Usefulness of Neospora caninum dense granule 1 based-iELISA for the detection of canine neosporosis
application/pdfNeospora caninum is an apicomplexan parasite that causes abortion in cattle and sheep. Dog
is the main host of N. caninum and infected one shows neurologic disorders. Accurate
diagnosis is important particularly under lacking of treatment and vaccination measures.
Herein, we targeted the evaluation of diagnostic ability of recombinant protein
NcGRA1(rNcGRA1) for detection of specific antibody (IgG) against N. caninum in mice and
dogs. The rNcGRA1 was expressed using the E. coli expression system. Diagnostic ability
of rNcGRA1 was tested against experimental mouse and dog sera, as well as field dog sera.
Simultaneously, same serum samples were tested against rNcSAG1 and Neospora lysate
antigen (NLA)-based iELISA. In the N. caninum experimentally infected mice, specific IgG
antibodies to NcGRA1 were detectable from 14 days post infection (dpi), and increased at
28 dpi with no detectable antibody levels in the negative control or Toxoplasma gondii
infected-mouse sera. In experimentally infected dog sera, the performance of rNcGRA1 was
proficient in demonstrating IgG antibody at 28 dpi. In case of field dog sera (N=18),
seropositive rate was 44.4% and 22.2% for rNcGRA1 and rNcSAG1-based iELISAs,
respectively. These results show that the rNcGRA1 is a novel useful diagnostic antigen for
serological detection of N. caninum in dogs.journal articl
Lactobacillus plantarum Shinshu N-07 isolated from fermented Brassica rapa L. attenuates visceral fat accumulation induced by high-fat diet in mice
application/pdfLactobacillus plantarum Shinshu N-07 (N07) and Lactobacillus curvatus #4G2 (#4G2)
were isolated from fermented Brassica rapa L. and selected as promising probiotics
with anti-adiposity activities based on in vitro assays. The anti-adiposity effects of these
two strains were investigated using a diet-induced obesity animal model. Epididymal
adipose tissue weight and adipocyte area were significantly lower and serum
triglycerides and glucose tended to be lower in mice fed the high-fat diet supplemented
with N07 compared with those fed the unsupplemented high-fat diet. Strain N07
suppressed hepatic steatosis, with accompanying downregulation of lipogenic genes in
the liver. Expression of inflammatory cytokines and macrophage infiltration markers
tended to be suppressed by N07 supplementation. Upregulation of uncoupling protein-1
in epididymal adipose tissue by N07 suggested that the transformation of white adipose
tissue to brown might have been induced. Intestinal microbiota analysis revealed that a
decrease in abundance of family S24-7 (phylum Bacteroidetes) following ingestion of
the high-fat diet was partly recovered by supplementation with N07. Changes in those
parameters were not observed in mice fed the high-fat diet supplemented with strain
#4G2, suggesting strain specificities. Thus, N07 is a potential probiotic strain that could
be used to develop functional foods that attenuate visceral fat accumulation after an
appropriate human intervention trial.journal articl
Seroepidemiological study of Toxoplama gondii in small ruminants (sheep and goat) in different provinces of Mongolia
application/pdfToxoplasmosis is caused by the protozoan parasite Toxoplasma gondii. Consumption of raw or undercooked meat is the main risk factor for acquiring T. gondii infection in humans. Meat and meat products derived from goats and sheep are mainly consumed in Mongolia; however, there is limited epidemiological information on T. gondii infection in small ruminants in this country. The main objective of the present study was to investigate the seroprevalence of T. gondii in sheep and goats in Mongolia. The seroprevalence of T. gondii IgG antibodies was determined by an indirect enzyme-linked immunosorbent assay based on the recombinant antigens of dense granule protein 7 of T. gondii. A total of 1078 goat and 882 sheep blood samples were collected from 17 of 21 provinces and the capital city of Mongolia. Overall, the seroprevalence of T. gondii among the goat and sheep samples was 32% and 34.8%, respectively. The seroprevalence among goat samples was significantly higher in western (42.7%) and eastern (45.6%) regions compared with other regions (24%). Additionally, the seroprevalence among sheep was significantly higher in eastern regions (55.4%) compared with other regions (26%–33%). Age, but not sex, was considered a risk factor for T. gondii seropositivity in goats, whereas no statistically significant differences were observed in sheep for age or sex. In conclusion, the present study demonstrates the high seroprevalence of T. gondii in small ruminants in Mongolia. Our results highlight that country-wide control measures are required to minimize infections in livestock.journal articl
Treatment Efficiency of Combination Therapy With Diminazene Aceturate and Quinapyramine Sulfate in a Horse With Dourine
application/pdfDourine is a lethal protozoan disease of equids, and it is caused by Trypanosoma equiperdum infection via coitus. To date, treatment strategies against the dourine are not recommended because of the frequent relapses; therefore, the World Organisation for Animal Health recommends the stamping-out policy for the control of dourine. Our previous studies have revealed a number of horses with dourine in Mongolia that is the fifth largest horse-breeding country. It is difficult to apply the stamping-out policy for cases of dourine in Mongolia because of an inadequate livestock guarantee system. Therefore, the development of effective treatment measures is an urgent need. In this study, an 8-year-old stallion was definitely diagnosed with dourine based on clinical signs, molecular analysis, and microscopic examination of trypanosomes. Combination therapy with diminazene aceturate and quinapyramine sulfate was applied. Before the treatment, the characteristic clinical signs of dourine were observed, and trypanosomes were detected in the urogenital tract mucosal swab samples by microscopic examination and polymerase chain reaction (PCR). Moreover, positive serological results were obtained. After the treatment, we observed an improvement in the health of the treated horse and no trypanosome infection in its urogenital tract by microscopic examination and PCR. Moreover, serological tests showed seronegative results. The horse has showed no relapse for at least 2.5 years after the treatment, and its reproductive ability has improved. Our result suggests that trypanosomes did not invade cerebrospinal fluid when we started the therapy. In conclusion, the combination therapy has therapeutic potential against dourine at an early phase.journal articl
