    Get PDF

    Get PDF

    Get PDF

The effects of postharvest carbon dioxide and a cold storage treatment on Tuta absoluta mortality and tomato fruit quality

    Get PDF
Tuta absoluta is an invasive pest species that affects tomatoes and other solanaceous crops and is found in Europe and other Mediterranean areas. Hitherto, fumigation with methyl bromide is the only measure used to control this pest during the postharvest period. Because of methyl bromide phytotoxicity and health, safety and environmental concerns, alternatives to this product need to be investigated. Therefore, the objective of this study was to determine the ability of T. absoluta to complete its preimaginal development on tomato fruit during the postharvest period and to evaluate the effectiveness of different supra atmospheric carbon dioxide (CO2) levels and cold storage treatments on T. absoluta control. T. absoluta was unable to complete its development from egg to adult on fruit of three tomato varieties. In contrast, T. absoluta completed its preimaginal development when more mature larvae were provided with the tomato fruit. The exposure of T. absoluta to a modified atmosphere of 95% CO2 at 25 °C for 48 h was effective for the control of all life stages, but negatively affect fruit quality. An increment in the exposure time to 72 h was necessary in order to obtain the same level of control at 40% CO2. A cold storage treatment at 1 °C for a total of 10 days was also effective for the control of the T. absoluta eggs. These two treatments did not negatively affect the quality of the fruit. Therefore, these appear to be effective alternatives to postharvest methyl bromide fumigation and future studies need to be planned to validate the commercial feasibility of these findings

Draft genome sequence for virulent and avirulent strains of Xanthomonas arboricola isolated from Prunus spp. in Spain

    Get PDF
Xanthomonas arboricola is a species in genus Xanthomonas which is mainly comprised of plant pathogens. Among the members of this taxon, X. arboricola pv. pruni, the causal agent of bacterial spot disease of stone fruits and almond, is distributed worldwide although it is considered a quarantine pathogen in the European Union. Herein, we report the draft genome sequence, the classification, the annotation and the sequence analyses of a virulent strain, IVIA 2626.1, and an avirulent strain, CITA 44, of X. arboricola associated with Prunus spp. The draft genome sequence of IVIA 2626.1 consists of 5,027,671 bp, 4,720 protein coding genes and 50 RNA encoding genes. The draft genome sequence of strain CITA 44 consists of 4,760,482 bp, 4,250 protein coding genes and 56 RNA coding genes. Initial comparative analyses reveals differences in the presence of structural and regulatory components of the type IV pilus, the type III secretion system, the type III effectors as well as variations in the number of the type IV secretion systems. The genome sequence data for these strains will facilitate the development of molecular diagnostics protocols that differentiate virulent and avirulent strains. In addition, comparative genome analysis will provide insights into the plant-pathogen interaction during the bacterial spot disease process

Salt-tolerant rootstock increases yield of pepper under salinity through maintenance of photosynthetic performance and sinks strength

The performance of a salt-tolerant pepper (Capsicum annuum L.) accession (A25) utilized as a rootstock was assessed in two experiments. In a first field experiment under natural salinity conditions, we observed a larger amount of marketable fruit (+75%) and lower Blossom-end Root incidence (-31%) in commercial pepper cultivar Adige (A) grafted onto A25 (A/A25) when compared with ungrafted plants. In order to understand this behavior a second greenhouse experiment was conducted to determine growth, mineral partitioning, gas exchange and chlorophyll a fluorescence parameters, antioxidant systems and proline content in A and A/A25 plants under salinity conditions (80 mM NaCl for 14 days). Salt stress induced significantly stunted growth of A plants (-40.6% of leaf dry weight) compared to the control conditions, while no alterations were observed in A/A25 at the end of the experiment. Accumulation of Na+ and Cl- in leaves and roots was similar in either grafted or ungrafted plants. Despite the activation of protective mechanisms (increment of superoxide dismutase, catalase, ascorbate peroxidase activity and non-photochemical quenching), A plants showed severely reduced photosynthetic CO2 assimilation (-45.6% of A(N390)) and substantial buildup of malondialdehyde (MDA) by-product, suggesting the inability to counteract salt-triggered damage. In contrast, A/A25 plants, which had a constitutive enhanced root apparatus, were able to maintain the shoot and root growth under salinity conditions by supporting the maintained photosynthetic performance. No increases in catalase and ascorbate peroxidase activities were observed in response to salinity, and MDA levels increased only slightly; indicating that alleviation of oxidative stress did not occur in A/A25 plants. In these plants the increased proline levels could protect enzymatic stability from salt-triggered damage, preserving the photosynthetic performance. The results could indicate that salt stress was vanished by the lack of negative effects on photosynthesis that support the maintained plant growth and increased marketable yield of the grafted plants. Diversity in the trifoliate orange taxon reveals two main genetic groups marked by specific morphological traits and water deficit tolerance properties

    No full text
Trifoliate orange (Poncirus trifoliata (L.) Raf.) is a very useful taxon for the citrus industry since this rootstock is immune to the Citrus Tristeza virus and confers cold tolerance. Numerous trifoliate orange varieties exist but little is known regarding their behavioural variability when subjected to abiotic constraints. The diversity of 74 P. trifoliata accessions maintained in the INRA-CIRAD Citrus Germplasm Collection was investigated using simple sequence repeat markers. Two major genetic groups were clearly identified as a few homonyms, intergroup or intra-group hybrids and doubled-chromosome tetraploid forms. The Group 1 phenotype was characterized by larger flowers and leaves and smaller seeds than Group 2. Tetraploid accessions showed larger leaves and heavier seeds than all other diploid accessions, regardless of genetic classification. Eight genotypes belonging to both genetic groups, as well as two hybrids between the two groups, were selected to investigate their water deficit tolerance. Stress was applied by withdrawing irrigation for 4 weeks. Physiological parameters such as leaf stomatal conductance, quantum yield of photosystem II electron transport, soil water potential, leaf osmotic potential and transpiration rate were estimated. Some varieties, such as Rubidoux 0101033, were clearly more tolerant to water deficit than others, such as Pomeroy 0101040 and Pomeroy 0110081. Interestingly, accessions that had the highest soil water potential and were the least affected by stress belonged to genetic Group 2. Conversely, trifoliate oranges of genetic Group 1 were the least tolerant

Can the Genetic Origin Affect Rabbit Seminal Plasma Protein Profile along the Year?

    No full text
Contents The study was designed to evaluate the influence of genetic origin on rabbit seminal plasma protein profile variation along the year. Seminal plasma of rabbits from line A (maternal line) and R (paternal line) collected during a natural year was subjected to polyacrylamide gel electrophoresis (SDS-PAGE). The electrophoretic profile of rabbit seminal plasma resulted in multiple protein bands of different intensity ranging from 9 to 240kDa. Results showed that seven protein bands were significantly different between genetic lines, and among these, three protein bands were significantly different between seasons. The differentially expressed proteins were identified by MALDI-TOF/TOF or LC-MS/MS analysis and were the following ones: FAM115E-like (220, 113 and 59kDa), ectonucleoside triphosphate diphosphohydrolase 3 isoform X2 (72kDa), annexin A5 (32kDa), lipocalin allergen Ory c 4 precursor (19kDa), and haemoglobin subunit zetalike (13kDa) between genetic lines and FAM115E-like (113kDa), haemoglobin subunit zetalike (13kDa) and -nerve growth factor (12kDa) between seasons. These results indicate that proteins from rabbit seminal plasma are under both seasonal control and genetic control. Furthermore, the differential presence of these proteins could be one of the causes explaining the differences observed in fertility and seminal parameters between these two lines in earlier studies

    Get PDF

Parasitism activity of Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Braconidae) and Aganaspis daci (Weld) (Hymenoptera: Figitidae) against Ceratitis capitata (Wiedemann) (Diptera: Tephritidae) under Mediterranean climatic conditions

Background: Ceratitis capitata (Wied.), the Mediterranean fruit fly, is one of the key pest species affecting citrus production around the Mediterranean coasts of Spain, Morocco and Tunisia. During the past decade the IVIA (Valencian Institute for Agricultural Research) has imported several parasitoid species to enhance a Biological Control (BC) program against this pest. Soon after the introduction of Diachasmimorpha longicaudata (Ashmead) in 2009, a native parasitoid, the figitid Aganaspis daci (Weld), was identified in Bétera, a town in the province of Valencia. This work will contribute to highlight the importance of the two species within the BC program against C. capitata in the study areas. Methods: To determine the influence of climatic factors on parasitism rate and Medfly mortality, apples artificially infested with Medfly larvae were individually exposed to five parasitoid couples for one week under natural conditions. Forty wood-framed mesh cages (twenty for each parasitism rate and immature development) of each parasitoid species were tested weekly over 10 weeks across one year. Results: Under Mediterranean climatic conditions, D. longicaudata exerted a high parasitism rate compared with A. daci. Extreme winter and summer temperatures seem to affect the immature development of both species. A higher immature mortality was observed for A. daci throughout the year, compared with D. longicaudata. Adult parasitoid species were capable of parasitizing C. capitata L2/L3 larvae at the extreme temperatures tested. Conclusions: This study suggests that A. daci shows a good performance as well as D. longicaudata against C. capitata under Mediterranean climatic conditions. Further studies are required to determine the influence of other climatic factors and whether the two parasitoid species act in synergism
