児童の自己調整能力が2Dメタバース上の仲間とのつながりへの意識に及ぼす影響
本研究の目的は,小学校で2Dメタバースを活用した自律的かつ対話的な交流活動において児童の自己調整能力が仲間とのつながり意識に及ぼす影響を示すことである.長崎と福島の児童計27名を対象に,2Dメタバース「oVice」を用いて被爆遺構や災害情報の発表と全体討論を行った.評価方法として,仲間とのつながり意識および自己調整方略について質問紙調査を行い,自己調整方略の上位群と下位群でつながり意識を比較した.マン・ホイットニーのU検定の結果,仲間とのつながりへの実感において上位群が下位群よりも平均値が有意に高かった.これにより,2Dメタバース上で仲間とのつながりを実感するためには,自己調整能力が一定の役割を果たすことが示唆された.また,上位群の自由記述に交流活動自体に関する肯定的な意見が見られ,自己調整能力が交流体験の質に影響する可能性が考えられたが,コミュニケーション自体の困難さが生じる可能性も示唆された.長崎大学教育学部紀要:人文科学 通巻 第91号 (Bulletin of Faculty of Education Nagasaki University: Humanities, Vol.91)長崎大学教育学部紀要, 11, pp.125-130; 2025departmental bulletin pape
証拠標本に基づく日本国内2例目のカドガワフエダイ Lutjanus johnii (Bloch, 1792) の形態学的特徴および生態に関する知見
Since Lutjanus johnii (Bloch, 1792) was first recorded from Kuroshima Island, Miyazaki Prefecture in 2011 and assigned the standard Japanese name “Kadogawa-fuedai”, no additional specimens have been reported from Japan. In 2024, we obtained a second Japanese specimen from off Kochidomari Port, Kagoshima Prefecture, marking the first domestic record in 13 years. This paper provides detailed measurements and a description of its external morphology, compares the specimen with previous records, and highlights issues related to distinguishing Lutjanus spp. found in Japan. Furthermore, the specimen was estimated to be a four-year-old male and exhibited a larger fork length than the average for four-year-old males in a previous study.カドガワフエダイは,2011年に宮崎県黒島で初めて記録され,標準和名が提唱されて以降,国内での新たな標本に基づく出現報告はない。著者らは,2024年に鹿児島県東風泊港沖から13年ぶりとなる国内2例目の標本1個体を入手した。本標本の外部形態の詳細な計測と記載および先行研究との比較を行い,日本産フエダイ属類似種との識別に関する問題点を明示した。また,本標本は雄の4歳と推定され,先行研究における雄の4歳の平均的な尾叉長よりも大きかった。長崎大学水産学部研究報告, 105, pp.10-19; 2025departmental bulletin pape
Polymerized Alizarin Red–Inorganic Hybrid Nanoarchitecture (PARIHN) as a Novel Fluorogenic Label for the Immunosorbent Assay of COVID-19
This study seeks to develop and implement a non-enzymatic fluorescent labeling for immunoassay and immunochromatographic assay (ICAs) targeting SARS-CoV-2, to meet the extensive interest and need for effective COVID-19 diagnosis. In this manuscript, we delineate the development, synthesis, and evaluation of a novel quinone polymer zinc hybrid nanoarchitecture, referred to as polymerized alizarin red–inorganic hybrid nanoarchitecture (PARIHN), which integrates an antibody for direct use in fluorescent immunoassays, offering enhanced sensitivity, reduced costs, and improved environmental sustainability. The designed nanoarchitecture can enhance the sensitivity of the immunoassay and enable rapid results without the complexities associated with enzymes, such as their low stability and high cost. At first, a chitosan–alizarin polymer was synthesized utilizing quinone–chitosan conjugation chemistry (QCCC). Then, the chitosan–alizarin polymer was embedded with the detection antibody using zinc ion, forming PARIHN, which was proven to be a stable label with the ability to enhance the assay stability and sensitivity of the immunoassay. PARIHN can react with phenylboronic acid (PBA) or boric acid through its alizarin content to produce fluorescence signals with an LOD of 15.9 and 2.6 pm for PBA and boric acid, respectively, which is the first use of a boric acid derivative in signal generation in the immunoassay. Furthermore, PARIHN demonstrated high practicality in detecting SARS-CoV-2 nucleoprotein in fluorescence (PBA and boric acid) systems with an LOD of 0.76 and 10.85 pm, respectively. Furthermore, owing to the high brightness of our PARIHN fluorogenic reaction, our labeling approach was extended to immunochromatographic assays for SARS-CoV-2 with high sensitivity down to 9.45 pg/mL.Biosensors, 15(4), art. no. 256; 2025journal articl

Improved transcriptome assembly and functional annotation of Pleurodeles waltl for regeneration research
In this study, we present an updated transcriptome assembly for the Iberian ribbed newt, Pleurodeles waltl (P. waltl), a widely used model organism in regeneration research. The existing publicly available transcriptome for this species is limited by the inclusion of only three libraries from the limb and two from the heart, tissues of particular interest for regeneration studies. Additionally, the previous annotation was limited, reducing the utility of the dataset for further in-depth research. To provide a more complete transcriptome with a more comprehensive annotation, we utilized 58 previously published and 9 newly sequenced libraries, expanding the available transcriptomic data for key tissues, especially limb and heart tissues. Our assessment demonstrates that the new assembly offers a more comprehensive representation of reads and proteins compared to previous versions. Furthermore, we significantly improved the functional annotation by using the Trinotate pipeline, which includes the identification of complete ORFs, Pfam motifs, gene names, GO terms, and KEGG Orthology, facilitating more robust transcriptomic analyses. We also examined various stages of limb regeneration and development, gaining insights into the key signaling pathways involved. Overdiagnosis of atypical lipomatous tumors/well-differentiated liposarcomas by morphological diagnosis using only HE stained specimens: a case-control study with MDM2/CDK4 immunostaining and MDM2/CDK4 fluorescence in situ hybridization
Nagasaki University (長崎大学)博士(医学)Background: Lipomatous tumors represent the most common type of soft tissue neoplasms. Mouse double minute 2 homolog (MDM2)/cyclin-dependent kinase 4 (CDK4) immunostaining is considered effective in differentiating between benign lipomas and intermediate malignant atypical lipomatous tumors/well-differentiated liposarcomas (ALT/WDLPSs). However, these tumors have traditionally been diagnosed histopathologically using hematoxylin and eosin-stained specimens, which is referred to here as morphological diagnosis. In this study, the accuracy of morphological diagnoses that had been made before MDM2/CDK4 immunostaining became available for distinguishing between lipoma and ALT/WDLPS was examined. Methods: The study participants were 109 patients with a morphological diagnosis of lipoma (68 patients) or ALT/WDLPS (41 patients) who had undergone surgical resection of the tumor in our hospital between 2009 and 2012. Tissue samples from all patients were used for MDM2/CDK4 immunostaining and the confirmation of MDM2/CDK4 amplification by fluorescence in situ hybridization (FISH). Results: Of the 41 patients with a morphological diagnosis of ALT/WDLPS, only 17 were positive for MDM2 FISH. In addition, one of the 68 patients with a morphological diagnosis of lipoma showed MDM2 amplification by FISH. When the definitive diagnosis of ALT/WDLPS was made by the positive results of MDM2 FISH, the sensitivity and specificity of morphological diagnosis were 41.5% and 98.5%, respectively. The sensitivity of MDM2 and CDK4 immunostaining was 55.6% and 40.0%, respectively, and their specificity was 87.0% and 84.6%, respectively. This indicates that the diagnostic accuracy of these immunostaining assays was not particularly high. The clinical features suggesting ALT/WDLPS were: patient age (older), maximum tumor diameter (large, cut-off value of 125 mm), tumor location (lower limb), and tumor depth (deep-seated). Conclusions: Morphological diagnosis alone can accurately diagnose lipomas. However, it has a propensity to overdiagnose ALT/WDLPS. Dennd2c Negatively Controls Multinucleation and Differentiation in Osteoclasts by Regulating Actin Polymerization and Protrusion Formation
Nagasaki University (長崎大学)博士(歯学)Osteoclasts are bone-resorbing multinucleated giant cells formed by the fusion of monocyte/macrophage lineages. Various small GTPases are involved in the multinucleation and differentiation of osteoclasts. However, the roles of small GTPases regulatory molecules in osteoclast differentiation remain unclear. In the present study, we examined the role of Dennd2c, a putative guanine nucleotide exchange factor for Rab GTPases, in osteoclast differentiation. Knockdown of Dennd2c promoted osteoclast differentiation, resorption, and expression of osteoclast markers. Morphologically, Dennd2c knockdown induced the formation of larger osteoclasts with several protrusions. In contrast, overexpression of Dennd2c inhibited the multinucleation and differentiation of osteoclasts, bone resorption, and the expression of osteoclast markers. Dennd2c-overexpressing macrophages exhibited spindle-shaped mononuclear cells and long thin protrusions. Treatment of Dennd2c-overexpressing cells with the Cdc42 inhibitor ML-141 or the Rac1 inhibitor 6-thio-GTP prevented protrusion formation. Moreover, treatment of Dennd2c-overexpressing cells with the actin polymerization inhibitor latrunculin B restored multinucleated and TRAP-positive osteoclast formation.
