Travelling with Interrail through Europe: A potential analysis in the context of climate change
Die vorliegende Bachelor-Thesis untersucht die Attraktivität und Zukunftsfähigkeit von Interrail im Kontext des fortschreitenden Klimawandels. Tourismus ist gleichermaßen Mitverursacher sowie Leidtragender von negativen Auswirkungen, weshalb Handlungsbedarf für zunehmende Nachhaltigkeit erforderlich ist. Dabei wird herausgestellt, dass Zugreisen klimafreundlich sind und dadurch Interrail in Europa eine klimafreundliche Reisealternative bietet. Mithilfe einer Potenzialanalyse basierend auf einer Literaturanalyse und einer quantitativen empirischen Erhebung werden die Attraktivität und Zukunftsfähigkeit von Interrail untersucht. Die Ergebnisse werden in einer SWOT-Analyse hinsichtlich der Stärken, Schwächen, Chancen und Risiken von Interrail analysiert. Die Ergebnisse zeigen, dass Interrail großes Potenzial als attraktive Reiseform besitzt, insbesondere aufgrund seiner Flexibilität, der zahlreichen Reisemöglichkeiten und der Budgetfreundlichkeit für die primäre Zielgruppe. Dennoch gibt es Bereiche die Handlungsbedarf erfordern, um das Produkt weiter zu verbessern und Hindernisse zu beseitigen, damit Reisende sich in Zukunft vermehrt für Interrail als klimafreundliche Alternative entscheiden
Near surface seismic inversion: quantitative interpretation of the physical properties of unconsolidated sediments
Active reflection seismic data offer unprecedented possibilities to image structures in the Earth’s interior. Employing inversion techniques, those images can be complemented by quantitative information making seismic interpretation less ambiguous and enabling the deduction of subsurface properties of relevance for applied research or engineering purposes. Mostly deep targets in lithified sediments or rocks have been targeted with seismic inversion studies and employed low resolution seismic sources. Constraints and methods established for this purpose are not applicable to the marine near surface, which is mainly comprised of unconsolidated sediments, and for which high resolution seismic signals are required. The comparatively few high resolution studies focussing on the marine near surface proofed the general feasibility of marine near surface seismic inversion, which sparked industry interest, e.g., for offshore wind farm investigations. However, the methods often require additional ground truthing data, or are not only based on the reflected wavefield, or methodological limitations impede a general application.
To invert marine near surface seismic data, this study focused on the optimization of data acquisition, and the development of inversion methods based exclusively on P-Wave reflection seismic data aided by empirical relations fitted to unconsolidated sediments. For the data acquisition optimization, the different types of unconsolidated sediments are characterized and detailed analyses of two types of near surface seismic sources are conducted. On this basis, numerical models and inversion trials are developed to test acquisition set-ups and to investigate the potential of the different inversion approaches for, among others, shear property estimation. These tests are then used to develop a catalogue of requirements for data acquisition and possible limitations are indicated. As a proof of concept for impedance inversion, seismic data from the North Sea and the Baltic Sea in offshore wind park areas have been inverted. For this purpose, algorithms have been improved and implemented for attenuation estimation and correction, low frequency trend generation by interval velocity estimation and conversion to density, band limited impedance inversion, merging of low frequency trends with band limited impedance as well as depth conversion. The resulting impedance profiles resemble a significant improvement for quantitative interpretation of geologically complex areas reducing interpretation biases and capture intra-unit lithologic variations and property boundaries which are not evident on seismic amplitude images. It is demonstrated that the impedance correlates well to Cone Penetration Test measurements and that the Soil Behaviour Type can be classified based on the impedance. Thus, these seismic inversion results improve the extraction of important sediment properties, e.g., for engineering purposes. For the estimation of shear properties, a pre-stack inversion algorithm with a steepest descent optimization scheme and a convolutional forward model based on unconstrained Zoeppritz P-Wave reflection coefficients is developed and tested. The initial model for the pre-stack inversion is based on impedance inversion results and fitted empiric relations. In the feasibility study conducted with data from the North Sea, the inversion scheme successfully predicted S-Wave velocities compared to available Cone Penetration Test ground truthing data, although the sensitivity for low S-Wave velocities is limited. A sensitivity analysis with synthetic test data indicates that this lack of sensitivity is a general limitation of P-Wave reflection data inversion.
In summary, in this thesis it is disclosed how marine near surface P-Wave reflection seismic data have to be acquired and inverted to deduce quantitative information reaching beyond and complementing qualitative standard interpretation results
Characterization of a limonene-degrading methanogenic enrichment culture
In this study, a limonene-degrading methanogenic enrichment culture was analyzed, revealing new insights into the predatory microbial interactions and potential limonene degradation pathways. The enrichment culture, established in 1999 using activated sludge obtained from a wastewater treatment plant, sustains a complex microbial community despite being supplied only with limonene as a carbon source.
Using long-read PacBio sequencing, a metagenome-assembled genome assigned to the
Syntrophobacteraceae family was identified as responsible for the initial step of limonene
degradation under methanogenic conditions. Within the genome of the Syntrophobacteraceae MAG, two operons were identified, with metatranscriptomic and metaproteomic data verifying the transcription of the genes and the presence of the proteins in the metaproteome. The initial activation of limonene is catalyzed by an enzyme annotated as benzylsuccinate synthase, which forms a new phylogenetic branch within the known and characterized fumarate-adding enzymes and was therefore named limonenylsuccinate synthase. Limonenylsuccinate synthase is possibly the first enzyme detected for monoterpene degradation in a methanogenic environment.
With 32 MAGs assembled from the metagenome, the microbial community in the enrichment culture is a complex syntrophic community. Most members of the community can be assigned to syntrophic bacteria, methanogenic archaea, or fermenters, who work together to degrade limonene. However, there is a predator thriving in the culture. Candidatus Velamenicoccus archaeovorus, a member of the candidate phylum Omnitrophota, that was visualized using CARD-FISH in combination with high-resolution microscopy and was found attached to the methanogen Methanothrix soehngenii and other microorganisms within the enrichment culture. Candidatus Velamenicoccus archaeovorus transcribes genes that allow it to attach to its prey, lyse the cells, and consume their biomass, showing similarities to the mechanisms of other predatory bacteria. Furthermore, a group I intron was detected in the predator’s 23S rRNA gene
and could be visualized using CARD-FISH in its prey cells, showing the transfer of mobile
genetic elements between prey and host.
The predation process introduces a new trophic level into the microbial community as it results in the release of necromass. The necromass, consisting of carbohydrates, lipids, and proteins from dead cells, is expected to be fermented by other members of the community, such as Anaerolineaceae and Lentimicrobium, closing the microbial loop and facilitating further syntrophic interactions. The complex interplay between syntrophic bacteria, methanogens, and the predatory bacterium illustrates the intricate dynamics within this anaerobic system. The discovery of the novel limonenylsuccinate synthase enzyme and the predatory role of Candidatus Velamenicoccus archaeovorus highlights the metabolic complexity of microbial biomass turnover in energy-limited ecosystems
Invertible neural networks and normalizing flows for image reconstruction
Datenbasierten Methoden, insbesondere Deep Learning Methoden, wurden in den letzten Jahren erfolgreich zur Lösung von verschiedenen Inversen Problemen angewendet. In der medizinischen Bildgebung, zum Beispiel in der Computertmographie (CT) oder Magnetresonanztomographie (MR), wurden Methoden entwickelt, die zu einer bemerkenswerten Steigerung der Bildqualität führen können. In meiner Promotion habe ich verschiedene Deep Learning Methoden zur Bildrekonstruktion analysiert und entwickelt. Mein Fokus lag in der Analyse von invertierbaren neuronalen Netzen. Invertierbare neuronale Netze bilden die Grundlage für die Implementation von generativen Normalizing Flows. Des Weiteren habe ich an verschiedenen Challenges teilgenommen, um diese neuartigen Deep Learning Methoden auf reale Problemstellungen anzuwenden und zu validieren.
Diese Arbeit besteht aus sechs Veröffentlichungen. In den ersten beiden Arbeiten wird die Anwendung von invertierbaren Netzwerken zur Approximation von a-posteriori Verteilungen im Kontext von verschiedenen medizinischen Bildgebungsproblemen untersucht. Zusätzlich haben wir an der Implementierung von invertierbaren Netzwerken zur Approximation der a-priori Verteilung gearbeitet. In der Helsinki Tomographie Challenge haben wir verschiedenen Ansätze für die CT-Rekonstruktion auf einem realen Datensatz ausgewertet und den zweiten Platz erreichen können. In dem letzten Abschnitt haben wir die Anwendung von Score-basierten Diffusionsmodellen, als Erweiterung der generativen Modelle in den anderen Veröffentlichungen, auf CT-Rekonstruktion und MR-Rekonstruktion untersucht. Insbesondere wurden Methoden für den Fall einer Diskrepanz der Training- und Testverteilung entwickelt. Außerdem haben wir notwendige Anpassungen für die Anwendung dieser Methoden auf Positronen-Emissions-Tomographie vorgenommen
Undaria pinnatifida: on the invasion potential of the commercially important Asian kelp in Europe
Non-native invasive species were the most common driver of species extinctions in the last 500 years and continue impact ecosystems worldwide. Globalization and population growth facilitate their spread and increase native ecosystems' vulnerability.
In light of the Climate Crisis, marine primary producers have gained increasing attention – as a potential solution and an impacted group. A marine primary producer that has gained much attention due to its near ubiquitous worldwide spread as an invasive species is the brown macroalga Undaria pinnatifida. Native to East Asia cultivated intensively, it was introduced to Europe in 1972. By 2009, it had spread until The Netherlands.
In this study, a new, self-sustaining population of U. pinnatifida was discovered Sylt, Germany, marking its to date (2024) northernmost introduced population. Two strikingly different phenotypes were observed, yet genetic analysis and field observations revealed high similarity and exchange between them. They were distinct from other European and native populations with the population in France (Brittany) the most likely source of the German U. pinnatifida.
Experiments demonstrated gametogenesis and better growth under cold conditions of the German gametophyte isolate and resulting sporophytes compared to a native Chinese isolate, indicating a potential cold adaptation. Further studies are required to prove the latter.
While U. pinnatifida is currently limited to primarily artificial substrates in the Wadden Sea, its increasing proximity to the rocky shores of Scandinavia raises questions about the likelihood of an establishment there. This spread could only be enabled by human vectors. Intact native communities have shown resilience to invasion by U. pinnatifida in many regions. Therefore, the best counteraction against the spread of U. pinnatifida, invasive species, and their unpredictable impacts is the protection and preservation of native communities that are under threat from Climate Change
Chancen und Potenziale der Partizipation von Nutzer:innen im Mehrgenerationenhaus MOBILE - eine quantitative Studie
Das Forschungsprojekt befasst sich in einem quantitativen Praxisforschungsprozess mit den Partizipationschancen von Nutzer:innen in einem Mehrgenerationenhaus in Bremen sowie mit dem Interesse der Nutzer:innen an mehr Partizipation vor Ort. Dafür wurde die Partizipationspyramide von Straßburger & Rieger (2019) aus Perspektive der Adressat:innen operationalisiert, um einen standardisierten Fragebogen für die Bearbeitung der Forschungsfragen erstellen zu können. In der Auswertung der erhobenen Daten lag der Fokus auf dem Faktor Alter und Partizipation. Auf den Faktor Geschlecht und Partizipation wurde skizzenhaft eingegangen. Abschließend wurde im Sinne der Handlungswissenschaft Soziale Arbeit Handlungsempfehlungen für die Praxis Sozialer Arbeit formuliert
The future of Workation - An analysis of the long-term potential of Workation based on the assessments of employees, employers and the accommodation sector
Workation ist ein neues Konzept im Rahmen von New Work, mit dem potenziell mobil Arbeitende die berufliche Tätigkeit mit einem Urlaub zeitweise kombinieren können. Aufgrund der jungen Entwicklung stellt sich die Frage nach dem langfristigen Potenzial von Workation und wie dieses von den verschiedenen Akteuren Arbeitnehmer, Arbeitgeber und dem Beherbergungssektor eingeschätzt wird. Durch eine umfassende Literaturrecherche und eine quantitative Befragung von Vertretern des Beherbergungssektors wurden die verschiedenen Perspektiven analysiert. Ziel dieser Arbeit war es insbesondere dem Tourismussektor, stellvertretend dem Beherbergungssektor, einen Mehrwert zu bieten. Die Ergebnisse verdeutlichen, dass langfristiges Potenzial gesehen wird und alle Akteure diverse Vorteile in Workation sehen. Somit kann geschlussfolgert werden, dass sich die Workationer zu einer für den Tourismussektor bedeutenden Zielgruppe entwickeln. Basierend auf diesen Erkenntnissen wurden konkrete Handlungsempfehlungen für den Beherbergungssektor formuliert, um die Zielgruppe optimal ansprechen zu können, wobei nicht jede Unterkunft aufgrund von externen Gegebenheiten wie der Lage oder Internetverbindung gleichermaßen vom Potenzial profitieren kann
Process development of electrode-based dielectrophoretic separators
Die Trennung von Partikeln ist ein Feld mit etablierten und robusten Verfahren. Ziel dieser Arbeit ist es, den Werkzeugkasten der auf Dielektrophorese basierenden Trennapparate um zwei Instrumente zu erweitern. Der erste Ansatz, der in dieser Arbeit vorgestellt wird, nennt sich frequenzmodulierte dielektrophoretische Partikelchromatographie (DPC). Der Ansatz wurde in experimentellen und numerischen Untersuchungen getestet. Dabei konnten die verschiedenen möglichen Trajektorien in einem DPC-Aufbau beschrieben und vorhergesagt werden. Der mikrofluidische Aufbau verfügt über Elektroden am Boden des Kanals, welche eine chromatographische Trennung ermöglichen. Das Verfahren wurde mit einfach detektierbaren, fluoreszierenden Polystyrol-Mikropartikeln getestet, und es konnten eine größenselektive Trennung von u.A. 2 μm und 3 μm Partikeln und eine Trennung anhand der Oberflächenfunktionalisierung von 2 μm Partikeln gezeigt werden. Der zweite Teil der Arbeit befasst sich mit der Erhöhung des Durchsatzes von DEP-Separatoren, welche Elektrodenarrays zur Manipulation von Partikeln verwenden. Hierbei wurde der Fokus auf die Beibehaltung der Selektivität gelegt. Es wurde ein Design auf Basis von speziell gefertigten, jedoch günstigen Leiterplatten (PCBs) entwickelt und zunächst mit Polystyrolpartikeln getestet. Die selektive Trennung einer Mischung aus 3 μm und 6 μm wurde bei einer Frequenz erreicht, die eine Trennung auf der Grundlage der unterschiedlichen Polarisierbarkeit der Partikel ermöglichte. Schlussendlich wurde der Aufbau mit leitfähigen Partikeln getestet. Gängige Elektrodenmaterialien von Lithium-Ionen-Batterien wurden ausgewählt, um die Trennbarkeit technisch relevanter Partikel zu zeigen. Die Experimente lassen schlussfolgern, dass Graphit bei geeigneten Parametern nahezu vollständig gefangen wird, wohingegen Lithiumeisenphosphat zum Großteil den Kanal wieder verlässt
Ansätze zur Vorhersage des Ermüdungsversagens von Klebverbindungen unter betriebsrelevanten Bedingungen
Adhesive bonding technology plays a major role in enabling multi-material engineering structures with lightweight potential. However, there is still a lack of knowledge about how operational conditions can affect the failure of adhesive joints under fatigue loading. Moreover, cost-intensive fatigue experiments are a limiting factor in addressing multiple operational conditions simultaneously.
The goal of the present work is to investigate approaches to predict the fatigue failure of bonded joints under operationally relevant conditions. The focus is to develop an understanding of the fatigue behavior of elastic and rigid bonding adhesives, whilst developing methods to reduce the number of experiments for the characterization of adhesives. In this work, the effect of multiaxial loading (considering non and proportional loading), varying mean stress, and stress concentrations are highlighted as operational-relevant conditions
Einzelfallorientierte Forschung in Logopädie/Sprachtherapie. Ein Tagungsbericht
Die Einzelfallforschung hat eine lange Tradition und Bedeutung für Disziplinen, die sich durch ein einzelfallorientiertes Vorgehen auszeichnen. Für die Logopädie/Sprachtherapie kann sowohl die Forschung, welche Evidenz von Maßnahmen darstellen soll, als auch die evidenzbasierten Praxis von diesem Forschungsdesign profitieren. Das Werk liefert einen Einblick in die aktuelle Diskussion der einzelfallorientierten Forschung in der Logopädie/Sprachtherapie und basiert auf den Erkenntnissen eines gleichnamigen Symposiums im September 2022. Es werden im ersten Teil zunächst einzelfallorientierte Forschungsprojekte vorgestellt, die beispielhaft das methodische Vorgehen abbilden. Danach liefern im zweiten Teil einschlägig versierte Fachleute Beiträge zu methodologischen Aspekten der Einzelfallforschung. Dadurch werden grundsätzliche erkenntnistheoretische und paradigmatische Diskussionen angeregt. Im dritten Teil werden diese Grundsatzdiskussionen durch die inhaltsanalytische Zusammenfassung der Kleingruppengespräche, die während des Symposiums zwischen allen Teilnehmenden stattfanden, dargestellt.VW Stiftun
