    Biochemical Studies to Understand Teratogenicity and Lethality Outcomes in Modified-FETAX

    The frog embryo teratogenesis assay-Xenopus (FETAX) is a standardized test used to assess the toxic and teratogenic effects of xenobiotics. With this test, toxic and/or teratogenic concentrations of xenobiotic substances can be determined using morphological parameters such as lethality, length, and malformations in stage 8–11 Xenopus laevis embryos after 96 h exposure. These parameters enable the determination of the median lethal and effective concentrations (LC50 and EC50), minimum concentration to inhibit growth (MCIG), and teratogenic index of the tested chemical to reveal the short-term effects of relatively high concentrations. On the other hand, although FETAX provides quantitative and qualitative data on teratogenicity and toxicity, the biochemical and molecular mechanisms of these effects cannot be explained. Recent studies have tried to elucidate the mechanisms causing malformations and to explain the underlying causes of toxicity and teratogenicity by biochemical marker analysis. This chapter describes methods to analyze modified-FETAX and some detoxification and oxidative stress-related biomarkers during the early embryonic development of X. laevis. © The Author(s), under exclusive license to Springer Science+Business Media, LLC, part of Springer Nature 2024

    Bu çalışma, enerji sektöründe ortaya çıkan uyuşmazlıkların çözümünde arabuluculuk ve tahkim süreçlerinin rolünü kapsamlı bir şekilde incelemektedir. Enerji projeleri, büyük yatırım gerektiren ve uzun vadeli taahhütler içeren karmaşık yapıları nedeniyle uyuşmazlıklara açık alanlardır. Bu bağlamda, arabuluculuk ve tahkim, taraflar arasında ortaya çıkan anlaşmazlıkların dostane ve hukuki bir zeminde çözülmesi için sıklıkla başvurulan alternatif uyuşmazlık çözüm yöntemleridir. Çalışmada, enerji sektöründe arabuluculuğun hız, maliyet etkinliği ve esneklik gibi avantajları vurgulanırken, tahkim süreçlerinin daha resmi ve bağlayıcı kararlar sunduğu ancak zaman zaman uzun ve maliyetli olabileceği belirtilmektedir. Ayrıca, bu yöntemlerin uluslararası düzeyde nasıl uygulandığı, Türkiye örneği ile küresel uygulamalar karşılaştırılarak değerlendirilmiş t ir. Önemli karar örnekleri ve tahkim kararları ışığında, enerji sektöründe arabuluculuk ve tahkim süreçlerinin etkinliği ve taraflar arasındaki iş birliği üzerindeki etkileri detaylı bir şekilde ele alınmıştır

    Adolescencia y creatividad. Una perspectiva libertaria: la de Luis Feduchi

    Given the loss of Luis Martínez-Feduchi, a Spanish adolescent mental health specialist, a review of his contributions to the adolescent clinical practice is carried out, particularly for the current pan-Hispanic cultural and social situation. We highlight his special attention to the freedom and autonomy of adolescents in all kinds of clinical settings, including the one that has to do with juvenile justice devices. In this sense, we remember their conceptions regarding the differences between tolerance and compromise in relation to their transgressions and/or disruptive behaviors and the need not to medicalize or marginalize the particular grief, conflicts, modes of expression and tasks of that vital stage. His contributions in those fields and devices are summarized, ending with a brief memory of the person.Ante la pérdida de Luis Martínez-Feduchi, especialista español en salud mental de la adolescencia, se realiza una revisión de sus aportaciones a la clínica de las adolescencias, en particular, para la situación cultural y social panhispánicas actuales. Se remarca su especial atención a la libertad y autonomía de los adolescentes en todo tipo de clínica, incluida la que tiene que ver con los dispositivos de Justicia de menores. En ese sentido, se resumen sus concepciones en relación con las diferencias entre tolerancia y transigencia con respecto a sus trasgresiones y/o comportamientos disruptivos y la necesidad de no medicalizar ni marginar los particulares duelos, conflictos, modos de expresión y tareas de esa etapa vital. Se resumen sus aportaciones en esos campos y dispositivos, para terminar con un breve apunte sobre el futuro de sus aportaciones

    Evaluation of busulfan, cyclophosphamide, and etoposide as a preparation regimen for autologous stem cell transplantation in Hodgkin's lymphoma patients

    BACKGROUND: Hodgkin's lymphoma (HL) exhibits a cure rate of 90% in patients diagnosed at an early stage and a cure rate ranging from 70% to 90% in patients diagnosed at an advanced stage. In the case of patients with relapsed/refractory HL (r/rHL), it is recommended to provide salvage chemotherapy initially, followed by autologous stem cell transplantation (ASCT). The ideal conditioning regimen for the transplantation process is still being investigated.OBJECTIVES: For individuals with r/rHL, high-dose chemotherapy combined with ASCT (HD-ASCT) is thought to be the most effective method of treatment. The purpose of this research was to evaluate the effectiveness and safety of the busulfan, cyclophosphamide, and etoposide (BuCyE) preparation regimen in r/rHL patients.MATERIALS AND METHODS: Retrospective analysis was conducted on the data of 67 lymphoma patients older than 18 years who had HD-ASCT with the BuCyE conditioning regimen between September 2014 and November 2021 (86 months). The research consisted of 34 r/r HL patients among them. A parenteral regimen of 0.8 mg/kg of busulfan every 6 h from day -7 to day -5, 50 mg/kg of cyclophosphamide on days -3 and -2, and 400 mg/m2 of etoposide on days -5 and -4 comprised the patient preparation regimen before ASCT. All data were collected from inpatient files and the Inonu University Turgut Ozal Medical Center Hospital Information System.RESULTS: The median age of the patients was 43 years, and 67.6% were males. The most common type of HL was nodular sclerosis, which was followed by mixed cellularity. The median time for platelet and neutrophil engraftment was 14 and 11 days, respectively. 5.0 x 106/kg was the median transplanted dose of CD34+ cells (2.1-13.55). Liver toxicity was observed in 6 (17.6%) patients. Eight patients suffered from pulmonary side effects. The median number of previous chemotherapies was 2 (2-4). In all lymphoma patients, the complete response rate was 61.8% (n = 21), whereas the disease progression rate was 32.3% (n = 11). Transplantation-related mortality on the 100th day was 8.8% (n = 3). Three-year overall survival was 57.17%.CONCLUSION: When the literature was reviewed, the studies with the BuCyE preparation regimen in patients with r/rHL were limited. This conditioning regimen was found to have fewer side effects and a lower cost. It can be preferable when compared to carmustine (BCNU), etoposide, cytarabine (ARA-C), and melphalan (known as BEAM) in r/rHL

    Pd-PEPPSI-type expanded ring N-heterocyclic carbene complexes: synthesis, characterization, and catalytic activity in Suzuki-Miyaura cross coupling

    In this work, we synthesized a series of six unsymmetrical benzimidazolium salts 2 and their pyridine-enhanced precatalyst preparation stabilization and initiation (PEPPSI)-themed palladium N-heterocyclic carbene complexes [PdCl2(NHC)(Py)]. All the products were isolated in satisfactory yields (75-85%). The synthesis of these novel palladium PEPPSI complexes involved in reacting NHC precursors with PdCl2 in pyridine at 60 degrees C in the presence of excess K2CO3. The structures of all compounds have been characterized by H-1 NMR, C-13 NMR, HRMS and IR spectroscopy, as well as elemental analysis techniques, which support the proposed structures. The catalytic activity of the six complexes was assessed in the Suzuki-Miyaura cross-coupling of phenylboronic acid and aryl halides.     An exploratory, single-center study of factors associated with child marriage among Syrian female adolescents residing in Turkey

    This exploratory study aimed to evaluate factors related with child marriage compared to unmarried status in female adolescent residents in a refugee camp in Turkey. The rate of child marriage and developmental status of offspring from those marriages were reported. A research team evaluated married female youth and their offspring for psychopathologies according to DSM-5 criteria and ascertained lifetime traumatic events among mothers. We compared the traumatic experiences and psychopathologies of married females and controls. Post-traumatic stress disorder (PTSD) was the most common diagnosis in both groups and child brides reported greater cumulative traumatic experiences and elevated rates of PTSD. More than one-tenth (i.e., 15.1%) of offspring of child brides displayed developmental delays and 12.1% were diagnosed with global developmental delay

    Chronic liver fibrosis induction in aging causes significant ultra-structural deterioration in liver and alteration on immune response gene expressions in liver-spleen axis

    The relationship between damage to the liver and spleen by aging and the immune response status in these two organs, which are anatomically and immunologically interconnected, is unknown. The authors investigated the histopathological, ultrastructural, and immunological effects of aging in young and aged fibrotic mice by using an experimental model. Four groups were planned, with 10 mice in each experimental group. The levels of fibrosis and ultrastructural destruction in the liver were determined by alpha-SMA staining and TEM analysis. Expression levels of immunity genes (Il2, Il4, Il6, Il10, Il12, Il17, Tnf, Ifng, Tgfb1, Gata3, Rorc, Tbx21, Foxp3, Ccl2, Ccr2, Cxcr3, Pf4, Cxcl10) were carried out by qRT-PCR. While structural disorders were detected in the mitochondria of aged healthy group, cellular destruction in the fibrosis-induced elderly group was at a dramatic level. Fibrosis induction in aged mice caused an elevation in the expression of chemokines (CCl2, CXCL10, CCR2) and cytokine (IL-17a) genes that induce autoinflammatory response in the liver.     Validity and Reliability Study of Turkish Version of Abbreviated Science Anxiety Scale

    The aim of this study is to adapt “Abbreviated Science Anxiety Scale”(ASAS) developed by Megreya et al. (2021) to Turkish and to investigate the reliability and validity of Turkish-ASAS. In the adaptation study, some steps were determined and implemented. After the English-Turkish translation validity of ASAS was ensured, data were collected within the scope of the survey study for reliability and validity studies. The study group in the scale adaptation study are experts (teaching staff in science education, Science teacher, and English teacher) and 410 middle school students (Grade 5, 6, 7 and 8). After the descriptive statistics and item analysis were made, the exploratory and confirmatory factor analysis was performed to reveal the factor structure. The adaptation stage findings shows the two-factor structure of ASAS. These factors are “learning science anxiety” and “science evaluation anxiety”. Cronbach Alpha reliability coefficient for ASAS-Turkish was calculated as 0.703. As in the original scale, ASAS-Turkish is a 5-point Likert type scale and consisting of 9 items. The results show that ASAS-Turkish is a short, valid and reliable tool. Low scores on the ASAS indicate low anxiety and high scores indicate high anxiety. It can be used to guide and enhance scientific study
