    Investigation of the Effect of Using Geogrids on the Performance of the Road in Pavements Constructed With Reduced Layer Thicknesses

    In this study, the structural performance of road pavements with reduced layer thicknesses at different rates using geogrids was examined, and the benefits of geogrids in preventing rutting were investigated. Within the scope of the study, a total of 16 test sections of 50 meters in length, 12 with geogrids and four without geogrids, were constructed on the Adana - Kozan road determined by the General Directorate of Highways of Turkey (KGM). The constructed sections were divided into two, and besides the reference sections, two types of geogrids with different geometric structures were used in successive sections. Some layer thicknesses used in the study were determined following the specification, and the other part was determined through a pavement design program. In addition, to compare the effect of using geogrids in different locations on the pavement performance in the application sections, the geogrid was used in two positions, on the subbase layer and between the base layer. Data from the load, displacement and temperature sensors placed between the layers during the construction phase and on-site Heavy Weight Deflectometer (HWD) data at certain time intervals were used to observe the structural performances of the constructed pavement sections. In addition, GPS and IRI measurements are made regularly. The traffic values of the trial section are also monitored. Examining the obtained data aims to compare the economic gain achieved by reducing the layer thicknesses, the cost to be spent for the geogrid, and to make cost-benefit analyses

    Development of Mg-Alginate Based Self Disassociative Bio-Ink for Magnetic Bio-Patterning of 3d Tumor Models

    Alginate forms a hydrogel via physical cross-linking with divalent cations. In literature, Ca2+ is mostly utilized due to strong interactions but additional procedures are required to disassociate Ca-alginate hydrogels. On the other hand, Mg-alginate hydrogels disassociate spontaneously, which might benefit certain applications. This study introduces Mg-alginate as the main component of a bio-ink for the first time to obtain 3D tumor models by magnetic bio-patterning technique. The bio-ink contains magnetic nanoparticles (MNPs) for magnetic manipulation, Mg-alginate hydrogel as a sacrificial material, and cells. The applicability of the methodology is tested for the formation of 3D tumor models using HeLa, SaOS-2, and SH-SY5Y cells. Long-term cultures are examined by Live/dead and MTT analysis and revealed high cell viability. Subsequently, Collagen and F-actin expressions are observed successfully in 3D tumor models. Finally, the anti-cancer drug Doxorubicin (DOX) effect is investigated on 3D tumor models, and IC50 values is calculated to assess the drug response. As a result, significantly higher drug resistance is observed for bio-patterned 3D tumor models up to tenfold compared to 2D control. Overall, Mg-alginate hydrogel is successfully used to form bio-patterned 3D tumor models, and the applicability of the model is shown effectively, especially as a drug screening platform

    Hardware acceleration with fpga Based electronic boards for machine learning

    Son yıllardaki makine ög˘renmesi algoritmalarındaki gelis¸meler uç cihazlardaki kullanımını da arttırmıs¸tır (Merenda et al., 2020). Makine ög˘renimi algoritmaları genel- likle GPU tabanlı bilgisayarlarda çalıs¸tırılmaktadır, bu da yüksek enerji tüketimi (De- sislavov et al., 2021), yog˘un donanım kaynag˘ı gereksinimleri ve büyük fiziksel boyutları (Liu et al., 2022) nedeniyle uç cihazlar için uygun olmamaktadır. Bu tez, donanım hızlandırıcısı olarak FPGA platformlarında makine ög˘renmesi algoritmalarının, özellikle derin sinir ag˘larının uygulanması ve çıkarım yapılmasını aras¸tırarak, düs¸ük güç tüke- timi, verimli donanım kullanımı ve yüksek çıkarım performansı elde etmeyi hedefle- mektedir. Bu sistemlerin uç cihazlara adaptasyonu için esneklig˘i ve verimlilig˘i artırmak amacıyla, CNV ag˘ının (Umuroglu et al., 2017b) daha hafif bir varyasyonu olan CNV light gelis¸tirilmis¸. Bu ag˘, PyTorch tabanlı bir araç olan Brevitas (Pappalardo et al., 2019) ile nicemleme-farkında-eg˘itim yöntemi, kullanılarak 1, 2, 4 ve 8-bit seviyelerine nicemleme yapılmıs¸tır. CNV light ag˘ı CIFAR-10, SVHN, GTSRB ve MNIST veri setleri üzerinde Brevitas ile eg˘itilmis¸tir. Modeller FINN çerçevesi (Umuroglu et al., 2017a) kullanılarak FPGA'ya sentezlenmis¸tir. Modeller en fazla, en az ve sabit FPS seviye donanım kul- lanımına göre ayarlanmıs¸tır. Xilinx XC7Z020-1CLG400C FPGA, modelin metriklerini deg˘erlendirmek ve raporlamak için kullanılmıs¸tır. GTSRB veri setinde, ikili (W1A1) nicemleme yapılmıs¸ CNV light ag˘ı, tüm donanım kullanımları için %95.12 dog˘ruluk ve en fazla donanım kullanımında 12,191 FPS performansı ve 3.20W güç tüketimi elde etti, minimum donanım kullanımı için ise 6 FPS ve 1.62W güç tüketti. Sonuçlar, FPGA'ların uç cihazlarda makine ög˘renmesi modellerini verimli ve ölçeklenebilir platformlar olarak kullanılabileceg˘ini göstermektedir.Developments in machine learning algorithms in recent years have also increased their use in end devices (Merenda et al., 2020). Machine learning algorithms are typically executed on GPU-based computers, which makes them unsuitable for edge devices due to their high power consumption (Desislavov et al., 2021), hardware resource requirements, and physical footprint (Liu et al., 2022). This thesis explores the deployment of machine learning algorithms on FPGA. This method aims to achieve low power consumption, efficient hardware utilization, and high inference performance. To improve efficiency, this research introduces the CNV light network, a simplified variation of the CNV network (Umuroglu et al., 2017b). The network was quantized to 1, 2, 4, and 8-bit levels using the Brevitas library (Pappalardo et al., 2019). The CNV light network was trained on the CIFAR-10, SVHN, GTSRB, and MNIST datasets with Brevitas. Custom parallelization configurations were utilized to explore maximum, minimum, and constant FPS hard- ware utilization levels using the FINN framework (Umuroglu et al., 2017a). The Xilinx XC7Z020-1CLG400C FPGA was employed for the evaluation and reporting of model metrics. Binary (W1A1) quantized CNV light network on the GTSRB dataset reached an accuracy of 95.12% for all hardware utilization cases and 12,191 FPS and 3.20W power consumption for maximum hardware utilization case, and 6 FPS and 1.62W power consumption for minimum hardware utilization case. The results show that FPGAs can be used as efficient and scalable platforms for deploying machine learning models

    Appropriation of forests and water resources in mount ida and surroundings: Dealing with socio-environmental problems by integrating biodiversity impact chain analysis into planning

    Bu tez, mevcut koruma eleştirileri ile kent ve planlama kuramındaki eleştirel tartışmalara dayanarak koruma ve planlama arasındaki bağlantıyı yeniden kurulmasının olanaklarını tartışmaktadır. Ayrıca Kaz Dağları örneği üzerinden planlamada yeni bir çevre koruma anlayışı geliştirmenin Türkiye için imkanlarını araştırmaktadır. Ekosistem hizmetlerinin değerlendirilmesi gibi piyasa temelli koruma anlayışları, geleneksel korumanın kapsamını genişletse de, bu yaklaşımlar yeni belirlenen bölgelerde 'yeşil gasp' uygulamalarına yol açması ve bu alanların dışında doğanın sınırsız bir şekilde sömürülmesini teşvik etmesi ile eleştirilmektedir. Bir yanda da sosyo-çevresel krizlerin ve yeni eşitsizlilerin üstesinden gelmeyi amaçlayan küçülme stratejilerinin mekansallaşması ve planlamayı ekonomik büyümeye odağından uzaklaştırarak büyüme sonrası bir planlama pratiğine geçişin gerekliliği tartışılmaktadır. Türkiye'de ise ekonomik büyümenin önceliklendirilmesi ve karar alma süreçlerinin merkezileştirilmesi nedeniyle, koruma bölgeleri içerisinde dahi yıkıcı faaliyetler ve 'daha yeşil' faaliyetler eş zamanlı olarak yürütülmektedir. Kaz Dağı Milli Parkı ve güney yamacı kısmen korunurken, çeperdeki ormanlarda yeni madencilik faaliyetleri ve kıyı şeridinde jeotermal enerji kaynaklı faaliyeler, ihracata yönelik politikalar nedeniyle ormanların ve su kaynaklarının çevre kirliliğine ve eşitsizliklere yol açacak şekilde tahsisi ile devam etmektedir. Kaz Dağları ormanları için yeni bir ekosistem hizmetleri projesi söz konusu olsa bile, yasal yetersizliklerin yanı sıra izole koruma kararları, ekonomik büyüme odaklı hedefler ve politikalar nedeniyle farklı ölçeklerde yeni eşitsizlikler ortaya çıkabilir. Bu doğrultuda tez, eşitsizlikleri ortaya çıkarmak, büyüme sonrası alternatifleri önermek ve politikleştirmek için bir araç olarak 'biyoçeşitlilik etki zinciri' analizi ile alan için 'koruma ölçekleri' önermektedir.This thesis discusses the potential of re-establishing the link between conservation and planning based on the existing conservation criticisms and critical debates in urban and planning theory. It also explores the possibilities of developing a new understanding of environmental protection in planning for Turkey through the Mount Ida case. While market-based conservation approaches, such as ecosystem services valuation, expand the scope of traditional conservation, they are criticized for leading to 'green grabbing' practices in newly designated areas and promoting unlimited exploitation of nature outside these areas. Concurrently, the spatialization of degrowth strategies aimed at overcoming socio-environmental crises and new inequalities, and the necessity of shifting planning away from economic growth focus towards a post-growth planning practice, are discussed. In Turkey, the prioritization of economic growth and the centralization of decision-making processes have led to the concurrent carrying out of destructive activities and 'greener' activities, even within protected regions. While Mount Ida National Park and its southern slope are partially protected, new mining activities in the peripheral forests and geothermal energy-based activities along the coastline continue with the allocation of forests and water resources in a way that leads to environmental pollution and inequalities due to export-oriented policies. Even if a new project of ecosystem services for the forests of Mount Ida is introduced, new inequalities may emerge at different scales due to isolated conservation decisions, economic growth-oriented targets and policies besides legal inadequacies. In this line, the thesis proposes 'conservation scales' for the area with 'biodiversity impact chain' analysis as a tool to expose inequalities, propose and politicize post-growth alternatives

    Design Alternatives of Light Shelves Using Altmann Linkage

    This paper proposes a novel new light shelf design with Altmann linkage using its kinetic principles: geometry and rotational angles. As previous studies explain a light shelf’s design in two ways: static and movable, the proposed one in this study has the potential to track the path of the sun due to its diagonal movement. The primary purpose is to direct the light shelf to intermediate directions, such as southeast and southwest, by utilizing the geometric properties of the Altmann linkage. The study explains how to dimension the links, calculate rotation angles, and model this device in Relux to test its daylight performance on specific dates in a year. A total of nine variations were analyzed during the three phases of design. They include shelf forms such as a rectangle, two rectangles, two squares, and varying link lengths, which define the distance to the windowsill. The final set of variations with two-square forms moving west and east successfully satisfied with sDA values as 71.52%, 72.99% (w), 75.92% (e); with ASE values as 8.83%, 8.56% (w), and 8.22% (e). This best design of Altmann linkage would be beneficial as an adaptive façade module that can direct daylight inside and achieve proper shading throughout the day and year. © 2024 The Author(s)

    Alternative Modelling Approaches for Frp Confined Concrete Columns

    Externally applied fiber-reinforced polymer (FRP) jackets represent a widely employed and effective technique for the strengthening and retrofitting of existing structures. This method not only provides a rapid solution but also offers enhanced ease of application, whether using single-type or hybrid FRP configurations. These materials contribute to the improvement of structural capacity while simplifying the implementation process. However, predicting the precise enhancements in load-carrying capacity and ductility of FRP-confined systems remains a challenging task. This highlights the necessity for a deeper understanding of these applications and the refinement of associated design methodologies. This thesis explores two primary approaches for modelling the behavior of FRPconfined concrete: finite element analysis (FEA) and analytical modelling. In the finite element framework, both the Concrete Damage Plasticity Model (CDPM) and the Drucker-Prager Plasticity Model (DPPM) are employed. However, due to the fundamentally different confinement mechanisms between FRP and steel, the direct application of these models to FRP-confined concrete is not fully adequate. Therefore, several modifications are introduced to improve the predictive accuracy of these models for both single-type and hybrid FRP-confined columns. Although various analytical models for FRP-confined concrete are available in the literature, there is a noticeable limitation in the research concerning the behavior of hybrid FRP systems. This study critically examines existing models and explores their adaptation for both single-type and hybrid FRP systems, with the results benchmarked against experimental data.Fiber takviyeli polimer (FRP) ile dıştan sargılama, mevcut yapıların güçlendirilmesi ve yenilenmesi için etkili ve hızlı bir yöntem olarak yaygın şekilde kullanılmaktadır. Bu malzemeler, tek tip veya hibrit FRP ceketler şeklinde uygulanabilir ve yapının taşıma kapasitesini artırmanın yanı sıra uygulama sürecini kolaylaştırmaktadır. Ancak, FRP sargılı sistemlerin sunduğu taşıma kapasitesi ve süneklik artışlarının yeterince öngörülememesi, bu yöntemlerin daha iyi anlaşılmasını ve tasarım ilkelerinin geliştirilmesini gerekli kılmaktadır. Bu tezde, FRP sargılı betonun davranışını modellemek için sonlu elemanlar analizi ve analitik modelleme olmak üzere iki farklı yaklaşım incelenmiştir. Sonlu elemanlar analizinde, Beton Hasar Plastisite modeli ve Drucker-Prager Plastisite modeli kullanılmıştır. Ancak, FRP'nin sağladığı sargılama basıncı, çelik sargılama basıncından farklı bir mekanizmaya sahip olduğu için bahsedilen plastisite modelleri doğrudan uygulanması mümkün olmamaktadır. Bu nedenle, hem tek tip hem de hibrit FRP sargılı kolonlar için modellerin doğruluğunu artırmaya yönelik iyileştirmeler yapılmıştır. Ayrıca, literatürde FRP sargılı betonun davranışını açıklayan birçok analitik model yer almakla birlikte, hibrit sistemlerin davranışını öngörmeye yönelik çalışmalar sınırlıdır. Bu kapsamda, mevcut analitik modeller değerlendirilmiş ve hem tek tip hem de hibrit LP sistemlerde yaygın olarak bilinen modellerin uygulanabilirliği araştırılmıştır. Elde edilen sonuçlar, deneysel verilerle karşılaştırılarak değerlendirilmiştir

    Design, prototyping and testing of modified altmann linkage networks To be used as deployable structures

    Bu tez çalışması, iki değiştirilmiş Altmann mekanizması ve bunlardan elde edilen katlanabilir ağ yapılar üzerine odaklanmaktadır. Değiştirilmiş mekanizmalar, genel doğru-simetrik Bricard mekanizmasının özel durumlarıdır. Bu mekanizmalar, orijinal Altmann mekanizmasına göre yapısal avantajlara sahip olup değiştirilmiş mekanizmalardan biri özgündür. Her iki mekanizmanın kinematik analizleri yapılmıştır. Bu mekanizmalarla oluşturulan katlanabilir ağ yapılar sınıflandırılmıştır. Ağ yapıların her birinin özellikleri ve hareket davranışlarını açıklamak için iki mekanizmadan biri seçilmiştir. Bazı ağ yapıların üretilmiş prototipleri sunulmuştur. Kubbe benzeri bir katlanabilir ağ yapısı vaka çalışması olarak seçilmiştir. Vaka çalışması, bir katlanabilir çadır mekanizmasının tasarımını, prototip üretimini ve imalat testlerini kapsamaktadır. İki fiziksel prototip tasarımı, yapısal benzetim çalışmaları ve bazı test sonuçları sunulmuştur. Tasarım geometrisinin parametrik modeli geliştirilerek Microsoft Excel®'de tanımlanmıştır. Katı model ortamına (Solidworks®) bu parametrik modelin aktarımını sağlamak için Excel eklentisi olarak özel bir API geliştirilmiştir. İlk prototipin statik yük koşulları altındaki doğrusal olmayan elastik deformasyon davranışını incelemek için benzetimler koşturulmuştur. Ön- ve son-işlemci olarak MSC Apex®, çözücü olarak ise MSC Nastran® kullanılmıştır. Sunulan sonlu eleman modelleri MSC Apex®'in API kütüphanesi kullanılarak oluşturulmuştur. Bu görev için bir başka özel API iş-akışı oluşturulmuştur. İlk prototipin üretimi ve test sonuçları sunulmuş olup ikinci prototipin üretim ve testleri ileriki çalışmalardır.This thesis focuses on the two modified Altmann linkages and deployable networks obtained from them. The modified linkages are special cases of general line-symmetric Bricard linkages. These linkages provide constructional advantages compared to the original Altmann linkage, and one of the modified linkages is novel for the literature. Kinematic analyses of both linkages are performed. The deployable networks composed of these linkages are classified. One of the two linkages is selected to describe the characteristics and motion behavior of each network. Built prototypes of some networks are presented. A dome-like deployable network is selected as a case study. The case study covers the design, prototyping and testing of a deployable tent mechanism. Two physical prototype designs, structural simulations and some test results are presented. The parametric model for the geometry of the designs is developed and implemented in Microsoft Excel®. A custom API is designed to integrate the parametric model into the solid modeling environment (Solidworks®) as an Excel add-in. Simulations are run to explore the nonlinear elastic deformation behavior of the first prototype under static load conditions. MSC Apex® is used as the pre- and post-processor, whereas MSC Nastran® is the solver. The presented finite element models are generated utilizing the API library of MSC Apex®. Another custom API workflow is introduced for this task. The construction and test results of the first prototype are presented, whereas the second prototype is to be built as a future study

    Multi-Layer Absorber Based on Plasmonic Resonances for Photovoltaic Applications at Visible Spectra

    This paper introduces a broadband absorber based on a multilayered, double-cylindrical-shaped metamaterial, numerically characterized for its performance. The structure comprises four interacting layers that generate plasmonic resonances. CST microwave simulations were conducted to analyze its absorption characteristics. The results demonstrate that the proposed metamaterial absorber achieves 99% absorption at 847 nm frequency region and 98% absorption in the 500-1200 nm frequency region. Additionally, polarization dependency analysis confirms that the absorber performs as a perfect, polarization-independent absorber across the studied frequency range. It exhibits high absorption in both TE and TM modes and remains unaffected by polarization or variations in the incident angle. Numerical simulations reveal that the absorption performance is driven by a combination of Fabry–Perot resonance effects, localized surface plasmons, and propagating surface plasmons. In summary, the proposed metastructure demonstrates omnidirectional absorption, polarization independence, and wide-angle incident absorption. This design shows significant potential for applications in photodetectors, active optoelectronic devices, and sensors. © IJCESEN

    Flexibility of Robots: Design, Modeling and Control

    Tüm malzemeler bir miktar esneklik sergiler. Sonuç olarak, herhangi bir malzemeden yapılmış bir robot manipülatörü bu yapısal esnekliği miras alır. Tipik olarak esnek robotların konumlandırma doğruluğu daha düşüktür. Bununla birlikte esneklik, kontrollü bir mekanik empedansa ve fiziksel insan-robot etkileşimlerinde artırılmış güvenliğe katkıda bulunduğundan avantajlı da olabilir. Temel zorluk esnek bir robotun giriş/çıkış performansını arttırmaktır. Bu performans büyük ölçüde üç temel bileşene dayanır: robotun mekanik yapısı, denetleyicisi ve denetleyici tarafından kullanılan matematiksel modelin doğruluğu. Bu bileşenlerde yapılan herhangi bir değişiklik veya ayarlama, genel giriş/çıkış performansını etkiler. Bu tez şu üç temel bileşeni kapsamaktadır: model, tasarım ve kontrol. İlk bölüm, çalışma sırasındaki titreşimleri tahmin etmek için aşırı kısıtlanmış bir paralel robot manipülatörünün direngenlik modelinin tasarlanmasına ayrılmıştır. İstenilen yörüngenin (veya girdinin) bu tahminlere göre ayarlanması, çıktı üzerindeki titreşim etkilerinin azaltılmasını mümkün kılar. İkinci kısım da istenilen mekanik empedans için esnek bir mafsal geliştirilir. Bu mafsal, küçük yükler altında esneklik göstermez, ancak etkileşim yükleri arttıkça yu\-mu\-şa\-yan davranışla esner. Mafsal, güvenli fiziksel insan-robot etkileşimlerini sağlarken robotun konumlandırma performansını düşük burulma seviyelerinde korur. Son olarak esnek robot için tasarlanan esnek mafsalı içeren bir denetleyici çerçevesi tasarlanmıştır. Kontrolcü, robotun mekanik durumları (sert ve esnek) arasında istikrarlı geçişler yapma ve istenen görevleri gerçekleştirme (yörünge izleme, istenen empedansı görüntüleme, kuvvet/konum izleme ve güvenliği sağlama) yeteneğine sahiptir.All of the materials exhibit some flexibility. Consequently, a robot manipulator constructed from any material inherits this structural flexibility. Typically, flexible robots have lower positioning accuracy. However, flexibility can also be advantageous, as it contributes to a controlled mechanical impedance and enhanced safety in physical human-robot interactions. The primary challenge is enhancing the input/output performance of a flexible robot. This performance largely relies on three key components: the robot's mechanical structure, its controller, and the accuracy of the mathematical model employed by the controller. Any alterations or adjustments made to these components impact the overall input/output performance. This dissertation encompasses these three key components: model, design, and control. The first section is dedicated to devising a stiffness model of an over-constrained parallel robot manipulator to estimate the vibrations during operation. Adjusting the desired trajectory (or input) based on these estimations makes it possible to mitigate the vibration effects on the output. The second part develops a flexible joint for the desired mechanical impedance. This joint exhibits no compliant deflection under small loads but deflects with a softening behavior as interaction loads increase. The joint maintains the robot's positioning performance at low torsion levels while ensuring safe physical human-robot interactions. Finally, a controller framework is designed for the flexible robot that contains the designed flexible joint. The controller is capable of stable transitions between the mechanical states of the robot (rigid and flexible) and achieving the desired tasks (trajectory tracking, displaying the desired impedance, force/position tracking, and ensuring safety)

    Development and Characterization of Affinity-Maturated Nanobodies Targeting B-Lymphocyte Antigen Cd19

    B-Cell Acute Lymphoblastic Leukemia (B-ALL) is a subtype of leukemia characterized by the uncontrolled proliferation of B-lymphocytes. The diagnosis of B-ALL relies on blood counts, immunophenotyping, cytogenetic analysis, and molecular tests. Treatment strategies for B-ALL include chemotherapy, radiotherapy, allogeneic bone marrow transplantation, and stem cell transplantation. In the diagnosis and treatment of B-ALL, antibodies or antibody fragments specific to CD19, a protein known as a B-lymphocyte antigen, are commonly used. However, producing antibodies specific to targeted antigens is often cost-prohibitive. Nanobodies, which constitute the heavy chain variable region of heavy-chain only camelid antibodies have been elucidated to be more advantageous. Three nanobodies (Nbs), which were chosen from a natural Nb-expressing phage display library in the study of (H. Wang et al. 2021a), with affinity to CD19 were tested. The nanobodies, which are derived from the heavy-chain antibodies of camelids and constitute the single-domain heavy variable region of the antibodies, were maturated in-silico. Two parent nanobodies, out of three, were chosen alongside four variants based on their in-silico performance. The purpose of this study is to verify the applicability of our in-house affinity maturation protocol (structure prediction, filtering, and scoring), by in-vitro production and functional characterization of affinity maturated variants against their parents nanobodies. Moreover, it is reflected on multiple paradigms concerning in-silico affinity maturation of nanobodies, and how models fare under the reality of experimentation.B-Hücre Akut Lenfoblastik Lösemi (B-ALL), B-lenfositlerin kontrolsüz çoğalması ile karakterize bir lösemi alt tipidir. B-ALL tanısı kan sayımları, immünfenotipleme, sitogenetik analiz ve moleküler testlere dayanır. B-ALL tedavi stratejileri arasında kemoterapi, radyoterapi, allojenik kemik iliği nakli ve kök hücre nakli bulunur. B-ALL'nin tanı ve tedavisinde, bir B-lenfosit antijeni olarak bilinen CD19 proteinine özgü antikorlar veya antikor parçaları yaygın olarak kullanılmaktadır. Ancak, hedef antijenlere özgü antikor üretimi genellikle yüksek maliyetlidir. Yalnızca ağır zincirli deve antikorlarının ağır zincir değişken bölgesini oluşturan nanokütlelerin (nanobody, Nb) daha avantajlı olduğu gösterilmiştir. (H. Wang ve ark., 2021a) çalışmasında, doğal bir Nb-ekspresyonlu faj gösterim kütüphanesinden seçilen ve CD19'a afinitesi olan üç nanokütle (Nb) test edilmiştir. Deve antikorlarının ağır zincirlerinden türetilen ve antikorların tek zincirli ağır değişken bölgesini oluşturan bu nanokütleler in-silico olarak olgunlaştırılmıştır. Üç nanokütleden ikisi, in-silico performanslarına dayanarak dört varyantla birlikte seçilmiştir. Bu çalışmanın amacı, kurum içinde geliştirilen afinite olgunlaştırma protokolünün (yapı tahmini, filtreleme ve derecelendirme) uygulanabilirliğini, afinitesi olgunlaştırılmış varyantların in-vitro üretimi ve fonksiyonel karakterizasyonuyla ana nanokütlelere karşı doğrulamaktır. Ayrıca, nanokütlelerin in-silico afinitesi olgunlaştırılmasıyla ilgili çeşitli paradigmalar ve modellerin deneysel gerçeklik karşısındaki durumu üzerine değerlendirmelerde bulunulmuştur
