Identification of PlexinD1 and AHDC1 as a putative interactors for Tip-1 protein
Abstract
The class 1 PDZ domain Tip-1 protein was first identified
as a binding partner for the Human T-Cell Leukaemia Virus
type 1 (HTLV-1) Tax oncoprotein. It was later shown to interact
with: the RhoA signaling effector Rhotekin, the Wnt signaling
effector β-catenin and the E6 oncoprotein from
high-risk HPV16 but not low-risk HPV6. These observations
suggested that Tip-1 may be an important “hub” protein that
is involved in pathways with a proven link to carcinogenesis.
Based on these findings, it was decided to further characterize
the cellular role of Tip-1 by carrying out a yeast two-hybrid
screen to identify new binding partners in order to uncover
potentially novel functions of this protein. This identified an
intracellular fragment of the trans-membrane receptor plexin
D1 and a C-terminal fragment of the AT hook DNA binding
containing 1 (AHDC1) protein which had a carboxyl terminal
PDZ binding domain consensus sequence. Both of these interactions
were confirmed by yeast mating assay which was also
used to show that mutant constructs of AHDC1 lacking the
carboxyl PDZ binding site did not bind Tip-1. Immunofluorescent
imaging of these proteins in HPV16 E6 expressing
human C33A cervical carcinoma cells suggested they may
co-localize.King Saud Universit
New oxorhenium (V) complex with an imidazol [NN] /hydantoin [SN] mixed ligand system, and radiochemical behavior of its oxotechnetium (V) complex analog
A [ReO(Imz)(Hyd)(H2O)2OH] complex was suc-cessfully synthesized by the ligand exchange method using oxorhenuim citrate and an imida-zole /Hydanton mixed ligand system. Geometry optimization of complex has been carried out using DFT at the B3LYP/LANL2DZ functional in singlet state. B3LYP predicated infrared spec-trum of the geometrically optimized structure using the same level of the theory and the same base set showed good agreement with experi-mentally observed values. The spin allowed singlet-singlet electronic transition of the [ReO(Imz)(Hyd) (H2O)2OH] complex was calcu-lated with time dependent density function the-ory (TD-DFT) and the UV-Vis spectra has been discussed on this basis. The complex was characterized using microanalysis and IR, UV-Vis, NMR and mass spectroscopic. The techne-tium tracer [99mTcO(Imz)(Hyd) (H2O)2OH] has also been synthesized by two methods using 99mTc-gluconate as a precursor or; by direct re-duction. The radiochemical purity of the com-plex was over 95% as measured by thin layer chromatography. In vitro studies showed that the complex possessed good stability under physiological conditions. The partition coeffi-cient indicated that the complex hydrophilic and the electrophoresis results showed that the complex cationic. Biodistrbution in mice showed that the complex accumulated in heart uptake of 9.53 ± 3.87% ID/gm at 5 min and good retention (6.37 ± 1.21)% ID/gm at 60 min. One hour after the injection, the heart/liver, heart/lung and heart/blood radioactivity ratios were 0.46, 1.04 and 0.56, respectively. These findings indicate that the complex might b
Energy storage through adsorption and desorption
Adsorption/desorption of water vapour on raw Saudi bentonite (RB) is proposed as a
heat energy storage. This is most readily achieved by adsorption and desorption of water vapour
on RB at different temperatures as a function of time. The RB subjected to preheating temperature of 200 _C, before subjecting to the adsorption process carried out. The IR spectra of RB before adsorption of water vapour at 298 and 313 K wereas studied. The adsorbed and desorbed water vapour from bentonite surfaces at 298 and 313 K was determined at different time. The adsorptive capacities of RB sample at 298 and 313 K were 0.0097 and 0.0141 mol/g of dry RB, respectively, after 72 h. The desorbed amounts are 0.0085 and 0.01 mol H2O/g of RB at 298 and 313 K, respectively after 72 h. A kinetic models of second order of the adsorption and desorption of water vapour fitted well the experimental data. Application of Van’t Hoff’s law at two temperatures (298 and 313 K) yields the adsorption and desorption enthalpy. The adsorption enthalpy (stored energy)
of RB increased with increasing contact time up to 5 h. At this time the maximum enthalpy was
about 30 kJ/g dry bentonite, at which the clay has lost all the energy that could be released due
to adsorption of water vapours. Then it shows a decrease in sorption energy when the time
increases. On the other hand, the desorption enthalpy increases gradually with the increase of
the time up to 72 h then become constant, maximum enthalpy was 14.99 kJ/g. The rate of water
vapour adsorption was found to be very high so that the extracted energy from the bentonite surface would not be a problem in any practical utilization of this system.King saud universit
A single-nucleotide polymorphism in the TP53 and MDM-2 gene modifies breast cancer risk in an ethnic Arab population.
Breast cancer is the most common oncological disease in women worldwide. Genetic predisposition to breast cancer can be associated with single-nucleotide polymorphisms (SNPs), which are observed in many women. Such gene polymorphisms, in combination with nutritional and environmental factors, can affect breast cancer development. The tumor suppressor TP53 and its negative regulator MDM2 play crucial roles in carcinogenesis. Previous case–control studies have revealed that TP53 72Arg > Pro and MDM2 309T > G polymorphisms contribute to the risk of common cancers. However, the relationship between these two functional polymorphisms and breast cancer susceptibility in the Saudi population has not been explored. In this study, we performed a case–control study of patients with breast cancer and healthy controls in a Saudi population using TaqMan-based real-time PCR. We found an increased breast cancer risk associated with the MDM2 GG [odds ratio (OR) = 2.79, 95% confidence interval (CI) = 2.04–3.92] and TG [OR = 1.43, 95% CI = 1.12–2.02] genotypes. An increased risk was also associated with the TP53 Pro/Pro genotype (OR = 2.19, 95% CI = 1.54–3.06) compared with the Arg/Arg genotype. The gene–gene interaction of MDM2 and TP53 polymorphisms increased breast cancer risk in a multiplicative manner (OR for the presence of both MDM2 GG and TP53 Pro/Pro genotypes = 7.71, 95% CI = 3.49–17.54). These findings suggest that polymorphisms of MDM2 and TP53 genes may be a genetic modifier for developing breast cancer in this ethnic population in the Arab world
Biochemical and molecular characterization of Haemophilus influenza isolated from Riyadh, Kingdom of Saudi Arabia
The objective of this study focused on the prevalence of Haemophilus influenza to confirm the colonies of H. influenza on the basis of their growth requirements and serotype distribution. This study prepared 80 isolates of H. influenze isolated from five different sources (eye, ear, sputum (SP), lower genital tract (TA), and nasopharyngeal (NPA)) with different ages for infants and elderly persons. The phenotypic characteristics, which included the biotype, serotype, antibiogram and β-lactamase production, were applied by using APINH and minimum inhibitory concentration (MIC). Also, the study focused on the identification of selected serotype using PFGE analysis. The discussion of this study differentiates the age groups occurrence in the isolates, alongside with non-typeable strain versus the typeable ones and their percentages in the sample of isolates. This clustering of most strains in one PFGE pattern might be explained with the colonel population structure of the encapsulated H. influenzaKing Saud Universit
Normal development of the Tibiofemoral angle in Saudi children from 2 to 12 years of age
The objectives of this study were to determine the normal chronological changes of the tibiofemoral (TF) angle in Saudi children and to compare between boys and girls regarding the development of the TF angle. Three hundred normal Saudi children with age range of 2-12 years participated in the study. They were selected from both sexes from nurseries and schools of five different areas in Riyadh. The subjects were divided according to their age into ten age groups. The TF angle was measured by plastic goniometers of different sizes. The results revealed a tendency of knock knees that decreased towards normal lower extremity alignment with age increment. There was no significant difference between boys and girls regarding the development of the TF angle (p=0.605). It was concluded that the normal Saudi children with age range of 2-12 years exhibit a valgus alignment of the lower extremities that decreases with age increment with no significant difference between boys and girls
"Low-Energy Radial Extracorporeal Shock Wave Therapy for Chronic Plantar Fasciitis: A Randomized Control Trial"
The purpose of this study was to investigate and clarify the value of low-energy radial extracorporeal shock-wave therapy (rESWT) to become a routine modality in the treatment of pain and functional disability associated with chronic plantar fasciitis. This study was double-blind, randomized, placebo-controlled trial. Forty six adult patients with unilateral plantar fasciitis were randomly assigned into two equal groups. The active treatment group (group I) received a total 3 treatments of shock wave exposure (3x2000 impulses) with 0.16 mJ/mm2, 2.5 bars and frequency of 8Hz given at weekly interval and the placebo group (group II) received the identical treatment protocol but shame exposed to shock-wave therapy. Pain and limitation of foot function were measured by Visual Analog Scale and Ankle-Hind Foot Scale respectively. The results revealed significant reduction of pain in both groups after 3 weeks of treatment (P=0.000 and 0.005 respectively). However there was a significant improvement in foot function in group 1 both after 3 weeks of treatment and 6weeks of follow up (P=0.000). Despite the small number of patients in this trial, low-energy rESWT was an effective non-invasive treatment method for chronic plantar fasciitis and may help the patients to avoid surgery
Appropriateness of Anaemia management in haemodialysis patients
The anemia of end stage renal disease (ESRD) is common and often severe complication that can be managed successfully by erythropoiesis-stimulating agents (ESA) administration. Aim: to investigate current practice of anaemia management in haemodialysis patients and to assess the appropriateness of anaemia management by comparing observed practice to the Kidney Disease Outcomes Quality Initiative (KDOQI) guideline recommendations. Setting and design: The study was conducted at two haemodialysis centers in Riyadh, Saudi Arabia. Data on anaemia parameters, comorbidities, ESA dosing and iron supplementation were collected. The data was collected for 7 months retrospectively from April to the end of May 2008 and prospectively from June to October 2008. Patients who were over 18 years of age with ESRD undergoing haemodialysis were included. Patients were excluded if they have cancer or receiving chemotherapy or radiotherapy. Results: Data were collected for 87 patients. Mean Hgb value for those patients was 11.16± 0.97 g/dL. Thirty nine of patients (45%) had mean Hgb values between 11.0 and 12.0 g/dL the target range recommended by KDOQI guideline. The mean weekly prescribed dose of erythropoietin was 8099± 5946 IU/Week (135 ±99 IU/Kg/Week). Information on ferritin concentrations was available for 48 (55%) patients. The mean serum ferritin concentration for those patients was 693 ±420.5 ng/mL. Fifty two patients had transferrin saturation (TSAT) values recorded. The mean TSAT value was 38.5% ±19.7. Conclusions: there is opportunity to improve anemia management in hemodialysis patients particularly thorough evaluation of causes of inadequate response rate and better monitoring and management of iron statusKing Saud Universit
Determination of Josamycin and Ciprofloxacin in their Pharmaceutical Dosage Forms by Spectrophotometry
Two simple, rapid and sensitive spectrophotometric methods for the determination of josamycin and ciprofloxacin in their pharmaceutical dosage forms are described. Method (A) is based on the reaction of aldehyde group of josamycin with 2,4-dinitrophenyl hydrazine in methanolic hydrochloric acid to yield a yellow product which absorbs maxima at 411 nm, whereas method (B) depends on the reaction of
ciprofloxacin with 1,2-naphthaquinone-4-sodium sulphonate in alkaline medium to form an orange coloured product measurable at 487 nm. The working conditions of both methods have been optimized and the reactions pathways were postulated. Regression analysis of Beer's law plots showed good correlation in the concentration ranges 10-160 and 5-120 μg/mL for josamycin and ciprofloxacin,
respectively. Detection and quantification limits for both methods are calculated. The methods were successfully applied to the determination of both drugs in bulk drugs and their formulations. Statistical treatment of the experimental results indicates that the accuracy and precision of the methods are analytically acceptable. The validity of the methods was evaluated by parallel determinations by established procedures and by recovery studies.King Saud Universit
