Proteinaceous components in goat amniotic fluid enhance the expression of keratinocyte differentiation-related proteins

    No full text
Amniotic fluid (AF) constitutes a dynamic environment containing diverse bioactive molecules derived from both maternal and fetal sources that support fetal development. As the fetus develops in continuous contact with AF, it is plausible that AF influences the formation of the skin epidermis. However, the mechanisms through which AF promotes keratinocyte differentiation remain largely unclear. Here, we showed that goat AF enhanced the expression of key functional proteins involved in epidermal barrier formation, including small proline-rich proteins, loricrin, and transglutaminase. We further obtained the bioactive fractions that promote the expression of these differentiation-related proteins through multistep protein fractionation via column chromatography. Proteomic analysis subsequently revealed 291 candidate proteins, including 85 distinct extracellular proteins, primarily grouped into calcium-binding proteins, proteases and their regulators, extracellular matrix components, and signaling molecules. Collectively, these results suggest that proteins secreted or released into AF contribute to establishing a microenvironment conducive to epidermal differentiation.Online Published: 25 September 2025journal articl

Pressure-flowrate characteristics of electrohydrodynamic conduction pump with rod-ring electrodes including reverse flow range

When attempting to control the heat transport by the loop heat pipe (LHP) to an extremely small amount using an electrohydrodynamic (EHD) pump, the EHD pump is exposed to conditions where the flow direction periodically changes. In this study, the pressure-flowrate (P-Q) characteristics of the EHD conduction pump, including the reverse flow regime, were investigated. The EHD pump consists of six pairs of rod-ring electrodes and is placed in a fluid loop with a mechanical pump to apply the reverse flow. The P-Q characteristics over the first and second quadrants were successfully measured. A EHD simulation model that is consistent with the pressure generated at zero flowrate is also developed, revealing the EHD field in the EHDpump.2025 IEEE International Conference on Dielectric Liquids (ICDL) (18-22 May 2025, Lodz, Poland)journal articl

Low-energy electron microscopy investigation of germanene segregation on Ag(111) thin films

    No full text
We used low-energy electron microscopy to investigate the formation processes of various surface structures on Ag(111) thin films grown on Ge(111) during Ge segregation. During annealing the Ag(111) samples cleaned by Ar+ sputtering, the Ag2Ge surface alloy first appeared along the Ag grain boundaries, signifying preferential diffusion of Ge atoms through these boundaries. Subsequent annealing at higher temperatures induced the phase transformation from the surface alloy to germanene. However, further higher temperature annealing caused back-diffusion of Ge atoms into the Ag thin films, reverting to the surface alloy. Formation of the ordered (7√7×7√7)R±19.1° germanene required cooling from high temperatures around 500 °C. However, high-temperature annealing occasionally resulted in the formation of three-dimensional (3D) Ge islands on the surface. During cooling, the 3D islands absorbed Ge atoms from the surrounding germanene, triggering phase transformation to the surface alloy in the vicinity of the islands.journal articl

Reticulon 4 reflects endoplasmic reticulum stress in arginine vasopressin neurons

    No full text
Objectives: Arginine vasopressin (AVP) is synthesized in the magnocellular supraoptic nucleus and paraventricular nuclei of the hypothalamus, where AVP neurons function under a consistently high demand for AVP production. AVP neurons are subject to endoplasmic reticulum (ER) stress even under basal conditions, and this ER stress is further exacerbated when AVP production increases due to dehydration. Reticulon (RTN) is essential for ER formation and stabilization and plays a critical role in membrane morphogenesis within the ER. This study aimed to investigate the expression of RTN family members in hypothalamic AVP neurons. Methods: Fluorescence immunohistochemistry and in-situ hybridization were employed to examine the expression of RTN family members in hypothalamic AVP neurons of adult male mice. Water deprivation and treatment with a chemical chaperone 4-phenylbutyric acid were used to increase and decrease the ER stress of AVP neurons, respectively. Results: Among the RTN family members, only RTN4 was found to be expressed in hypothalamic AVP neurons. RTN4 was colocalized with ER organelle markers, including immunoglobulin heavy chain binding protein and calnexin. Furthermore, RTN4 expression increased during ER stress induced by water deprivation. On the other hand, increased RTN4 expression by water deprivation was attenuated by 4-phenylbutyric acid treatment. Conclusions: Our results suggest that RTN4 expression in AVP neurons is closely associated with ER stress caused by increased protein production in neuroendocrine cells.journal articl

Relationship between support and job satisfaction among nurses working in residential long-term care facilities: a cross-sectional study

The quality of care in care facilities for older adults varies according to the employees’satisfaction level. This study aimed to clarify the relationship between the level of support from supervisors and colleagues and job satisfaction among nurses working in care facilities for older persons. Using a cross-sectional design, we analyzed the responses of 295 nurses in long-term care facilities in Japan to a questionnaire survey. The surveyed items included the nurses’ satisfaction with their current job (the dependent variable), and the degree of support from supervisors and colleagues. Of the 295 nurses, 166 (56.3%) were satisfied with their nursing jobs. Additionally, 125 (42.4%) nurses had low support from both supervisors and colleagues, 110 (37.3%) had high support from either supervisors or colleagues, and 60 (20.3%) had high support from both. There was a positive association between support from supervisors and nurses’ satisfaction. Furthermore, based on the criterion of low support from supervisors and colleagues, high support from either or both was significantly associated with nurses’ satisfaction (an odds ratio of 2.04 in the case of support from either and 4.02 in the case of support from both). These results suggest that there should be strategic hiring and training of managers who can support staff, as well as the strengthening of teamwork.departmental bulletin pape
