    Summary Pancreatic cancer still has dismal survival rates and high rates of early recurrence despite improvements of multimodal treatment options and more and more aggressive surgical approaches in recent years. Thus, precise and personalized management strategies to improve patient outcomes are needed. Circulating tumor DNA (ctDNA), a component of cell-free DNA (cfDNA) in body fluids, harbors genetic and epigenetic signatures of tumors and can be detected noninvasively for example through simple blood collections or peritoneal fluid during staging laparoscopy (liquid biopsies). This biomarker provides real-time insights into systemic tumor burden, heterogeneity, and genetic profile and has been proven to be of significant prognostic relevance for several gastrointestinal malignancies. Furthermore, the testing of ctDNA has emerged as a pivotal prognostic biomarker to indicate patients with high biological risk for recurrence and worse overall survival. Especially in pancreatic cancer, it has been shown that preoperative ctDNA detectability in peripheral blood is associated with systemic tumor burden (even volumetric). This indicates potential micrometastatic or subclinical disseminated disease, suggesting a benefit from neoadjuvant chemotherapy to address the systemic component of the disease prior to surgery. Furthermore, dynamic changes in ctDNA during systemic treatment can predict therapeutic response and guide adjustments in treatment regimens. Postoperatively, ctDNA presence could assist in detecting minimal residual disease which also predicts early relapse, facilitating timely intervention (or immediate adjuvant chemotherapy as already shown in colorectal cancer, e.g., Dynamic trial). Therefore, perioperative ctDNA detection has the potential to refine the management of pancreatic cancer, enhancing decision-making processes and optimizing personalized treatment approaches in serving as an additional highly sensitive biomarker for guiding treatment decisions between upfront surgery and neoadjuvant chemotherapy in patients with resectable pancreatic cancer. The first ever personalized change of treatment decision from upfront surgery (by current gold standard staging) to neoadjuvant chemotherapy based on additional liquid biopsy results was documented in March 2024 (Linz, Austria)

    Package for calculation of electrinic properties with tight-binding model

    Due to research results in recent years, it has become increasingly important to have accurate modeling and prediction of electronic properties in materials. This is important not only for fundamental research but also for industrial applica tions, especially in the field of semiconductor devices or nanoscale electronics. To achieve this goal, computational methods are needed that can model the quan tum mechanical behavior of electrons in solids. A widely used method in this context is the tight-binding (TB) model, which provides a good balance between computational efficiency and accuracy required to describe electronic structures of complex systems. This thesis shows the development of a software package created to calculate the electronic properties of a system using the tight-binding model. There are many motivations for the design of such a package. On one hand, the goal is to have a user-friendly tool that allows researchers to efficiently and easily simulate elec tronic band structures or similar related properties. Previous tools often have the problem that even though they provide very accurate results, they are sometimes very computationally intensive, especially for larger systems, if they use other models such as density functional theory (DFT). Therefore, in contrast to those, the tight binding model is very popular for simulations of larger systems.     Die vorliegende Diplomarbeit untersucht die Auswirkungen von Homeoffice auf die Mitarbeiterzufriedenheit in der österreichischen Versicherungsbranche. Vor dem Hintergrund der COVID-19-Pandemie hat sich Homeoffice als ein zentrales Element der modernen Arbeitswelt etabliert, was sowohl Chancen als auch Herausforderungen für Unternehmen und Mitarbeitende mit sich bringt. Ziel der Arbeit ist es, die Wechselwirkungen zwischen Homeoffice und zentralen Faktoren der Arbeitszufriedenheit – wie Autonomie, Workload, Anerkennung und der wahrgenommenen Bedeutung von Aufgaben – zu analysieren. Dabei greift die Arbeit auf theoretische Modelle wie das Job Characteristics Modell und die Zwei-Faktoren-Theorie zurück und testet empirisch die formulierten Hypothesen mittels einer quantitativen Befragung. Die Ergebnisse dieser Arbeit zeigen, dass Homeoffice die Arbeitszufriedenheit in der Versicherungsbranche sowohl positiv als auch negativ beeinflussen kann, abhängig von individuellen Merkmalen wie Alter, Geschlecht und beruflicher Position sowie von organisationalen Rahmenbedingungen. Insbesondere die erhöhte wahrgenommene Autonomie und die Flexibilität bei der Gestaltung von Arbeitszeit und -ort wirken sich positiv auf die Zufriedenheit der Mitarbeitenden aus. Gleichzeitig konnte festgestellt werden, dass die wahrgenommene Anerkennung durch Führungskräfte sowie die Bedeutung der ausgeführten Aufgaben im Homeoffice einen besonders starken Einfluss auf die Zufriedenheit haben. Darüber hinaus wurde deutlich, dass Homeoffice nicht zu einer signifikanten Veränderung der Arbeitsbelastung führt, jedoch die Qualität der Kommunikation und die Möglichkeit zum informellen Austausch unter Kollegen beeinflusst. Die Arbeit liefert praxisrelevante Empfehlungen zur Balance zwischen Homeoffice und Büroarbeit, um sowohl die Bedürfnisse der Mitarbeitenden als auch die Leistungsfähigkeit der Unternehmen langfristig sicherzustellen.The present thesis examines the impact of remote work (home office) on employee satisfaction in the Austrian insurance sector. Against the backdrop of the COVID-19 pandemic, remote work has become a central element of the modern working environment, presenting both opportunities and challenges for companies and employees. The aim of this study is to analyze the interactions between remote work and key factors of employee satisfaction, such as autonomy, workload, recognition, and the perceived significance of tasks. The study builds on theoretical frameworks such as the Job Characteristics Model and the Two-Factor Theory, while empirically testing the formulated hypotheses through a quantitative survey. The results of this research demonstrate that remote work can influence employee satisfaction in the insurance sector both positively and negatively, depending on individual characteristics such as age, gender, and professional position, as well as organizational conditions. In particular, increased perceived autonomy and flexibility in scheduling and workplace location positively impact employee satisfaction. At the same time, it was found that perceived recognition by supervisors and the significance of tasks performed in a remote setting play a particularly strong role in shaping satisfaction. Furthermore, the study reveals that remote work does not lead to a significant change in workload but does affect the quality of communication and opportunities for informal exchanges among colleagues.     Due to the recent advances in machine learning as well as in the field of physics-based fluid simulation, combining the two becomes a feasible idea. Novel Reinforcement Learning (RL) algorithms offer higher sample efficiency in addition to less sensitivity to hyperparameters when compared to earlier approaches. This is especially true for the Soft Actor-Critic algorithm that has risen drastically in popularity since its inception. The mentioned advantages makes RL algorithms more suited than ever for both complex simulated problems and real-world applications. Therefore, one aspect that has historically been challenging in the field of robotics can be approached in such a manner, namely fluid-handling. The reason behind the difficulty is the intricate dynamic behavior of fluids that is computationally expensive to simulate, or has been simulated using models that simplify the dynamics greatly. Inefficient but accurate calculations take too long and simple models did not capture effects such as waviness, fluid separation or sloshing to a sufficient degree. Therefore, both approaches are not ideally suited for calculating robot trajectories and even less so for real-time controllers. In recent years, the Smoothed Particle Hydrodynamics (SPH) method has been continuously advanced on by both the physics simulation and the computer graphics communities. The result is a method to simulate fluids both efficiently and accurately and, therefore, SPH has gained a lot of traction. As such, the SPH method offers a balance between accuracy and computational cost that works for training a machine learning model which can be used for trajectory planning, or, once trained, even as a real-time controller. This thesis aims at combining a SPH simulation with RL to learn how to handle fluids.     Basierend auf den bereits im Deskriptiven Bericht dargestellten Überlegungen wurden Fragen inhaltlich gruppiert und Skalen zusammengestellt. Damit wird einerseits das Ziel verfolgt, die teilweise etwas unübersichtlichen Aussagen zusammenzufassen und übersichtlich strukturierte Ergebnisdarstellungen zu ermöglichen, und andererseits aber auch insgesamt die Ergebnisse auf eine zuverlässigere Basis zu stellen und Interpretationen eine höhere Aussagekraft zu geben. Diese Erstellung von Skalen auf Schüler:innen- bzw. Lehrkraft-Ebene ermöglicht darüber hinaus auch, Zusammenhänge innerhalb der Fragebögen genauer zu untersuchen. Bei der Erstellung der Skalen wurde darauf geachtet, dass die dafür verwendeten Fragen inhaltlich zusammenpassen, so dass sie nicht mehrere Themenbereiche gleichzeitig abbilden, sondern nach Möglichkeit nur eng definierte Konstrukte

    Die vorliegende Diplomarbeit behandelt die Bedeutung der menschlichen Grundbedürfnisse in Verbindung mit dem subjektiven schulischen Wohlbefinden. Da sich das schulische Wohlbefinden sowohl auf die Leistung als auch auf die Gesundheit der Schülerinnen und Schüler auswirken kann, ist es von Bedeutung, sich näher mit dem schulischen Wohlbefinden auseinanderzusetzen. Da jedoch die Definition des Wohlbefindens sehr breit ist und verschiedene Definitionen vorherrschen, schafft die vorliegende Arbeit einen Überblick über den Forschungsstand mithilfe einer Metaanalyse. Im Zuge dessen wurde mit einer Definition des schulischen Wohlbefindens gearbeitet, die sich am Hedonismus orientiert und sich das Wohlbefinden demnach aus dem positiven Affekt, dem negativen Affekt sowie der Zufriedenheit zusammensetzt. Dabei wurden außerdem die Determinanten der Autonomie, der Kompetenz sowie der sozialen Eingebundenheit nach der Selbstbestimmungstheorie herangezogen. Im Zuge der Arbeit sind 36 Studien in die Auswertungen eingeflossen, die sich auf die Primarstufe und die Sekundarstufe beschränken. Dabei konnte festgestellt werden, dass vor allem die Zufriedenheit als Komponente des schulischen Wohlbefindens am häufigsten beforscht ist. Bezüglich der Instrumente lässt sich erkennen, dass es eine Vielzahl an Instrumenten gibt, die eingesetzt werden, wobei einzelne häufiger eingesetzt werden. Hinsichtlich der Korrelation der Wohlbefindensindikatoren und den Determinanten der Selbstbestimmungstheorie zeigten die standardisierten Korrelationskoeffizienten, die mithilfe von RStudio berechnet wurden, meist kleine bis mittlere Effekte. Die größten Effektstärken in Bezug auf die Korrelation mit dem positiven Affekt weisen dabei die Autonomieunterstützung, die Selbstwirksamkeit und die soziale Eingebundenheit auf. Hinsichtlich des negativen Affekts gibt es die höchste Effektstärke zwischen dem negativen Affekt und der Autonomieunterstützung. Betrachtet man die Zufriedenheit, so sind die Effektstärken bei der Korrelation zwischen der Zufriedenheit und der Autonomieunterstützung, der Kompetenzunterstützung und dem Lese-Selbstkonzept am höchsten.eingereicht von Theresa Maria PöhackerDiplomarbeit Johannes Kepler Universität Linz 202
