Benfotiamine in high carbohydrate diet enhances growth and reproductive performance of the Asian native catfish Clarias macrocephalus subadults
Clarias macrocephalus has been extirpated in most regions in the Philippines and as a preliminary study towards its reintroduction, we aimed at enhancing the growth and reproductive performance of the broodstock. Several studies have identified thiamine deficiency as a factor in the extirpation of some important fish species in the wild. The present study aimed to determine the effects of elevated dietary carbohydrates and benfotiamine, a more bioavailable analog of thiamine (vitamin B1). Three diets were used, namely, a commercial diet specific for catfish was used as the control diet (Com), a high carbohydrate (HC) diet, and a benfotiamine-supplemented HC diet (HCB). Results show that survival of the subadult catfish in captivity for 10 weeks was remarkably high, exhibiting an average of 99% for all three treatments (P>0.05). HCB diet resulted in significantly higher final average body weight (FABW), weight gain (WG), specific growth rate (SGR), and significantly best food conversion ratio (FCR) (P0.05). For the reproductive performance, female Asian catfish fed the HCB diet exhibited significantly heavier ovary and liver (P0.05). GSI of the female catfish and fecundity were significantly higher in the HCB group (P0.05). In the male catfish, testes were significantly heavier in the HCB group (P0.05). HSI, male LW, and LL were significantly higher in the HC and HCB groups (P>0.05) than in the Com group (P0.05). In conclusion, elevating dietary carbohydrates in the diet from 7.70% to 22.73% (i.e., Com to HC diet, respectively) did not negatively affect the growth and reproductive performance of the Asian catfish. Further supplementation of benfotiamine at 0.02% to the HC diet enhanced significantly both the growth and reproductive performance of the Asian catfish
Transcriptome Analysis of Metapenaeus affinis Reveals Genes Involved in Gonadal Development
Metapenaeus affinis is a crustacean with important commercial value in the fishery of the South China Sea. Overfishing has resulted in the decline of the wild population and germplasm degradation. However, there is little background knowledge about its gonadal development, and there is a lack of research on the development of this species. To better understand the molecular regulatory mechanisms during gonadal development, here, we performed RNA-Seq on immature and mature ovaries and compared their transcriptomic signatures. 126,930,488 and 122,677,356 clean sequencing reads were obtained from the Illumina sequencing platform, respectively. 394 differentially expressed genes (DEGs) were identified, of which 136 were up-regulated, and 258 were down-regulated. Further analysis revealed rich transcriptional sequences, which have homology with genes related to reproduction and development. Expression patterns of COX, GPX, E3s, PCNA, STPK, and other genes were changed during ovarian development. Validation by qRT-PCR demonstrated the reliability of RNA-Seq. This study has made a significant contribution to the currently available sequence data of M. affinis and provided reference data for the development of genetic and breeding work
Effects of salinity and alkalinity on growth and survival of all-male giant freshwater prawn (Macrobrachium rosenbergii De Man, 1879) juveniles
All-male giant freshwater prawns (AMGFPs) have been a popular crop cultivated in the Mekong Delta, Vietnam, due to their proven production efficiency compared to all-female or mixed-sex prawn cultures. However, the crucial water quality factors impacting AMGFP aquaculture efficiency have yet to be elaborately investigated. Two separate experiments were randomly arranged with three replicates to evaluate the effects of salinity or alkalinity on the growth and survival of AMGFP juveniles during the grow-out period. The results show that the prawn survival rate in the salinity range of 0–15‰ varied from 66.1 to 74.8% and in a salinity range of 0–5‰ was relatively low compared to the range of 10-15‰; however, the difference was not significant among salinities after 90 days of culture (p > 0.05). All the prawn growth performance parameters significantly decreased with increasing salinities of 0, 5, 10, and 15‰ after 30, 60, and 90 days of culture (p 0.05), and both were significantly higher than those at salinities of 10 and 15‰ (p < 0.05) after 90 days of culture. In addition, the survival rate reached 82.5–84.4% and did not significantly differ among alkalinities of 80, 100, 120, 140, and 160 mgCaCO3 L−1. However, the growth performance parameters and yield of AMGFPs at an alkalinity of 160 mg L−1 were significantly higher than those at lower alkalinities (80, 100, 120, and 140 mg CaCO3 L−1) after 90 days of culture. Therefore, it is recommended that a salinity range of 0–5‰ and alkalinity of 160 mgCaCO3 L−1 is optimal for the growth-out culture of AMGFP juveniles
