Multicomponent Strategies for GPCR Targeting: Allosteric Modulation of Dopamine Receptor D2 and Development of Potent and Selective CB2R Agonists
This PhD thesis focuses on the development of novel, potent, and selective ligands for two therapeutically relevant GPCR targets: the dopamine receptor D2 and the cannabinoid receptor CB2. Different isocyanide-based multicomponent strategies were designed to generate focused small molecule collections that enabled the interrogation of the selected targets, the identification of new lead compounds and the evaluation of the most salient features of the SAR within these series. The first chapter deals with the optimization of novel Melanostatin-inspired allosteric modulators of the D2R. Chapter two documents a systematic MCR-assisted pharmacomodulation process that enabled the dissociation of the promiscuous cannabinomimetic profile of model designer drugs to identify potent, selective, and biased CB2R agonists. The research encompasses molecular design strategies, synthesis, biological evaluation, and structure-activity relationships. Preliminary evidence supporting the attractive antiproliferative activity of selected CB2R agonists in glioblastoma cell lines is also provided.2025-05-1
Mapping light pollution with integrated indicators derived from all-sky hemispheric radiance data
It has been calculated in up to one million directions in the sky
the radiance produced by a single source of light in the 0.12 km
- 527 km range and for different light source and observing site
altitudes.
Using these data, light pollution indicators’ discrete Point
Spread Functions (PSF) were computed: radiance averaged
over all the hemisphere, at 30° altitude, in the first 10° above
the horizon, and horizontal irradiance. From the discrete PSFs,
analytical PSFs were retrieved, permitting to obtain the
indicators values in a much faster way compared to the
traditional path, with saving times of the order of 10⁴-10⁶. As
applications, it has been evaluated the light pollution at all the
world’s major professional astronomical observatories and
maps of Iberian peninsula for all the studied indicators were
produced
Beyond phase-in: assessing impacts on disinformation of the EU Digital Services Act
This work proposes a comprehensive research agenda to empirically evaluate the real-world impacts of the European Union’s
Digital Services Act (DSA) on combating online disinformation. It provides background on the DSA’s context, mechanisms,
timeline, and expected efects on platforms to situate the need for rigorous impact assessment. A detailed legal, technical,
psychological, behavioral and ethical critique reveals meaningful gaps in the DSA requiring ongoing regulatory refnement
and oversight. Most critically, the paper puts forth an encompassing framework spanning computational analytics, interviews,
ethnography, surveys, discourse analysis and mixed methods to rigorously assess the DSA’s multi-dimensional efects on
complex factors enabling disinformation proliferation. Priorities include evaluating notice-and-takedown efcacy, advertising
transparency improvements, risk assessment outcomes, oversight integration, and procedural shifts in platform governance.
Coordinated eforts between researchers, regulators and platforms are needed to address methodological challenges around
isolating DSA impacts amidst an evolving EU regulatory landscape, constrained data access from platforms, and difculties
generalizing fndings across the sociotechnical diversity of platforms and national contexts in EU Member States.Contribution from the ITN project NL4XAI and HYBRIDS which has received funding from the European Union’s Horizon 2020 research and innovation programme under the Marie Skłodowska-Curie grant agreement No. 860621 and 101073351.

Enriching interactive explanations with fuzzy temporal constraint networks
Humans often use expressions with vague terms which play a fundamental role for effective communication. These expressions are successfully modeled with fuzzy technology, but they are not usually integrated yet with Natural Language Processing models and techniques. Large-scale pre-trained language models yield excellent results in many language tasks, but they have some drawbacks such as their lack of transparency and thorough temporal reasoning capabilities. Therefore, the use of such models may provoke inconsistent or incorrect dialogues in the context of conversational agents which were aimed at providing users of intelligent systems with interactive explanations.
In this paper, we propose a model for fuzzy temporal reasoning to overcome some inconsistencies detected in pre-trained language models in a specific application domain of a conversational agent carefully designed for providing users with explanations which are endowed with a good balance between naturalness and fidelity. More precisely, starting from a knowledge graph that provides an intuitive representation of the entities and relations in the application domain, we describe how to map the temporal information onto a fuzzy temporal constraint network. This formalism allows to represent imprecise temporal information and provides mechanisms for checking consistency in conversations.
In addition, as a proof of concept, we have developed TimeVersa, a conversational agent which integrates the proposed model into an application domain (i.e., a virtual assistant for tourists) that requires handling imprecise temporal constraints. We illustrate in a use case how the agent can identify temporal inconsistencies and answer queries related to temporal information properly. Results after a user study report that users' perception of consistency is significantly higher in a conversation with TimeVersa than in a similar conversation using the well-known GPT-3 Large Language Model, when vague temporal information is involved. The proposed approach is a step forward for developing conversational agents operating in application domains that require temporal reasoning under uncertainty.Mariña Canabal-Juanatey is a PhD Researcher supported by the Galician Ministry of Culture, Education, Professional Training and University (ED481A 2022/212). Drying kinetics modeling of hot air drying of emulsion templated oleogels employing hydroxypropyl methylcellulose as structuring agent
This work focused on the drying of O/W emulsions employed to obtain templated structured oils with hydroxypropyl methylcellulose (HPMC) as oleogelator. Sunflower oil/water emulsions with different HPMC content (1%–3% w/w) were dried at different air temperatures (from 70 to 100 °C) employing two different initial thicknesses (0.25 and 0.50 cm). Drying kinetics showed the existence of an initial constant drying rate period followed by a falling drying rate period below critical moisture content. This critical content was constant (0.38 kg water/kg dry solid) independently of drying conditions and HPMC content. Drying rate decreased linearly with moisture content during the falling rate periods. A 3-parameter model was proposed to simulate drying kinetics in the range of drying temperature and HPMC content of tested oleogels. Physical properties of fresh and stored (20 days) oleogels such as oil binding capacity, color, and hardness were measured to evaluate drying conditions effect on product quality. Temperatures above 70 °C were necessary to promote the HPMC gelation. Best characteristics of oleogels in terms of texture, color and oil binding capacity were obtained with HPMC content of 2% w/w, 0.25 cm of emulsion thickness and 80 °C of drying temperature. These results are useful for the design of large-scale dryers for this new kind of productsThis work was funded by MCIN/AEI/10.13039/501100011033 and, as appropriate, by the “European Union NextGenerationEU/PRTR (grant CNS2022-135217)S
