曽我恭平君提出の論文「AMPA型グルタミン酸受容体の機能解析を指向した可視化および光活性制御技術の開
発」は、神経伝達物質受容体であるAMPA型グルタミン酸受容体（以下、AMPA受容体）を標的とし、可視化プ
ローブの開発と、時空間分解能高く光活性制御が可能なケミカルツールの開発について論じている。各章の概要は
以下の通りである。
第1章では、本研究に関連する研究背景について述べている。はじめに、神経科学分野においてAMPA受容体の
機能解析が重要であることを示した。次に、AMPA受容体の可視化および活性制御法の従来法を概観し、利点と問
題点について説明した。機能解析のためには遺伝子工学的手法では不十分であることを説明し、本論文では小分子
ベースのツール開発を行うことを示した。
第2章では、AMPA受容体を迅速に可視化するための新規ケミカルプローブの開発について述べている。AMPA
受容体のリガンドをベースにしたプローブのデザインを行い、使用する色素によってAMPA受容体に対する親和性
が異なることを明らかにした。そのメカニズムを解明するために、X線結晶構造解析を行った。その結果から、色
素と受容体が相互作用していることを示した。X線結晶構造解析は、名古屋大学の卓越大学院プログラム「トラン
スフォーマティブ化学生命融合研究大学院プログラム（GTR）」の融合研究の一環として、東北大学多元物質科学
研究所の南後研究室とともに行われた。
第3章では、開発した可視化プローブを利用して、神経細胞に内在的に発現するAMPA受容体の可視化について
述べている。最初に、AMPA受容体を迅速かつ可逆的に標識できることを示し、本手法の利便性を述べた。さら
に、記憶・学習の基盤である長期増強時において、AMPA受容体がシナプスに集積する現象を捉えることに成功し
た。最後に、過去に開発した別の可視化方法と組み合わせることで、これまで理解が不十分であったAMPA受容体
の動態変化メカニズムの解析を行なった。その結果、AMPA受容体は主にエキソサイトーシスによってシナプスに
集積することを明らかとした。
第4章では、時空間分解能高くAMPA受容体活性を制御可能な、新規光活性制御技術の開発について述べてい
る。化合物の電子求引性に着目したユニークなデザインを行い、光で親和性が低下する光分解性アンタゴニストの
開発を行なった。光分解性アンタゴニストと、従来型のケージド化合物を組み合わせることで空間分解能高い活性
制御が可能であることを示した。最後に、海馬培養神経細胞を使用し、1細胞選択的な細胞の活性化が可能である
ことを示した。
　第5章では、本論文をまとめ、今後の展望について述べている。
以上のように本論文では、AMPA受容体の可視化を指向した、新規プローブの開発を行なった。本手法により、
従来なしえなかった内在のAMPA受容体の簡便な可視化が可能となった。また、光制御可能な新規分子を開発し、
AMPA受容体活性制御に新たな技術の提供を行った。本論文を通じて得られた結果は、ケミカルバイオロジーおよ
び神経科学分野の発展に寄与するものであると判断される。よって、本論文の提出者である曽我恭平君は博士（工
学）の学位を受けるに十分な資格があると判断した。

骨格筋は人体総質量の約 40％を占める最大の臓器の 1 つであり、運動器としての役割に
加えて、全身の代謝恒常性維持に重要なエネルギー代謝器官である。近年では、骨格筋は
マイオカインと呼ばれる生理活性物質を分泌する“内分泌器官”としても注目されており、
他臓器と相互に影響を及ぼすことで全身の健康維持に寄与している。これにより、骨格筋
は運動器や代謝器官としてだけでなく、免疫や神経、認知機能などにも関わる多様な健康
関連機能に寄与する臓器として再評価されている。実際に、骨格筋量や筋力は健康寿命と
密接に関連しており、筋量や筋力が低いと、死亡率や心血管疾患、生活機能低下のリスク
が高まることが報告されている。一方、骨格筋の減少は、運動不足、加齢、栄養不良、慢
性炎症、神経変性など様々な要因で生じる。特にサルコペニアは、加齢に伴う筋肉量や筋
力の低下を特徴とし、運動機能や生活の質の低下、転倒・骨折のリスク上昇、さらには死
亡率の増加など深刻な影響を及ぼす。日本の高齢者では約 8.2％がサルコペニアに該当し、
年間の累積発症率は 2.0％とされる。今後、認知症やフレイルとの関連も踏まえ、サルコペ
ニアの早期診断・予防・介入は、医療費の抑制や健康寿命の延伸に直結する重要課題とな
る。そのため、骨格筋機能の維持・向上に関する基礎研究と社会実装は、公衆衛生上、極
めて重要である。サルコペニアをはじめとする筋機能の低下に対する予防や介入手段とし
て、運動療法が広く推奨されている。しかしながら、高齢者にとっては運動実施のハード
ルが高く、継続的な介入が困難である場合も少なくない。そのため、日常の食事を通じて
骨格筋の機能低下を予防することは、現実的かつ重要なアプローチと考えられる。

近年、MPS（Microphysiological Systems）は、創薬研究や毒性評価、疾患モデリングの分
野で注目を集めている。MPS は、生体内に近い環境を再現した in vitro評価系であり、従来
の 2 次元培養と比べて、細胞や組織の機能的応答を引き出せることが特徴である。骨格筋
分野でも、3 次元構造や電気・機械刺激を活用した骨格筋 MPS の開発が進み、従来評価が
困難であった筋収縮力の定量化が可能となった。これにより、筋疾患やサルコペニアに対
する機能性食品成分や候補薬剤の有効性を、in vitro で評価する取り組みが加速している。
所属研究室では、骨格筋 MPSの筋収縮力を非侵襲的に評価するマイクロデバイスを開発し、
デキサメタゾンによる筋収縮力低下や、IGF-1による筋収縮力の回復を確認してきた。さら
に近年では、96 ウェルプレートの筋収縮力測定システムを構築し、抗筋萎縮ペプチドのス
クリーニングにも応用している。これらのことから、本マイクロデバイスを用いた骨格筋
MPS 評価系は、骨格筋に対する機能性成分の評価や探索への応用が期待される。 
そこで本研究の目的は、骨格筋MPS を用いて、サルコペニアなどに対する筋機能維持に
有効な食品成分を評価することである。本論文は、5章で構成されており、結果は第 2章か
ら第 4章の 3部で構成される。 
第 2 章では、L-アンセリンのヒト骨格筋細胞に対する作用を検証した。L-アンセリンは、
カルノシンやバレニンと同様にイミダゾールペプチドの一種であり、1929 年にガチョウか
ら初めて同定された。L-アンセリンは鳥類や、カジキやカツオといった遊泳能力の高い回遊
魚に豊富に含まれ、さまざまな食事から摂取されている。L-アンセリンは筋損傷の改善作用
や下肢筋力の増加作用が報告されており、筋機能の維持に寄与する可能性が示されている。
これらの知見から、L-アンセリンは筋分化や筋収縮力を高める可能性があると仮説を立て、
この仮説を検証するため、三次元培養におけるヒト骨格筋 MPS を構築し、L-アンセリンが
筋収縮力に及ぼす直接的な効果を評価した。まず初代ヒト骨格筋細胞を用いた平面培養に
おいて、qPCR および免疫染色を実施した。その結果、L-アンセリンは筋短径の増加および
筋分化を促進することが示唆された。さらに、骨格筋 MPS を用いた筋収縮力測定により、
L-アンセリンの筋収縮力増加作用が確認された。また、構成アミノ酸である β-アラニンお
よび 1-メチルヒスチジンでは効果が見られなかったことからジペプチド構造が重要である
可能性が示された。加えて、ヒスタミン受容体拮抗薬を用いた検討により、L-アンセリンの
筋収縮力増加作用および筋分化促進作用にはヒスタミン受容体 1（H1R）が関与する可能性
が示された。 
第 3 章では、多様な生理活性を有するポリフェノールであるケルセチンについて、ヒト
骨格筋細胞における筋線維タイプへの影響を検証した。ケルセチンは抗酸化・抗炎症作用
に加え、筋萎縮の抑制効果やミトコンドリア機能の活性化が報告されており、筋機能低下
の予防効果が期待される。まず、初代ヒト骨格筋細胞を用いた平面培養においてケルセチ
ンは、筋分化の促進および遅筋タイプのミオシン重鎖の発現を増加させることが示された。
さらに、AMPKの活性化作用および PGC1αの遺伝子発現増加が示され、AMPK/ PGC1α経
路を介して遅筋線維を誘導することが示唆された。また、ミトコンドリアスーパーコンプ

レックス形成および持久力に関与する Cox7A2Lの遺伝子発現を促進させ、ミトコンドリア
への酸素供給に関与する myoglobin（Mb）の遺伝子発現も増加させた。加えて、ヒト骨格筋
MPS を活用することで、ケルセチンが骨格筋細胞に作用し、Time to peak tension（TPT）お
よび Half relaxation time（1/2RT）、筋持久力を増加させ、結果として遅筋型筋線維様の表現
型を示すことが明らかとなった 
第 4章では、より生体模倣性の高い in vitroモデルの構築を目指し、初代ヒト骨格筋細胞
および高齢ヒト血清を用いた加齢性筋萎縮評価系を開発した。高齢ヒト血清は、従来の筋
萎縮誘導剤であるデキサメタゾンと同様に平面培養における筋萎縮および骨格筋 MPSにお
ける筋収縮力低下を誘導した。いずれの処置においても筋分解マーカーである Trim63 およ
び Fbxo32の遺伝子発現を増加させた。RNA-seq解析によりそのメカニズムの差異を検討し
たところ、高齢ヒト血清は主に NF-κB 経路を介して作用する一方、デキサメタゾンは FoxO
経路を介していた。実際に qPCR を用いた遺伝子発現評価において、高齢ヒト血清は NF-κB
経路の下流にある炎症マーカー（IL-1β、IL-6、CXCL8 および PTGS2）の遺伝子発現を増加
させたが、デキサメタゾンは CXCL8 および PTGS2 の発現には影響せず、IL-1β および IL-6
の遺伝子発現を低下させた。また、高齢ヒト血清は FoxO1 の遺伝子発現に影響せず、FoxO3
の遺伝子発現をわずかに増加させたが、デキサメタゾンはいずれの遺伝子発現も促進させ
た。タンパク質のリン酸化を評価したところ、高齢ヒト血清は NF-κB p65と IκBα のリン酸
化を促進した。これらのことから高齢ヒト血清は NF-κB 経路を介して筋萎縮を誘導する可
能性が示された。さらに、ケルセチンによる評価では、平面培養における筋萎縮および骨
格筋 MPS における筋収縮力低下を抑制した。メカニズムを検討した結果、ケルセチンは
NF-κB 経路の抑制を介し、炎症および筋萎縮マーカーの発現を抑制することにより、筋萎
縮および筋収縮力低下を抑制する効果が認められた。これらのことから本評価系は、生体
模倣性の高い加齢性筋萎縮モデルとして、食品成分の有効性評価への応用の可能性が示さ
れた。 
第 5章では、以上の結果を総括し、ヒト骨格筋細胞および骨格筋 MPS を用いた評価系が、
サルコペニアの予防やパフォーマンス向上を目的とした食品成分のスクリーニングや作用
機序解明に有効であることを改めて述べた。また、L-アンセリンとケルセチンに着目し、初
代ヒト骨格筋細胞や骨格筋 MPS、またヒト血清を用いて筋収縮力や収縮特性に及ぼす影響
を評価した結果について考察した。本アプローチはこれまでのサルコペニアや筋萎縮に対
する in vitro評価で課題となっていた「げっ歯類とヒトの種差による反応性の違い」や「従
来の平面培養系では筋収縮評価が困難さ」を克服した研究である。ヒト骨格筋細胞および
骨格筋MPS を用いた in vitro 評価を用いることで、サルコペニア対策に寄与する様々な食品
成分の評価が可能になると考えられる。また、本結果で得られた L-アンセリンおよびケル
セチンについては、サルコペニアの予防やパフォーマンスの向上を目的とした食事・治療
戦略において、有望な介入手段となる可能性がある。骨格筋MPS 技術は現在も成長を続け
る分野であり、本研究がMPS 分野の発展に寄与し、さらには、有効性成分の探索に貢献す

ることを期待する。これにより、サルコペニア対策として様々な食品成分が見出され、筋
機能低下に対する有効な介入策が見出されることを願う。

刘 媛媛氏の博士論文「Synthesis of Nanoparticles with Hierarchical Structure for Oxygen Reduction Reaction
Catalysts(階層構造を有するナノ粒子の合成と酸素還元反応触媒への応用)」は、水素社会に不可欠なPEMFCの触媒に
関する研究であり、酸素還元および水素酸化反応に関するカーボンシェル触媒などの合成と特性評価を実施している。
第1章では、PEMFCの動作原理と研究動向を概観し、ORRの遅い反応速度が性能向上を制約するボトルネックであること
を明確化している。さらに、触媒の組成、構造、電気化学特性に関する最新の文献をレビューし、特に白金系触媒の研究進
展と課題を整理した。最後に本論文の研究目的と章立てを提示し、以降の議論の基盤を築いている。
第2章では、SPスパッタリング法を用いたPt系触媒の作製可能性を検討し、パルス電流強度、放電時間、電解液温度といっ
たプロセスパラメータが粒子径や分布に及ぼす影響を解析している。また、Ni(NO₃)₂やCo(NO₃)₂などの電解液中金属イオン
が触媒形成とORR性能に与える効果を評価し、金属ドーピングが性能向上に寄与する可能性を示している。さらに、PtIr合
金ナノ粒子の合成を試み、後続の三元合金研究の基盤を確立している。
第3章では、SPスパッタリングにより調製したPt触媒の構造・電気化学特性を、pH 4.4・7・8・11の電解質中で比較した。特
にpH 8で合成した触媒は表面炭素層構造を形成し、ラマン分光、TEM、XPS、XAFSによる評価でその特性を明らかにして
いる。その結果、pH 8触媒は質量活性約500 mA/mgを示し、市販Pt/C触媒（256 mA/mg）の約2倍に達している。
第4章では、PtPd-PtIr合金電極を用いたSPスパッタリング法により、PtPdIr三元合金触媒を合成した。Ir含有量は電極組
成に比例して増加し、約10％で安定化することを確認している。得られたナノ粒子は1–3 nmに制御可能で、炭素担体上に
均一分散した。粒子-c（Pt:Pd:Ir = 79:14:7）はORR・HORの双方で最適な性能を示し、XPS解析によりPd(0)/Pd(II)のバ
ランスが電子構造を調節して性能向上に寄与すること得ている。
第5章では、電解沈積法によるPtCu合金触媒の調製を行い、Cu²⁺濃度が沈積挙動および生成物組成に与える影響を系統
的に解析している。さらに、0.1 M HClO₄および0.1 M KOH中でのORR特性をCV・LSVにより評価している。その結果、一
部の試料は市販触媒を凌駕する性能を示し、本手法の有効性と合金構造制御の可能性を実証している。
第6章では、本論文全体を総括し、各章の成果を整理するとともに、SPスパッタリング法を基盤とした触媒設計の新たな展開
方向を提示している。
以上より、本論文は、水素社会に必要な新たな燃料電池触媒の設計指針を示唆しており、その成果は工学分野において、
きわめて重要な価値を有し、刘　媛媛氏は博士（工学）の学位に相当する内容と判断できる。

This doctoral thesis addresses fundamental challenges in the control of large-scale 
multi-agent systems (MASs), such as fleets of drones or autonomous vehicles. As these 
systems scale up, two core issues arise: communication delays between agents and a 
lack of network scalability. These factors deteriorate control performance, slow 
convergence, and, in practical scenarios, can lead to issues such as collisions and traffic 
congestion. To address these problems, the thesis introduces novel control 
methodologies based on predictor-based delay compensation and hierarchical network 
strategies, offering both theoretical advancements and practical implications. 
Conventional delay compensation methods, particularly predictor-feedback techniques, 
have proven effective in single-agent systems. However, their direct application to 
MASs has long been considered fundamentally infeasible. In multi-agent scenarios, 
each agent typically lacks access to the current values of the control input of others, 
which are essential for classical predictor-based compensation. This information 
unavailability precludes precise prediction of other agents' states, even in 
disturbance-free environments. Most previous studies have instead employed truncated 
predictors or low-gain approximations, which guarantee only stability rather than 
accurate prediction. In contrast, this thesis proposes a breakthrough: a fully 
non-truncated, trust-based predictor framework specifically tailored for MASs with

constant communication delays. 
 
The core idea of the proposed delay compensation is a fictional yet practically realizable 
assumption: each agent "trusts" that the others can perfectly predict the current global 
state and act accordingly, as if no delays exist. This mutual trust enables each agent to 
infer the delayed control inputs of others using only known dynamics and control gains. 
Through theoretical analysis, the thesis shows that this assumption becomes valid if it 
initially holds over a certain interval, and can thereafter be sustained. The prediction 
method achieves high accuracy, even under disturbances, and theoretical guarantees of 
system stability are provided through Lyapunov functions and Linear Matrix Inequality 
(LMI) formulations. Numerical simulations demonstrate that system performance with 
the proposed method approaches that in the delay-free case. 
 
In addition to communication delays, the thesis tackles the scalability problem of MASs 
by introducing a systematic hierarchical control framework. As MASs grow in size, 
conventional flat network structures (e.g., ring or random topologies) suffer from severe 
limitations in convergence speed and communication burden. Hierarchical network 
structures, inspired by natural and engineered systems, offer a promising alternative 
due to their capacity for information aggregation, structural sparsity, and robustness. 
However, such structures risk degrading control performance by restricting the 
granularity of exchanged information. 
 
This work provides a quantitative foundation for hierarchical control design by 
employing Linear Quadratic Regulator (LQR) theory. The key contribution is an 
algebraic characterization of when the globally optimal controller, without any imposed 
structural constraints, naturally adopts a hierarchical structure. To this end, the thesis 
introduces the concept of hierarchical algebras and hierarchical network vector spaces. 
These tools allow the derivation of sufficient conditions for a class of hierarchical 
performance outputs under which the solution to the algebraic Riccati equation, 
associated with the optimal control problem, inherits a hierarchical structure. 
Importantly, the method accommodates heterogeneous agent groupings, where group 
sizes and dynamics may differ, significantly extending the scope beyond previous 
literature. Simulation studies confirm that hierarchical LQR controllers designed under 
these principles maintain near-optimal performance while drastically reducing 
communication complexity.

Recognizing the importance of stochastic disturbances in real-world high-speed MASs, 
such as turbulence affecting aerial drones, the thesis further extends the hierarchical 
control framework to stochastic systems. This extension is motivated by scenarios like 
drone formations tracking high-speed targets (e.g., Formula 1 racing cars), where noise 
and uncertainty are inevitable. By analyzing the structure of the Stochastic Algebraic 
Riccati Equation (SARE), the thesis identifies conditions under which the optimal 
stochastic feedback gain preserves a hierarchical form. A corresponding hierarchical 
stochastic optimal controller is then designed and tested in simulation, showing 
remarkable performance even in the presence of significant noise. 
 
Overall, this dissertation makes three major contributions: (1) the development of a 
non-truncated predictor-based delay compensation method for MASs that enables 
accurate state estimation and stable control under communication delays; (2) the 
establishment of a quantitative hierarchical control theory that achieves the 
compatibility of optimality and scalability using algebraic tools; and (3) the extension of 
this framework to stochastic environments, ensuring applicability in uncertain, 
real-world conditions.  
These contributions are theoretically rigorous, computationally feasible, and practically 
impactful. They collectively advance the field of decentralized control and provide a 
blueprint for the next generation of intelligent, large-scale, cooperative systems. 
 
 
 
Chapter 1 motivates the study of large-scale multi-agent systems (MASs) and outlines 
their applications in areas such as smart mobility, drone swarms, and distributed 
energy systems. It highlights two core challenges: communication delays and lack of 
scalability. The chapter introduces predictor-based compensation and hierarchical 
control as key strategies, and frames the thesis structure around addressing these 
challenges. 
 
In Chapter2, foundational concepts and notation are introduced, including vector spaces, 
matrix algebra, Kronecker products, and LQR framework. These tools are essential for 
formulating structured control laws and analyzing system properties in later chapters. 
 
Part Ⅱ begins with the problem of communication delays in MASs. Chapter 3 proposes 
a novel trust-based predictor method, where agents assume others act as if there is no

delay. Each agent uses known dynamics and control laws to reconstruct delayed state 
information. The method’s correctness is validated theoretically via Lyapunov analysis 
and LMIs, and demonstrated through simulations. 
 
Part Ⅲ  explores how hierarchical communication structures, which aggregate 
information from groups of agents, affect control performance.  
Chapter 4 defines a generalized hierarchical system model, introduces algebraic tools to 
describe network structure, and sets up the hierarchical optimal control problem. 
 
 
Chapter 5 identifies conditions under which the global LQR optimal controller naturally 
possesses a hierarchical structure. Using hierarchical algebra, it proves that under 
certain structural assumptions, information aggregation does not degrade optimality. 
Simulations validate that performance remains high while communication load is 
reduced. 
 
Within Part Ⅳ, Chapter 6 justifies the need for stochastic control due to real-world 
uncertainties, such as turbulence in drone operations. It investigates the structure of 
the stochastic algebraic Riccati equation and derives sufficient conditions for the 
optimal control law to inherit a hierarchical structure. 
 
Building on Chapter 6, Chapter 7 develops a stochastic hierarchical optimal control 
framework. It designs a controller that achieves robust performance in noisy 
environments while maintaining a structured, scalable form. Simulation results show 
strong performance under disturbances. 
 
In Chapter 8, the thesis is summarized, emphasizing three main contributions: (1) a 
trust-based delay compensation method for MASs, (2) a quantitative theory for 
designing hierarchical optimal controllers, and (3) an extension of this theory to 
stochastic systems. The work is positioned as a step forward toward scalable, robust, 
and decentralized MAS control.

     Dairy products are an excellent source of essential nutrients and play an important 
role in maintaining a healthy diet. In Cambodia, the demand for milk has grown to meet 
domestic needs. In 2020, the country imported approximately $56.3 million worth of milk, 
making it the 28th-largest milk importer worldwide. To reduce this dependency on foreign 
production and strengthen the local dairy industry, the Cambodian government has taken 
proactive measures to promote domestic dairy production. However, there are still 
significant challenges, in particular, knowledge and services related to dairy farming are 
limited in Cambodia. 
     Mastitis is the most concerning disease, causing the most economic losses for the 
dairy industry worldwide. The most common cause of mastitis is an intramammary infection 
with bacterial pathogens. The teat represents the first-line defense mechanism of the bovine 
mammary gland against microbial invasion. Its keratin layer plays a crucial role by forming 
a protective plug within the tip of the teat canal, effectively blocking and neutralizing 
potential pathogens. However, this natural barrier can be compromised after milking 
procedures or due to damage to the teat skin, thereby increasing the risk of bacterial entry 
through the teat canal and subsequent colonization of the mammary tissue. After the 
invasion of bacteria, an acute phase of udder defense occurs, involving the immediate 
elimination of invading pathogens by local phagocytes, the release of inflammatory 
substances, especially chemoattractants, and the migration of polymorphonuclear leukocytes 
into the infected udder. This latter process causes an increase in somatic cell count and low 
milk quality. The recruited polymorphonuclear leukocytes act to kill invading bacteria in 
the infected quarter. If the bacteria are destroyed, a spontaneous cure occurs, resulting in 
only a transient intramammary infection. Occasionally, the defense mechanisms of the

infected mammary gland are unable to eliminate pathogens, especially those such as S. 
aureus and S. agalactiae. As a result, the remaining pathogens can cause either chronic 
intramammary infection or a severely increased immune response, leading to clinical 
mastitis. To reduce the mastitis problem, a mastitis control program has been established to 
minimize new intramammary infection rates through a prevention program and eliminate 
existing intramammary infections using an optimal treatment program. 
     Pathogenic bacteria of mastitis can be divided into two primary sources. One is 
contagious pathogens, which spread from cow to cow, usually during milking, and the other 
is environmental pathogens, which are found in the surroundings of cattle, like bedding, 
manure, or soil. Understanding the type of bacteria is crucial for developing the appropriate 
mastitis control measures in Cambodia. 
     In Cambodia, dairy farming is still in the early stages and is mainly managed by 
smallholder farmers. However, the epidemiology of mastitis, the primary pathogens 
involved, and the factors affecting milk quality that cause economic impacts are still not 
well understood, making it difficult to control mastitis effectively. The overall objective of 
this dissertation is to understand the current situation of mastitis, identify the major 
pathogens and their modes of spread, and examine how mastitis affects milk quality in 
Cambodian dairy farms. This knowledge will be essential for improving udder health and 
helping the sustainable growth of the dairy sector in Cambodia. 
     In Chapter 2, a longitudinal study was conducted to determine the association 
between the dynamic status of primary pathogens responsible for mastitis and seasonal 
variations in a Cambodian smallholder dairy farm. In addition, the proportions of transient 
and chronic intramammary infection (IMI) episodes, as well as the duration of IMI, were 
examined between the primary mastitis pathogens. Through the present study, S. uberis, S. 
chromogenes, and S. gallolyticus were identified as the primary organisms responsible for 
the mastitis epidemic on a Cambodian dairy farm. These bacteria had higher ratios of 
chronic episodes than the other mastitis bacteria found. In addition, S. chromogenes had a 
longer time to cure than pathogens other than S. uberis and S. gallolyticus. The results of 
Chapter 2 indicate that the lack of an optimal mastitis control program in the Cambodian 
smallholder dairy farm causes uncommon mixed infections, uncommon mastitis pathogens, 
and uncommon chronic S. chromogenes infections. 
     In Chapter 3, the genetic diversity and transmission dynamics of S. chromogenes in 
IMI among dairy cows were investigated on two smallholder farms in Cambodia. The 
distribution of transient and persistent episodes of S. chromogenes was explored using 
pulsed-field gel electrophoresis (PFGE) to determine whether S. chromogenes exhibited 
characteristics of a genetically contagious pathogen capable of spreading via multiple 
transmission routes. The possibility of quarter-to-quarter, cow-to-cow, and farm-to-farm 
transmission was also investigated. A total of S. chromogenes isolates (n=235) were 
collected and categorized into 13 PFGE types and 48 subtypes. Among these, the majority 
of types, including PFGE types B, C, D, F, G, I, J, K, L, and M, along with their subtypes, 
that exhibited 100% genetic similarity, were identified as environmental strains. In contrast, 
only three PFGE types, A, E, and H, were identified as contagious strains. These types were 
associated with persistent infections and demonstrated the capacity to spread between cows 
and between mammary quarters. The findings of Chapter 3 support the hypothesis that while 
S. chromogenes is predominantly an environmental pathogen, certain genotypes exhibit 
contagious transmission, contributing to the persistence of mastitis and herd-level 
outbreaks. 
     In Chapter 4, the potential relationship between cisternal milk composition and 
somatic cell counts (SCC) in relation to mastitis pathogens was investigated. The 
differences in milk composition and SCC among mastitis pathogens, farms, and IMI status 
were determined on three dairy farms (a small farm, a medium-sized farm, and an industrial 
farm) in Cambodia. The prevalence of IMI varied significantly among farms, with 
coagulase-negative Staphylococci (CNS) being the dominant pathogen. Streptococcus spp. 
and mixed infections were associated with the highest SCC, while gram-negative bacteria 
and CNS caused moderate increases. Mixed IMI significantly reduced lactose and 
solids-not-fat levels, correlating with elevated SCC. Farm management influenced milk 
composition, likely due to better feed quality and lower incidence of mastitis. 
Pathogen-specific differences were evident, with Streptococci causing the highest SCC, 
followed by CNS and gram-negative bacteria. The results of Chapter 4 indicate that 
Streptococcus spp. caused the highest SCC, while mixed CNS-streptococci infections led to 
the greatest lactose reduction. Farm management significantly affected milk quality, with 
superior feed correlating to lower SCC and mastitis incidence. 
     In conclusion, the mastitis control program needs to be emphasized to minimize the 
mastitis problem in Cambodia, with a focus on both known major mastitis pathogens, such 
as S. uberis and emerging mastitis pathogens, such as the chronic S. chromogenes with 
capability on various routes of mastitis transmission, as the present dissertation has shown 
that this bacterium appears to be associated with reduced milk quality and economic losses. 
This dissertation highlights that mastitis is a significant issue in Cambodian dairy farms, 
necessitating the implementation of mastitis control measures, including improved hygiene, 
control strategies, and effective management to reduce infections and enhance productivity.

With global food demand projected to rise by 70% by 2050, the need for sustainable 
pest management strategies has become increasingly urgent to ensure food security while 
minimizing harm to human health and the environment. Transgenic crops offer a promising 
avenue to address challenges such as crop losses and the overuse of chemical pesticides by 
enhancing agricultural sustainability and resilience. 
Phytosterols, sterols found in plants, are vital components of plant cell membranes, 
contributing to membrane integrity, signaling, vesicle trafficking, and stress responses. Over 
250 unique phytosterol structures have been identified in plants. Insects, which cannot 
synthesize sterols de novo, must acquire them through their diet. Because sterols are essential 
for insect cell membranes and hormone biosynthesis, modifying plant sterol composition 
presents a novel target for pest control. 
This study focuses on Brassica napus (canola), one of the world’s most important oilseed 
crops, which is increasingly threaten by pests and diseases. In particular, the tobacco cutworm 
Spodoptera litura (Fabricius) is a destructive pest that affect over 300 crops worldwide. This 
research investigates the potential of genetically modifying canola to alter its sterol 
composition as a menas to reduce S. litura performance. Specifically, the study explores: (1) 
sterol utilization in S. litura through artificial diet assays; (2) the effects of stigmasterol-
enriched canola lines generated by overexpressing Cytochrome P450 710A1 (CYP710A1); and 
(3) the impact of introducing the Sterol methyltransferase (CrSMT) gene from 
Chlamydomonas reinhardtii ( a green alga), which leads to reductio of a utilizable sterol (β-
sitosterol), and the production of non-utilizable sterol.  
In Chapter 2 of the thesis, to evaluate the effects of individual sterols under controlled 
conditions, a sterol-free artificial diet was specifically formulated for Spodoptera litura. Two 
groups of sterols were tested: Δ5-sterols (brassicasterol, campesterol, β-sitosterol, cholesterol, 
and stigmasterol) and Δ4-sterol ((+)-4-cholesten-3-one). The results indicated that S. litura 
could utilize Δ5-sterols, whereas the Δ4-sterol, such as (+)-4-cholesten-3-one, was not suitable 
for development. The findings in Chapter 2 provide foundational insights into the sterol 
utilization capacity of S. litura and demonstrate that sterol structure and metabolic 
compatibility are critical determinants of larval performance. The sterol-free artificial diet 
developed in this study offers a robust platform for dissecting sterol function and metabolism 
in insect herbivores. These insights may have broader applications in pest management 
strategies, particularly through targeted manipulation of plant sterol profiles. 
In Chapter 3, to explore the broader applications of multiple sterols under more 
practical conditions, genetically modified canola (Brassica napus), one of the world’s most 
important oilseed crops and currently ranked as the third-largest globally, was used in feeding 
experiments. Building on the findings from Chapter 2, which demonstrated that individual 
sterols influence Spodoptera litura development and survival, Chapter 3 shifts focus to the 
effects of more complex sterol mixtures that better reflect natural plant-insect interactions. 
Specifically, this chapter examines how genetic modification of B. napus to alter sterol 
composition impacts herbivore performance. We utilized transgenic B. napus lines 
overexpressing the Cytochrome P450 710A1 (CYP710A1) gene. This gene functions as a C-22 
sterol desaturase, catalyzing the conversion of β-sitosterol to stigmasterol in the sterol 
biosynthesis pathway in plants. In this study, three CYP710A1 B. napus lines, including 
18(J29a), 19(J37c), and 21(J51m), were selected based on their sterol profiles, and wild-type 
(WT) B. napus was used as the control treatment. The B. napus was grown under glasshouse 
conditions. Then, the leaf was freshly cut and S. litura were fed on transgenic modified B. 
napus lines based on their distinct sterol profiles. S. litura body weight and survival rates 
improved, and no adverse effects were observed in midgut histology when fed on the CYP710A1 
B. napus lines such as 18(J29a), 19(J37c), and 21(J51m). Chapter 3 demonstrates the potential 
of engineering B. napus sterol profiles, particularly through stigmasterol enrichment, to 
enhance resistance to various biotic stresses. This approach can be tailored to target specific 
pests while preserving ecological balance. These findings contribute to a broader 
understanding of how sterol metabolic engineering can be leveraged as a pest management 
tool and highlight the importance of species-specific evaluations when designing such 
interventions. 
In Chapter 4, another transgenic line was developed by transferring the Sterol 
methyltransferase gene from Chlamydomonas reinhardtii (a green alga) (CrSMT) to Brassica 
napus. This enzyme plays a crucial role in the sterol biosynthesis pathway by catalyzing 
methylation at the C-24 position of sterol intermediates, resulting in reduced utilizable sterols 
such as β-sitosterol and production of non-utilizable sterols such as spinasterol, a sterol not 
typically found in canola. Feeding assays were conducted in two phases. In phase one, insects 
were fed on 5-week-old B. napus from CrSMT_33, CrSMT_35C, and CrSMT_35J lines, which 
showed distinct sterol profiles. In phase two, 7-week-old B. napus from CrSMT_27 and 
CrSMT_31 lines, selected for unique sterol compositions, were used to assess age-dependent 
sterol effects, and also to screen more CrSMT B. napus lines. Spodoptera litura fed on CrSMT 
B. napus lines such as CrSMT_35J, CrSMT_27, and CrSMT_31 had pronounced negative 
effects, influencing developmental duration, lethality, and impaired midgut and integument 
structure. In this Chapter, the combination of sterol deficiency (particularly β-sitosterol) and 
the presence of non-preferred sterol (spinasterol) disrupts normal growth, metabolism, and 
midgut integrity in S. litura, supporting sterol profile manipulation as a promising pest 
management.   
In summary, Spodoptera litura was able to utilize brassicasterol, campesterol, β-
sitosterol, cholesterol, and stigmasterol, but not (+)-4-cholesten-3-one or spinasterol. The 
presence of non-utilizable sterols resulted in reduced body weight, extended developmental 
duration, increased lethality, and structural impairments in the midgut and integument. 
These sterols also led to a significant reduction in sterol content within both larval and pupal 
stages. Among the tested transgenic lines, Brassica napus CrSMT_35J, CrSMT_27, and 
CrSMT_31 showed strong potential to negatively impact insect development and survival. 
These findings highlight the critical role of sterol composition in shaping herbivore 
performance and suggest that manipulation of plant sterol profiles may offer a targeted 
approach for pest management.   
However, this study has some limitations that require further validation and future 
investigation. First, although feeding insects artificial diets with defined sterols allows for 
precise analysis of sterol metabolism, future studies should explore mixtures of utilizable (e.g., 
cholesterol, β-sitosterol) and non-utlizable sterols (e.g., cholestenone, cholestenol) to better 
understand their roles in insect development and hormonal regulation. 
Second, to assess the broader applicability of sterol-based plant modifications, their 
effects should be tested on various insect species, both generalists and specialists, using 
different transgenic lines (e.g., CYP710A1, CrSMT). 
Third, combining transgenic lines may enhance pest resistance. For instance, crossing 
CYP710A1 and CrSMT lines could result in stronger and broader protective effects. 
Finally, examining sterol biosynthesis and distribution across different plant tissues 
and developmental stages could identify key vulnerabilities and inform more targeted pest 
management strategies.

Drylands are one of the most widespread habitats in the world, but managing them presents 
challenges due to their unique ecological characteristics, particularly in the face of climate 
change and increasing human demands. Establishing protected areas (PAs) is considered an 
effective strategy for promoting the sustainability of these ecosystems. Launched in 2013, the 
National Desert Parks (NDPs) project in China represents an innovative approach to balancing 
conservation and utilization as a means of combating desertification, marking a significant 
advancement in the country’s PA system. This study provides a comprehensive evaluation of 
the NDP policy’s implementation by reviewing its overall development, the ecological impacts 
of the parks, and their effectiveness from the perspective of stakeholders. 
 
The study first reviewed the progress of the NDP policy and all designated parks, using the 
status of parks that were either retained or excluded from the PA integration and optimization 
efforts as a baseline. The analysis referenced announcements released by the government, news, 
as well as information shared by individuals on popular social media, life service platforms, and 
navigation applications in China. As of the end of 2024, a total of 99 dryland NDPs have been

designated, with 81 currently active, covering a total area of 2858 km2. These designations were 
primarily concentrated in the early phases of the project, with Xinjiang receiving the highest 
number of approvals. However, only about one-third of these designations have shown progress 
in terms of tourism development, while the others either lack detailed information or have not 
yet been started. Eighteen designations have been merged, split, or cancelled from the list of 
NDPs, primarily due to land-use conflicts. The study highlighted the varying levels of 
development among the designated parks and pointed out the existence of “paper parks”, which 
challenge the regulatory framework of the policy. 
 
Among the NDPs with well-developed tourism, the study selected five of them with clear 
boundaries to assess their ecological effectiveness, which was indicated by their environmental 
dynamics over the decade since the policy was announced. This assessment utilized multiple 
remote sensing (RS) indicators capable of dryland monitoring, examining changes in vegetation 
cover, bare land extent, soil particles, and surface moisture. Additionally, it analysed the 
environmental trends of each park in relation to contemporaneous climatic variation to evaluate 
their adaptability to changing climate conditions. Of the parks studied, two showed ecological 
improvements that outweighed the impacts of climate variations, while another two maintained 
environmental stability, showing significant effects from climatic changes. In contrast, one park 
experienced deterioration due to shifting climatic conditions. The study also conducted on-site 
verification in two parks where conditions improved and deteriorated. The findings provided 
further insights into their environmental trends. In addition, field surveys revealed discrepancies 
between the actual park implementation and the official declaration, particularly regarding the 
park boundaries and functional zoning related to conservation efforts. Similar inconsistencies 
were observed in the other parks through high-resolution Google base images. 
 
The study also evaluated the NDP’s effectiveness from stakeholders’ perspectives. This was 
accomplished through a case study that utilized questionnaires to investigate the various 
ecosystem services provided by the park designation, taking into account economic, social, 
intangible, and environmental benefits. It also assessed the park’s management and the public’s 
attitudes toward the park and the NDP policy. A total of 190 residents and tourists participated

in the data collection. Overall, the responses indicated a recognition of the park’s effectiveness 
in providing benefits, particularly in terms of intangible and environmental aspects. However, 
there was an imbalanced distribution of values among stakeholders with different social 
attributes. Residents, particularly villagers and those with lower education levels, reported a 
lower perception of the benefits. Additionally, the feedback highlighted issues in park 
management, including insufficient public participation, delayed dividends, and inadequate 
treatment of nearby private attractions. Despite these management shortcomings, there was still 
general support for the park’s future and the NDP policy among the stakeholders who responded, 
largely due to a variety of benefits offered by the park. 
 
In summary, the study highlighted the progress made in implementing China’s NDP policy and 
demonstrated its significance through successful cases in ecological conservation and the 
enhancement of human well-being. However, the study also identified challenges that threaten 
its sustainability, which need to be addressed further for all NDPs. Currently, there are apparent 
differences in the development of designated parks, with some parks significantly lagging and 
some still facing shortcomings. As a result, more efforts are required in the future to achieve the 
policy’s targets. For future research, a broader range of NDPs should be included in assessments 
when their boundaries can be accurately tracked, using RS data with higher resolution, 
conducting more detailed field investigations, increasing stakeholders’ involvement, as well as 
performing comparative analysis.

１．背景
本稿は2024年度の春学期に日本語研修コースで実施
した書道の授業の報告である。
日本語研修コース（EJ コース）は，大使館及び大
学推薦による研究留学生，教員研修生のためのコース
である。学生は日本語学習経験がない。到達目標は
CEFR の A2レベル，JLPT の N4レベルを目安とし
ている。授業は月曜から金曜まで毎日90分2コマの授
業が行われる。テキストは『NEJ: A New Approach to 
Elementary Japanese』を使用している。2024年春学
期の学生数は8名で，すべて非漢字圏の学生である。
このクラスで学期の後半に書道の授業を行った。
福光（2005）は留学生の書道の授業の目標として，
①筆を使って墨書することに慣れ親しむ，②日常の文
字を正しく美しい形で書くことができるように練習す
る，③書道史を通して書体成立の変遷を学び，文字に
対する理解・関心を高める ，④自らの書作品を創り，
また鑑賞することを通して 書道の芸術性を理解し， 
それを各自の言葉で話すことができるようになる，の
4点を留学生に対する書道の授業の目標としたが，本
授業では，筆を使って墨書し，自らの作品を完成させ
ることとした。
２．授業の進め方とポイント
授業の1週間前に，次週は書道を行うことを伝え，
書道で書きたい漢字の言葉を各自考えておくよう，ま
た，汚れてもよい服を着てくるよう指示した。書道用
具を購入する必要がないことを説明した。
1時限目は短期交換留学受入れプログラム（NJ1）
（9名）との2クラス合同で，2時限目は EJ コースの
学生のみで行った。
机は4つで1つの島を計5つ作り，その上に汚れ防
止のため新聞紙を敷いて，学生に好きなところに座ら
せた。
初対面の学生も多かったので，授業開始時に簡単な
自己紹介をしてから，授業に入った。
まず，書道は漢字文化をもつ中国で発達し，日本へ
は６世紀から7世紀頃，仏教とともに写経として書道
が伝わったこと，約3000年前鹿の骨に書かれた漢字
（甲骨文字）が最古だと言われていること，楷書・草
書・行書などの書体，墨，硯，筆，文鎮など書道の道
具をパワーポイトで簡単に説明した。次に，約4分の
動画（1）を視聴し，書道道具の配置，書く際の姿勢，筆
の持ち方，筆の使い方を確認した。学生にワンセット
ずつになった書道用具を配布し，動画で見たとおりに
配置，準備をさせた。数ページからなる手本の冊子も
配布した。
ひととおり準備が整ったところで，基礎練習を開始
した。学生たちを教師の机近くに来させ，書いて見せ
たあと，各自の机に戻り，練習させた。まず，漢字の
一で横線を，川で縦線の練習をしたが，始筆と終筆に
苦戦していた。絵画のように同じところをなぞる二度
書き，三度書きをする学生も見られた。何回書いても
形がとれない学生には，手に触れてもいいか確認した
うえで，教師が手を添えて，書き方を指導した。教師
がリードして書くことで，力の入れ方や筆を運ぶ方向
などが矯正できる。「才」で左払いとはねを練習し，
さらに「木」で右払いを練習した。はねと左払いは特
に難しいようだった。基礎練習の締めくくりとして，
「永」の練習をした。「永」は永字八法といって，書道
に必要とされる基本的な技法が使われる文字だからで
ある。基本的な練習をしたあと，学生が書きたい漢字
1文字を選ばせ，練習した。手本がないので，その場
で書き，渡して，各自練習させた。
学生が選んだのは，「美」「金」「力」「春」，「魚」な
どであったが，中には改善のように2文字の語を書く
学生もいた。半紙に漢字1文字を書き，小筆を使って
カタカナで名前を書き作品を賛成させた。NJ1の学生
は1限のみの参加だったので，ここで退出した。
EJ の学生は引き続き2限も書道を行った。

各自書きたい言葉を考えてきたので，その言葉を確
認し，練習させた。学生によっては，画数が多かった
り，形がとりにくかったりするものや，日本語としては
なじみのない言葉もあったので，教師と相談し，題を変
更する学生もいた。書道の題はどのようなものがある
のか，事前に学生に伝えるとよかったかもしれない。
2限目は，まず半紙で学生が考えてきた語句を書く
練習を行った。文字数が増え，難易度が増した。何回
か練習したところで書初め用紙を配布し，清書させ
た。縦長の用紙に複数の文字を書くので，バランスが
とりにくそうだったが，みな作品を完成させることが
できた。作品は教室にしばらく展示した。
書道授業の翌週に学生に感想を聞いたところ，「1
回では足りない，もっとやりたい」「楽しかった」な
ど，体験型の活動を評価する意見や，「筆を使って書い
てみて，筆順の重要性を理解できた」など，書道体験
を通じて漢字書写への気づきを述べる学生もいた。
今回の書道の授業については，初級後半になり，学
ぶべき文法項目も増え，やや疲れ気味の学生の息抜き
の意味もあったが，それ以上に学生は学びがあったよ
うだ。
３．今後に向けて
本稿では2024年春学期に初級クラスで行った書道の
授業を報告した。今回は書くだけにとどまってしまっ
たが，書く前の活動として，書道で好まれる題につい
て話し，日本文化に触れさせることもできた。また，
書いた後の活動としては，学生が書いた作品の読み方
や意味，その語を選んだ理由などを説明したり，互い
の作品を鑑賞して感想を言い合ったりすることもでき
た。そうすれば，書道を通して日本文化に触れるのみ
でなく，日本語学習にもつなげられ，意義深い授業が
行えると思う。限られた時間を有効に使い，豊かな学
びを実現するには，どのような授業を行うべきかさら
に考えたい。
