I joined the Institut Pasteur in September 1962 for what was supposed to be a short internship, but I worked
there for 33 years.
At the time, I had left medicine and the preparation for the “Internat” fascinated by the lectures given by
Jacques Monod at the Faculté des Sciences. He accepted me in his department, despite the fact, as he told me,
that I “belonged to medicine and to the female sex”. It was there that I prepared my doctorate in science under
the supervision of my husband, Henri Buc. I then joined François Jacob’s laboratory, which I left in 1992.
I do not want to recount here the magnificent career of François Gros. He was successively an eminent
researcher, attracting many great scientists to him and collaborating with the best international specialists of
the time; then director of the Institut Pasteur, professor at the Collège de France, member of the Académie
des Sciences, of which he became permanent secretary; and scientific advisor to François Mitterrand.
In fact, I hardly knew him at all during that time.
It was only after my retirement in the early 2000s that I had the great good fortune to interact with him, an
interaction that lasted until his death. I would like to mention three moments that were particularly important
to me:
(1) My new involvements brought me closer to medicine and Africa. At the time, I was actively involved in
a Franco-African medical association that wanted to organize neonatal screening for sickle cell anaemia in the
Democratic Republic of Congo. This genetic disease, the most common in the world, is terribly painful and
potentially fatal in homozygotes (children affected in Africa rarely live beyond the age of five) and is linked
to a mutation in a gene that encodes one of the haemoglobin chains. To carry out this screening, we needed
specialized equipment and funding. Stuart Edelstein, a member of the association’s scientific advisory board
had just published a book, Biology of a Myth, about the cultural implications of sickle cell anaemia. At his
suggestion, I contacted François Gros in 2004. He, at the time honorary permanent secretary of the Académie
des sciences, reviewed with much good will the project I submitted to the Académie. Thanks to him, we
received major grants for two consecutive years.
(2) François Gros then generously associated me with COPED (Committee for Developing Countries), which
he had created within the Academy in 1996. In order to promote scientific exchanges with the countries of
North and sub-Saharan Africa, COPED organizes forums on specific issues (science, health, energy resources,
demography, etc.). I took part in two of these, as François had asked me to write the proceedings. This
heavy workload enabled me to meet African leaders, academics, scientists and members of Academies, and
to become aware of their problems and possible advances in the context of each country. The 2009 forum
focused on education, from primary school to university. The 2011 forum was entitled “Water, Health and
Agriculture in Africa”. Participants endeavoured to take stock of the water resources available in their own
countries. Meanwhile, French and African researchers reported on cutting-edge methodological approaches
to sanitation and risk prevention.
My commitment to the work of COPED lasted almost three years. It enabled me to devote myself to more
human investments than those offered by pure research. Very gradually, by listening to François and working
with him, I really got to know him. I appreciated his selfless love for science and his often exhausting efforts to
make forums useful in terms of their spin-offs, often in fields far removed from his initial training.
(3) It was then that I returned to medicine, focusing on its socio-cultural aspects in collaboration with
Professor Frédéric Galactéros. Professor Galactéros, a leading specialist in sickle cell anaemia, asked me to
interview patients about their experiences, their feelings and their methods of coping with the interferences
between their work and their illness. These interviews became the subject of a book entitled La maladie
génétique au quotidien. La drépanocytose, histoires de vies. Once again, François’ collaboration was invaluable:
he encouraged us to keep to the spirit of our approach (giving a voice to those who usually do not speak out);
he read over my texts and made suggestions to make the book more accessible. He wrote a beautiful preface for
this first book (there was a second one) and helped with its publication at the Presses Universitaires de France.
My relationship with François Gros was a lasting one. Quite often, right up to the end, I would push the door
to his new office on Quai Conti, at the top of the tower where one stands close to both the Louvre and the sky.
I kept him updated on my new projects concerning sickle cell patients. But our conversations were often more

personal. I would tell him about my questions. He would tell me about the many requests for his services that
he was trying to meet. We talked about the world around us. . .
François had a very rare and precious gift: the ability to welcome all kinds of people, to listen to them with
kindness and to bring out the best in them. With his usual modesty, he used to tell me that he was “just an
impresario”. On the contrary, Henri Buc and I are deeply convinced that this intelligent and generous heart,
which never said no, was the foundation and driving force behind a humanistic vision of science.
After my conversation with him, I would leave the Institut de France in a serene frame of mind. It had
become a tradition for me to cross the Pont des Arts and then walk joyfully through the Cour Carrée du Louvre.

Claude Combes left us on July 8, 2021. He was
eighty-five years old. In recent years, his health had
kept him away from the activities of the Academy of
Sciences where he had been elected correspondent
in 1996, then member in 2004.
At first tempted by literary studies after a bac-
calaureate in philosophy, he finally chose the life
sciences, brilliantly obtained the agregation, and
then defended his thesis at the University of Mont-
pellier under the direction of Louis Euzet, creator of
the “Montpellier–Perpignan” school of parasitology.
Another great parasitologist, Jean-Antoine Rioux,
inspired his first steps as a researcher in the
Pyrenees. In 1961, Claude was appointed assis-
tant at the University of Perpignan, then a scientific
university college. Throughout his life, he remained
faithful to this University despite numerous national
and international requests.
Claude Combes was a teacher-researcher with an
exceptional charisma, a master unanimously appre-
ciated by his students, a talented lecturer, inhabited
by the passion of transmitting scientific knowledge,
its construction, its interrogations.
His research and that of his laboratory have led
to innovative advances in the transmission, ecology
and evolution of parasites. They have largely con-
tributed to the knowledge of the relationships be-
tween the genetic diversity of pathogens and their
medical properties, based on the model of schisto-
somiasis, the second largest parasitic endemy in the
world with more than 250 million people affected in
tropical regions.
Claude Combes was one of the first researchers to
demonstrate the role of parasites in the structuring,
functioning and evolution of life. He was able to draw
attention to these usually invisible elements of nat-
ural communities, constantly interacting with other
living beings in singular associations, often revealing
the mechanisms of natural selection.
He developed the notion of sustainable interac-
tion to define the durability of the host–parasite re-
lationship as opposed to the predator–prey relation-
ship. He symbolized the two stages—encounter and
compatibility—of the constitution of a host–parasite
system by the notion of filters, or diaphragms, reg-
ulating encounters and compatibilities: diaphragms
that the host strives to keep closed while the parasite
keeps opening them. This inspired him to study host–
parasite interaction through the prism of evolution-
ary theory, with both partners engaged in an unlim-
ited arms race.
Thus considered, sustainable interactions repre-
sent a textbook case of coevolution. They are con-
structed through a sequence of reciprocal selective
pressures: the selection of encounter genes in the
parasite is matched by the selection of avoidance
genes in the host; the selection of exploitation genes
in the parasite is matched by the selection of para-
site elimination genes in the host. This Darwinian
approach to the pathogen–host relationship has be-
come established in the field of infectious diseases,
and even in the understanding of the dynamics of
cancer cells.
Claude Combes and his team discovered that
schistosomes multiply by indefinite replication of
sporocysts in their mollusk vectors. This replication
gives rise to successive generations of larvae, which
allowed the laboratory to achieve a world first: the ex-
perimental cloning of a parasitic metazoan by micro-
surgical transplantation, a process that allows the in-
definite conservation of specific genotypes, selected
for their medical interest.
The laboratory of Claude Combes was among
the first to perceive the interest of molecular bi-
ology tools in the field of the evolutionary ecol-
ogy of parasites. This approach led to the idea that
Schistosoma, a genus whose species are all human
pathogens, comprises four distinct lineages, and that
these schistosomes parasites of humans result from
lateral transfers having occurred about 2 million
years ago, from species that evolved either in ro-
dents or in ungulates. It also appeared that the schis-
tosomes of the New World have a recent origin,
linked to the displacement of infested African pop-
ulations from the 16th century on. Finally, a phylo-
genetic study led the laboratory to describe a new
species of schistosome parasite of humans, Schisto-
soma guineensis, present around the Gulf of Guinea.
Author of more than 200 articles and a dozen
books, Claude Combes has directed some thirty the-
ses. He has participated, often as president, in about
fifty thesis juries and in numerous evaluation com-
mittees, always highly appreciated for the extent of
his knowledge and the quality of his opinions. He
launched the laboratory of animal biology at the Uni-
versity of Perpignan, associated with the CNRS and
a WHO collaborating center, a laboratory whose in-
fluence is still developing today. Claude Combes was

awarded several scientific prizes and recognitions.
He was, in addition to member of the Academy of
Sciences, a member of the Academy of Agriculture of
France.
Several of his books have particularly marked
the parasitic ecology [1–6]. Among them, “Interac-
tions durables. Écologie et évolution du Parasitisme”
translated as “Parasitism: Ecology and Evolution of
Intimate Interactions” [1]. In the preface to this trans-
lation, David Simberloff wrote: “That one author can
draw so easily and appropriately on examples from
plants and animals, microorganisms and macroor-
ganisms, genes and ecosystems, and the medical and
molecular literature, is the mark of a virtuoso scien-
tist”. In a review of “The Art of being a Parasite” [3],
Robert Poulin stated in the journal Parasitology: “We
know that parasites are wonderful creatures. But con-
vincing the average person of this simple fact is a
monumental challenge. On the whole, Combes suc-
ceeds superbly”.
This “monumental challenge”, Claude Combes
will still take it up brilliantly in his “Darwin, dessine-
moi les hommes” [5], book which approaches the his-
tory of the living, “a grandiose history which gave
to the human beings the capacity to ask questions
without losing the one to be amazed”. This book was
awarded the prize “La Science se livre” in 2007.
Last year, a PhD student, Rasha Shraim, titled a ca-
reer column in Nature: “How philosophy makes me
a better scientist”. Claude would certainly have ap-
preciated it: he knew so well how to take a step back,
to encourage reflection, to be enthusiastic about the
lessons he drew from the observation of the Pyrenean
flora and fauna. . . as well as from the mysteries of the
biological cycle of schistosomes.

Un moment tenté par des études littéraires après
un baccalauréat de philosophie, il avait finalement
choisi les sciences naturelles, obtenu brillamment
l’agrégation, puis soutenu sa thèse à l’Université de

sitologie. Un autre grand parasitologue, Jean-Antoine

fidèle à cette Université en dépit de nombreuses sol-

Claude Combes a été un enseignant-chercheur au
charisme exceptionnel, un maître unanimement ap-
précié par ses étudiants, un conférencier talentueux,
habité par la passion de transmettre le savoir scien-
tifique, sa construction, ses interrogations.
Ses recherches et celles de son laboratoire ont
conduit à des avancées novatrices sur la transmis-
sion, l’écologie et l’évolution des parasites. Elles ont
largement contribué à la connaissance des relations
entre la diversité génétique des agents pathogènes
et leurs propriétés médicales, à partir du modèle
des schistosomoses, deuxième endémie parasitaire
au monde avec plus de 250 millions de personnes af-
fectées dans les régions tropicales.
Claude Combes a été un des premiers chercheurs
à démontrer le rôle des parasites dans la structura-
tion, le fonctionnement et l’évolution du vivant. Il
a su attirer l’attention sur ces éléments habituelle-
ment invisibles des communautés naturelles, en in-
teraction constante avec les autres êtres vivants dans
des associations singulières, souvent révélatrices des
ressorts de la sélection naturelle.
Il développa la notion d’interaction durable pour
définir la pérennité de la relation hôte–parasite par
opposition à la relation prédateur–proie. Il symbol-
isa les deux temps — rencontre et compatibilité —
de la constitution d’un système hôte–parasite par
la notion de filtres, ou diaphragmes, régulant les
rencontres et les compatibilités : des diaphragmes
que l’hôte s’efforce de maintenir fermés alors que
le parasite n’a de cesse de les ouvrir. Cette notion
lui inspirera l’étude de l’interaction hôte–parasite

à travers le prisme de la théorie évolutionniste, les
deux partenaires se livrant à une course illimitée aux
armements.
Ainsi considérées, les interactions durables
représentent un cas d’école de coévolution. Elles
se construisent au fil d’un enchaînement de pres-
sions sélectives réciproques : à la sélection de gènes
de rencontre chez le parasite, répond la sélection
de gènes d’évitement chez l’hôte ; à la sélection
de gènes d’exploitation chez le parasite, répond la
sélection de gènes d’élimination du parasite chez
l’hôte. Cette approche darwinienne de la relation
pathogène–hôte s’est imposée dans le domaine des
maladies infectieuses, voire dans la compréhension
de la dynamique des cellules cancéreuses.
Claude Combes et son équipe ont découvert que
les schistosomes se multiplient par réplication in-
définie des sporocystes dans les mollusques vecteurs.
Cette réplication donne naissance à des générations
successives de larves, ce qui permit au laboratoire de
réaliser une première mondiale : le clonage expéri-
mental d’un métazoaire parasite par transplantation
microchirurgicale, un procédé qui autorise la con-
servation indéfinie de génotypes précis, sélectionnés
pour leur intérêt médical.
Le laboratoire de Claude Combes fut parmi les
premiers à percevoir l’intérêt des outils de la bi-
ologie moléculaire dans le domaine de l’écologie
évolutive des parasites. On doit à cette dé-
marche l’idée que Schistosoma, genre composé
d’espèces toutes pathogènes de l’homme, com-
prend quatre lignées distinctes et que ces schisto-
somes parasites de l’homme sont issus de trans-
ferts latéraux survenus il y a environ 2 millions
d’années, à partir d’espèces qui ont évolué soit
chez des rongeurs, soit chez des ongulés. Il apparut
aussi que les schistosomes du Nouveau Monde
ont une origine récente, liée aux déplacements
de populations africaines infestées à partir du
XVIesiècle. Enfin, une étude phylogénique a con-
duit le laboratoire à décrire une nouvelle espèce
de schistosome parasite de l’homme, Schistosoma
guineensis, présente sur le pourtour du Golfe de
Guinée.
Auteur de plus de 200 articles et d’une douzaine
d’ouvrages, Claude Combes a dirigé une trentaine
de thèses. Il a participé, souvent comme président,
à une cinquantaine de jurys de thèse et à de nom-
breuses commissions d’évaluation, toujours très
apprécié pour l’ampleur de ses connaissances et la
qualité de ses avis. Il a fondé le laboratoire de bi-
ologie animale de l’université de Perpignan, associé
au CNRS et centre collaborateur de l’OMS, un lab-
oratoire dont le rayonnement se développe encore
de nos jours. Récompensé, ainsi que ce laboratoire,
par plusieurs reconnaissances et prix scientifiques,
Claude Combes était, outre membre de l’Académie
des sciences, membre de l’Académie d’agriculture de
France.
Plusieurs de ses livres ont particulièrement mar-
qué l’écologie parasitaire [1–6]. Parmi eux, « In-
teractions durables. Écologie et évolution du Para-
sitisme », traduit sous le titre de « Parasitism : Ecology
and Evolution of Intimate Interactions » [1]. Dans la
préface à cette traduction, David Simberloff écrira :
« Qu’un auteur puisse s’appuyer aussi facilement
et de manière aussi appropriée sur des exemples
de plantes et d’animaux, de microorganismes et de
macroorganismes, de gènes et d’écosystèmes, ainsi
que sur la littérature médicale et moléculaire, est la
marque d’un scientifique virtuose ». Dans une anal-
yse de « The Art of being a parasite » [3], Robert Poulin
précisera dans la revue Parasitology : « Nous savons
que les parasites sont des créatures merveilleuses.
Mais convaincre le citoyen moyen de ce simple fait
est un défi monumental. Au total, Combes y parvient
superbement ».
Ce « défi monumental », Claude Combes le
relèvera encore avec brio dans son « Darwin, dessine-
moi les hommes » [5], livre qui aborde l’histoire du
vivant, « une histoire grandiose qui a donné aux hu-
mains la capacité de poser des questions sans perdre
celle de s’émerveiller ». Un livre distingué par le prix
“La Science se livre” en 2007.
L’an dernier, une étudiante PhD, Rasha Shraim,
intitulait une chronique « carrières » de la revue
Nature : « Comment la philosophie fait de moi un
meilleur scientifique ». Claude aurait certainement
apprécié : il savait si bien prendre du recul, inciter à
la réflexion, s’enthousiasmer des leçons qu’il tirait de
l’observation de la flore et de la faune pyrénéenne. . .
comme des mystères du cycle biologique des schis-
tosomes.

It is obvious, but sometimes it needs to be re-
minded, researchers need data to generate and test
novel hypotheses. The more the dataset is of high
quality, the more robust the scientific results and
predictions are produced. And the more widely the
data is shared, the more verifiable and reproducible
the results are. In human research, many initiatives
have been made to produce large-scale data, ranging
from national registries that include the entire pop-
ulation, to the establishment of cohorts of varying
sizes. In 2006, the UK Biobank biomedical database
and resource was created, and it has revolutionized
research on several levels, particularly because the
data is accessible to the scientific and medical com-
munities [1].
The initial budget of UK Biobank was £62 mil-
lion, funded by the UK government and the Well-
come Trust. The objective was to collect data on
500,000 participants aged 40–69 years old over four
years and then follow them for at least 30 years.
The database was opened to researchers in March
2012, hosting demographic, medical, psychologi-
cal, socio-economic data (from interviews or online
questionnaires), biological samples (e.g., urine and
blood), genetic data and imaging data. Ten years
later, more than 28,000 researchers had access to this
data.
Why is UK Biobank revolutionizing research? Un-
like national registries where data access is limited,
researchers from all countries can access UK Biobank
data after their research project was approved and
their IT security to host the requested data was ver-
ified. Although genetic data is considered sensitive,
UK Biobank has obtained informed consent from
participants to share it. There is a periodical update
sent to all registered researchers to ask them to re-
move participants who chose not to share their data
anymore.

More than five petabytes of genetic data are
available including >500,000 microarrays, >400,000
whole-exome and >200,000 whole-genome se-
quences [2]. These data allow researchers to ex-
plore the genetic contribution to many traits
and diseases. Among the major results, this re-
source has been key to estimate the heritabil-
ity of more than 4000 phenotypic traits and are
available online at the Benjamin Neale laboratory
(https://nealelab.github.io/UKBB_ldsc/index.html).
Genetic variations have been identified as risk fac-
tors and predictive models have been proposed for
diseases such as diabetes, cardiovascular disease and
cancer [3]. You can interrogate if a gene is associated

Epidemiological and genetic data are also linked
to imaging data. The goal is to have magnetic reso-
nance images (MRI) of the brain, heart, and abdomen
for 100,000 participants (already 50,000 available).
The combined analysis of genotyping and brain MRI
data has allowed, among other things, to the estima-
tion of the heritability of interindividual differences
in brain anatomy [4, 5].
Regarding COVID-19, all registered researchers
received messages informing them of the collec-
tion of new data collected in relation to COVID-19
and encouraging them to submit research projects
on the pandemic. More than 740 researchers re-
sponded, and more than 148 papers were published.
A specific UK Biobank study also collected new data
on 20,000 volunteers who were either original UK
Biobank participants or their children or grandchil-
dren over the age of 18. A total of 6.6% of partici-
pants had already been infected by May/June 2020
and this rate increased to 8.8% by the end of No-
vember 2020. This study was one of the first to show
that antibodies produced following natural infec-
tion can protect most people from further infection
for at least 6 months (The UK Biobank SARS-CoV-

I have only scratched the surface of the find-
ings and the possibilities offered by UK Biobank. I
have not mentioned all the richness and reliabil-
ity of the data as well as the next objectives such
as the linkage with other registries such as death,
cancer, . . . I also omitted the ability to share re-
sults of the research projects directly via the UK
Biobank portal, and the description of the limita-
tions and biases of representativeness of UK Biobank
which are well documented [6]. All this informa-
tion is available on the UK Biobank website https://
www.ukbiobank.ac.uk/. Again, for scientists to work,
there is a need for large amounts of high qual-
ity and accessible data. There is an urgent need to
support such initiatives in other countries to repli-
cate the UK Biobank results, to increase the diver-
sity of people studied and to detect associations
that are specific to countries with different health
systems.

Plus les données sont de qualité et en grande quan-
tité, plus les résultats scientifiques et les prédictions
sont solides. Plus les données sont partagées, plus les
résultats obtenus sont vérifiables et réplicables. Con-
cernant la recherche sur la population humaine, de
nombreuses initiatives ont été mises en place afin
d’obtenir des données à grande échelle allant de reg-
istres nationaux incluant l’ensemble de la popula-
tion, à l’établissement de cohortes de plus ou moins
grandes tailles. En 2006, la base de données biomédi-
cales et de ressources UK Biobank est créée et a
révolutionné la recherche à plusieurs niveaux, en
particulier en rendant les données accessibles aux
scientifiques [1].
Le budget initial de UK Biobank était de 62 mil-
lions de livres, financées par le Royaume-Uni et le
Welcome Trust. L’objectif était de collecter en qua-
tre ans des données sur 500 000 personnes de 40 à
69 ans, puis de suivre ces dernières sur au moins
30 ans. La base a été ouverte aux chercheurs en
mars 2012 et elle héberge, entre autres, des don-
nées démographiques, médicales, psychologiques,
socio-économiques (issues d’interviews ou de ques-
tionnaires en ligne), des prélèvements biologiques

UK Biobank révolutionne la recherche car con-
trairement aux registres nationaux dont les don-
nées sont peu accessibles, la base est accessible aux
chercheurs de tous les pays après validation de leur
projet de recherche et vérification de la sécurité in-
formatique dont ils disposent pour héberger les don-
nées qu’ils souhaitent récupérer. Bien que les don-
nées génétiques soient considérées comme sensi-

Les données épidémiologiques et génétiques sont
aussi reliées à des données d’imagerie. L’objectif
de UK Biobank est d’avoir des images par réso-
nance magnétique (IRM) du cerveau, du cœur et de
l’abdomen pour 100 000 participants (50 000 don-
nées sont déjà accessibles). L’analyse combinée des
données de génotypage et d’IRM cérébrales a permis,
entre autres, d’estimer l’héritabilité des différences
interindividuelles pour l’anatomie du cerveau [4, 5].
Concernant la recherche sur la COVID-19, tous
les chercheurs dont les projets de recherche avaient
été acceptés par UK Biobank recevaient des mes-
sages les alertant du recueil de nouvelles données
en rapport avec la COVID-19 et les incitant à dé-
poser de nouveaux projets de recherche. Plus de 740
chercheurs ont répondu et plus de 148 articles ont
été publiés. Une étude ancillaire de UK Biobank a
aussi collecté des données sur 20 000 volontaires
qui étaient soit des participants originaux de UK
Biobank, soit leurs enfants ou leurs grands enfants

Je n’ai effleuré que la surface des découvertes is-
sues des données de la base UK Biobank. Je n’ai pas
mentionné toute la richesse et la fiabilité des don-
nées accessibles, ainsi que les prochains objectifs.
Par exemple, le chaînage avec d’autres registres, tels
que celui des décès, des cancers, . . . Je n’ai pas non
plus mis en avant le partage des métadonnées issues
des projets (déposées par les chercheurs et accessi-
bles via le portail de UK Biobank), ni fait la descrip-
tion des biais de représentativité de UK Biobank, qui
existent bien évidement mais qui sont bien docu-
mentés [6]. Toutes ces informations sont disponibles
sur le site web officiel de la base : https://www.
ukbiobank.ac.uk/. Je le répète, pour que les scien-
tifiques puissent travailler, il faut des données en
grande quantité, de très bonnes qualités et acces-
sibles. Il est urgent de soutenir de telles initiatives
dans d’autres pays afin de répliquer les résultats is-
sus de UK Biobank, d’augmenter la diversité des per-
sonnes étudiées et de pouvoir détecter des effets spé-
cifiques de certains pays ayant des systèmes de santé
différents.

Since their discovery by Antoni van Leeuwenhoek
in 1674, it has long been believed that bacteria are
only a few µm in length. This remains true in the
vast majority of cases. But, in the last decades, gi-
ant bacteria have been observed, often initially con-
fused with eukaryotic organisms. These bacteria can
reach a size of tens to few hundreds µm. They have
been described in at least four phyla [1, 2]. Some
giants are even visible to the naked eye. In 1993,
Esther Angert et al. discovered a giant bacterium
(Epulopiscium fishelsoni) in the intestinal tract of a
Red Sea surgeonfish (Acanthurus nigrofuscus). Ini-
tially identified as a eukaryotic protist, it was in fact
a sulfur-oxidizing Gram-positive bacterium measur-
ing 200–700 µm in length [3]. In 1997, Heide Schultz
et al. from the Max Planck Institute for Marine Micro-
biology, discovered a giant bacterium in Walvis Bay
(Namibia) from a very unusual oxygen-free marine
sediments, very rich in sulfides and organic matter
mostly formed by dead diatoms. These bacteria mea-
sure 100 to 300 µm in diameter on average, but can
reach 750 µm. To the naked eye, these cocoidal bac-
teria look like white pearls. It is a sulfur-oxidizing bac-
terium named Thiomargarita nabimiensis, oxidizing
hydrogen sulfide (H2S) into elemental sulfur and us-
ing nitrate as electron acceptor. Vacuoles containing
sulfur and nitrate can be visualized, allowing the bac-
teria to survive long periods of nitrate and hydro-
gen sulfide starvation [4]. This bacterium, like others
of this group, also has self-splicing introns, typically
found only in eukaryotes [5].
1. Discovery of Thiomargarita magnifica
Jean-Marie Volland et al. have just published in Sci-
ence the discovery of a new giant bacterium that they

call Thiomargarita magnifica [6]. This is a collabora-
tive work between French teams from the University
of the West Indies, the Museum of Natural History,
Sorbonne University, and American teams from the
University of California, Berkeley. In 2009, one of the
co-authors, Dr. Olivier Gros, a biologist from the Uni-
versity of the West Indies, France, has observed white
filaments of 0.9 cm to 2 cm attached to the sub-
merged leaves of an aquatic plant (Rhizophora man-
gle) in the mangrove forests of Guadeloupe [7]. In
fact, the present study revealed that it was a sulfur-
oxidizing bacterium of the γ-proteobacteria family,
as T. namibiensis.
The authors give a very detailed morphological
and genomic description of this singular bacterium.
By various imaging techniques, it appears that each
filament consists of a single cell along most of its
length, without any dividing septum, with a par-
tial constriction of the apical pole end where buds
(daughter cells) can separate from the filament. The
bacterium has a large central vacuole along the fil-
ament that corresponds to 75% of the total volume
of the bacterium. This vacuole contains high con-
centrations of nitrate, allowing prolonged survival
in the absence of nitrate. The cytoplasm is thus re-
stricted to a thin outer layer of 0.5 to 2 µm at the cell
periphery. This reduction of the cytoplasm would
minimize the dependence on chemical diffusion of
nutrients since, unlike eukaryotes; bacteria do not
have an active intracellular transport system. In the
cytoplasm, numerous white vesicles of about 2 µm
in diameter form refractive sulfur granules. In addi-
tion, the cytoplasm contains bubble-like membrane
organelles containing nuclear material with DNA fib-
rils like the cell nuclei of eukaryotes. These vacuole
membranes contain ATP synthases producing ATP,
as the mitochondria of eukaryotes. Virtually no bac-
teria or archaea are known to segregate their genetic
material as the nucleus of eukaryotes with its nuclear
membrane. The rare exceptions with membrane-
enclosed nucleoids are reported in one species of
Atribacteria and some species of Plancotomycetes
that have a membrane compartment containing
DNA [8, 9].
T. magnifica is a polyploid bacterium, like all giant
bacteria previously described [10]. It contains an av-
erage of 37,000 copies of the genome per mm of fila-
ments. Its genome has been sequenced from five in-
dividual cells showing a nucleotide identity greater
than 99.5%. The genome size is estimated at 11–
12 Mb, compared to the average bacterial genome
size of 4.2 Mb. The genome of T. magnifica con-
tains more than 11,788 genes (compared to 3935
genes on average in bacteria). The genome of this
chemoautotrophic bacterium includes a large num-
ber of genes dedicated to sulfur oxidation and car-
bon fixation. Polyploidy is a means of decreasing se-
lection pressure on genes, allowing intracellular gene
duplications, reassortments of divergences leading to
extreme intracellular genetic diversity in some giant
sulfur-oxidizing bacteria [10].
2. Giant bacteria
Why means this gigantism? Giant bacteria belong to a
few highly specialized bacterial groups. They live un-
der extreme conditions of pH, temperature or pres-
sure, and often thrive in carbon and nutrient rich
environments. For most bacteria, it is advantageous
to be very small, because their metabolism depends
on the diffusion of molecules across the cytoplas-
mic membrane, a process that is very fast and ef-
ficient only over short distances of a few microme-
ters or less. Longer distances greatly slow down the
diffusion of molecules. Eukaryotic protozoa, which
are 10 to 20 µm long and can sometimes reach 3 to
4 cm [11], have overcome these constraints by devel-
oping protein transporters and a system of organelles
to organize subcellular functions. This could explain
the strategy of giant bacteria, which tends to increase
their surface area relative to their volume, by re-
stricting their cytoplasm. In the case of T. magnifica,
we also observe compartmentalization, especially of
DNA, as in eukaryotes. Many questions remain unan-
swered, notably about the metabolism and functions
of the organelles of these giant bacteria. As the au-
thors suggest, this could be solved by culturing this
bacterium with a growth time of two weeks, similar
to that of other species of Thiomargarita sp. [4].
Research on the biology of giant bacteria is an-
other unexpected step towards understanding the
evolution of biological complexity of living cells,
leading to the emergence of eukaryotic cells. Do the
innovations of life come from the adversity of hostile
environments?

ne mesuraient que quelques µm de longueur. Cela
reste vrai dans la très grande majorité des cas. Mais,
on a observé dans les dernières décennies des bac-
téries géantes, souvent confondues au départ avec
des organismes eucaryotes. Ces bactéries peuvent at-
teindre une taille de quelques dizaines à quelques
centaines deµm. Elles ont été décrites dans au moins
quatre phyla [1, 2]. Certaines géantes sont même vi-
sibles à l’œil nu. En 1993, Esther Angert et al. de
l’Indiana University a mis au jour une bactérie géante
(Epulopiscium fishelsoni) dans le tractus intestinal
d’un poisson-chirurgien de la mer Rouge (Acanthu-
rus nigrofuscus). D’abord identifié comme un pro-
tiste eucaryote, il s’agissait en réalité d’une bactérie
sulfureuse à Gram positif mesurant 200–700 µm de
longueur [3]. En 1997, Heide Schultz et al. du Max
Planck Institute for Marine Microbiology a découvert
une bactérie géante à Walvis Bay en Namibie dans
les sédiments marins très inhabituels, sans oxygène,
très riche de sulfures et de matières organiques for-
mées de diatomées mortes. Cette bactérie mesure
100 à 300 µm de diamètre en moyenne, mais peut
atteindre 750 µm. À l’œil nu, ces bactéries coccoïdes
ont l’aspect de perles blanches. Il s’agit d’une bacté-
rie sulfureuse nommée Thiomargarita nabimiensis,
capable d’oxyder l’hydrogène sulfuré (H2S) en soufre
élémentaire et d’utiliser le nitrate comme accepteur
d’électrons. On peut visualiser des vacuoles conte-
nant le soufre et les nitrates et permettant aux bacté-
ries de survivre à de longues périodes de restrictions
en nitrate et en hydrogène sulfuré [4]. Cette bacté-
rie comme d’autres de ce groupe, possède aussi des
introns auto-épissés, habituellement retrouvés uni-
quement chez les eucaryotes [5].
1. Découverte de Thiomargarita magnifica
Jean-Marie Volland et al. viennent de publier dans
Science la découverte d’une nouvelle bactérie géante
qu’ils appellent Thiomargarita magnifica [6]. Il s’agit
d’un travail collaboratif entre des équipes françaises
de l’université des Antilles, du Muséum d’Histoire
naturelle, de Sorbonne Université, et des équipes
américaines de l’université de Californie, Berkeley.
Cet organisme singulier avait été trouvé en 2009
par le Dr. Olivier Gros, un des co-auteurs, biolo-
giste de l’université des Antilles. Dans les forêts de
mangrove de la Guadeloupe, il avait observé des
filaments blancs de 0,9 cm à 2 cm, attachés aux
feuilles immergées d’une plante aquatique (Rhizo-
phora mangle) [7]. En réalité il s’agissait d’une bac-
térie sulfureuse de la famille des γ-proteobacteria,
comme T. namibiensis.
Les auteurs font une description morphologique
et génomique très détaillée de cette singulière bac-
térie. Par diverses techniques d’imagerie, il apparaît
que chaque filament est constitué d’une seule cellule
sur presque toute sa longueur, sans septum de divi-
sion, avec une constriction partielle de l’extrémité du
pôle apical où des bourgeons (cellules-filles) se sé-
parent du filament. La bactérie présente une vaste
vacuole centrale tout au long du filament qui corres-
pond à 75% du volume total de la bactérie. Cette va-
cuole contient de fortes concentrations de nitrates,
ce qui permet une survie prolongée en l’absence de
nitrates. Le cytoplasme est ainsi restreint à une mince
couche externe de 0,5 à 2 µm en périphérie de la cel-
lule. Cette réduction du cytoplasme minimiserait la
dépendance à la diffusion chimique des nutriments
puisque, à la différence des eucaryotes, les bacté-
ries n’ont pas de système de transport intracellulaire
actif. Dans le cytoplasme, on distingue au micro-
scope électronique de nombreuses vésicules claires
d’environ 2 µm de diamètre qui forment des gra-
nules réfringents de soufre. De plus, le cytoplasme
contient des organelles membranaires en forme de
bulles contenant du matériel nucléaire avec des fi-
brilles d’ADN comme les noyaux cellulaires des eu-
caryotes. Ces membranes contiennent des ATP syn-
thases qui produisent de l’ATP, à l’instar des mito-
chondries des eucaryotes. Pratiquement aucune bac-
térie ni archée n’est connue pour ségréger son maté-
riel génétique, à la façon du noyau des eucaryotes qui
possède une membrane nucléaire. Les rares excep-
tions sont des nucléoïdes entourés de membranes
dans une espèce de Atribacteria et certaines espèces
de Plancotomycetes qui présentent un compartiment
membranaire contenant de l’ADN [8, 9].
Comme toutes les bactéries géantes déjà dé-
crites [10], T. magnifica est une bactérie polyploïde.
Elle contient en moyenne 37 000 copies du génome
par mm de filaments. Son génome a été séquencé

à partir de cinq cellules individuelles montrant une
identité nucléotidique supérieure à 99,5%. La taille
du génome est estimée à 11–12 Mb, comparati-
vement à la moyenne des génomes bactériens de
4,2 Mb. Le génome de T. magnifica contient plus de
11 788 gènes (contre 3935 gènes en moyenne chez les
bactéries). Le génome de cette bactérie chimioauto-
trophe comprend un grand nombre de gènes dévolus
à l’oxydation du soufre et à la fixation du carbone. La
polyploïdie est un moyen de décroître la pression de
sélection sur les gènes, permettant des duplications
de gènes intracellulaires, des réassortiments, des
divergences menant à une diversité génétique intra-
cellulaire extrême dans certaines bactéries géantes
oxydant le soufre [10].
2. Les bactéries géantes
Que signifie ce gigantisme ? Les bactéries géantes
appartiennent à quelques groupes bactériens hau-
tement spécialisés. Elles vivent dans des conditions
extrêmes de pH, de température ou de pression, et
prospèrent souvent dans des environnements riches
de carbone et de nutriments. Pour la plupart des bac-
téries, il est avantageux d’être très petites, car leur
métabolisme dépend de la diffusion des molécules
à travers la membrane cytoplasmique, un processus
très rapide et efficace seulement sur de courtes dis-
tances de quelques micromètres ou moins. De plus
grandes distances ralentissent fortement la diffusion
des molécules. Les protozoaires eucaryotes, qui me-
surent 10 à 20 µm et peuvent parfois atteindre 3 à
4 cm [11], se sont affranchis de ces contraintes en
élaborant des transporteurs protéiques et un sys-
tème d’organelles pour organiser les fonctions sub-
cellulaires. Cela pourrait expliquer la stratégie des
bactéries géantes qui tend à augmenter leur surface
par rapport à leur volume, tout en en restreignant
leur cytoplasme. Dans le cas de T. magnifica, on ob-
serve en plus une compartimentalisation de l’ADN,
comme chez les eucaryotes. De nombreuses ques-
tions restent en suspens, notamment sur le métabo-
lisme et les fonctions des organelles de ces bacté-
ries géantes. Comme le suggèrent les auteurs, elles
pourraient être résolues par la mise en culture de
cette bactérie dont le temps de croissance serait de
deux semaines, similaire à celui d’autres espèces de
Thiomargarita [4].
Les recherches sur la biologie des bactéries
géantes permettent un pas de plus vers la com-
préhension de l’évolution de la complexité biolo-
gique des cellules vivantes, aboutissant à l’émer-
gence des cellules eucaryotes. Les innovations de la
vie viennent-elles de l’adversité des milieux hostiles ?
