Giardíase como preditor de desnutrição Infantil em regiões em desenvolvimento
RESUMO
As infeções parasitárias intestinais são uma preocupação de saúde pública a nível global, especialmente em populações vulneráveis das áreas tropicais e subtropicais. Um dos principais agentes de infeção intestinal é Giardia duodenalis, que se encontra presente de forma ubíqua, tornando muito difícil o seu controlo. A desnutrição infantil é, e continuará a ser, um motivo de preocupação, principalmente em crianças até aos 5 anos de idade. Apesar de nos países em vias de desenvolvimento, as infeções parasitárias e a desnutrição infantil coexistirem, a associação de infeção por G. duodenalis e a desnutrição infantil sempre foi um ponto de estudo, embora ainda não se tenha conseguido chegar a um consenso. Diversos estudos foram realizados com o intuito de determinar essa mesma associação, e uma meta-análise dos trabalhos publicados poderá vir a ser um ponto de partida para avaliar a robustez das conclusões descritas por esses autores. Após pesquisa bibliográfica de artigos publicados nas bases de dados Pubmed, e SciELO contendo as palavras “Giardia” e “malnutrition” foram selecionados 19 artigos para este estudo. A realização desta meta-análise, com base nas publicações selecionadas, teve como objetivo principal gerar dados que ajudem a fundamentar a hipótese de existir uma associação entre a infeção por G. duodenalis e os seguintes parâmetros, indicadores de desnutrição infantil: baixo peso, nanismo e baixo índice de massa corporal. Os resultados obtidos neste estudo indicam que as crianças infetadas com G. duodenalis têm mais odds de virem a sofrer de baixo peso ou nanismo, comparativamente a crianças não parasitadas por este protozoário, sugerindo que a giardíase possa constituir um fator preditor de desnutrição infantil.ABSTRACT
Intestinal parasitic infections are a significant concern in global public health, especially among vulnerable populations in tropical and subtropical regions. One of the common infectious agents is Giardia duodenalis, which is ubiquitously present worldwide, making its control challenging. Child malnutrition is, and will continue to be, a major concern, particularly in children under 5 years of age. Although parasitic infections and child malnutrition coexist in developing countries, research on the association between G. duodenalis infection and child malnutrition has yet to reach a consensus. Several authors have explored this potential link, and a meta-analysis of their publications could provide valuable insights into the findings of these studies. After conducting a bibliographic search of articles published in the PubMed and SciELO databases using the keywords “Giardia” and “malnutrition,” 19 articles were selected for this study. The meta-analysis performed on the selected articles aimed to gather evidence for a possible association between G. duodenalis infection and specific indicators of child malnutrition, including underweight, stunting, and low body mass index. The results of this study indicate that children infected with G. duodenalis have higher odds of suffering from underweight or stunting compared to children not parasitized by this protozoan, suggesting that giardiasis may be a predictor of child malnutrition
Hidrogel Compósito de Polímero/Nanofibras de Base Biológica para Aplicações Biomédicas
A degeneração do disco intervertebral (DIV), muitas vezes iniciada no núcleo pulposo (NP), está associada à dor lombar, que é uma das principais causas de incapacidade a nível mundial e afeta 80% da população mundial. O tratamento desta condição é um desafio, pois os tratamentos atuais, limitam a capacidade do movimento do DIV e não apresentam a capacidade de restaurar a estrutura e as funções do DIV na totalidade. Neste trabalho, pretende-se desenvolver uma alternativa menos invasiva capaz de devolver as funções biomecânicas do NP danificado, como um hidrogel injetável à base de quitosano, caracterizado por ser um material compósito de biopolímeros, e que combine a reticulação via reações de base Schiff e as propriedades termorresponsivas de um LCST (Temperatura Inferior de Solubilidade Crítica), como a hidroxipropilcelulose oxidada (HPC-Ox).
Para tal foram testados vários sistemas, sendo o escolhido constituído por 2% quitosano, 2% celulose nanocristalina, 6% glicerofosfato e HPC-Ox (variável entre 2, 4 e 6% m/m). Através dos ensaios reológicos verificou-se que as ligações covalentes dominaram o sistema de reticulação, que o aumento de HPC-Ox aumenta as propriedades mecânicas, e apresentam curvas de viscosidades semelhantes às de um sistema líquido cristalino. Foi possível verificar que os hidrogéis apresentaram capacidades de absorção máximas, em solução tampão fosfato salina (PBS) a 37 ºC, entre 548 e 230%. Ensaios de compressão não confinados a 37 ºC nestes hidrogéis originaram módulos de Young médios de 17, 43 e 58 kPa, ou seja, apresentam propriedades mecânicas promissoras para imitar o NP saudável. A análise MEV demonstrou que estes hidrogéis apresentam estruturas porosas complexas e observar a organização helicoidal originária de fases líquidas cristalinas na parede dos poros.
Os resultados obtidos até ao momento são bastante promissores para a fase inicial de desenvolvimento deste novo sistema de hidrogel para aplicações biomédicas, tais como, o tratamento do NP.Degeneration of the intervertebral disc (IVD), often beginning in the nucleus pulposus (NP), is associated with low back pain, which is one of the main causes of worldwide disability and affects 80% of the world's population. Treating this condition is a challenge, as current treatments limit the movement capacity of the IVD and do not have the capacity to restore the structure and functions of the IVD in its entirety. This study aims to develop a less invasive alternative capable of restoring the biomechanical functions of the damaged NP, such as an injectable chitosan-based hydrogel, characterized by being a biopolymer composite material, which combines cross-linking via Schiff base reactions and the thermoresponsive properties of an LCST (Low Critical Solution Temperature), such as hydroxypropyl cellulose oxidized (HPC-Ox).
To achieve this goal, various systems were tested, with the chosen one consisting of 2% chitosan, 2% nanocrystalline cellulose, 6% glycerophosphate and HPC-Ox (varying between 2, 4 and 6% m/m). The rheological tests showed that covalent bonds dominated the crosslinking system, that the increase in HPC-Ox improved the mechanical properties and that the flow curves were similar to those of a liquid crystalline system. It was possible to verify that the hydrogels showed maximum absorption capacities, in phosphate buffered saline (PBS) at 37 ºC, of between 567 and 237%. Unconfined compression tests at 37 ºC on these hydrogels resulted in average Young's moduli of 17, 43 and 58 kPa, i.e. they have promising mechanical properties for imitating healthy NP. SEM analysis showed that these hydrogels have complex porous structures and observed the helical organization originating from liquid crystalline phases in the pore wall.
The results obtained so far are very promising for the initial development phase of this new hydrogel system for biomedical applications, such as the treatment of NP
AAV2 capsid fusion of large proteins by rational design: Insertion sites breakdown
Gene Therapy is one of the most extensively studied therapeutic modalities in both preclinical and clinical contexts and have shown efficacy in treating a wide range of diseases. Adeno-associated virus (AAV) is a commonly used viral vector in gene therapy due to its capability to safely and efficiently deliver therapeutic genes to target cells. Despite its success, AAV vector’s effectiveness can be enhanced through modifications to its capsid, the protein shell that protects the viral genome, by improving transduction, altering tropism, and reducing immune responses. Capsid engineering, particularly the insertion of larger proteins, remains an area of active research to further expand AAV vector's therapeutic potential. This technique requires separating the capsid viral proteins (VP1, VP2, and VP3) expression to allow for precise protein insertions into the AAV vector capsid sites. Despite previous studies, there is still a need to understand how these insertions affect the functionality of proteins over 20 kDa and the functionality of the vector capsid.
In this study we evaluated the insertion of mCherry protein (~28kDa) into three specific regions of the AAV2 capsid (residues 453 and 587 in VP1, and the N-terminal residue 138 in VP2), using four capsid mosaic constructs. We successfully validated these constructs by transient transfection, where insertion sites presented between a 15 and 148-fold decrease in mCherry-positive cells compared to the control. Following this, the engineered AAV2/mCherry mosaic vectors were produced and characterized by assessing their total particles, viral genome and transducing titers. No major losses in total particles and viral genome particles per mL were observed compared to the WT AAV2 vector control. Nevertheless, in terms of transducing units, there was a more prominent decrease in titers than in the control. Additionally, the effects of mCherry insertion on the AAV2 vector capsid's performance were evaluated in vitro. In VP1 insertion at the 453 residue, we observed that the mCherry peak was at 48 hours with almost 20% of mCherry positive cells. In VP2, results presented the highest value at 24 hours, with around 75% of mCherry positive cells maintaining the signal at 48 hours. This, overall, indicated a 4-fold difference in the mCherry signal between insertion sites in VP1 and VP2.
These findings allowed for a comparison of the three different insertion sites and their impact on overall AAV2 vector functionality and protein expression, indicating that the best hotspot for insertion of proteins ≥20kDa is residue 138 followed by 453 residue. This work lays the groundwork for the development of more effective AAV-based vectors, advancing gene therapy and expanding its potential to treat a wider range of genetic disorders.A terapia génica é uma das modalidades terapêuticas mais investigadas, tanto no contexto pré-clínico como clínico, e tem-se revelado eficaz no tratamento de uma diversificada gama de doenças. O vírus adeno-associado (AAV) é um vetor viral amplamente utilizado na terapia génica devido à sua capacidade de fornecer genes terapêuticos às células-alvo de forma segura e eficiente. Apesar do seu sucesso, a eficácia do AAV pode ser melhorada através de modificações na sua cápside, o invólucro proteico que protege o genoma viral, melhorando a transdução, alterando o tropismo e reduzindo as respostas imunitárias. A engenharia da cápside, em particular a inserção de proteínas , continua a ser uma área de investigação ativa para expandir ainda mais o potencial terapêutico do vetor AAV. Esta técnica requer a separação das proteínas da cápside viral (VP1, VP2 e VP3) para permitir a inserção precisa de proteínas na cápside do AAV. Apesar de estudos anteriores, ainda há necessidade de compreender como estas inserções afetam quer a funcionalidade destas proteínas com mais de 20 kDa quer a funcionalidade da capsíde do vetor.
Neste estudo, foi testada a inserção de uma proteína mCherry (~28kDa) em três regiões diferentes da cápside do vetor AAV2 (resíduos 453 e 587 na VP1, e o resíduo N-terminal 138 na VP2), usando quatro construções de mosaico da cápside. As construções foram validadas com êxito por transfecção transiente. Observou-se que a mcherry na cápside apresentava uma diminuição de intensidade 15 a 148 vezes nas células positivas para mCherry, em comparação com o controlo (mCherry livre). Em seguida, os vetores mosaicos AAV2/mCherry foram produzidos e caracterizados através da avaliação das suas partículas totais, genoma viral e títulos de transdução. Não foram observadas perdas significativas nas partículas totais e nas partículas do genoma viral por mL em comparação com o controlo do vetor WT AAV2. No entanto, nas unidades de transdução, verificou-se uma diminuição mais proeminente dos títulos em comparação com o controlo. Adicionalmente, os efeitos da inserção de mCherry no desempenho da cápside do vetor AAV2 foram avaliados in vitro. Na inserção da VP1 no resíduo 453, foi observado que o pico de percentagem de células mCherry positivas ocorreu às 48 horas com quase 20% de células positivas para mCherry. Na VP2, os resultados apresentaram o valor mais alto às 24 horas, com cerca de 75% das células positivas para mCherry mantendo o sinal às 48 horas. No geral, isto indicou uma diferença de 4 vezes no sinal mCherry entre os locais de inserção em VP1 e VP2.
Estes resultados permitiram comparar três locais diferentes de inserção na cápside e o seu impacto na funcionalidade da proteína heteróloga e no vetor AAV2, indicando que o melhor local para a inserção de proteínas ≥20kDa é o resíduo 138, seguido do resíduo 453. Este trabalho estabelece as bases para o desenvolvimento de vetores de AAVs mais eficazes, fazendo avançar a terapia génica e expandindo o seu potencial para tratar uma gama mais vasta de doenças genéticas
The Influences of Trust and AI Ethics on AI Use Acceptance in Politics: A Public Perception Survey

the context of politics. The research addresses the growing concern about AI's impact on
decision-making processes and democratic politics, filling a gap in understanding how the
public views AI's role in these areas. It explores the influence of AI trust and explainable AI on
acceptance of AI use and tests the Attitudes towards AI scale against it. Furthermore, the
influence of AI ethics on AI trust is included. Using a cross-sectional research design, an online
questionnaire survey is analyzed using the PLS-SEM method to find statistical relationships.
Trust and positive attitudes towards AI are found to be the most important factors influencing
the acceptance of AI use in politics, while AI ethics influence trust in AI. Therefore, the study
suggests actions for policymakers and politicians to foster trust in AI technologies through
transparent communication, clear privacy policies, and continuous education on the topic,
leading to more understandable and transparent AI systems. It also shows that the public
trusts politicians more when AI is used for simple and administrative tasks than for decisionmaking tasks. According to the findings, the public should engage in discussions around the
topic to strengthen AI democracy and to guide where AI should and should not be used in
politics. Further research should focus on different areas of AI, such as generative AI, robotics,
or expert systems, in order to gain deeper insights. On the other hand, the topic in general,
but especially AI ethics in politics, should be studied longitudinally to understand the rapidly
evolving developments in the topic
How can the customer journey of b lab be improved with the implementation of engagement initiatives
The proliferation of sustainability has been verified in recent years, with its corporate adoption
matching the cultural phenomenon. This paper consolidates the results of theoretical research,
primary data from interviews and a survey, and documents provided by B Lab to study the
Customer Journey of the NGO. Following a thorough customer pain point analysis, initiatives
were proposed in each stage of the Customer Journey to improve customer engagement.

The Impact of Email Marketing Automation on Customer Engagement and Retention: A Case Study of Adobe Campaign Classic

automation tool used by businesses worldwide with many advantages and possibilities to
improve specially with the new technologies coming up day per day. This study investigates
the impact of email marketing automation on customer engagement and retention, with a
focus on campaigns executed through Adobe Campaign. By analyzing metrics from loyaltyfocused, new member onboarding, subscription-based, and reactivation campaigns, the study
demonstrates the effectiveness of automated email strategies. Notably, the high engagement
rates and a 0% opt-out rate across all campaigns underscore the platform's ability to maintain
customer interest and foster loyalty. Semi-structured interviews conducted with marketing
professionals reveal both the strengths and challenges of using Adobe Campaign, highlighting
its effective personalization capabilities and technical difficulties. While the findings affirm the
positive impact of email marketing automation, they also point to areas for improvement,
particularly in technical support and user experience. The study's limitations include a
restricted data range, potential subjective biases in qualitative analysis, and unaccounted
external variables. Future research should aim to address these limitations to provide a more
comprehensive understanding of email marketing automation's role in customer engagement

Novel Affinity-Based Technologies for Targeting Distinct Biological Systems
O desenvolvimento de estratégias de purificação baseadas em afinidade foi impulsionado pelas vantagens teóricas que esta modalidade oferece para o bioprocessamento de PMTAs (produtos medicinais de terapia avançada), podendo resolver problemas atuais da indústria. A cromatografia de afinidade permitiu que a bioprodução de anticorpos monoclonais (mAbs) atingisse novos patamares, provocando a indústria a desenvolver estas tecnologias para outras biomoléculas, como vetores virais. O foco deste trabalho foi a caracterização/otimização de duas estratégias baseadas em afinidade para a purificação de diferentes biomoléculas, mAbs e vetores lentivirais (LVs). A primeira estratégia, focada em cromatografia, visou o uso de diferentes ligandos de afinidade (Z6Alk-Cys e Petasis-Ugi), imobilizados em diferentes tipos de meios cromatográficos. O ligando com melhor desempenho foi o Z6Alk-Cys imobilizado em esferas de agarose, onde uma recuperação de 83% (rendimento total de 31%) de IgG adsorvido foi obtida. O mesmo ligando foi imobilizado em membranas, porém, nenhuma recuperação foi conseguida. A outra estratégia foi um método não cromatográfico de purificação de LV, baseado em separação de fase líquida através de ultracentrifugação ou filtração de fluxo tangencial (TFF). A separação ocorre devido à presença de um copolipeptídeo termorresponsivo com afinidade para este alvo. Dois lotes desta solução de copolipeptídeo foram avaliados. Uma taxa de recuperação de TU (unidades de transdução) de 100% foi alcançada em escala de bancada e à escala de 200 mL (TFF), um rendimento total de 49,0 ± 25,5% foi obtido. A necessidade de um entendimento mais profundo da termorresponsividade deste copolipeptídeo ficou clara antes de prosseguir com o aumento de escala, tendo esta sido conseguida através de DLS e SEC-HPLC, cujos resultados também estão apresentados. O foco deste trabalho foram tecnologias promissoras baseadas em afinidade e também as adversidades enfrentadas durante o seu desenvolvimento, mostrando que otimização ainda é necessária para seu potencial uso comercial.The development of affinity-based purification strategies was supported by the theoretical advantages this modality poses for ATMPs (advanced therapeutic medicinal products) bioprocessing, which could tackle current industry problems. Affinity chromatography allowed the bioproduction of monoclonal antibodies (mAbs) to reach new heights and the industry has moved towards developing these technologies for other biomolecules, such as viral vectors. This work was focused on process optimization and technology characterization for two affinity-based strategies for different biomolecules, mAbs and lentiviral vectors (LVs). The first strategy was chromatography-focused, using two different affinity ligands (Z6Alk-Cys and Petasis-Ugi) for mAbs, immobilized in different types of chromatographic media. The best performing ligand was Z6Alk-Cys immobilized in agarose beads, where a bound IgG recovery rate of 83% (overall yield of 31%) was obtained. The same ligand was immobilized in novel membrane constructs based on fibers electrospun in a polyamide mesh, however, no recovery was obtained. The other strategy was a non-chromatographic LV purification method, based on liquid phase separation through ultracentrifugation or tangential flow filtration (TFF). Separation occurs due to the presence of a thermo-responsive copolypeptide with affinity to this target. Two batches of this copolypeptide solution were evaluated. A TU (transducing unit) recovery rate of 100% was achieved at benchtop scale and at 200 mL TFF scale, an overall yield of 49 ± 25.5% was obtained. The need for deeper understanding of this copolypeptide thermo-responsiveness was made clear before proceeding with scale-up, this was addressed by DLS and SEC-HPLC analysis which results are also reported. This work highlights promising affinity-based technologies and the adversities faced during their development, showing optimization is still needed for their potential commercial use
Fotogrametria e SIG como contributos à potencialização de Métodos para Aquisição de Dados em Pedreiras de Rocha Ornamental: O Estudo de Caso da Solancis
O presente trabalho centra-se no desenvolvimento de uma metodologia que possibilita a otimização das tarefas relacionadas com a aquisição de dados e com a partilha dos mesmos, no que toca aos blocos de rocha ornamental, produzidos pelas empresas extrativas. A Solancis - Sociedade Exploradora de Pedreiras SA, permitiu o trabalho de campo e a recolha de informação nas suas pedreiras, possibilitando a elaboração deste projeto. Para a realização da metodologia, foram determinadas várias etapas: a criação de um formulário digital, que permite recolher dados alfanuméricos e fotográficos sobre as caraterísticas dos blocos; a elaboração de um método de levantamento fotogramétrico com recurso a smartphone; o desenvolvimento de um método de processamento das imagens, de forma a gerar modelos 3D de blocos; a criação de ficheiros de vídeo, que permitam a partilha destes modelos; a elaboração de tutoriais para a realização da recolha de dados, destinada aos colaboradores da Solancis. Deste modo, procura-se impulsionar o Setor da Pedra Natural com o desenvolvimento de métodos tecnológicos e produtos digitais inovadores, alinhando os objetivos do projeto com a agenda Sustainable Stone by Portugal, que visa potenciar a eficiência da indústria extrativa, assim como promover a sustentabilidade.This work focuses on the development of a methodology that makes it possible to optimize the tasks related to data acquisition and sharing, regarding ornamental stone blocks produced by extractive companies. Solancis - Sociedade Exploradora de Pedreiras SA, allowed fieldwork and the collection of information in its quarries, making it possible to prepare this project. In order to carry out the methodology, several stages were determined: the creation of a digital form, which allows the collection of alphanumeric and photographic data on the characteristics of the blocks; the development of a photogrammetric survey method using a smartphone; the development of an image processing method, in order to generate 3D models of the blocks; the creation of video files, which allow these models to be shared; the development of tutorials for carrying out data collection, aimed at Solancis employees. The aim of this project is to boost the natural stone sector by developing innovative technological methods and digital products, aligning the project's objectives with the Sustainable Stone by Portugal agenda, which aims to boost the efficiency of the extractive industry and promote sustainability
Zebrafish avatars to forecast hormone therapy response in breast cancer
Breast cancer (BC) stands as one of the most prevalent cancers and a leading cause of mortality among women. Approximately 70% of all BC cases are characterized as hormone-receptor-positive (HR+). For these patients, treatment mainly comprises endocrine therapies (ET), inhibiting estrogen signaling by blocking receptors or hindering estrogen production [84, 85, 86]. Consequently, the growth and spread of hormone-sensitive cancer cells become compromised. The use of ET has significantly reduced BC-related mortality. However, despite its effectiveness, 15-20% of tumors are intrinsically resistant to treatment [87] and 30-50% eventually develop resistance [88]. Moreover, these cancers can lie dormant, and metastasis may emerge many years after diagnosis [88].
Thus, a test that could anticipate which patients are resistant to ET could help guide treatment decisions, sparing patients from unnecessary toxicities.
My host Lab has been developing the zebrafish xenograft model (zAvatars), as a quick in-vivo screening platform for drug testing and personalized therapy. zAvatars are generated by injecting human tumor cells obtained from surgery or biopsies, into zebrafish embryos [89, 90, 91]. zAvatars offer speed (10 days assay), cellular resolution, and the ability to perform numerous transplants simultaneously. Most importantly, they can be used to detect changes in cell mortality, proliferation rate, and even some cancer hallmarks like angiogenesis and metastasis formation.
Our laboratory has shown that the zebrafish xenograft model has resolution to distinguish sensitivity/resistance towards multiple chemotherapies in the context of different cancer types. However, it is important to note that most of these therapies targeted molecular pathways within the tumor cells (i.e. cell-autonomous). While this is the case for two ETs, Tamoxifen and Fulvestrant, Letrozole on the other hand also acts systemically, by inhibiting the production of estrogen not only by tumor cells but also throughout the patient’s tissues.
The primary goal of this project was to validate zebrafish xenografts for BC within the context of ET.
We started by developing, characterizing, and optimizing our model of HR+ BC zebrafish xenografts. These xenografts were generated with the T47-D cell line, a Luminal A type, HR+ BC cell line, therefore being representative of the most common type of BC. We found that these cells implanted more efficiently in the brain than in the PVS, the most usual injection site.
By treating T47-D xenografts with these 3 ETs, we found that all ETs were able to significantly induce Act.Caspase-3 expression, a marker for apoptosis. We also registered a decrease in cell proliferation upon Letrozole administration.
Subsequently, we were also able to generate patient-derived xenografts or zAvatars with a sample from a stage IV metastatic BC patient with the Luminal B subtype. The zAvatars were sensitive to the 3 ETs. Overall, my work suggests that the zebrafish xenograft model has the resolution necessary to screen sensitivity to ET, including Letrozole, as long as the cancer cells tested express the aromatase-coding gene CYP19A1.O cancro da mama (BC) é um dos tipos de cancro mais prevalentes e uma das principais causas de mortalidade entre as mulheres. Cerca de 70% de todos os casos de BC são classificados como positivos para recetores hormonais (HR+). Para estes pacientes, o tratamento consiste principalmente em terapias endócrinas (ET), que inibem a sinalização de estrogénio ao bloquear os seus recetores ou ao impedir a produção do mesmo [84, 85, 86]. Consequentemente, o crescimento e a disseminação das células cancerígenas sensíveis a hormonas ficam comprometidos. O uso de ET reduziu significativamente a mortalidade relacionada ao BC. No entanto, apesar da sua eficácia, 15-20% dos tumores são intrinsecamente resistentes ao tratamento [87] e 30-50% eventualmente desenvolvem resistência [88]. Além disso, estes cancros podem permanecer dormentes, e podem surgir metástases muitos anos após o diagnóstico inicial [88].
Consequentemente, um teste capaz de antecipar quais os pacientes resistentes a ET, poderia ajudar na decisão de tratamento, poupando os pacientes de toxicidade desnecessária.
O nosso laboratório tem vindo a desenvolver o modelo de xenógrafos de peixe-zebra (zAvatars), com o intuito de desenvolver uma terapia personalizada para cada doente. Os zAvatars são gerados através da injeção de células tumorais humanas, obtidas a partir de cirurgias ou biópsias, em embriões de peixe-zebra, que receberão posteriormente as mesmas opções terapêuticas que o paciente [89, 90, 91]. Os zAvatars oferecem rapidez (ensaio de 10 dias), resolução celular e a capacidade de realizar múltiplos transplantes simultaneamente. A sua principal vantagem é que podem ser usados para detetar alterações na mortalidade celular, na taxa de proliferação e até mesmo em “hallmarks” do cancro, como a angiogénese e a formação de metástases.
O nosso laboratório demonstrou que este modelo tem resolução necessária para distinguir sensibilidade/resistência a múltiplas quimioterapias no contexto de diferentes tipos de cancro. No entanto, é importante ter em conta que a maioria dessas terapias visa atacar as vias moleculares dentro das células tumorais (ou seja, vias célula-autónomas). Embora este seja o caso de dois tratamentos endócrinos, Tamoxifeno e Fulvestranto, o Letrozol, por outro lado, também atua sistemicamente, inibindo a produção de estrogénio, não apenas pelas células tumorais, mas também em todos os tecidos do paciente.
O principal objectivo deste projeto foi validar os xenógrafos de peixe-zebra para BC no contexto de ET.
Começámos por desenvolver, caracterizar e otimizar o nosso modelo de xenógrafos de BC HR+ em peixe-zebra. Estes xenoenxertos foram gerados com a linha celular T47-D, uma linha celular HR+ de BC do subtipo Luminar A, sendo, portanto, representativa do tipo mais comum de BC. Descobrimos que estas células implantam mais eficientemente no cérebro do que no PVS, o local de injeção mais comum.
Ao tratar o xenógrafos de T47-D com estas três ET, descobrimos que todas as ET eram capazes de induzir significantemente um aumento na expressão de Act.Caspase-3, um marcador de apoptose. Também fomos capazes de observar uma diminuição na proliferação celular após a administração de Letrozol.
Subsequentemente, conseguimos ainda gerar um xenógrafo a partir de uma amostra de um paciente, um zAvatar, a partir de uma amostra de fluido ascítico de um paciente com BC metastático no estadio IV, do subtipo Luminal B. O meu trabalho sugere que este modelo de xenógrafos de peixe-zebra tem a resolução necessária para triar sensibilidade a ET, incluindo letrozol, desde que as células cancerígenas expressem o gene que codifica a aromatase, CYP19A1
