    Development of a generic topical dosage form using Design of Experiments (DoE): optimizing formulation and process parameters for enhanced efficacy and quality

    Semi–solid dosage forms constitute a significant proportion of pharmaceutical dosage forms. They serve as a vehicle for drug delivery typically administered through the skin. Their unique properties allow them to adhere to the application surface and remain there for extended periods. As semi–solid formulations, ointments are primarily used to provide protective and emollient effects on the skin and deliver the desired treatment for specific conditions. For the development of generic topical products, it is mandatory to ensure therapeutic equivalence by matching the reference product both qualitatively (Q1, with the same components), and quantitatively (Q2, with the same components and concentrations). The objective of this study was to develop a generic topical product with a therapeutic equivalence to a commercialized ointment product, employing the Quality by Design (QbD) approach to guarantee the quality of the desired product target. The first step consisted of identifying the critical properties (emollient concentration) and their process parameters (mixing time and rate) concerning product development. Secondly, a screening test was conducted to determine the emollient concentration range (30–35%, w/w) for further studies. Then, a risk assessment was conducted to evaluate the most critical factors: emollient concentration, mixing rate, and time. A central composite design was then performed to analyze the effects of the independent variables. The characterization of the ointment formulations was carried out according to a guideline provided by the European Medicine Agency (EMA), assessing physical properties, including rheological behaviour, and product performance. The results of the three–factor studies demonstrated that emollient concentration was the critical parameter influencing the quality attributes of the design of experiments formulations. An optimization design was conducted using Design–Expert® 13 software to achieve the final formulations (FF) composition. Accordingly, six final formulations (FF 1– FF 6) were prepared comprising emollient concentrations between 30.000 and 34.054%, w/w, mixing rates of 200 to 360 rpm, and mixing times between 40 and 60 minutes. The final formulation (FF 1) of this study was then tested on a 0.5 kg laboratory scale, revealing discrepancies in product performance during the scaling process from 50 g to 0.5 kg. The findings demonstrate that the final formulation successfully achieved therapeutic equivalence to the reference product. The comparable outcomes suggest that the final formulation can potentially offer an alternative therapeutic option to the commercialized product.Formas farmacêuticas semi–sólidas constituem uma proporção significativa das formas farmacêuticas. Atuam como um veículo para o transporte de medicamentos que são, tipicamente, administrados através da pele. As suas propriedades únicas permitem que se mantenham na superfície de aplicação durante longos períodos. As pomadas são semi–sólidas usados, principalmente, para fornecer efeitos protetores e emolientes na pele. Para o desenvolvimento de um producto tópico genérico é obrigatório garantir a equivalência terapêutica qualitativa (Q1, com os mesmos componentes) e quantitativa (Q2, com os mesmos componentes e concentrações). O objetivo deste estudo foi desenvolver um produto tópico genérico com equivalência terapêutica a uma pomada comercializada, utilizando uma abordagem Quality by Design (QbD) para garantir a qualidade do produto desejado. O primeiro passo consistiu em identificar as propriedades críticas (concentração de emoliente) e os parâmetros de processos (velocidade e tempo de agitação) de relativos ao desenvolvimento do produto. De seguida, um teste preliminar foi conduzido de forma a determinar uma gama de concentrações de emoliente para estudos futuros. Posteriormente, foi realizada uma avaliação de risco para avaliar os fatores mais críticos: concentração de emoliente, velocidade e tempo de agitação. Um desenho composto central foi então realizado para analisar os efeitos das variáveis independentes. A caracterização das formulações de pomada foi realizada segundo uma diretriz fornecida pela Agência Europeia de Medicamentos (EMA), no qual as propriedades físicas, incluindo o comportamento reológico, e desempenho do produto foram avaliadas. Os resultados do estudo dos três fatores demonstraram que a concentração de emoliente foi o parâmetro mais crítico, influenciando, assim, a qualidade das formulações. Um desenho de otimização foi realizado usando o software Design–Expert® 13, para obter a composição das formulações finais (FF). Foram preparadas seis formulações finais (FF1– FF 6) com concentrações de emoliente entre 30.000 e 34.054% m/m, velocidades de agitação de 200 a 360 rpm, e tempo de agitação entre 40 e 60 minutos. A formulação final (FF 1) foi testada numa escala laboratorial de 0.5 kg, o que revelou discrepâncias no desempenho do produto, de 50g para 0.5 kg. Os resultados demonstram que a formulação final alcançou com sucesso equivalência terapêutica do produto de referência. Os resultados sugerem que a formulação final pode potencialmente oferecer uma opção alternativa ao produto comercializado

    Develop and Perform Experiments in a Spray Dryer Equipment in order to Improve Process Sustainability

    Hovione aims to reduce 40% of carbon emissions by 2030, and currently, all solvents used in the spray drying process are incinerated or go for treatment, and fresh solvents are used. To increase the process's sustainability, economy, and productivity, it is essential to reuse these solvents. However, the reutilization is particularly challenging with the use of binary mixtures, as it requires precise knowledge of the solvents ratio to adjust quantities for their use in new batches in production. This is the primary objective of this work: to develop a strategy for re-adjust the quantities of binary mixtures in production without the need of laboratory tests. The composition of the mixtures can be obtained from their density and temperature, variables easily measured by mass flow meters in production. In this dissertation, models were developed to determine the solvent ratio for three mixtures: dichloromethane/methanol, acetone/water, and ethanol/water. The dichloromethane/methanol mixture model was created using data from the DynoChem program, which the values were validated through laboratory experiments, and a RMSE of 0.0035 g/cm³ was obtained. For acetone/water and ethanol/water, the Dynochem's data was not valid, so new models were developed based on laboratory tests, obtaining RMSEs of 0.0071 g/cm³ and 0.0054 g/cm³, respectively. Furthermore, an Excel tool was created to know the quantity of solvents to be loaded into the reactor, based on density, temperature, and mass of the binary mixture. It was also studied the efficiency of the solvent reuse through simulations carried out in Aspen Plus; they showed that the efficiency stabilizes after several batches, with significant financial savings: 1510€ per batch for dichloromethane/methanol, 3870€ for acetone/water, and 2030€ for ethanol/water. If solvent reuse had been implemented in the fiscal year 2023, Hovione would have saved at least 19.5 k€, €77.6 k€, and €35.7 k€ for the respective mixtures.A Hovione ambiciona reduzir as emissões de carbono em 40%, e atualmente todos os solventes utilizados no processo de secagem por pulverização são incinerados ou vão para tratamento, e solventes frescos são utilizados. Para se aumentar a sustentabilidade, economia e produtividade do processo, é essencial reutilizar estes solventes. No entanto, reutilização é particularmente desafiante com a utilização de misturas binárias, uma vez que requer um conhecimento preciso do rácio de solventes nas misturas, de forma a ajustar-se quantidades para a sua utilização em novos lotes na produção. Este é o principal objetivo deste trabalho: desenvolver uma estratégia para se re-ajustar as quantidades de solventes nas misturas na produção, sem a necessidade de testes laboratoriais. A composição das misturas pode ser obtida a partir da densidade e temperatura das misturas, variáveis facilmente medidas pelos caudalímetros na produção. Na presente dissertação desenvolvaram-se modelos para determinar o rácio de solventes para três misturas: diclorometano/metanol, acetona/água e etanol/água. O modelo da mistura diclorometano/metanol foi criado com recurso a dados do programa DynoChem e foi validado através de dados experimentais, tendo-se obtido um RMSE de 0.0035 g/cm³. Para a acetona/água e etanol/água, o DynoChem não era válido, pelo que foram desenvolvidos novos modelos baseados em testes laboratoriais, obtendo-se RMSEs de 0.0071 g/cm³ e 0.0054 g/cm³, respetivamente. Além disso, foi criada uma ferramenta Excel de forma a saber-se a quantidade de solventes a carregar ao reator, a partir da densidade, temperatura e massa das misturas binárias. Foi também estudada a eficiência da reutilização de solventes, através de simulações no Aspen Plus, que mostraram que a eficiência estabiliza após vários lotes, com poupanças financeiras significativas: 1510€ por lote para a mistura diclorometano/metanol, 3870€ para acetona/água e 2030€ para etanol/água. Se a reutilização de solventes tivesse sido implementada no ano fiscal de 2023, a Hovione teria poupado, pelo menos, 19.5 k€, 77.6 k€ e 35.7 k€ para as respetivas misturas

    Feasibility study of EWOD-based pumping mechanism for on-chip valvaless microfluidic applications

    When faced with the miniaturization challenges that mechanical micropumps pose, the current state of the art does not offer a fabrication flow that is CMOS compatible with reasonable sample yield (more then 50pc of the working devices after the fabrication flow) nor compatible with high ionic concentration liquids. To fulfill this gap, this thesis aims at studying the possible use of electrowetting on dielectric (EWOD) as the pumping mechanism behind a valveless on-chip micropump. To achieve this goal, the role of ion concentration in EWOD performance was studied by testing phosphate buffer saline (PBS), com-monly used in place of bodily fluids in testing in terms of ion concentration and molarity, and de-ionized water (DIW) through contact angle (CA) measurements, as well as impedance spectroscopy (IS) tests. Simulation algorithms were created to both simulate the frequency-dependance of EWOD performance of the droplets, and to study the influence of design parameters in final pump performance. Microchips were fabricated in a cleanroom environment and assembled to experimentally test the feasibility of the chosen pumping mechanism. The EWOD performance studies revealed that the higher the ion concentration of the droplet, the higher the achieved CA change when applying the same voltage, and the broader the usable frequency range before reaching its cutoff. The testing of the microchips with an optical microscope was successful in proving the possibility of using EWOD as a flow generation method, with electric actuation resulting in a 15 µm oscillatory motion at the outlet’s liquid-air interface. The main focuses of future studies for this project should be the improvement of the fabrication process and the introduction of an element that stimulates directional flow to the device, effectively turning into a micropump.Ao enfrentar os desafios de miniaturização que microbombas mecânicas apresentam, o panorama atual de microfluídica não oferece um fluxo de fabricação compatível com CMOS com um rendimento razoável de amostras (mais de 50% dos dispositivos funcionais após o fluxo de fabricação) nem compatível com líquidos de alta concentração iónica. Para preencher esta lacuna, esta tese visa estudar o possível uso de electrowetting em dielétrico (EWOD) como o mecanismo de bombeamento por trás de uma microbom-ba on-chip sem válvulas. Para alcançar este objetivo, o papel da concentração iónica no desempenho de EWOD foi estudado testando tampão fosfato-salino (PBS), comumente usado em vez de fluidos corporais em testes em termos de concentração iónica e molaridade, e água desionizada (DIW), através de medições de ângulo de contato (CA), bem como testes de espectroscopia de impedância (IS). Foram criados algorit-mos de simulação para simular a dependência do desempenho de EWOD das gotas na frequência aplicada, e para estudar a influência dos parâmetros de design no desempenho final da bomba. Microchips foram fabricados em ambiente de sala limpa e montados para testar experimentalmente a viabilidade do mecanis-mo de bombeamento escolhido. Os estudos de desempenho de EWOD revelaram que, quanto maior a concentração iónica da gota, maior a mudança de CA alcançada ao aplicar a mesma voltagem, e mais ampla a gama de frequências uti-lizável antes de atingir o seu corte. Os testes de microchips com um microscópio óptico foram efetivos em provar a possibilidade de usar EWOD como um método de geração de fluxo, com a atuação elétrica re-sultando num movimento oscilatório de 15 µm na interface líquido-ar da saída do dispositivo. Os principais focos de estudos futuros para este projeto devem ser a melhoria do processo de fabricação e a introdução de um elemento que estimule o fluxo direcional para o dispositivo

    Improve spray dryer control strategy with redundant devices in order to avoid equipment interlock

    Hovione's pharmaceutical operations sector experienced in 2021 and 2022 several unexpected shutdowns. The parameter that led to more shutdowns was the oxygen concentration in Spray Dryers' gas circuit, resulting in estimated losses of around 560 000€. Aiming to mitigate this problem, this dissertation proposes a control strategy using redundant oxygen concentration measuring devices. The initial step consisted in assessing the feasibility and redundancy of having two oxygen analyzers in the Spray Drying process. It was concluded that for a model equipment (Spray Dryer 84) having two oxygen analyzers was feasible and, due to their location, operating conditions, and output values behavior, they can be considered redundant. The subsequent step involved designing an enhanced control strategy capable of handling simultaneous readings from both analyzers during operation, as well as managing any signal loss from either analyzer. This strategy consisted in analyzing the discrepancy between the values and their comparison with the maximum value of oxygen concentration allowed. After the strategy development, it is expected that this strategy passes in software and real process tests. Moreover, the objective is to apply the control strategy herein proposed to 12 additional Spray Dryers currently in operation at Hovione, which lack a redundant analyzer. To evaluate the required investment for this implementation and the most cost-effective oxygen analyzer under consideration: the paramagnetic or the tunable diode laser spectrometer, an economic analysis was performed. The conclusion was that the paramagnetic oxygen analyzer is the most suitable due to a 41.7% lower investment cost than the tunable diode laser spectrometer and greater compatibility with the process. In particular, the investment in the paramagnetic oxygen analyzer has a payback time of 8 months and a return on investment of 66%. In conclusion, the improved control strategy herein developed makes it possible to reduce unexpected shutdowns, thus increasing process robustness, and importantly it can be extended to 12 additional Spray Dryer units currently in use at Hovione, offering a rea-sonable return on investment and a favorable pay-back time.O sector de operações farmacêuticas da Hovione teve em 2021 e 2022 várias paragens inesperadas. O parâmetro que levou a mais paragens foi a concentração de oxigénio no circuito de gás do secador por atomização, resultando em perdas estimadas de cerca de 560 000€. Com o objetivo de mitigar este problema, esta dissertação propõe uma estratégia de controlo utilizando analisadores redundantes na medição da concentração de oxigénio. O passo inicial consistiu em avaliar a viabilidade e redundância de ter dois analisadores de oxigénio no processo de secagem por atomização. Concluiu-se que, para um equipamento (Secador por atomização 84), a existência de dois analisadores de oxigénio é viável e que, devido à sua localização, condições de operação e comportamento dos valores das medições, podem ser considerados redundantes. O passo seguinte consistiu em desenvolver uma estratégia de controlo, capaz de ter em conta leituras simultâneas de ambos os analisadores durante o funcionamento, bem como gerir eventuais perdas de sinal de qualquer um deles. Após o desenvolvimento da estratégia, espera-se que esta seja aprovada em testes de software e de processos reais. Além disso, o objetivo é aplicar a estratégia de controlo proposta em 12 secadores por atomização em funcionamento na Hovione, que não possuem um analisador redundante. Para avaliar o investimento necessário para esta implementação e o analisador de oxigénio mais rentável a considerar, considerando o paramagnético ou o espetrómetro laser de díodo sintonizável, foi feita uma análise económica. A conclusão foi que o analisador de oxigénio paramagnético é o mais adequado devido a um custo de investimento 41,7% inferior ao do espetrómetro e a uma maior compatibilidade com o processo. Em particular, o investimento no analisador paramagnético tem um tempo de retorno de 8 meses e um retorno de investimento de 66%. Em conclusão, a estratégia de controlo desenvolvida torna possível reduzir paragens inesperadas, aumentando assim a robustez do processo, e mais importante ainda, pode ser estendida a 12 unidades adicionais de secagem por atomização atualmente em uso na Hovione, oferecendo um retorno de investimento e um tempo de retorno favorável

    Synthesis of pharmaceutically relevant N-heterocycles via metal-catalysed reactions

    N-heterocycles are an important class of compounds present in many bioactive mole cules, constituting a valuable source of therapeutic agents in the pharmaceutical industry. Par ticularly, carbolines and quinolines are notable due to their diverse biological properties and structural diversity. In this work different approaches were explored towards the carboline nucleus. The method investigated relied on a palladium-catalysed denitrative C−H amination, as the key step to achieve the saturated carboline scaffold. The first step involved the preparation of an N,N diarylamine via nucleophilic aromatic substitution reaction using the commercially available 3-fluor-2-nitropyridine and p-anisidine, leading to the desired product in 82% yield. After N protection, a palladium-catalysed denitratrive C−H amination led to the formation of the car boline in 27% yield, constituting of a new and promising route. The synthesis of tetrahydrocarbolines was also considered, and two different approaches were investigated. The first consisted of using properly protected L-threonine and aniline to generate a chiral key intermediate, to be further converted into several chiral C1-substituted tetrahydro carbolines, relying on a metal-catalysed reaction. However, this approach did not afford the desired compound. The second approach consisted of using an allylic amine, pre pared via a molybdenum-catalysed amination reaction, to achieve a key intermediate for further reaction with an aromatic amine, followed by palladium-catalysed cyclization. However, the allylic amine underwent an intramolecular cyclization leading to the formation of a quinoline, by C−H activation reaction, along with the acetylated aza-Cope rearrangement product. This fascinating result, lead us to explore this reaction. The reaction was optimized, and different allylic amines were prepared in yields up to 86%. The reaction proved to be wide in scope, and quinolines were obtained in yields up to 59%, via an unprecedented Pd-catalysed C−H activa tion reaction of allylic amines. The reaction is still being investigated, consisting of a promising and simple route towards biologically relevant quinolines.Os N-heterociclos são uma classe importante de compostos presentes em muitas molé culas bioativas, constituindo uma fonte valiosa de agentes terapêuticos na indústria farmacêu tica. Em particular, as carbolinas e as quinolinas são notáveis devido às suas diversas proprie dades biológicas e diversidade estrutural. Neste trabalho, foram exploradas diferentes abordagens para o núcleo da carbolina. O método investigado baseou-se numa aminação C−H desnitrativo catalisada por paládio, como passo fundamental para obter a estrutura da carbolina saturada. A primeira etapa envolveu a preparação de uma N,N-diarilamina através da reação de substituição aromática nucleofílica utilizando a 3-fluor-2-nitropiridina e a p-anisidina disponíveis no mercado, conduzindo ao pro duto desejado com um rendimento de 82%. Após N-proteção, uma aminação C−H desnitrativa catalisada por paládio conduziu à formação da carbolina em 27% de rendimento, constituindo uma nova e promissora via. A síntese de tetrahidrocarbolinas foi também considerada, tendo sido investigadas duas abordagens diferentes. A primeira consistiu na utilização de L-treonina e anilina devidamente protegidas para gerar um intermediário chave quiral, a ser posteriormente convertido em várias tetrahidrocarbolinas quirais C1-substituídas, com base numa reação catalisada por metal. No entanto, esta abordagem não permitiu obter o composto desejado. A segunda abordagem consistiu na utilização de uma amina alílica, preparada através de uma reação de aminação catalisada por molibdénio, para obter um intermediário-chave para uma reação posterior com uma amina aromática, seguida de ciclização catalisada por paládio. No entanto, a amina alílica sofreu uma ciclização intramolecular que conduziu à formação de uma quinolina, por reação de ativação C−H, juntamente com o produto de rearranjo aza-Cope acetilado. Este resultado fascinante levou-nos a explorar esta reação. A reação foi otimizada e foram preparadas diferentes aminas alílicas com rendimentos até 86%. A reação provou ser de âmbito alargado, tendo sido obtidas quinolinas com rendimentos até 59%, através de uma reação de ativação C−H de aminas alilicas sem precedentes catalisada por Pd. A reação ainda está a ser investigada, consistindo numa via simples e promissora para a obtenção de quinolinas biologicamente relevantes

    Does User-Generated Content (UGC) related to food tourism influence the destination choices of tourists?

    Dissertation presented as the partial requirement for obtaining a Master's degree in Data Driven Marketing, specialization in Marketing Research and CRMAccording to the World Food Travel Association, food tourism contributes significantly to a destination's economic benefit, accounting for 25%. In this sense, it is of interest to continue to analyse what motivates tourists to have food experiences at the destinations. Existing literature already underscores the proven significance of user-generated content (UGC) and the impact of food experiences on destination choices. Therefore, in this research, we propose develop a research model that allow us to understand the impact of food tourism related UGC on tourists’ decisionmaking. This research model is based on the theory of planned behaviour, which is commonly used to explain attitude-behaviour relations in the tourism context, including the tourists’ destination choices. Based on 292 responses, findings reveal that food tourism related UGC positively impact the behaviour intention to select a travel destination, and that relationship is mediated by subjective norms and perceived control over the behaviour. This academic research will allow us to deepen the knowledge of how food tourism UGC shapes travel decision-making. Additionally, it will enable us to provide insights about the role of digital platforms in shaping tourists' food behaviours and experiences to destination industry professionals. This, in turn, will empower them to refine marketing strategies and enhance visitor experience design within the dynamic landscape of food tourism

    Key motivations in energy community engagement: Impacts on adopting emerging technologies

    Dissertation presented as the partial requirement for obtaining a Master's degree in Information Management, specialization in Knowledge Management and Business IntelligenceThis research study aims to investigate the main motivations behind energy community engagement and its subsequent impact on the adoption of technologies. The leading question tries to understand what leads people to join energy communities. Individuals can be motivated to be in an energy community for several reasons, including environmental concerns and a commitment to community well-being. Energy communities are presented as a response to the environmental crisis, promoting resource management, consumption reduction, and lower ecological impact. They empower citizens by placing them at the centre of the energy value chain, endorsing local and autonomous energy generation. To explore this, an online questionnaire was distributed among individuals, gathering 300 completed responses. The results may indicate that empowerment serves as an interesting moderator of joy, improving the behaviour intention to participate and the engagement levels within energy communities. This suggests that positive emotions can significantly influence individuals' willingness to adopt emerging technologies and participate in sustainable energy practices. The rise of technologies that optimize the utilization of renewable resources further enhances this engagement. The findings highlight that motivations play a crucial role in energy community engagement, influencing perceptions of new technologies and supporting a global energy transition. By extending the hedonic-motivation system adoption model (HMSAM) through the lens of sustainable development, the study underscores the importance of fostering a sense of empowerment and environmental awareness to drive meaningful participation in energy communities

Banks have been improving how they provide services to their customers and mobile apps have allowed them to create a more accessible way to connect with them. One of the features that users value is robo-advisors, which provide customers with guidance in their investments. By using the expectation confirmation model, we show the continuance usage of robo-advisors. The results based on 152 individuals demonstrated that satisfaction and perceived usefulness of robo-advisors influence continuous usage. Using the mediation effect of satisfaction, we demonstrate how personalization and perceived anthropomorphism influence continuous intention
