    Turning by-products into health: fermentation-driven production of bioactive compounds for cardiovascular disease prevention

    Cardiovascular diseases (CVDs) remain the leading cause of death globally and pose a growing public health burden due to their economic impact on healthcare systems. Metabolic conditions and unhealthy lifestyles contribute to the progression of conditions closely linked to CVDs. Functional foods containing bioactive compounds have been demonstrated as a valid dietary strategy to mitigate risk factors associated with CVDs, offering multi-targeted preventive effects. However, their production can vary in methods, safety, yield, sustainability, and cost, influencing their accessibility and widespread use. Fermentation is an amply validated method for the design of functional foods, offering a sustainable and cost-effective approach to industrial production of bioactive compounds. Moreover, using agri-food by-products can further reduce costs and increase the process sustainability and product bioactivity. This process enables the biosynthesis and enrichment of key cardioprotective metabolites, which have been shown to exert antihypertensive, lipid-lowering, antioxidant, and other health-promoting activities. The review provides an overview of recent studies on bioactive compounds such as monacolin K (MK), peptides, γ-aminobutyric acid (GABA), phenol, and other health-promoting molecules. Here, cardiovascular benefits and the role of fermentation in their production from low-cost substrates are discussed to meet growing global demands for functional foods

    The presentation focused on the concept of cultural infrastructure, asking whether it can serve as a governance framework that integrates the approaches of commoning and infrastructuring. Through an interdisciplinary lens, and embracing a reading of property rights in light of Article 42.2 of the Italian Constitution, it examined different models of collective cultural management of goods, highlighting their points of convergence and the potential to subsume them under the broader notion of cultural infrastructure

    Traces and Memories of a Rural Reuse of Roman Amphitheatres: Archaeological Restoration and Landscape

    Following the rise of Christianity and the severe economic crisis that affected the late Roman empire in Europe, Roman amphitheatres faced different fates, influenced by functional, social and economic factors. Urban systems gradually pushed amphitheatres out of the main urban core. They became isolated, and their ruins were repurposed for agriculture and for the defence from nearby cities. Over the years, these structures were buried to make way for new terraces or agricultural fields. In some other cases, they were altered by overlapping layers of other elements for residential or productive purposes. Despite this, the ancient amphitheatres have endured and drawn attention due to their ruins, their landscape and the narrative of their palimpsest. This narrative highlights the archaeological component and tells the story of their transformation over time. By focusing on significant case studies, such as the Anfiteatro Campano of Santa Maria Capua Vetere and the amphitheatre of Cuma, this study aims to examine the vernacular values of the ancient ruins, as well as the outcomes of the farmers activities –such as drainage channels or signs of ploughing and cultivation– on archaeology and landscape. The study proposes possible strategies and criteria for conserving the archaeological sites and enhancing the use of the storytelling, to communicate both their original story and their agricultural reuse over the years

    Il quinto volume della collana “Progettare Archeologia”, a cura di Oreste Lubrano e Valerio Tolve, raccoglie gli esiti di due edizioni del workshop in seno al programma di Master in “Museografia, Architettura e Archeologia, Progettazione Strategica e Gestione Innovativa delle Aree Archeologiche” dell’Accademia Adrianea di Architettura e Archeologia, ospitato e organizzato dall’Università degli Studi di Napoli “Federico II”. Quello tra i due istituti è ormai un sodalizio assolutamente collaudato nel tempo. A Napoli infatti l’Accademia Adrianea non ha solo trovato un supporto logistico e una sede di lavoro, ma un proficuo sostegno scientifico che rappresenta il valore aggiunto di quest’esperienza: la possibilità di confrontarsi tanto con il ricco patrimonio archeologico campano, ma soprattutto l’occasione di misurarsi con un metodo di lavoro che interpreta la necessaria innovazione a partire da referenti eccellenti prelevati dallo spessore storico della nostra antica disciplina, sottolineando la stessa dimensione ‘archeologica’ dell’architettura e del suo patrimonio. L’overture di Pier Federico Caliari tende ad inquadrare il tema specifico del workshop – e di questo volume – entro il quadro complessivo di quella particolare branca dell’architettura che si è affermata nel tempo – e non a caso particolarmente in ambito mediterraneo: la cosiddetta ‘archeologia da architetti’. Seguono una serie di saggi di carattere teorico, a firma dei docenti che hanno curato e condotto le due edizioni del workshop su Poseidonia/Paestum – Gaetano Fusco, Federica Visconti, Renato Capozzi, Francesca Coppolino, Oreste Lubrano e Valerio Tolve – ai quali è affidato l’inquadramento teorico del metodo operativo e del punto di vista. La sezione dei progetti, a cura di Nicola Fortunato, Oreste Lubrano e Valerio Tolve, è invece dedicata all’illustrazione degli esiti progettuali che, ancorché limitati e costretti nel tempo breve del workshop, mostrano soluzioni site specific per le aree individuate entro il vasto ambito del Parco Archeologico di Paestum. Chiudono il volume i ‘punti di vista’ di Stefania Pollone, Mariateresa Giammetti, Angela D’Agostino e Claudia Sansò che, ospiti negli anni nei jury di critica finale, hanno voluto intrecciare, nell’occasione di questa pubblicazione, le loro personali esperienze di ricerca con la dissertazione dei progetti degli studenti

    Highly variable genomic methylation in the Beckwith-Wiedemann syndrome associated with multi-locus imprinting disturbances

    Background: The expression of imprinted genes, which depends on their gamete of origin, is regulated by DNA sequences characterized by differential methylation between the maternal and paternal alleles (also known as germline differentially methylated regions or gDMRs). The most common molecular defect associated with Beckwith-Wiedemann syndrome (BWS), a condition linked to overgrowth and tumours, is the loss of methylation of the KCNQ1OT1-TSS gDMR located on chromosome 11p15.5 (also known as IC2 LoM). Approximately one-third of BWS patients with IC2 LoM exhibit multi-locus imprinting disturbances (MLID). While maternal-effect variants in proteins of the oocyte subcortical maternal complex (SCMC) have been linked to MLID, the underlying mechanisms and health impact of this epigenetic disturbance remain unclear. Results: We used the Infinium EPIC methylation array to investigate whole-genome CpG methylation in 64 BWS patients with IC2 LoM and 37 control subjects. We distinguished two patient groups, one with a variable methylation level of 24 gDMRs and the other with single-locus IC2 LoM. We observed that the mothers of the former patient group carried more variants in maternal-effect genes than those of the latter group, and 50% of them, but none of the latter group had variants in the SCMC genes. Additionally, in the former group, the mothers were older at the time of pregnancy, and the patients showed higher variation in methylation levels of thousands of CpGs located in non-imprinted loci, including protochaderins and cancer-associated genes. We found no differences in clinical features or in the incidence of assisted reproductive technology between the two patient groups. However, multiple affected siblings and recurrent miscarriages were observed only among cases with biallelic maternal-effect SCMC gene variants. Conclusions: This study demonstrates that the BWS patients with MLID exhibit highly variable methylation changes that affect both imprinted and non-imprinted loci in a seemingly stochastic manner throughout the genome. These findings support the hypothesis that MLID results from the interaction of maternal-effect genes and environmental factors in aged oocytes, leading to disordered DNA methylation in the whole genome. Future research should investigate whether and how these epimutations impact the health of affected individuals, particularly in adulthood
