Continuous Experience Sampling and the Affect-Embodiment Coherence
video/mp4Emotional states are a core element for well-being, and social communication and interactions. Therefore, a positive affective state when interacting with a product or service is a signal of design success. Emotional states consist of a felt affect triggered or elicited by a particular stimulus. These stimuli can be either endogenous, e.g., a memory; or exogenous, e.g., the smile of another person. Emotions vary within an instant, and a single stimulus might cause several emotions or a blend of them. Emotions are dynamically inter-related responses, including bodily changes such as facial expressions, posture, and other autonomic changes. Finding clear signatures on the intricate relationship between body changes and subjective experiences is one of the major challenges in affective science. It is still under debate whether bodily changes precede or follow the subjective experience of emotion. Currently, experience sampling is done mainly through self-report. However, self-report might be biased or interrupt the user experience. Bodily changes can provide an alternative measurement with higher temporal resolution. However, many of these cues can also be controlled voluntarily, and therefore have the same biases as self-report. I will introduce a series of studies, algorithms, and systems we have developed for measuring, predicting, and supporting personalized health and wellbeing. Brassinosteroids are required for efficient root tip regeneration in Arabidopsis
Compared with other organisms, plants have an extraordinary capacity for self-repair. Even if the entire tissues, including the stem cells, are resected, most plant species are able to completely regenerate whole tissues. However, the mechanism by which plants efficiently regenerate the stem cell niche during tissue reorganization is still largely unknown. Here, we found that the signaling mediated by plant steroid hormones brassinosteroids is activated during stem cell formation after root tip excision in Arabidopsis. Treatment with brassinazole, an inhibitor of brassinosteroid biosynthesis, delayed the recovery of stem cell niche after root tip excision. Regeneration of root tip after resection was also delayed in a brassinosteroid receptor mutant. Therefore, we propose that brassinosteroids participate in efficient root tip regeneration, thereby enabling efficient tissue regeneration to ensure continuous root growth after resection.journal articl
Nitrogen represses haustoria formation through abscisic acid in the parasitic plant Phtheirospermum japonicum
Parasitic plants are globally prevalent pathogens that withdraw nutrients from their host plants using an organ known as the haustorium. The external environment including nutrient availability affects the extent of parasitism and to understand this phenomenon, we investigated the role of nutrients and found that nitrogen is sufficient to repress haustoria formation in the root parasite Phtheirospermum japonicum. Nitrogen increases levels of abscisic acid (ABA) in P. japonicum and prevents the activation of hundreds of genes including cell cycle and xylem development genes. Blocking ABA signaling overcomes nitrogen’s inhibitory effects indicating that nitrogen represses haustoria formation by increasing ABA. The effect of nitrogen appears more widespread since nitrogen also inhibits haustoria in the obligate root parasite Striga hermonthica. Together, our data show that nitrogen acts as a haustoria repressing factor and suggests a mechanism whereby parasitic plants use nitrogen availability in the external environment to regulate the extent of parasitism.journal articl
A guiding role of the Arabidopsis circadian clock in cell differentiation revealed by time-series single-cell RNA sequencing
Circadian rhythms and progression of cell differentiation are closely coupled in multicellular organisms. However, whether establishment of circadian rhythms regulates cell differentiation or vice versa has not been elucidated due to technical limitations. Here, we exploit high cell fate plasticity of plant cells to perform single-cell RNA sequencing during the entire process of cell differentiation. By analyzing reconstructed actual time series of the differentiation processes at single-cell resolution using a method we developed (PeakMatch), we find that the expression profile of clock genes is changed prior to cell differentiation, including induction of the clock gene LUX ARRYTHMO (LUX). ChIP sequencing analysis reveals that LUX induction in early differentiating cells directly targets genes involved in cell-cycle progression to regulate cell differentiation. Taken together, these results not only reveal a guiding role of the plant circadian clock in cell differentiation but also provide an approach for time-series analysis at single-cell resolution.journal articl
Accessibility of ARPES for Three-dimensionally Architected Si{111}7×7 Facet Surfaces on Micro-patterned Si(110)
The creation of three-dimensional (3D) geometrical shapes with atomically ordered surfaces and the investigation of their physical properties are major steps contributing to the development of a new paradigm in surface science. We produced a 3D-patterned Si sample with atomically flat and reconstructed {111} facet surfaces, and investigated its structural and physical properties. To apply the conventional techniques in surface science to 3D samples with various oriented surfaces, instead of two-dimensional planar samples, an appropriate relationship between the crystallographic surface ordering on the 3D-architected surfaces and the angle-resolved photoelectron spectroscopy (ARPES) setup considering the configuration in 3D space is indispensable. The distinctive and complex low-energy electron diffraction (LEED) patterns reflecting the 3D-arranged facet surfaces showed the realization of atomically reconstructed facet surfaces on 3D-patterned Si. Surface states of the 3D-patterned Si{111} surfaces are mapped by ARPES by considering the 3D geometrical relationship. The selection of the appropriate alignment of the incident electron beam (light) for the target surfaces allows the clear observation of the band dispersion from the produced {111}7×7 facet surfaces in 3D space. Our demonstration of accessibility of ARPES technique could provide useful guidelines for new methodologies, giving a fundamental understanding of 3D-shape-induced novel functionalities.journal articl
Lensless dual-color fluorescence imaging device using hybrid filter
In this study, a dual-band hybrid filter that achieves high excitation light rejection performance in a lensless imaging system was fabricated and incorporated into an imaging device. The hybrid filter consisted of interference and absorption filters, and a fiber optic plate (FOP). The interference filters were attached to both sides of the FOP, which was placed on top of the absorption filter to suppress the decrease in spatial resolution. In addition, the lamination order was optimized to achieve a high fluorescence observation performance. The fabricated hybrid filter was mounted on an image sensor and had the ability to indicate the green and red fluorescence components.journal articl
