    First detection of Theileria equi in free-roaming donkeys (Equus africanus asinus) in Sri Lanka

    application/pdfEquine piroplasmosis (EP) is a tick-borne disease caused by Theileria equi and Babesia caballi in equids, including horses, donkeys, zebras, and mules. It is globally endemic with significant economic impact on the equine industry. Infected animals may serve as carriers, and they may be a source of infection for ticks, thereby posing a great challenge for disease management. Sri Lanka is a tropical country, where infections by various tick-borne parasites are common among livestock animals. However, infections by T. equi and B. caballi remain unstudied in Sri Lanka. Therefore, in the present study, we conducted an epidemiological survey to investigate the presence of T. equi and B. caballi in apparently healthy free-roaming donkeys. Blood samples were randomly taken from 111 donkeys in Mannar (n = 100) and Kilinochchi (n = 11) districts in Sri Lanka. Thin blood smears were prepared from the blood samples and subjected to microscopic examination. Additionally, blood DNA samples were prepared and screened for T. equi and B. caballi infections using species-specific PCR assays. Our results showed that 64 (57.7%) and 95 (85.6%) of the donkeys were positive for T. equi by microscopy and PCR, respectively. However, all samples were negative for B. caballi. Phylogenetic analysis of the T. equi 18S rRNA sequences detected two distinct genotypes, namely C and D. To our knowledge, this is the first report of T. equi in Sri Lanka and of genotype C in donkeys.     Role of dense granule antigen 7 in vertical transmission of Neospora caninum in C57BL/6 mice infected during early pregnancy

    application/pdfNeosporosis is a parasitic disease affecting the health of dogs and cattle worldwide. It is caused by Neospora caninum, an obligate intracellular apicomplexan parasite. Dogs are its definitive host, it mostly infects livestock animals, especially cattle that acts as intermediate host. It is necessary to have well-established models of abortion and vertical transmission in experimental animals, in order to determine basic control measures for the N. caninum infection. We evaluated the role of N. caninum dense granule antigen 7 (NcGRA7) in the vertical transmission of N. caninum using the C57BL/6 pregnant mouse model. We inoculated mice on day 3.5 of pregnancy with parental Nc-1 or NcGRA7-deficient parasites (NcGRA7KO). Post-mortem analyses were performed on day 30 after birth and the surviving pups were kept until day 30 postpartum. The number of parasites in the brain tissues of offspring from NcGRA7KO-infected dams was significantly lower than that of the Nc-1-infected dams under two infection doses (1 × 106 and 1 × 105 tachyzoites/mouse). The vertical transmission rates in the NcGRA7KO-infected group were significantly lower than those of the Nc1-infected group. To understand the mechanism by which the lack of NcGRA7 decreases the vertical transmission, pregnant mice were sacrificed on day 13.5 of pregnancy (10 days after infection), although parasite DNA was detected in the placentas, no significant difference was found between the two parasite lines. Histopathological analysis revealed a greater inflammatory response in the placentas from NcGRA7KO-infected dams than in those from the parental strain. This finding correlates with upregulated chemokine mRNA expression for CCL2, CCL8, and CXCL9 in the placentas from the NcGRA7KO-infected mice. In conclusion, these results suggest that loss of NcGRA7 triggers an inflammatory response in the placenta, resulting in decreased vertical transmission of N. caninum.journal articl

    Longitudinal genetic analysis of semen production traits in Holstein bulls according to a random regression animal models on age

    application/pdfHere we used random regression animal models (RRAMs) to investigate genetic change over age in the semen volume (VOL) and sperm concentration (CON) of Holstein bulls. We used 35,294 collection records from 1284 Holstein bulls and their 4166 pedigree records. The models included year and month of collection, collection place, collection method, and number of collections attempted for each day and month of age (second-order regressions) as fixed effects; technician as a random effect; and additive genetic and permanent environment as random regressions (first-order regressions). We examined two RRAMs with homogeneous and heterogeneous residual variances (RRAM1 and RRAM2, respectively). By using RRAM1, heritability for VOL and CON increased from 0.08 to 0.61 and 0.18 to 0.57, respectively, between 10 and 126 months of age. By using RRAM2, heritability for VOL increased from 0.11 to 0.28 between 10 and 24 months of age for young bulls and increased from 0.08 to 0.48 between 25 and 126 months of age for mature bulls; heritability for CON ranged from 0.18 to 0.19 for young bulls and increased from 0.10 to 0.48 for mature bulls. Posterior genetic correlations between young ages and older ages were strongly positive for VOLs but weak for CONs. © 2022 Japanese Society of Animal Science.journal articl

    Diurnal Change of the Photosynthetic Light-Response Curve of Buckbean (Menyanthes trifoliata), an Emergent Aquatic Plant

    application/pdfUnderstanding plant physiological responses to high temperature is an important concern pertaining to climate change. However, compared with terrestrial plants, information about aquatic plants remains limited. Since the degree of midday depression of photosynthesis under high temperature depends on soil water conditions, it is expected that emergent aquatic plants, for which soil water conditions are always saturated, will show different patterns compared with terrestrial plants. We investigated the diurnal course of the photosynthetic light-response curve and incident light intensity for a freshwater emergent plant, buckbean (Menyanthes trifoliata L.; Menyanthaceae) in a cool temperate region. The effect of midday depression was observed only on a very hot day, but not on a moderately hot day, in summer. The diurnal course of photosynthetic light-response curves on this hot day showed that latent morning reduction of photosynthetic capacity started at dawn, preceding the apparent depression around the midday, in agreement with results reported in terrestrial plants. We concluded that (1) midday depression of emergent plants occurs when the stress intensity exceeds the species’ tolerance, and (2) measurements of not only photosynthetic rate under field conditions but also diurnal course of photosynthetic light-response curve are necessary to quantify the effect of midday depression.journal articl

    Virucidal Activities of Acidic Electrolyzed Water Solutions with Different pH Values against Multiple Strains of SARS-CoV-2

    application/pdfSevere acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a threat to human health. Acidic electrolyzed water (AEW) has recently been suggested to demonstrate virucidal activity. Many types of AEW with different pH values, generated by the electrolysis of different chemicals, such as sodium chloride, potassium chloride, and hydrochloric acid, are commercially available. In this study, we compared the virucidal activities of these types of AEW against SARS-CoV-2, including the ancestral strain and variant Alpha, Beta, Gamma, Delta, and Omicron strains. Virus solution (viral titer, 6.9 log10 50% tissue culture infective dose [TCID50]/mL) was mixed with AEW (free available chlorine concentration, 34.5 ppm) at mixing ratios of 1:9, 1:19, and 1:49. At mixing ratios of 1:9 and 1:19, AEW with a pH of 2.8 showed stronger virucidal activities than AEW with a pH of 4.1 to 6.5 against the SARS-CoV-2 ancestral strain in 20 s. From the strongest to the weakest virucidal activity, the AEW pH levels were as follows: pH 2.8, pH 4.1 to 5.4, pH 6.4 to 6.5. At a ratio of 1:49, the viral titers of viruses treated with all AEW solutions at pH 2.8 to 6.5 were almost below the detection limit, which was 1.25 log10 TCID50/mL. The virus inactivation efficiency of AEW was reduced in the presence of fetal bovine serum and other substances contained in the virus solution used in this study. AEW with pH values of 2.8 to 6.5 showed virucidal activity against all of the tested SARS-CoV-2 strains, including the ancestral and variant strains.
