Internal leaks in industrial valves pose a critical issue that can lead to significant financial impacts, including energy wastage, product loss, and elevated operational costs. Furthermore, these leaks compromise worker safety by exposing personnel to potential accidents and hazardous environments. While monitoring techniques are available on the market, none effectively combine simplicity, reliability, and cost-efficiency. This work aimed to develop a continuous monitoring system based on acoustic emission technology, capable of accurately detecting and quantifying internal valve leaks. The system is also designed to be easy to implement and economically viable, allowing for large-scale application in industrial environments. To achieve this goal, the project involved design, development, numerical simulation, production, and experimental validation of acoustic amplifiers, specifically engineered to capture targeted frequencies and efficiently filter acoustic signals. Improvements were made to an experimental demonstrator, particularly in the valve actuator system. The results obtained demonstrate that the developed system is effective in detecting internal leaks, confirming its technical and economic viability. Implementing this solution has the potential to substantially reduce financial losses and enhance operational safety in industries that rely on valves, representing a promising approach to addressing a recurring issue

Chromatography is widely used in biopharmaceutical purification, particularly for therapeutic antibodies. However, traditional methods, such as using Protein A resin, are costly and present challenges like ligand leakage and antibody denaturation. Additionally, the process requires large amounts of water and generates significant chemical waste, raising environmental concerns. This thesis focuses on the development of affinity ligands to reduce production costs while maintaining efficiency in antibody purification. By leveraging virtual compound libraries, a scaffold was used to generate millions of potential ligands. This approach reduces the number of experimental candidates while ensuring high chemical diversity. The virtual library was screened to identify ligands with strong binding potential to the antibody. The top-performing ligands were further analyzed to gain insights into their molecular interactions. Two ligands showed strong potential for purification. These findings represent a promising advance toward more sustainable and cost-effective antibody purification.

    No full text
The aim of this study was to develop a generic analgesic and antipyretic drug in powder form for oral solution, with the primary goal of filling a gap in the Portuguese market. Firstly, existing formulations in other markets were analysed and the choice of excipients, defining the reference drug. The Design of experiments (DoE) was employed in the formulation study to assess the influence of sweetener, glidant, diluent and active pharmaceutical ingredient (API) on the powder's physicochemical properties, yielding a total of 18 experimental runs. It was concluded that the diluent Pearlitol®200 SD, representing 87% of the formulation, is the critical factor for adequate flowability. Two formulations were selected, according to the flowability. Solubility tests showed that the API is very soluble, confirming that this active substance belongs to Class I of the BCS System. The formulations prepared exhibited similar physicochemical characteristics to the reference drug, however, the absence of flavouring resulted in a less favourable sensory experience. Analyses of particle size and loss on dying confirmed the similarity between the formulations. The active substance dosage results showed values in range of 95% and 103%. The analysis of impurities by HPLC demonstrated the results were within the specifications, with impurity C of 0.18% in greater quantity, due to this being an active metabolite of the API, impurity A of 0.02% only present in the reference drug and an unknown impurity of 0.04%. The analytical matrix and API samples confirmed the absence of interference in the results. The final formulation chosen contains sodium cyclamate as a sweetener, blackcurrant flavouring, glidant, Pearlitol®200 SD and API, optimising taste and physicochemical properties

    No full text
The objective of this study was to evaluate washed cork stoppers passing through the chromatographic modules of NDtech 2.0, assess their impact on the accurate detection and quantification of 2,4,6-trichloroanisole (TCA), and develop a new chromatographic method capable of analyzing these stoppers within a maximum time frame of 20 seconds. Cork is widely recognized as the most appropriate material for sealing alcoholic beverages. Natural cork stoppers stand out as the superior choice for sealing wines, not only for their ability to preserve the quality and characteristics of the beverage, enabling its evolution and maturation, but also for being considered the most environmentally sustainable material. However, cork may contain TCA in its volatile fraction, which is considered the primary compound responsible for organoleptic deviations in wine, characterized by musty and moldy odors, giving rise to the expression "cork taint." To address this issue, Amorim Cork developed NDtech®, a process capable of individually and non-invasively identifying the amount of TCA present in analyzed stoppers, thereby removing contaminated stoppers from the production flow. NDtech stoppers have the highest added value within the company, and their demand in the wine market has been successful and is growing. However, the inability to analyze washed stoppers imposes limitations on production, creates logistical complications, and incurs additional costs. In this project, the chromatographic performance of NDtech 2.0 modules was evaluated in the analysis of stoppers from the reactive washing process, where different levels of hydrogen peroxide were used. Various types of chromatographic columns, injection volumes, and temperatures applied to the chromatographic system were tested and analyzed. The study concluded with the implementation of the best-evaluated modifications in a chromatograph on the NDtech 2.0 production line, including the development of several new chromatographic methods and tests of a final method with a 16-second analysis capability over 24 hours. Further in-depth study of this method, as well as its validation, are still necessary

    No full text
The objective of this study is to increase the recovery of protein and starch in the wet milling process by optimizing the fiber washing stage through the addition of enzymatic hydrolysis. The influence of different multi-enzymatic complexes on the release of starch and protein retained in the suspension was studied. Laboratory assays, with an incubation time of 3 hours at a temperature of 122 °F (50 °C), resulted in a increase in starch separation, and a increase in protein separation from the fiber. These enzymes catalyze the hydrolysis of the fiber, enabling the extraction of both free starch and starch bound to the cell wall retained in the fiber, thereby increasing the efficiency of the washing stage. Additionally, the most suitable stage in the washing system for enzymatic hydrolysis was investigated. A laboratory assay using suspension samples from the process concluded that it is possible to separate 1% more starch from the fiber with the substrate/suspension from the 5th washing stage (intermediate/final stage of the system) compared to the substrate/suspension from the 2nd washing stage (initial stage). Another objective of this study was to evaluate the influence of enzymatic hydrolysis on the reduction of fiber moisture at the outlet of the press, using E2 without incubation in the previous stage. To achieve this, an industrial trial was conducted by adding 1.2 L/h of enzyme in the sixth fiber washing stage, resulting in a reduction in suspension moisture. The enzymes reduce the fiber's capacity to retain water, thereby increasing the efficiency of the dehydration stage. A less moist suspension requires less drying, reducing the energy costs of this operation

This work focuses on the development of an analytical method by liquid chromatography (HPLC) for two pharmaceutical products with the same active substance but in different salts, aiming for the efficient quantification of active substances and preservatives. From the optimization of the method, as described in the Pharmacopeias, resulted in shorter run times of 15 minutes and simplified the procedure, reducing both financial and time costs. Additionally, it allowed the use of the same method for both products and enabled the quantification of preservatives present in one of the formulations. The mobile phase used was Buffer:Methanol (25:75), with a flow rate of 1.2 ml/min, an injection volume of 25 μL, and detection at 254 nm, using a reversed-phase C18 column. The pre-validation tests were conducted using the respective reference products, except for accuracy and degradation, which were verified in the laboratory batch (DSD009). These tests revealed that the quantification of active substances and preservatives showed repeatability – %RSD between 0.82 and 1.24 – both active substances and preservatives remained stable for at least 3 days after the preparation of the solutions, and it was also verified that the method is considered accurate – with a recovery of 98.8% and %RSD of 0.84. The forced degradation tests indicated that the product does not significantly degrade under basic conditions, but even when degraded by about 93% in acidic conditions along with peroxide, the impurity peaks detected still showed good resolution. Solubility tests in different media resulted in the selection of 80% Phosphate Buffered Saline (PBS) + 20% Ethanol as the receiving phase in the in vitro release tests (IVRT). In the IVRT tests, quantifications were performed using the developed analytical method. Reference product 1 and its respective test product showed similar release behaviors. Reference product 2 did not reach the 70% release target after 24 hours, resulting in the suspension of the corresponding batch evaluation

Optimization of Bioink Formulations for Peripheral Nerve Regeneration

    No full text
Peripheral nerve injuries are a common health problem that affects millions of people worldwide, often depriving them of normal nerve function and sensitivity and is accompanied by pain and discomfort. Serious nerve injuries may lead to a large gap between the two nerve stumps and require surgical intervention. Nerve guidance conduits (NGCs) are, in a simplified way, hollow tubes that are seeded into nerve endings and supply guidance so that the correct nerve regeneration may occur. Currently, one of the most promising treatment options for peripheral nerve injuries is a class of NGCs made from biodegradable materials. These materials can be absorbed by the body at a rate that does not interfere with nerve regeneration, and synergy can be achieved with the addition of cells/growth factors or drugs (e.g. anti-inflammatories). Previously, the University of Coimbra (Portugal) conducted two research projects on peripheral nerve regeneration of the rat sciatic nerve through the production of: 1) membranes and 2) hollow NCGs, both using a biomaterial ink composed of Dextran and Poly (ε-caprolactone) polymers. However, for scale-up purposes, an adaptation of the same formulations was replaced by a 3D printing manufacturing method (Digital Light Processing, DLP) now capable of printing personalized NGCs. Here, I present the synthesis of the polymers for the ink, the characterization of UV-cured hydrogels, and the 3D printing of NGCs from three formulations regarding their swelling capacities, hydrolytic degradation, mechanical, photo rheological and thermal properties that demonstrate individual properties with potential for customizable NGCs. A detailed empirical study of the ink performance and printing parameters was also performed. The incorporation of Gallic acid into the printed tubes was proven to be successful with a controlled release of the molecule over 48h.As lesões de nervos periféricos são um problema de saúde comum que afeta milhões de pessoas em todo o mundo, muitas vezes privando-as da função sensorial e motora nervosa, sendo frequentemente acompanhadas de dor e desconforto.

    No full text
The aim of this study was to assess the feasibility of anaerobic co-digestion (AcoD) with fats, oils and grease (FOG) at FA da Guia wastewater treatment plant. FOG is a substrate with a high theoretical methane production potential, ranging from 0,7 to 1,1 m³ CH4/kg VS. The interest in testing this substrate arose from Águas do Tejo Atlântico's project to study the feasibility of co-digesting and valorising this type of waste to Increase the energy produced at the FA da Guia. To this end, a biochemical methane potential (BMP) test was carried out to replicate real digester conditions. Digested sludge was used as an inoculum, thickened sludge as a substrate and FOG as a co-substrate. Three conditions were tested with different volumes of added FOG: 0% FOG (blank), 5% FOG and 10% FOG. Although preliminary, the results of the BMP test allow to conclude that the addition of FOG did not show any inhibitory effect on anaerobic digestion and that it considerably increased the potential for methane recovery. The test with 10% FOG recorded the highest specific methane production, with a maximum value of 176,2 NmL/g VS. The addition of FOG resulted in at least a 47% increase in specific methane production compared to the blank test. However, it was clear that it is essential to carry out more tests. The duration of the tests should be increased, and new conditions and pre-treatments should be tested in order to ensure that methane production and the efficiency of the digestion process are maximised
