Systematic mapping of experimental approaches to studying common mycorrhizal networks in arbuscular mycorrhiza

Common mycorrhizal networks (CMN) capture the imagination of researchers and the public alike and have played a large role in communicating about mycorrhiza in general. With many of the claims about the functional importance of CMN recently under intense scrutiny, it becomes important to assess the literature on this topic. We systematically map experimental approaches, uncovering large data gaps, with evidence from field studies and data on ecosystem processes lacking. Very few studies meet the strictest recommended criteria, limiting our understanding of CMN-mediated effects on plants and soils. A renewed research effort needs to unravel the contribution of CMNs under global environmental change.

- Common mycorrhizal networks (CMN) are a research area within mycorrhizal ecology, studying the effects of mycorrhizal fungi linking plant roots. Experimentally studying the functioning of such networks is challenging because of the presence of many different types of network links that give rise to potentially confounding effects.
- Here, we collect, collate, and describe the research evidence for common mycorrhizal networks, specifically for arbuscular mycorrhiza (AM), using a systematic mapping approach.
- We find that not all studies tested networks formed exclusively by AM fungi, but that other filamentous fungi were present and that very few articles report on experiments fulfilling the current stringent definition (demanding hyphal continuity between plant roots). Furthermore, most research is limited to controlled environmental conditions with certain plant, fungal, and resource-transfer variables measured, while microbial community responses or ecosystem processes are neglected.
- Given the broad interest in this topic, we see a need to strengthen the evidence base on “common mycorrhizal networks” in AM fungi, necessitating a renewed research effort focusing on a range of levels of mechanistic resolution (from simple to complex networks with and without hyphal continuity). Rekonstruktion kontinuierlicher Stimulusrepräsentationen im visuellen Arbeitsgedächtnis, in der Wahrnehmung und in der räumlichen Aufmerksamkeit
In recent years, there have been considerable advances in our understanding of how the human brain represents different kinds of visual stimuli. In particular, machine learning approaches have made it possible to decode or classify visual stimulus features from the multivariate neural signal, allowing for the distinction of different stimulus categories. However, many visual features are continuous in nature, meaning that they are not perceived in categories but in a dynamic, gradual way. This calls for feature-continuous analysis methods that can establish a detailed mapping between the continuous stimulus features and their corresponding representational spaces.
Here, we contributed to the study of continuous feature representations by developing new approaches for the pre-processing and analysis of neuroimaging data, with the aim to reconstruct continuous, periodic stimuli from neural responses. First, we developed feature-space smoothing (FSS), a pre-processing method to considerably increase the signal-to-noise ratio of neural data in a continuous setting. Second, we implemented a periodic support vector regression (pSVR), which allows for the reconstruction of continuous stimulus features from the combined pattern of neural responses. We successfully applied these methods in the context of two separate neuroimaging studies. In Study 1 (see Printing copy of the publication), we used fMRI to investigate the strength of working memory representations of continuous orientations in the visual areas of participants with particularly strong or weak visual imagery ability. In contrast to our initial predictions, we found no significant difference in the amount of reconstructed information between the two groups. Instead, working memory signals were highly predictive of behavioral task performance, in both groups. We conclude that working memory representations in early visual cortex do not necessarily rely on vivid visual images. In Study 2 (currently in preparation for publication), we used EEG to investigate the neural representation of a continuous motion stimulus in two distinct frequency bands, ERP- and alpha-activity. We could successfully reconstruct direction-specific information from the sustained EEG signal. However, only the ERP activity systematically reflected the stimulus information throughout the entire trial, while informative alpha-activity was restricted to the response period of the trial. We conclude that the ERP activity encodes a perceptual signal, while the alpha-band reflects the locus of spatial attention, consistent with previous literature.
Together, these results show that our novel analysis techniques are a valuable contribution to the research of neural representations in a feature-continuous setting, using different kinds of neuroimaging data (fMRI, EEG) and cognitive processes (working memory, perception, spatial attention). Thus, we introduce FSS and pSVR as useful additions to the neuroscientific toolkit.In den letzten Jahren wurden erhebliche Fortschritte bei der Frage erzielt, wie visuelle Stimuli im menschlichen Gehirn repräsentiert werden. Insbesondere durch die Entwicklung neuartiger Machine Learning Verfahren ist es nun möglich, unterschiedliche Stimulus-Kategorien aus multivariaten neuronalen Signalen zu dekodieren. Viele visuelle Merkmale werden jedoch im Gehirn nicht als Kategorien wahrgenommen, sondern graduell. Dies erfordert Analysemethoden, die einen kontinuierlichen Zusammenhang zwischen diesen Stimuli und den entsprechenden neuralen Repräsentationen herstellen können.
Ziel dieser Arbeit ist es, einen Beitrag zur Erforschung kontinuierlicher Stimulusrepräsentationen zu leisten. Dafür haben wir neue Methoden für die Verarbeitung und Analyse von Neuroimaging-Daten entwickelt, mit dem Ziel, kontinuierliche, periodische Reize aus neuronaler Aktivität zu rekonstruieren. Dies umfasst Feature-Space Smoothing (FSS), eine Methode zur Verbesserung des Signal-Rausch-Verhältnis kontinuierlicher neuronaler Daten, und eine periodische Support-Vektor-Regression (pSVR) für die Rekonstruktion kontinuierlicher Stimulusmerkmale aus multivariaten neuronalen Signalen. Diese Methoden wurden in zwei separaten Neuroimaging-Studien angewendet. In Studie 1 (siehe Printing copy of the publication) haben wir mittels fMRT die während einer Kurzzeitgedächtnis-Aufgabe in visuellen Gehirnarealen gespeicherten Repräsentationen von kontinuierlichen Orientierungs-stimuli, spezifisch bei Studienteilnehmern mit sehr stark oder schwach ausgeprägter bildlicher Vorstellungskraft, untersucht. Entgegen unseren Annahmen fanden wir keinen signifikanten Gruppenunterschied in der rekonstruierbaren Information. Stattdessen korrelierten die Signale in beiden Gruppen stark mit den Verhaltensdaten. Daraus schließen wir, dass im Kurzzeitgedächtnis behaltene Repräsentationen in frühen visuellen Arealen nicht zwingend auf bildliche Vorstellungskraft angewiesen sind. In Studie 2 (in Vorbereitung) haben wir neuronale Repräsentation eines kontinuierlichen Bewegungsstimulus in zwei EEG-Frequenzbändern (ERP und Alpha-Band) untersucht. Wir konnten erfolgreich richtungsspezifische Informationen aus dem EEG-Signal rekonstruieren. Allerdings spiegelte nur die ERP-Aktivität die Stimulusinformation über den kompletten Versuchszeitraum wider, während sich das informative Alpha-Signal auf den Zeitraum um die Verhaltensantwort beschränkte. Wir schließen daraus, dass die ERP-Aktivität ein Wahrnehmungssignal kodiert, während das Alpha-Band den Lokus der räumlichen Aufmerksamkeit widerspiegelt.
Zusammengefasst zeigen die Ergebnisse, dass unsere neuen Analysetechniken gut geeignet sind, um neuronale Repräsentationen kontinuierlicher Merkmale zu untersuchen. Dies gilt für verschiedene Datenquellen (fMRI, EEG) und kognitive Prozesse (Kurzzeitgedächtnis, Wahrnehmung, räumliche Aufmerksamkeit). FSS und pSVR sind daher nützliche methodische Ergänzungen für die Analyse neurowissenschaftlicher Datensätze
Verbesserung der motorischen Funktion von Patient:innen nach operativer Entfernung eines Hirntumors mittels repetitiver transkranieller Magnetstimulation
Healthy motor function depends on the delicate interplay between both hemispheres of
the brain. Resection of a brain tumor can disrupt this communication by introducing a
lesion to the motor network of one hemisphere. This alteration of interhemispheric communication has been associated with motor deficits even in the absence of direct structural
damage to the core motor system.
Repetitive transcranial magnetic stimulation (rTMS) is a non-invasive tool that has been
shown to modulate activity of stimulated brain areas as well as connected networks. This
hemisphere-specific modulation has been used to improve recovery of motor deficits in
stroke patients, but limited evidence is available for patients with brain tumors.
Therefore, the aim of this thesis was to develop an rTMS-based treatment protocol for
rehabilitation of postoperative motor deficits in brain tumor patients. This goal was addressed
in three studies:
Study 1 recruited 20 healthy subjects to assess the efficacy of low-frequency rTMS (5 15-
minute sessions, 1Hz) on electrophysiological outcomes and motor function compared to
a control group. We observed changes in outcomes in a hemisphere-specific way. Further,
stimulation effects were also visible in behavioral outcomes, thus suggesting that
rTMS can be used to modulate motor function in patients. Since observed effect sizes
were rather small, longer stimulation protocols might be needed to induce clinically meaningful
changes.
In a randomized clinical trial (Study 2), we applied the same rTMS intervention for 7 sessions
(1Hz, 15 minutes) in 30 patients with motor deficits after brain tumor resection as
adjuvant treatment during the immediate postoperative period. No group differences were
found for the primary endpoint, the Fugl Meyer Score 3 months postoperatively. However,
rTMS improved motor function in the stimulated hand muscles one month postoperatively
compared to the control group. Further, a subgroup of patients with a motor eloquent
ischemia also showed improvements in global neurological function. In contrast, generalization
of effects to other muscle groups and functional impairments was limited, thus
suggesting a high specificity of induced effects and the need for patient stratification.
Finally, Study 3 evaluated the safety of a twice-daily, low-frequency rTMS protocol (14
sessions, 1 Hz, 30 minutes) to address the low effect sizes observed in the previous
studies. This intensified rTMS did not induce more side effects than standard once-daily rTMS. Therefore, intensified rTMS is a promising candidate to increase the stimulation
dose within time-limited clinical settings.
Taken together, these results show the benefit of rTMS to facilitate recovery of motor
function in brain tumor patients postoperatively. They further point to the need for adequate
patient stratification and personalization of stimulation protocols.Eine gesunde Motorik hängt entscheidend vom Zusammenspiel der beiden Hirnhälften
ab. Die Resektion eines Hirntumors kann diese Kommunikation durch Erzeugung einer
Läsion im motorischen Netzwerk einer Hemisphäre stören. Diese Veränderung der interhemisphärischen Kommunikation wurde mit motorischen Defiziten in Verbindung gebracht,
selbst wenn keine direkte strukturelle Schädigung des motorischen Kernsystems
vorliegt.
Die repetitive transkranielle Magnetstimulation (rTMS) ist eine nicht-invasive Methode,
mit der nachweislich die Aktivität der stimulierten Hirnareale sowie der damit verbundenen
Netzwerke moduliert werden kann. Ziel dieser Arbeit war daher die Entwicklung eines
rTMS-basierten Behandlungsprotokolls für die Rehabilitation von postoperativen motorischen
Defiziten bei Hirntumorpatienten. Dieses Ziel wurde in drei Studien verfolgt:
In Studie 1 wurden 20 gesunde Probanden rekrutiert, um Effekte der niederfrequenten
rTMS (5 Sitzungen, 1 Hz, 15 Minuten) auf elektrophysiologische und motorische Zielparameter
im Vergleich zu einer Kontrollgruppe zu untersuchen. Wir beobachteten hemisphären-
spezifische Veränderungen der Parameter nach rTMS, welche sich auch auf
Verhaltensoutcomes erstreckten. Dies deutet darauf hin, dass rTMS zur Modulation der
motorischen Funktion bei Patienten eingesetzt werden kann, wobei längere Stimulationsprotokolle erforderlich sein könnten, um klinisch bedeutsame Veränderungen zu bewirken.
In einer randomisierten klinischen Studie (Studie 2) wendeten wir dieselbe rTMS-Behandlung
über 7 Sitzungen (1 Hz, 15 Minuten) bei 30 Patienten mit motorischen Defiziten nach
einer Hirntumorresektion unmittelbar postoperativ an. Für den primären Endpunkt, den
Fugl Meyer Wert 3 Monate postoperativ, waren keine Gruppenunterschiede nachweisbar.
Allerdings verbesserte die rTMS die motorische Funktion der stimulierten Handmuskeln
einen Monat postoperativ im Vergleich zur Kontrollgruppe. Darüber hinaus zeigte eine
Untergruppe von Patienten mit motorisch eloquenten Ischämien Verbesserungen der globalen
neurologischen Funktion. Die hohe Spezifität der induzierten Effekte bezüglich der
stimulierten Muskeln deutet zudem die Notwendigkeit einer adäquaten Patientenstratifizierung
an.
In Studie 3 wurde die Sicherheit eines zweimal täglich durchgeführten rTMS-Protokolls
(14 Sitzungen, 1 Hz, 30 Minuten) untersucht, um die in den vorangegangenen Studien
beobachteten geringen Effektgrößen zu adressieren. Diese intensivierte rTMS verursachte
nicht mehr Nebenwirkungen als die standardmäßige einmal tägliche rTMS. Daher
scheint die intensivierte rTMS eine vielversprechende Option zur Erhöhung der Stimulationsdosis in zeitlich begrenzten klinischen Situationen.
Insgesamt deuten diese Ergebnisse auf den Nutzen der rTMS zur beschleunigten Rehabilitation
der motorischen Funktion bei Hirntumorpatienten nach einer Operation hin. Sie
zeigen außerdem die Notwendigkeit einer adäquaten Patientenstratifizierung und Personalisierung der Stimulationsprotokolle auf

Health-related quality of life after critical illness: construct validation of two new items for application in ambulatory care
Hintergrund: Die intensivmedizinische Behandlung hat sich in den letzten Jahren ver-bessert, weist aber noch immer Defizite auf: Zum einen sind evidenzbasierte Qualitäts-indikatoren (QI) unzureichend implementiert, zum anderen ist die gesundheitsbezogene Lebensqualität (Englisch: Health-related Quality of Life, HrQoL) bei Überlebenden ver-mindert und wird entgegen Empfehlungen nur selten erhoben. Kurze Messinstrumente könnten das ambulante Screening auf Folgeerscheinungen unterstützen und weitere Informationen zu zugrundeliegenden Beschwerden liefern, sind diesbezüglich jedoch noch nicht validiert. Ziel der Arbeit war es, eine Konstruktvalidierung von zwei Items zur subjektiven mentalen und körperlichen Gesundheit durchzuführen und so kurze Messin-strumente für die hausärztliche Versorgung zu validieren.
Methodik: Im Rahmen einer multizentrischen, stepped-wedge cluster-randomisierten kontrollierten Studie von intensivmedizinischen Patient*innen wurde der Einfluss einer telemedizinischen Visite auf die QI-Adhärenz untersucht. In einer Sekundäranalyse wur-de die HrQoL von Überlebenden analysiert. Diese wurde mittels EuroQol (EQ)-5D-5L in Nachsorgeuntersuchungen erhoben. Zum gleichen Zeitpunkt wurden Patient*innen auf einer Skala von 0 (am schlechtesten) bis 10 (am besten) nach ihrem mentalen und kör-perlichen Gesundheitsstatus in der letzten Woche befragt. Es wurde untersucht, wie gut der Index-Wert des EQ-5D-5L mittels verschiedener Regressionsanalysen vorhergesagt werden konnte. Schließlich verglichen wir den Gesundheitsstatus nach intensivmedizini-scher Therapie mit spezifischen körperlichen und seelischen Beeinträchtigungen.
Ergebnisse: Insgesamt konnten 1463 Patient*innen an 10 Clustern in die Studie einge-schlossen werden. Die Adhärenz aller QI bis auf QI 1 konnten durch die Intervention gesteigert werden. In der Nachsorge von 850 Patient*innen wies der Index-Wert des EQ-5D-5L (Median 0,78 [IQR 0,45-0,91]) eine hohe Korrelation zur mentalen (Spe-arman: 0,59) und körperlichen Gesundheit (Spearman: 0,68) auf. Die Kombination beider Items (nRMSE 0,164; nMAE 0,118) konnte den EQ-5D-5L Index-Wert besser vorhersa-gen als die visuelle Analogskala (nRMSE 0,175; nMAE 0,124) des EQ-5D. Unter Be-rücksichtigung weiterer Kovariaten konnte das Modell zusätzlich verbessert werden (nRMSE 0,133; nMAE 0,1). Zu den häufigsten, angegebenen Beschwerden bei niedriger mentaler Gesundheit gehörten neben Stimmungsschwankungen auch Niedergeschla-genheit und Schlafprobleme. Schwierigkeiten beim Laufen oder Treppensteigen, Kraftlo-sigkeit, Abgeschlagenheit und Schmerzen zählten zu den häufigsten Einschränkungen bei niedriger körperlicher Gesundheit.
Schlussfolgerung: Eine telemedizinische Visite erhöht die intensivmedizinische Be-handlungsqualität. In der ambulanten Nachsorge kann durch eine subjektive Bewertung des mentalen und körperlichen Gesundheitsstatus die HrQoL angenähert und mögliche zugrundeliegende Beschwerden abgeleitet werden.Background: Overall care for critical ill patients improved in the last few years but is still deficient. First, indicators of quality (QI) of evidence-based medicine do not seem to be fully implemented. Second, survivors of critical illness frequently report a health-related quality of life (HrQoL) worse than the general population. Although recommended, HrQoL after discharge of affected patients is often not assessed. Novel short items could facilitate screening in primary care, but these items are not yet validated. Aim of this work was, to perform a construct validation of two items for subjective mental and physi-cal health and thus validate new measurement instruments for primary care.
Methods: In an analysis of a multicentre, stepped-wedge cluster-randomized controlled trial of patients with critical illness the influence of telemedical visits on QI-adherence was assessed. In a secondary analysis, we analysed a cohort of critical illness survivors. As part of post-ICU discharge follow-ups, the EuroQol (EQ)-5D-5L was used to measure HrQoL. Subjective health was further evaluated by letting patients rank their mental and physical health status in the previous week from 0 (worst) to 10 (best). Various regressi-on models have been used to predict the EQ-5D-5L index value. Finally, we compared the subjective health status after critical illness with different specific physical and mental conditions patients suffered from.
Results: At 10 participating clusters 1463 patients could be enrolled. QI performance increased significantly for all QI but QI 1 via the telemedical intervention. We conducted follow-up assessments with 850 patients. In the analysis the EQ-5D-5L index value (me-dian 0.78 [IQR 0.45-0.91]) was higher correlated with physical (Spearman 0.68) than with subjective mental health (Spearman 0.59). Both items together (nRMSE 0.164; nMAE 0.118) predicted the EQ-5D-5L index value better than the EQ-5D visual analogue scale (VAS) (nRMSE 0.175; nMAE 0.124). Model performance was further increased by including additional covariates (nRMSE 0.133; nMAE 0.1). The most common symptoms reported by patients with low mental mental health ratings included mood changes, de-pressive symptoms, and difficulties sleeping. Patients with low physical health ratings most frequently reported difficulties walking or climbing stairs, lack of strength, fatigue, and pain.
Conclusion: Telemedical visits increase quality of care for critical ill patients. Short, subjective ratings of mental and physical health can be applied in the ambulatory follow-ups of survivors of critical illness to approximate their HrQoL. These items may also al-low inferences on specific underlying conditions of ICU survivors
Klinische und genetische Risikofaktoren für die Entwicklung reif oder unreif geborener Kinder
Auf der NICU ist das Frühgeborene verschiedenen Risiken für seine weitere Entwicklung ausgesetzt. Diese Habilitationsschrift fasst eigene Arbeiten zusammen, die darauf abzielen, Faktoren zu identifizieren, die (patho)physiologische Prozesse beeinflussen, und die zugrundeliegenden molekularen Mechanismen zu untersuchen, um Hinweise auf sinnvolle Lösungsansätze zu finden.
So wurde eine PCR-Diagnostik zum Nachweis der Besiedlung mit dem kritischen NICU-Keim Serratia marcescens etabliert und in der Praxis validiert, damit schneller Kohortierungs- und Barrieremaßnahmen getroffen und so Ausbruchsgeschehen verhindert werden können. Durch dieses Screening verbesserte sich die Handlungsfähigkeit der Hygienebeauftragten und die Patientensicherheit an der Klinik für Neonatologie an der Charité. In einem anderen Projekt wurde untersucht, ob eine genetische Variante (SNP rs1617640) im Promotor des Erythropoietin(EPO)-Gens einen Risikofaktor für die Entstehung einer schweren Retinopathie des Frühgeborenen (ROP ≥3°), mit oder ohne Behandlung mit rekombinantem Epo (rhEpo), darstellte. Im Studienergebnis zeigte sich kein Zusammenhang zwischen der EPO-Promotor-SNP-Variante und schwerer ROP bei Frühgeborenen <1.500 g Geburtsgewicht, unabhängig von der Behandlung mit rhEpo. Weitere potenzielle Risikofaktoren untersuchte ich in einer Studie zur Vermeidung Transfusions-assoziierter Erkrankungen. Es ist noch wenig verstanden, welchen Effekt die Transfusion adulter Thrombozyten auf das neonatale hämostatische System mit relativ hypofunktionellen Thrombozyten besitzt. Daher wurde in der eigenen Arbeit der Einfluss von Präparationsart, Lagertemperatur und –dauer auf die Freisetzung sog. Biological Response Modifiers (BRMs) untersucht, die in Vorversuchen eine höhere Konzentration in adultem als in neonatalem Blut gezeigt hatten. Zur Vermeidung einer Exposition mit unnötig hohen Mengen solcher adult-spezifischer BRMs kann man auf Grundlage der Ergebnisse empfehlen, immer das frischste Thrombozytenkonzentrat zu verwenden. Der Überzeugung folgend, dass unreife Frühgeborene durch Optimierung der Umgebungsbedingungen die zur Verfügung gestellten Ressourcen möglichst effizient zur Reifung nutzen können, wurde die Frage nach dem Mechanismus der endogenen zirkadianen Regulation der Epo-Produktion aufgegriffen. Zusammenfassend lässt sich sagen, dass die normoxische zirkadiane Epo-Expression transkriptionell durch Bindung der zirkadianen Masterregulatoren CLOCK/BMAL1 an eine E-Box-Sequenz im EPO-Promotor reguliert ist und dass der chronotherapeutisch optimale Applikationszeitpunkt von rhEpo während der Nacht liegt. Einen weiteren Schwerpunkt der eigenen Arbeit stellt die Erforschung molekularer Mechanismen der Adaptation an intrauterine und postnatale Veränderungen des Sauerstoffpartialdrucks (pO2) dar. So sind die typischen Frühgeborenen-Erkrankungen ROP und Bronchopulmonale Dysplasie (BPD) u.a. die Folge eines postnatal zu hohen pO2. Intrauterin herrscht ein physiologisch so niedriger pO2, sodass es zur Stabilisierung des Transkriptionsfaktors Hypoxie induzierbarer Faktor (HIF) kommt. Bei Frühgeburt verlässt das Kind diese physiologisch-hypoxische Umgebung zu früh, sodass es zu einer „Vollbremsung“ hypoxisch vermittelter Entwicklungsprozesse, vor allem der Gefäßentwicklung, kommt. In der Lunge der sich entwickelnden Maus zeigte die eigene Arbeit, dass HIF nur intrauterin stabilisiert wurde, die An- oder Abwesenheit jedoch keinen Einfluss auf die Expression typischer HIF-Zielgene hatte. Das Sauerstoffsensing-System besteht aus HIF und den HIF-destabilisierenden Prolylhydroxylase(PHD)-Enzymen. Das HIF-PHD-System war in Lungen ex vivo Organkulturen jedes Entwicklungsalters (embryonal, fetal, neonatal, adult) funktionsfähig. Die Analyse von Single cell-mRNA-Sequenzierdaten zeigte, dass vor allem die Phd1-Hif1-Achse, unterstützt von der Phd3-Hif3-Achse während der intrauterinen Entwicklung, für die Lungenentwicklung relevant waren. Zudem legten die Daten die Vermutung nahe, dass eine postnatale HIF2α-Aktivität trotz hohen Umgebungs-pO2 ein Schlüssel für die korrekte Vaskularisierung und damit Alveolarisierung der Lunge sein könnte. Das würde für Strategien sprechen, die eine extrauterine Fortsetzung der durch Frühgeburt und/oder Sauerstoffsupplementation unterbrochenen bzw. gestörten Lungenangiogenese und alveolarisierung fördern
Fermi-level pinning of Yu-Shiba-Rusinov states in a superconductor with weakly broken spin-rotational invariance
As is well known, magnetic impurities adsorbed on superconductors, e.g., of the s-wave type, can introduce a bound gap-state (Yu-Shiba-Rusinov resonance). We here investigate within a minimal model how the impurity moment arranges with respect to a weak homogeneous internal magnetic field employing a fully self-consistent mean-field treatment. Our investigation reveals a critical window of impurity-to-substrate coupling constants, J. The width of the critical region, SJ, scales like SJ B/A, where B is the magnitude of the internal field, that breaks the spin-rotation symmetry, and A is the bulk order parameter. While tuning J through the window, the energy of the Yu-Shiba-Rusinov (YSR) resonance is pinned to the Fermi energy epsilon F and the impurity moment rotates in a continuous fashion. We emphasize the pivotal role of self-consistency: In treatments ignoring self-consistency, the critical window adopts zero width, SJ = 0; consequently, there is no pinning of the YSR-resonance to epsilon F, the impurity orientation jumps and therefore this orientation cannot be controlled continuously by fine-tuning the coupling J. In this sense, our study highlights the significance of self-consistency for understanding intricate magnetic interactions between superconductive materials and Shiba chains
Temporal dynamics of antimicrobial resistance gene abundances in chicken manure and anaerobic digestate
Introduction
Antimicrobial resistance (AMR) can spread in microorganisms through the transfer of antimicrobial resistance genes (ARGs). Livestock husbandry is one of the pathways for AMR emergence and transmission. Chicken manure contains valuable nutrients for agricultural field fertilization and can be used as input material for biogas production by anaerobic digestion (AD). However, usually, chicken manure also contains quite high levels of ARGs. In this study, we investigated the presence and temporal dynamics of ARGs against different antibiotic classes in chicken manure and anaerobic digestate as a source of AMR spread.
Methods
To get an overview of the ARG profiles, we quantified the abundances of 374 ARGs by high-throughput (HT)-PCR. We studied eight selected ARGs ( tetA , tetX , sul1 , sul2 , lnuF , emrD , aadA , and tnpA ) using qPCR in chicken manure from different flocks and animal ages and in digestate from different AD time points.
Results
Chicken manure showed higher amounts of ARGs compared to digestate, which was characterized by a higher ARG diversity. We observed that the effect of chicken age differed between the flocks. ARG abundances in digestate from different time points and different treatment conditions did not exhibit major changes.
Conclusion
The flocks’ variability had no relevant effect on ARG abundances in chicken manure, likely due to similar growth conditions. However, manure ARG content increased with the age of the chickens. In our experimental batch setup, AD was more effective in reducing AMR microorganisms than reducing ARGs. Further investigations on process optimization or the application of pre-treatment methods could enhance ARG reduction. Notably, pre-mixing chicken manure with material from a biogas plant prior to processing resulted in a lower ARG load compared to untreated chicken manure
Multiparous and primiparous mothers partially differ in how they allocate maternal behaviour in captive western lowland gorillas (Gorilla gorilla gorilla)
In primates, mothers are crucial for the survival and integration of young offspring into their social group. Usually, mothers adjust their behaviour to the offspring’s age and needs, but maternal experience might modulate developmental changes in such allocation. In this study, we conducted behavioural observations on 7 mother-offspring dyads of captive western lowland gorillas ( Gorilla gorilla gorilla ) to assess whether multiparous mothers adjusted to the changing requirements of their offspring differently from primiparous mothers, and better facilitated their social integration into the group. Our results showed that, compared to primiparous mothers, multiparous mothers were more likely in body contact with younger offspring and less with older offspring. However, maternal experience neither predicted nor mediated how likely mothers were to start or end body contact, nor did it mediate developmental changes in the offspring’s social behaviour toward other group members. Our study provides preliminary evidence of some limited differences in how primiparous and multiparous mothers allocate maternal behaviour in western lowland gorillas
Retrospective analysis comparing different pedicles of the nipple-areola complex after inverted-T reduction plasty regarding cosmetic and operative parameters
Background: Despite numerous innovations in screening methods and treatment, breast cancer is still the most common cancer in women in the European Union and is the third most common type of cancer worldwide. Therefore, there is a continued need for improvement in the methods of breast cancer treatment. Surgical intervention is one of the cornerstones in the treatment of breast cancer. In this regard, breast cancer surgery has evolved over the decades with an aim to achieve a curative as well as aesthetically pleasing result with less perioperative complications. At the Kliniken Essen-Mitte (KEM), one of the modified surgical techniques is retention of the
mediocranial pedicle of the nipple-areola complex during the standard inverted-T reduction
mammoplasty.
The aim of the presented analysis is to describe the modified surgical technique with emphasis on operative and patient-centered parameters, adverse events in the mediocranial pedicle compared to other pedicles, possible risk factors and patient satisfaction assessed by the validated BreastQ questionnaire.
Methods: Retrospective data from 387 inverted-T reduction mammoplasty procedures performed
at the KEM Breast Center between May 2014 and September 2019 were analyzed. The procedures were categorized into three subgroups according to indication: plastic surgery intervention, tumor position-adapted intervention, and reduction mammoplasty of the contralateral breast to address breast asymmetry following breast conserving surgery, hereafter referred to as adaption to tumor side. Data evaluated included 17 clinical and surgical parameters, and patient satisfaction assessed using the BreastQ questionnaire preoperatively, and at 6 weeks and 6 months after the operation.
Statistical analysis was performed using SPSS 23.
Results: Postoperative complication rates (26.6%) were low as described in the published
literature. In our analysis, the main complications were wound healing disorders (18.1%) with
1.8% nipple-areola-complex-related, 13.2% superficial and 3.1% deep. Tumor position-adapted
interventions were more likely to have postoperative complications (p = 0.003), mainly surgical revisions (n = 37; 9.5%, p < 0.001), compared to procedures with an adaption to tumor side (n =
22; 5.7%) or plastic surgery (n = 25; 6.5%). Comorbidities such as diabetes mellitus and nicotine abuse correlated with an increased incidence of adverse events. Patient satisfaction was
consistently high at 6 weeks and 6 months postoperatively. An inverted-T reduction mammoplasty with retention of the mediocranial pedicle was found to be associated with a better outcome in terms of the overall result of surgery and in the physical, psychological, and sexual quality of life.
Conclusion: To summarize, the reduction mammoplasty according to the modified technique at KEM is a safe and effective method, accompanied by high patient satisfaction.Hintergrund: Trotz zahlreicher Innovationen der Screeningmethoden und der Behandlung bleibt das Mammakarzinom die vorherrschende Krebsart bei Frauen in der Europäischen Union und zählt global zu den 3 häufigsten Tumorerkrankungen. Eine wichtige Säule in der Therapie des Mammakarzinoms ist die operative Therapie. Diesbezüglich sind über Jahrzehnte zahlreiche Modifikationen erfolgt, welche den Anspruch haben neben einem kurativen ebenfalls ein
komplikationsloses und ästhetisch ansprechendes Ergebnis zu erzielen. Eine modifizierte Operationstechnik wird im Brustzentrum der Kliniken Essen-Mitte (KEM) praktiziert. Hierbei erfolgt eine mediokraniale Stielung des Mamillenareolakomplexes (MAK) in Kombination mit der standardisierten Schnittführung entsprechend eines umgekehrten Ts (KEM-Standard).
Ziel der vorliegenden Arbeit ist es diese modifizierte Reduktionsplastik der KEM zu beschreiben
mit Schwerpunkt auf der Darstellung von operativen und patientenzentrierten Parametern,
unerwünschten Ereignissen bei der mediokranialen Mamillenstielung im Vergleich zu anderen
Stielungen, möglichen Risikofaktoren und der Patientinnenzufriedenheit erfasst mithilfe des standardisierten BreastQ-Fragebogens.
Methoden: Es wurden retrospektiv 387 invertierte T-Reduktionsplastiken ausgewertet, welche zwischen Mai 2015 und September 2019 im Brustzentrum der KEM durchgeführt worden sind.
Die Reduktionsplastiken wurden entsprechend der Indikation in 3 Subgruppen unterteilt: Tumorlageadaptierter, Plastischer und Angleichungseingriff der Gegenseite nach erfolgter
brusterhaltender Therapie. 17 klinische und operative Parameter wurden erhoben, sowie die Zufriedenheit mittels BreastQ-Fragebogen präoperativ, nach 6 Wochen und 6 Monaten erfasst.
Die statistische Analyse erfolgte mittels SPSS 23.
Ergebnisse: Die postoperativen Komplikationsraten (26,6%) waren vergleichsweise niedrig wie in der Literatur beschrieben. Hauptkomplikationen waren in unserer Analyse Wundheilungsstörungen (18,1%) mit 1,8% MAK-bezogenen Wundheilungsstörungen, 13,2%
oberflächlich und 3,1% tief. Bei tumorlageadaptierten Reduktionsplastiken zeigten sich signifikant häufiger postoperative Komplikationen (p = 0,003), hauptsächlich operative Revisionen (N = 37; 9,5%, p < 0,001), im Vergleich zu Angleichungsoperationen (N = 22; 5,7%) oder plastischen Eingriffen (N = 25; 6,5%). Nebendiagnosen wie Diabetes mellitus und Nikotinabusus korrelierten mit einem erhöhten Auftreten unerwünschter Ereignisse.
Die Patientinnenzufriedenheit erreichte 6 Wochen und 6 Monate postoperativ ein konstant hohes Niveau. Eine invertierte T-Reduktionsplastik mit mediokranialer Stielung korrelierte in Bezug auf
das Gesamtergebnis der Brust sowie bei der physischen, psychischen und sexuellen Lebensqualität mit einer höheren Zufriedenheit.
Schlussfolgerung: In Zusammenschau unserer Ergebnisse ist die modifizierte Technik der
Reduktionsplastik der KEM eine sichere und effektive Methode, welche zudem zu einer hohen
Patientinnenzufriedenheit führt
