Abstract. I joined the laboratory of François Gros as a young student in the mid-1980s and worked on
the characterization of the β-tropomyosin gene in chicken and the regulation of alternative splicing of
its transcript, under the supervision of Marc Fiszman. In particular, I was interested in how secondary
structures of the RNA influence the recognition of exons specifically used in muscle cells. I will recall a
few memories on how interacting with François on this project shaped my perception of the scientific
process and of the relationships between models and data. Later I worked on many aspects of RNA
biology, from transcription to mRNP biogenesis and non-coding RNAs.

I joined François’ laboratory in November 1984,
coming from the University of Pisa, as part of the
exchange program between the Scuola Normale of
Pisa and the Ecole Normale Supérieure (ENS). This
was after completing my third year of University,
which today would be equivalent to a Master 1 de-
gree. At the time, François was leading the Unit of
Biochemistry at the Institut Pasteur and was also di-
recting a laboratory at the Collège de France. The
Unit of Biochemistry housed several research groups,
primarily focused on the biochemistry of muscle,
and later, muscle development: the groups of Marc
Fiszman, Robert (Bob) Whalen and Margaret Buck-
ingham, who a few years later became indepen-
dent and founded her own unit in the same de-
partment. Initially, the three groups were interested
in the biochemistry and expression of contractile
proteins, with Bob and Margaret focusing more on
myosins and their mRNAs, respectively, and Marc on
tropomyosin. I had been accepted to work in Marc’s
group.
Marc had set up a nice cellular system whereby
differentiation of chicken myoblasts infected by a
thermosensitive mutant of the Rous sarcoma virus
could be induced by shifting the culture tempera-
ture to 42 °C [1]. In Marc’s group the expression
of tropomyosin isoforms induced specifically during
differentiation was studied biochemically.
But the molecular biology era had started with the
isolation of cDNAs that could be used for charac-
terizing the many different mRNAs coding for con-
tractile proteins, for analyzing gene expression and—
importantly—for the characterization of genes. Arti-
cles had been published by the group of Margaret on
the structure of the myosin light chain genes [2, 3].
In Marc’s lab my task was to isolate the gene cod-
ing for β-tropomyosin in chicken: we had a cDNA
clone provided by David Helfman and in Marc’s
group the expression of the isoforms of chicken
tropomyosin had been extensively studied during
muscle development at the protein level. When we
determined the structure of the gene we discovered

that it was alternatively spliced, with two couples of
exons used in a mutually exclusive manner, exon 6A
and 9B in myoblast, 6B and 9A in myotubes. The big
question was then how the “right” exons are chosen.
At that time, the understanding of splicing regula-
tion was in its early stages. The discovery that genes
were composed of exons and introns had occurred
less than a decade earlier [4, 5] and many examples
of alternatively spliced genes were emerging, starting
in the early and mid 80s [6].
While there was much speculation and research
on how the choice of alternatively spliced exons was
made, the underlying mechanisms remained largely
unknown. This was the challenge we faced. One
of my best souvenirs in François’ laboratory revolves
around these studies, and reflects the great oversight
he provided, even from a somewhat distant eye!
I had analyzed the sequence of the alternatively
spliced region around exons 6A and 6B using the
available tools of the time, such as early versions of
the RNA folding algorithm created by Zuker [7], now
called Mfold [8]. I had proposed a model suggesting
that exon 6B could not be used in myoblasts because
it was sequestered within a secondary structure of the
primary transcript [9]. Upon differentiation, an al-
ternative secondary structure would form around the
other exon (6A), simultaneously freeing the muscle-
specific exon and sequestering the exon used in un-
differentiated cells (Figure 1).
I was very proud of my model, the demonstration
of which, in my naiveness, I had taken for granted.
Around that time the group of Bernard Dujon had
joined the Institute. I remember seeking the opin-
ion of Alain Jacquier, an RNA expert in Bernard’s
team and later on an inspirational friend. Alain was
very skeptical about the possibility that such a struc-
ture would form in the cell, which of course left me
in deep despair. I remember walking back to the
lab, clutching my now seemingly useless model, and
meeting François, who read the disappointment on
my face. François was a very busy person, I was
not even sure he knew who I was. But, of course,
this encounter was a bit like the branch you grab
when falling off a cliff, and I started to explain con-
fusedly the reason of my despair. I certainly did
not know that François had fallen into the “magic
RNA potion” in his youth: “When Monod asked me
what I would like to work on, now that I was in his
lab . . . my decision was taken. I would, from then
on become an RNA biochemist, an RNA man! Noth-
ing concerning RNA and its role in protein synthesis
should be foreign to me.” [10]. François invited me
into his office and patiently listened to my story.
When I was done, he thought for a couple of minutes,

and, of course, agreed with Alain (. . . falling down
the cliff again). He said: “Domenico, ne tombez pas
amoureux d’un modèle, tôt ou tard il vous décevra
(don’t fall in love with a model, sooner or later it will
disappoint you)”—I like to think he was wittily play-
ing with the double meaning of the word “model”.
But then he added: “. . . mais soyez persévérant pour
le démontrer (but be perseverant in proving it”.
This was of course the net that saved my fall.
Many mutations and compensatory mutations later,
the model was partially proven [11], and secondary
structures of the primary transcript are now believed
to have important roles in alternative splicing [12–
15].
I like to think of this anecdote as nicely summariz-
ing François’ legacy. We should be dreamers, some-
times visionaries, persistent in pursuing demonstra-
tive science, but never in love with models. I am cer-
tain that I interpret the thoughts of many colleagues
and friends of that period in thanking François for
building the great and supportive environment that
made possible a very nice moment of science in the
Unit of Biochemistry of the Institut Pasteur.

François Gros est décédé en 2022. Il était un des pionniers de la biologie moléculaire naissante et, pendant la
majeure partie de sa carrière scientifique, il travailla à l’Institut Pasteur en assurant sa direction, de 1976 à 1981.
Il a participé avec Jacques Monod et François Jacob, au développement du concept de l’ARN messager. Durant
un séjour au sein du laboratoire de Jim Watson, à Harvard, il révèlera en effet l’existence d’un ARN instable, en
utilisant des approches biochimiques, ce qui fit l’objet d’une publication majeure – Gros et al. dans la revue
Nature (1961). Cela a été essentiel à la confirmation de l’hypothèse de l’ARN messager à laquelle il continua à
contribuer à son retour à l’Institut Pasteur.
Ultérieurement, dans son propre laboratoire, il a mené à bien la recherche sur la transcription et la
traduction de l’ARNm dans les bactéries ainsi que celle concernant des systèmes eucaryotes ; ceci, dans un
premier temps, à l’Institut de Biologie Physico-Chimique (IBPC) puis, à partir de 1972, à l’Institut Pasteur.
Il s’est intéressé ensuite aux changements dans l’expression des gènes durant la différentiation cellulaire, en
utilisant des cellules musculaires comme système modèle ainsi que des cellules neuronales, dans son second
laboratoire, au Collège de France. Ceux qui ont eu la chance de travailler avec lui ont largement bénéficié de sa
connaissance scientifique et de son soutien généreux.
Il a aussi apporté une importante contribution à l’organisation de la recherche et de la politique scientifiques
en France. Il fut directeur de l’Institut Pasteur (1976-1981) et conseiller scientifique en chef auprès des Premiers
ministres (1981-1985) durant la présidence de F. Mitterrand. Pendant sa carrière, il a participé à la rédaction de
nombreux rapports importants, à la fois pour le gouvernement et ensuite, en tant que secrétaire perpétuel de
l’Académie des sciences (1991-2000) où il a fortement soutenu la coopération scientifique dans les pays en voie
de développement, particulièrement en Afrique.
En avril 2023, un colloque a été organisé pour lui rendre hommage, avec une matinée à l’Institut Pasteur
consacrée à l’ARN, où sont intervenus notamment Venki Ramakrishnan et Emmanuelle Charpentier, lauréats
du prix Nobel. Dans l’après-midi, à l’Institut de France, la suite de la conférence l’honora pour ses contributions
à l’Académie.
Ce volume des Comptes Rendus Biologies rend également hommage à François Gros. Il contient un texte
biographique écrit par C. Petit et P. Kourilsky, professeurs au Collège de France ; l’article suivant est rédigé par
J.-P. Changeux qui le connut depuis le temps où ils travaillaient tous les deux avec Jacques Monod. Les articles
suivants sont écrits par des collaborateurs ayant travaillé avec lui, dans son laboratoire à l’IBPC, à l’Institut
Pasteur et au Collège de France. Figurent également deux articles sur les ARN, provenant d’orateurs ayant pris
part au colloque en son hommage, puis d’autres, écrits par des collègues ayant rappelé ses actions à l’Académie.

I met François Gros in 1976, when the two of us dined together in Moscow on the way to an interminable trip
that took us to Tashkent, Vientiane, and then Hanoi, where we were received by Prime Minister Phan van Dong
and General Giap. It was the beginning of a great friendship that lasted until the end.
At the time, François Gros was a professor at the Collège de France and director of the Institut Pasteur. The
following year, he was elected a correspondent of the Académie des sciences, and a member in 1979. In 1991,
he was elected permanent secretary, succeeding Alfred Jost, who had died prematurely. He was fond of telling
the story of how he accepted this post after a conversation in front of the magnolias in the second courtyard
with Jean Hamburger, whose persuasiveness I can personally attest to.
His term of office lasted 10 years (June 10, 1991 – December 31, 2000), 10 years of happiness but also
of hard work and considerable investment. François Gros made a major contribution to the renovation
and modernization of our Académie, both through the many scientific initiatives he led and through the
development of our activities, which led to the reform of our statutes in 2003, led by Jean Salençon and Roger
Monnier, with the support of Claude Allègre, then French Minister of Education. The reform was not completed
until three years after the end of his term of office, but François Gros played a very important role in shaping
the ideas that led to the success of this major reform. In fact, the end of his term did not prevent him from
continuing his commitment to the Académie as honorary permanent secretary. In fact, at the age of 96, he
continued to visit his office at the Académie every day, as committed as ever to his work and much sought after
for his experience and the depth of his opinions.
In close collaboration with Paul Germain, and later with Jean Dercourt, he promoted the development of
the Applications Committee of the Académie des Sciences, the CADAS, which a few years later gave rise to the
Académie des Technologies. More generally, he got on very well with Jean Dercourt, the permanent secretary
of the First Division, whose personality, temperament and interests he himself recognised as complementary.
He also got on well with the five successive Presidents: Jean Hamburger, Jacques Friedel, Marianne Grunberg-
Manago, Jacques-Louis Lions and Guy Ourisson.
François Gros’ work as permanent secretary was essentially scientific, placing his immense knowledge and
synthesising skills at the Académie’s disposal. Together with Pierre Buser, Jean Rosa, Michel Tellier and François
Mathey, he played an active role in the revival of the Comptes Rendus in the biology and chemistry series.
François Gros was the first to open up the vast field of relations between science and society by setting up a
committee which he kindly entrusted to me to chair. A few years later, this committee became the Science,
Ethics and Society Committee.
François Gros coordinated, and in fact largely wrote, many of the reports on science and technology that
the Académie produced during this period. He was particularly involved in the preparation of an outstanding
report on the genome, in which he was able to express the depth of his culture and thought, Development and
application of genomics [1]. Other reports include those on animal and human physiology, the plant world,
genetically modified plants, and pharmaceuticals.
In 2006, he personally coordinated a major report entitled Science and Developing Countries [2], a subject
that was particularly close to the heart of the Pasteurian he was.
He endeavoured to create close links between the Académie and the public authorities. Thus, President
Jacques Chirac approached the Académie on the vast subject of the expected development of science in the
first decades of the 21st century, particularly in relation to the environment and living conditions, information
sciences and health. He set up the 2000 Committee, chaired by Jacques-Louis Lions. The drafting of this report
generated a great deal of enthusiasm among all those involved, and all the members of the delegation who
presented the report to the President at the Elysée Palace have fond memories of it. Still on the subject of
relations with the public authorities, it is worth mentioning the extremely high number of opinions requested
from the Académie at the time by the Ministries, the Parliamentary Office for Scientific and Technological
Choices, and sometimes also by the Senate, the Economic and Social Council, and so on.
He was involved in the organisation of a number of high-level symposia, in particular on the Enlightenment,
with the German Academy of Sciences, the Leopoldina, and on biology and developing countries. François
Gros also made great efforts to forge closer links with foreign Academies, not only with the major European

Academies, in particular the German Academy, with which the Humboldt Prize was created, but also with
the Academies of developing countries, as part of the creation of the Committee for Developing Countries
(COPED).
His many personal contacts enabled him to establish highly interesting collaborations. He developed a close
working relationship with the Weismann Institute of Science, which led to the creation of FIRAS (Fondation
internationale pour le rayonnement de l’Académie des sciences) with Robert Parianti. This enabled him to raise
substantial funds to support his work at COPED, as well as the renovation of the Pasteur House in Arbois, which
coincided with a major celebration of the centenary of Louis Pasteur’s death. On another front, he worked
closely with Alain Mérieux, with whom he developed a close friendship. He took an active part in the work of
the Board of Directors of the Fondation Christophe et Rodolphe Mérieux at the Institut de France and in the
creation of a prestigious prize to support research into infectious diseases in developing countries in the most
effective way.
I won’t go into detail about his major scientific contributions, but I will highlight the modesty with which he
spoke about them, a modesty that did not prevent him from being very determined. In a scientific environment
that is not always benevolent, I never heard any criticism of him or his activities. On the contrary, I would like
to stress his great humanity, his kindness, his generosity and his exceptional availability, both to his friends and
colleagues, French and foreign, and to the staff of the Académie, who learned to turn to him for the most diverse
problems. As he used to say with humour, his problem was that he did not dare to contradict his interlocutors.
His radiant personality was such that we feel he is still with us and must continue to inspire us.

ouvrage, le professeur Harris réexamine en détail les expériences décisives de Gros et al [1] et d’autres, et
continue à conclure que c’est en fait l’ARN ribosomal qui est le messager !
Lorsque j’ai obtenu mon diplôme de biochimie à Cambridge en 1974, le sujet de l’ARNm était encore
d’actualité et j’ai eu la chance de réaliser un projet de premier cycle sur la traduction de l’ARN dans des
lysats de réticulocytes avec Tim Hunt et Richard Jackson. Cela m’a persuadé de poursuivre dans le domaine de
l’ARNm pour mon doctorat, au Beatson Institute de Glasgow, et ensuite lors de mon postdoctorat avec François
à l’Institut Pasteur.
2. À propos des populations d’ARN dans les cellules animales et du nombre de gènes
Maintenant que l’ARN pouvait être isolé, notre travail au Beatson Institute, et dans le cadre d’études menées
en parallèle par Nabeel Affara dans le laboratoire de François à Pasteur, par le laboratoire de John Bishop à
Édimbourg, par le laboratoire d’Eric Davidson à Los Angeles et par d’autres, consistait à tenter de concevoir
des méthodes expérimentales permettant d’étudier les populations d’ARN dans les cellules animales. Nous
avons utilisé une enzyme nouvellement découverte, la transcriptase inverse, pour faire des copies de tous les
ARNm polyadénylés. Dans des « conteneurs » expérimentaux hautement miniaturisés, d’un volume de l’ordre
du microlitre, nous avons mélangé les ARN et des copies d’ADN marquées par radioactivité, puis nous avons
étudié la formation des duplex ARN-ADN sur des périodes pouvant aller jusqu’à une semaine.
Les résultats de ces expériences nous ont permis d’estimer le nombre de gènes exprimés dans une cellule
animale, et le nombre de ces gènes communs à un autre type de cellule. Compte tenu de toutes les incertitudes
liées aux calculs, les résultats sont étonnamment cohérents d’une étude à l’autre, la plupart des cellules et
tissus animaux exprimant entre 10 000 et 20 000 gènes [3]. Ce chiffre est du même ordre que le nombre de
gènes humains obtenu vingt ans plus tard grâce au séquençage du génome [4].
3. À propos de l’ARNm non polyadénylé
En 1978, incité par mon ami Nabeel Affara, j’ai déménagé à l’Institut Pasteur pour travailler avec François. Je
me souviens encore d’un de mes premiers échanges par courrier avec François, qui était un très bon épistolier,
alors que j’étais encore à Glasgow. Il exprimait l’espoir qu’une « excellente articulation » s’établirait entre nous.
J’aime à penser que ce fut le cas !
À l’Institut Pasteur, j’ai poursuivi l’étude des ARNm. Dans toutes les études sur l’ARNm mentionnées
précédemment, nous avions défini expérimentalement l’ARNm comme étant polyadénylé, puisque nous
l’avions isolé par chromatographie sur des résines d’oligo(dT)-cellulose, ou de poly(U)-Sépharose. Cependant,
une fraction importante de l’ARNm (définie par sa traduction dans un lysat réticulocytaire) ne se liait ni
à l’oligo(dT) ni au poly(U). Nous avons donc essayé de voir si cette fraction pouvait coder des protéines
différentes, et représenter de ce fait un type différent d’ARN messager.
En analysant les produits de traduction de préparations poly(A)+ et poly(A)− hautement purifiées, grâce
à une électrophorèse bidimensionnelle, nous avons montré que les deux populations codaient des protéines
similaires, mais en quantité très différentes [5]. Une classe de protéines qui était presque entièrement obtenue à
partir d’ARNm poly(A)− était celle codant pour les histones. Nos études ont montré que, mis à part les histones,
l’ARNm poly(A)- ne contient pas un autre sous-ensemble particulier d’ARNm. Ce travail et la figure décisive du
gel 2D ont été cités dans le livre Molecular Cell Biology de Darnell, Lodish et Baltimore [6]. Peu de travaux ont
été réalisés depuis sur cette question, mais ceux qui l’ont été confirment notre conclusion [7].

4. À propos de l’avènement du clonage génétique
Les études précédentes sur les populations d’ARNm représentaient les dernières de l’« ère pré-clonage ».
L’avènement de la technologie de l’ADN recombinant allait révolutionner nos approches expérimentales en
biologie moléculaire, et le laboratoire de François ne faisait pas exception (voir son chapitre sur Le « boum » du
génie génétique [8]). Un projet de clonage de gènes a été mis en place dans le groupe de Margaret Buckingham,
et j’ai pris la responsabilité de cloner les gènes de l’actine de la souris. Je me souviens encore avoir traversé la
rue du Dr Roux pour discuter avec François Rougeon, l’un des spécialistes mondiaux du clonage de gènes, qui
m’a prévenu que cela risquait d’être compliqué car les ARNm de l’actine étaient moins abondants que l’ARNm
de la globine et les autres ARNm qui avaient été clonés jusqu’alors ! Par conséquent, nous avons soumis les ARN
à une séparation par gradient de sucrose afin d’enrichir le(s) ARNm d’actine avant leur clonage !
Le clonage de gènes était un défi pour quelqu’un qui n’avait jamais fait de travaux expérimentaux avec des
plasmides bactériens, ni même avec des bactéries ! Néanmoins, l’abondance des ARNm d’actine (en particulier
après les avoir sélectionnés en fonction de leur taille !) a fait qu’une centaine de bactéries recombinantes
ont été largement suffisantes pour isoler plusieurs ADNc d’actine distincts, y compris deux ADNc d’actine
musculaire différentes [9]. Ce travail nous a permis de caractériser, pour la première fois, l’expression des
actines musculaires et cardiaques dans le muscle squelettique [10].
5. À propos de la rétro-transcription de l’ARNm polyadénylé et l’intégration des copies d’ADN dans
le génome
Les ADNc d’actine nouvellement clonés ont permis de répondre tout d’abord à la question du nombre de
gènes d’actine dans les génomes de mammifères. Nous avons constaté que dans l’ADN de la souris, en plus
des six gènes d’actine que les travaux de séquençage des protéines laissaient présager, il existait également
une famille de séquences faiblement apparentées, dont certaines avaient été récemment amplifiées dans
le génome [11]. Bien qu’ils n’aient pas été séquencés, la longueur de ces gènes de type actine dans l’ADN
génomique nous a amenés à prédire qu’ils pourraient représenter des pseudogènes sans intron, générés par
transcription inverse des ARNm de l’actine et insertion dans le génome. Ce processus a été confirmé depuis,
avec le séquençage de plus de 8 000 rétropseudogènes dans le génome humain [12].
Il est quelque peu surprenant que ces études sur la réintégration des copies d’ARNm dans le génome aient
été largement ignorées lors du récent débat sur la possibilité de réintégration du vaccin à ARN contre la Covid-
19 dans le génome. Alex Whiting, écrivant en 2020 [13], a cité le professeur Michel Goldman de l’Université libre
de Bruxelles qui a déclaré que « les vaccins à ARNm ne modifient pas votre ADN. Certains craignent que les
vaccins à ARNm ne modifient l’ADN des personnes. Mais cette idée est “complètement fausse” et n’a “aucune
base scientifique” ».
Il semblerait, en fait, que ce soit l’affirmation de l’impossibilité de réintégration dans le génome qui soit
complètement fausse [14]. Combattre les théories du complot sur les dangers potentiels des vaccins à ARN ne
doit pas conduire à ignorer des faits potentiellement contradictoires !
6. À propos de la nature des séquences régulatrices des gènes
Le « graal » des biologistes moléculaires dans les années 1980 était l’identification des séquences régulant
l’expression des gènes dans les cellules de mammifères. Pour y parvenir, je devais poursuivre les travaux sur
les gènes de l’actine dans le laboratoire de Larry Kedes à l’université de Stanford. Outre les 9 000 kilomètres
qui séparaient Paris de Palo Alto, la principale différence entre les deux laboratoires était que le laboratoire de
Kedes travaillait sur des gènes d’actine humains et non sur des gènes d’actine de souris. Après quelques mois
de travail expérimental assez laborieux pour identifier le site d’initiation de la transcription du gène de l’actine
cardiaque humaine, j’allais avoir de la chance.

C’était l’époque, en 1984, où Apple lançait le premier Macintosh, et une offre spéciale était faite aux
travailleurs de Stanford. C’est donc sur mon propre Macintosh, à l’aide d’un programme de comparaison
d’ADN très rudimentaire, que la séquence régulatrice « CCArichGG » a été identifiée à mon domicile de
Mountain View [15]. Depuis, il a été démontré que ces séquences d’ADN CCArGG sont plus répandues que
nous ne l’avions initialement prévu et qu’elles jouent des rôles plus diversifiés. En effet, trente-sept ans plus
tard, les mécanismes moléculaires de liaison des facteurs de transcription à ces séquences sont toujours en
cours d’élucidation [16]. Je me sens extrêmement chanceux d’avoir pu participer, grâce à l’aide de François, à
cette période si passionnante de la recherche.

Il ressort clairement de ce qui précède que les premières années de ma carrière de chercheur ont été forte-
ment marquées par le temps que j’ai passé dans le laboratoire de François Gros. Je serai toujours reconnaissant
à François de m’avoir permis de venir travailler dans son laboratoire, de m’avoir laissé choisir mes sujets de re-
cherche et de m’avoir si souvent offert son temps précieux et ses conseils. Parmi les autres personnes qui m’ont
beaucoup aidé pendant mon séjour à Pasteur, je voudrais également remercier Nabeel Affara, qui m’a persuadé
de venir à Paris, et Margaret Buckingham, avec qui je devais travailler sur le projet de clonage de gènes. Enfin,
et surtout, c’est dans le laboratoire de François que j’ai rencontré ma femme Catherine et que j’ai acquis la
nationalité d’un pays qui est le mien depuis maintenant quarante-cinq ans!
