    No full text
This study aims to present a simple and effective carrier matrix to immobilize laccase as opposed to complex and tedious immobilization processes and also to use it in the removal of textile dyes. For this purpose, Cobalt (Co) and Copper (Cu) based metal-organic frameworks (MOFs) were prepared and laccase was immobilized on two different MOFs via encapsulation. The characterization outcomes showed that laccase was well immobilized into MOF supports. Optimum pH and temperature were found for Lac/Co-MOF (pH 4.5 at 50 degrees C) and Lac/Cu-MOF (pH 5.0 at 50 degrees C). The Km (0.03 mM) and Vmax (97.4 mu mol/min) values of Lac/Cu-MOF were lower than those of Lac/Co-MOF (Km = 0.13 mM, Vmax = 230.7 mu mol/min). The immobilized laccases showed good reusability as well as improved resistance to temperature denaturation and high storage stability. For instance, the Lac/Co-MOF and Lac/Cu-MOF retained more than 58% activity after 4 weeks of storage at room temperature. Meanwhile, Lac/Co-MOF and Lac/Cu-MOF maintained 56.5% and 55.8% of their initial activity, respectively, after 12 reuse cycles. Moreover, thermal deactivation kinetic studies of immobilized laccases displayed lower k value, higher t1/2, and enhancement of thermodynamic parameters, which means better thermostability. Finally, the decolorization activities for the Lac/Co-MOF were 78% and 61% at the 5th cycle for Reactive Blue 171 an

Effects of curcumin on arginase enzyme activity, ornithine and nitric oxide levels in experimental breast cancer model

    No full text
Breast cancer accounts for almost 30% of all cancer types, making it the most common type of tumor among women in the world. Arginase, an essential enzyme of the urea cycle, leads to the formation of urea and ornithine from L-arginine using the same substrate as nitric oxide synthase (NOS). Arginase has been reported to be higher in cancer patients and can be used as a useful biomarker. In this study, we aimed to investigate the effects of curcumin, an anticarcinogenic agent, on arginase enzyme activity, ornithine, and nitric oxide (NO) levels in experimental breast cancer model in mice. 43 male Balb/c mice were used, and Erhlich acid tumor model was created in the study. Mice were divided into five groups as healthy control group, curcumin treatment before tumor formation, curcumin treatment after tumor formation and cancer control groups. 100 mg/kg curcumin were given orally. Serum and tissue arginase enzyme activities, NO levels and tissue ornithine levels were determined spectrophotometrically. Increased serum arginase activity decreased with curcumin treatment, but this difference was not statistically significant. On the other hand, decreased NO levels were increased with curcumin treatment. In tumor tissue, arginase activity and ornithine levels decreased significantly with curcumin treatment and tissue NO levels increased significantly with curcumin treatment. In our study, we show that curcumin may have a protective effect on the development of breast cancer by inhibiting arginase enzyme activity and ornithine levels, which are the precursors of polyamines, as well as inducing NO production via NOS. As a promising anticancer agent, the net effects of curcumin in this mechanism should be supported by more advanced studies and new parameters. © 2022 Ondokuz Mayis Universitesi. Beta-glucan effects on 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) toxicity in liver and brain

    No full text
2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is a common environmental contaminant that is toxic to brain, heart, kidney and liver. TCDD toxicity is due to free radical formation. Beta-glucan is an antioxidant that exhibits beneficial effects on health. We investigated the effects of beta-glucan on brain and liver tissues of rats with TCDD induced toxicity. We used female rats divided into four groups: control, TCDD group treated with TCDD 2 mu g/kg/week, beta-glucan group treated with 50 mg/kg/day beta-glucan for 3 weeks, TCDD + beta-glucan group treated with 2 mu g/kg/week TCDD and 50 mg/kg/day beta-glucan together for 3 weeks. We found that the thiobarbituric acid reactive substance (TBARS) levels were increased significantly in the TCDD group compared to the other groups. Glutathione (GSH) levels, and superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities were reduced in the TCDD group compared to the control group. SOD, CAT, GPx activities and GSH levels were increased in the TCDD + beta-glucan group. Histopathological observations were consistent with our biochemical findings. The oxidative stress and histopathology caused by TCDD were ameliorated by beta-glucan treatment. Beta-glucan should be explored for preventing brain and liver damage caused by TCDD toxicity

Economic history has witnessed many approaches that try to explain the reasons for development differences between countries. At the beginning of these approaches are the differences in physical capital and technological progress put forward by R. Solow in the 1950s. But these two variables fell short of explaining why some countries are richer and some poorer. At this point, it is necessary to increase the physical capital, create the technological infrastructure, increase the production capacity, etc. The issue that events may be related to the functioning of institutions has come to the fore. There is a large literature on the subject that institutions are the structures that regulate social and economic life and that they are the basis of sustainable economic growth. In order to find an answer to the question of how institutions and institutional quality affect the economic performance of countries, Brazil, Russia, India, China, South Africa and Turkey (BRICS-T) countries with identical economic structures were examined in the study. Panel Data Analysis methods were used in the study, in which annual data for the 2002-2020 period were used. The institutional quality indicators in the study, taken from the World Governance Indicators, are government effectiveness (GE), political stability and non-violence (PS), rule of law (RL), regulatory quality (RQ), freedom of expression and accountability (VA) and corruption control (CC). In the study, according to the PVECM (Long Term) causality test results, a causal relationship was found from institutional quality factors to economic growth in the long run. According to Emirmahmutoğlu and Köse (2011) panel causality test results, there is a causality relationship from a different institutional quality factor to growth in each country. In general, we found that there is causality running from political stability and non-violence (PS), freedom of expression and accountability (VA) and regulatory quality (RQ) to growth. Finally, institutional quality factors that positively affect the economic growth of countries were found to be regulatory quality (RQ), corruption control (CC) and rule of law (RL)

Historical city centers are important places for cities in terms of their cultural, historical, economic, and social characteristics. Due to the rapid economic growth and excessive population increase experienced in the recent period, most cities have faced an uncontrolled growth. Because of the related pressures, historical city centers have been adversely affected in terms of their physical, functional, economic, and social structure. This study aims to develop spatial strategies to contribute to the solution of the problem mentioned above. It focuses on the spatial configuration of the study area and utilizes the space syntax method and parameters, as well as a comparative analysis of some key indicators. The study presents the results of the spatial strategy developed through the identification of points of special interest (SA), strategic priority areas (SPA), and urban sub-foci (USP). As a result of the analysis and the suggestions developed, it has been revealed that there is a significant increase in the socio-spatial characteristics of both the city area and the historical city center.Cukurova University [FDK-2016-7736]The study was funded by Cukurova University Research Projects Unit.Project No: FDK-2016-773
