 シトクロム P450 は様々な生物種が持つ酸化酵素で、疎水性化合物の酸化反応を触媒することで生
合成や代謝等の生命現象に関わっている。その過程において、P450は不活性な C–H結合に直接官能
基を導入するため、合成化学への応用を目指して多くの研究と機能改変が数多く実施されてきた。数
十万種以上と報告される P450の中でも、巨大菌由来のCYP102A1 (P450BM3) は長鎖脂肪酸に対して
最速のモノオキシゲナーゼ活性を有することから、盛んに研究対象として扱われてきた。しかし他の
多くの酵素と同様に、P450BM3 は天然基質である長鎖脂肪酸の水酸化反応に最適化した構造と機能
を持つことから、大きく構造の異なる有機小分子等の非天然基質の酸化反応はほとんど進行せず、機
能の改変が必要となる。P450BM3の機能改変手法として、構成アミノ酸の置換を行う変異導入法や、
変異導入を繰り返す「指向性進化法」も非常に強力な手法として知られ、広く用いられている。一方、
申請者の所属研究室ではこれまでに、長鎖脂肪酸を模倣して合成された「デコイ分子」を P450BM3に
添加することで、変異導入なしにベンゼンやプロパンなどの非天然基質の効率的な水酸化を可能とす
る「基質誤認識システム」を開発している。また、非天然基質の水酸化が可能となるよう進化させた
P450BM3 変異体に対してデコイ分子を作用させることで、活性がさらに向上することが報告されて
おり、基質誤認識システムがこれまでに開発された P450BM3 変異体に対しても有効であると示され
ている。本論文は、基質誤認識システムの利用に適した P450BM3 変異体を指向性進化によって開発
し、細菌由来の天然分子への新規機能付与を達成した報告である。 
 第二章では、一部の細菌が分泌する物質をデコイ分子として利用する P450BM3 変異体の作製を、
指向性進化法によって実施した。菌体間コミュニケーション物質であるN-acyl homoserine lactone (acyl-
HSL) は一部のデコイ分子と類似のアシルアミノ酸骨格を有し、優れた膜透過性を示すことから、菌
体内反応系でのデコイ分子としての利用が期待できる。HSLの一種であるN-decanoyl HSL (C10-HSL) 
をデコイ分子として用い、菌体内ベンゼン水酸化反応を指標として、結晶構造解析などの結果を踏ま
えた半合理的設計により、五世代に渡る P450BM3の指向性進化を行った。最終的に基質結合部位に

は P25V, R47W, T49L, Q73G, A74V, L188P の変異が導入され、C10-HSL をデコイ分子として利用
可能となった V-19A14 変異体を開発した。V-19A14 変異体発現大腸菌は C10-HSL 存在下でのベ
ンゼン水酸化において GC 収率 46%を示し、C10-HSL は高い活性を誘起するデコイ分子の一つ
となった。既存のデコイ分子の中でも高い膜透過性を有するものは少ないが、C10-HSL は高い
膜透過性を活かして菌体内反応に利用する上で非常に有利であることが示された。 
 第三章では、開発した変異体の酵素触媒としての機能に加え、結晶構造解析によって C10-HSL
との結合を調査した。C10-HSL 存在下かつ精製酵素の状態で、V-19A14 によるベンゼン水酸化
反応の触媒回転頻度は毎分 475 回転に達した。この値は、全ての P450 によるベンゼン水酸化活
性の中で最速であり、高い応用可能性が示唆された。さらに本論文中ではトルエンやプロパン
などの非天然基質の水酸化に関しても記述する。結晶構造解析から、デコイ分子である C10-HSL
が既存のデコイ分子と同様の様式で変異体と結合していることが明らかとなった。一方で、過
去の報告では見られなかったアミノ酸残基に、水分子を介した水素結合が確認された。進化の
世代を経るごとに、前述の水素結合に関係するアミノ酸残基と水分子の数が変化することも確
認され、これが C10-HSL との結合解離定数の変化に大きく寄与していると考えられる。変異が
導入された R47W と T49L に着目すると、野生型ではフェニル基など嵩高い部分構造のデコイ
分子に適した空間が、アミノ酸側鎖が嵩高く変化することで C10-HSL のラクトン環構造に適し
た結合部位へと変化したことが示された。最後に、V-19A14 変異体の応用利用例として、HSL
合成酵素との共発現大腸菌を用いた自己完結型の菌体反応系を開発した。ベンゼン水酸化反応
において生成したフェノール収率は高くないものの、合成酵素の導入のみでデコイ分子の合成
と外部添加を不要とする反応系が達成され、今後更なる活性の向上が期待できる。 
 第四章では、植物ホルモンの一種であるサイトカイニン (CK) の水酸化を P450BM3 変異体
で試みたため報告する。広く用いられる F87A 変異体や、計算科学的手法を利用した変異体ラ
イブラリーを用いて CK 水酸化に挑戦したが、これを効率よく消費し水酸化する変異体の開発
には至らなかった。しかし、比較的大きい分子サイズの基質を標的とする基質誤認識システム
の反応の報告は少なく、今後の発展へ向け貴重な参考情報としての利用が期待される。 
 第五章では、システインのチオール基と共有結合を形成するマレイミド化合物を、システイ
ンを導入した R47C 変異体へ修飾した複合体を用い、基質誤認識システムの反応機構において
デコイ分子の解離が必須かどうかを検証した。N-Dodecyl maleimide (C12-Mal) は高い効率で
P450BM3 の R47C 変異体と共有結合で複合化されることが示唆された。C12-Mal を複合化した
変異体は、既存のデコイ分子である C10-Phe を添加した場合と同等にベンゼンとナフタレンの
水酸化反応を触媒することが明らかとなった。さらに C12-Mal を複合化した変異体にベンゼン
を滴定すると、ヘムの吸収スペクトル変化が観測され、デコイ分子が結合した状態でもベンゼ
ンが活性中心であるヘムへアクセス可能であることを強く示唆する結果を得た。 
 以上、申請者は、デコイ分子の利用に最適な P450BM3 変異体の開発を通じて、基質誤認識シ
ステムを利用した非天然基質水酸化反応の機能をさらに向上させるとともに、細菌由来の天然
分子に新たな機能を付与することに成功した。通常、指向性進化によって開発された変異体は
変異の蓄積により、安定な野生型から大きく構造が変化し不安定となるが、強く結合するデコ
イ分子と共存させて進化を行うことで、高い活性を示す変異体を比較的安定な構造を維持した
まま開発することが可能となる。以上の成果は、指向性進化による変異体酵素の開発と基質誤
認識システムの適用を別々に、単純な組み合わせで行うのみでは不可能な機能付与であると考
えられ、新しい酵素工学的アプローチとして高い有用性と応用可能性を有すると期待できる。

本研究は、核酸分子を基盤とした医薬品の機能向上を目指し、新たな化学修飾の
開発を中心に取り組んだ意欲的かつ先駆的な試みである。その目的は、薬理効果や
生物学的安定性を改善し、核酸医薬の可能性をさらに広げることであり、以下の主
要な研究成果を挙げた。 
まず、新規のリン酸プロドラッグとして、フルオロホスホルアミデート（FPA）
基を開発した。この還元応答性 FPA 基をゲムシタビンに導入し、ゲムシタビン耐性
細胞株でも高い抗がん活性を示すことを確認した。また、細胞毒性やアセチルコリ
ンエステラーゼ阻害活性が低いことも示され、高活性かつ低毒性のリン酸プロドラ
ッグとして有望であることが明らかになった。 
次に、核酸系逆転写酵素阻害剤（NRTI）として 2'-β-メチルセレニル修飾を施した
新規化合物を開発した。この修飾は、糖パッカー構造を C3'-エンドコンフォメーシ
ョンへとシフトさせ、HIV-1 逆転写酵素に対する基質として機能することを示した。
さらに、この化合物は従来の NRTI に耐性を示す株にも抗 HIV-1 活性を保持し、高
濃度でも細胞毒性が低いことが確認された。一方で、ピリミジン誘導体の細胞毒性
が強いことが示唆され、核酸塩基構造との関係についてさらなる研究が必要である。 
さらに、siRNA の応用を指向した安定なリン酸誘導体として、ケト-ホスホネート
の開発に取り組んだ。この誘導体は効率的に合成可能であることが示されたが、オ
リゴ核酸合成への応用においては課題が残された。特に、二置換基を導入する際の
困難さが明らかになり、試薬と反応条件のさらなる最適化が求められる。 
最後に、光応答性セレン修飾核酸の開発を行い、光照射による鎖切断反応を実証
した。この化学修飾を用いて、光照射によって切断可能な環状 DNA の合成を目指
した実験では、部分的な成功が見られたが、DNA リガーゼを用いたライゲーション
が進行しないという課題が残された。この結果は、光応答性核酸の新たな応用可能
性を示す一方で、さらなる改良が必要であることを示している。 
これらの研究成果は、核酸医薬の分野における基盤の一端を形成するものであり、
薬理効果や安定性の向上に寄与する可能性がある。しかし、各研究においていくつ
かの課題が明確化されたことから、独創性と実用化に向けた完成度の両面で改良の
余地があると考えられる。プレゼンテーションでは、研究内容をおおむね明確かつ
論理的に説明しており、成果の意義を伝える努力が見られた。 
総括すると、申請者は新たな核酸誘導体の開発を通じて、各種核酸医薬の高機能化
に一定の成果を上げた。医薬化学の観点から実用化に向けては、さらなる活性向上、
合成法の効率化、安全性の向上が求められるものの、本研究は新たな視点に基づき、
核酸医薬の発展に一定の貢献を果たしたと考えられる。以上の理由により、申請者は
博士(理学)の学位を授与される十分な資格があるものと認められる。

This thesis investigates the ferroelectric properties of Dion-Jacobson-type layered 
perovskites and exfoliated 2D nanosheets, presenting a systematic study that brings a 
bridge from three-dimensional bulk systems to two-dimensional nanosheets. The work 
explores the structural, electrical, and ferroelectric characteristics of these materials, 
aiming to unlock their potential for advanced microelectronic applications.  
Chapter 1 provides a foundational introduction to ferroelectricity, discussing its 
underlying mechanisms, categories, and unique behaviors, including polarization 
hysteresis and domain dynamics in Chapter 1.1. It delves into the structural 
characteristics of ABO3 perovskites and their ferroelectric behaviors in Chapter 1.2. 
Furthermore, the layered perovskites, such as Dion-Jacobson, Ruddlesden-Popper, and 
Aurivillius phases, were discussed regarding their sophisticated chemical compositions 
and adjustable structures in Chapter 1.3.  The chapter highlights the significance of 
size effects in ferroelectric materials, emphasizing the challenges and opportunities in 
nanoscale ferroelectrics in 1.4. The unique swelling property and exfoliability of 
nanosheets with the atomic-scale thickness nanosheets of the layered perovskites were 
introduced in 1.5. The Dion-Jacobson-phase perovskites with the monovalent interlayer

cations exhibit great potential to be exfoliated into their 2D forms, offering pathways to 
explore the tailored properties both in 3D bulk systems and 2D nanosheets. Under this 
motivation, the purpose of this research was represented in Chapter 1.6.  
Chapter 2 investigates the lead-based Dion-Jacobson perovskite, CsPb2Nb3O10, 
detailing its synthesis through solid-state reactions and its structural characterization 
via X-ray diffraction, Raman spectroscopy, and second harmonic generation (SHG). 
CsPb2Nb3O10 crystallizes in a tetragonal system with a polar space group P4mm. The 
intrinsic ferroelectric properties are explored, revealing a spontaneous polarization of 
7.93 μC cm-² and a phase transition at 260°C. The chapter underscores the material's 
out-of-plane polarization and its unique characteristic with the negligible contribution 
of Pb2+ to the polarization compared to other Bi- and Pb-based layered ferroelectrics, 
indicating the potential of the CsPb2Nb3O10 as a robust ferroelectric material, providing 
opportunities for further investigation of ferroelectrics in its 2D forms. 
Chapter 3 transitions to the mechanical exfoliation of CsPb2Nb3O10, analyzing the 
thickness-dependent ferroelectric behavior of the exfoliated nanosheets. This chapter 
starts from the argument of the exfoliability of van der Waals and non-van der Waals 
materials, pushing the boundaries of the difficulties of the mechanical exfoliation of 
non-van der Waals materials, in which CsPb2Nb3O10 was examined as a potential 
exfoliable material. As expected, CsPb2Nb3O10 exhibited high cleavability with no 
remarkable variation in the structure and composition after the mechanical exfoliation 
process. The ferroelectric properties of the nanosheets with thicknesses ranging from 
7.5 nm to 50 nm were characterized. The results highlight the ability of the exfoliated 
nanosheets to retain ferroelectric behavior at the nanoscale and exhibit 
thickness-dependent properties, paving the way for applications in 2D ferroelectric 
devices. 
Chapter 4 explores the chemical exfoliation of Pb2Nb3O10 nanosheets. The synthesis, 
structural analysis, and ferroelectric characterization of these nanosheets are presented, 
showcasing their tunable properties and the advantages of chemical exfoliation in 
producing high-quality nanoscale ferroelectrics. Pb2Nb3O10 nanosheets retain 
ferroelectric behaviors even with an atomic thickness of 1.8 nm, pushing the boundaries 
of layered perovskite nanosheet thickness limitations. This chapter demonstrates the 
potential of chemically exfoliated nanosheets in flexible and miniaturized electronic 
devices.

Chapter 5 focuses on the fabrication and characterization of multilayer films of 
Pb2Nb3O10 nanosheets through layer-by-layer assembly techniques. The multilayer 
films [Pb2Nb3O10]n exhibited high quality with the individual nanosheets arranged edge 
to edge. PFM measurements verified the ferroelectric behavior of the films, revealing 
butterfly-shaped amplitude-voltage loops and 180° phase reversals indicative of 
polarization switching. Thickness-dependent electrical properties, including coercive 
voltage and withstanding voltage, were observed, highlighting the tunability of the 
films' properties. This chapter illustrates the potential of controlled assembly 
techniques in tailoring the properties of ferroelectric thin films for specific applications 
like ferroelectric tunnel junctions. 
Chapter 6 expands the scope to lead-free Dion-Jacobson-type perovskites, specifically 
K(Sr, Ca)m-1Nb3Tim-3O3m+1 (m=3~5) with the property tailoring by chemical 
manipulating and atomic-scale engineering. The synthesis, structural characterization, 
and exfoliation of these lead-free nanosheets are detailed. The nanosheets exhibit 
precise thickness control, corresponding to their respective m values, and maintain 
unique 2D morphologies with flat surfaces and distinct crystalline edges. Notably, 
Sr3Nb3TiO13 nanosheets (m=4) display both out-of-plane and in-plane ferroelectric 
signals, while Ca4Nb3Ti2O16 (m=5) nanosheets exhibit proper ferroelectricity with 
distinct out-of-plane polarization, highlighting the remarkable effect of atomic-scale 
engineering for properties tailoring, with applications in environmentally friendly 
electronic devices. 
Chapter 7 synthesizes the findings of the thesis, providing a comprehensive conclusion 
on the ferroelectric properties of Dion-Jacobson perovskites across dimensional scales. 
The chapter also offers an outlook on future directions, emphasizing the importance of 
advancing exfoliation techniques, exploring new material compositions, and integrating 
nanoscale ferroelectrics into next-generation microelectronic devices. 
This thesis advances the understanding of Dion-Jacobson-type perovskites, highlighting 
their unique properties and transformative potential in nanoscale ferroelectric 
applications. The findings offer valuable insights into the design and development of 
high-performance materials for energy storage, sensing, and miniaturized electronics.

 S＋3Eを前提としたカーボンニュートラルの 2050年実現に向けて、CO2排出量の多くを
占める電力部門と運輸部門における排出削減が求められている。電力部門では、2024 年時
点で日本の発電電力の 7 割程度を占める火力発電の CO2排出量削減が課題となっている。
特に、水素・アンモニア発電や炭素貯蔵・再利用を前提とした脱炭素型火力発電の高効率
化が重要である。一方で、運輸部門では、自動車の電動化が期待されている。これらの目
標を達成するためには、発電機用固定子コイルや、EV用パワーモジュールにおける固体絶
縁材料の絶縁破壊強度の向上が不可欠である。 
 固体絶縁材料の絶縁破壊強度を向上させる材料技術として、セラミックナノ粒子を分散
させたナノコンポジット化が注目されている。エポキシ樹脂にルチル晶酸化チタンナノ粒
子を添加することで、絶縁破壊強度が向上することが明らかになっている。しかしながら、
ナノコンポジットの絶縁破壊強度向上メカニズムは未解明であり、このメカニズムの解明
は、最適な材料選定や製造工程の最適化に資する。特に、大きな凝集体が残存していると、
ルチル晶酸化チタン/エポキシ樹脂ナノコンポジットの絶縁破壊強度は低下することが明ら
かにされている。そのため、粒子凝集を除いた状態での絶縁破壊強度の評価が必要である
が、ナノ粒子はサイズに対する表面積が大きく凝集しやすいため、粒子凝集を除いた状態
でナノ粒子材質の違いと絶縁破壊強度の関係は十分に解明されていない。 
また、エポキシ樹脂の絶縁破壊メカニズムとして電子なだれ破壊が想定される。この破

壊過程は、まず初めに材料内に電子が注入され、電界により加速されて格子に衝突して電
離を引き起こす過程が繰り返されることで電子密度が増倍する。電子の持つエネルギーに
よって樹脂分子鎖が切断され最終的に絶縁破壊に至る。しかし、破壊過程のうちどの要素
が絶縁破壊強度を決定する要因となるかは不明であり、破壊過程の各要素と絶縁破壊強度
との相関を取得する必要がある。さらに、ナノコンポジットの絶縁破壊強度向上メカニズ
ムを解明するためには、各要素においてナノ粒子が及ぼす影響を明確にすることが必要で
ある。 
 そこで、本研究では、ナノコンポジットの絶縁破壊強度向上メカニズムを解明すること
を目的とし、ナノコンポジットの絶縁破壊強度と粒子材質の関係、絶縁破壊に至る過程の
各要素に与えるナノ粒子の働き、破壊過程の各要素と絶縁破壊強度との相関を実験的に評
価した。得られた実験結果を統合し、ナノコンポジット化による絶縁破壊強度向上メカニ
ズムを提唱した。 
 第 2 章では、ナノコンポジットの絶縁破壊強度に対する粒子材質の違いを評価する上で
必要不可欠な粒子凝集を除去したナノコンポジットを作製し、作製した試料の粒子分散状
態を、走査型電子顕微鏡（SEM）と粒子分散処理後の粒子充填率により評価した。用いた
ナノ粒子は、シリカ、アルミナ、アナターゼ晶酸化チタン、ルチル晶酸化チタンの 4 種類
とし、超音波照射と遠心分離法を用いて、ナノ粒子の分散処理を行った。超音波照射を用
いることでエポキシ樹脂ナノコンポジット内部のナノ粒子凝集体を解砕することができる。
遠心分離法では、試料に遠心力を加えることで残存している凝集体を沈めて分離すること
ができる。SEM画像の観察結果からいずれの材質のナノ粒子においても、分散処理を施し
たナノコンポジットでは凝集体が除かれて、ナノ粒子が単分散していることを確認した。
また、分散処理後の各種ナノコンポジットの粒子充填率は 0.1~0.3 vol%と同程度であり、
粒子材質の違いを評価するためのナノコンポジット試料を作製できた。 
 第 3 章では、絶縁破壊強度が向上するナノ粒子材質および粒子分散状態を明らかにする
ため、第 2 章で作製したナノコンポジット試料を用いて、ナノ粒子材質および粒子分散状
態の異なるエポキシ樹脂ナノコンポジットのインパルス絶縁破壊強度を評価した。その結
果、ナノ粒子凝集体が分離・除去されると、無充填エポキシ樹脂に比べて低電界における
絶縁破壊確率が低下し、絶縁破壊が起こりにくくなる。また、ルチル晶酸化チタンはエポ
キシ樹脂に比べたインパルス絶縁破壊強度の向上効果を示し、この効果がルチル晶酸化チ
タン特有であることを明らかにした。次に、インパルス絶縁破壊強度の向上効果が得られ
たルチル晶酸化チタン/エポキシ樹脂ナノコンポジットにおいて、発電機の運転電圧である
交流電圧下における絶縁破壊強度向上効果を明らかにするため、粒子分散状態を変化させ
たナノコンポジットを作製し、交流絶縁破壊強度を評価した。その結果、ナノ粒子を分散
させることで、インパルス絶縁破壊強度と同様に交流絶縁破壊強度の向上が確認された。
以上のことから、エポキシ樹脂ナノコンポジットの絶縁破壊強度が向上するナノ粒子材質
および粒子分散状態を明確にできた。この結果に基づき、次章以降で絶縁破壊に至る過程

の各要素に与えるナノ粒子の働きを検討するため、絶縁破壊強度が向上するルチル晶酸化
チタンにおいてその粒子分散状態を変化させた試料を用いた。 
 第 4 章では、絶縁破壊強度向上メカニズムの解明に向けて、電界発光特性を測定するこ
とで、絶縁破壊に至る初期段階で発生する電荷注入現象について評価した。粒子分散状態
の異なるルチル晶酸化チタン/エポキシ樹脂ナノコンポジットを用いて電界発光開始・消滅
電界を求め、ナノ粒子および凝集体が電界発光開始・消滅電界の値を変化させる要因を電
子の得るエネルギーの観点から検討した。その結果、凝集体を含むナノコンポジットでは、
電界発光開始・消滅電界が無充填エポキシ樹脂よりも低いことを明らかにした。これは、
凝集体周囲の広い高電界領域により、電子が高いエネルギーを得るためである可能性を示
した。一方、分散状態が良好なナノコンポジットでは電界発光開始・消滅電界が無充填エ
ポキシ樹脂よりも高いことを明らかにした。これは、分散ナノ粒子が電子の移動を妨げ、
エネルギーを減少させるためである可能性を示した。以上のことから、絶縁破壊に至る初
期段階で発生する電荷注入現象においてナノコンポジット中に注入された電子のエネルギ
ーは、分散ナノ粒子の存在により減少し、凝集体の存在により増加する可能性を明らかに
した。 
 第 5 章では、絶縁破壊強度向上メカニズムの解明に向けて、熱刺激電流（TSC）特性、
ガラス転移温度を測定することで、電荷のトラップや樹脂分子鎖の運動について評価した。
TSC 測定時のコレクティング電界印加時において生じる 2 つのピーク波形が双極子の脱分
極か、トラップ電荷の脱トラップのどちらに起因するかをコレクティング電界の有無やガ
ラス転移温度の実験結果を比較することで明らかにし、TSC 波形から脱分極、脱トラップ
に必要なエネルギーをそれぞれ推定することで各エネルギーにナノ粒子および凝集体が及
ぼす効果を検討した。その結果、低温側の TSCピークは樹脂分子運動の活発化に伴う双極
子の脱分極に、高温側の TSCピークはトラップ電荷の脱トラップに起因することを明らか
にした。分散ナノ粒子は、ガラス転移温度の低下や双極子脱分極の活性化エネルギーの増
加を引き起こすことを明らかにした。さらに、高温側の TSC波形の解析から、分散ナノ粒
子の添加がエポキシ樹脂に浅いトラップを新たに生じさせることを示した。一方、凝集体
の存在は、ガラス転移温度を上昇させる、またはナノ粒子添加によるガラス転移温度低下
効果を妨げることを明らかにした。さらに、低温側の TSC波形の解析から、凝集体が双極
子の脱分極の活性化エネルギーを減少させることを示し、高温側の TSC 波形の解析から、
凝集体がエポキシ樹脂に深いトラップを新たに生じさせることを示した。以上のことから、
絶縁破壊過程のうち、電子加速に関連する電荷のトラップ、樹脂分子鎖の切断に関連する
樹脂分子鎖の運動に対して、分散ナノ粒子は浅いトラップを導入、樹脂分子鎖の運動を抑
制し、凝集体は深いトラップを導入、樹脂分子鎖の運動を促進させることを明らかにした。 
 第 6 章では、前章までに得られた実験結果をもとに絶縁破壊メカニズムを考察し、分散
ナノ粒子および凝集体が絶縁破壊強度を変化させるメカニズムを検討した。絶縁破壊過程
の各要素と絶縁破壊強度の相関を取得することで、絶縁破壊強度に強く関連する要素を明

らかにし、それをもとに絶縁破壊メカニズムを考察した。その結果、電界発光開始電界と
双極子脱分極の活性化エネルギーが絶縁破壊強度と強く相関することを明らかにし、以下
の絶縁破壊メカニズムを示した。（1）まず、絶縁材料内に電子が注入されて高電界中を移
動することで電子が高エネルギー状態に遷移する。（2）そして、その高エネルギー状態の
電子からエポキシ樹脂分子がエネルギーを得ることで、樹脂分子鎖が変位する。これによ
って、樹脂分子間の距離の増加や分子鎖の切断が生じることで、電子移動が容易になり絶
縁破壊が生じる可能性が考えられる。分散ナノ粒子による絶縁破壊強度向上メカニズムと
しては、以下の可能性を示した。上述した破壊メカニズムのうち(1)の過程で、分散ナノ粒
子が電子移動を妨げ、高エネルギー化を阻害することと、(2)の過程で、樹脂と粒子の強い
結びつきにより樹脂分子鎖の変位に必要なエネルギーが増加する。一方、凝集体による絶
縁破壊強度低下メカニズムとしては、凝集体周囲の広い高電界領域で電子が高エネルギー
化し、高エネルギー化した電子がエポキシ樹脂中の双極子の回転を促進することで、樹脂
分子間の距離の増加や分子鎖の切断を低電界で引き起こす。さらに、各実験項目と絶縁破
壊強度を比較し、絶縁破壊強度の予測式を算出した結果、電界発光や TSCといった非破壊
試験を用いて絶縁破壊強度を線形近似で表現できる可能性を示した。 
以上より、本研究では絶縁破壊過程の各要素に関する特性評価を行い、それらの実験結
果からルチル晶酸化チタン/エポキシ樹脂ナノコンポジットの絶縁破壊強度向上メカニズム
を明らかにした。この研究成果は、メカニズムをもとにしたより高い絶縁破壊強度を有す
るエポキシ樹脂ナノコンポジット材料の開発に寄与するものと考えられる。

澤田悟君提出の博士論文「回転デトネーションエンジンの推力軸周りに発生するトルクの機構解明と方向制御」
では，実験計測の結果を基に，回転デトネーションエンジン（RDE）の推力軸周りに発生するトルクの生成機構
および特性を考察している．また，このトルクの源となる回転デトネーション波の伝播方向の制御を通して，トル
クの効果的な生成および方向の制御を実験的に示している．各章の概要は以下の通りである．
[1] 第１章では，序論として，デトネーションの特性，デトネーションを熱機関に応用した際の利点，その熱機関
の一種でありなおかつ本論文で対象とするRDEの概要，その内部で周回伝播する回転デトネーション波の伝播特
性，それに伴う周方向の流れおよびRDEに加わる力の特性について述べられている．
[2] 第２章では，RDE内部の円環経路上を回転デトネーション波が周回伝播することによって誘起される周方向の
流れ，並びに，その流れが燃焼室壁面に与える周方向の摩擦の特性を簡易的な計算モデルから考察されている．そ
して，回転デトネーション波の近傍での流れは，顕著に大きな周方向の運動量を有し，これによって周囲の燃焼室
壁面には，回転デトネーション波の伝播方向と同方向の強い剪断応力が作用することが示されている．さらに，そ
の機構を基に，燃焼室壁面に作用する周方向の摩擦力の方向と回転デトネーション波の伝播方向が一致すること，
供給条件および回転デトネーション波の伝播形態に対する周方向の摩擦の特性について整理されている．
[3] 第３章では，RDEの推力軸周りのトルク・推力の実験計測に用いた機器（六軸力覚センサ），燃焼室内部を撮
影した画像から回転デトネーション波の伝播形態を評価する手法に関して，それらの原理，並びに，それらの手法
を用いることが十分適切であったことが示されている．
[4] 第４章では，RDEの作動試験の中で，その推力軸周りのトルクを実験計測した結果が示されている．そして，
第２章の計算で示された通り，燃焼室壁面に生じる周方向の摩擦力の方向が，回転デトネーション波の伝播方向と
一致し，それに応じたトルクがRDEに発生することが実験的に示されている．加えて，宇宙空間でのRDEの作動
試験においても，回転デトネーション波に由来するトルクが発生したこと，供試体の角速度がそれに応じて時間変
化したことが示されている．以上の実験結果から，回転デトネーション波の波面近傍での周方向の剪断応力が見積
もられており，地上・宇宙での試験並びに第２章での計算結果が互いに整合する結果であったことが示されてい
る．そして，周方向の流れおよび力が，RDEの推力性能に与える影響についても評価されており，およそ1%未満
と小さいこと，スロートの有無並びに背圧の低下に伴う周方向の流れ・力への寄与が小さいことが考察されてい
る．
[5] 第５章では，デトネーション領域と壁面の接触，並びに，回転デトネーション波の伝播速度がトルクの生成に
おいて重要であることが実験的に示されている．内壁を有する場合のRDEと比較して，内壁を取り外すことで燃
焼室空間を径方向に対しても大幅に拡大させた場合には，回転デトネーション波の伝播速度の二乗が増大し，デト
ネーションの領域と接触する壁面の範囲の減少と拮抗することで，内壁の有無によるトルクの大きさの差異が小さ
いことが実験的かつ明確に示されている．
[6] 第６章では，前章まで取り扱った摩擦由来のトルクとは異なり，螺旋状に分布する凸部を有した側壁およびそ
の凸部の前後で生じる圧力差を利用したトルクの生成機構について，実験結果を基に述べられている．そして，そ
のトルクの生成機構および特性，トルクの大きさの向上が明確に示されている．加えて，上記の壁面形状を利用し
た回転デトネーション波の伝播方向・トルクの方向の制御を実験的かつ明確に達成しており，その機構についても
考察されている．
[7] 第７章では，本研究の結論を示している．
以上のように本論文では，回転デトネーション波に由来して周方向の力を生成する上で，波面近傍での流れが有
する周方向の運動量・デトネーション領域と接触する壁面の範囲が重要であることが明らかにされている．また，
周方向の力が発生したとしても，RDEの軸方向推力を十分担保し得る結果が示されている．その一方で，実験計
測の結果を基に，トルクの大きさの向上・方向の制御の手法が明確に示され，RDEに生じるトルクの活用方法が
示されている．さらに，本論文で示された回転デトネーション波の伝播方向の制御は，これまで長年の間，デト
ネーションエンジンの研究分野で未達成であり，高い新規性を有する．本論文は，RDEの周方向の力という観点
から実験的にRDE内部の物理構造を深く研究しており，デトネーションの基礎物理の解明，応用の実現性の向上
に大きく貢献するものであると判断できる．よって，本論文の提出者である澤田悟君は博士（工学）の学位を授与
されるに十分な資格があると本論文審査委員会は判断する．

The Finite-Difference Time-Domain (FDTD) method, developed by K. S. Yee in 1966, is 
a numerical method for solving the time evolution of electromagnetic fields by 
approximating Maxwell’s equations in both time and space with the finite difference of 
the second-order accuracy. A staggered grid (Yee grid) system which ensures that 
Gauss’s law for both electric and magnetic fields is always satisfied. Owing to this 
advantage and the simplicity of the numerical algorithm, the FDTD method has been 
regarded as the standard numerical method for electromagnetic fields for over half 
century. In plasma kinetic simulations, the FDTD method is used as a numerical 
method for electromagnetic analyses. This method is also used for acoustic analyses. 
However, the FDTD method has limitations, particularly in the occurrence of 
numerical oscillations in discontinuous waveforms and continuous waveforms with 
steep gradients. The numerical oscillations are caused by an error in the phase velocity, 
which depends on frequency and wavenumber. To reduce the numerical error in the 
phase velocity, higher-order finite differences are used. The FDTD(2,4) method uses the 
fourth-order spatial difference. However, the Courant condition becomes more restricted 
by using higher-order finite differences in space and a larger number of dimensions (𝐶 =
𝑐∆𝑡/∆𝑥 < 1/√𝑛  and 𝐶 < 6/7√𝑛  in 𝑛  dimensions with FDTD(2,2) and FDTD(2,4), 
respectively), resulting in more consumption of computational time.

In the present study, new explicit and non-dissipative FDTD schemes are 
proposed in two and three dimensions, which relaxes the Courant condition and reduces 
numerical errors. The proposed schemes are developed by adding higher-degree 
difference operators with optimized coefficients to the time-development equations of 
the conventional schemes. The time-development equations of the present schemes are 
based on the Taylor expansion of Maxwell’s equations, which incorporating odd-degree 
difference operators only. One-dimensional and Laplacian types of the higher-degree 
difference operators are considered for relaxation of the Courant condition and 
reduction of numerical errors. 
First, the one-dimensional third-degree difference operators are added to the 
time-development equations of FDTD(2,4) with coefficients. These operators, with 
second- and fourth-order accuracies, are optimized using dispersion relations derived 
via Fourier analysis. The Courant conditions can be relaxed and numerical errors can 
be reduced by adjusting the coefficients. An optimal coefficient of the third-degree 
difference operators is obtained with brute-force search using the dispersion relations so 
that numerical instabilities are suppressed and numerical errors are minimized in the 
entire wavenumber space. These schemes are stable with large Courant numbers up to 
1 in both two and three dimensions. The numerical errors of the present schemes are 
smaller than those at small Courant numbers where the conventional schemes are 
stable (i.e., 0.65 times smaller in three dimensions). At large Courant numbers, however, 
the large numerical errors remain with the present schemes based on FDTD(2,4). The 
numerical dispersion relations show that there exists a large anisotropy in the phase 
velocity errors with the present schemes. Test simulations show that anisotropic 
numerical oscillations appear. 
Second, the one-dimensional third- and fifth-degree difference operators are 
added to the FDTD(2,6) scheme. Following two schemes are developed: FDTD(2,6) with 
fourth-order third-degree difference operators, and FDTD(2,6) with fourth-order 
third-degree and second-order fifth-degree difference operators. The dispersion relations 
of the schemes based on FDTD(2,6) are derived in the same way as those based on 
FDTD(2,4). A set of optimal coefficients of the third- and fifth-degree difference 
operators are obtained with brute-force search. These schemes are also stable with large 
Courant numbers up to 1. The numerical errors of the present schemes based on 
FDTD(2,6) are not reduced significantly. The numerical dispersion relations show that 
the schemes based on FDTD(2,6) has only about 0.9 times smaller numerical errors 
than those based on FDTD(2,4), although the numerical accuracies of the difference 
operators are higher. Therefore, a straightforward extension of the present schemes to

FDTD(2,8) is not effective. 
Third, third-degree Laplacian operators, which are composed of first-degree 
and Laplacian differences, are added to the time-development equations of FDTD(2,4) 
with coefficients. Third-degree difference operators with second- and fourth-order 
accuracies are considered. The dispersion relations of the schemes with Laplacian 
operators (L-schemes) are derived in the same way as the schemes with 
one-dimensional difference operators (D-schemes). A set of optimal coefficients is also 
searched in the same way. The numerical dispersion relations show that the 
anisotropies are reduced with the schemes with Laplacian operators. The numerical 
errors of L-schemes are about a half of those of D-schemes. Test simulations show that 
L-schemes suppress waveform anisotropies and numerical oscillations more effectively 
than D-schemes. For further reduction of numerical errors, other higher-degree 
difference operators including Laplacian need to be appended to the time-development 
equations as well as the third-degree difference operators. 
The computational time with the present schemes for each time step are longer 
than those with the conventional scheme because the computational time increases as 
the number of operations increase. However, the present schemes are stable at large 
Courant numbers. The total computational time with the present schemes are 
0.54–0.84 and 0.54–0.67 times shorter than those with the conventional scheme in two 
and three dimensions, respectively. 
These novel FDTD schemes are well-suited for applications electromagnetic, 
optical and acoustic analyses, offering improved efficiency without compromising 
accuracy. Furthermore, the way of the relaxation of the Courant condition is able to be 
applied to other explicit numerical methods. 
The present schemes are able to be applied into plasma kinetic simulations. In 
plasma kinetic simulations, where the computational cost for particles far exceeds that 
for electromagnetic fields, the proposed schemes significantly reduce overall 
computational time in nearly the same ratio with extensions of time steps. For the 
implementation, a correction of current density is necessary to suppress numerical 
errors in the charge conservation law, which is left numerical errors, is left as a future 
study.

A power semiconductor module consists of a few switching devices to realize high 
power density and good thermal performance. Recent technical trends have introduced 
wide bandgap switching devices such as silicon carbide (SiC) and gallium nitride (GaN), 
providing high switching frequency and high switching speed. These switching devices 
improve the efficiency of power electronic systems with their superior switching 
characteristics. However, high dv/dt and di/dt of switching devices generate 
considerable electromagnetic interference such as conducted common-mode noise, 
which requires careful designs to minimize parasitic inductance and capacitance of 
power modules. Since the power module works as a noise source and introduces 
common-mode noise propagation path, various studies have been conducted to reduce 
electromagnetic interference by power module design.  
Chapter 1 introduces the architectures and packaging methods of power 
semiconductor modules. Noise propagation in the power modules are described and 
various state-of-the-art power modules and literature are reviewed. The objective and 
the outline of this dissertation are finally explained. 
Chapter 2 shows the proposed power module packaging method to reduce 
conducted common-mode noise. This study realizes low common-mode capacitance by

etching a specific portion of bottom copper layer in a substrate. The proposed power 
module does not compromise size, thermal performance, and top copper pattern of 
power modules. The etched region is filled with air which has the lowest permittivity in 
Chapter 2. An epoxy is applied to support the substrate and to prevent encapsulant 
from entering the etched region. Thermal design and prototyping of the proposed power 
module are elaborated. The common-mode noise reduction is experimentally verified, 
and the same thermal performance is also confirmed by its structure function. However, 
high thermal stress to the baseplate solder is observed by thermal cycling tests, though 
the power module effectively reduced the CM noise.  
Chapter 3 utilizes a low-permittivity epoxy instead of air to improve mechanical 
reliability. The thermal stress to the baseplate solder layer is decreased, maintaining 
large common-mode noise reduction. Since the air has the lowest permittivity, i.e., 1, the 
proposed power module shows slightly high CM capacitance. Manufacturing process 
and packaging method to inject the low-permittivity epoxy is also explained. Thermal 
cycling tests prove the higher mechanical reliability. 
The power modules proposed in Chapter 2 and Chapter 3 requires additional 
manufacturing process for the epoxy. Chapter 4 utilizes silicone gel to fill the etched 
region using conventional encapsulation stage. This method does not need additional 
manufacturing process. Since silicone gel is a soft material and has high surface 
tackiness and low viscosity, it provides higher manufacturability than using epoxy. No 
cracks, delamination, and voids are observed even after the thermal cycling tests, 
showing almost equivalent common-mode noise reduction. 
A different design approach with a new vertical shunt resistor is introduced in 
Chapter 5. A new configuration of the shunt resistor is designed and this vertical shunt 
resistor is packaged beneath the lead of the power module. It does not increase the size 
of the substrate or common-mode capacitance and realizes current sensing. Several 
advantages such as low equivalent series inductance, high thermal resistance, and 
materials are also presented. Design guidelines for sensing points are also discussed. 
 Chapter 6 concludes this dissertation. In addition to silicone gel used in 
Chapter 4, epoxy encapsulation is considered for the future works. Bottlenecks 
experienced in this study are introduced and their reasons are investigated. A new 
encapsulation method of transfer-molding and a new substrate design are suggested to 
improve the power module packaging.

 新美陽子は、イネの節間伸長機構を解明するため、透明化技術、局所的遺伝子発現
解析、ホルモン定量、Click-iT EdU イメージングアッセイなどを組み合わせ、成長ステ
ージの移行過程における時空間的な解析を行い、ジベレリン（GA）生合成遺伝子の発
現調節、GA の輸送、および GA 感受性の時空間的な制御によって、イネの茎伸長が
精緻に調節されている可能性を示した。以下にその要旨を記載する。 
 
 作物の短稈性は耐倒伏性を付与する重要な農業形質であり、茎伸長に関する研究は
これまでに数多く行われてきた。特に「緑の革命」において利用された半矮性品種の
作出は、イネやコムギにおける最も著名な育種例である。化学肥料の普及により多収
が可能となった一方で、過剰な茎伸長が倒伏を引き起こすという問題が発生した。こ
の課題を解決するために短稈で耐倒伏性の品種が開発され、現在も広く利用されてい
る。これらの品種育成に用いられた遺伝子（イネ：sd1、コムギ：Rht-B1b、Rht-D1b）
が現在も育種に用いられていることは、茎制御が農業において極めて重要であること
を物語っている。 
イネの節間伸長には植物ホルモンである GA が重要な役割を果たすことが知られてお
り、GA と茎伸長の関係については多くの知見が蓄積されている（Sasaki et al., 2002; 
Fukazawa et al., 2017; Nagai et al., 2020）。矮性変異体の研究からは GA 生合成酵素
をコードする遺伝子（Sakamoto et al., 2004）や GA の情報伝達経路（Ueguchi-Tanaka 
et al., 2005）についても明らかにされてきた。しかし、GA と茎伸長の関係には未解
明な点が多く残されている。本研究では、イネの節間伸長機構を解明するため、透明
化技術、局所的遺伝子発現解析、ホルモン定量、Click-iT EdU イメージングアッセイ
などを組み合わせ、成長ステージの移行過程における時空間的な解析を行った。 
 
第二章 短日条件下でのイネの実験系構築 
本研究では、バイオトロンを用いた短日条件（10 時間明期/14 時間暗期）で Nipponbare
（Oryza sativa cv.）を育成し、約 45 日で出穂させる実験系を構築した。この系では
6 葉齢で相転換が起こり、11 葉齢が止め葉となる。この環境を用いることで安定的か
つ効率的に研究を進めることが可能となった。 
花芽形成遺伝子 OsMADS15 の発現を指標とする蛍光マーカーmOrange を導入した
形質転換体イネを用い、透明化技術を用いた観察により相転換期を 6 葉齢と特定した。
また、透明化技術を最適化することで、茎や葉の連続的な立体観察を実現した（Niimi 
et al., 2022）。

第三章 GA 生合成遺伝子の発現動態と節間伸長 
相転換前後における GA 生合成遺伝子の発現動態を解析するため、3～9 葉齢の茎頂メ
リステム（SAM）や葉身をサンプリングし、次世代シーケンサーによる遺伝子発現解
析を実施した。CRISPR-Cas9 による機能破壊株の解析を通じて、GA20ox1、GA20ox2、
GA3ox2 を節間伸長に寄与する遺伝子候補として同定した。 
局所的発現解析では、8 葉齢の幼穂で GA 生合成遺伝子が発現し、上位節間や維管束
周辺でも弱い発現が確認されたが、11 葉齢（止め葉期）では発現が低下していた。ホ
ルモン定量では 6 葉齢や 8 葉齢での GA 濃度上昇は確認されず、10 葉齢や 11 葉齢で
も大きな変化は見られなかった。活性型 GA として検出されたのは開花期の葯におけ
る GA4 と GA7 のみであった。これらの結果から、節間伸長には葯（穂）で生成され

第四章 節間伸長過程における細胞分裂と細胞伸長 
Click-iT EdU イメージングアッセイおよびトリパンブルー染色を用い、伸長中の節間
での髄腔形成および介在分裂組織の局在を可視化した。これにより、節間伸長過程に
おける細胞分裂および細胞伸長の空間的・時間的動態を明らかにした。 
 
本研究の意義と展望 
本研究では、透明化技術、局所的遺伝子発現解析、ホルモン定量、Click-iT EdU イメ
ージングアッセイなど多角的な手法を用い、成長ステージにおける時空間的な解析を
行った。このアプローチにより、節間伸長に関与する分子メカニズムの新たな知見を
得た。イネの成長過程では形態変化に伴い遺伝子発現やホルモン濃度が大きく変動す
るため、従来の点的・線的な解析にとどまらず、立体的かつ包括的なアプローチを取
ることで理解が深まることが示された。得られた知見は、イネだけでなく他の作物種
の成長制御にも応用可能であり、今後の植物科学および農業生物学における研究に貢
献することが期待される。さらに本研究で示唆された仮説の検証を進め、イネ節間伸
長機構のさらなる解明を目指していく。 
 
したがって、本委員会は本論文が博士（農学）の学位論文として十分な価値を有す
ると認め、論文審査に合格と判定した。
