Protocolo para obtener células en suspensión de Artemia franciscana para realizar electroforesis unicelular alcalina
Unicellular electrophoresis is a highly sensitive assay to detect DNA damage caused by various physical, chemical and biological agents. Despite Artemia franciscana is an important experimental model in different areas, there are no studies that refer to genotoxicity in Artemia through this assay. The aim of this study was to establish the protocol to obtain a cell suspension of A. franciscana appropriate in quality and quantity to assess single DNA strand breaks through the comet assay in the future studies.La electroforesis unicelular es un ensayo muy sensible para detectar el daño en el ADN provocado por diferentes agentes físicos, químicos y biológicos. A pesar de que Artemia franciscana es un modelo experimental muy importante en diferentes áreas, en la literatura no se encuentran estudios en los que se lleve a cabo esa prueba de genotoxicidad con células de esta especie. The binomial Gracilaria parvispora I.A. Abbott (Gracilariaceae, Rhodophyta) is assigned to gracilarioid plants growing in the coastal lagoons and estuaries of Laguna Superior, Inferior, Mar Muerto, and Mar Tileme from Gulf of Tehuantepec, in the South Central Mexican Pacific (Oaxaca-Chiapas). The taxon is described in detail and its taxonomic relationships ith other similar taxa are briefly discussed. This species has a tropical-subtropical distribution in the Pacific Ocean.El binomio Gracilaria parvispora I.A. Abbott (Gracilariaceae, Rhodophyta) es asignado a plantas "gracilarioides" presentes en las Lagunas Superior, Inferior, Mar Muerto y Mar Tileme del Golfo de Tehuantepec, Pacífico centro-sur mexicano (Oaxaca-Chiapas). El taxon es descrito detalladamente y se discute brevemente su relación con otros taxa similares. La especie tiene una distribución tropical-subtropical en el Oceáno Pacífico
Aquatic macrophytes tolerance to domestic wastewater and their efficiency in artificial wetlands under greenhouse conditions
Aquatic and semi-aquatic plant species of three different water qualities were inventoried, two of the El Tunal river and one of one of its tributaries, considering its content of dissolved oxygen, soluble phosphates, nitrate, ammonia, fecal coliforms, total suspended solids, and measurements of pH and electrical conductivity. A MANOVA/ANOVA demonstrated significant differences among parameters and sites. Twenty-eight species were identified; from them: Schoenoplectus americanus, S. tabernaemontani and Eleocharis densa were selected. All three were grown successfully under greenhouse conditions. Adaptation to local wastewater was evaluated using 5 micro-units: one control with wastewater (WW), another with gravel (G), and three sub-surface flow wetlands, one for each of the three selected plants, in duplicate. For ammonia and phosphate concentration, the systems with gravel removed 96 - 98%, and 99 - 100%, respectively. Fecal coliforms content was reduced about the same in all systems, 98.5 - 98.7%. No significant differences were found in removal of fecal coliforms and ammonia across time or among species. Removal of ions (98% in 48 h) was due mainly to the gravel used as support, for its ionic exchange capacity. Nonetheless, the three selected species are considered as appropriate for wetland construction because they are native, abundant, tolerant to local conditions, easy to propagate and establish, and highly tolerant to wastewater in their place of origin. Its dense growing habit would represent also a refuge for wildlife, another goal for constructing a wetland in the area
Análisis químico-biológico para determinar el estatus trófico de la Laguna de Tres Palos, Guerrero, México
An evaluation of the main physicochemical and biological parameters was carried out in Tres Palos coastal lagoon in 2003; the aim of the study was to evaluate the environmental quality and the trophic status of the lagoon, and to compare these results with the data obtained in 1971. The salinity, temperature and dissolved oxygen remained similar: oligohaline (2-4 ups), warm (28-31°C), with dissolved oxygen oversaturation at the surface and hypoxia or anoxia at the bottom. However, other characteristics changed considerably. In 1971 the coastal lagoon was eutrophic with high densities of phytoplankton (8 x 103 cel mL-1), and blooms of cyanophytes (i.e., Mycrocystis, Anabaena, Spirulina). Thirty-two years later, the trophic status of Tres Palos coastal lagoon evolved to hypereutrophy. Phytoplankton biomass (?100 mg chlorophyll a m-3) and density (i.e., >10 x 104 cel mL-1) increased in response to the large concentration of available phosphorus (3.6 to 80.6 µM); the cyanophyta continued as the dominant group but with different representative species (i.e., Aphanocapsa delicatissima, Merismopedia punctata, Chroococcus dispersus var. minor, Raphidiopsis curvata and Pseudoanabaena limnetica). The ß-mesosaprobic organisms dominated and revealed the large amount of organic matter coming from allochthonous (urban and industrial wastewater) and autochthonous (phytoplankton) sources.Se llevó a cabo una evaluación de los principales parámetros fisicoquímicos y biológicos de la Laguna de Tres Palos en 2003; el objetivo fue evaluar la calidad ambiental y estatus trófico correspondiente y finalmente, hacer una comparación con las características evaluadas en 1971. La salinidad, temperatura y oxígeno disuelto permanecieron similares: oligohalina (2-4 ups), cálida (28-31°C), con sobresaturación superficial y condiciones de hipoxia o anoxia en el fondo. Sin embargo, otras características se modificaron dramáticamente. En 1971 la laguna era eutrófica con elevadas densidades de fitoplancton (8 x 103 cel mL-1) y florecimientos de cianofitas (i.e., Mycrocystis, Anabaena, Spirulina). Treinta y dos años después, la condición trófica de Tres Palos evolucionó hacia la hipereutrofia. La biomasa (?100 mg clorofila a m-3) y densidad (p. ej., >10 x 104 cel mL-1) del fitoplancton aumentaron en respuesta a la gran cantidad de fósforo disponible (3.6 a 80.6 µM); las cianofitas continuaron siendo el grupo dominante pero las especies representativas cambiaron (i.e., Aphanocapsa delicatissima, Merismopedia punctata, Chroococcus dispersus var. minor, Raphidiopsis curvata y Pseudanabaena limnetica). Los organismos ß- mesosaprobios fueron dominantes, revelando una elevada concentración de materia orgánica la cual proviene de fuentes tanto alóctonas (aguas residuales urbanas e industriales) como autóctonas (fitoplancton)
Florecimientos de dinoflagelados nocivos en la costa Pacífica de Costa Rica
Since 1979 has been reported harmful microalgae blooms (HAB) on the Pacific coast of Costa Rica, it was identified until 2005 at least 13 taxa of dinoflagellates. In recent years these phenomena have intensified both spatially and temporarily, discolorations occur almost uninterrupted; they are distributed for months and which extend along the coast causing death fish and smelly in the water. Such massive FAN started with Pyrodinium bahamense, whose discoloration in 2000 lasted for more than one year and in which morphotypes were observed the two varieties of the species, coexisting alongside with Gymnodinium catenatum and causing serious cases of paralytic shellfish poisoning by seafood consumption (PSP). Later, from September 2003 until June 2004, Cochlodinium cf. polykrikoides produced discolorations in the entire Pacific coast followed by extensive stains of Akasiwo sanguinea and Gymnodinium instriatum. The latest event was observed in June 2005 when Alexandrium monilatum produced extensive discolorations with chains formed by more than 100 cells. The HAB of dinoflagellates are now common along the coasts of Costa Rica, possibly as a result of the current environmental conditions that favor the massive proliferation of invasive species (aggressive), which can even affect other sites in the American Pacific as the case of P. bahamense var. compressum which can be moved towards Mexico along the Central Pacific Coast through the Costa Rica Current Flow and Mexican Western Current.Desde 1979 se han reportado florecimientos de especies de microalgas nocivas (FAN) en la costa Pacífica de Costa Rica, reconociéndose hasta el 2005 al menos 13 taxa de dinoflagelados. En años recientes estos fenómenos se han intensificado tanto espacial como temporalmente, produciéndose discoloraciones casi ininterrumpidas que se extienden por toda la costa produciendo mortalidad de peces y fetidez en el agua. Tales FAN masivos se iniciaron con Pyrodinium bahamense en el 2000, quien perduró por más de un año y en el cual se observaron morfotipos correspondientes a las dos variedades de la especie, coexistiendo junto a Gymnodinium catenatum y ocasionando serios casos de Intoxicación Paralizante por Consumo de Mariscos (IPM). Posteriormente, de septiembre del 2003 hasta junio del 2004, Cochlodinium cf. polykrikoides produjo discoloraciones en toda la costa Pacífica, seguidas por manchas extensas de Akasiwo sanguinea y Gymnodinium instriatum. El evento más reciente se observó en junio del 2005 cuando Alexandrium monilatum produjo extensas discoloraciones con cadenas formadas por más de 100 células. Los FAN de dinoflagelados ahora son comunes en las costas de Costa Rica, posiblemente como resultado de las condiciones ambientales actuales que favorecen la proliferación masiva de especies invasivas (agresivas), las cuales incluso pueden afectar otros sitios del Pacífico americano como es el caso de P. bahamense var. compressum que puede desplazarse hasta México, a lo largo de la costa del Pacífico centroamericano a través de la Corriente Costera de Costa Rica y la Corriente Occidental Mexicana
Influence of phytoplankton diets on the ingestion rate and egg production of Acartia clausi and A. lilljeborgii (Copepoda: Calanoida) from Bahía de La Paz, Gulf of California
Different phytoplankton diets were tested on Acartia clausi and A. lilljeborgii from Bahía de La Paz to determine their effect on survival, egg production, and ingestion rate. Female copepods were fed diatom strains (Chaetoceros sp., Cylindrotheca closterium, Odontella longicruris, and Dytilum brightwelli), dinoflagellate strains (Scrippsiella sp., Gyrodinium sp., Prorocentrum micans, and P. rhathymum), and one Raphidophyceae (Chattonella sp.). After 24 h of incubation in darkness at 24 °C, survival with various phytoplankton diets was above 91%. Gyrodinium sp. produced the lowest survival in both copepod species (44.5% in A. clausi and 89.6% in A. lilljeborgii). Dinoflagellate diets provided the highest egg production. A. clausi had higher egg production when fed with P. rhathymum, P. micans, Gyrodinium sp., Scrippsiella sp., and Chattonella sp. A. lilljeborgii had a higher egg production with Scrippsiella sp., P. micans, Gyrodinium sp., and Chaetoceros sp. Ingestion rates were higher in both Acartia species with two diatom diets (O. longicruris and Chaetoceros sp.), P. rhathymum and Chattonella sp. These results suggest that both Acartia species respond to nutritional quality of phytoplankton in a short time. Higher ingestion rates did not necessarily result in higher egg production, suggesting that the link between ingestion and egg production may be in what is eaten, rather than in how much. The response in egg production seems to be species specific, but in general was higher with dinoflagellate diets, suggesting a higher food quality compared with diatoms (excepting Chaetoceros sp.)
