Imaging of Tau and Microtubules to Study Mechanisms of Tau Pathologies and Neurodegeneration
The nervous system undergoes constant remodeling and adapting of its structural organization to fulfill its function, the propagation and processing of information. The structural backbone of neurons are arrays of microtubules (MTs) maintaining their specialized morphologies and serving as railway system for the transport of cargoes and information. The functional regulation of this complex system is based on many different factors, while microtubule-associated proteins (MAPs) like tau being one of them. The tau protein is of particular interest as it plays a central role in numerous neurodegenerative diseases, called tauopathies, where Alzheimer’s Disease (AD) is the most common and prominent example. Hallmark of tauopathies such as AD is the aggregation and increased phosphorylation of tau, while also many other post-translational modifications (PTMs) of the protein are described and linked to neurodegeneration.
Scope of this thesis are investigations on how the neuronal MTs, MT-dynamics and MT-dependent transport are affected by post-translational changes of the tau protein. A new type of toxic gain of function of tau is described, as it impairs axonal transport when cleaved at a diseases relevant site and microtubule-targeting drugs are introduced as pharmacological modifiers of MAP-microtubule interaction. The effect of MT-stabilization by Epothilone D on the MT-cytoskeleton as well as the effect of a tau knockout is further investigated by cutting-edge microscopy (DNA-PAINT SMLM) and algorithm-based quantification of the MT array. Changes of the MT organization and structure on the level of individual MTs upon stabilization and tau knockout are described. Accumulating evidence suggests that pathologically modified monomeric and soluble oligomeric forms of tau should be considered as harmful tau species. Hence, a cell-based model allowing investigations of disease-like modified tau, its oligomerization, and MT-interaction is implemented enabling the screening for modulators of early-stage aggregation and phosphorylation of tau. Leitkultur
Der folgende Beitrag setzt sich mit der Geschichte, dem Gebrauch und der Kritik am Begriff Leitkultur auseinander und wird die sprachlichen wie sozialen Referenzen der Leitkulturdebatten sichtbar machen. Der Beitrag wird auch zeigen, welche politische Leerstelle der Begriff besetzte, an welchen Punkten sich die Für- und Widerreden unterscheiden und überschneiden. Seit der Leitkulturdebatte, die Thomas de Maizière 2017 mit zwei zuspitzenden Aussagen auslöste – „Wir geben uns zur Begrüßung die Hand.“ und „Wir sind nicht Burka.“ –, konstituiert sich die Verwendung des Begriffs in einer identitätspolitisch kaum mehr vermittelbaren polarisierten Konstellation. Sie schwankt zwischen der Negation des Nicht-Deutschen – exemplarisch dafür stehen Aussagen wie „Der Islam gehört nicht zu Deutschland“ (AfD 2021) – und der Negation des Deutschen – zum Beispiel in Aussagen wie „Wir brauchen eine jüdisch-muslimische Leitkultur“ (Czollek 2020). Beide Positionen hatte der Begriff in der Geschichte seiner Verwendung bislang nicht eingenommen. Es war der syrischstämmige Politikwissenschaftler Bassam Tibi, der den Begriff mit seinem Buch Europa ohne Identität. Leitkultur oder Wertebeliebigkeit 1998 als erster ins Spiel brachte. Er setzt ihn darin ein gegen Begriffe wie Volk, gegen Redeweisen von ‚unseren Gepflogenheiten‘ oder gegen jede Rede von einem Vorrang einer Ethnie oder Religion. Tibi unternimmt den Versuch, mit dem Begriff Leitkultur im Gegenteil eine demokratische zivilisatorisch-politisch europäische Identität zu beschwören (Tibi 1998: 83). Allein die unterschiedlichen Verwendungsweisen des Begriffs von Tibi über de Maizière bis zur AfD und Czollek zeigen, welch vielschichtigen Weg dieser Begriff zurückgelegt hat
Health science research approaches and practical approaches in the prevention of occupational dermatoses
Der Schutz, die Erhaltung sowie die Förderung der Gesundheit der Bevölkerung sind zentrale Public-Health-Anliegen, die angesichts aktueller Entwicklungen nicht nur in Deutschland zunehmend an Bedeutung gewinnen. Die Gesundheitswissenschaften, die sich durch Inter- und Transdisziplinarität, Politikrelevanz sowie einen Bevölkerungs- und Systembezug auszeichnen, sind in besonderer Weise geeignet, um in verschiedenen Anwendungsfeldern – z. B. in der Berufsdermatologie – relevante Frage- und Problemstellungen mit empirischen Forschungsmethoden zu adressieren. Die Prävention beruflich bedingter Hauterkrankungen ist angesichts der großen Zahl der Verdachtsanzeigen, der Anerkennungsraten, der hohen Versorgungskosten sowie letztlich aufgrund des gesetzlichen Versorgungsauftrages ein legitimes Anwendungsfeld für die gesundheitswissenschaftliche Forschung und Praxis. Vor diesem Hintergrund sind die Ziele der vorliegenden kumulativen Dissertation, anhand von vier Untersuchungen spezifische und bis dato offene Forschungsfragen innerhalb des Anwendungsfeldes ‚Prävention beruflich bedingter Hauterkrankungen‘ zu adressieren sowie auf diese Weise aufzuzeigen, wie durch gesundheitswissenschaftliche Forschung Ansätze zur Lösung praxisrelevanter Probleme entwickelt werden können. Hierzu werden in der vorliegenden Arbeit Merkmale und Strukturen gesundheitswissenschaftlicher Forschungsprozesse herausgearbeitet. Diese Ausführungen bilden den Rahmen zur Verortung der Originalarbeiten im wissenschaftlichen Kontext. Die vier Untersuchungen adressieren nicht nur spezifische Forschungsfragen innerhalb des Anwendungsgebietes, sondern stellen außerdem die Pluralität der forschungsmethodischen Herangehensweisen heraus, die sich aus den vielfältigen Bezugsdisziplinen der Gesundheitswissenschaften ergeben. Auf diese Weise zeigen die Untersuchungen auf, inwiefern den Merkmalen Inter- und Transdisziplinarität Rechnung getragen werden kann, wenngleich sich dabei Optimierungspotenzial hinsichtlich der disziplinübergreifenden Zusammenarbeit andeutet. Darüber hinaus verdeutlichen die Untersuchungen, inwiefern Probleme aus der Praxis aufgegriffen und Forschungsergebnisse in die Praxis transferiert werden können. Um langfristig die Gesundheit der Bevölkerung zu schützen, zu erhalten und zu fördern, ist es die Aufgabe der Gesundheitswissenschaften, durch die regelgeleitete und qualitätsgesicherte Durchführung empirischer Forschungsvorhaben unter Einbeziehung von Akteuren der Praxis eine wissenschaftliche Grundlage für Public Health zu schaffen. Hierfür ist eine strukturelle (z. B. finanzielle und institutionelle) Stärkung der Gesundheitswissenschaften essenziell. Darüber hinaus sollten Inter- und Transdisziplinarität weiter gefördert werden. Nicht zuletzt ist eine Intensivierung des Austausches zwischen Akteuren der Wissenschaft und Entscheidungsträgern aus Praxis und Politik sowie gleichermaßen zwischen den genannten Akteuren und der Bevölkerung anzustreben
The Expansion of the Shanghai Cooperation Organization SCO
The Shanghai Cooperation Organization SCO with headquarter in Beijing was founded in 2001 as successor of the Shanghai Five group from 1996. The SCO stepwise expanded by new member states and in 2022/23, Saudi Arabia, Qatar and Egypt got the status of dialogue partners; United Arab Emirates, Bahrain, and Kuwait may follow soon. The SCO is an inter-governmental organization based on the principle of non-interference in sovereign states’ internal affairs formulated as ‚Shanghai Spirit’. The SCO has an institutional structure with a Secretariat, a Regional Anti-Terrorism Structure (RATS, Tashkent), regular meetings, multi-national military exercises (Peace Mission) and it supports economic cooperation between member states. The SCO is an alternative global governance model to the alliance model of the West (NATO, EU, Quad) and covers now most of the Eurasian landmass and approximately 40% of the world population, but it is not a military alliance and does not aim to integrate the member states into supranational frameworks. The initial focus on security later shifted to economic issues and now also to energy policy. As various member states have their own geopolitical agenda (e.g., China, Russia, India, Iran and so on), Western analysts sometimes doubt the relevance of the SCO, but it allows its member states a diversification and balancing of their security-related, economic, and diplomatic activities, by this getting more political freedom of action. The SCO activities are well aligned with Chinas ‘circle of friends’ and partnership concept of foreign policy and many SCO states have a bilateral partnership with China, e.g., Saudi-Arabia and Iran. Based on this, the co-incidence of the SCO expansion and the trilateral agreement from 2023 between China, Saudi-Arabia, and Iran to restore the Saudi-Iranian relations is not surprising. The United Arab Emirates also have good relations to China and are now restoring their relation to Iran as well. In summary, the current SCO expansion is a further step to a multipolar world order and is challenging the security architecture of the United States for the MENA region
Location factor night-time economy - Promotion and development of the night-time economy as a task of municipal economic development agencies?
Ziel dieser Dissertation ist es, Möglichkeiten zur Förderung und Entwicklung der Nachtökonomie durch die kommunale Wirtschaftsförderung zu erörtern und einen Beitrag zum besseren Verständnis des institutionellen Umfelds der Nachtökonomie zu leisten. Um dieses Ziel zu erreichen, wird eine institutionentheoretische Perspektive eingenommen und die Forschungsstränge der Standortfaktorenforschung, kommunalen Wirtschaftsförderung und nächtlichen Ökonomien werden miteinander verknüpft. Hierdurch können die Beteiligten, deren Austausch- und Interaktionsformen sowie den vorherrschenden Rahmenbedingungen im institutionellen Umfeld der Nachtökonomie analysiert werden. Diese Erkenntnisse wurden für die Entwicklung von Handlungsansätzen zur Förderung der Nachtökonomie durch die kommunale Wirtschaftsförderung berücksichtigt.
Die Ergebnisse zeigen, dass das institutionelle Umfeld der Nachtökonomie sehr komplex sowie kontext- bzw. standortabhängig ist. Eine Vielzahl privatwirtschaftlicher, städtischer und weiterer Stakeholder prägt mit ihren teils gegensätzlichen Zielvorstellungen das institutionelle Umfeld der Nachtökonomie. Je nach Standort unterscheiden sich die Anzahl der Beteiligten, ihre Austauschbeziehungen und Vernetzungsformen. Diese drücken sich in den formellen bzw. formalisierten sowie insbesondere den informellen Institutionen aus. Letzteren kommt für den Wissensaustausch im Kontext der Nachtökonomie eine besondere Bedeutung zu. Unterschiedliche Ausprägungen der Embeddedness und institutionellen Dichte konnten in den Städten der Fallstudie identifiziert werden.
Die kommunale Wirtschaftsförderung wird als eine von vielen Beteiligten im institutionellen Umfeld der Nachtökonomie ausgemacht. Je nach Ausstattung und Ressourcen sowie Standortgegebenheiten ist diese unterschiedlich stark in das institutionelle Umfeld der Nachtökonomie eingebettet. Deutlich wird zudem die Diskrepanz zwischen der Bewertung der Bedeutung der Nachtökonomie und der Verankerung des Themas als Aufgabenfeld in den Wirtschaftsförderungen. So sehen 97 % der befragten Wirtschaftsförderungen die Bedeutung der Nachtökonomie für die Standortwahl von Fachkräften, aber nur 1/3 der Wirtschaftsförderungen hat die Nachtökonomie als eines ihrer Arbeitsthemen verankert. Dabei zeigen sich starke Anknüpfungspunkte an die bestehenden Aufgabenfelder der kommunalen Wirtschaftsförderung.
Aus den Erkenntnissen zum institutionellen Umfeld der Nachtökonomie und zur kommunalen Wirtschaftsförderung wurde ein integrierter Ansatz zur Förderung der Nachtökonomie entwickelt. Dieser bezieht die relevanten Stakeholder des institutionellen Umfelds ein und bietet der kommunalen Wirtschaftsförderung Raum zur Förderung der Nachtökonomie. Aus einer institutionentheoretischen Perspektive kommt der kommunalen Wirtschaftsförderung in diesem Kontext die Rolle des Institutional Entrepreneurs zu. Durch ihre Kompetenzen und die Einbringung zusätzlicher Ressourcen kann sie die Rahmenbedingungen für den institutionellen Wandel hin zu effizienteren Institutionen ermöglichen.
Für den theoretischen Diskurs zur Nachtökonomie leisten die Ergebnisse einerseits einen wichtigen Beitrag zum besseren Verständnis des institutionellen Umfelds. Andererseits wurde mit der Forschungsarbeit der Nachtökonomie-Diskurs um eine institutionentheoretische Perspektive weiterentwickelt und der Diskurs zu den Aufgabenfeldern der kommunalen Wirtschaftsförderung um das Thema Nachtökonomie erweitert
Power Consumption Modeling and Estimation for Software Applications in Model-Driven Development of Embedded Systems
Nowadays, embedded systems are ubiquitous and inherent in almost all areas of life. In recent years, trends like the Internet of Things (IoT) have been a primary driver for the growing embedded systems market. Many of those IoT devices are battery-powered and more resource-constrained. In addition to economic constraints like total costs and short time-to-market, technical constraints lead to multiple challenges in embedded software development. For battery-powered systems, electrical energy is one of the most critical constraints. For instance, uncontrolled power consumption or an exhausted energy source caused by software applications may lead to failure and costly damage to the device or the environment. Developers often lack knowledge and suitable design concepts to specify, implement, and evaluate energy-efficient software applications. Additionally, constantly changing technologies, extensive functionalities, and various requirements further increase the complexity of embedded software applications while making their development a critical and complex task. To manage the complexity of software applications and the development process, methodologies such as Model-driven Development (MDD) have gained importance. However, power-related non-functional aspects are insufficiently considered in MDD. This thesis addresses the aforementioned gaps and presents a novel framework for energy-aware software design patterns. Developers and engineers may use the framework to specify and describe design patterns addressing power-related issues of software applications. The introduced design pattern template provides a set of metrics to describe possible energy savings and the effort-saving ratio when applying a design pattern. The template also contains a unified graphical representation to visualize the effects of design patterns. In addition, a first catalog of energy-aware design patterns is provided, which may be used to design software applications in MDD and traditional development. To further enhance the development of energy-efficient software applications in MDD, this thesis also introduces a novel power consumption estimation approach for models based on the Unified Modeling Language (UML). The approach is specifically designed for early development stages when optimizations are most effective. A concept for hardware component models is presented, which can be integrated into the software application model. With the provided UML profile, aspects related to power and timing can be modeled. In addition, methods for indirect and direct power analysis are introduced. While the indirect power analysis is based on simulated hardware behavior, the direct power analysis relies on a real hardware platform and a measuring device. Along with the novel and formal description of energy bugs, software applications can be evaluated and energy-related issues detected. A real-world example of an IoT sensor node and a proof-of-concept implementation of the power consumption estimation approach illustrate the application of proposed modeling and estimation concepts. Moreover, the detection of energy bugs is demonstrated, and the accuracy of the analysis methods is compared. Additionally, the overall performance of the direct power analysis is investigated in depth. The results have shown that the concepts and approaches are suitable for analyzing and predicting the power consumption of software applications in early development phases. Furthermore, the process can be integrated into existing development workflows to support developers using MDD to design energy-efficient software applications

Cultural Attachment Theory - How children form relationships in three cultural contexts in Costa Rica
Attachment theory originates in a unique historical and cultural context, yet its assumptions are considered universal. A Euro-American middle-class pattern of caregiving and early relationship formation has quickly been regarded the single pathway for optimal development and the only conceptual lens to study relationships worldwide, increasingly being applied in non-Western contexts and to cultural minorities in Western immigration states. The purpose of this study is to revise and expand the monocultural assumptions of attachment theory by examining early attachment development in three cultural contexts in Costa Rica: Urban middle-class San José, rural Guanacaste, and rural indigenous Bribri Talamanca. Using an ethnographic and culture-conscious approach, semi-structured caregiver interviews, ethnographic observations, and recording of videos and photographs were conducted with 30 families per sample with a child between 6 and 28 months of age. Results revealed profound differences in the size of caregiver and attachment networks, the roles and conceptions of caregivers and attachment figures, type and context of caregiver-infant interactions, and the ways and modes through which children form attachments. While the San José sample resembled Western middle-class families in their caregiving networks and beliefs, children in Guanacaste and Bribri Talamanca showed larger networks, however differently composed. In both rural samples, the mother did not necessarily function as the primary caregiver, and children learned to form close relationships with multiple caregivers and the extended family from an early age. Further, there is evidence that not only sensitive caregiving is the main entry to attachment, but primary care, such as feeding, can also serve as an important attachment mechanism. These findings suggest that there is more variety in attachment patterns than attachment theory implies and promotes. It is argued that attachment has not only the function of providing security, but also serves as a platform for cultural learning and is therefore culturally specific in nature. The study highlights the need for culturally conscious attachment research and a shift in practice toward more inclusive and less normative approaches to cultural differences in attachment development and caregiving
A closer look at the sources of variability in scalar implicature derivation: a review
For more than 20 years, studies in experimental pragmatics have provided invaluable insights into the cognitive processes involved in deriving scalar implicatures and achieving inferential comprehension. However, the reports have always contained a notable degree of variability that remained inadequately discussed in the literature. For instance, upon closer inspection of the experimental record, one can always find a group of individuals who tend to be largely pragmatic, overwhelmingly logical, or sometimes mixed not showing a clear preference. There also exist newly-devised paradigms that prompt a radically different type of response than other paradigms, and thus new evidence casting doubt on long-established findings in the field. More recent research on scalar diversity further suggests that differences in the semantic structure of scalar words can lead to differing rates of scalar implicatures and can modulate the time invested in pragmatic processing. Indeed, one can contend that the current empirical landscape on scalar implicatures can be characterized as having three primary sources of variability: inter-individual, methodological, and linguistic. What factor or factors are behind these patterns of variability, and how can we interpret them in light of a pragmatic theory? This paper has a 2-fold objective: one is to review the previous experimental record on scalar implicatures from variability-based lenses, and the other to discuss the factor(s) that could account for this observed variability in the literature. Skin picking: A problem among healthcare workers?
Skin picking (SP) with manipulation of the skin using nails or other tools leads to various degrees of skin damage. Clinical signs of SP on the hands include erythema, erosions and bleeding wounds, crusts, hyperkeratosis, and often loss of the cuticles.
