    Physicochemical analysis of selected recreational pools in Dasmariñas City, Cavite and detection of Cryptosporidium parvum and Giardia lamblia using loop-mediated isothermal amplification

    Going to swimming pools are one of the affordable recreational activities for many Filipinos. Nonetheless, the physicochemical characteristics of these pools might contribute to the presence of waterborne parasites such as Cryptosporidium parvum and Giardia lamblia. Various studies in the Philippines suggest the isolation of these parasites in local water sources, including these pools. Thus, this study aims to determine the presence of Cryptosporidium parvum and Giardia lamblia in the recreational pools of Dasmariñas, Cavite using a quantitative turbidimetric loop-mediated isothermal amplification (LAMP) and correlate it to the selected swimming pool’s physicochemical characteristics such as temperature, pH, total dissolved solids, turbidity, nitrate, and residual chlorine. The workflow of the study involved vacuum filtration of the collected water samples, extraction of the genomic DNA, analysis of the extracted DNA using UV-vis spectroscopy and gel electrophoresis, LAMP testing, and turbidimetric analysis of the LAMP. Results showed that the swimming pools maintained acceptable physicochemical characteristics, except for nitrate. Also, both Cryptosporidium parvum and Giardia lamblia are absent in all the swimming pools tested attributed to their strict adherence to sanitation protocols. Furthermore, there is no significant correlation between the presence of the parasites based on the turbidity of the LAMP and the physicochemical characteristics of the swimming pools, except for nitrate and turbidity of water samples for Giardia lamblia, potentially due to interfering chemicals in the water sample

    Cutting-edge Bioinformatics strategies for synthesizing Cyclotriazadisulfonamide (CADA) analogs in next-Generation HIV therapies

    Cyclotriazadisulfonamide (CADA) is a macrocyclic compound known for its unique mechanism in inhibiting HIV infection by downregulating the CD4 T-cell receptor, a crucial entry point for the virus. Unlike other antiretrovirals, CADA exhibits activity against a wide range of HIV strains, as all HIV variants require CD4 binding for infection. Furthermore, CADA has shown a synergistic effect with clinically approved anti-HIV drugs, offering potential for enhanced therapeutic strategies (Vermeire & Schols, [65]). One proposed mechanism for CADA’s inhibition of the CD4 receptor involves blocking the gates of the Sec61 channel, thereby preventing its translocation. However, CADA suffers from poor solubility and bioavailability. To address this, the study aimed to design CADA analogs with improved binding to the Sec61 channel, enhanced bioavailability, and reduced toxicity. The analogs were designed using SeeSAR, with Avogadro and Meeko used for 3D configuration and pseudoatom placement, respectively. AutoDock Vina version 1.2.4 was employed to predict the binding energies of these analogs. Of the 113 analogs designed, 93 demonstrated a more negative binding energy to the Sec61 channel compared to CADA. Structure-binding energy analyses were done to the top-binding analogs to show favorable structural modifications. Enzyme-ligand interactions were analyzed to elucidate the forces contributing to these binding energies. Additionally, 33 of the 113 analogs were deemed bioavailable using a bioavailability criteria specific for macrocycles. Toxicity predictions using PASS Online and StopTox identified analogs JGL023, JGL024, JGL032, and JGL047 as potential drug candidates. Molecular dynamics simulations using Gromacs-2020.4 revealed that JGL023 and JGL032 exhibited the most favorable binding to the Sec61 channel, as determined by evaluating ligand and residue flexibility, compactness, contact frequency, motion pathways, free energy, and other relevant parameters. Synthetic routes for these four analogs were proposed for future studies. The results of this study offer a new perspective on developing drugs to inhibit HIV entry

    Identifying the presence of soil-transmitted helminths in Lactuca sativa (lettuce) and Brassica oleracea var. capitata (cabbage) from kadiwa market and central market, Dasmariñas City, Cavite

    Soil-transmitted helminths (STH), also known as intestinal nematodes, require soil to reach the infective stage, commonly including Ascaris lumbricoides (giant roundworm), Trichuris trichiura (whipworm), Hookworm. The main purpose of the study is to determine the presence of Soil- transmitted helminths in Lactuca sativa (Lettuce) and Brassica oleracea var. capitata (Cabbage) from Kadiwa and Central Market in Dasmariñas, Cavite. In addition to that, the researchers utilized a descriptive method and sedimentation technique to identify the STH present in the said vegetables. The results showed that 13 STH are present in Kadiwa Market and 18 STH in Central Market out of the 70 samples tested from each market. The most common STH seen in both markets is the Ascaris lumbricoides egg. While there is a presence of STH from the samples, there is no significant association between L. sativa and B. oleracea var. capitata samples and parasite detection, and no significant association between market source (Kadiwa and Central Market) and parasite detection. Results indicates that there is insufficient evidence that one vegetable type is more likely to contain parasite than the other, and not enough evidence to say that vegetables from one market are more likely to have parasites than those from the other. Further studies are necessary for the establishment of a well-grounded information leading to a better understanding regarding food safety awareness

    Lichen Planus Pigmentosus and Vitiligo in a 61-Year-Old Filipino Man: Case Report

    Pigmentary disorders have been implicated in causing psychosocial turmoil in patients as they can cause some degree of cosmetic disfigurement. Lichen planus pigmentosus (LPP) presents as ashy, dermatosis-like eruptions on sun-exposed areas, particularly on the head, neck, and earlobes. On the other hand, vitiligo is a chronic disorder that appears as depigmented patches on the skin. A 61-year-old man with Fitzpatrick skin phototype IV presented to us initially with LPP but eventually also developed vitiligo. The patient was treated with low-dose oral isotretinoin for LPP and topical tacrolimus 0.1% ointment for both LPP and vitiligo with a good clinical outcome. One case of segmental vitiligo and zosteriform LPP, affecting a 22-year-old Indian woman, has been previously reported in the English-language literature. An autoimmune etiology that causes melanocytorrhagy may be a plausible hypothesis for the coexistence of these 2 conditions
