Comparative Evaluation of Masson's Trichrome and Picrosirius Red Staining for Digital Collagen Quantification Using ImageJ in Rabbit Wound Healing Research
The therapeutic potential of Pluronic F127 (PF127) hydrogel loaded with adipose-derived stromal vascular fraction (AdSVF), mesenchymal stem cells (AdMSC), and conditioned media (AdMSC-CM) for repairing full-thickness skin wounds was evaluated using a rabbit model. The rabbits were randomly divided into eight groups with six animals each and treatment was given as per the predetermined protocol (3 doses at one-week interval): Group A (Control), Group B (AdSVF), Group C (AdMSC), Group D (AdMSC-CM), Group E (PF127), Group F (AdSVF + PF127), Group G (AdMSC + PF127), and Group H (AdMSC-CM + PF127). Skin tissue samples were collected from the healing wounds on day 28 for staining and collagen quantification. Collagen density (Area %) was quantified using tissue sections stained with Masson's Trichrome (MT) and Picrosirius Red (PSR) stain using the Colour Deconvolution plugin of ImageJ and RGB stack method, respectively. These techniques function based on separating different colour channels in the stained tissue sections to isolate the collagen fibers and then quantifying them through thresholding and image analysis. Across the treatment groups, both staining methods generally showed a trend of increased collagen density compared to the control group. For most groups, PSR staining consistently indicated slightly lower collagen densities than MT staining. However, the overall trends were similar in both staining. The comparison between PSR and MT staining methods revealed that both techniques effectively assess collagen density in healing wounds. However, there were subtle differences in the absolute values obtained, with PSR staining tending to yield slightly lower collagen density measurements than MT. These differences can be attributed to the distinct mechanisms of these staining methods. Therefore, both staining methods can digitally quantify collagen density in wound healing research
Ovine pulmonary adenocarcinoma (OPA) in sheep: an update on epidemiology, pathogenesis and diagnosis
Ovine pulmonary adenocarcinoma (OPA) is a spontaneous lung tumor in sheep caused by Jaagsiekte sheep retrovirus (JSRV) belonging to the Retroviridae. The primary aim of this review work is to give brief insights into the epidemiological aspects of OPA based on a meta-analysis of available research work. This review article also discussed pathogenesis, diagnostic tests and control strategies available for OPA in Sheep. This will help in developing future strategies for disease-free status in India. This disease is endemic in Europe, Africa, Asia, and American continents, causing significant economic losses due to chronic respiratory illness and persistent infections in flocks. The virus is unique among retroviruses with selective affinity to lungs and is the only virus known to cause spontaneous lung tumors in sheep. The incubation time ranges for sheep with naturally occurring OPA ranged from one to four years. There are two pathological forms of the disease: classical and atypical. At an early stage, OPA is difficult to detect in sheep due to a lack of preclinical diagnostic methods, as JSRV is poorly immunogenic and doesn't induce an immune response. PCR, histopathology, and immunohistochemistry are recommended methods for OIE diagnosis. To become a JSRV-free country, mandatory surveillance, detection, and removal of positive animals are required, as OPA is difficult to control due to a lack of vaccines and preclinical diagnostic tests. Due to its similar histological and molecular pathogenesis to that of human lung cancer, OPA is considered an ideal large animal model of human lung adenocarcinoma
Effect of probiotics on the histomorphometry characteristics of Mus musculus Jejunum infected by Salmonella gallinarum
Salmonellosis is a disease caused by Salmonella gallinarum, which can cause digestive tract infections. Probiotics are good microorganisms for the host because they can increase the normal bacteria flora in the digestive tract. They can maintain the intestinal mucosal barrier and prevent bacterial adhesion. This study aimed to determine the histomorphometric characteristics of the jejunum from the intestines of mice (Mus musculus) after being infected with S. gallinarum. A total of 20 mice, 4-6 weeks, were divided into four research groups: P1 (probiotics and S. gallinarum infection), P2 (probiotic administration), P3 (S. gallinarum infection), and P4 (control). The probiotics used contain microorganisms such as Lactobacillus casei, Saccharomyces cerevisiae, and Rhodopseudomonas palustris, dissolved in distilled water in a ratio of 1:1000. Probiotics were given orally at 0.5 ml for 7 days. S. gallinarum infection was given orally, with a volume of 0.5 ml (1.5 x 108 CFU/ml). The results showed that the mean score of intestinal lesions differed between groups. The width of the villi, the thickness of the mucosa, and the depth of the intestinal crypts were significantly different. The best result of histology findings was in the group of mice that were induced with probiotics (P2)
Optimization of Total Flavonoid Extraction From the Helicteres hirsuta Lour. Roots by Bath Ultrasound Assisted method and cytotoxic activities of these Flavonoids
This study was carried out to optimize the various approaches to analyze the effects of various variables on the total flavonoid content extraction from the roots of Helicteres hirsuta L. The existence of various compounds in the methanol fraction was accessed by using LC-MS/MS analysis. The results of the study identified the ideal parameters such as times (30 minutes); methanol solvent concentration (50%); ultrasonic frequency (12 Hz); and material/solvent ratio [1:30 (w/v)] for extracting the highest total flavonoids from the roots of H. Hirsuta. The study's results suggested that the total flavonoid value was 3.52684 (mg Catechin/g extract). The verified experiment obtained an actual value of 5.205 (mg Catechin/g extract). Further, the results of the study suggested the presence of 20 compounds of a flavonoid nature (66.667%) appearing in the purified methanol fractional extract. These compounds can inhibit DPPH free radicals at 50%, with an IC50 value of 536.760 g/mL, and they also have inhibitory activity on the growth of cancer cell lines with IC50 values ranging from 115.81 and 219.17g/mL. The human leukemia cell line (HL-60) exhibits the most significant cytotoxic response to a methanol extract from H. hirsuta root with an IC50 value of 115.81 g/mL
Comparative analysis of antioxidant activities of Vitex negundo and Ficus carica leaf extracts
Humans have been dependent on nature for various vital supplies and resources for a long time. Most biotechnological and pharmacological industries use chemicals and active compounds to treat diseases or make medications isolated from natural resources. A variety of plants have been explored for research of which Vitex negundo and Ficus carica are also examples as they are strong candidates for their potential antioxidant properties. In the current research, the anti-oxidant activities of V. negundo and F. carica leaf extracts were evaluated. The antioxidant activities of selected plants were analyzed using DPPH and FRAP assay. The results obtained from the DPPH assay indicated that methanolic extracts of V. negundo showed the highest inhibition of 90.07±1.17 percent at 1000 µl with IC50 value of 415.98 µg/ml followed by ethyl acetate and chloroform extracts (64.05±0.89 and 54.39±0.99 percent, respectively) with IC50 value of 751.96 µg/ml and 896.55 µg/ml when compared to F. carica extracts which showed highest inhibition of 75.75±1.08 percent at 1000 µl with IC50 value of 475 µg/ml followed by ethyl acetate and chloroform extracts (51.94±0.79 and 44.21±0.60 percent respectively) with IC50 value of 967.51 µg/ml and 1092.48 µg/ml. On comparing both plants, FRAP results indicated that methanol extracts of V. negundo showed the highest FRAP value (1042.1±0.98 µM) followed by ethyl acetate and chloroform extracts, which shows 996.6±1.25 µM and 949.6±1.63 µM at 1000 µl whereas F. carica showed highest FRAP value (995.6±1.35µM) followed by ethyl acetate and chloroform extracts, which shows 987.6±1.05µM and 447.6±1.01µM at 1000 µl. The results of the study can be concluded that among the tested extracts, the best antioxidant potential was exhibited with V. negundo leaf extracts, especially in methanol extracts
Beneficial impacts of goat milk on the nutritional status and general well-being of human beings: Anecdotal evidence
Goats provide an essential food supply in the form of milk and meat. Goat milk has distinct qualities, but it shares many similarities with human and bovine milk regarding its nutritional and therapeutic benefits. Because of their different compositions, goat and cow milk products could have different tastes, nutrients, and medicinal effects. Modification in composition aid of goat milk determining the viability of goat milk processing methods. Comparatively, goat's milk has higher calcium, magnesium, and phosphorus levels than cow's or human milk but lower vitamin D, B12, and folate levels. Goat milk is safe and healthy for infants, the old, and healing ailments. Capric, caprylic, and capric acid are three fatty acids that have shown promise as potential treatments for various medical issues. Considering the benefits and drawbacks of goat milk over cow milk is essential; goat milk is more digestible, has unique alkalinity, has a better buffering capacity, and has certain medicinal benefits. Acidifying goat milk shrinks fat globules and makes protein friable (with less αs1-casein and more αs2-casein). Goat milk treats malabsorption illnesses because it has more short- and medium-chain triglycerides that give developing children energy. In wealthy countries, goat milk and its products—yoghurt, cheeses, and powdered goods—are popular with connoisseurs and persons with allergies and gastrointestinal issues who need alternative dairy products. A food product category containing fermented goat milk with live probiotic microbes appears promising nutritionally and medicinally. This article presents anecdotal evidence of the therapeutic effects of consuming goat milk for human health and its nutritional value
Effect of human β-Globin second intron on transient gene expression in mammalian cell lines
Exogenous protein expression in mammalian cells is necessary to produce therapeutic proteins and modern medical applications like developing DNA vaccines and gene therapy. This study examines the human-Globin (hBG) second intron's capacity for intron-mediated enhancement (IME) in various mammalian cell lines. Our study's main aim is to investigate the effect of the incorporation and arrangement of the second intron of the human Beta-globin gene into the pVAX-1 expression cassette on improving the expression of foreign genes. Two plasmids were constructed, one with the hBG second intron positioned upstream and the other downstream in the expression cassette. EGFP expression was evaluated at the mRNA and protein levels after transfection using Lipofectamine 2000 using One-way ANOVA analysis. Results showed that the pVAX-1 harbouring the hBG second intron did not lead to enhanced transient EGFP expression and did not exhibit Intron Mediated Enhancement (IME) in tested mammalian cell lines. Further investigations are necessary to understand factors contributing to the lack of enhancement and explore alternative intron options for optimizing foreign gene expression in cell lines
Acid Rain and Seed Germination: A Predictive Model Using ML-based CART Algorithm
The impact of acid rain on the germination of seeds is a significant concern in agricultural and environmental studies. Acid rain, characterized by elevated acidity levels due to pollutants like sulfur dioxide and nitrogen oxides, can adversely affect the germination process of various plant species. The objective of this study was to evaluate the impact of simulated acid rain (SAR) on the germination of Brinjal (Solanum melongena Linn.) and Cowpea (Vigna unguiculata ssp. cylindrica L. Walpers) crops. The experiments were conducted using eight plastic trays of approximately 25 cm. x 30 cm dimensions. Four trays were used for experiments with brinjal seeds (Set I), while the other four were used for cowpea seeds (Set II). One tray from each set was used as positive control and treated with normal pH 5.6, while the other three trays from each batch were treated with SAR solutions of pH 4.5, 3.5, and 2.5. Brinjal seed germination percentage and seed vigor were inferior to Cowpea seeds. The seeds treated with SAR (pH 4.5, 3.5, and 2.5) showed hindered seed germination. Furthermore, a more significant inhibitory effect was observed at lower pH values. The mean germination percentage of seeds was highest for standard SAR (pH 5.6) in the case of Brinjal seeds, while it was recorded lowest for Cowpea seeds. The results indicate that plants do not respond uniformly to SAR. To investigate the behavior of the simulated acid rain data, a Machine Learning-based Decision Tree Algorithm was employed to identify and optimize conditions. Cowpea was predicted to get 95% seed germination, whereas brinjal would only be 64% in acid rain of pH value 5.05 for 36 hours. In conclusion, utilizing a Machine Learning-based CART algorithm has provided valuable insights into predicting the germination behavior of seeds under the influence of acid rain
Effect of Transpiration on the Monocot Ornamental Plants Leave Anatomy
Transpiration refers to the loss of water from leaves, and increased levels can lead to changes in leaf morphology and anatomy, affecting the total thickness. This study aims to determine the effect of transpiration on leaf anatomy, particularly thickness, in six types of monocots ornamental plants, namely Rhoeo discolor (L'Her.) Hance ex Walp., Hymenocallis littoralis (Jacq.) Salisb., Cordyline fruticosa (L.) A. Chev., Chlorophytum laxum R. Br, Dracaena reflexa Lam, and Aglaonema commutatum Schott. The study procedures were conducted using a Factorial Completely Randomized Design (Factorial CRD) with an experimental approach. The first factor was the type of plant, while the second was the condition before and after transpiration. The data obtained were analyzed using ANOVA, followed by LSD and Pearson correlation tests. The results showed that the plant type factor significantly affected the thickness of leaf tissues. The conditions before and after transpiration also significantly impacted all leaf tissues except for the lower epidermis. Furthermore, this finding was supported by the positive correlation between the thickness shrinkage of the upper epidermis-mesophyll and transpiration. The results also revealed that the mesophyll of R. discolor, C. laxum, D. reflexa, and A. commutatum differentiated into palisade and spongy layers, but there was no differentiation in the other two species. The transpiration rate was observed to change along with the specific anatomical structure of the leaf tissues. The lowest rate was found in R. discolor with thicker hypodermis tissue, while the highest was in C. laxum with thinner mesophyll
Contraceptive efficacy and antioxidant potential of Leptadenia reticulata bark extracts in male albino rats
Birth control measures available are primarily for women which are hormonal supplements that are increasing cancer risks and reproductive health issues. Male contraceptive options are effective and available, i.e. barrier methods and vasectomy. Condoms are failure-prone and single-use, while a vasectomy is a permanent sterilization method done surgically, and reversion is not always successful and expensive. A promising oral male contraceptive drug candidate is yet to be discovered. This study investigated the contraceptive efficacy and antioxidant potential of various extracts of Leptadenia reticulata bark in male rats. To study the effects of various extracts (ethanolic and petroleum ether) of L. reticulata bark in male rats, oral administration at the dose level of 250 mg/kg body weight/ day was done for 60 days. Observations were made for body and organ weight, hematology, serum biochemical chemistry, testosterone and antioxidants, lipid profile, sperm parameters (density and motility) and histological changes (reproductive organs). As compared to control in treated groups (TP and bark petroleum ether extract), a significant reduction (P ≤ 0.001) was perceived in sperm motility and density, as well as reproductive organ weight, serum testosterone, and serum antioxidant parameters like SOD. Histological observations revealed arrest in spermatogenesis and reduced seminiferous tubule diameter, mature Leydig cells, secondary spermatogonia, and spermatids which caused a substantial increase in LPO and GSH. From the research findings, it can be concluded that bark petroleum ether extract of L. reticulata possesses contraceptive potential in male albino rats and can serve as a safe and reversible oral contraceptive for males
