Isolation, structure elucidation and testing of secondary metabolites from fungi of the genus Lactarius and Mycena

    No full text
This thesis is about the isolation and structure elucidation of so far uninvestigated secondary metabolites from fruiting bodies of the mushrooms Lactarius circellatus, Lactarius trivialis and Mycena zephirus. In the Spiteller group L. circellatus has already been investigated before for its content of sequiterpenoids and five unknown and several known sequiterpenoids have been identified and their structures have been elucidated. However, their stereo configuration, the proton and carbon resonances in their NMR spectra have not been fully assigned so far. Therefore, NMR spectra of the previously isolated sequiterpenes were recorded again. Moreover, fruiting bodies of L. circellatus were extracted again and investigated for the presence of new compounds, resulting in the isolation of the so far unknown sequiterpenoid 12-hydroxy-15-carboxyblennin A (19) together with hydroxyblennin A (34). A careful analysis of the coupling constants in the 1H NMR, the analysis of the NOE correlations in the NOESY spectra, and a detailed analysis of the correlations in the HMBC spectra in respect to their dihedral angle according to the Karplus relationship allowed to determine the relative configuration of all isolated sequiterpenoids, including the so far unpublished new sequiterpenoids 12-hydroxy-15-carboxyblennin A (19), 7,15-dihydroxyblennin A (20), 2,3-epoxylactarorufin A (21),14-carboxy-deoxylactarorufin A (22), and 13-hydroxy-14-carboxy-deoxylactarorufin A (23). To determine the absolute configuration of the isolated sequiterpenoids, CD spectra were recorded and compared with the corresponding calculated CD spectra. By these means the absolute stereochemistry of the sequiterpenoids 19 to 23 could be determined. However, the measured and calculated CD spectra of 22 and 23 were not matching perfectly. So far, there have been no reports about pigments from L. circellatus, despite the fact that it exhibits on the surface of its cap a grayish green color similarly to L. blennius. Consequently, the pigment was isolated and its structure elucidated to be blennione (35), the same pigment that is present in L. blennius. Blennione (35) was found to be much more abundant in L. circellatus than in L. blennius, the mushroom it was originally isolated from. Some biological tests were performed but blennione (35) displayed no activity. In addition, tiny amount of a pink pigment from L. blennius was found related to lilacinone (37), a pink pigment that was isolated and structurally elucidated before from L. lilacinus. Fresh fruiting bodies of Lactarius trivialis display an intensive violet color on the cap surface that is fading away upon ageing of the fruiting bodies. The responsibe pigments have not been investigated so far. Therefore, fruiting bodies of L. trivialis were subjected to a comprehensive investigation of their secondary metabolites with a focus on their pigments. Not entirely unexpectedly, the isolation of the pigments from the skin cap turned out to be difficult due to the low stability of the pigments, especially when methanol was used as a solvent. However, an extraction with acetone with subesquent column chromatography on Sephadex LH20 with acetone as a solvent followed by purification of the pigments with semi-preparative HPLC resulted in the isolation of five colored compounds, that were named trivialines A (47), B, C, D, and E. Due to their HR-ESI-MS data, the compounds five pigments are closely related to each other and show a high degree of unsaturation. Only in the case of trivialine A (47) and trivialine B the isolated amount was sufficient to record a full set of one and two dimensional NMR spectra. Consequently, the structure elucidation focussed on these two compounds, both exhibiting a molecular formula of C32H20N4O6. Due to the high degree of unsaturation, the low number of unexchangable protons, leading to a lack of corellations in the HMBC, the structure elucidation turned out to be very challenging. Nevertheless, for trivialine A (47) a tentative structure was proposed. Moreover, fruiting bodies of L. trivialis were investigated for the presence of new sesquiterpenoids. Thus, the new compound 14-hydroxylactarorufin B (53) was isolated and its structure elucidated. In addition, the lactarorufin B (27) and 14-hydroxyblennin A (52), both known from other mushrooms, were also identified to be present in L. trivialis. So far, 14-hydroxyblennin A (52) was reported only from Russula sanguinea, but the published NMR data fit perfectly to 15-hydroxyblennin A (34), while the NMR data from the corresponding sesquiterpene from L. trivialis fit very well to the structure of 14-hydroxyblennin A (52). Consequently, the structure in the above mentioned manuscript about R. sanguinea has to be revised to 15-hydroxyblennin A (34). Apart from mushrooms of the genus Lactarius, in this thesis also fruiting bodies of Mycena zephirus were investigated in respect to their secondary metabolite content and the structures of two compounds were elucidated to be N5-isopentylglutamine (54) and N5-2-methylbutylglutamine (55), respectively

Designing for Diverse Needs: Technology-supported Well-being and Empowerment of Children and their Care Ecosystem

    No full text
In our increasingly digitised society, children interact with technologies from a young age, leading to both benefits, e.g. learning opportunities, and risks, e.g. increased anxiety. Many of these technologies are not designed for children, necessitating an investigation into their impact on children's well-being and their design. Power dynamics between adults and children are critical, particularly for neurodivergent children like those with ADHD, whose interactions with technology are influenced by their diagnoses and adults' perceptions. This thesis advocates for creating technologies that empower both neurotypical and neurodivergent children, incorporating their perspectives to shape the technologies they use and enhancing their agency to support their well-being. Contributing to the evolving narrative in HCI of designing meaningful interactions, it examines how to design for both the well-being and empowerment of children and their care ecosystem, considering diverse interests and needs, focusing particularly on children with ADHD, the most prevalent neurodevelopmental diagnosis in children. This work designs a range of technologies and investigates the interactions of children and their care ecosystems with them. The primary research question guiding this thesis is: How can technologies be designed to empower and support the well-being of both ADHD and neurotypical children and their care ecosystems? Overall, this thesis enhances understanding of strategies and aspects technologies can use to empower and support the well-being of ADHD and neurotypical children and their care ecosystems. It presents the design and evaluation of prototypes using these strategies and proposes a new agenda for ADHD technology research that designs "beyond symptoms," advocating for the active inclusion of multiple care ecosystem stakeholders. It also offers a refined understanding of technology-supported emotional expression and reflection processes to foster children's well-being

An engagement-aware recommender system for people with dementia

    No full text
With the number of people with dementia (PWD) growing worldwide and no cure available, there is an urgent need for tertiary prevention strategies to reduce symptoms, slow disease progression, and improve patients' quality of life. Technology-driven approaches, such as tablet-based activation systems, are well-suited for this purpose as they are portable and can offer diverse contents to meet different capabilities, needs, and interests. However, for these systems to benefit cognitive functioning, well-being, and quality of life, they must engage users effectively, which varies greatly among individuals. Thus, tablet-based systems should be able to monitor and interpret users’ engagement levels to personalize the sessions. This thesis examines a tablet-based system called I-CARE which aims to promote cognitive, social, and physical activation for PWD. It analyzes interactions between PWD and their caregivers (CGs) using a multimodal dataset gathered in a semi-supervised care setting, with engagement levels annotated by human raters for detailed analysis. The dissertation explores the recognition of PWD's affective states, including engagement and activation preferences, based on their verbal and non-verbal responses. Moreover, recognized engagement levels are used as feedback for a recommender system designed to support PWD and CGs in selecting appropriate contents, enabling beneficial activation sessions. A study conducted in a real-world setting evaluated the system's effectiveness and identified challenges in dynamic scenarios. Last, this thesis investigates the automatic detection of spontaneous emotional tears, a key social signal, based on facial and postural behaviors in healthy individuals, which could enhance perceptive abilities across various applications

Applications of cavity ring-down Sspectroscopy in environmental physics: Rayleigh scattering cross-sections and airborne measurements of peroxy radicals

    No full text
This doctoral thesis aims at the improvement of the airborne measurement of peroxy radicals by focusing on two applications of cavity ring-down spectroscopy (CRDS): the determination of the EM radiation extinction cross-section of gas molecules, and the determination of gas molecule concentrations with the knowledge of the EM radiation extinction and the EM radiation extinction cross-sections. The experimental part of the work is based on the use of the Peroxy Radical Chemical Enhancement and Absorption Spectrometer (PeRCEAS) instrument which combines the peroxy radical chemical amplification (PERCA), for the amplified conversion of peroxy radicals in NO2, with a sensitive NO₂ detection by CRDS. For the first CRDS application, three setups were used for the determination of Rayleigh scattering cross-sections σ_(Rayl.)of atmospheric gases at 408 nm. Thus, the EM radiation extinction by N₂, O₂, Ar, CO, CO₂, N₂O, CH₄, and synthetic air (SA) was measured using step pressure changes and pressure ramps in optical cavities. The experimentally determined σ_(Rayl.) were compared with calculated values based on literature refractive index (n) and King correction factors. The calculated and measured σ_(Rayl.) agreed within 0.6 %, 2.4 %, 1.2 %, 2.2 %, and 1.5 % for CO₂, N₂, O₂, SA, and Ar, respectively, at ~ 408 nm. The measured σ_(Rayl.) for N₂O and CH₄ confirmed the improved accuracy of the most recently published determinations of their refractive indexes. The CO σ_(Rayl.) experimentally determined for the first time in this work at 408 nm, is 4.1 % higher than the calculated value resulting from extrapolation, suggesting the need for improved knowledge of the refractive index and King correction factors in the blue spectral region. The second CRDS application is a pre-requisite for the acquisition of airborne peroxy radical data and involves the deployment of PeRCEAS on an airborne platform for the measurement of the total sum of peroxy radicals which react with NO (RO_2^*) during the EMeRGe (Effect of Megacities on the transport and transformation of pollutants on the Regional and Global scales) project. The retrieved RO_2^* were then compared with the results of four atmospheric models and with calculations based on photostationary steady state (PSS) assumptions. The box model and the PSS calculations are constrained to the airborne measurements. Three case studies/scenarios were investigated: 1) close to Rome and Manila's major population centres, 2) long-range transported air masses from megacities in the Western Pacific, and 3) over a very populated area in Taiwan during different take-off and landing scenarios. Generally, models agree with measurements within their uncertainties, but they face difficulties with short-term variations. Notably, in Rome, models underestimated measurements by up to 80% at lower altitudes close to the boundary layer, while in Manila the box model underestimated 30% of the observations. In the Taiwan cases, the models showcased a 2 to 5 times underestimation in capturing short-term RO_2^* variability in more polluted, aerosol-rich conditions. A re-evaluation method, adjusting the ratio of different peroxy radicals in the RO_2^* calculations, reduced differences between the box model and PSS results by up to 40%. Overestimations by the PSS calculations were linked to high NO levels, organic nitrate formation, and aerosol-induced radical losses unaccounted for in current calculations. The findings underscore the capabilities of the models investigated and highlight the necessity for the speciation of radical measurements to improve the understanding of the short-term variability of RO_2^* in complex polluted areas

A myriad of processes and systems in Chemical- and other fields of Engineering Science are multiphase. Applications that involve processes containing multiple phases of increasing geometric complexity have become increasingly common in recent years. Attempts to model and simulate systems with a large number of interfaces with complex geometries is often limited by the interface tracking and accuracy of the curvature computation which is a large source of error of the numerical method. This work resolves the long standing difficulty in computing the curvature of interfaces accurately and efficiently. In this work a new practical tool was developed that can be implemented in a wide array of state-of-the art methods. It is shown that the rigorous derivation of the curvature of interfaces allows for an accurate, or near-exact computation of curvature for simulation of three-phase systems. Additionally, it is demonstrated that a special trapezoidal integration error can be used to provide an estimate of continuity of the interface, which is useful for ensuring dynamic accuracy in three-phase simulations by controlling the refinement of a numerically captured fluid-fluid interface that is manifold. It is further shown that the method for computing curvatures developed herein can be computed exactly for arbitrary fluid-fluid interfaces that are manifold. The extent to which an interface is manifold can be tracked by computing the topology of the simplicial complex using computationally efficient methods developed especially for this application. Since the method for computing curvatures are near-exact and mesh independent, a very large tolerance can be used in large scale simulation of complex three-phase materials. In principle, it is only necessary to track that the interface is continuous. Therefore, the complex can be refined in such a way that the error matches the desired tracking of non-solid volume due to porosity and asperities in three-phase systems. The result of this work can be implemented in any formulation where the underlying geometry of the model is manifold, and is especially useful in the fields of multiphase flow, thermodynamics, materials engineering and surface energy minimisation

How adhesives should flow during manufacturing

    No full text
This thesis proposes a methodology to optimize the application pattern of adhesively bonded joints, so that the liquid adhesive automatically flows into the desired bond shape during manufacturing — leaving no spots over- or underfilled, thus the adhesive is used in the most efficient way. Besides preventing adhesive excess and waste that eliminates the need for cleaning and reworking steps after joining, this optimization approach ultimately leads to a more ecological and economic adhesive manufacturing processes. To achieve this goal, an optimization algorithm is developed that iteratively modifies the shape of adhesive application pattern until it flows into the desired bond shape. Established methods from topology optimisation for solids are used, re-interpreted, and applied in the context of fluid mechanics in a new way. At each iteration, the resulting distribution obtained after squeezing an initial adhesive application pattern is calculated. The resulting distribution is then compared to the desired adhesive distribution, and overflown adhesive is traced back to its origin in the initial pattern (using the calculated velocity and direction). These overflown areas are then removed from the initial pattern and the iteration loop continues until the resulting distribution matches the desired bond geometry within an acceptable tolerance. This optimization strategy relies on the accurate simulation of squeeze flow processes for which a special numerical model was chosen. This model was extensively validated in a comprehensive experimental study in advance, to prove its suitability. By combining the Hele-Shaw approach, a method to investigate squeeze flow processes, with Particle Image Velocimetry, a method to measure fluid flows, it is demonstrated that the simulation model is sufficiently accurate and can be implemented to calculate the quantities that are required by the optimization algorithm. Further, the surface structure has proven to be of secondary importance and doesn't need to be considered in the optimization. The algorithm was applied to optimize a variety of different application patterns. It was found that the optimized shapes have recurring shapes such as pointy fingers which flow into corners and bays that flow into straight edges. Furthermore, quantities like the degree of compression, number of cells and considered flow-laws were varied to investigate their influence on the algorithms results. However, it was found that the algorithm is mesh and flow-law independent, which highly improves its applicability and simplicity for practical use. It is capable to identify optimal patterns for frequently used adhesive layer thicknesses and degrees of compression. Finally, the optimized patterns obtained by theoretical analysis were validated through real world experiments, that confirmed their ability to flow into the desired shapes. Overall, this thesis presents an innovative approach to optimize adhesive application patterns. Due to the performed optimization, the environmentally questionable adhesive waste and costly post-cleaning work can be eliminated — thus, a more ecological and economical manufacturing process is achieved
