From Exploration to Critique: Catalyzing Critical Inquiry With Cultural Collection Visualizations
This article examines whether visualizations in cultural heritage fields can move beyond their traditional roles of exploration and analysis to facilitate higher order cognitive processes central to the practice of “critique.” Drawing on arts and humanities perspectives, we define critical cognition as a mode of inquiry that explores and evaluates the potentiality of cultural objects and topics—and interrogates the associated representation and research systems that shape them—against the background of their actual state. We propose a framework for thinking about the critical potential of visualization systems and outline design strategies that enhance critical cognition and encourage responsive action. To that end, we examine multiple system components, including culture and context, collection practices, digital modeling, visualization, and research practices. We argue that by incorporating critical perspectives on these system components into interface design, we can empower users, foster more reflective and nuanced engagement with cultural collections, deepen understanding of their social, historical, and epistemic contexts, and ultimately strengthen trust in visualization methods within the arts and humanities

Genomische Insertionen in der Vielfalt menschlicher Antikörper
Human antibodies variety is crucial for normal immune system performance. Antibodies are synthesized by the B cells and diversified by genomic rearrangement of V(D)J segments, somatic hypermutation and isotype switching. Recently, fragments of LAIR1 and LILRB1 genes were found to be inserted in the immunoglobulin heavy chain locus of B cells extracted from malaria-exposed individuals. These insertions encoded an additional domain binding a broad spectrum of repetitive interspersed family (RIFIN) proteins of malaria-causing Plasmodium falciparum. This doctoral study aims to address the following questions: what is the frequency of such events, how malaria exposure contributes to it, and what can be a mechanism of these phenomena. To profile the insertions incorporated in the antibody genes, a technique called suppression PCR was developed. It predominantly amplifies the products of a certain length, allowing to selectively enrich the insertion-containing immunoglobulin transcripts. Applying this method to 56 donors from Africa and Europe, we detected 1822 chimeric heavy chain immunoglobulin transcripts in 89.3% of the donors. The frequency of the insertions varied between 1.67·10-6 and 1.74·10-4 per B cell and did not differ significantly between donors of African and European origin. The insertions were incorporated either in between the VDJ segments of the variable domain (VDJ inserts) or in between the variable and constant domains (J-CH1 inserts). We characterized the insertions based on their genomic origin, developmental stage of B cells in which they were detected, GC composition, expression level, proximity to telomeres, sources of DNA damage, and AID or RAG off-targets. These features allowed us to classify the insertion-containing transcripts into four classes: fragments of nuclear genome (nucVDJ) and mitochondrial DNA (mtVDJ) incorporated in the variable domain, nuclear DNA originating from telomere- and R-loop-proximal regions (telJC), and AID-mediated insertions originating from ERFS-proximal genomic regions (nucJC). 13.2% of the insertions did not interfere with normal heavy chain reading frame and a set of the insertions bears somatic hypermutations, pointing at the affinity maturation. A selection of chimeric antibodies were expressed in vitro as grafts on various antibody backbones. While most of the chimeric antibodies lost the original target binding activity, a subset of them acquired an unspecific binding, providing a putative explanation of chimeric antibody presence in human repertoire. Collectively, this dissertation sheds light on a new layer of antibody diversification.Die Vielfalt der menschlichen Antikörper ist entscheidend für die normale Leistung des Immunsystems. Antikörper werden von den B-Zellen synthetisiert und durch genomische Umlagerung von V(D)J-Segmenten, somatische Hypermutation und Isotypwechsel diversifiziert. Kürzlich wurde festgestellt, dass Fragmente der LAIR1- und LILRB1-Gene in den Immunglobulin-Schwerketten-Locus von B-Zellen eingefügt sind, die aus Malaria-exponierten Personen extrahiert wurden. Diese Insertionen kodierten für eine zusätzliche Domäne, die ein breites Spektrum von Proteinen der repetitiven interspersierten Familie (RIFIN) des Malaria-verursachenden Plasmodium falciparum bindet. Diese Doktorarbeit zielt darauf ab, die folgenden Fragen zu beantworten: Wie häufig treten solche Ereignisse auf, wie trägt die Malaria-Exposition dazu bei und was kann ein Mechanismus für diese Phänomene sein? Um die in die Antikörpergene eingebauten Insertionen zu profilieren, wurde eine Technik namens Suppressions-PCR entwickelt. Es verstärkt vor allem die Produkte einer bestimmten Länge und ermöglicht so eine selektive Anreicherung der insertierten Immunglobulin-Transkripte. Bei der Anwendung dieser Methode auf 56 Spender aus Afrika und Europa konnten wir bei 89,3% der Spender 1822 chimäre Immunglobulin-Transkripte der schweren Kette nachweisen. Die Häufigkeit der Insertionen variierte zwischen 1,67·10-6 und 1,74·10-4 pro B-Zelle und unterschied sich nicht signifikant zwischen Spendern afrikanischer und europäischer Herkunft. Die Insertionen wurden entweder zwischen den VDJ-Segmenten der variablen Domäne (VDJ-Inserts) oder zwischen der variablen und der konstanten Domäne (J-CH1-Inserts) eingebaut. Wir charakterisierten die Insertionen anhand ihres genomischen Ursprungs, des Entwicklungsstadiums der B-Zellen, in denen sie nachgewiesen wurden, der GC-Zusammensetzung, des Expressionsniveaus, der Nähe zu Telomeren, den Quellen von DNA-Schäden und AID- oder RAG-Off-Targets. Diese Merkmale ermöglichten es uns, die insertierungshaltigen Transkripte in vier Klassen zu klassifizieren: Fragmente des Kerngenoms (nucVDJ) und der mitochondrialen DNA (mtVDJ), die in die variable Domäne eingebaut sind, Kern-DNA, die aus Telomer- und R-Loop-proximalen Regionen stammt (telJC) und AID-vermittelte Insertionen, die aus ERFS-proximalen Genomregionen (nucJC) stammen. 13,2% der Insertionen störten den normalen Leserahmen der schweren Kette nicht und ein Satz der Insertionen weist somatische Hypermutationen auf, die auf die Affinitätsreifung hinweisen. Eine Auswahl chimärer Antikörper wurde in vitro als Transplantate auf verschiedenen Antikörpergrundgerüsten exprimiert. Während die meisten chimären Antikörper ihre ursprüngliche Zielbindungsaktivität verloren, erlangte eine Untergruppe von ihnen eine unspezifische Bindung, was eine mögliche Erklärung für das Vorhandensein chimärer Antikörper im menschlichen Repertoire darstellt. Psychosomatische Merkmale chronischer tinnituskorrelierter Belastung
Chronic Tinnitus denotes the longstanding, conscious awareness of a tonal or composite noise for which there is no identifiable external acoustic source. Depending on the psychological makeup of affected individuals, tinnitus can be highly distressing. Current guidelines view the symptom together with its cognitive-emotional processing as a psychosomatic phenomenon – and suggest (1) medico-audiological treatment for underlying medical influences – if known, and (2) psychological treatment for highly distressed individuals. Anchored within a psychosomatic vulnerability-stress-coping framework, the present thesis sets out to examine chronic tinnitus-related distress. It asks two questions:
1. In how far do direct or indirect psychological factors – that may or may not correlate with ‘somatic’ variables – influence individuals’ experience of a tinnitus symptom as distressing? And:
2. In how far are psychosomatic (i.e. psychologically or medico-audiologically anchored) treatment approaches effective in ameliorating tinnitus-related distress?
Chapter 1 [Introduction] briefly reviews evidence on vulnerability-stress-coping contributors to chronic tinnitus symptomatology and related distress-reactions. Chapter 2 [Vulnerability] presents the first two studies of the thesis, which examine chronic tinnitus-related distress at the junctions of (1) biological markers of tinnitus symptom vulnerability or perceived stress experiences and (2) psychological vulnerability-stress interactions. Chapter 3 [Stress] presents the third study, which investigates, whether transdiagnostically relevant psychological variables underlie chronic tinnitus- and pain-related distress experiences. Chapter 4 [Coping] examines the efficacy of psychosomatic treatment approaches. The final three studies of this thesis examine (1) whether a transdiagnostic psychological treatment approach ameliorates different functional symptom clusters, (2) whether a ‘somatic’ treatment approach (hearing amplification) bears psychological benefit, and (3) psychological effects on hearing amplification via hearing aid use-time. Ventricular Assist Device Implantation for Ischemic Cardiomyopathy Study – VADIICS
Patientinnen und Patienten, die bei koronarer Herzkrankheit (KHK) an Herzinsuffizienz versterben, nehmen eine beachtenswerte Gruppe innerhalb der Todessursachenstatistiken der westlichen Welt ein. Eine der häufigsten invasiven Behandlungsmethoden ist die aortokoronare Bypass-Operation (ACB-OP). Bei Patientinnen und Patienten mit stark fortgeschrittenem Krankheitsbild ist die Datenlage zur optimalen Versorgung jedoch un-eindeutig. Die Kunstherztherapie bietet sich als vielversprechende Alternative für ausge-wählte Patientinnen und Patienten an. Vor diesem Hintergrund stellt sich die Frage, ob diese der ACB-OP bei Patientinnen und Patienten mit KHK und fortgeschrittener Herzinsuffizienz überlegen/gleichwertig ist.
Es erfolgte eine retrospektive Datenauswertung von Patientinnen und Patienten, die an KHK und fortgeschrittener Herzinsuffizienz (LVEF < 26 %) litten und entweder ein Kunstherz erhielten oder mittels ACB-OP versorgt wurden. Die Behandlung erfolgte zwischen 2015 und 2018 am Deutschen Herzzentrum Berlin (DHZB). Eingeteilt wurde in permanente Herzunterstützungssystem-(Assist Device)-Kohorte (n = 97) und aortokoronare Bypass-Kohorte (n = 100). Als primärer Endpunkt wurde die Einjahresmortalität bestimmt. Folgend konnten die einflussreichsten Risikofaktoren für Therapieversagen (Versterben und/oder Einsatz von Extracorporeal-Life-Support-Systemen und/oder permanenten Her-zunterstützungssystemen) während des stationären Aufenthalts durch eine Subgrup-penanalyse der aortokoronaren Bypass-Kohorte identifiziert werden. Aus diesen identifi-zierten Risikofaktoren wurde ein Risiko-Score gebildet.
Es zeigte sich kein signifikanter Unterschied im Einjahres-Überleben zwischen den Kohorten (p = 0,598; HR(LVAD) = 0,80). Aufgrund der geringen Fallzahlen zu Rehospitalisierung und Lebensqualität erfolgte die Auswertung deskriptiv und vergleichend. In der Bypass-Kohorte kam es zu 33 Rehospitalisierungen (34,55 %). Eine Rehospitalisierung in der Assist Device Gruppe erfolgte 197 Mal (57,29 %). Numerisch fand sich in Bezug auf die Lebensqualität ein Unterschied zwischen den Kohorten. So betrug der Median der visuellen Analogskala-(VAS)-Daten in der Assist-Device-Gruppe 62,5, bei 48 nacher-fassbaren Patientinnen und Patienten. Der Median der VAS-Daten in der Bypass-Gruppe betrug 55, bei 28 nacherfassbaren Patientinnen und Patienten.
Die Subgruppenanalyse und Modellselektion für den Risiko-Score der aortokoronaren Bypass-Kohorte identifizierte Diabetes mellitus (OR = 44,66), reduzierte LVEF (OR = 1,11) und Höhe des INTERMACS-Levels (OR = 4,09) als einflussreichste Risikofaktoren.
Die Assist Device Therapie stellt einen vielversprechenden therapeutischen Ansatz bei Patientinnen und Patienten mit KHK, fortgeschrittener Herzinsuffizienz und entsprechenden Risikofaktoren für ein Therapieversagen der ACB-OP dar. Weitere prospektive, mul-tizentrische Studien können helfen, eine klare Behandlungsempfehlung für diese Patienten- und Patientinnengruppe bereitzustellen.Patients suffering from heart failure and coronary artery disease represent a remarkable group in the Western world’s cause mortality statistics. Coronary artery bypass grafting is the mainstay of surgical treatment. However, there’s a lack of eminent data on optimal treatment for patients with the severest forms of heart failure. Assist-Device-Implantation may be a promising alternative for selected patients. The question is whether it’s superior or equal to coronary artery bypass graft surgery in this population.
A retrospective data analysis was performed on patients suffering from coronary artery disease and severe heart failure (left ventricular ejection fraction < 26 %), treated with either coronary artery bypass graft surgery or with assist device implantation. Treatment was performed at the German Heart Center Berlin (DHZB) between 2015 and 2018. Patients were divided in an artificial heart cohort (n = 97) and a coronary artery bypass graft surgery cohort (n = 100). One year mortality was defined as the primary endpoint. In addition, a subgroup analysis identified major risk factors for treatment failure (death and/or need for extracorporeal life support and/or assist device implantation) in the coronary artery bypass surgery cohort. These were used to create a risk score. There was no significant difference in one-year survival between the cohorts. Due to the small number of cases of rehospitalization and quality of life cases, the analysis was descriptive and comparative. In the bypass cohort, patients had to be rehospitalized 33 times. This represented 34,55 % of the patients who could be followed up. In comparison, rehospitalization occurred 197 times in the assist-device-group. This represented 57,29 % of the patients who could be followed up.
Numerically, there was a difference in quality of life between the cohorts. The median VAS data in the assist device group was 62,5, with 48 patients available for follow up. The median VAS data in the bypass group was 55, with 28 patients available for follow up.
Subgroup analysis and model selection for the risk score of the aortocoronary bypass cohort identified diabetes mellitus (OR = 44,66), reduced left ventricular function (OR = 1,11) and INTERMACS level (OR = 4,09) as the most influential risk factors.
Assist-Device-Treatment may be an important therapeutic approach in patients with coronary artery disease, severe heart failure, and major risk factors for coronary artery bypass graft surgery treatment failure. Further prospective, multicenter studies may help to establish a clear treatment recommendation for this patient population
Understanding change from the patient perspective in a transdiagnostic Internet-based intervention for emotional disorders: a qualitative content analysis
Qualitative investigations that openly explore changes and facilitators of changes from the patient’s perspective might offer valuable insights on impacts of therapy and helpful and hindering aspects. Our aim for this study was to explore the perspective of patients on a transdiagnostic Internet-based intervention to understand (1) which changes (positive as well as negative effects) responders and non-responders experienced, and (2) which aspects of the intervention they found helpful or hindering in facilitating those changes. We interviewed 21 patients that showed response or non-response to treatment using the Change Interview Schedule following a 10-week Internet-based intervention based on the Unified Protocol. Interviews were analyzed using qualitative content analysis. Both responders and non-responders reported positive changes, with few negative changes mentioned. Across both groups, increased positive affect was reported most frequently (81%). Both groups reported helpful factors, with guidance mentioned most frequently across both responders and non-responders (85.7%). Mainly, aspects of the specific framework were perceived as hindering (e.g. lack of personalization) (66.7%). Overall, patients reported mostly positive impacts, even if they did not respond to treatment. Results highlighted that what patients find helpful or hindering is individual
Einflussfaktoren auf das corneale Endothel organkultivierter Spenderaugenhornhäute
Neben spenderabhängigen Einflussfaktoren und einem unvermeidbaren Endothelzellverlust im Rahmen einer passiven Organkultivierung, auch wenn diese aktuell den Goldstandard darstellt, hat die Augenhornhautbank Einfluss auf die Endothelzelldichte von Augenhornhäuten.
Die fehleranfällige Bestimmung der cornealen Endothelzelldichte von Spenderaugenhornhäuten lässt sich durch eine computerbasierte Analyse zweifelsfrei positiv beeinflussen. Wir konnten zeigen, dass die entwickelte Bildanalysesoftware vollautomatisch valide und reproduzierbar die zentrale Endothelzelldichte bestimmt.
Unsere Untersuchungen fanden einen relevanten Endothelzellverlust durch die Verwendung einer stärker hypoosmolaren Substanz für die lichtmikroskopische Visualisierung dieser Zellen. Durch die Verwendung einer möglichst gering hypoosmolaren Substanz mit einer niedrigen Expositonsdauer kann die Endothelzellschicht im Rahmen der Organkultivierung von Spenderaugenhornhäuten geschützt werden.
Hypotherme und hypertherme Temperaturänderungen zeigten in unseren Arbeiten in einem Bereich zwischen +4 und +42°C keinen Effekt auf die cornealen Endothelzellen von organkultivierten Augenhornhäuten. Temperaturen unterhalb des Gefrierpunktes und über +42°C müssen, z.B. während des Transportes von Augenhornhauttransplantaten zum Empfänger, streng vermieden werden, da ansonsten eine komplette Nekrose der empfindlichen Endothelzellschicht eintritt.
Die aktuellen Forschungen zu einer aktiven Organkultivierungsform von Spenderaugenhornhäuten ergaben vielversprechende Ergebnisse hinsichtlich einer Protektion der Endothelzellen. Dabei muss auf die bekannten positiven Effekte der etablierten Organkultur nicht verzichtet werden. Solle sich dieser Ansatz erfolgreich in die Routine etablieren lassen, wird es voraussichtlich zu einer echten Evolution der Konservierung von Spenderaugenhornhäuten kommen

Surveillance of Strongylus vulgaris and other Strongylus spp. in equids is important for targeted intervention in parasite control, requiring reliable routine diagnostic methods.

Methods
During the routine diagnostic investigations, in total 712 strongyle-egg positive equine faecal samples were cultured. Third-stage larvae (L3) were morphologically differentiated. For molecular validation, samples were examined using S. vulgaris real-time PCR and Strongylus edentatus/Strongylus equinus/Strongylus asini high-resolution melting PCRs.
Results
Based on 28S rRNA PCR, 594 samples positive for nematode DNA were included in the study. The inter-rater reliability to compare morphological and molecular species identification was fair for Strongylus spp. without species identification and for S. edentatus, slight for S. equinus and poor for S. vulgaris. The frequency based on morphological and molecular data in this study were for S. vulgaris 0% and 0.8%, respectively, for S. edentatus 0.3% and 1.5%, respectively, and for S. equinus 2.0% and 0.2%, respectively. Based on molecular analyses, one sample obtained from a domestic horse contained S. asini DNA, which was confirmed by sequencing.
Main limitations
For many samples, no or only incomplete data regarding clinical history, the exact geographical location and whether samples were obtained on individual or farm level, were available.
Conclusions
Results of morphological and molecular examination methods of strongyle L3 from equine samples can differ substantially. Further evaluation of these methods is required to provide reliable and cost-effective methods of screening equine parasites. Further studies using approaches suitable to detect S. asini are needed to evaluate its clinical and epidemiological relevance
Der umsetzbare Verlust von SLF2 treibt die B-Zell Lymphomgenese voran und beeinträchtigt die DNA Reparatur
Large-scale sequencing studies provide a precise characterization of the genomic landscape of aggressive human B-cell lymphomas (BCLs). However, the large num-ber of the genetic and non-genetic alternations identified in these studies make it chal-lenging to pinpoint functionally relevant drivers of lymphomagenesis. Here, we lever-aged a functional in vivo genome-scale screening experiment in a model of B-cell lymphomagenesis with a set of known ATM and ATR substrates to identify genes in-volved in DNA damage response (DDR), and specifically identified SMC5-SMC6 complex localization factor 2 (SLF2) as a tumor suppressor of B-cell lymphomagenesis and functionally relevant to DDR. To unravel the mechanism associated with SLF2 loss, we performed global transcriptome and proteomic profiling of SLF2-deficient B-cell lymphoma cells and found that SLF2 is an important regulator of ATR-CLSPN-CHK1 axis. Mechanistically, SLF2 loss resulted in an accumulation of DNA double-strand breaks (DSBs) as indicated by increased .H2.AX expression and foci formation.
Further, SLF2 deficiency led to compromised the activation of the DDR checkpoint ki-nase CHK1 and depleted expression of CHK1 upstream regulator CLSPN. Importantly, SLF2 was also suppressed in human BCL and low SLF2 expression was associated with adverse prognosis in the cohorts of diffuse large B-cell lymphoma (DLBCL) pa-tients. The B-cells lacking SLF2 are more resistant to cyclophosphamide, a key com-ponent of standard immunochemotherapeutic regiment for BCL patients. Of note, the accumulation of DNA damage in the cells due to SLF2 loss alternates the post-translational SUMOylation pathway, thus leads to the sensitivity towards pharmacolog-ical inhibition of SUMOylation in these cells. Moreover, this co-dependency confers synthetic lethality to a clinically applicable SUMOylation inhibitor and CHK1 inhibitors, which was verified in both syngeneic and xenografted mouse model.
In summary, we here identified SLF2 as a novel tumor suppressor of B-cell lym-phomagenesis and a regulator of the ATR-CHK1 pathway. Furthermore, we reveal that co-targeting of the SUMOylation and DDR pathway as a promising clinical strategy for the treatment of aggressive human cancer.Groß angelegte Sequenzierungsstudien ermöglichen eine genaue Charakterisierung der Genomlandschaft aggressiver menschlicher B-Zell-Lymphome (BCLs). Allerdings macht es die große Anzahl der in diesen Studien identifizierten genetischen und nichtgenetischen Veränderungen schwierig, funktionell relevante Treiber der Lym-phomagenese zu identifizieren. Hier nutzten wir ein funktionelles In-vivo-Screening-Experiment im Genommaßstab in einem Modell der B-Zell-Lymphomgenese mit einer Reihe bekannter ATM- und ATR-Substrate, um Gene zu identifizieren, die an der DNA-Schadensreaktion (DDR) beteiligt sind, und identifizierten speziell SMC5 -SMC6-Komplex-Lokalisierungsfaktor 2 (SLF2) als Tumorsuppressor der B-Zell-Lymphomgenese und funktionell relevant für DDR. Um den mit dem SLF2-Verlust ver-bundenen Mechanismus aufzuklären, führten wir ein globales Transkriptom- und Pro-teomprofil von SLF2-defizienten B-Zell-Lymphomzellen durch und stellten fest, dass SLF2 ein wichtiger Regulator der ATR-CLSPN-CHK1-Achse ist. Mechanistisch gese-hen führte der SLF2-Verlust zu einer Anhäufung von DNA-Doppelstrangbrüchen (DSBs), was durch eine erhöhte .H2.AX-Expression und Herdbildung angezeigt wur-de.
Darüber hinaus führte ein SLF2-Mangel zu einer beeinträchtigten Aktivierung der DDR-Checkpoint-Kinase CHK1 und einer verminderten Expression des CHK1-Upstream-Regulators CLSPN. Wichtig ist, dass SLF2 auch in menschlichem BCL un-terdrückt wurde und eine niedrige SLF2-Expression mit einer ungünstigen Prognose in den Kohorten von Patienten mit diffusem großzelligem B-Zell-Lymphom (DLBCL) verbunden war. Die B-Zellen, denen SLF2 fehlt, sind resistenter gegen Cyclophos-phamid, einen Schlüsselbestandteil der standardmäßigen immunchemotherapeuti-schen Behandlung von BCL-Patienten. Bemerkenswert ist, dass die Anhäufung von DNA-Schäden in den Zellen aufgrund des SLF2-Verlusts den posttranslationalen SUMOylierungsweg verändert und somit zu einer Empfindlichkeit gegenüber einer pharmakologischen Hemmung der SUMOylierung in diesen Zellen führt. Darüber hin-aus verleiht diese Co-Abhängigkeit einem klinisch anwendbaren SUMOylierungsinhi-bitor und CHK1-Inhibitoren synthetische Letalität, was sowohl im syngenen als auch im xenotransplantierten Mausmodell verifiziert wurde.
Zusammenfassend haben wir hier SLF2 als einen neuartigen Tumorsuppressor der B-Zell-Lymphomagenese und einen Regulator des ATR-CHK1-Signalwegs identifiziert.
