Towards the establishment of a reference method for Mycobacterium avium drug susceptibility testing

    No full text
"Mycobacterium avium is the most common non-tuberculous mycobacteria (NTM) associated with human infections. Treatment of M. avium disease is challenging, with microbiological eradication typically requiring a multidrug antibacterial regimen over a prolonged period. This scenario worsens as no standardized drug susceptibility testing guidelines are available. This work aimed to evaluate the provisional European Committee for Susceptibility Testing (EUCAST) reference protocol for minimum inhibitory (MIC) determination of slow growing NTM, correlate it with established molecular determinants and examine its usefulness for testing new drugs.(...)"N/

This report was written as part of the Supervised Teaching Practice (STP) of the Master’s Degree in Teaching Music Education in Basic Education of the Faculty of Social and Human Sciences in Lisbon. This report portrays the work carried out in the context of an internship in a public school in the district of Lisbon, in the school year of 2023/2024. Initially, the report seeks to critically and succinctly describe the entire experience that made up the STP. The first stage is the contextualisation of the public school where the internship took place and an analysis of the pedagogical practice, taking into account the guidelines that apply to the subject of Musical Education. This section also includes a characterisation of the classes, including a description and a reflection on the experience gained with the Specialised Support Unit for Students with Multidisabilities. The analysis and reflection on the observation and teaching components, as well as a description of the participation in projects, events and meetings are also part of this chapter. At the end, there is a critical reflection on the role of the Musical Education teacher, as well as a general critical observation of the pedagogical practice. Next, it is presented the implementation, analysis and reflection of the project based on the theme of Soundscapes. The project and the activities carried out in a 5th grade class, with a focus on musical exploration, improvisation and its interconnection with poems written by the heteronyms of the poet Fernando Pessoa, are portrayed. Using Soundscapes as a stimulus for musical creation and improvisation, this project seeks to reflect on the importance that this theme has, not only in the creative skills of the students, but also in its transversality to other areas of knowledge. Starting with vocal exploration, this project reflects not only on the possibilities that this theme brings with regard to individual musical creation, but also in small groups and in its potential as a pedagogical tool using poetry as the creative material for a musical performance. Once the project’s outputs have been presented and analysed, some considerations are made, as well as the implications for the future of the project

Funding Information: This research was funded by FCT/MCTES (PIDDAC, Portugal) under the programmatic projects UIDB/50020/2020, UIDP/50020/2020 and LA/P/0140/2020; and by the European Commission through GLYCOTwinning (Grant Agreement: 101079417), and Horizon 2020 research and innovation programme under the EJPRD COFUND-EJP N 825575 (EJPRD/0001/2020). Publisher Copyright: © 2025 IEEE.GNE myopathy (GNEM) is a rare adult-onset disease characterized by atrophy and weakness of skeletal muscles caused by mutations in the GNE gene. There is no approved therapy, but clinical trials with supplementation with N-acetylmannosamine (ManNAc) alleviated the GNEM-associated decrease in sialylation. Nevertheless, oral intake of ManNAc has shown low absorption and high gastrointestinal adverse effects. In this regard, the aim of this work was to explore an alternative delivery route for ManNAc directly targeting the muscle tissues. For this we investigated the thermal responses of Pluronic F-127 gel and its muscle adhesion after subcutaneous injection, and the kinetics of release of gelencapsulated ManNAc in phosphate buffered saline medium. The results obtained show the thermal response of Pluronic F-127 represents a unique opportunity for its injectability and function as a drug delivery system directly acting on the muscle surrounding area and showing gel stability in the target area. Moreover, the continuous delivery of ManNAc from the gel (20% Pluronic F-127) was detected during 3h, at a continuous rate and slower than using a simple ManNAc solution. In conclusion, the data obtained in this work supports the possible use of the Pluronic F-127 thermoreversible hydrogel loaded with ManNAc in GNE myopathy therapy and research.authorsversionpublishe
