    本发明公开了一种天然抗菌肽及其在抗菌方面的应用。本发明的抗菌肽来源于单环刺螠。通过检索单环刺螠基因数据库,筛选分析获得抗菌肽成熟肽序列,其氨基酸序列如序列表中SEQ ID NO : 1所示。通过对比分析,该天然抗菌肽与目前已知所有抗菌肽的氨基酸序列存在明显差异,属于一种新型抗菌肽。抗菌实验结果表明,本发明的抗菌肽对革兰氏阳性细菌、革兰氏阴性细菌和真菌具有极强的抗菌活性,抗菌作用广谱高效,可用于制备抗菌、抑制细菌生长的药物、防腐剂、兽药、动物饲料以及化妆品

    The identification of an increasing number of aryl organophosphate esters (aryl-OPEs) in environmental samples has led to growing attention recently. Due to the potential adverse effects on human health and environment, development of new analytical methods for sensitive and selective determination of aryl-OPEs in complex matrices is urgently needed. Here, a novel analytical method for the identification and determination of trace amounts of aryl-OPEs in water samples is developed by using melamine sponge@heteropore covalent organic framework (MS@HCOF) based on vortex-assisted extraction (VAE) prior to UHPLC-MS/MS analysis. The MS@HCOF was rationally designed and synthesized through an in-situ growth strategy and exhibited superior selectivity toward aryl-OPEs compared with that of MS@singlepore COF (MS@SCOF) due to steric effect. A systematic optimization was conducted on important parameters of VAE, resulting in the successful extraction of nine aryl-OPEs in just 6 min. Under optimized conditions, the limits of detection (S/N = 3) and quantification (S/ N = 10) were within the ranges of 0.001-0.027 and 0.005-0.091 ng/L for nine aryl-OPEs, respectively. The validated method was proven applicable to real water samples, i.e., the recoveries were 65.3-119.5 % for seawater, 59.4-112.9 % for effluent, and 76.0-117.4 % for tap water. Furthermore, the adsorption mechanisms were explored through density functional theory (DFT) calculations. DFT results revealed that a notable selective enrichment capacity of MS@HCOF towards aryl-OPEs stems from 7C-7C conjugation and hydrogen bonding. The established method benefits from the advantages of high selectivity and sensitivity for the ultra-trace determi-nation of aryl-OPEs

    Anti-stokes fluorescence of phycobilisome and its complex with the orange carotenoid protein

    Phycobilisomes (PBSs) are giant water-soluble light-harvesting complexes of cyanobacteria and red algae, consisting of hundreds of phycobiliproteins precisely organized to deliver the energy of absorbed light to chlorophyll chromophores of the photosynthetic electron-transport chain. Quenching the excess of excitation energy is necessary for the photoprotection of photosynthetic apparatus. In cyanobacteria, quenching of PBS excitation is provided by the Orange Carotenoid Protein (OCP), which is activated under high light conditions. In this work, we describe parameters of anti-Stokes fluorescence of cyanobacterial PBSs in quenched and unquenched states. We compare the fluorescence readout from entire phycobilisomes and their fragments. The obtained results revealed the heterogeneity of conformations of chromophores in isolated phycobiliproteins, while such heterogeneity was not observed in the entire PBS. Under excitation by low-energy quanta, we did not detect a significant uphill energy transfer from the core to the peripheral rods of PBS, while the one from the terminal emitters to the bulk allophycocyanin chromophores is highly probable. We show that this direction of energy migration does not eliminate fluorescence quenching in the complex with OCP. Thus, long-wave excitation provides new insights into the pathways of energy conversion in the phycobilisome

    Rapid antibiotic screening based on E. coli apoptosis using a potentiometric sensor array

    Phenotypic antimicrobial susceptibility testing enables reliable antibiotic screening but requires multiple strategies to identify each phenotypic change induced by different bactericidal mechanisms. Bacteria apoptosis with typical phenotypic features has never been explored for antibiotic screening. Herein, we developed an antibiotic screening method based on the measurement of antibiotic-induced phosphatidylserine (PS) exposure of apoptotic bacteria. Phosphatidylserine externalization of E. coli that can be widely used as an apoptosis marker for antibiotics with different antibacterial mechanisms was explored. A positively charged PS-binding peptide was immobilized on magnetic beads (MBs) to recognize and capture apoptotic E. coli with PS externalization. Apoptotic E. coli binding led to the charge or charge density change of MBs-peptide, resulting in a potential change on a magneto-controlled polymeric membrane potentiometric sensor. Based on the detection of apoptotic E. coli killed by antibiotics, antibiotic screening for different classes of antibiotics and silver nanoparticles was achieved within 1.5 h using a potentiometric sensor array. This approach enables sensitive, general, and timesaving antibiotic screening, and may open up a new path for antibiotic susceptibility testing

    Distribution and influencing factors of macrobenthos on three seagrass beds in the intertidal zone of Shandong province, China

    Introduction The macrobenthos plays a vital role within the ecosystem of seagrass beds, with its characteristics and spatial distribution serving as indicators of the well-being of the seagrass beds. Methods In August 2018, three seagrass beds located in the Yellow River Estuary of Dongying, the west coast of Yantai, and Swan Lake of Weihai, were investigated to compare the ecological influences of seagrass habitat on the benthic environment and macrobenthic community. Within each seagrass bed, porewater, sediment, and macrobenthos were sampled from three separate stations (center of seagrass bed, edge of seagrass bed and bare area). Results and discussion One-way ANOVA showed significant differences (p < 0.05) in environmental factors and macrobenthos species, abundance, biomass and diversity indices among the three seagrass beds. The present data did not show significant impacts on habitat and macrobenthos in the different coverage areas of seagrass beds at the investigated spatial scales, though crustacea and some carnivores were relatively more inclined to inhabit areas with higher seagrass densities. Aquaculture and eutrophication may trigger the loss of seagrass bed habitats, that affects macrobenthic biodiversity, and conservation measures are needed to protect seagrass bed habitats

    Field Determination and Spatial Distribution of Se (IV) in Coastal Seawater of China

    A portable electrochemical system based on rGO/AuNPs/Nafion was constructed for selenium (IV) field determination in coastal seawater of the North Yellow Sea, East China Sea and South China Sea. This system demonstrated a low determination limit (0.03 mu g/L), a wide linear range of concentration (0.1-30 mu g/L), exceptional reproducibility and repeatability, and excellent selectivity. Each individual, entire test could be accomplished within 8 min. In Yantai Sishili Bay, the Se (IV) concentrations ranged from 0.16-3.59 mu g/L (average 1.43 mu g/L). In Shengshan Island, the Se (IV) concentrations ranged from 0.20-5.18 mu g/L (average 2.06 mu g/L). In Zhelin Bay, the Se (IV) concentrations ranged from 0.87-4.87 mu g/L (average 3.13 mu g/L). The average concentrations of Se (IV) in surface seawater showcased an increasing trend from the north to the south of China, suggesting Zhelin Bay > Shengshan Island > Sishili Bay. The system develops a new, competitive method for water quality assessment, and contributes to the accurate study of the biogeochemical cycles of selenium

    Nitrite-dependent anaerobic methane oxidation (N-DAMO) in global aquatic environments: A review

    The vast majority of processes in the carbon and nitrogen cycles are driven by microorganisms. The nitritedependent anaerobic oxidation of methane (N-DAMO) process links carbon and nitrogen cycles, offering a novel approach for the simultaneous reduction of methane emissions and nitrite pollution. However, there is currently no comprehensive summary of the current status of the N-DAMO process in natural aquatic environments. Therefore, our study aims to fill this knowledge gap by conducting a comprehensive review of the global research trends in N-DAMO processes in various aquatic environments (excluding artificial bioreactors). Our review mainly focused on molecular identification, global study sites, and their interactions with other elemental cycling processes. Furthermore, we performed a data integration analysis to unveil the effects of key environmental factors on the abundance of N-DAMO bacteria and the rate of N-DAMO process. By combining the findings from the literature review and data integration analysis, we proposed future research perspectives on N-DAMO processes in global aquatic environments. Our overarching goal is to advance the understanding of the NDAMO process and its role in synergistically reducing carbon emissions and removing nitrogen. By doing so, we aim to make a significant contribution to the timely achievement of China's carbon peak and carbon neutrality targets
