Pathogenic variants of sphingomyelin synthase SMS2 disrupt lipid landscapes in the secretory pathway
Sphingomyelin is a dominant sphingolipid in mammalian cells. Its production in the trans-Golgi traps cholesterol synthesized in the ER to promote formation of a sphingomyelin/sterol gradient along the secretory pathway. This gradient marks a fundamental transition in physical membrane properties that help specify organelle identify and function. We previously identified mutations in sphingomyelin synthase SMS2 that cause osteoporosis and skeletal dysplasia. Here, we show that SMS2 variants linked to the most severe bone phenotypes retain full enzymatic activity but fail to leave the ER owing to a defective autonomous ER export signal. Cells harboring pathogenic SMS2 variants accumulate sphingomyelin in the ER and display a disrupted transbilayer sphingomyelin asymmetry. These aberrant sphingomyelin distributions also occur in patient-derived fibroblasts and are accompanied by imbalances in cholesterol organization, glycerophospholipid profiles, and lipid order in the secretory pathway. We postulate that pathogenic SMS2 variants undermine the capacity of osteogenic cells to uphold nonrandom lipid distributions that are critical for their bone forming activity
Interplay between SERCA, 4E-BP, and eIF4E in the Drosophila heart
Appropriate cardiac performance depends on a tightly controlled handling of Ca2+ in a broad range of species, from invertebrates to mammals. The role of the Ca2+ ATPase, SERCA, in Ca2+ handling is pivotal, and its activity is regulated, inter alia, by interacting with distinct proteins. Herein, we give evidence that 4E binding protein (4E-BP) is a novel regulator of SERCA activity in Drosophila melanogaster during cardiac function. Flies over-expressing 4E-BP showed improved cardiac performance in young individuals associated with incremented SERCA activity. Moreover, we demonstrate that SERCA interacts with translation initiation factors eIF4E-1, eIF4E-2 and eIF4E-4 in a yeast two-hybrid assay. The specific identification of eIF4E-4 in cardiac tissue leads us to propose that the interaction of elF4E-4 with SERCA may be the basis of the cardiac effects observed in 4E-BP over-expressing flies associated with incremented SERCA activity
Free word association analysis of Germans' attitudes toward insects
In this paper, the results of two free word association tests were analyzed to gain insight into Germans' attitudes toward insects. We used a novel approach in the form of an association network with the help of the software “Gephi” that was originally developed for social network analysis. The influence of attitude on the willingness to donate (WTD) and actual donation to an insect conservation project was investigated as well. Data collection was conducted via an online questionnaire (n = 515; Mage = 49.36, SD = 16.73; female = 50.1%). For the first test, participants listed three associations for the prompt “insect.” The associations were assigned to the three components of attitude: affective, cognitive, and behavioral. For the second test, participants named insects they pictured when thinking about “insects.” The results were taxonomically classified. The WTD was assessed with a Likert-type scale and an actual donation could be made at the end of the online questionnaire. “Bee” was the most frequently named association, followed by “useful,” “nature,” “pollination,” and “pesky.” “Pesky” was most often named with “useful,” indicating that being aware of insects' usefulness is not enough to supersede negative associations. In the second test, only 6% of the associations were on the species level, which suggests little taxonomic knowledge about or interest in insects. Linear regression revealed that positive affective associations had a positive influence on the WTD and negative affective associations had a negative influence on the WTD, both with a small effect size. We advise educating people not only about the usefulness of insects but also fostering positive, personal encounters to increase positive affective associations and decrease negative ones

Der Begriff hat seinen Ursprung in der völkerrechtlichen Unterscheidung von Vertragsparteien und dritten Staaten. Er wird zur Bezeichnung von Angehörigen von Staaten benutzt, die nicht Mitglied der Europäischen Union sind. Nach einer Phase, in der es keinen einheitlichen Sprachgebrauch gab, hat sich Ende der 1990er Jahre der Begriff Drittstaatsangehörige durchgesetzt und er wird generell in der rechtswissenschaftlichen Literatur und in der amtlichen Sprache der EU-Rechtsvorschriften verwendet. Seine Funktion besteht darin, die grundlegende Unterscheidung zwischen Unionsbürger:innen und Staatsangehörigen anderer Staaten zu kennzeichnen. Während die einen weitgehende Freizügigkeit genießen, steht den anderen Freizügigkeit nur dann zu, wenn sie ihnen ausdrücklich im Unionsrecht eingeräumt wird. Nicht erfasst werden mit dem Begriff die erheblichen Unterschiede zwischen verschiedenen Gruppen von ‚Drittstaatsangehörigen‘. Dennoch ist seine Verwendung unumstritten
Navigation Control and Path Planning for Autonomous Mobile Robots
Mobile robots need to move in the real world for the majority of tasks. Their control is often intertwined with the tasks they have to solve. Unforeseen events must have an adequate and prompt reaction, in order to solve the corresponding task satisfactorily. A robust system must be able to respond to a variety of events with specific solutions and strategies to keep the system running. Robot navigation control systems are essential for this. In this thesis we present a robot navigation control system that fulfills these requirements: Move Base Flex.
Furthermore, the map representation used to model the environment is essential for path planning. Depending on the representation of the map, path planners can solve problems like simple 2D indoor navigation, but also complex rough terrain outdoor navigation with multiple levels and varying slopes, if the corresponding representation can model them accurately. With Move Base Flex, we present a middle layer navigation framework for navigation control, that is map independent at its core. Based on this, we present the Mesh Navigation Stack to master path planning in complex outdoor environments using a developed mesh map to model surfaces in 3D. Finally, to solve path planning in complex outdoor environments, we have developed and integrated the Continuous Vector Field Planner with the aforementioned solutions and evaluated it on five challenging and complex outdoor datasets in simulation and in the real-world.
Beyond that, the corresponding developed software packages are open source available and have been released to easily reproduce the provided scientific results
Assembly and composition of the cECM is critical for heart physiology
The present study focuses on the cardiac function of Drosophila melanogaster. Drosophila heart parameters are evolutionarily conserved, making Drosophila a useful human heart disease model. This model enables the in vivo investigation of physiological and genetic methods. This thesis is subdivided into four parts: parts 1-3 comprise the introductions of three publications, and part 4 presents unpublished data. The first publication is about the heart physiology of Drosophila. It explains how intracardiac valve cells work and proves their participation in blood flow directionality. A databased model shows the orientation of myofibrils within the valve cell. The myofibrils allow the valve cells to oscillate between a roundish and elongated cell shape. A toll-GFP enhancer line was shown to mediate strong reporter gene activity in the intracardiac valve of third instar larvae, pupae and adults. Transmission electron microscopy (TEM) analyses and immunohistochemical studies showed the differentiation of larval valve cells for the first time. The second publication focuses on the cardiac extracellular matrix (ECM), which contains two unique proteins - Lonely heart (Loh) and Pericardin (Prc). The study demonstrated that Loh is crucial for Prc recruitment to the developing matrix. Loh is anchored to the ECM by its thrombospondin type 1 repeat (TSR1-1) with its embedded putative glycosaminoglycan (GAG)-binding side. The N-terminus of Loh is proposed to face the plasma membrane. Prc is presumably recruited by two Loh TSR1 domains (TSR1-2 and TSR1-4). Nearly all Drosophila tissues, except salivary glands, create Prc networks through ectopic Loh expression. The study also found that the amount of Prc and Loh in the cardiac ECM influences heart function. The third publication investigated a set of neuropeptides and their ability to modulate cardiac function in third instar larvae. The results showed that 11 of the 19 tested peptides significantly affected the heart function in semi-intact larvae. Furthermore, the peptides’ in vivo relevance was tested through the knockdown of chronotropic peptide precursors. The study found that a RNAi mediated knockdown of all respective peptide precursors affected the heart rate. By combining semi-intact heart preparations and in vivo analyses, we identified several heartbeat-modulatory peptides in Drosophila. The unpublished data introduces a new software program called HIRO. It is written in Java, platform-independent and can easily detect the heart rhythm. Only mild anaesthesia and basic equipment are needed to record the Drosophila heartbeat. HIRO was used to show the influence of the RNAi-mediated downregulation of critical ECM proteins in Drosophila third instar larvae. The screen revealed Myospheroid and Laminin A as promising candidates that can significantly affect the heart parameters. HIRO is optimised for future applications and can be used as a high-throughput screening software with a simple setup. Taken together, this thesis provides new insights into the physiology and function of the Drosophila heart. The developed software HIRO comes with a user-friendly interface and a step-by-step introduction to easily conduct heart parameter measurements. HIRO will help to expand our knowledge of the fundamental processes in the model organism Drosophila melanogaster
Synthese, Charakterisierung und Untersuchung der spektroskopischen Eigenschaften von Nanopartikeln mit Gagarinit- und Apatitstruktur
Hexagonales ß-NaREF4 und Strontiumchlorapatit (Sr5(PO4)3Cl) haben sich als Wirtsgitter
für Ionen der Lanthanoide bewährt und bilden die Basis für eine Reihe von
(Nano-)Materialien mit hervorragenden Lumineszenzeigenschaften. Die Struktur der ß-
Natriumseltenerdtetrafluoride leitet sich von der des natürlich vorkommenden Minerals
Gagarinit (NaCaYF6) ab. In der hier vorliegenden Arbeit wurde die Synthese von Nanopartikeln
mit Gagarinitstruktur und der Zusammensetzung ß-Na1,5- x/2MExRE1,5- x/2F6
(ME = Ca2+, Sr2+) untersucht, sowie deren Dotierung mit dreiwertigen Lanthanoidionen.
Es wird gezeigt, dass die Nukleations- und Wachstumsmechanismen von
Nanopartikeln mit Gagarinitstruktur grundsätzlich den bekannten Mechanismen bei
ß-NaREF4-Nanopartikeln entsprechen. Es wird ferner gezeigt, dass beim Übergang von
ß-NaREF4 zu ß-Na1,5- x/2MExRE1,5- x/2F6 (ME = Ca oder Sr) eine Stabilisierung der kubischen
a-Phase von NaREF4 und eine Verringerung der Anzahl an Keimen der ß-Phase
auftritt. Zusätzlich werden durch den Einbau von Sr2+ die Gitterparameter vergrößert,
wodurch sich a-NaSrGdF6 ideal als Schalenmaterial zur Umhüllung von ß-NaREF4-
Kernen der leichten Lanthanoide eignet. Die Ergebnisse der Röntgenfluoreszenzanalyse
und die spektroskopische Untersuchung von ß-Na1,5- x/2SrxEu1,5- x/2F6-Nanopartikeln legen
nahe, dass das Erdalkaliion die Natrium- und Seltenerdionen auf der 1f-Position
im Kristallgitter von ß-NaREF4 ersetzt. Die Ergebnisse der Fluoreszenzspektroskopie
des Eu3+-dotierten Materials zeigen insbesondere, dass der Einbau von Sr2+-Ionen die
lokale Symmetrie der optisch aktiven Eu3+-Ionen erhöht.
Weiterhin wird gezeigt, dass sich Sr5(PO4)3Cl-Nanopartikel mithilfe von Phosphatsalzen
organischer Basen auch in dem von der NaREF4-Nanopartikelsynthese
bekannten Lösungsmittelgemisch Ölsäure/Octadecen herstellen lassen. Durch Variation
der Anionenkonzentrationen, Einbau von verschiedenen Dotierungsionen und
Anwendung von Kern-Schale-Synthesemethoden lassen sich Nanopartikel mit Apatitstruktur
in unterschiedlichen Größen und Morphologien herstellen. Neben kleinen
sphärischen Partikeln können durch geeignete Wahl der genannten Parameter sowohl
entlang der c-Achse elongierte Stäbchen gewonnen werden, als auch in c-Richtung
gestauchte, plättchenförmige Partikel, die sich entlang dieser Achse in langen Ketten
anordnen

Eine kritische Einordnung des parlamentarischen Diskurses um Rassismus im Vergleich der Jahre 2011/12 und 2020
Nach dem rassistischen Anschlag in Hanau am 19.02.2020 und dem Mord an George Floyd am 25.05.2020 wurde oftmals von einem Wandel im gesellschaftlichen und medialen Diskurs um Rassismus gesprochen. Ob sich dieser Wandel auch im parlamentarischen Diskurs des deutschen Bundestags widerspiegelt, untersucht dieses Working Paper. Es widmet sich der bislang kaum beforschten Frage, wie Rassismus in parlamentarischen Debatten thematisiert wird und inwiefern Veränderungen im Sprechen über Rassismus im Bundestag zu beobachten sind. In einer vergleichenden Diskursanalyse von Plenarprotokollen der Jahre 2011/12 (Debatten nach der NSU-Selbstenttarnung) und 2020 werden Kontinuitäten, aber auch Diskontinuitäten im Diskurs über Rassismus herausgearbeitet.
Während sich einerseits Veränderungen dahingehend zeigen, dass Rassismus im Jahr 2020 deutlich häufiger explizit angesprochen wird und zunehmend rassismuskritische Positionen im Bundestag vertreten werden, konnten vor dem Hintergrund rassismuskritischer Theorie aber auch zahlreiche Strategien der Dethematisierung von Rassismus identifiziert werden, die sowohl 2011/12 als auch 2020 den Diskurs prägen
