Versuche zu Wachstum und Sporulation des Welkepilzes Verticillium nonalfalfae
Invasive, gebietsfremde Arten wie der Götterbaum (Ailanthus altissima) können negative Auswirkungen auf die Biodiversität von Pflanzenpopulationen haben und, im Fall des Götterbaumes, sogar Probleme für die Infrastruktur und menschliche Gesundheit verursachen. Die bisherige Bekämpfung des Götterbaumes ist aufwändig, zumeist ineffektiv, teuer und erfolgt oft unter Zuhilfenahme chemischer Herbizide. Bei dem in Österreich per Notfallzulassung verwendeten und sehr wirksamen biologischen Mykoherbizid Ailantex® handelt es sich um ein Isolat von Verticillium nonalfalfae (Vert56). In dieser Arbeit wurden die optimalen Wachstums- und Sporulationsbedingungen dieses Welkepilz-Isolats untersucht. Die o.g. Parameter wurden in zwei in vitro-Experimenten, einmal in völliger Dunkelheit und einmal in einer Langtag-Photoperiode (16 h Licht und 8 h Dunkelheit) getestet. In beiden Versuchen wurde das Wachstum des V. nonalfalfae-Isolats Vert56 bei vier verschiedenen Temperaturstufen (17,5 °C, 20 °C, 22,5 °C, 25 °C), auf insgesamt sechs verschiedenen Nährmedien (MEA, PDA, PEA, AiPDA, AiMEA und AiA), fünf davon in drei verschiedenen Konzentrationen (0,5 %, 1 % und 1,5 %), erhoben und danach die Sporulation der Kulturen sowie die Keimfähigkeit der Konidien ermittelt. Die optimale Wachstumstemperatur von V. nonalfalfae (Isolat Vert56) in vitro lag bei 22,5 °C, wobei das schnellste Wachstum auf MEA (0,5 %) stattfand. Die höchste Sporulation wurde bei Dunkelheit und den Temperaturen 17,5 °C und 20 ° C auf PDA nachgewiesen. Bei den Nährmedien PDA und MEA stiegen die Sporenzahlen mit zunehmender Nährmedien-Konzentration. Die höchsten Keimfähigkeits-Raten wurden konzentrations-unabhängig bei Licht und 17,5 °C erreicht. Tendenziell hemmte Licht das Pilzwachstum, ein Einfluss des Lichts auf die Sporulation konnte nicht nachgewiesen werden. Establishment and validation of in vitro models of the blood-saliva barrier
Biologische in-vitro Modelle der Blut-Speichel Schranke bieten die Möglichkeit den Transport von Biomarkern vom Blut in den Speichel (oder umgekehrt) zu untersuchen, die sich für die Diagnose von Erkrankungen nützlich erweisen, sowie Therapieansätze für die lokale oder systemische Anwendung zu testen.Es ist essenziell, dass diese Barriere-Modelle eine ausgeprägte parazelluläre Barriere und Expression von relevanten Markern aufweisen, um vergleichbar zu den natürlich vorkommenden Barrieren zu sein. Heutzutage werden hauptsächlich in-vitro Modelle basierend auf Primärzellen verwendet, die den Nachteil haben wegen ihrer limitierten Kultivierungsdauer nur eine eingeschränkte Charakterisierung zu ermöglichen. Deshalb liegt der Fokus vermehrt auf in-vitro Modelle, die auf Tumorzelllinien und immortalisierten Zellen basieren, aber oft nur eine schwache Ausprägung der parazellulären Barriere aufweisen.Um diese Hürde zu überbrücken, wurden im Zuge dieser Doktorarbeit in-vitro Modelle der Blut-Speichel Schranke basierend auf Tumorzelllinien und immortalisierten Zellen mithilfe von Kultivierungsbedingungen und Medienzusammensetzungen auf ihre Ausbildung einer parazellulären Barriere optimiert. Zusätzlich wurden diese Modelle auf ihre Expressionsmuster relevanter Marker auf der mRNA/Protein-Ebene untersucht und unter anderem mit Biopsien oder in-vivo Literaturdaten verglichen.Angewendet wurden die charakterisierten in-vitro Modelle schließlich für Transportstudien mit Biomarkern, die im Blut vorkommen, um ihre Anwendung in der Speicheldiagnostik zu evaluieren und letzten Endes für Entzündungsstudien der Mundschleimhaut, die mit nichtsteroidale Antirheumatika (NSAR) behandelt wurden.Zusammengefasst, wurden hier Epithelmodelle Blut-Speichel Schranke basierend auf die Mundschleimhaut, Unterkieferspeicheldrüse (Glandula submandibularis) und Ohrspeicheldrüse (Glandula parotis) etabliert, optimiert und für Untersuchungen von molekularen Transportsystemen eingesetzt.Biological in vitro models of the blood-saliva barrier are beneficial for analysis of the transport of biomarkers and drugs from blood to saliva or vice vera, either by supporting the development of new non-invasive diagnostic strategies or improving the understanding of the mechanism behind local drug deliveries.For laboratory data to have relevance in vivo, applied barrier models are required to have similarities to the biological tissue, such as a stringent paracellular barrier and expression of the relevant markers. So far, the majority of available models are either based on primary cells, where the potential of a detailed characterisation prior to experimental studies is limited, or on cell lines originating from either tumour or immortalisation, where expression of tight junction proteins for the formation of the paracellular barrier is subpar.To overcome the limitation of current cell line-based models of the blood-saliva barrier, their paracellular properties were optimised by systematic testing of cultivation condition and media composition. Expression of marker panels at the mRNA and protein level was performed and compared to biopsy samples or available in vivo data.After thorough characterisation the models were applied for transport studies of serum biomarkers to evaluate their potential for salivary diagnostics. Active transport mechanisms, potentially involved in the transport of those biomarkers, were identified and may explain the correlation between clinical serum and saliva data. Riparian habitats of the Austrian Danube - how have they developed and what have we lost?
Das Christian Doppler Labor (CDL-MERI) erforscht die Meta-Ökosystemdynamik in Flusslandschaften an der österreichischen Donau mit dem Ziel, nachhaltige ökologische Maßnahmen für das zukünftige Donaumanagement zu entwickeln. Im Rahmen des Projekts wird in dieser Masterarbeit die Veränderung der Uferlebensräume entlang eines 160 km langen Abschnitts der österreichischen Donau von Melk bis zur slowakischen Grenze untersucht. Die Auswirkungen von Einstauungen und Schifffahrt auf Uferhabitate werden durch sorgfältige Kartierung von mehr als 400 Kilometern Uferlinie im Jahr 1959 (vor dem Bau von Wasserkraftwerken) und 2021 untersucht. Die Studie untersucht die Veränderungen in der Habitatzusammensetzung und die Auswirkung des Wellenschlags und bestätigt, dass bereits 1959 große Teile der Ufer des Untersuchungsgebiets mit Blockwurf und Buhnen befestigt waren. Bis 2021 nahmen die Kiesbänke und -flächen weiter ab, insbesondere in den eingestauten Abschnitten. Diese Veränderung kann auf die Verlagerung von fluvialen zu lakustrinen Eigenschaften zurückgeführt werden, da die Stauanlagen die lotische Dynamik und das Sedimentregime verändern. Dies wirkt sich insbesondere auf die Fischlebensräume rheophiler Jungfische aus. Die vor Wellenschlag geschützten Lebensräume nahmen nur gering ab, ihre Qualität veränderte sich jedoch erheblich. Betonstrukturen, Steinschüttungen und biologisch gestaltete Ufer nahmen zu. Die ökologischen Folgen der Veränderung sind insbesondere hinsichtlich reduzierter Überlebensraten von Fischlarven relevant und unterstreichen die Notwendigkeit nachhaltiger Uferbewirtschaftung. Anhand der Kartierung wurde ein Datensatz erstellt, der für künftige flussökologische Arbeiten zu Verfügung steht. Die Datenbank der Uferhabitate kann dazu beitragen, nachhaltige Modelle und Strategien für das Management der Donau zu entwickeln.The Christian Doppler Laboratory (CDL-MERI) explores Meta-Ecosystem Dynamics in Riverine Landscapes at the Austrian Danube, aiming to develop sustainable ecological measures for future Danube management. Within the project, this master thesis explores the transformation of riparian habitats along a 160 km long section of the Austrian Danube from Melk to the Slovakian border. The impact of impoundments and navigation on bank types and wave-action, pressures common to rivers globally, are studied through meticulous mapping of more than 400 kilometres of shoreline in 1959 (before hydropower construction) and 2021. The study explicitly explores changes in habitat composition and wave exposure, confirming that already in 1959 large parts of the study area’s bank were fortified with riprap. By 2021 gravel banks and bars declined, particularly in impounded segments. The change may be attributed to the shift of fluvial to lacustrine qualities due to impoundments disrupting the lotic dynamics and sediment regimes, impacting fish habitats, especially for juveniles of rheophilic species. Habitat protected from wave action declined marginally but changed substantially in quality, with an increase of concrete, riprap and biologically engineered banks. The study underscores the vulnerability of native fish fauna, emphasizing the ecological consequences of altered riparian dynamics. Ecological implications point to the jeopardy of larval fish survival, emphasizing the need for sustainable management of riparian habitats. The study aims to create a comprehensive dataset that can be provided and utilized as support for future river ecology research, which may, in turn, contribute to developing sustainable river management strategies. Exploration of genome-wide DNA methylation patterns in table beet
Epigenetische Veränderungen, insbesondere die DNA-Methylierung, sind von großer Bedeutung für die Stabilität des Genoms, die Genexpression und die Aktivität transponierbarer Elemente. Die herkömmliche Bisulfit-Sequenzierung ist der Goldstandard, aber der Abbau der DNA-Proben beeinträchtigt die Analyse der genomweiten Cytosin-Methylierung. In dieser Studie wurden Methylierungsmuster in der Roten Rübe untersucht, wobei der Schwerpunkt auf charakteristischen Genen (Betalainsynthese- und Rhizomania-Resistenzgene) und transponierbaren Elementen (TEs) lag. Es wurden zwei neuartige Sequenzierungsansätze verwendet: enzymatische Methylsequenzierung (EM-seq) und PacBio-Sequenzierung. Im Gegensatz zu chemischen Methoden werden bei diesen Ansätzen Enzyme oder die Kinetik der DNA-Polymerase zur Identifizierung methylierter Basen eingesetzt. Die Rote Rübe (Beta vulgaris ssp. vulgaris), eine wirtschaftlich wichtige Kulturpflanze, wird selektiv wegen ihrer charakteristischen roten Wurzeln angebaut. Anhand von Blatt-DNA wurden genomweite Cytosin-Methylierungsmuster analysiert, insbesondere in CG-, CHG- und CHH-Motiven (wobei H = A, C oder T). Die CG- und CHG-Methylierung war genomweit und in TEs stärker ausgeprägt, wobei PacBio mehr Methylierung nachwies als EM-seq. Die Studie zeigte auch, dass TEs der Klasse I (Copia, Gypsy, LINE/L1) stärker methyliert sind als TEs der Klasse II (DNA-Transposons). Darüber hinaus wurden bei beiden Sequenzierungsansätzen in den untersuchten Genen und ihren regulatorischen Regionen konsistente Methylierungsmuster beobachtet. Diese Studie zeigt die Wirksamkeit von EM-seq und PacBio-Sequenzierung bei der Untersuchung der DNA-Methylierung in Pflanzen am Beispiel der Roten Rübe. Die Übereinstimmung der mit diesen Technologien beobachteten DNA-Methylierungsmuster unterstreicht ihre Komplementarität und ihr Potenzial für die künftige DNA-Methylierungsforschung.Epigenetic modifications, particularly DNA methylation, play a crucial role in shaping biological processes, including genome stability, gene expression regulation, and transposable element activities. While traditional whole genome bisulfite sequencing, has been a gold standard for DNA methylation exploration, it suffers from DNA sample degradation, hence compromising genome-wide cytosine methylation accuracy. This study investigates genome-wide methylation patterns in table beet, focusing on specific characteristic genes (Betalain and Rhizomania disease resistance gene) and transposable elements (TEs), utilizing two novel sequencing approaches: enzymatic methyl sequencing (EM-seq) and PacBio sequencing. Unlike chemical conversion methods, these approaches employ enzymes and kinetics to identify cytosine methylation, mitigating sample degradation concerns. Table beet (Beta vulgaris ssp. vulgaris), an economically significant crop with distinctive red roots, has been selectively cultivated for its betalain pigmentation. Leaf DNA was utilized to examine the genome-wide cytosine methylation patterns, particularly in CG, CHG, and CHH contexts, (with H= A, C, or T). Overall, CG and CHG methylation were at a higher level genome-wide and within the explored TEs, with PacBio sequencing detecting more methylation than EM-seq. The study also found that cytosine methylation levels were higher in class 1 TEs (Copia, Gypsy, LINE/L1) compared to class 2 TEs (DNA Transposon), with PacBio sequencing detecting more CG context methylation than EM-seq in TEs. Furthermore, consistent methylation patterns were observed across the two sequencing approaches in the genes explored and their regulatory regions. This study underscores the efficacy of EM-seq and PacBio sequencing in studying DNA methylation in table beet. Exploring the micro-scale biogeochemistry of nutrient and contaminant hot spots in soil with chemical imaging
Die räumliche Verteilung von pH-Werten, Flüssen labiler Elemente und Enzymaktivitäten kann mit Chemical Imaging (Optoden, Diffusive gradients in thin films, Zymographie) zerstörungsfrei, hochauflösend und in situ untersucht werden. In dieser Dissertation wurden diese Techniken in unterschiedlichen Kontexten miteinander kombiniert um Interaktionen und Prozesse der Nähr- und Schadstoffdynamik im Boden rund um Wurzeln oder Metallverbindungen zu erforschen und Potentiale und Limitierungen zu diskutieren. Im ersten Teil wurde der Einfluss von Bodentextur und Wurzelhaaren auf Phosphor(P)-Mobilisierungsprozesse untersucht. Unter Laborbedingungen wurde um junge Lupinenwurzeln eine erhöhte Phosphatase-Aktivität, sowie eine Verarmung an labilem P beobachtet. Das weist darauf hin, dass der hohe P-Bedarf von Lupinen und Mikroorganismen in der Rhizosphäre nicht durch die Aktivität von Phosphatasen allein gedeckt werden konnte. Im Feldversuch ermöglichten Wurzelfenster die Untersuchung von Maiswurzeln zu Beginn des generativen Wachstumsstadium. Statt P-Verarmungszonen wurden erhöhte Flüsse von labilem P und Versauerung rund um wachsende Wurzelspitzen beobachtet, was die Bedingungen für Phosphatase-Aktivität dort optimierte, wo Phosphor präferiert von der Wurzel aufgenommen wird und darauf hindeutet, dass diese Rhizosphären-Prozesse gemeinsam zur Phosphorversorgung beitragen. Im zweiten Teil wurde das Zusammenspiel von pH-Änderungen und Mobilität von Schrotbestandteilen während der Verwitterung von Blei und Wolfram-Schrotkugeln im Boden untersucht. Trotz des geringen Anteils waren insbesondere Nickel und Antimon aufgrund ihrer Freisetzung und Mobilität im Boden bedeutender als die Schrothauptbestandteile. Die Kombination von Chemical Imaging Methoden ermöglichte mehrere Prozesse in der Rhizosphäre oder rund um Kontaminationsquellen zu verknüpfen und ein umfassenderes Verständnis von Phosphor-Kreisläufen in der Rhizosphäre und der Verwitterung von Schrotkugeln im Boden zu erhalten.Chemical imaging allows to non-destructively visualize the distribution of pH (planar optodes), labile ions (diffusive gradients in thin films) and active enzymes (zymography) at high resolution and in situ. The objectives of this work were to combine imaging techniques in different contexts to explore interactions and processes influencing the fate of nutrients and contaminants in soil around roots (rhizosphere) or metal compounds and to discuss methodological merits and limitations. The first part aimed to study phosphorus (P) cycling in rhizosphere by investigating the effects of soil texture and root hairs on the distribution and intensity of root-induced pH changes, acid phosphatase activity (APA) and labile P. Under laboratory conditions, APA was high along the roots but could not compensate for the high demand, rapid uptake, and immobilization of P, resulting in depletion of labile P around young roots of blue lupin (Publication I). Custom-made root windows allowed for imaging of maize roots at the transition from vegetational to reproductive growth under field conditions (Publication II). Acidification was linked to root growth, establishing optimum conditions for APA which co-localised with elevated P fluxes at the positions where P is mainly taken up by the root indicating that these rhizosphere processes jointly contribute to P nutrition. The second part investigated the interplay of weathering-induced pH changes and corresponding mobility of lead(Pb)- and tungsten(W)-based shotgun pellet components in soil (Publication III). Images and batch incubations revealed that minor shot components such as nickel in the W-shot and antimony in the Pb-shot are of major importance due their release and mobility. Development of perfusion control strategies for integrated continuous biomanufacturing
Ein Trend in der biopharmazeutischen Industrie investiert derzeit in die Entwicklung von kontinuierlichen und integrierten Fertigungsprozessen, die die Produktivität steigern und gleichzeitig Prozessdimensionen reduzieren und unproduktive Stillstandszeiten beseitigen könnten.Vor diesem Hintergrund wurden integrierte Steuerungsstrategien für einen Perfusionsprozess im Rahmen einer kontinuierlichen Bioverarbeitung im kleinen Maßstab entwickelt. Strategien zur Verbesserung in Bezug auf das Prozessdesign wurden in einem skalierbaren Aufbau etabliert und in Verbindung mit einer nachgeschalteten Produktaufreinigung ganzheitlich getestet. Um Entwicklungszeiten zu beschleunigen wurden reversible und irreversible Auswirkungen auf das zelluläre Verhalten bei Verschiebung von Prozessparametern innerhalb eines Experiments wurden in Minibioreaktoren getestet. Kritische Qualitätsattribute wie Antikörperglykation wurden untersucht und die erstellten Modelle auf Perfusionsprozesse übertragen. Die Aufmerksamkeit wurde außerdem auf Kontrollstrategien in der Zellkultureinheit gelenkt, um Schwankungen oder Verunreinigungen in den verschiedenen Parametern zu minimieren. Dabei konnte mit der Implementierung einer umgekehrten tangentialen Flussfiltrationssteuerung in der Zellrückhaltevorrichtung die Membranverschmutzung minimiert und kontrollierte Erntebedingungen erzielt werden. Darüber hinaus verbesserte die Implementierung von modellprädiktiven Kontrollstrategien in der Perfusionseinheit die Prozessleistung deutlich und trug dazu bei, Prozessschwankungen in den Flussraten oder den Produktkonzentrationen, die zum direkt integrierten Aufreinigungsschritt transportiert werden, zu reduzieren. Die implementierten Strategien verringerten den Kontrollaufwand in den nachfolgenden Prozessschritten und zeigten große Möglichkeiten zur Lösung der Herausforderungen und Bedürfnisse, die integrierte kontinuierliche Bioprozesse in der Antikörper Herstellung mit sich bringen.The biopharmaceutical industry is currently investing great efforts in establishing continuous and integrated manufacturing processes that could increase productivity while reducing process dimensions and eliminating unproductive downtimes. However, there are still certain challenges that require continued development for the implementation of continuous processes in industry.The work in this thesis aimed to develop integrated solutions and control strategies for a perfusion process in the context of integrated continuous bioprocessing. Based on a four-step approach, a perfusion procedure using a recombinant CHO cell line was developed at small-scale, scaled up, and tested for feasibility in conjunction with downstream capture. Strategies to improve cell performance in terms of media and process design were established in a scale-down setup: Reversible and irreversible impact on cellular behavior upon intra-experimental process parameter shifts were identified in spin tube bioreactors to accelerate development times. Critical quality attributes such as antibody glycation were studied in fed-batch cultures and the model transferred to perfusion processes. Attention has been drawn to more advanced control strategies in the cell culture unit, where product or impurity variations in perfusion harvest were minimized to allow long-term steady state conditions. The implementation of dynamic or reverse tangential flow filtration control in the cell retention device proved to minimize membrane fouling and allowed controlled product purity levels. In advance, model predictive control strategies significantly improved process performance and helped to reduce process variations in perfusion harvest flow rates or product concentrations. The implemented strategies reduced control efforts in the subsequent unit operations and showed great opportunities to resolve the challenges and needs provided by integrated continuous bioprocesses in mAb manufacturing. The impacts of climate change on mountain agriculture in Europe : future socio-ecological trajectories
Europäische Landwirt*innen stehen nicht nur vor der Herausforderung, sich an sinkende Erträge auf Grund von Klimawandel anzupassen, sondern müssen auch sozioökonomische Veränderungen, wie Reformen der Gemeinsamen Agrarpolitik, in ihre Entscheidungen miteinbeziehen. Neben biophysikalischen und sozioökonomischen Rahmenbedingungen ändert sich auch die intrinsische Motivation der Landwirt*innen, insbesondere im Zuge der Hofübergabe an die nächste Generation. In meiner Dissertation untersuche ich, wie sich Klimawandel, wechselnde sozioökonomische Rahmenbedingungen sowie intrinsische Faktoren auf die Entscheidungsfindung von Landwirt*innen hinsichtlich Landnutzung, Betriebsmanagement und Anpassung an den Klimawandel auswirken. Der regionale Fokus liegt auf Europäischen Bergregionen, da diese komplexen Veränderungen nicht nur die individuelle Landbewirtschaftung, sondern auch die Landnutzungsdynamik zwischen Flachland und Bergland beeinflussen werden. Mit Hilfe des agenten-basierten Modells SECLAND ABM habe ich für Studienregionen in Österreich und Frankreich mögliche zukünftige Landnutzungspfade in sozioökonomisch und klimatisch verschiedenen Szenarien simuliert. Die Ergebnisse prognostizieren einen weiteren Rückgang landwirtschaftlicher Betriebe gekoppelt mit der Verwaldung aufgegebener Flächen (v.a. extensives Grünland). Die Szenario-Unterschiede zeigen, dass externe Bedingungen diese Entwicklung beschleunigen oder abfedern können. Während Klimawandel in einigen Untersuchungsgebieten positiv auf die langfristige Ertragsentwicklung gewirkt hat, war der Einfluss von klimawandelbedingten Wetterextremen auf die landwirtschaftliche Produktion durchwegs negativ. Die Arbeitsbelastung hat sich als Schlüsselfaktor für die Entscheidungsfindung gerade im Hinblick auf Klimawandelanpassung herauskristallisiert. Finanzielle Anreize haben nur eine begrenzte Motivation und soziale Aspekte, wie die Arbeitsbelastung, müssen dringend stärker in Nachhaltigkeitszielen berücksichtigt werden.European farmers are facing not only challenges from anthropogenic climate through decreasing yields but also socio-economic changes such as reforms of the common agricultural policy guidelines. Biophysical and socioeconomic framework conditions, also intrinsic motivations of farmers change, especially during the phase of handing over a farm to the next generation. This dissertation investigates how climate change, changes in the socioeconomic conditions as well as intrinsic factors will affect the decision-making of farmers regarding land use, farm management and climate change adaptation. The regional focus lies on mountain regions in Europe, as these complex dynamics will not only affect individual farm and land management but also collective land-use dynamics between lowlands and mountain areas. Investigating these dynamic developments, I developed and applied the agent-based model SECLAND ABM to three case studies in Austria and France and simulated socio-ecological development trajectories for three socio-economically and climatically diverging future scenarios. Across all scenarios, the simulation results depicted the continued decline of active farms and agricultural land with a forest transition on abandoned areas, especially grasslands. However, the differences among the scenarios highlighted that changing scenario conditions can accelerate or buffer this development. While some aspects of climate change also had some positive long-term effects in some study sites, climate change related, extreme weather events always had negative effects on agricultural production. The workload of farms emerged as essential constraint for farmers’ decision-making for farm management but also for climate change adaptation. Financial incentives provide only partial motivation and their impact is limited. Process development for the production of polyhydroxyalkanoates from volatile fatty acids
.Developing biorefineries is an important part of the solution towards environmental sustainability. To make biorefineries a reality, it is crucial to develop the conversion of platform compounds like volatile fatty acids (VFA) into high-value products, such as biopolymers. Polyhydroxyalkanoates (PHA) are biopolymers important for many application sectors, ranging from agriculture to biomedicine. This thesis investigates the potential for the production of polyhydroxyalkanoates from residue-based VFA.In a first step, the potential of the Pseudomonas genus to valorize VFA by producing mcl-PHA was explored. The investigation focused on VFA from anaerobic digestion of pressed sugar beet pulp. The acidification was optimized and scaled up in a 250 L reactor, reaching 27.8 g/L of VFA. Furthermore, the P. putida and P. citronellolis were grown on the digestion-based substrate producing 15.2 g/L and 15.6 g/L of biomass containing mcl-PHA.The mcl-PHA production was further scaled up to a 500-litres. The biomass reached up to 15.2 g/L, with 24.9 % of mcl-PHA. The produced polymer had a molecular weight of 536 kDa. The most prevalent monomer fraction was 3-hydroxydecanoate (3HD) with 60.7 %, followed by 3-hydroxyoctanoate (3HO) with 37.3 %. Additionally, 11 % of monomers side chains had double bounds.Finally, the problem of high dilution of VFA from anaerobic digestion was addressed. The study introduces a complex cell recycle fed-batch system using Bacillus megaterium for poly(3-hydroxybutyrate) production from acetic acid. This indirect pH-stat feeding regime successfully fed the strain without inhibition while keeping the process volume constant. Entscheidend für ihre Umsetzung ist die Entwicklung von Verfahren zur Umwandlung von Plattformverbindungen wie flüchtige Fettsäuren (VFA) in hochwertige Produkte wie Biopolymere. Polyhydroxyalkanoate (PHA) sind Biopolymere, die vielseitig eingesetzt werden können. Diese Arbeit untersucht das Potential zur Produktion von PHA aus reststoffbasierten VFA.Zunächst wurde das Potential der Gattung Pseudomonas zur Verwertung von VFA zur Produktion von mcl-PHA getestet. Der Fokus lag dabei auf VFA aus der Vergärung von Zuckerrübenpressschnitzeln. Der Versäuerungsprozess wurde optimiert und in einem 250 L Reaktor hochskaliert, wobei eine Konzentration von 27.8 g/L VFA erreicht wurde. P. putida und P. citronellolis wurden auf dem vergärungsbasierten Substrat kultiviert, wobei 15,2 und 15,6 g/L Biomasse produziert wurden.Die Produktion von mcl-PHA wurde in einem weiteren Schritt auf 500 L skaliert. Die Biomasse erreichte Werte von bis zu 15,2 g/L mit einem Gehalt von 24,9 % PHA. Das Polymer hatte ein Molekulargewicht von 536 kDA. Die vorherrschende Monomerfraktion war 3-Hydroxydekanoat mit 60,7 %, gefolgt von 3-Hydroxyoctanoat mit 37,3 %. Außerdem wiesen 11 % der Seitenketten der Monomere Doppelbindungen auf.Zuletzt wurde das Problem der hohen Verdünnung von VFA aus der anaeroben Vergärung adressiert. Es wird ein komplexes Fed-Batch System mit Zellrückführung vorgestellt, wobei Bacillus megaterium zur Produktion von Poly(3-hydroxybutyrat) aus Essigsäure verwendet wird. Das indirekte pH-Stat Substratzufuhrsystem versorgte den Stamm ohne Auftreten von Inhibierung, während das Prozessvolumen konstant gehalten wurde. Als Ergebnis wurde eine Biomassekonzentration von 19,1 g/L mit einem PHA-Gehalt von 70,2 % erzielt. Oxidative modification of cellulose : access to renewable thermoplastics
Plastik ist in unserer modernen Welt sowohl ein Fluch als auch ein Segen. Auf der einen Seite handelt es sich um ein äußerst vielseitiges Material, das Anwendungen im Bauwesen, im Gesundheitswesen, in der Verpackungs- und Transportindustrie findet. Auf der anderen Seite hat der weltweite Einsatz von erdölbasiertem Plastik zur Erschöpfung fossiler Brennstoffe und zu einer ernsthaften Plastikverschmutzung geführt, die sowohl die Umwelt als auch die menschliche Gesundheit gefährdet. Um dieser Herausforderung zu begegnen, widmet sich die vorliegende Arbeit der Entwicklung von alternativen Materialien aus Cellulose, einer erneuerbaren Ressource, mit dem Ziel, erdölbasierte Kunststoffe zu ersetzen. Allerdings kann Cellulose im Gegensatz zu herkömmlichem Plastik nicht allein durch Erhitzen verformt werden. Dies ist jedoch möglich, wenn Cellulose mittels Periodat-Oxidation in "Dialdehyde Cellulose" (DAC) umgewandelt und anschließend weiter modifiziert wird. In dieser Arbeit wurden verschiedene Modifikationswege untersucht, um unterschiedliche Seitengruppen einzuführen, und ihren Einfluss auf die Glasübergangstemperatur (Tg) zu analysieren. Dadurch wurden thermoplastische Materialien mit Tg-Werten von 23 bis 112 °C synthetisiert, die vergleichbare Eigenschaften wie konventionelle Kunststoffe (z. B. PET, PLA und PS) aufwiesen. Die Variation der Flexibilität des Polymergerüsts und der Seitengruppen ermöglichte die Anpassung der thermischen Eigenschaften für verschiedene Anwendungen. Im Verlauf dieser Forschung wurden grundlegende Herausforderungen im Zusammenhang mit der Periodat-Oxidation von Cellulose, deren Folgechemie und analytische Fragestellungen behandelt, um die Gesamtprozesse zu optimieren. Insgesamt stellt die Arbeit einen wichtigen Beitrag zur Entwicklung nachhaltiger Materialien dar, da sie einen Weg zur Ersetzung gebräuchlicher Kunststoffe durch cellulosebasierte Alternativen aufzeigt.Plastic is both a curse and a blessing in our modern world. On one hand, it is an incredibly versatile material, finding applications in everything from construction and healthcare to packaging and transportation. On the other hand, the widespread use of petroleum-based plastics has led to the depletion of fossil fuels and a serious issue of plastic pollution, posing threats to the environment and human health. To address this challenge, the present work is dedicated to developing alternative materials derived from cellulose, a renewable resource, aimed at replacing petroleum-based plastics. However, native cellulose cannot be melted and reshaped through heating alone, unlike plastics. One way to plasticize cellulose is through periodate oxidation to generate "dialdehyde cellulose" (DAC), which is then further derivatized. In this study, various modification pathways were explored to introduce different side chains along the polymer backbone, examining their influence on the thermal properties, particularly on the glass transition temperature (Tg). Cellulose-based thermoplastics with Tg values ranging from 23 to 112 °C were successfully generated, comparable to those of fossil-based alternatives (e.g., PET, PLA, and PS). A strategy was developed to tune the Tg by controlling the polymer backbone's flexibility and the composition of the side groups. Throughout the course of this research, fundamental challenges related to the periodate oxidation of cellulose, subsequent chemistry, and cellulose analytics were addressed to optimize the overall processes. Overall, the work represents an important contribution to the development of sustainable materials, as it provides a pathway to replace commonly used plastics with cellulose-based alternatives. Barriers in Austrian river ecosystems: how fragmentation metrics help to explain impacts on fish assemblages
Die Fragmentierung von Flüssen gilt als eine der Hauptbelastungen für Flussökosysteme und verantwortlich für den Rückgang wandernder Fischarten. Aus der Perspektive der Wiederherstellung der Durchgängigkeit ist es notwendig Prioritäten zu setzen, dort wo es für Fische am vorteilhaftesten ist. In dieser Arbeit wird zum ersten Mal die Fragmentierung von Flüssen und ihre Auswirkungen auf Ebene der Flussabschnitte und -segmente auf die Fischbestände in Österreich untersucht. Die Intentionen dieser Studie waren: (1) die Methode von Cooper et al. (2017) auf das österreichische Flussnetzwerk zu adaptieren, (2) das Ausmaß der Flussfragmentierung zu analysieren und wie es sich zwischen Ökoregionen, Fischregionen und Flussgrößen unterscheidet, (3) und die Auswirkungen der Flussfragmentierung auf Fischgemeinschaften in einer mehrfach beeinträchtigten Umwelt zu bewerten. Die Berechnung der Flussfragmentierung basiert auf einer räumlichen Analyse, bei der 60 253 Barrieren analysiert wurden und die zu 19 Fragmentierungsmetriken führt (z. B. Dichte der Barrieren, Entfernung der Barrieren). Die Ergebnisse zeigen, dass die Flussfragmentierung je nach Ökoregion, Fischregion und Flussgröße unterschiedlich ist. Die höchste Barrieredichte wurde in der Alpenregion und entlang der Hauptflüsse beobachtet. Insbesondere die Ökoregion 'Dinarischer Westbalkan' wies ein stark fragmentiertes Flussnetz auf. Außerdem zeigen die Ergebnisse einen drastischen Rückgang der frei fließenden Flussabschnitte und Einzugsgebiete in ganz Österreich. Obwohl eine Veränderung der Fischbestände aufgrund der Flussfragmentierung nicht beobachtet werden konnte, zeigten zusätzliche Stressfaktoren von Barrieren, Auswirkungen. Diese Studie unterstützt die Anwendung einer neuen Methode zur Berechnung der Flussfragmentierung und zur Bewertung der Auswirkungen der Flussfragmentierung auf die Fischbestände. Darüber hinaus hilft sie bei der Bewertung zukünftiger Entwicklung von Barrieren und Flussrenaturierungmaßnahmen.River fragmentation is seen as one of the key pressures on riverine ecosystems and is responsible for the decline of numerous migratory fish species. From a restoration perspective, it is necessary to prioritize river section where it is most beneficial for fish. For the first time, this thesis evaluates river fragmentation at two spatial scales, reach and segment and its impact on fish assemblages, in Austria. The intentions of this study were: (1) to adapt the method from Cooper et al. (2017) to the Austrian river network, (2) to analyze the extent of river fragmentation and how it differs between ecoregions, fish regions, and river sizes, (3) and to evaluate the effect of river fragmentation on fish assemblages in a multi stressor environment. The calculation of river fragmentation is based on a spatial analysis, considering 60 253 barriers and resulting in 19 fragmentation metrics (e.g. barrier density, barrier distance). The outcome showed that river fragmentation differs across ecoregions, fish regions, and river sizes. The highest barrier density was observed in the alpine region and along mainstem rivers. In particular, the ecoregion ‘Dinaric Wester Balkan’ showed a highly fragmented river network. Moreover, the results highlight a drastic decline in free-flowing river sections and catchment areas throughout Austria. Even though a shift in fish assemblages due to river fragmentation could not be observed, additional stressors induced by barriers showed an impact.This study supports the application of a new method to calculate river fragmentation and assess the effect of river fragmentation on fish assemblages.
