The role of program duration on student exchange outcomes: a cultural competency continuum perspective
In an era marked by rapid globalization, the ability to navigate multicultural environments and engage in meaningful cross-cultural interactions has become an essential competency for the modern workforce. In response to these global trends, Taiwan has strategically pursued international academic collaboration to enhance the global competitiveness of its students. This study investigates the impact of student exchange programs on the development of cultural competence, with a specific focus on how participation in such programs influences students' progression along the cultural competence continuum. Utilizing qualitative methods, this research conducts in-depth interviews with students who have participated in international exchange programs to explore their experiences of cultural adjustment, intercultural communication, and collaborative learning in diverse settings. Furthermore, the study compares the cultural learning outcomes of long-term and short-term exchange participants to assess how the duration of cultural immersion affects students’ ability to develop cultural awareness and competence. The findings aim to contribute to the ongoing discourse on international education by identifying key factors that support effective cultural learning. These insights are expected to inform the design and implementation of future exchange programs, enabling institutions and policymakers to better prepare students for global engagement. Models and algorithms for production planning under uncertain demand
Manufacturing companies need to efficiently coordinate complex production processes with multiple stages on scarce resources, in order to maintain competitiveness. Optimizing inventory levels for car components in the automotive industry, minimizing production costs for wafer production in the cost intensive semiconductor industry or guaranteeing high service levels in the retail industry are only three out of many available examples. These use cases share very similar production planning problems at the core, which can be formulated as mixed integer linear programs. In the medium term, production quantities for components and end items need to be determined in order to efficiently meet customer demand, while minimizing the resulting costs. In the short term, the resulting production orders need to be scheduled on the shop floor, aiming to minimize objectives such as makespan and tardiness. This thesis addresses medium term production planning by investigating multi-item multi-echelon capacitated lot sizing under uncertain demand. Short term production scheduling is additionally considered in several aspects within the thesis. First, by enriching medium term lot sizing models with short term components, such as setup carry-overs and subsequently by focusing on production scheduling problems as they arise in real-world settings.
Customer demand, which has a major influence on lot sizing decisions, is subject to uncertainty. Incorporating this uncertainty into optimization models is a crucial task in order to generate production plans that perform well, even when the actually realized customer demand is not known in advance. The main part of this thesis is represented by the first three parts and deals with different approaches of optimization under uncertainty to integrate stochastic demand into multi-item multi-echelon capacitated lot sizing.
In part 1, a simulation-optimization framework is introduced, combining a simulation environment with a medium term production planning component in order to perform rolling horizon planning. The forecast evolution approach is used within the simulation to generate customer demands that are updated over time and therefore represent the uncertainty that is also observed in reality. Since classical Material Requirements Planning (MRP) is still the de facto industry standard, it is used as a baseline planning method. However, MRP comes with two major drawbacks. Firstly, it lacks to consider resource capacity restrictions which prohibits the method from anticipating bottlenecks in the production system. Secondly, it does not consider demand uncertainty. In order to resolve these issues we introduce mixed integer linear programming models for the considered lot sizing problem that can be solved by commercial solvers to generate production plans replacing standard MRP. A deterministic optimization model, as well as a scenario-based two-stage stochastic programming model are presented. All three planning methods, namely MRP, deterministic and stochastic optimization are then evaluated and compared in a numerical experiment, where we investigate the impact of customer update behaviour, demand variation and resource utilization.
The presented two-stage stochastic lot sizing model is examined in more detail in part 2, where we additionally consider the possibility of setup carry-over. The combination of medium and short term planning within the simulation-optimization framework has shown that the conservation of setup states between periods is an important feature allowing to generate more realistic production plans. Setup carry-overs allow to anticipate short term effects by considering partial scheduling decisions. Moreover, we investigate budget-uncertainty robust optimization, which is an alternative approach to consider uncertainty within an optimization model. Robust optimization is especially interesting for large problem instances and cases with many scenarios, because the resulting robust models can be solved significantly faster than the stochastic programming models. The general concept of robust optimization follows a worst-case approach, however, by choosing suitable uncertainty budgets it can also generate production plans with competitive average performance compared to stochastic optimization. In order to predict the suitability of both approaches for a given problem instance we solve the respective optimization models for a large set of test instances and fit conditional inference trees to the resulting out-of-sample performances.
In part 3, the two-stage stochastic model with setup carry-overs is then extended to the more realistic multi-stage case, where production quantities can be adjusted at multiple periods in time, based on the most recent demand information. This modelling approach better represents how decisions are made in reality and is especially interesting in the case, where also components face external customer demand. Multi-stage stochastic programming models usually rely on scenario-tree representations, which lead to extremely large mathematical models because the scenario trees grow exponentially in the number of decisions stages. It is therefore very challenging to solve the resulting multi-stage models in a compact form. To resolve this issue we propose a progressive hedging algorithm that decomposes the problem by scenario. By reducing the number of scenario subproblems and leveraging parallel computing we aim to improve the solution procedure for problems that are extremely difficult to solve as a compact optimization model. We further adapt metaheuristic adjustment strategies that aim to lead the algorithm towards a consensus more efficiently. Moreover, we investigate two different variants to calculate this consensus in each iteration, namely a variant relying on averaging and another one using majority voting.
Finally, the fourth part consists of two conference publications and focuses on short term production scheduling and investigates cases that complicate the process in real-world applications, namely a large number of operations and complex scheduling constraints. First, a literature review provides an overview of different methods to tackle large scale production scheduling problems. Especially advanced metaheuristics combining population- and trajectory based approaches, as well as constraint programming techniques are successful in the literature to tackle these challenging problem instances. The gained insights are then used to address a real-world use case, where ideas of population-based metaheuristics are integrated into a trajectory-based method, namely a Tabu Search. Elite solutions are collected during the search procedure and used to extract promising solution patterns. These promising patterns are then conserved, while perturbation operators destroy large parts of the remaining incumbent solution in order to foster diversification.Fertigungsunternehmen müssen komplexe Produktionsprozesse mit mehreren Stufen auf knappen Ressourcen effizient koordinieren, um wettbewerbsfähig zu bleiben. Die Optimierung der Lagerbestände für Autoteile in der Automobilindustrie, die Minimierung von Produktionskosten in der kostenintensiven Halbleiterindustrie oder die Sicherstellung hoher Verfügbarkeiten im Einzelhandel sind nur drei von vielen Beispielen. Diese Anwendungsfälle teilen ähnliche Produktionsplanungsprobleme, die als gemischt-ganzzahlige lineare Programme formuliert werden können. Mittelfristig müssen Produktionsmengen für Komponenten und Endprodukte bestimmt werden, um die Kundennachfrage zu erfüllen und gleichzeitig die entstehenden Kosten zu minimieren. Kurzfristig müssen die daraus resultierenden Produktionsaufträge in der Produktionsstätte aufgeteilt und eingeplant werden, während Gesamtdurchlaufzeit oder Verspätungen minimiert werden sollen. Diese Arbeit befasst sich mit der mittelfristigen Produktionsplanung, indem die kapazitätsbeschränkte Losgrößenplanung für mehrere Produkte und mehrere Produktionsstufen unter unsicherer Nachfrage untersucht wird. Die kurzfristige Produktionsplanung wird zusätzlich in mehreren Aspekten behandelt. Kurzfristige Planungskomponenten werden in mittelfristige Losgrößenmodelle integriert und Feinplanungsprobleme mit typischen Eigenschaften aus realen Anwendungen werden untersucht.
Die Kundennachfrage, die einen wesentlichen Einfluss auf Losgrößenentscheidungen hat, ist von Unsicherheiten geprägt. Die Berücksichtigung dieser Unsicherheit in Optimierungsmodellen ist eine wesentliche Aufgabe, um Produktionspläne zu erstellen, die auch dann gut funktionieren, wenn die tatsächlich realisierte Kundennachfrage im Voraus nicht bekannt ist. Der Hauptteil dieser Arbeit besteht aus den ersten drei Teilen und befasst sich mit verschiedenen Ansätzen der Optimierung unter Unsicherheit, um stochastische Nachfrage in die kapazitätsbeschränkte Losgrößenplanung zu integrieren.
Im ersten Teil wird ein Simulations-Optimierungs-Framework vorgestellt, das eine Simulationsumgebung mit einer mittelfristigen Produktionsplanungskomponente kombiniert, um rollierende Planungen durchzuführen. Der sogenannte Ansatz der Forecast-Evolution wird in der Simulation verwendet, um Kundennachfragen zu generieren. Diese Nachfragen werden laufend aktualisiert und bilden so die Schwankungen und Unsicherheiten realer Nachfragedaten ab. Da die klassische Materialbedarfsplanung (MRP) nach wie vor der Industriestandard ist, wird sie als Referenzmethode verwendet. Allerdings hat MRP zwei wesentliche Nachteile. Erstens berücksichtigt es keine Kapazitätsrestriktionen, was die Methode daran hindert, Engpässe im Produktionssystem vorherzusehen. Zweitens wird die Nachfrageunsicherheit nicht berücksichtigt. Um diese Probleme zu lösen, stellen wir gemischt-ganzzahlige Optimierungsmodelle für das betrachtete Losgrößenproblem vor, die von kommerziellen Solvern gelöst werden können. Dabei sollen Produktionspläne erzeugt werden, um so klassiches MRP zu ersetzen. Ein deterministisches Optimierungsmodell, sowie ein szenariobasiertes zweistufiges stochastisches Optimierungsmodell, werden präsentiert. Die drei Planungsmethoden – MRP, deterministische und stochastische Optimierung – werden in einer Simulationsstudie evaluiert und verglichen. Dabei werden verschieden Kundenverhalten, Nachfrageschwankungen und Auslastungen der Ressourcen untersucht.
Im zweiten Teil wird das vorgestellte zweistufige stochastische Losgrößenmodell weitergehend untersucht. Es wird die zusätzliche Möglichkeit betrachtet, ein Produktionssetup in die nächste Planungsperiode zu übertragen, also Produktion kontinuierlich fortzusetzen. Die Kombination von mittelfristiger und kurzfristiger Planung innerhalb des Simulations-Optimierungs-Frameworks hat gezeigt, dass es für realistische Produktionsplanung von wesentlicher Bedeutung ist, Setup-Zustände auch zwischen den einzelnen Perioden zu berücksichtigen. Die Übertragung von Produktionssetups erlaubt es, kurzfristige Effekte in der mittelfristigen Planung zu berücksichtigen. Angewandt auf dieses erweiterte Losgrößenplanungsproblem untersuchen wir robuste Optimierung mit Budgetunsicherheit. Robuste Optimierung ist ein alternativer Ansatz zur Berücksichtigung von Unsicherheiten in Optimierungsmodellen. Diese Herangehensweise ist besonders interessant für große Probleminstanzen und Situationen in denen viele Szenarien benötigt werden. Die resultierenden robusten Modelle können signifikant schneller gelöst werden als stochastische Optimierungsmodelle. Obwohl robuste Optimierung einem Worst-Case-Ansatz folgt, können damit Produktionspläne erstellt werden, die auch im Mittel zu kostengünstigen Produktionsplänen führt, insbesondere wenn geeignete Unsicherheitsbudgets gewählt werden. Um die Eignung beider Ansätze, also robuste und stochastische Optimierung, für eine gegebene Probleminstanz zu bestimmen, lösen wir die jeweiligen Optimierungsmodelle für eine große Menge von Testinstanzen. Dann werden sogenannte Inference Trees verwendet um die erwartete Performance der Modelle vorherzusagen.
Im dritten Teil wird das zweistufige stochastische Modell auf den realistischeren mehrstufigen Fall erweitert, bei dem die Produktionsmengen zu mehreren Zeitpunkten basierend auf den neuesten Nachfrageinformationen angepasst werden können. Dieser Modellierungsansatz stellt besser dar, wie Entscheidungen in der Realität getroffen werden. Mehrstufige stochastische Optimierungsmodelle basieren üblicherweise auf Szenariobaum-Darstellungen, die zu extrem großen mathematischen Modellen führen. Die Szenariobäume wachsen exponentiell mit der Anzahl der Entscheidungsstufen. Es ist daher eine große Herausforderung, die resultierenden mehrstufigen Modelle in kompakter Form zu lösen. Deshalb wird in dieser Arbeit ein Progressive-Hedging-Algorithmus präsentiert, der das Problem in kleinere Teilprobleme zerlegt. Für jedes Szenario des Szenariobaums wird dabei ein deterministisches Losgrößenproblem gelöst. Durch die Reduzierung der Anzahl dieser Teilprobleme und Parallelisierung sollen Probleme schneller gelöst werden, für die ein kompaktes Modell nur in sehr langer Rechenzeit gelöst werden könnte. Es werden zwei Varianten zur Berechnung einer Konsens-Lösung untersucht. Der Progressive-Hedging-Algorithmus wird dabei in dieser Arbeit um Adaptions-Strategien erweitert, die die Suche nach einem Konsens effizienter gestalten sollen.
Schließlich konzentriert sich der vierte Teil, bestehend aus zwei Konferenzpublikationen, auf die kurzfristige Produktionsfeinplanung. Der Fokus liegt auf Problemeigenschaften, die den Feinplanungsprozess in realen Anwendungen erschweren, nämlich eine große Anzahl von Operationen und komplexe Planungsrestriktionen. Zunächst wird ein Literaturüberblick über verschiedene Methoden zur Bewältigung großer und komplexer Produktionsplanungsprobleme gegeben. Insbesondere fortgeschrittene Metaheuristiken, die Populations- und Trajektorienbasierte Ansätze kombinieren, sowie Techniken des Constraint Programming, sind in der Literatur erfolgreich. Die gewonnenen Erkenntnisse werden dann an einem realen Anwendungsfall getestet. Es werden Ideen von populationsbasierten Metaheuristiken in eine Trajektorien-basierte Methode, nämlich eine Tabu-Suche, integriert. Sogenannte elitäre Lösungen werden während des Suchverfahrens gesammelt und genutzt, um vielversprechende Lösungsmuster zu extrahieren. Diese vielversprechenden Muster werden dann konserviert, während Störungsoperatoren große Teile der verbleibenden Lösung zerstören, um eine größere Diversifikation zu erreichen.Author Dipl.-Ing. British Journal of Dermatology / Evidence of immunogenic cell death (ICD) and ICD-dependent dendritic cell activation induced by extracorporeal photopheresis in patients with leukaemic forms of cutaneous T-cell lymphoma
Abstract Background Despite novel therapeutic options, the long-term management of cutaneous T-cell lymphoma (CTCL) remains challenging. Extracorporeal photopheresis (ECP) is an immunomodulating photochemotherapy associated with higher overall survival when used for the treatment of leukaemic forms of CTCL. Its exact mode of action has not been fully elucidated. Immunogenic cell death (ICD) is pivotal in cancer immunotherapy, marked by the release of damage-associated molecular patterns that enhance dendritic cell (DC) maturation and cytotoxic T-lymphocyte responses. Objectives To explore ICD in patients with leukaemic forms of CTCL during ECP and its effect on DC activation. Methods We conducted in vitro studies with peripheral blood mononuclear cells (PBMCs) from healthy donors and ex vivo experiments with white blood cells (WBCs) from patients with leukaemic forms of CTCL undergoing ECP. We assessed cell viability, apoptosis and ICD markers [ATP, high mobility group box 1 protein (HMGB1), calreticulin] using flow cytometry, enzyme-linked immunosorbent assay and quantitative polymerase chain reaction. Engulfment assays evaluated DC activation by ECP-treated CD4+ T cells. Results ECP-treated healthy PBMCs and WBCs from patients with leukaemic forms of CTCL showed a significant induction of ICD hallmarks, including ATP release, HMGB1 secretion and calreticulin surface exposure. In patients with leukaemic forms of CTCL, calreticulin exposure was mainly present in CD4+CD26– T cells, indicating greater ICD susceptibility of malignant T cells. ECP-treated CD4+ T cells were phagocytosed by DCs, a process that was found to be dependent on ICD signals. Conclusions ECP induces ICD in malignant T cells and, to a lesser extent, in healthy T cells, facilitates DC activation. These findings suggest that ECP enhances targeted immune responses against malignant T cells in leukaemic forms of CTCL, offering new insights into its therapeutic mechanisms and potential applications in cancer immunotherapy

Univ.-Prof. Dr. Horst Aspöck 85 – a pioneer of Medical Parasitology in Austria and of research on the Neuropterida. The renowned and internationally well-known parasitologist and entomologist Univ.-Prof. Dr. Horst Aspöck celebrated his 85th birthday on July 21 2024. This is an occasion to document his extensive lifework. The present paper comprises an account of his life in tabular form, a complete list of his publications and – besides several other documentations (co-authors, search terms, dedications) – a list of all new taxa described by him, largely with his wife and colleagues from many other countries. His opus presently comprises 813 publications (among them several books and without abstracts), largely in well-respected journals. Aspöck leaves an inerasable trace, not only by his activity at the Medical University, but he also shaped and still shapes Neuropterology together with his congenial wife, Ulrike, as only few other scientists. It is not a surprise that Aspöck still presents lectures (including compulsory ones) and that he continues to have close contacts to students and colleagues at the universities
Statistical Inference using Truncated Normal Distribution
The effective population size (Ne) is an important metric for understanding the factors that cause changes in allele frequencies in population genetics. The current technique for estimating this effective population size, which uses a Gaussian mixture model and the EM algorithm, can be challenging because the number of mixture components in this application is typically large and unknown. Therefore, an alternative approach using the truncated normal distribution is described. Such statistical inference methods based on truncated or censored samples have been used in a variety of areas, including quality control, biological studies, official statistics, and life science problems.
The main goal of this thesis is to compare the two methods and to describe in which situations each method works best. This will be done through a simulation study consisting of three steps. First, both methods are applied to simulated normally distributed data, then to simulated genome data without linkage, and finally to simulated genome data with linkage.
Both methods work similarly well when the data is not affected by selection. Summary Management of oligometastatic disease in colorectal cancer is increasingly focused on local treatment strategies that can significantly enhance survival outcomes. The integration of surgical resection, thermal ablation and catheter-directed therapies, as well as ablative radiotherapy, and the combination with systemic therapy are critical components of a multidisciplinary approach to optimize patient care. The purpose of this manuscript is to review current definitions of oligometastatic cancer and summarize the current literature describing treatment and outcomes of local therapy in patients with oligometastatic colorectal cancer
Interventionelle Therapie bei Neugeborenen mit Aortenklappenstenose
Zusammenfassung
Hintergrund: Eine schwere kongenitale Aortenklappenstenose kann sich bereits in der Neonatalperiode manifestieren und erfordert ein unmittelbar therapeutisches Eingreifen. Die Behandlung der kongenitalen Aortenklappenstenose kann sowohl interventionell als auch chirurgisch erfolgen. Bei der Ballondilatation, die die interventionelle Therapiemethode darstellt, wird die stenosierte Klappe durch einen Ballon aufgedehnt. Die optimale Therapiestrategie sowie Risikofaktoren für ein schlechtes Outcome sind jedoch bis heute Diskussionsthema.
Ziel: Das primäre Studienziel war die Evaluierung des Outcomes von Neugeborenen mit kongenitaler Aortenklappenstenose hinsichtlich der Wiedereingriffsrate, möglicher Komplikationen sowie der Mortalität nach interventionellem Eingriff. Es wurde angenommen, dass Neugeborene mit Aortenklappenstenose und trikuspid angelegter Aortenklappe ein besseres Outcome nach interventioneller Therapie aufweisen als jene mit bikuspider Klappenmorphologie.
Methoden: Im Rahmen der vorliegenden Arbeit wurde eine monozentrische Studie mit retrospektiver Datenanalyse aller Patient*innen durchgeführt, die aufgrund einer kongenitalen Aortenklappenstenose in der Neonatalperiode zwischen 1.Jänner 2000 und 30.September 2022 am Kinderherzzentrum Linz, Med Campus IV des Kepler Universitätsklinikums primär interventionell mittels Ballondilatation behandelt wurden. Es erfolgte eine rein deskriptive Datenauswertung.
Ergebnisse: Das Studienkollektiv umfasste 38 Patient*innen mit überwiegend männlichen Neugeborenen (männlich:weiblich = 3,22:1). Die häufigste postinterventionelle Komplikation war eine signifikante akute Aortenklappeninsuffizienz, die bei 34,2% der Patient*innen festgestellt wurde. In 35 Patient*innen (92,1%) mit verfügbaren Werten wurde der Peak-to-Peak-Gradient von durchschnittlich 63,43 (±28,32) auf 23,54 (±13,56) mmHg reduziert. Kein Neugeborenes verstarb intraprozedural. Die 1-Jahres- sowie 5-Jahres-Überlebensrate betrug 88,1 (±5,6) %. Die Reinterventionsfreiheit lag nach einem Jahr bei 50,8 (±8,6) % und nach fünf Jahren bei 43,1 (±8,9) %. Patient*innen ohne Reintervention hatten echokardiographisch im Median einen etwas höheren Aortenklappenanulus-, einen höheren Aorta ascendens- und einen höheren Aortenisthmus-Z-Score, sowie einen leicht geringeren linksventrikulären Durchmesser und eine bessere linksventrikuläre Funktion als Patient*innen mit Reintervention. 17 von 21 (81%) Patient*innen mit echokardiographisch beurteilbarer Klappenmorphologie wiesen eine funktionell bikuspide Aortenklappe auf. Die 1-Jahres-Überlebensrate für Patient*innen mit funktionell bikuspider Aortenklappe lag bei 92,9 (±6,9) % und jene von Patient*innen mit nicht funktionell bikuspider Aortenklappe bei 50 (±25) %. Die Reinterventionsfreiheit der Patient*innen mit funktionell bikuspider Aortenklappe betrug nach einem Jahr 57,7 (±13,2) %. Patient*innen mit nicht funktionell bikuspider Aortenklappe benötigten alle innerhalb eines Jahres nach der primären Ballondilatation eine Reintervention.
Schlussfolgerungen: Komplikationen und Reinterventionen nach primär interventioneller Behandlung von Neugeborenen mit Aortenklappenstenose waren häufig. In einigen Patient*innen konnten Folgeeingriffe jedoch langfristig vermieden werden. Weitere prospektive Studien sind nötig, um den Einfluss der Aortenklappenmorphologie auf das Outcome in interventionell behandelten Neugeborenen mit Aortenklappenstenose zu evaluieren.Abstract
Background: A severe congenital aortic valve stenosis can, in rare cases, manifest in the neonatal period requiring immediate therapeutic intervention. Treatment for congenital aortic valve stenosis can be either interventional or surgical. Balloon dilation represents the interventional treatment method in which the stenosed valve is dilated using a balloon. However, the optimal treatment strategy and risk factors for unfavorable outcomes remain a topic of discussion.
Aim: The primary objective of this study was to evaluate the outcome of neonates with congenital aortic stenosis in terms of complications, reinterventions and mortality after interventional treatment. It was hypothesized that neonates with aortic valve stenosis and tricuspid aortic valve anatomy would experience better outcomes after interventional treatment than those with a bicuspid aortic valve.
Methods: In this single-center retrospective study, data from all patients who were primary treated with ballon dilatation for aortic valve stenosis in the neonatal period between January 1st, 2000, and September 30, 2022 at the Pediatric Heart Center Linz, Med Campus IV of the Kepler University Hospital, were evaluated. The data were analyzed descriptively.
Results: The study cohort comprised 38 patients, primarily male newborns (male:female = 3,22:1). The most common postinterventional complication was significant acute aortic valve insufficiency, which was found in 34,2% of the patients. In 35 (92,1%) patients with available values the peak-to-peak gradient was reduced from an average of 63,43 (±28,32) to 23,54 (±13,56) mmHg. There was no intraprocedural death. The overall survival rate was 88,1 (±5,6) % at one year and at five years. Freedom from reintervention was 50,8 (±8,6) % at one year and 43,1 (±8,9) % at five years. In patients who did not undergo reintervention, the median z-scores of the aortic anulus, the aortic isthmus and the ascending aorta, based on echocardiographic measurements, were slightly higher, left ventricular diameter was slightly smaller and left ventricular function was better than in patients who underwent reintervention. Out of 21 patients, in whom the morphology of the aortic valve could be assessed by echocardiography, 17 (81%) were found to have a functional bicuspid aortic valve. Survival at one year was 92,9 (±6,9) % in the subgroup of patients with a functional bicuspid aortic valve and 50 (±25) % in the subgroup of patients with non-functional bicuspid valve. In patients with functional bicuspid aortic valve freedom from reintervention was 57,7 (±13,2) % at one year. All patients with a non-functional bicuspid aortic valve required reintervention within one year after the primary ballon dilatation.
Conclusion: Complications and reinterventions after primary interventional treatment of neonatal aortic valve stenosis were frequently observed. Nevertheless, in some patients, long term prevention of reinterventions was achieved.
