Endothelin-1 (ET-1) is a potent vasoconstrictor hormone that has been identified as an important factor
responsible for the development of cardiovascular dysfunctions. ET-1 exerts its vasoconstrictor activity
through two pharmacologically distinct receptors, ETA and ETB that are found in vascular smooth muscle
cells (VSMCs) and the vasodilator activity through an ETB receptor located on endothelial cells. This study
aimed to show the impact of 1µM L-arginine (LA), 100µM tetrahydrobiopterin (BH4), and their combined
effect on ET-1 activity in both lead-treated and lead-untreated rat aortic rings. This means, investigating how
endothelial dysfunction reverses the role of nitric oxide precursor and cofactor. In this study, Rat aortic rings
have been pre-incubated with BH4, LA and their combination. Subsequently, the aortic rings were preincubated with 200µM N-Nitro-L-arginine methyl ester (L-NAME) and 0.5µM BQ-123. Then, the vascular
response to cumulative doses of rat ET-1 was analyzed in each of the above-mentioned groups (LA, BH4, LA
& BH4, L-NAME, BQ-123), in the presence and absence of lead acetate 1µM Pb (C2H3O2)2. ET-1 efficacy and
potency were significantly decreased in the presence of LA, BH4, and LA and BH4 combination in the untreated
group, while it significantly increased in the presence of lead. In the second trial of experiments ET-1 efficacy
markedly decreased in BQ-123- incubated cells in both lead-treated and untreated aortic rings. In the presence
of lead, the efficacy of ET-1 was raised with the use of L-NAME. In conclusion, LA and BH4 can be considered
pharmacological agents to alter the potency of ET-1-induced vasoconstriction and concomitantly lower blood
pressure
Down-regulation of abaI, abaR, Bap and OmpA genes in Acinetobacter baumannii by ethanol extract of Glycyrrhiza glabra after toxicity assessment
Acinetobacter baumannii is a pathogen that has caused rising concerns within healthcare facilities in recent years. As antibiotic overuse and resistance rise, natural remedies with the potential have received attention as antibiotics that might have fewer side effects and lower resistance. Glycyrrhiza glabra was used to investigate its effects on A. baumannii's quorum sensing and biofilm production abilities. In this study, the toxicity assessment of Glycyrrhiza glabra L. extract on rats, the phytochemical analysis and the quantitative measurement for the association of the biofilm reduction with the active components in the plant was determined. The results indicated ciprofloxacin and gentamicin were the most effective antibiotics and that various capabilities of biofilm-productions were demonstrated, only four percent of the samples established robust biofilm, while 40% to 56% demonstrated weak to moderate biofilm production, respectively. Phytochemical qualitative testing of ethanol leaf extracts from Glycyrrhiza glabra showed the existence of flavonoids, alkaloids, phenolic, tannic acid, and terpenoids, but no saponins. Assessment of toxicity revealed a low hazard, with an LD50 of 4.95 g/Kg. Our results showed that the extract's SICs elucidated a substantial quantitative decrease in biofilm production by the bacterial isolates, including the reference ATCC strain, which is known to be a potent biofilm producer. As a conclusion, biofilm creation in Acinetobacter baumannii has been shown to be greatly reduced by G. glabra extract
New Tricholomalides D–G from the Mushroom Tricholoma ustaloides Grown in an Italian Beech Wood
Four novel seconeodolastane diterpenoids, named tricholomalides D–G, were isolated, together with the known tricholomalide C, from the fruiting bodies of Tricholoma ustaloides Romagn., a species belonging to the large Tricholoma genus of higher mushrooms (Basidiomycota, family Tricholomataceae). They were isolated through multiple chromatographic separations, and the structures, including the absolute configuration, were established through a detailed analysis of MS, NMR, and CD spectral data and comparison with related compounds reported in the literature, which has been thoroughly revised
Evaluation of dietary rosemary leaf powder on growth, carcass composition and hemato-biochemical profiles of common carp (Cyprinus carpio) reared in cage system
This study was conducted to evaluate the effect of rosemary (Rosmarinus officinalis) leaf powder (RLP) at 0.0% (control), 0.5%, 1% and 2% as a dietary supplement for eight weeks, on the growth performance, carcass composition and haemato-biochemical parameters. One hundred twenty common carp (Cyprinus carpio) (21.11±0.17g) were randomly distributed to 12 cages (0.2 m3) with three replicates for 56 days. The results showed that RLP-supplemented diets achieved significant enhancement of growth performance (FBW, SGR and WG) and feed utilization (FCR, FCE and PI) compared with a control group. In addition, TFI and PI were significantly improved in 0.5 and 2% RLP-supplemented groups. RLP had no significant effects on body composition except moisture content which was significantly increased in fish fed with a 1% supplemented diet and crude protein which was significantly decreased in fish fed with a 1% supplemented diet compared to the control diet. The results showed a significant increase in RLP-supplemented diets in RBC counts, LYM, MCV, hematocrit, total protein, albumin and globulin compared with the control group. WBC counts significantly increased in fish fed the 0.5 and 2% RLP diets, and hemoglobin significantly elevated in fish fed the 1% RLP diet. Glucose, GRA, MCH, MCHC and MPV were significantly decreased in the group fed the RLP diets compared with the control group. The AST, ALT, LDL, VLDL, CHO and TG levels showed a significant decrease in RLP compared to the control diet (P 0.05). The urea and creatinine levels were higher in the RLP groups compared with the control (P < 0.05). Dietary RLP 1% increased lipase when compared to the control group. However, amylase levels in fish fed 0.5 and 1% RLP has significantly improved, while fish fed 2% RLP has significantly decreased when compared to the control diet. Phosphorus encapsulated gallium nitride and aluminum nitride nanotubes as nonenzymatic sensors for fructose, glucose, and xylose sugars as biomarkers for diabetes-mellitus: Outlook from computational study
Excessive sugar consumption has been correlated with various adverse health outcomes, encompassing both short-term and long-term implications for human well-being. Traditional approaches for sugar detection, such as chromatography, spectroscopy, and enzymatic assays, necessitate significant time, specialized equipment, and expertise. In this study, we explore the potential of phosphorus-doped Gallium nitride (P@GaNNT) and aluminum nitride (P@AlNNT) nanotubes as novel means to detect three distinct sugars: fructose (F), glucose (G), and xylose (X). To investigate their capabilities, we employ density functional theory (DFT) computations at the B3LYP-D3(BJ)/def2-SVP methodology. The molecular orbital analysis of the complexes provided evidence of reduced energy gap (Eg) values compared to the surfaces in their pristine states. The X_P@AINNT interaction was the most stable complex, with an energy gap (Eg) value of 4.408eV while G_P@AINNT was the most reactive complex, with an Eg value of 0.545eV. When these complexes were evaluated in a solvent (water), their stability was found to be higher than their reactivity, as evidenced by the increased Eg values for each complex. Results from topological studies (QTAIM and NCI) showed the presence of covalent, electrostatic, and weak van der Waals interactions among atoms in these systems. The adsorption energies for F_P@AlNNT and F P@GaNNT indicated that fructose was chemisorbed onto P@AlNNT and P@GaNNT, with values of -1.442eV and -1.469eV, respectively. On the other hand, glucose and xylose were found to be physiosorbed on P@GaNNT and P@AlNNT, based on the positive results from their adsorption. This study demonstrated the potential of P@AlNNT and P@GaNNT as valuable tools for sugar detection
Pomegranate Juice Attenuates Anti-ulcer in Indomethacin-induced Peptic Ulcer
Introduction: Pomegranate (Punica granatum L.) is a well-known fruit that grows in tropical and subtropical areas of the world. In this study, we wanted to find out how pomegranate juice (POMJ) affected rats with gastric ulcers caused by standard drugs. Indomethacin is the most common drug causes gastric ulcer also, most of the NSAID drugs have harmful side effects, so studies have focused on finding an alternative natural solution. Methods: In this study, 25 male albino rats were used and were split into four groups of five rats each. These groups were called control, pomegranate, indomethacin, and standard. The examination took one week to complete. Indomethacin saline suspension (100 mg/kg rat weight) caused gastric ulcers. Pomegranate peel juice (5-10%) reduced stomach ulcer area and ulcer index, gastric juice volume, and acidity. Pomegranate juice restores stomach mucus content and tissue at the histological level. Results: Rats that were given indomethacin and then given pomegranate juice were significantly less likely to get a gastric ulcer. It also lowered the ulcer index to 0.7093±0.36 showing that 52.25 % prevention. Conclusion: The study's macroscopical and microscopical results showed that pomegranate juice might be able to reduce the ulceration caused by indomethacin in a rat model. Pomegranate as a protective food supplement against gastric ulcers
Effect of Plumbago zeylanica on Analgesia and Arthritis
Background: Rheumatoid arthritis is a chronic inflammatory joint condition that causes oxidative damage and inflammation. The current study evaluates the safety and effectiveness of an 85 percent methanolic extract of Plumbago zeylanica (PZ). Methods: The acute toxicity study is used to calculate the LD50 value in the safety profile. The hot plate and tail immersion procedures are both used to examine the analgesic effect in the efficacy profile. Paw edema caused by carrageenan was investigated for its ability to reduce inflammation and complete Freund's adjuvant produced arthritis was tested for anti-arthritic activity. Results: Various phytochemical analyses, including qualitative and quantitative analyses of 85 % methanolic extract and raw materials. The extract exhibits analgesic effects by increasing the reaction time in both the hot plate and tail immersion procedures. In Freund's adjuvant-induced paw edema, the anti-inflammatory activity of methanolic extract is 30.42 % and 16.90 %, respectively. Conclusion: The PZ extract raises the level of antioxidants and lowers the oxidative stress caused by arthritis caused by complete Freund's adjuvant. PZ has a lot of secondary metabolites that can help with pain, inflammation, and free radicals
Current landscape of miRNAs and TGF‐β signaling in lung cancer progression and therapeutic targets
Lung cancer (LC) is the primary reason for cancer-associated fatalities globally. Due to both tumor-suppressing and tumor-promoting activities, the TGF-β family of growth factors is extremely essential to tumorigenesis. A non-coding single-stranded short RNA called microRNA (miRNA), which is made up of about 22 nt and is encoded by endogenous genes, can control normal and pathological pathways in various kinds of cancer, including LC. Recent research demonstrated that the TGF-β signaling directly can affect the synthesis of miRNAs through suppressor of mothers against decapentaplegic (SMAD)-dependent activity or other unidentified pathways, which could generate allostatic feedback as a result of TGF-β signaling stimulation and ultimately affect the destiny of cancer tissues. In this review, we emphasize the critical functions of miRNAs in lung cancer progression and, more critically, how they affect the TGF-β signaling pathway, and explore the role of both the TGF-β signaling pathway and miRNAs as potential therapeutic targets for improving the treatments of LC patients
Essential Oil Composition of calotropis procera (Aiton) Dryand
The use of herbal medicines is significant in healthcare initiatives, particularly in underdeveloped nations. All plants or parts of plants are thought to be possible sources of therapeutic substances, which includes a surprisingly comprehensive definition of what constitutes a medical plant. The literature and our own research both suggest that few studies exist that describe the chemical composition of Calotropis procera outside of Nigeria. Therefore, the purpose of this research was to describe the constituents of Calotropis procera essential oil, which is found in the northern part of Nigeria. Hydro distillation was used to distil the oil. An Agilent (Series MSD) gas chromatography 7890A/5975C linked mass spectrometer was used for the analysis of the oil. In the analysed essential oil, monoterpenes (60.4% of the oil) were the most prevalent compound, followed by other substances (23.81%) and sesquiterpenoids (15.67%). Phellandrenebeta-> (8.70), decanal 4Z-> (8.48), and perilla ketone (7.77) % are the three most prevalent substances. This examination will provide as a starting point for more study on the subject plant
