    Evidence for peroxisomal redundancy among the glucose-6-phosphate dehydrogenase isoforms of Arabidopsis thaliana

    The oxidative pentose phosphate pathway (OPPP) plays an important role in the generation of reducing power in all eukaryotes. In plant cells, the OPPP operates in several cellular compartments, but as full cycle only in the plastid stroma where it is essential. As suggested by our recent results, OPPP reactions are also mandatory inside peroxisomes, at least during fertilization. For the first enzyme of the OPPP, glucose-6-phosphate dehydrogenase (G6PD), we previously showed that one Arabidopsis isoform (G6PD1) can be directed to peroxisomes under specific circumstances. Since g6pd1 knock-out plants are viable, we aimed at elucidating potential redundancy regarding peroxisomal targeting among the other G6PD isoforms. Localization studies of so far cytosolic annotated G6PD5 and G6PD6 (both ending -PTL>) using different reporter fusions of full-length versus the last 50 amino acids revealed that GFP-C-short versions are efficiently imported into peroxisomes. Modification of the final tripeptide to a canonical peroxisomal targeting signal type 1 (PTS1) also resulted in peroxisomal localization of the full-length versions and revealed that G6PD5/6 import may occur as homo- or heterodimer. Interestingly, the new noncanonical PTS1 motif is highly conserved among the cytosolic G6PD isoforms of the Angiosperms, whereas members of the Poaceae (rice and maize) possess two variants, one ending with an additional amino acid (-PTLA>) and the other one extended by a stronger PTS1 motif. From both evolutionary and physiological perspectives, we postulate that G6PD import as homo- and heterodimer restricted the acquisition of more efficient peroxisomal targeting motifs to leave some G6PDH activity in the cytosol

    Oculomotor behavior can be adjusted on the basis of artificial feedback signals indicating externally caused errors

    Whether a saccade is accurate and has reached the target cannot be evaluated during its execution, but relies on post-saccadic feedback. If the eye has missed the target object, a secondary corrective saccade has to be made to align the fovea with the target. If a systematic post-saccadic error occurs, adaptive changes to the oculomotor behavior are made, such as shortening or lengthening the saccade amplitude. Systematic post-saccadic errors are typically attributed internally to erroneous motor commands. The corresponding adaptive changes to the motor command reduce the error and the need for secondary corrective saccades, and, in doing so, restore accuracy and efficiency. However, adaptive changes to the oculomotor behavior also occur if a change in saccade amplitude is beneficial for task performance, or if it is rewarded. Oculomotor learning thus is more complex than reducing a post-saccadic position error. In the current study, we used a novel oculomotor learning paradigm and investigated whether human participants are able to adapt their oculomotor behavior to improve task performance even when they attribute the error externally. The task was to indicate the intended target object among several objects to a simulated human-machine interface by making eye movements. The participants were informed that the system itself could make errors. The decoding process depended on a distorted landing point of the saccade, resulting in decoding errors. Two different types of visual feedback were added to the post-saccadic scene and we compared how participants used the different feedback types to adjust their oculomotor behavior to avoid errors. We found that task performance improved over time, regardless of the type of feedback. Thus, error feedback from the simulated human-machine interface was used for post-saccadic error evaluation. This indicates that 1) artificial visual feedback signals and 2) externally caused errors might drive adaptive changes to oculomotor behavior

    On the evolution of plate tectonics under the influence of intrinsically dense LLSVPs

    Große, heterogene Strukturen an der Kern-Mantel-Grenze, sogenannte Large Low Shear Velocity Provinces (LLSVPs), könnten als Relikte früherer Differenzierungsprozesse über weite Teile der Erdgeschichte hinweg existiert haben. Sind diese, wie häufig angenommen, dichter als der umgebende Erdmantel, könnten sie die Mantelkonvektion stabilisieren und die Plattentektonik beeinflussen. Die hier eingesetzten Computermodelle liefern Einblicke in physikalische Prozesse und Entwicklungstendenzen, die für die tektonische Geschichte der Erde relevant sein könnten. LLSVPs könnten Stil und Beginn erster Plattenbewegungen beeinflusst und die Entstehung moderner Plattentektonik verlangesamt haben. Für LLSVPs mit moderaten Ausmaßen könnte die Mantelkonvektion zunächst ausschließlich unter einer starren Erdoberfläche stattgefunden haben, was frühe Plattenbewegungen deutlich verzögert hätte. Dieses Szenario ließe sich zudem mit aktuellen Hypothesen zu Vorläufern der modernen Plattentektonik verknüpfen

    Predicting Locomotion Intention using Eye Movements and EEG with LSTM and Transformers

    Die Vorhersage zukünftiger Bewegungsabläufe auf der Grundlage intrinsischer Daten dient vielen Zwecken, z. B. der Optimierung der Nutzung des physischen Raums in Virtual-Reality-Umgebungen und der Verbesserung der Steuerung elektronischer Hilfsmittel für Patienten mit motorischen Beeinträchtigungen. Die Vorhersage menschlicher Fortbewegungsabsichten erweist sich jedoch aufgrund der hochkomplexen und nichtlinearen Wechselwirkungen zwischen den relevanten Parametern als schwierig. Tiefe neuronale Netze bieten einen erheblichen Vorteil gegenüber herkömmlichen Ansätzen, um diese Herausforderung zu bewältigen. Wir behandeln diese Aufgabe als ein Problem der Zeitreihenvorhersage und vergleichen LSTM-Netzwerke mit Transformatormodellen. Ein besonderer Aspekt unserer Arbeit ist die Betonung der Augenbewegungen als zentrales Merkmal, was zu den neuartigen Vorhersagefähigkeiten beiträgt. Neben den Blickdaten evaluieren wir die Hinzunahme von EEG als Datenquelle für diese Vorhersageaufgabe, die in Gehirn-Computer-Schnittstellen verwendet werden soll. Um dies zu erreichen, haben wir zwei Experimente zur Datenerfassung in benutzerdefinierten virtuellen Umgebungen durchgeführt, die verschiedene Aufgaben mit Joystick-Steuerung beinhalten. Wir stellen diese neuen Datensätze in Verbindung mit dieser Arbeit vor. Die Ergebnisse zeigen, dass Blickdaten ein wertvolles Werkzeug für die Vorhersage der Fortbewegung in verschiedenen Kontexten sind, selbst wenn es keine starke und direkte Verbindung zwischen Blick und zukünftigen Wegpunkten gibt. Transformer-Modelle konnten eine bessere Leistung erzielen als LSTM-Netzwerke, und wir kommen zu dem Schluss, dass für eine erfolgreiche Vorhersage in verschiedenen Situationen Datensätze erforderlich sind, die ein breites Spektrum von Bewegungsszenarien enthalten.Predicting future locomotion based on intrinsic data serves many purposes, including optimizing the utilization of physical space in virtual reality environments and enhancing the control of electronic aids for patients with motor impairments. However, predicting human locomotion intentions proves challenging due to the inherent difficulty arising from the highly complex and nonlinear interactions among the relevant parameters. Deep neural networks offer a significant advantage over conventional approaches in addressing this challenge. We treat this task as a time series prediction problem and compare LSTM networks to transformer models. A distinctive aspect of our work is our approach’s emphasis on eye movements as a central feature, contributing to its novel predictive capabilities. Besides gaze data, we evaluate the addition of EEG as a data source for this prediction task to be used in brain-computer interfaces. To achieve this, we conducted two data collection experiments in custom virtual environments that feature different tasks utilizing joystick control. We present these novel datasets in conjunction with this work. The results demonstrate that gaze data proves to be a valuable tool for locomotion prediction in different contexts, even when there is not a strong and direct connection between gaze and future waypoints. Transformer models were able to achieve better performance than LSTM networks, and we conclude that successful prediction across diverse situations requires datasets containing a wide range of movement scenarios

    Preventing smartphone induction of magnet mode in cardiac implantable electronic devices

    Aims: Wireless charging capable smartphones may interact with cardiac implantable electronic devices (CIED). We hypothesized that magnetic shielding with a steel plate placed at the back of a smartphone may prevent interaction. Methods and results: Sixteen CIED (6 pacemakers and 10 implantable cardioverters/defibrillators) from all manufacturers were consecutively implanted in a subcutaneous and submuscular location in an isolated porcine thorax and connected to an interactive heart simulator. Two smartphones (Apple iPhone 14 and Google Pixel 8 Pro) were placed on top of the implantation site, and signs of magnet mode induction were recorded. The Apple iPhone induced magnet mode in seven of 16 (44%) subcutaneously placed CIED. Placed in a magnetic phone case, the Apple iPhone induced magnet mode in six of 16 (38%) devices. Magnet mode induction was successfully prevented in all cases by placing a 1 mm thin steel plate at the back of the smartphone. The Google Pixel did not induce magnet mode in any of the 16 devices, even with a magnetic case. Submuscular CIED placement resulted in no occurrence of magnet mode induction by any smartphone. All devices were still able to be interrogated and responded to a CIED magnet. Conclusion: The Apple iPhone induces magnet mode in close to half of subcutaneously placed CIED due to its MagSafe magnet. Neither a smartphone utilizing the Qi wireless charging standard nor magnetic smartphone cases confer the same risk. Submuscular CIED placement or magnetic shielding with a steel plate prevents magnet mode induction. The future Qi2.0 standard may exacerbate interaction risks

    PrimarWebQuests im bilingualen Mathematikunterricht:Potenziale des fachsprachlichen und fachbezogenen Kompetenzerwerbs an der Schnittstelle sprachlich-bilingualen, mathematischen und digitalen Lernens

    Diese interdisziplinär zu verortende Arbeit fokussiert den bilingualen Mathematikunterricht und ist dem national und international relevanten Bereich Mathematik und Sprache zuzuordnen. Der Erkenntnisgewinn dieser Untersuchung trägt zur empirischen Fundierung möglicher Lernsettings bilingualen Mathematikunterrichts bei. Hierzu wird das fachbezogene und fachsprachliche Lernen bilingual unterrichteter Lernender eines vierten Schuljahres hinsichtlich des Größenbereichs Längen untersucht und der Beitrag des Unterrichtsansatzes PrimarWebQuest geklärt. Die Studie ist in der qualitativen Sozial- und Unterrichtsforschung zu verorten und folgt einer rekonstruktiv-interpretativen Methodologie. Als Potenzial des Settings der Studie zeigt sich aus der sprachlich-bilingualen Perspektive ein Kompetenzzugewinn der Lernenden in beiden verwendeten Sprachen sowie hinsichtlich der metasprachlichen Reflexion. Darüber hinaus kann ein fachbezogener Kompetenzzugewinn aus der mathematischen Perspektive festgehalten werden, da die Lernenden ihre Längenkonzepte ausschärfen konnten und in der Lage sind, die Strukturen der zwei Längenmaßsysteme zu vergleichen. Gerahmt werden diese Potenziale aus der digitalen Perspektive durch den Beitrag des Unterrichtssettings, welches neben einem Zugewinn notwendiger Kompetenzen in einer digitalen Gesellschaft auch sprachsensibles und interkulturelles Lernen ermöglicht und somit den Grundsätzen des Content and Language Integrated Learnings folgt
