Clostridium perfringens toxins and AHDS. These toxins may be partially responsible for the epithelial cell injury, but the pathogenesis of AHDS is still not fully understood. Recent studies have suggested that severe damage to the intestinal mucosa and associated barrier dysfunction can trigger chronic gastrointestinal illnesses. Besides bloodwork and classical markers for AHDS such as protein loss and intestinal bacterial dysbiosis, we focused mainly on the plasma-proteome to identify systemic pathological alterations during this disease and searched for potential biomarkers to improve the diagnosis. To accomplish the goals, we used liquid chromatography-mass spectrometry. We compared the proteomic profiles of 20 dogs with AHDS to 20 age-, breed-, and sex-matched control dogs. All dogs were examined, and several blood work parameters were determined and compared, including plasma biochemistry and cell counts. We identified and quantified (relative quantification) 207 plasmatic proteins, from which dozens showed significantly altered levels in AHDS. Serpina3, Lipopolysaccharide-binding protein, several Ig-like domain-containing proteins, Glyceraldehyde-3-phosphate dehydrogenase and Serum amyloid A were more abundant in plasma from AHDS affected dogs. In contrast, other proteins such as Paraoxonase, Selenoprotein, Amine oxidases, and Apolipoprotein C-IV were significantly less abundant. Many of the identified and quantified proteins are known to be associated with inflammation. Other proteins like Serpina3 and RPLP1 have a relevant role in oncogenesis. Some proteins and their roles have not yet been described in dogs with diarrhoea. Our study opens new avenues that could contribute to the understanding of the aetiology and pathophysiology of AHDS

    Bloodwork is a widely used diagnostic tool in veterinary medicine, as diagnosis and therapeutic interventions often rely on blood biomarkers. However, biomarkers available in veterinary medicine often lack sensitivity or specificity. Mass spectrometry-based proteomics technology has been extensively used in the analysis of biological fluids. It offers excellent potential for a more comprehensive characterization of the plasma proteome in veterinary medicine.In this study, we aimed to identify and quantify plasma proteins in a cohort of healthy dogs and compare two techniques for depleting high-abundance plasma proteins to enable the detection of lower-abundance proteins via label-free quantification liquid chromatography-mass spectrometry. We utilized surplus lithium-heparin plasma from 30 healthy dogs, subdivided into five groups of pooled plasma from 6 randomly selected individuals each. Firstly, we used a commercial kit to deplete high-abundance plasma proteins. Secondly, we employed an in-house method to remove albumin using Blue-Sepharose.Among all the samples, some of the most abundant proteins identified were apolipoprotein A and B, albumin, alpha-2-macroglobulin, fibrinogen beta chain, fibronectin, complement C3, serotransferrin, and coagulation factor V. However, neither of the depletion techniques achieved significant depletion of highly abundant proteins. Despite this limitation, we could detect and quantify many clinically relevant proteins. Determining the healthy canine proteome is a crucial first step in establishing a reference proteome for canine plasma. After enrichment, this reference proteome can later be utilized to identify protein markers associated with different diseases, thereby contributing to the diagnosis and prognosis of various pathologies

    Diplomarbeit - Veterinärmedizinische Universität Wien - 2024In der Humanmedizin ist die Arthroskopie erstmalig 1918 beschrieben worden und seitdem ein fester Bestandteil der Orthopädie. In den letzten Jahrzehnten gab es viele Innovationen auf dem Gebiet der minimalinvasiven Orthopädie. Einer dieser Innovationen ist das Nadelarthroskop. Der Umfang dieser Instrumente beträgt < 2 mm, wodurch die Möglichkeiten von minimalinvasiven Eingriffen neugestaltet wurden. Das Ziel dieser Arbeit war es daher festzustellen, welche Indikationen für die Nadelarthroskopie in der Human- und Veterinärmedizin in der Literatur bereits beschrieben wurden. Es wurde eine systematische Literaturrecherche nach Prisma Richtlinien in Onlinedatenbanken wie Pubmed, Science direct und Google Scholar und in veterinärmedizinischen Universitätsbibliotheken durchgeführt. Die Studien wurden anschließend nach den Kriterien der Cochrane Musculoskeletal Group (Santesso et al. 2006) in Evidenzklassen eingeteilt. Insgesamt konnten 36 Publikationen, welche in dem Zeitraum September 2010 – September 2023 veröffentlicht wurden, in diese Übersichtsarbeit inkludiert werden. Dabei konnten 21 Studien der Humanmedizin und 15 Studien der Veterinärmedizin zugeteilt werden. Ihrem Studiendesgin entsprechend, konnten nur 3 Arbeiten die Kriterien für die Einstufung in die Evidenzklasse „Silber“ erfüllen. Die Nadelarthroskopie stellte sich in nahezu allen eingeschlossenen Studien als eine äußerst effektive und komplikationsarme intraartikuläre Untersuchungsmethode dar. Die häufigste Indikation war eine diagnostische Arthroskopie. Therapeutische Indikationen wurden bisher nur von wenigen Vorreitergruppen erforscht, doch auch hier erwies es sich als ein vielversprechendes Instrument. In the recent decades, there have been many innovations in the field of minimally invasive orthopedics. One of these innovations is the needle arthroscope. Needle arthroscopes are very small arthroscopes. The diameter of these instruments is under 2 mm, which has reshaped the possibilities of minimally invasive procedures. This review attempts to determine which indications for needle arthroscopy in human and veterinary medicine have already been described in the literature. Using different search engines such as Pubmed, Science direct, Google Scholar and the local veterinary university libraries catalogues, a systematic literature search was performed. The studies were then classified into evidence classes according to the criteria of the Cochrane Musculoskeletal Group (Santesso et al. 2006). A total of 36 publications published between September 2010 and September 2023 were included in this review. Of these, 21 studies could be assigned to human medicine and 15 studies to veterinary medicine. Only 3 papers met the criteria for classification to the "silver" evidence class. Needle arthroscopy proved to be an effective intraarticular examination method in the included studies. The most frequently indication was the diagnostic arthroscopy. So far, therapeutic indications have only been researched by a few pioneers, here it could also be a promising tool. Needle arthroscopy can improve the diagnosis of both animal and human patients with musculoskeletal complaints. There are only a few literatures available on therapeutic indications, especially in veterinary medicine. However, it is argued that it does provide advantages over conventional arthroscopy in human medicine. In general, the amount of available literature is still too small and not of sufficient quality, so further scientific work is essential

    Comparison of Transformers with LSTM for classification of the behavioural time budget in horses based on video data

    This study compares the performance of Transformers with LSTM for the classification of the behavioural time budget in horses based on video data. The behavioural time budget of a horse consists of amount of time of the activities such as feeding, resting, lying, and moving, which are important indicators of welfare and can be a basis of pain detection. Video technology offers a non-invasive and continuous monitoring approach for automated detection of horse behaviours. Computer vision and deep learning methods have been used for automated monitoring of animal behaviours, but accurate behaviour recognition remains a challenge. Previous studies have employed Convolutional LSTM models for behaviour classification, and more recently, Transformer-based models have shown superior performance in various tasks. This study proposes a multi-input, multi-output classification methodology to address the challenges of accurately detecting and classifying horse behaviours. The results demonstrate that the multi-input and multi-output Transformer model achieves the best performance in behaviour classification compared with single input and single output strategy. The proposed methodology provides a basis for detecting changes in behaviour time budgets related to pain and discomfort in horses, which can be valuable for monitoring and treating horse health problems

    Evaluierung der Stresslevel von Servicehunden mittels Speichelcortisolwerten im Rahmen ihres Alltags im Vergleich zu Familienhunden

    Service dogs support people with motor disabilities, work together with their owners on a daily basis and are on constant alert. It is therefore of scientific interest whether this is associated with increased stress levels. In this study, the cortisol levels in the saliva of eleven service dogs and eight family dogs without special tasks were compared in order to determine their stress levels. It was investigated whether service dogs have increased stress levels compared to family dogs. The results are relevant in terms of the animals\u27 health, as prolonged stress can lead to illness, among other things. As cortisol levels can vary greatly over the course of a day and in order to obtain representative results, the samples were taken three times a day for seven consecutive days and, where possible, at the same times. In order not to falsify the stress level, the measurements were taken by the owners themselves and in their own homes. It was found that there were no significant differences with regard to the time of sampling and that service dogs had significantly lower cortisol levels than family dogs. Possible reasons could be the regulated daily routine of service dogs and a strong trust between humans and dogs, which may result in lower stress levels. Due to the relatively small sample size in this study, the results should be interpreted with caution and replicated with a larger number of dogs.Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Servicehunde unterstützen Personen mit motorischen Einschränkungen, arbeiten täglich mit ihren BesitzerInnen zusammen und stehen unter ständiger Alarmbereitschaft. Daher ist es von wissenschaftlichem Interesse, ob dies mit einer erhöhten Stressbelastung einhergeht. In der vorliegenden Arbeit wurde der Cortisolwert im Speichel von elf Servicehunden und acht Familienhunden ohne besondere Aufgaben miteinander verglichen, um deren Stressbelastung eruieren zu können. Es wurde untersucht, ob Servicehunde erhöhte Stresspegel im Vergleich zu Familienhunden aufweisen. Die Ergebnisse sind relevant im Bezug auf die Gesundheit der Tiere, da länger anhaltender Stress unter anderem zu Erkrankungen führen kann. Da Cortisolwerte im Verlauf eines Tages stark variieren können und um repräsentative Ergebnisse zu erhalten, wurden die Proben an sieben aufeinanderfolgenden Tagen drei Mal täglich und möglichst zu gleichen Uhrzeiten entnommen. Um den Stresspegel nicht zu verfälschen, wurden die Messungen von den BesitzerInnen selbst und im eigenen Heim durchgeführt. Es stellte sich heraus, dass keine signifikanten Unterschiede im Hinblick auf den Zeitpunkt der Probenahme vorliegen und dass Servicehunde signifikant niedrigere Cortisolwerte als Familienhunde aufweisen. Mögliche Gründe könnten ein geregelter Tagesablauf von Servicehunden und ein starkes Vertrauen zwischen Mensch und Hund sein, wodurch es möglicherweise zu einer geringeren Stressbelastung kommt. Aufgrund der relativ kleinen Stichprobengröße in dieser Studie sollten die Ergebnisse mit Vorsicht interpretiert werden und mit einer größeren Anzahl an Hunden repliziert werden

    Development of an in vitro model for inducing cellular senescence in ovine chondrocytes and synoviocytes: Implications for osteoarthritis research

    Diplomarbeit - Veterinärmedizinische Universität Wien - 2024Osteoarthritis (OA) ist eine degenerative Gelenkerkrankung, die durch das Vorhandensein von alternden Zellen gekennzeichnet ist. Chondrozyten und Synoviozyten sind die vorherrschenden Zelltypen im Gelenk und spielen eine entscheidende Rolle beim chronischen Fortschreiten der OA, wenn sie in einen Zustand der Seneszenz eintreten. Es wird angenommen, dass chronische Entzündungen, die durch verschiedene Faktoren wie Genetik oder Gelenkverletzungen ausgelöst werden, die zelluläre Homöostase dieser Zellen stören und sie in einen zellulären Stresszustand versetzen. Stressbedingte Zellschäden, insbesondere unter Bedingungen eines homöostatischen Ungleichgewichts wie Sauerstoffmangel, können zu zellulärer Dysfunktion führen und die Zellen möglicherweise in die Seneszenz treiben. Sobald Zellen in die Seneszenz eintreten, verbleiben sie für unbestimmte Zeit in einem Zustand des Wachstumsstillstands. Seneszente Zellen produzieren SASP-Faktoren (Senescence-Associated Secretory Phenotype), darunter entzündungsfördernde Zytokine wie IL-6 und IL-8, die zur Degeneration des Knorpels beitragen. Dies wiederum führt zu einer Kaskade von mehr Chondrozyten und Synoviozyten, die aufgrund der durch die OA verursachten erhöhten Stressumgebung um die Läsion in die Seneszenz eintreten. Das Verständnis des seneszenten Zustands von Chondrozyten und Synoviozyten ist entscheidend, um in diesen Zyklus einzugreifen und ihn zu unterbrechen. Ziel dieser Studie war es, die Seneszenz in Chondrozyten und Synoviozyten mit Hilfe von Dexamethason (20 μM, 10 μM, 5 μM und 1 μM) und Doxorubicin (200 nM, 100 nM, 50 nM und 25 nM) chemisch zu induzieren und das Seneszenzinduktionsprotokoll zu optimieren. Beide Medikamente wurden mit verschiedenen Assays und Tests getestet, um eine Dosistitration und einen direkten Vergleich zu ermöglichen. Die Ergebnisse zeigten, dass Dexamethason bei keiner der getesteten Konzentrationen toxische Auswirkungen auf die Zellen hatte, wie der MTT-Lebensfähigkeitstest zeigte. Eine mindestens 72-stündige Inkubation mit Dexamethason in allen Konzentrationen (20 μM, 10 μM, 5 μM und 1 μM) war erforderlich, um die Seneszenzassoziierten Genexpressionen in Chondrozyten wirksam hochzuregulieren, und eine Inkubation von 6 Tagen war die beste Dauer. In Chondrozyten zeigte 1 μM Dexamethason die stärkste Hochregulierung von Seneszenz-assoziierten Genen, was durch die 3-tägigen und 6- tägigen Titrationsversuche bestätigt wurde. Für Synoviozyten wurde 5 μM Dexamethason als optimale Konzentration ermittelt. Doxorubicin war bei 200 nM zytotoxisch und 100 nM waren zytotoxisch, wobei die Lebensfähigkeit der Zellen deutlich reduziert war. Doxorubicin induzierte die höchste Expression von seneszenzassoziierten Genen sowohl in Chondrozyten als auch in Synoviozyten bei einer Konzentration von 200 nM. Allerdings zeigte der MTT-Test, dass die Konzentrationen von 200 nM und 100 nM Doxorubicin toxisch waren und zum Zelltod führten. Um dies zu vermeiden, wurde eine Konzentration von 50 nM als nicht toxisch identifiziert, die aber immer noch in der Lage ist, Veränderungen in der Genexpression auszulösen, die auf Seneszenz hindeuten. Zusammenfassend lässt sich sagen, dass die Studie einen Einblick in die optimalen Bedingungen und chemischen Wirkstoffe für die Auslösung von Seneszenz in Chondrozyten und Synoviozyten gibt. Diese Ergebnisse bilden die Grundlage für künftige Studien, die darauf abzielen, die Seneszenz umzukehren und potenzielle Behandlungen für OA zu entwickeln. Es sei darauf hingewiesen, dass unsere In-vitro-Ergebnisse zwar Aufschluss über den seneszenten Zustand geben, aber möglicherweise nicht vollständig die In-vivo-Bedingungen der OA widerspiegeln. Weitere Untersuchungen, wie z. B. Beta-Galaktosidase-Assays und Zytokinanalysen, werden empfohlen, um unser Verständnis des seneszenten Zustands zu vertiefen.Diploma thesis - University of Veterinary Medicine Vienna - 2024Osteoarthritis (OA) is a degenerative joint disease characterised by the presence of senescent cells. Chondrocytes and synoviocytes are the predominant cell types in the joint and play a critical role in the chronic progression of OA when they enter a state of senescence. It is thought that chronic inflammation, triggered by various factors such as genetics or joint injury, disrupts the cellular homeostasis of these cells, causing them to undergo cellular stress. Stress-induced cellular damage, particularly under conditions of homeostatic imbalance such as oxygen deprivation, can lead to cellular dysfunction, potentially pushing cells into senescence. Once cells enter senescence, they remain in a state of growth arrest for an indefinite period of time. Senescent cells produce Senescence-Associated Secretory Phenotype (SASP) factors, including pro-inflammatory cytokines such as IL-6 and IL-8, which contribute to cartilage degeneration. This in turn leads to a cascade of more chondrocytes and synoviocytes entering senescence due to the increased stress environment around the lesion caused by OA. Understanding the senescent state of chondrocytes and synoviocytes is critical to intervene and interrupt this cycle. The aim of this study was to chemically induce senescence in chondrocytes and synoviocytes using Dexamethasone (at 20 μM, 10 μM, 5 μM and 1 μM) and Doxorubicin (at 200 nM, 100 nM, 50 nM and 25 nM) and to optimise the senescence induction protocol. Both drugs were tested using various assays and tests to allow dose titration and direct comparison. The results showed that Dexamethasone had no toxic effects on cells at any of the concentrations tested, as indicated by the MTT viability assay. A minimum of 72 hours incubation with Dexamethasone at all concentrations (20 μM, 10 μM, 5 μM, and 1 μM) was required to effectively upregulate senescence-associated gene expressions in chondrocytes, and 6 days incubation was the best duration. In chondrocytes, 1 μM Dexamethasone showed the highest up-regulation of senescence-associated genes, which was confirmed by the 3-day and 6-day titration assays. For synoviocytes, 5 μM Dexamethasone was identified as the optimal concentration. Doxorubicin was cytotoxic at 200 nM and 100 nM was cytotoxic with reduced cell viability. Doxorubicin induced the highest expression of senescence-associated genes in both chondrocytes and synoviocytes at a concentration of 200 nM. To overcome this, a concentration of 50 nM was identified as non-toxic but still capable of inducing gene expression changes indicative of senescence. In conclusion, the study provides insight into the optimal conditions and chemical agents for inducing senescence in chondrocytes and synoviocytes. These findings lay the groundwork for future studies aimed at reversing senescence and developing potential treatments for OA. It should be noted that although our in vitro results shed light on the senescent state, they may not fully reflect the in vivo conditions of OA. Further investigations, such as beta-galactosidase assays and cytokine analysis, are recommended to further our understanding of the senescent state

    Can a gradual weaning and separation process reduce weaning distress in dam-reared dairy calves? A comparison with the two-step method

    The weaning and separation phase remains one of the biggest challenges for cow-calf contact systems, but a gradual process that better mimics the naturally occurring reduction in milk intake has not yet been scientifically investigated. Therefore, the aim of our study was to compare behavioral and physiological indicators of distress in 3-mo-old dam-reared dairy calves (with previous full-time cow-calf contact) weaned and separated either via gradual reduction of contact time with the dam (GR; 1 wk of half-day contact, 1 wk of morning contact, and 1 wk of fence-line contact before complete separation, n = 18) or via 2-step weaning using a nose flap (NF, 2 wk of access to the dam with a nose flap, 1 wk of fence-line contact before complete separation, n = 18). Behavior was recorded 1 wk before (or for lying 3 wk before) weaning start and during the 3 wk weaning and separation period with direct observations on 4 d/wk or via accelerometers (locomotor play, lying behavior). Blood and fecal samples were taken twice per week from weaning start until 3 wk after weaning start. Calves were weighed weekly. Statistical analysis was conducted using (generalized) linear mixed models. Over the whole weaning and separation phase, NF calves showed a stronger decrease in the number of lying bouts, amount of locomotor play, and ADG, as well as a higher increase in TMR feeding time compared with GR calves, whereas GR calves vocalized more often and showed more searching behavior than NF calves. Also, the neutrophil:lymphocyte ratio of NF calves was elevated on d 3 after insertion of the nose flaps compared with baseline, but showed no change for GR calves on any sampling day. Overall, results point toward a favorable effect of a gradual weaning strategy on reduction of weaning and separation distress in dam-reared dairy calves, but the method requires further improvement from the protocol used in our study
