Evaluation of monoamine oxidase B fluctuation in liver fibrosis cell and mice models via a specificity fluorescent probe
Liver fibrosis can result in cirrhosis and cancer, leading to damage to liver function and even endangering life. Early diagnosis of fibrosis is essential for optimizing the therapeutic schedule and improving the cure rates. Recent researches have shown that monoamine oxidase B (MAO-B) could be used as the marker for the early diacrisis of liver fibrosis. However, there still lacked strategies for accurate in situ monitoring of MAO-B. In this work, we developed a fluorescence probe YXHcy-NH2 for detecting MAO-B activity in living cells and in vivo. MAO-B activates the probe within 100 minutes and emits bright fluorescence at the fluorescence emission peak. The results displayed that the fluorescence intensity of fibrotic LX-2 cells was ten times that of normal cells, and the fluorescence brightness of liver tissues in liver fibrotic mice was five times higher than that of normal mice, so our probe could successfully distinguish between fibrotic cells and normal cells, as well as liver fibrotic mice and normal mice. Our study provides an easy, rapid, and high sensitive method for in situ in vivo imaging of MAO-B. It may become an effective mean for early diagnosis of liver fibrosis
Long-term variations and trends of tropospheric and ground-level NO 2 over typical coastal areas
Tropospheric and ground-level levels of nitrogen dioxide (NO 2 ) may have changed due to environmental policies and technological changes. Here, multiyear ground-based measurement (2014 - 2022) and satellite-derived retrieval (October 2004 to May 2023) datasets of NO 2 concentrations are used to study the long-term groundlevel and tropospheric NO 2 patterns and trends at four time scales (interannual, quarterly, monthly, and hourly) over the three regions with the highest global NO 2 concentrations. Several tropospheric NO 2 hotspots are observed over industrialized and highly populated coastal zone areas. Over the past 18 years, these hotpots have generally decreased due to the implementation of strong and effective environmental protection policies. A significant trend reversal was observed in 2011 in East-central China. The maximum values of tropospheric NO 2 occurred in winter over East-central China, Western Europe and East-central USA, with concentrations of 13.010, 4.243 and 4.149 x 10 15 molecules cm -2 , respectively. The Bohai Sea exhibited disproportionately high tropospheric NO 2 levels, suggesting it as a significant contributor to regional nitrogen pollution. Ground-level NO2 shows similar temporal patterns to tropospheric concentrations, with over 82.8 % of sites in East-central China exceeding 30 mu g m - 3 , compared to 12.8 % in Western Europe and 3.8 % in East-central USA. The analysis of diurnal variations in ground-level NO 2 concentrations shows a significantly decrease during the daytime compared to nighttime, reflecting that the peak occurs at approximately 08:00 and 20:00 LT, while the lowest concentrations at around 12:00 LT. These insights clarify the impacts of environmental policies on NO 2 levels and identify critical areas for targeted pollution control, enhancing our understanding of NO 2 dynamics in highpollution regions globally
Realistic Nanoplastics Induced Pulmonary Damage via the Crosstalk of Ferritinophagy and Mitochondrial Dysfunction
The smaller size fraction of plastics may be more substantially existing and detrimental than larger-sized particles. However, reports on nanoplastics (NPs), especially their airborne occurrences and potential health hazards to the respiratory system, are scarce. Previous studies limit the understanding of their real respiratory effects, since sphere-type polystyrene (PS) nanoparticles differ from NPs occurring in nature with respect to their physicochemical properties. Here, we employ a mechanical breakdown method, producing NPs directly from bulk plastic, preserving NP properties in nature. We report that among four relatively high abundance NP materials PS, polyethylene terephthalate (PET), polyvinyl chloride (PVC), and polyethylene (PE) with a size of 100 nm, PVC induced slightly more severe lung toxicity profiles compared to the other plastics. The lung cytotoxicity of NPs is higher than that of commercial PS NPs and comparable to natural particles silicon dioxide (SiO2) and anatase titanium dioxide (TiO2). Mechanistically, BH3-interacting domain death agonist (Bid) transactivation-mediated mitochondrial dysfunction and nuclear receptor coactivator 4 (NCOA4)-mediated ferritinophagy or ferroptosis are likely common mechanisms of NPs regardless of their chemical composition. Insights into the discovery and intervention of metalloproteinase in marine hazardous jellyfish
The proliferation of toxic organisms caused by changes in the marine environment, coupled with the rising human activities along the coastal lines, has resulted in an increasing number of stinging incidents, posing a serious threat to public health. Here, we evaluated the systemic toxicity of the venom in jellyfish Chrysaora quinquecirrha at both cellular and animal levels, and found that jellyfish tentacle extract (TE) has strong lethality accompanied by abnormal elevation of blood biochemical indicators and pathological changes. Joint analysis of transcriptome and proteome indicated that metalloproteinases are the predominant toxins in jellyfish. Specially, two key metalloproteinases DN6695_c0_g3 and DN8184_c0_g7 were identified by mass spectrometry of the red blood cell membrane and tetracycline hydrochloride (Tch) inhibition models. Structurally, molecular docking and kinetic analysis are employed and observed that Tch could inhibit the enzyme activity by binding to the hydrophobic pocket of the catalytic center. In this study, we demonstrated that Tch impedes the metalloproteinase activity thereby reducing the lethal effect of jellyfish, which suggests a potential strategy for combating the health threat of marine toxic jellyfish
Rapidly reducing cadmium from contaminated farmland soil by novel magnetic recyclable Fe3O4/mercapto-functionalized attapulgite beads
Reducing cadmium (Cd) content from contaminated farmland soils remains a major challenge due to the difficulty in separating commonly used adsorbents from soils. This study synthesized novel millimeter-sized magnetic Fe3O4/mercapto-functionalized attapulgite beads (MFBs) through a facile one-step gelation process incorporating alginate. The MFBs inherit the environmental stability of alginate and enhance its mechanical strength by hybridizing Fe3O4 and clay mineral components. MFBs can be easily separated from flooded soils by magnets. When applied to 12 Cd-polluted paddy soils and 14 Cd-polluted upland soils, MFBs achieved Cd(II) removal rates ranging from 16.9% to 62.2% and 9.8%-54.6%, respectively, within a 12-h period. The MFBs predominantly targeted the exchangeable and acid soluble, and reducible fractions of Cd, with significantly enhanced removal efficiencies in paddy soils compared to upland soils. Notably, MFBs exhibited superior adsorption performance in soils with lower pH and organic matter (OM) content, where the bioavailability and mobility of Cd are heightened. The reduction of Cd content by MFBs is a sustainable and safe method, as it permanently removes the bioavailable Cd from soil, rather than temporarily reducing its bioavailability. The functional groups such as -SH, -OH, present in attapulgite and alginate of MFBs, played a crucial role in Cd(II) adsorption. Additionally, attapulgite and zeolite provided a porous matrix structure that further enhanced Cd(II) adsorption. The results of X-ray photoelectron spectroscopy suggested that both chemical precipitation and surface complexation contributed to Cd(II) removal. The MFBs maintained 87.6% Cd removal efficiency after 5 regeneration cycles. The surface of the MFBs exposed new adsorption sites and increased the specific surface area during multiple cycles with Cd-contaminated soil. This suggests that MFBs treatment with magnetic retrieval is a potentially effective pathway for the rapid removal of Cd from contaminated farmland soils
Global prevalence of microplastics in tap water systems: Abundance, characteristics, drivers and knowledge gaps
Tap water is a main route for human direct exposure to microplastics (MPs). This study recompiled baseline data from 34 countries to assess the current status and drivers of MP contamination in global tap water systems (TWS). It was shown that MPs were detected in 87 % of 1148 samples, suggesting the widespread occurrence of MPs in TWS. The detected concentrations of MPs spanned seven orders of magnitude and followed the linearized log-normal distribution (MSE = 0.035, R2 = 0.965), with cumulative concentrations at 5th, 50th and 95th percentiles of 0.028, 4.491 and 728.105 items/L, respectively. The morphological characteristics were further investigated, indicating that particles smaller than 50 mu m dominated in global TWS, with fragment, polyester and transparent as the most common shape, composition and color of MPs, respectively. Subsequently, the SHapley Additive exPlanations (SHAP) algorithm was implemented to quantify the importance of variables affecting the MP abundance in global TWS, showing that the lower particle size limit was the most important variables. Subgroup analysis revealed that the concentration of MPs counted at the size limit of 1 mu m was >20 times higher than that above 1 mu m. Ultimately, current knowledge gaps and future research needs were elucidated
Epidermal oxysterols function as alarm substances in zebrafish
Alarm substances signal imminent predation thread and enable anti -predation strategies. In shoaling fish, alarm cues diffuse from injured skins that induce intense fear and anti -predation behaviors in other members. While these "fear substances"are shown to be present in numerous fishes and thought to exist in roughly 8,000 Ostariophysan species, their chemical nature remains largely unknown. We posited that fish alarm cues comprise small compounds and induce specific behaviors characteristic of fish exposed to skin extracts. Using the behaviors as bioassays, we tracked the alarm function of zebrafish skin extract to two compounds, 24-methyl-5 a- cholestane-3 a, 7 a, 12 a, 24,28-pentahydroxy 28 -sulfate, an oxysterol sulfate, and 5 a- cyprinol sulfate. At concentrations of less than one nanomolar, each compound induced antipredator behaviors and increased cortisol levels in zebrafish. Their mixture, at the natural ratio, replicated the skin extract in eliciting the full suite of anti -predator behavior patterns. Our findings reveal a molecular mechanism whereby fish escape predation danger
Morphological, Microstructural, and In Situ Chemical Characteristics of Siderite Produced by Iron-Reducing Bacteria
Dissimilatory iron-reducing bacteria (DIRB) oxidize organic matter or hydrogen and reduce ferric iron to form Fe(II)-bearing minerals, such as magnetite and siderite. However, compared with magnetite, which was extensively studied, the mineralization process and mechanisms of siderite remain unclear. Here, with the combination of advanced electron microscopy and synchrotron-based scanning transmission X-ray microscopy (STXM) approaches, we studied in detail the morphological, structural, and chemical features of biogenic siderite via a growth experiment with Shewanella oneidensis MR-4. Results showed that along with the growth of cells, Fe(II) ions were increasingly released into solution and reacted with CO32- to form micrometer-sized siderite minerals with spindle, rod, peanut, dumbbell, and sphere shapes. They are composed of many single-crystal siderite plates that are fanned out from the center of the particles. Additionally, STXM revealed Fh and organic molecules inside siderite. This suggests that the siderite crystals might assemble around a Fh-organic molecule core and then continue to grow radially. This study illustrates the biomineralization and assembly of siderite by a successive multistep growth process induced by DIRB, also provides evidences that the distinctive shapes and the presence of organic molecules inside might be morphological and chemical features for biogenic siderite
Molecular imprinting-based triple-emission ratiometric fluorescence sensor with aggregation-induced emission effect for visual detection of doxycycline
The development of high-performance sensors for doxycycline (DOX) detection is necessary because its residue accumulation will cause serious harm to human health and the environment. Here, a novel tri-emission ratiometric fluorescence sensor was proposed by using "post-mixing" strategy of different emissions fluorescence molecularly imprinted polymers with salicylamide as dummy template (DMIPs). BSA was chosen as assistant functional monomer, and also acted as sensitizers for the aggregation-induced emission (AIE) effect of DOX. The blue-emitting carbon dots and the red-emitting CdTe quantum dots were separately introduced into DMIPs as the response signals. Upon DOX recognition within 2 min, blue and red fluorescence of the tri-emission DMIPs sensor were quenched while green fluorescence of DOX was enhanced, resulting in a wide range of color variations observed over bluish violet-rosered-light pink-orange-yellow-green with a detection limit of 0.061 mu M. The sensor possessed highly selective recognition and was successfully applied to detect DOX in complicated real samples. Moreover, with the fluorescent color collection and data processing, the smartphone-assisted visual detection of the sensors showed satisfied sensitivity with low detection limit. This work provides great potential applications for rapid and visual detection of antibiotics in complex substrates
