Antifouling, antibiofilm and ecotoxicological evaluation of paints with marine actinomycetes extracts
Marine biofouling has posed challenges to the nautical industry and all the industries that have underwater infrastructures, contributing to negative environmental impacts. Currently, main mitigation strategies of this phenomenon include antifouling methods with biocidal compounds; however, their toxicity has inspired a search for sustainable, cost-effective and environmentally safe alternatives to combat this phenome-non. Natural compounds from marine organisms such as actinomycetes represent a promising source for environmentally friendly antifouling compounds. This study explored the antifouling and antibiofilm potential of crude extracts derived from actinomycetes containing secondary metabolites with antifouling activity as additives to nautical paints. Exposure bioassays on model marine organisms were conducted to evaluate the antifouling capacity of the extracts containing napyradiomycins or marinones when incorporated in a biocide free marine paint. In addition, the antibiofilm capacity of the paints with the extracts was also evaluated. The paints containing the extracts in their composition demonstrated good antifouling power, as well as the ability to inhibit bio-film formation, thus being strong candidates for use as agents to combat both marine microfouling and macrofouling. Biochemical assays on oxidative stress biomarkers (including GST, SOD, CAT, TAC and lipid peroxidation) further suggested napyradiomycin-based and marinone-based paints as eco-friendly products, being marinone-derived paint safer for marine species compared to the paint containing napyradiomycins.A bioincrustação marinha tem colocado desafios à indústria náutica e a todas as indústrias que possuem infraestruturas subaquáticas, contribuindo para impactos ambientais negativos. Atualmente, as principais estratégias de mitigação deste fenómeno incluem métodos anti-incrustantes com compostos biocidas; no entanto, a sua toxicidade inspirou a procura de alternativas sustentáveis, económicas e ambientalmente seguras para combater este fenómeno. Compostos naturais de organismos marinhos, como as actinomicetas, representam uma fonte promissora de compostos anti-incrustantes ecológicos. Este estudo explorou o potencial anti-incrustante e antibiofilme de extratos brutos derivados de actinomicetas que produzem metabolitos secundários com atividade anti-incrustante como aditivos em tintas náuticas. Foram realizados bioensaios de exposição em organismos marinhos modelo para avaliar a capacidade anti-incrustante dos extratos contendo napiradiomicinas ou marinonas quando incorporados numa tinta marinha isenta de biocida. Além disso, foi também avaliada a capacidade antibiofilme das tintas com os extratos. As tintas contendo os extratos na sua composição demonstraram um bom poder anti-incrustante, bem como capacidade de inibir a formação de biofilme, sendo assim fortes candidatas para utilização como agentes de combate tanto à micro- como à macroincrustação marinha. Os ensaios bioquímicos de biomarcadores de stress oxidativo (incluindo GST, SOD, CAT, TAC e peroxidação lipídica) sugeriram ainda as tintas à base de napiradiomicinas e à base de marinonas como produtos amigos do ambiente, sendo a tinta derivada de marinonas mais segura para as espécies marinhas em comparação com a tinta contendo napiradiomicinas
Troubleshooting do Transporte Pneumático de pó de PEAD
The Pneumatic Powder Conveying System in section 80 of the HDPE plant was the Pneumatic Powder Conveying System in section 80 of the high-density polyethylene (HDPE) plant was refurbished between 1990 and 1991 to transport up to 20 t/h. The powder is carried from the drying hopper (Purge Bin H-3081) to Silos H-8001/2 using nitrogen in a closed circuit, with automatic oxygen and hydrocarbon control. Transport is carried out by C-8001 Blowers, and there are filters to protect the compressors. Finally, there is an exchanger in the system to prevent overheating.
The aim of this work is to study the root causes that can lead to the plant being shut down or to a reduction in operating flow. the “5 Whys?” quality method was used. The novelty of this work lies in the fact that it is an exhaustive analysis identifying root causes and proposing solutions with an economic analysis of each one, something that has never been done for the company's pneumatic conveying system.
Out of a total of twenty-four potential root causes, six were identified for the problem highlighted: No level in the Purge Bin; accumulation of polymer “webs” / “splinters”; the filters in the S-8012 Bag Filter are damaged; the flow interval is lower than recommended; ineffective washing of the E-8004 exchanger; lack of an exchanger upstream of the C-8001 Blowers. Seven proposals for improvement were put forward, three of which are of no cost to the company and can be implemented immediately: operating with the level in the Purge Bin; regularized inspection of the filters in the S-8012 Bag Filter; regularized checking of the nitrogen injection flow rate in the lubrication system of the V-8001 Rotary Valve. Of all the proposals for improvement, four require investment on the part of the company: chemical cleaning; recommissioning of the E-8003 exchanger; purchase of a flanged exchanger to replace the E-8003; round vibrating screen. Each of the recommendations has a benefit for the company of around 1 million euros per year.
In addition, laboratory tests were carried out to see if there was any relationship between the grids and the pneumatic conveying failures.O Sistema de transporte Pneumático de Pó da secção 80 da fábrica de polietileno de alta densidade (PEAD) foi reformado entre 1990 e 1991 para transportar até 20 t/h. O pó é transportado da tremonha de secagem (Purge Bin H-3081) para os Silos H-8001/2 utilizando azoto em circuito fechado, com controlo de oxigénio e hidrocarbonetos em automático. O transporte é feito pelos Blowers C-8001, e há filtros para proteger os compressores. Por último, existe um permutador no sistema para evitar o superaquecimento.
Este trabalho tem como objetivo estudar quais são as causas-raiz que podem levar à sua paragem ou à redução de caudal de operação. Assim, foi utilizada a ferramenta da qualidade denominada como “5 Porquês?”. A novidade deste trabalho remete para o facto de se tratar de uma análise exaustiva com identificação de causas-raiz e propostas de soluções com análise económica de cada uma, algo que nunca foi feito para o transporte pneumático da empresa.
De um total de vinte quatro potenciais causas-raiz foram identificadas seis para o problema evidenciado: Inexistência de nível no Purge Bin; acumulação de “teias” / “farripas” de polímero; os filtros do Filtro de Sacos S-8012 estão danificados; intervalo do caudal abaixo do recomendado; lavagem ineficaz do permutador E-8004; inexistência de um permutador a montante dos Blowers C-8001. Foram apresentadas sete propostas de melhoria, sendo que três não apresentam qualquer custo para a empresa e podem ser implementadas imediatamente, sendo estas: operar com nível no Purge Bin; inspeção aos filtros do Filtro de Sacos S-8012 regularizada; verificação regularizada do caudal de injeção de azoto no sistema de lubrificação da Válvula Rotativa V-8001. De todas as propostas de melhoria, quatro requerem investimento por parte da empresa: limpeza química; recolocação em serviço permutador E-8003; compra de um permutador flangeado de forma a substituir o E-8003; peneiro vibratório redondo. Cada uma das recomendações tem um benefício para a empresa de cerca de 1 milhão de euros ao ano.
De forma complementar foram realizados ensaios no laboratório de modo a tentar perceber se existia alguma relação entre os grades com as falhas do transporte pneumático
Galerie Pedre Daupias. Uma coleção de pintura em Lisboa
Pedro Eugénio Daupias (1818-1900), 1.º visconde e 1.º conde de Daupias, foi um dos mais importantes colecionadores em Portugal no último quartel do século XIX. Das várias coleções que reuniu, a de pintura, pela excelência de algumas peças, é a que ganha maior destaque para a História da Arte, estando alinhada com o gosto parisiense finissecular, embora as suas tendências ecléticas o tivessem levado a reunir todo o tipo de objetos artísticos: escultura, cerâmica, têxteis, mobiliário, ourivesaria, entre outros. A dispersão da coleção começou em 1892, quando uma seleção das melhores pinturas foi à praça em Paris. Em 1894, realizou-se um leilão dedicado às artes decorativas, novamente na capital francesa. E, dez anos após a morte do colecionador, em 1910, foi vendido num leilão em Lisboa o remanescente da coleção. O catálogo que o conde fez publicar em 1882, dedicado à pintura e escultura modernas da sua coleção, é um instrumento essencial para o seu conhecimento, tendo sido uma das bases mais importantes do presente trabalho.The French-Portuguese Pedro Eugénio Daupias (1818-1900), 1st Viscount and 1st Count Daupias, was one of the most important collectors in Portugal in the last quarter of the 19th century. Of the collections he gathered, the one on painting, for the excellent quality of some of its pieces, is the one that gets the attention from art historians, being aligned with the Parisian taste, even if his eclectic taste led him to collect all kinds of artistic objects: sculpture, ceramics, textiles, furniture, silverware and miscellaneous. In 1882, the Count published a catalogue dedicated to modern painting and sculpture. The dispersion of his collection began in 1892, when a selection of the best paintings was auctioned in Paris. In 1894, there was another auction dedicated to the decorative arts, again at the French capital. And ten years after the collector's death, in 1910, the remains of his collection were sold at yet another auction in Lisbon
Regulation of Oxidative Stress Response in Streptococcus pneumoniae : The Protective Role of RNase R
Streptococcus pneumoniae is a Gram-positive, aerotolerant opportunistic pathogen, recognised on the WHO’s priority list as the leading agent causing bacterial pneumonia. Our immune system responds by mobilising phagocytes that induce oxidative burst, generating reactive oxygen species (mainly H2O2, O2- and HOCl). Remarkably, this bacterium produces H2O2 and uses it as a competitive advantage against other pathogens. Paradoxically, S. pneumoniae lacks the main detoxifying enzymes, raising the question of how it handles oxidative stress. Previous results from our laboratory highlighted a key role of the exoribonuclease RNase R, in resistance to H2O2.
RNase R is a solo member of the RNB family in S. pneumoniae and with the distinctive capability of degrading secondary RNA structures, suggesting a vital role in its metabolism. This work aims to unravel how RNase R promotes the survival of the pneumococcus under oxidative stress.
Throughout this work, three pneumococcal strains were used, TIGR4 wild type, the isogenic mutant lacking RNase R (Δrnr), and the complemented strain expressing RNase R in trans (Δrnr+R). Beyond revisiting H2O2 effects, we performed susceptibility and viability assays to evaluate the effect of RNase R on the resistance to the other main oxidising agents of the oxidative burst, O2- and HOCl. On the other hand, to identify H2O2-induced genes and small non-coding RNAs (ncRNAs), RNA-sequencing was performed under H2O2 stress. The best candidates were experimentally validated via RT-qPCR and Northern blot.
The results suggest that the strain lacking RNase R behaves differently depending on the stress challenge presented. The enzyme is critical in the resistance to hydrogen peroxide but does not seem to deeply affect S. pneumoniae resistance against superoxide anion and hypochlorous acid-induced stress. Moreover, we also observed higher lipid peroxidation in Δrnr under H2O2-induced stress, which is probably a result of our previous discovery that RNase R has an effect on membrane lipids.
Under H2O2 oxidative stress, the genomic landscape was altered in Δrnr as compared to the WT, with 104 transcripts down- and 95 up-regulated in the mutant strain. Key affected genes include ABC transporters (piuBCDA, piaBA, mntE), regulatory elements (ciaR, glnR), DNA-binding proteins (yaaA), and enzymes (SP_1911, nanA). 19 transcripts were selected for validation, among which 17 had its expression variation confirmed by RT-qPCR and 5 were further studied by Northern blot. Globally, 58 ncRNAs were differentially expressed in the Δrnr strain under stress, 10 of which were identified for the first time in this study.
Overall, our results offer new insights into the RNase R regulatory network and suggest that RNase R affects the levels of several transcripts with pivotal roles in the pneumococcus oxidative response. This suggests that RNase R might be a key enzyme in the complex and integrated network of mechanisms of the oxidative stress response, and we are only starting to uncover its role.Streptococcus pneumoniae é um agente patogénico Gram-positivo, aerotolerante, sendo o maior agente causador de pneumonia bacteriana e por isso reconhecido pela OMS como pertencente à lista dos agentes prioritários. O sistema imune responde-lhe mobilizando fagócitos que induzem o chamado oxidative burst, onde são geradas espécies reativas de oxigénio (principalmente H2O2, O2- e HOCl). Curiosamente pneumococos é capaz de produzir H2O2 e usa-o como uma vantagem competitiva contra outros patogénicos. Paradoxalmente, S. pneumoniae não apresenta os principais enzimas responsáveis pela destoxificação celular, levantando a questão de como é capaz de lidar com o stress oxidativo. Trabalhos anteriores do nosso laboratório destacaram um papel vital da exoribonuclease RNase R na resistência ao H2O2. A RNase R é o único representante da família RNB em S. pneumoniae e apresenta a capacidade distinta de degradar RNA em cadeia dupla, indicando um papel crucial no metabolismo. Este trabalho tem como objetivo desvendar como é que RNase R promove a sobrevivência dos pneumococos durante o stress oxidativo.
Ao longo deste trabalho foram usadas três estirpes de pneumococos: a estirpe selvagem TIGR4, o mutante isogénico sem a RNase R (Δrnr) e a estirpe complementar que expressa a RNase R em trans (Δrnr+R). Realizámos ensaios de suscetibilidade e viabilidade para determinar o efeito da ausência da RNase R na resistência aos outros principais agentes oxidantes do oxidative burst, o O2- e o HOCl. Em paralelo foi realizada sequenciação de RNA para identificar todos genes e pequenos RNAs não codificantes (ncRNAs) induzidos pelo H2O2. Os melhores candidatos foram validados experimentalmente por RT-qPCR e Northern blot.
Os resultados sugerem que a estirpe sem RNase R apresenta comportamentos distintos dependendo do tipo de stress a que a bactéria é exposta. Este enzima apresenta um papel vital na resistência ao H2O2 mas não parece afetar muito a resistência ao O2- e ao HOCl. Além disso conseguimos observar uma maior peroxidação lipídica no Δrnr, o que pode ser um reflexo do efeito do enzima na membrana lipídica.
Sob stress induzido por H2O2, o panorama genómico está alterado no Δrnr em comparação com o WT, com 104 transcritos sub- e 95 sobre-expressos, no Δrnr. Alguns dos principais genes afetados incluem os transportadores ABC (piuBCDA, piaBA, mntE), elementos regulatórios (ciaR, glnR), proteínas de ligação ao DNA (yaaA) e enzimas (SP_1911, nanA). 19 transcritos foram selecionados para validação experimental, 17 dos quais foram confirmados por RT-qPCR e 5 foram estudados em mais detalhe por Northern blot. 58 ncRNAs estão diferencialmente expressos e 10 deles foram identificados pela primeira vez neste estudo.
Em suma, os nossos resultados revelam novos elementos na rede de regulação da RNase R, sugerindo que esta afeta os níveis de expressão de vários genes com papeis fulcrais na resposta oxidativa em pneumococos. Tal parece indicar que a RNase R é um enzima chave na rede complexa e integrada de mecanismos de resposta ao stress oxidativo, que apenas agora começamos a desvendar
Understanding a new respiratory route in Geobacter: characterization of the inner membrane associated cytochrome CbcS
Geobacter species are recognized for their unique metabolic capabilities, especially their ability to use various extracellular compounds, such as insoluble metals, as electron acceptors in the process of extracellular electron transfer (EET). Among these, Geobacter sulfurreducens has been the most thoroughly investigated. Studies using gene knockout and proteomics have identified essential proteins involved in EET mechanisms. A crucial step in this process involves electron transfer from the menaquinone pool to the quinone-cytochrome c oxidoreductase complexes located in the inner membrane, which then assist the transfer to their redox partners in the periplasm. G. sulfurreducens genome codes for six inner membrane oxidoreductase gene clusters, three of which - CbcL, ImcH and CbcBA – have been already associated to specific metabolic pathways.
This thesis focused on cloning, expressing, purifying and biochemical characterizing CbcS, a multiheme cytochrome c from the Cbc4 complex, a recently identified quinone-cytochrome c oxidoreductase complex. The tetraheme periplasmic domain of CbcS was cloned into four different constructs and heterologously expressed in E. coli. The purification of CbcS was optimized and several techniques were used to explore its structural and functional properties. Potentiometric redox titrations were conducted to determine the reduction potential of the protein at pH 7 and pH 8, while Nuclear Magnetic Resonance (NMR) was employed to analyze the spin state of the heme groups and to confirm the hemecore arrangement predicted by AlphaFold. In the context of the EET pathways of G. sulfurreducens, the electron transfer between CbcS and PpcA was monitored by NMR.
The results obtained in this thesis offer new insights towards a new respiratory route in Geobacter.A espécie Geobacter é reconhecida pelas suas capacidades metabólicas únicas, especialmente na capacidade que tem em utilizar diversos compostos presentes no exterior da célula, tais como metais insolúveis, como aceitadores de eletrões no processo de transferência de eletrões extracelular (TEE). Dentro desta espécie, Geobacter sulfurreducens é a que tem sido mais extensivamente estudada. Diversos estudos de deleção de genes e proteómica identificaram as proteínas essenciais que estão envolvidas no processo de TEE. Uma etapa crucial deste processo envolve a transferência de eletrões das menaquinonas para os complexos de oxidorredutases quinona-citocromo c localizados na membrana interna, que depois auxiliam a transferência para os seus parceiros redox no periplasma. O genoma de G. sulfurreducens codifica para seis grupos de genes de oxidorredutases da membrana interna, três dos quais – CbcL, ImcH e CbcBA – foram já associados a vias metabólicas especificas.
Esta tese focou-se na clonagem, expressão, purificação e caracterização bioquímica do CbcS, um citocromo c multihémico que pertence ao complexo Cbc4, um complexo de oxidorredutases quinona-citocromo c recentemente identificado. O domínio periplasmático tetrahémico do CbcS foi clonado de quatro formas diferentes e expresso heterologamente em E. coli. As condições de purificação do CbcS foram otimizadas e diversas técnicas foram utilizadas para explorar as suas propriedades estruturais e funcionais. Foram efetuadas titulações potenciométricas para determinar o potencial de redução da proteína a pH 7 e a pH 8. A técnica de Ressonância Magnética Nuclear (RMN) foi utilizada para analisar o estado de spin dos hemos e para confirmar o arranjo tridimensional previsto pelo modelo AlphaFold. No contexto das vias de TEE em G. sulfurreducens a transferência de eletrões entre CbcS e PpcA foi monitorizada por RMN.
Os resultados obtidos nesta tese oferecem novas perspetivas sobre uma nova via respiratória na espécie Geobacter
Otimização da recuperação de hexano no processo de produção do PEAD
A fábrica de PEAD da Repsol Polímeros (Sines) utiliza hexano como o solvente responsável pela formação da suspensão no reator, tendo depois uma secção de recuperação destinada a este composto. O presente trabalho foi desenvolvido no âmbito da otimização da recuperação de hexano.
Observou-se que os locais que representam maior perda de solvente são os condensadores de compostos voláteis orgânicos (VOC's), tendo-se procedido à simulação de seis condensadores no software Aspen HYSYS V12.1. As simulações foram primeiramente calibradas utilizando os dados de design dos respetivos equipamentos, podendo depois ser aplicadas a casos concretos a fim de proceder à sua verificação. Para esta análise foram considerados os três grades mais produzidos anualmente, tendo-se realizado uma simulação por grade e por equipamento. Concluiu-se que o condensador E 7008 é aquele onde se obtém uma maior recuperação e o E 3007 é o que apresenta menor recuperação, sendo o responsável por 58% das perdas atuais.
Foi possível realizar análises de sensibilidade às variáveis que mais influenciam a recuperação de hexano: o caudal de utilidade (brine) e a temperatura de entrada da utilidade. A par com as análises de recuperação de hexano realizaram-se também gráficos custo-benefício para auxiliar na elaboração das propostas de melhoria. Verificou-se que um aumento em, aproximadamente, 15% no caudal total de brine resulta numa recuperação adicional de 238 t/ano. A redução da temperatura de entrada do brine em 2ºC, resulta num aumento da recuperação em 96 t/ano.
A nível das alterações do caudal de circulação da utilidade nos equipamentos, a proposta realizada envolve um gasto adicional de 32 mil €/ano, com um ganho adicional de 323 mil €/ano. No que diz respeito à diminuição da temperatura do brine, a proposta realizada envolve um gasto adicional de 12 mil €/ano, com um ganho adicional de 125 mil €/ano, considerando preços médios. Aplicando as duas propostas, há um aumento na recuperação em 398 t/ano e obtém-se o lucro mais elevado para o caso médio e melhor caso. Considerando o pior caso, deve dar-se prioridade ao aumento do caudal de circulação de brine.The HDPE plant of Repsol Polímeros (Sines) uses hexane as the solvent responsible for the formation of the suspension in the reactor and has a recovery section dedicated to this compound. This work was carried out within the scope of optimizing hexane recovery.
It was observed that the locations representing the greatest solvent loss are the volatile organic compound (VOC) condensers. Consequently, six condensers were simulated using the Aspen HYSYS V12.1 software. The simulations were initially calibrated using the design data of the respective equipment, allowing them to later be applied to specific cases for verification. For this analysis, the three most annually produced grades were considered, with a simulation performed for each grade and each piece of equipment. It was concluded that condenser E 7008 yields the highest recovery, while E 3007 demonstrates the lowest recovery, being responsible for 58% of current losses.
Sensitivity analyses were performed on the variables that most influence hexane recovery: the utility flow rate (brine) and the inlet temperature of the utility. Alongside hexane recovery analyses, cost-benefit graphs were also created to aid in the development of improvement proposals. It was found that an approximate 15% increase in the total brine flow rate results in an additional recovery of 238 tons per year. A reduction of the brine inlet temperature by 2°C leads to an increase in recovery by 96 tons per year.
Regarding changes in the circulation flow rate of the utility in the equipment, the proposal involves an additional cost of €32,000 per year, with an additional gain of €323,000 per year. Concerning the reduction of brine temperature, the proposal entails an additional cost of €12,000 per year, with an additional gain of €125,000 per year, considering average prices. By applying both proposals, there is an increase in recovery of 398 tons per year, achieving the highest profit in both the average and best-case scenarios. In the worst-case scenario, priority should be given to increasing the circulation flow rate of the brine

Housing policies in Portugal have been extensively discussed in academia. This study draws
conclusions from the research question: Is the housing package "Mais Habitação" (Law n. º
71/XV/1.ª) intergenerationally fair? The CGF and SOIF toolkit, which analyses
intergenerational fairness of public policies, was employed in this study. The preliminary
findings indicate that the policy is probably intergenerationally fair. The research clarifies the
nuances of housing policy in Portugal and presents actionable recommendations, marking a
pioneering effort in housing policy evaluation

intentions, the influence on mothers' self-image, and the role of trust in online communities.
Using a survey of 260 new mothers analyzed through regression and mediation models, the
findings reveal that engagement with mom influencers significantly increases the intention to
purchase baby essentials. This effect is notably mediated by the trust established within
online mother communities, highlighting the pivotal role of these virtual support networks.
However, the direct impact on self-image was minimal, suggesting that while influencers
shape purchasing decisions, they do not significantly alter self-perception. These insights
underscore the importance of influencer credibility and community trust in shaping
consumer behavior. Practical implications for marketers include building authentic
relationships with mom influencers and fostering supportive online communities to target
new mothers effectively. The study also challenges assumptions about the role of self-image
in consumer behavior, suggesting it may vary by context

environmental awareness, and demand for eco-friendly services/products are putting
considerable stress on the energy sector to change from fossil fuels to renewable energies.
Nevertheless, this is impossible and impractical in light of the current demand for energy, and
we are thus obliged to find a solution in which we reduce our energy consumption or use
energy in a more sustainable way, or both. Smart home technologies enable governments,
companies, and individuals to achieve their goals, gain direct and indirect benefits regarding
energy savings, and consume resources more conscientiously. This study investigates the
variables that may affect individuals’ adoption of this type of technologies. We apply the
unified theory of acceptance and use of technology (UTAUT) model to create our research
model and add three new variables: perceived risks, perceived environmental benefits, and
digital literacy. We conducted a survey, obtaining 171 responses and analysed the data with
the partial least squares method (PLS), allowing us to verify the validity of our model. The
