    Recently, the Constitutional Court issued a ruling confirming the constitutionality of two court resolutions that had ordered the vaccination of a person with disabilities against the will of their legal guardian. The purpose of this article is to analyze what would have happened if these events had occurred today, after the entry into force of Law 8/2021, which regulates a new system of support for legal capacity in favor of people with disabilities. After analyzing the new support measures that have been incorporated into the Civil Code, it is concluded that there is a serious mismatch between the Civil Code and Law 41/2002Recientemente, el Tribunal Constitucional ha dictado una sentencia por la que se confirmaba la constitucionalidad de dos resoluciones judiciales que habían ordenado la vacunación de una persona con discapacidad en contra de la voluntad de su tutor. El objeto de este artículo es analizar qué hubiera ocurrido de haberse producido esos hechos en la actualidad, tras la entrada en vigor de la Ley 8/2021 que regula un nuevo sistema de apoyo a la capacidad jurídica en favor de las personas con discapacidad.     Association of a haplotype in the NRG1 gene with schizophrenia: a case-control study

    Objectives: Schizophrenia (SZ) is a severe multifactorial disease. NRG1 is a gene acting in the development of SZ. A number of NRG1 single nucleotide polymorphisms (SNPs) and their haplotypes are associated with SZ. In the present study, we investigated the association of a NRG1 haplotype (G-C in rs6988339-rs3757930 frame) which was reported to be associated with SZ, and two other SNPs in the same gene (rs74942016, rs80127039) whose rare missense alleles were found in SZ patients. Also, we analyzed disease associations of potential new haplotypes constructed by the variants of these SNPs.Methods: We genotyped 4 SNPs in a sample consisting of 302 SZ patients and 333 controls from a local Turkish population. We tested the disease associations of these variants at single SNP, haplotype and diplotype levels in case-control design.Results: At single SNP level, the CC genotype of rs3757930 was associated with SZ (p=0.038). The previously reported association of G-C haplotype in rs6988339-rs3757930 frame was absent (p=0.416), but we found another haplotype (C-G in rs3757930-rs74942016, p=0.018) and three diplotypes (A-C/G-C diplotype of rs6988339-rs3757930 frame, C-G/C-G diplotype of rs3757930-rs74942016 frame, and A-C-G/G-C-G diplotype of rs6988339-rs3757930-rs74942016 frame) associated with schizophrenia in our sample.Conclusions: Our study indicated the associations of a SNP, a haplotype, and a diplotype of NRG1 with schizophrenia and supported the involvement of NRG1 gene in the development of the disease.     Impacts of different triathlon races on systemic cytokine profile and metabolic parameters in healthy individuals: a systematic review

    The present systematic review aimed to discuss the impacts of different triathlon protocols on the level of pro and anti-inflammatory cytokines, as well as biomarkers related to the performance of healthy individuals. Four databases [PubMed (28 articles), Scopus (24 articles), Science Direct (200 articles), and SPORT Discus (1101 articles) were assessed. The eligibility criteria were applied, and the selected articles were used in the peer review, independently, as they were identified by March 2022. Of the 1359 articles found, 10 were included in this systematic review. Despite the difference in triathlon protocols, it was observed an increase in pro and anti-inflammatory cytokines including IL-4 and IL-10, and chemokines, such as IL-8 and MCP-1. Moreover, the anti-inflammatory serum levels increase after triathlon. Overall, the studies also reported enhancement in the serum levels of cortisol, creatine kinase, C reactive protein, Endothelial Growth Factor, Vascular Endothelial Growth Factor, Myostatin, Lactate dehydrogenase, free fatty acids, and lactate dehydrogenase in triathlon athletes. This systematic review indicates that different triathlon race promotes an acute elevation of circulating cytokines and chemokines levels which return to standard levels after triathlon races. The findings of this systematic review demonstrate that the modulation of inflammatory parameters may be associated with an increase in metabolic indicators (CK, Cortisol, and LDH) after the end of different types of triathlon races.We would like to thank all authors for their fundamental contribution to the preparation of this systematic review.We would like to thank all authors for their fundamental contribution to the preparation of this systematic review

    Investigation of the role of ADAMTS-13 in the development of thrombosis in Behçet's patients

    Amaç: Behçet hastalığının vasküler tutulumunda, inflamasyon, tromboz ve koagülasyon süreçlerini bir arada değerlendirip, hastaların ilaç kullanım durumlarını inceleyerek, bu süreçlerde etkisi olabilecek parametrelerin serum düzeylerini belirlemek, bu parametrelerin vasküler tutulum ve hastalık aktivitesini belirlemedeki rolünü araştırmak amaçlanmıştır. Materyal ve metot: Çalışmaya 60'ı aktif dönemde ve 32'si vasküler tutulum öyküsüne sahip 98 Behçet hastası ile 50 gönüllü sağlıklı kişi dahil edilmiştir. Katılımcılardan alınan kanlar santrifüj edilip serum ve plazmalar -80°C'de dondurularak analiz gününe kadar saklanmıştır. Serum ADAMTS-13 ve Lp(a) düzeyleri ELISA yöntemiyle; D-dimer, fibrinojen, vWF düzeyleri immunotürbidimetrik yöntemle; IL-6 ve HCY düzeyi elektrokemilüminesans yöntemle; hs-CRP düzeyleri nefelometrik yöntemle; vitamin D düzeyleri ise LC-MS/MS yöntemiyle analiz edilmiştir. Bulgular: Behçet hastalarında ADAMTS-13, Lp(a), HCY, hs-CRP düzeyleri kontrol grubuna göre anlamlı olarak daha yüksek, vitamin D düzeyleri ise anlamlı olarak daha düşük bulunmuştır; ADAMTS-13 düzeyi aktif dönemde olan hastalarda remisyon döneminde olan hastalara göre anlamlı olarak daha düşükken fibrinojen ve hs-CRP aktif dönemde olan hastalarda remisyon döenmindeki hastalara göre daha yüksek bulunmuştur; steroid kullanan hastaların ADAMTS-13 düzeyleri steroid kullanmayan hastalara göre anlamlı olarak daha yüksek bulunmuştur. Behçet hastalarında ADAMTS-13 düzeyi ile hs-CRP arasında anlamlı negatif ilişki bulunmuştur. Sonuç: Sonuçlarımız ADAMTS-13 düzeylerinin hastalık aktivitesini yansıtmada gösterge olabileceğini göstermiştir. Ayrıca steroid kullanımının ADAMTS-13 düzeylerini artırmada etkili olduğu ve ADAMTS-13 düzeylerinin yüksekliğinin antiinflamatuvar etkide de rolü olabileceği görülmüştür.Aim: In our study, we aimed to evaluate the inflammation, thrombosis and coagulation processes together in the vascular involvement of Behçet's disease, to examine the drug use status of the patients, to determine the serum levels of the parameters that we think may have an impact on these processes, and to investigate the role of these parameters in determining vascular involvement and disease activity. Material and Method: 98 Behçet's patients and 50 healthy volunteers were included in the study. 60 of 98 patients are in the active phase of the disease and 32 have a history of vascular involvement. After venous blood was collected from the participants and the blood was centrifuged, serum and plasma were frozen at -80°C and stored until the day of analysis. Serum ADAMTS-13 and Lp(a) levels by ELISA method; D-dimer, fibrinogen, vWF levels were determined by immunoturbidimetric method; IL-6 and HCY levels by electrochemiluminescence method; hs-CRP levels by nephelometric method; Vitamin D levels were analyzed by LC-MS/MS method. Results: ADAMTS-13, Lp(a), HCY, hs-CRP levels were found to be significantly higher and vitamin D levels were significantly lower in Behçet's patients than in the control group; while ADAMTS-13 levels were significantly lower in patients in the active period than in patients in the remission period, fibrinogen and hs-CRP were found to be higher in patients in the active period than in patients in the remission period; ADAMTS-13 levels of patients using steroids were found to be significantly higher than patients not using steroids. According to the correlation analysis, a significant negative relationship was found between ADAMTS-13 level and hs-CRP in Behçet's patients. Conclusion: Our results showed that ADAMTS-13 levels may be an indicator of disease activity. It has also been observed that steroid use is effective in increasing ADAMTS-13 levels and that high ADAMTS-13 levels may also play a role in the anti-inflammatory effect

    A Modified Dijkstra Algorithm for ROS Based Autonomous Mobile Robots

    Abstract ? Autonomous Mobile Robots (AMRs) are frequently used in many fields of technology. In this study, an AMR was designed to execute different path planning algorithms. Firstly, working principle, system architecture and motion planning of AMR are presented. Then, a map for the current environment is produced by a Robot Operating System (ROS) powered AMR which was designed for this study. The AMR locates itself on the produced map with the aid of an integrated Light Detection and Ranging sensor (LIDAR). The locomotion of AMR to a user-defined target on the produced map is performed by an optimal path based on AMR's own navigation plan. Two different path planning algorithms, which are Dijkstra’s algorithm and a modified version of Dijkstra’s algorithm, are executed on a cost-effective AMR platform, which has the capability of Simultaneous Localization and Mapping (SLAM). The reason why Dijkstra algorithm is handled in this study rather than A*, D* and RRT algorithms is that this algorithm is a basic and widely used algorithm. Dijkstra’s algorithm is modified, and pros and cons of the modified algorithm are analysed compared to Dijkstra algorithm. The proposed algorithm and navigation of AMR are tested both in real time in real world and as a simulation in Gazebo. Two algorithms were compared according to the results obtained from the robot locomotion both in real application and simulation environment. It is observed that the modified version of the Dijkstra’s algorithm comparatively yielded a bit more satisfactory results in the aspect of path planning

    A Fecal-Microbial-Extracellular-Vesicles-Based Metabolomics Machine Learning Framework and Biomarker Discovery for Predicting Colorectal Cancer Patients

    Colorectal cancer (CRC) is one of the most common and lethal diseases among all types of cancer, and metabolites play a significant role in the development of this complex disease. This study aimed to identify potential biomarkers and targets in the diagnosis and treatment of CRC using high-throughput metabolomics. Metabolite data extracted from the feces of CRC patients and healthy volunteers were normalized with the median normalization and Pareto scale for multivariate analysis. Univariate ROC analysis, the t-test, and analysis of fold changes (FCs) were applied to identify biomarker candidate metabolites in CRC patients. Only metabolites that overlapped the two different statistical approaches (false-discovery-rate-corrected p-value 0.70) were considered in the further analysis. Multivariate analysis was performed with biomarker candidate metabolites based on linear support vector machines (SVM), partial least squares discrimination analysis (PLS-DA), and random forests (RF). The model identified five biomarker candidate metabolites that were significantly and differently expressed (adjusted p-value < 0.05) in CRC patients compared to healthy controls. The metabolites were succinic acid, aminoisobutyric acid, butyric acid, isoleucine, and leucine. Aminoisobutyric acid was the metabolite with the highest discriminatory potential in CRC, with an AUC equal to 0.806 (95% CI = 0.700-0.897), and was down-regulated in CRC patients.     Designing of rapid assay for the detection of RdRp/Orf1ab specific to SARS-CoV-2

    SARS-CoV-2 is still threat and mostly used detection method is real time reverse transcriptase polymerase chain reaction (rRT-PCR) for the open reading frame (Orf1ab), RNA-dependent RNA polymerase (RdRp), nucleocapsid (N) and envelope (E) genes of virus. However, rRT-PCR may have false negative rate for the nucleic acid detection. Since the RdRp/Orf1ab has high sensitivity for the molecular detection, two sandwich models, Model 1A-Model 1B, based on hybridization on lateral flow assay (LFA) were designed here and applied with the synthetic and clinical samples of RdRp/Orf1ab. In this purpose colloidal gold nanoparticles (AuNPs) were used as label. Membranes having different flow rate, three oligonucleotide probe concentrations and running buffers were used. Although synthetic target sequence was recognized by all the LFAs, PCR products obtained from either the synthetic plasmid DNA or oro/nasopharyngeal swabs were detected by Model 1 A using W12 mem-brane. Designed strip assays detected the RdRp/Orf1ab of the clinical samples as 100% sensitivity and specifity. It means that they might be used for the detection of virus and can be modified for the recognition of mutant genes of virus.     The effect of comics technique on teaching stories for hearing impaired individuals

    İşitme engelli bireyler, normal işiten bireylere göre kendilerine anlatılan konuları anlamakta birtakım güçlükler yaşamaktadır. İşitme engelli bireylere eğitim veren eğitimciler ise ilgili dersi öğrencilere anlatırken zorluk çekmektedirler. Bu zorlukların sebeplerinden biri de anlattıkları konuyu görselleştirerek anlatmak zorunda olmalarıdır. Çoğunluğu soyut konular olan din derslerinde bu zorluk bir hayli artmaktadır. Bu araştırmada, bu sorun üzerine yoğunlaşılmış, eğitimcinin konuyu daha kolay ve anlaşılır bir şekilde anlatabilmesi, öğrencilerin de konuyu daha iyi anlayabilmesi için çizgi roman tekniğinin etkililiği araştırılmıştır. Bu amaçla, fil kıssası çizgi romana aktarılarak, çizgi romanın işitme engelli bireylerde, anlatılan kıssanın öğrenilmesi üzerindeki etkililiği araştırılmıştır. Araştırmada kontrollü öntest-sontest deneysel desen kullanılmıştır. Araştırmanın çalışma grubu, İstasyon Gençlik Merkezi Kur'an Kursuna devam eden 14 işitme engelli öğrenciden oluşmaktadır. Bu öğrenciler deney ve kontrol grubu olmak üzere iki gruba ayrılmıştır. Araştırmaya katılan bu gruplara deney öncesinde ön test, uygulama sonrasında ise son test uygulanmıştır. Araştırmanın nicel verileri, bir deney ve bir kontrol grubuna uygulanan "başarı testinden" elde edilmiştir. Araştırmada, işitme engellilerin eğitiminde görsel içerikli materyal kullanılmasının, işitme engelli öğrencinin, konu daha kolay öğrenmesini sağladığı ve öğrenciyi derse karşı daha ilgili ve motive hale getirdiği, dersi öğreten öğreticiye kolaylık sağladığı sonucuna ulaşılmıştır. Lecturers who teach hearing-impairment individuals have difficulties while having classes with them. One the reasons of these difficulties is having to lecture them by visualizing the subjects in their curriculum. That handicap escalates in religious subjects which have abstract themes. In this research, that handicap is zoomed in and the efficacy of comic books technique is gone through in order the fact that lecturers are able to teach individuals with hearing-impairment easier and cognisably as well as the learners comprehending the topic much better. Hence, efficacy of comic books is observed on the learning outcomes by narrating ' The Incident Of Elephant' on a comic book. Controlled pre-test /post-test experimental approach is used in this research. The participants of the research consists of the fourteen year old individuals with hearing impairment studying 'İstaston Youth Center Quran Course'. Students are divided into two groups as contolled group and experimental group. Those students are made to take a pre-test before the experiment and later a post-test after the experiment. Quantitative data are obtained through the achievement test that are applied on both contolled and experimental groups. Consequently in this research, it is acknowledge that using visual materials in teaching process has a significant effect on learning the subject smoothier, increasing students' eagerness and motivation towars the subject and providing convenience to the students.
