    Aktivitas Sitotoksik Ekstrak Etanol Daun Aglaia elliptica Blume terhadap Galur Sel Kanker Serviks (HeLa)

    Cytotoxicity tests on ethanol extract of Aglaia elliptica Blume leaves and its fractions on cervical cancer cell line (HeLa) were carried out. Dried powder of Aglaia elliptica leaves was extracted using ethanol andthen fractionated by n-hexane, ethyl acetate, and buthanol solvents. The cytotoxicity tests using MTT method (prolipheration inhibiton test) on HeLa cell lines were conducted for ethanol extract, n-hexana, ethyl acetate, and buthanol lractions.The results showed that the ethanol extract of Aglaia elliptica and its fractions have inhibition effect against HeLa cell line with IC50 of 282.44 ppm, 177.64 ppm, 16.03 ppm, 211.77 ppm, 181.33 ppm for ethanol extract, n-hexane, ethyl acetate, buthanol, and Water fractions respectively

    The synergic effects of n-butanolic fraction of secondary metabolite of endophytic fungus 1.3.11 (FB) and doxorubicin (Dox) on cell cycle regulation and the expression of Bcl-2 gene expression were investigated on MCF-7 and T47-D cells by iiow cytometry and immunocytochemical techniques respectively. The results showed that after 12 hours of incubation period with FB at its IC50 dose, MCF-7 cell cycle was inhibited at G1 phase while Dox inhibited the cell cycle at G2/M phase. Similar results were observed in T47-D cells when incubated with Dox and FB individually under the same treatment condition. Further treatment was then performed to these cells where both Dox and FB were combined at their IC50 and  lC50 dose and added to incubate with the cells over 12 hours period. Interestingly, the modified treatment combination showed that MCF-7 and T47-D cell cycle regulation were inhibited at G2/M phase. Our immunocytochemical study also showed no significant inhibition suppression of Bel-2 gene expression in both MCF-7 and T47-D cells when compared with their corresponding positive control after treatment with FB and Dox or both combined FB and Dox at 1C50 and  IC50 dosage over 15 hours incubation

    Uji Aktivitas Isoflavon Tempe terhadap Penghambatan Enzim α-Glukosidase dan Pengaruhnya pada Tikus Hiperglikemik

    Isoflavone is a bioactive compound found in fermented soy bean or tempe, and found to have beneficial effect on human health. The objectives of this research were to assay the activity of tempe isoliavone in inhibiting the α-glucosidase enzyme in vitro and to determine the dosage of antihyperglycemic activity of isoflavone tempe. inhibitory effects of tempe isoflavone on α-glucosidase enzyme was tested by spectrofotometry method while in vivo evaluation are conducted using rats under hyperglycemic condition. A total of thirty five male Spraque Dawley rats, which is two months old were used in this study. The rats were divided into seven groups: (1) negative control group; (2) positive hyperglycemic group; (3-6) positive hyperglycemic group and treated with tempe isoflavone with dosages of 100, 200, 300, and 400 mg/kg body weight (bw); and (7) positive hyperglycemic group treated with acarbose dosage of 4,5 mg/kg bw. Blood glucose level was evaluated every 0, 30, 60, 120, 180, and 240 minutes following treatment using Blood Blucose Test Meter GlucoDr. The result showed that the inhibitory effect of tempe isoflavone on α-glucosidase enzyme was 11.89%, with IC50 1.4 mg, and this mechanisms are caused by daidzein and genistein. lsoflavone of tempe with dose of 400 mg/kg bw showed an antihyperglycemie effect.Isoflavon adalah senyawa bioaktif pada tempe yang dilaporkan mempunyai efek positif terhadap kesehatan, Penelitian ini bertujuan untuk mengetahui aktivitas penghambatan enzim α-glukosidase oleh isoflavon tempe secara in vitro dan efek antihiperglikemik in vivo pada tikus putih hiperglikemia. Uji aktivitas in vitro daya hambat isoflavon tempe terhadap enzim α-glukosidase menggunakan metode spektrofotometri, sementara evaluasi in vivo menggunakan tikus dalam keadaan hiperglikemik_ Penelitian ini menggunakan 35 ekor tikus putih jantan galur Spraque Dawley berumur dua bulan. Tikus dibagi menjadi tujuh kelompok perlakuan: (1) kelompok kontrol negatif; (2) kelompok kontrol positif hiperglikemia; (3-6) kelompok hiperglikemia dan diberi isoflavon tempe masing-masing dengan dosis 100, 200, 300, dan 400 mg/kg bb per oral; dan (7) kelompok kontrol obat positif diberi obat acarbose 4,5 mg/kg bb. Kadar glukosa darah diukur setiap 0, 30, 60, 120, 180, dan 240 menit setelah perlakuan menggunakan alat Blood Glucose Test Meter GIucoDr Hasil penelitian meriunjukkan bahwa isoflavon tempe mempunyai daya hambat in vitro terhadap aktivitas enzim α-glukosidase dengan inhibisi sebesar 11,89% dan nilai IC50 sebesar 1,4 mg, dan dapat dinyatakan bahwa mekanisme daya hambat ini disebabkan oleh daidzein dan genistein. Uji aktivitas antihiperglikemik isoflavon tempe pada berbagai dosis menunjukkan bahwa pada dosis 400 mg/kg bb mempunyai daya antihiperglikemik

    Phalerin is an active component of mahkota dewa (Phaleria macrocarpa (Scheff) Boerl) proven to have an anti-inflammation effect. The labeling of phalerin with gamma emitting radionuclides was aimed to study its pharmacokinetic behavior and particularly to trace its metabolites. The labeling with 131I was caried out using iodogen as oxidator. Radiolabeled compound was characterized by high performance liquid chromatography (HPLC) using C- 18 column eluted with methanol 70% and detected with UV detector (λ=291 nm) and by thin layer chromatography (TLC) using silica gel strips eluted with chloroform-methanol (9:2), and labeling efficiency was determined using the same TLC system. Purification of radiolabeled product Was carried out using size exclusion chromatography (Sephadex G-25 column) eluted with 0.05 M phosphate buffer pH 7.4. Biodistributions of 131l-phalerin in various organs of normal and inilammation-induced mice were observed at 1, 4, and 24 hours post-intravenous injection. Radiochemical purity of 131l-phalerin was 90.2 ± 2.8% and increased to 96.0 ± 0.4% after purification. Radioactivities in inflamed tissue at 1, 4, and 24 hours post-injection were respectively 1.6 times, 1.4 times, and 1.3 times higher than that in normal tissue, The results showed a significant uptake of radiolabeled phalerin in inflamed tissue

    Medicinal plant called “sarang semut”, Myrmecodia pendens Merr. & Perry (Rubiaceae), is an epiphyt plant traditionally used in Papua for healing various diseases such as hyperuricemia, rheumatic, and cancer. A chemical compound recognized as xanthinoxidase inhibitor has been isolated from n-buthanol fraction of the plant extract. The isolation was carried out by column chromatography (SiO2, CHCI3-MeOH = 5:1-1:1) and the purification Was conducted using high pressure liquid chromatography (RP, C-18; MeOH-water = 5 : 1). The activity test for Xanthinoxidase inhibitor Was performmed by using modilied Noro method. The pure compound showed xanthinoxidase inhibitor activity of IC50  at 79,77%

    Pengaruh Lupene-ol dan Lupene-on dari Aegle marmelos Correa terhadap Pelepasan Enzim β-hexoaminidase dari Sel Mast

    20(29)-lupene-3α-ol and 20(29)-lupene-3-on are pentacyclic triterpenes isolated from Aegle marmelos Correa collected in Yogyakarta, Indonesia. Their molecular structures were confirmed in Universiti Putra Malaysia. These compounds were obtained from petroleum ether extract of the plant’s stem bark. This study investigated the effects of the compounds on the β -hexoaminidase enzyme release from mast cell culture (RBL-2H3 cell line). DNP24-BSA was used as immunologic inducer for the β -hexoaminidase release from the mast cells. The enzyme release was determined by colorimetric methods with a substrate, p-nytrophenyl-2-acetamydo-2-deoxy- β -D-gluko-pyranocide, and a microplate reader at 405 nm. lt was found that treatment with 20 µg/ml DNP24-B SA could stimulate the release of β -hexoaminidase from RBL-ZH3 cells by 3O.83 ± 1.10%. Lupene-ol and lupene-on showed inhibitory effects on the β -hexoaminidase release from RBL-2H3 cells induced by DNP24-BSA in dose-dependent manner. At the dose of 100 µM, lupene-ol and lupene-on inhibited  the β -hexoaminidaserelease by 35.69i4.7 5% and 39.19 9.56%, respectively. The ICN values of their effects on DNP24-BSA experiments were 59.40 µM and 72.51 µM. The results suggested that the inhibitory effect of lupene-ol and lupene-on on the β-hexoaminidase release involved mechanisms related to the interaction of IgE on the RBL-2H3 cells surface or intracellular signal transductions found in the mast cell degranulation.     Penggunaan Obat Herbal pada Pasien Kanker Serviks

    Herbal medicines have widely been used to treat many type of diseases despite the advance of standard or conventional therapy. In fact, many people in Indonesia use medicinal plant as their customary part of life. Therefore, it is necessary to further explore the use of herbal medicines through modern perception. ln this study we would like to know the frequency and species of herbal medicines used among cervical cancer patients in National Cancer Hospital Dharmais, Jakarta, Indonesia and also to assess the relationship between age, education, jobs, stage of cancer, and payment status of the patients. The results were 61,8% patients used herbal medicines. The herbal medicines used most were mahkota dewa (Phaleria macrocarpa Scheetf Boerl.) 35.3%, temu putih (Curcuma zedoaria Rosc.) 32.4%, and buah merah (Pandanus conoideus Lam) 17,6%. There was a significant relationship (p=0,039) between the use of herbal medicines and stage of cancer but no relationship between age, education, occupation, income and payment status with the use of herbal medicines

    Analisis Mutu Pelayanan Instalasi Farmasi pada RSUD Cilacap dan RS PKU Muhammadiyah Karanganyar

    An investigation on the quality service at the Pharmacy Department of Cilacap Local Government Hospital (CLGH) and Mohammadiyah Hospital in Karanganyar (MHK) which covered five service dimensions, i.e. reliability, responsiveness, assurance, empathy and tangible factors, has been conducted, The quality of services was analyzes by using the SERVQUAL gap model. The study Was a non-experimental descriptive research. The data were collected using purposive sampling with questionnaires as the research tools. The results showed that in the pharmacy departments, of both CLGH and MHK), the above-mentioned pharmacy service dimensions observed performed below customer expectations. The highest gap score in CLGH is on responsiveness followed by tangible factors, reliability, empathy and assurance. The biggest gap score in MHK is on tangible factors, followed by empathy, assurance, responsiveness and reliability, Loyalty of customer to pharmacy department of CLGH for regular patient is around 50,6%, MHK 39,5%, neutral customer 35,1%, and non-loyal customer 25,4%. The results indicated that both hospitals need to strengthen quality services of their pharmacy departments, particularly on service dimensions which have high score gap.Penelitian terhadap mutu pelayanan lnstalasi Farmasi Rumah Sakit di Rumah Sakit Umum Daerah (RSUD) Cilacap dan Rumah Sakit PKU Muhammadiyah di Karanganyar yang mencakup lima dimensi pelayanan, yaitu reliability, responsiveness, assurance, empathy dan tangible factors, telah dilakukan. Mutu pelayanan dianalisis menggunakan gap model SERVQUAL. Penelitian ini non-eksperimental deskriptif, data dihimpun menggunakan sampling purposif dengan kuesioner sebagai alat penelitian. Hasil menunjukkan bahwa di lnstalasi Farmasi kedua rumah sakit, baik RSUD Cilacap maupun RS PKU Muhaminadiyah Karanganyar, semua dimensi layanan farmasi kinerjanya di bawah harapan pelanggan. Nilai gap tertinggi di RSUD Cilacap adalah pada responsiveness yang diikuti oleh tangible factors, reliability, empathy dan assurance, Nilai gap tertinggi RS PKU Muhammadiyah Karanganyar adalah pada tangible factors, diikuti oleh empathy, assurance, responsiveness dan reliability. Kesetiaan pelanggan lnstalasi Fannasi RSUD Cilacap untuk pasien regular sekitar 50,6%, RS PKU Muhammadiyah Karanganyar 39,5%, pelanggan netral 35,1 %, dan pelanggan yang tidak setia 25,4%. Hasil penelitian ini mengindikasikan bahwa kedua rumah sakit tersebut perlu meningkatkan kualitas layanan farmasinya, terutama pada dimensi pelayanan dengan nilai gap yang tinggi

    Abstract: In order to develop the applications of radiation for biomaterial preparation, a non-toxic superabsorbent has been synthesized. A series of superbasorbent hydrogel were prepared from aqueous solution containing 5% polyethylene oxide (PEO) with different concentration of alginate (0.5-2.0%) were irradiated at the doses of 20, 30, and 40 kGy by gamma radiation.The gel fraction, swelling kinetics and the equilibrium degree of swelling (EDS) of the hydrogels were studied, It was found that the incorporation of even 2% alginate (sodium salt) increased the EDS of the hydrogels from 20g/g to 320 g/ g. At a dose of 20 kGy, PEO-alginat hydrogels with high gel fraction (  ̴ 8O%) and very high EDS (  ̴ 320 g/g) were obtained. The hydrogels were also found to be sensitive to the ionic solution of NaCl. The PEO-alginate hydrogels could be considered as candidate biomaterials in health care

    Formulation of lozenges containing: A) green tea extract; B) green tea extract with sodium ascorbate; and C) green tea extract with grape seed extract have been carried out. Antioxidant activities were compared to the activity of raw active materials. Lozenges were tested for physical characteristics, hedonic values, dissolution rate, and stability during storage, Physical tests indicated that the lozenges comply with the Farmakope Indonesia IV requirements. Determination of IC50 antioxidant activity using DPPI-I free radical scavenger showed that IC50 of formula A, B, and C were 844.04 µg/mL, 221.80 µg/ mL, and C 57.4 µg/mL, respectively. Meanwhile, IC50 value of green tea extract, sodium ascorbate, and grape seed extract were 4.29 µg/mL, 12.95 µg/mL, and 1.96 µg/ml., respectively. Accelerated stability test were carried out based on the EGCG concentration observed by using HPLC and then calculated by Arrhenius equation showed that the shelf life of lozenges of formula A, B, and C Were 2,11 years; 1.87 years; and 2,73 years, respectively. The result showed that the lozenge containing green tea extract and grape seed extract (formula C) is the best formula based on FI IV requirements, antioxidant activity and lozenges stability.Telah dilakukan fomiulasi tablet hisap dengan bahan aktif utama: A) ekstrak teh hijau; B) ekstrak teh hijau dan natrium askorbat; C) ekstrak teh hijau dan ekstrak biji anggur, Aktivitas antioksidan tablet hisap dibandingkan dengan aktivitas bahan baku aktif. Tablet hisap diuji karakteristik Hsik, uji kesukaan (nilai hedonik), uji disolusi, dan uji stabilitas dalam penyiinpanan. Uji karakteristik fisik menunjukkan bahwa tablet hisap memenuhi ketentuan-ketentuan Farmakope Indonesia IV Penetapan aktivitas antioksidan tablet hisap dengan metode DPPH memberikan nilai IC50 formula A, B, dan C masingmasing adalah 844,04 µg/mL, 221,80 µg/ml., dan C 57,4 µg/mL. sementara nilai ICM) ekstrak teh hijau, natrium askorbat, dan ekstrak biji anggur masing-masing adalah 4,29 µg/ml., 12,95 µg/mL, dan 1.96 µg/mL. Pengujian stabilitas dipercepat dilakulcan berdasar kadar EGCG dengan KCKT yang kemudian dihitung menggunakan persamaan Arrhenius menunjukkan bahwa waktu simpan formula A, B, dan C masing-masing adalah 2,11 tahun; 1,87 tahun dan 2,73 tahun_ Hasil menunjukkan bahvva tablet hisap mengandung ekstrak teh hijau dan ekstrak biji anggur (formula C) aclalah tablet hisap yang terbaik
