Comparative genomic study of blaKPC-2-carrying multidrug-resistant Klebsiella pneumoniae in the United Arab Emirates
AbstractWe performed whole-genome sequencing of three multidrug-resistant KPC-producing Klebsiella pneumoniae isolates recovered from patients in a tertiary-care hospital in Abu Dhabi, UAE, and conducted a comparative genomic analysis with previously reported blaKPC−2-positive isolates from the region. The study aimed to characterize acquired antimicrobial resistance genes (ARGs), assess their phylogenetic context, and analyze the plasmids carrying blaKPC−2. Between 2017 and 2018, 162 carbapenem-resistant K. pneumoniae (CRKP) isolates were collected from four tertiary care hospitals in Abu Dhabi. Antibiotic susceptibility was determined using the Vitek2 system, and PCR was used to detect carbapenemase genes. Whole-genome sequencing was performed for the three blaKPC-2-positive isolates using Illumina NovaSeq, and assemblies were generated with SPAdes 3.9. PlasmidSPAdes and PLACNETw were used to reconstruct blaKPC-2-carrying plasmids, which were then compared with plasmids described in earlier UAE reports. The three K. pneumoniae isolates belonged to sequence types ST11, ST20, and ST231, respectively. All harbored the blaKPC−2 gene on IncFII-type plasmids of varying sizes. Our comparative analysis demonstrated variability in the genetic platforms carrying blaKPC−2, with two transposons (Tn1721a and Tn4401b) identified as the main structures associated with this gene in UAE isolates. Conclusions: Our findings highlight the diversity of genetic platforms carrying blaKPC-2 in the UAE, particularly the frequent association with IncFII plasmids and their potential role in the dissemination of antimicrobial resistance. Although blaKPC-2 is a globally widespread carbapenemase gene, it has been rarely reported in the United Arab Emirates (UAE). To date, only two studies describing blaKPC-2-carrying Klebsiella pneumoniae isolates have been published from the region—in 2015 and 2019. Our study presents a comprehensive genomic analysis of blaKPC-2-positive isolates collected in 2017–2018 from tertiary care hospitals in the UAE. Compared to previous reports, this study employed advanced bioinformatics tools and conducted deep genomic surveillance, including resistome, plasmidome, transposon mapping, and phylogenetic comparisons with public genomes. We identified distinct genetic platforms carrying blaKPC-2, notably the association with IncFII plasmids and the continued presence of the Tn4401b transposon. Although blaKPC-2 is present in the UAE, it remains underreported, likely due to limited regional sequencing efforts. This study provides essential baseline genomic data and a comparative framework to support ongoing and future surveillance of antimicrobial resistance in the Gulf region
Bisparse blind deconvolution through hierarchical sparse recovery
The hierarchical sparsity framework , and in particular the HiHTP algorithm(Hierarchical Hard Thresholding Pursuit), has been successfully applied to many relevant communication engineering problems recently, particularly when the signal space is hierarchically structured. In this paper, the applicability of the HiHTP algorithm for solving the bi-sparse blind deconvolution problem is studied. The bi-sparse blind deconvolution setting here consists of recovering hand bfrom the knowledge of h∗(Qb), where Qis some linear operator, and both band hare assumed to be sparse. The approach rests upon lifting the problem to a linear one, and then applying HiHTP, through the hierarchical sparsity framework . Then, for a Gaussian draw of the random matrix Q, it is theoretically shown that an s-sparse h∈Kμand σ-sparse b∈Knwith high probability can be recovered when μ≳slog(s)2log(μ)log(μn)+sσlog(n)

sie die soziale Teilhabe und die Identitätsentwicklung. Trotz des Prinzips der Chancengleichheit bleibt der Bildungserfolg in Deutschland nach wie vor stark von der sozialen Herkunft und somit vom familiären Kontext abhängig. In dieser Dissertation wird das
Zusammenspiel verschiedener Aspekte des familiären Zusammenlebens und ihr Einfl uss
auf die Bildung von Kindern untersucht.
Die Analysen basieren auf Daten der Startkohorte 3 (SC3) des Nationalen Bildungspanels
(NEPS) sowie des Beziehungs- und Familienpanels (pairfam). Im Fokus stehen Kinder, die mindestens die vierte und höchstens die zehnte Klasse besuchen. Die Ergebnisse zeigen, dass die Qualität der innerfamiliären Beziehungen eine Rolle für die Bildung dieser Kinder spielt. In der deutschen Forschung fand dieses Thema bislang wenig Beachtung, ebenso wie die Einbeziehung der Wahrnehmung und Perspektive der Kinder auf die Familie. Untersucht wird insbesondere, wie Veränderungen innerhalb der Familie - sowohl hinsichtlich ihrer Form als auch der Beziehungsqualität - die Mathematikleistungen und Bildungsaspirationen von Kindern beeinflussen. Ein besonderer Fokus liegt dabei auf dem Vergleich zwischen Kindern, die mit beiden leiblichen Elternteilen in einem Haushalt aufwachsen, und solchen, die dies nicht tun.

Despite the principle of equal opportunities, educational success in Germany remains highly dependent on social background, and thus, family context. This dissertation examines how various aspects of family life influence children's educational outcomes.
The analyses are based on data from the Starting Cohort 3 (SC3) of the National Educational Panel Study (NEPS) and the Panel Analysis of Intimate Relationships and Family Dynamics Study (pairfam). The focus is on children attending at least the fourth and at most the tenth grade. The results show that the quality of relationships within the family does play a role in these children's education. This topic has received little attention in German research so far, as has the inclusion of children's perceptions and perspectives on the family. The effects of changes within the family, in terms of both form and relationship quality, on children's mathematics performance and educational aspirations are being researched in particular. The comparison between children who grow up with both biological parents in one household and those who do not is also examined.
The findings of this dissertation emphasize the significant impact of family relationship dynamics on educational processes and highlight the importance of considering the perspectives and needs of children. The results show that comprehensive support, including emotional and material support, is essential for avoiding negative impacts on school performance, particularly in unstable family situations. The results provide valuable insights for family and education policy, stressing the need for inclusive support services that provide all children with equal educational and developmental opportunities, regardless of their family situation. Algae-dominated metaproteomes uncover cellular adaptations to life on the Greenland Ice Sheet
Eukaryotic algae-dominated microbiomes thrive on the Greenland Ice Sheet (GrIS) in harsh environmental conditions, including low temperatures, high light, and low nutrient availability. Chlorophyte algae bloom on snow, while streptophyte algae dominate bare ice surfaces. Empirical data about the cellular mechanisms responsible for their survival in these extreme conditions are scarce. This knowledge gap was addressed by quantifying proteins for both algal taxa from samples on the southern margin of the GrIS. We show that the streptophyte glacier ice algae have a relative enrichment in proteins involved in environmental signaling and nutrient transport, indicative of cellular readiness to dynamically respond to extreme GriS environmental cues, linked, for example, to photoprotection and the rapid update of scarce nutrients. In contrast, the chlorophyte snow algae have a high abundance of proteins linked to lipid and nitrogen metabolisms, providing evidence for the biological processes sustaining the cellular carbon and nitrogen stores necessary for survival in an oligotrophic environment. We also identify proteins in both taxa linked to the synthesis and breakdown of key cellular pigments. Our study gives novel insights into the cellular biology of these algae and their adaptation to extreme environments
Accelerated ultrafast demagnetization of an interlayer-exchange-coupled Co/Mn/Co trilayer
We investigate the ultrafast magnetization dynamics of an interlayer-exchange-coupled Co/Mn/Co trilayer system after excitation with an ultrafast optical pump. We probe element- and time-resolved ferromagnetic order by x-ray magnetic circular dichroism in resonant reflectivity. We observe an accelerated Co demagnetization time in the case of weak total parallel interlayer coupling at 9.5 ML Mn thickness for antiparallel alignment of both Co layers compared to parallel alignment as well as for parallel alignment in the case of strong parallel interlayer coupling at 11 ML of Mn. From ab initio time-dependent density functional theory calculations, we conclude that optically induced intersite spin transfer of spin-polarized electrons from Co into Mn acts as a decay channel to enhance and accelerate ultrafast demagnetization. This spin transfer can only take place in case of a collinear Mn spin structure. We argue that this is the case for antiparallel alignment of both Co layers at 9.5 ML Mn thickness and parallel alignment in case of 11 ML of Mn. Our results point out that an antiferromagnetic spacer layer and its spin structure have a significant effect on the magnetization dynamics of adjacent ferromagnetic layers. Our findings provide further insight into fundamental mechanisms of ultrafast demagnetization and may lead to improve dynamics in multilayered systems for faster optical switching of magnetic order
Validation of a protocol for mapping of the supplementary motor area using navigated transcranial magnetic stimulation in healthy subjects
Hintergrund: Für die Durchführung komplexer Bewegungen spielt neben dem primären Motorkortex (M1) auch das supplementär motorische Areal (SMA) eine zentrale Rolle. Unser Verständnis der genauen Funktion und Lokalisation dieses räumlich nicht klar abgegrenzten Areals ist dabei noch unvollständig. Ziel der hier vorliegenden Arbeit war es deshalb, ein vorgeschlagenes Protokoll zur Kartierung des SMA mittels repetitiver navigierter transkranieller Magnetstimulation (rnTMS) an gesunden Pro-banden zu validieren und im Zuge dessen, die Darstellung der unteren Extremität im SMA genauer zu lokalisieren sowie Effekte hemisphärischer Dominanz zu untersuchen.
Methodik: In diese Studie wurden 30 gesunde Probanden (Alter 36,0±15,1 Jahre, Altersspanne 21-67 Jahre; 14 weiblich) eingeschlossen. Um den Einfluss hemisphärischer Dominanz auf die Organisation des SMA zu untersuchen, erfolgte eine Kartie-rung dieses Areals für beide Hemisphären mittels rnTMS-basierter Stimulation (20 Hz, 120% Ruhemotorschwelle (RMT)) mit Hilfe einer einhändigen Finger-Tapping-Aufgabe. Nachfolgend wurde die Spezifität der Effekte durch Ergänzung des Nine-Hole-Peg-Tests (NHPT) geprüft sowie eine mögliche Kompensation durch das kontra-laterale SMA mittels beidhändigem Finger-Tapping untersucht. Anschließend erfolgte die rnTMS-basierte Untersuchung der unteren Extremität mit Hilfe einer Zehen-Tapping-Aufgabe (20 Hz, 140% RMT).
Ergebnisse: In Bezug auf die hemisphärische Dominanz konnten keine signifikanten Unterschiede eruiert werden. Die Kartierung war für 24 von 30 Probanden für die obere und für 27 von 30 Probanden für die untere Extremität möglich. Eine erfolgreiche Kartierung stellte sich durch eine Abnahme der motorischen Perfomance bei der Finger- beziehungsweise Zehen-Tapping-Aufgabe unter Stimulation dar. Im Falle der oberen Extremität war eine signifikante Reduktion der Anzahl der Finger-Taps im Vergleich zur Ausgangsbasis erkennbar (p<0,01), die beim Wechsel zu beidhändigem Finger-Tapping zumindest teilweise kompensiert werden konnte (p<0,01). Fehler in der Bewegungsausführung der unteren Extremität traten bei der Stimulation weiter kaudal gelegener Anteile der SMA-Region im Vergleich zur oberen Extremität auf, wobei klare Grenzen nicht erkennbar waren.
Schlussfolgerung: Das mit dieser Arbeit validierte Protokoll ermöglicht die nicht-invasive rnTMS-basierte Kartierung des SMA. Durch die ergänzende Testung der unteren Extremität, kann eine mögliche somatotopische Organisation des SMA untersucht werden. Fehlende Einflüsse bezüglich der hemisphärischen Dominanz sowie die beobachtete Kompensation durch das kontralaterale SMA unterstützen die These, dass die Rehabilitation SMA-assoziierter Defizite maßgeblich von der Rekrutierung der gesunden Hemisphäre bestimmt wird.
Zukünftige Validierungen an Patienten sind erforderlich, um das Protokoll langfristig im klinischen Alltag für die präoperative Planung sowie zur Risikoabschätzung postoperativer Defizite zu etablieren.Background: The supplementary motor area (SMA) plays a key role in executing complex movements alongside the primary motor cortex (M1). However, the precise function and localisation of the SMA are not yet fully understood. Therefore, the purpose of this study was to validate a protocol for mapping of the SMA using repetitive navigated transcranial magnetic stimulation (rnTMS). Additionally, it aimed to investigate a more precise localisation of the lower extremity representation within the SMA as well as effects of hemispheric dominance.
Methods: In this study, 30 healthy subjects (age 36.0±15.1 years, range 21-67 years; 14 female) were included. To investigate the influence of hemispheric dominance on the organisation of the SMA, mapping of this area was performed for both hemi-spheres using rnTMS (20 Hz, 120% resting motor threshold (RMT)) while subjects per-formed a unimanual finger tapping task. The specificity of the effects was verified by adding the Nine-Hole Peg Test (NHPT) and a potential compensation by the contrala-teral SMA was explored using a bimanual finger tapping task. Subsequently, the lower extremity was examined based on a toe tapping task (20 Hz, 140% RMT).
Results: No significant differences regarding hemispheric dominance could be ob-served. Mapping was possible in 24 out of 30 subjects for the upper and in 27 out of 30 subjects for the lower extremity. Successful mapping was indicated by a decrease in motor performance during the finger or toe tapping task while stimulating. For the upper extremity, a significant reduction in the amount of finger taps compared to base-line was found (p<0.01), which could be at least partially compensated when switching to bimanual finger tapping (p<0.01). Errors in movement execution of the lower extremity occurred when stimulating more caudal parts of the SMA region compared to the upper extremity, although clear boundaries could not be determined.
Conclusion: The protocol validated by this dissertation provides the opportunity to non-invasively map the SMA using rnTMS. By including tests for the lower extremity, poten-tial somatotopic organisation of the SMA can be investigated. The lack of effects re-garding hemispheric dominance and the observed compensation by the contralateral SMA support the hypothesis that rehabilitation of SMA-associated deficits is signifi-cantly influenced by the recruitment of the healthy hemisphere.
Future validations in patients are necessary to establish the protocol for clinical use in preoperative planning and to assess the risk of postoperative deficits
Modelling triaxial deformation of continental tectonics
Polymodal faulting, which comprises three or more coeval fault sets, has been documented from centimeter to kilometer scale in nature. Differing from previously well-known conjugate, or bimodal, fault patterns under biaxial or plane strain, the polymodal faulting phenomenon is considered to represent triaxial deformation or strain (e.g., constriction and flattening strain). Triaxial tectonic regimes can take place variably in obliquely divergent, transcurrent, and convergent settings, allowing the coeval existence of different tectonic regimes (e.g., thrust, strike-slip, and normal faulting). The classical and widely accepted Mohr-Coulomb failure criterion can predict the orientation of conjugate fault planes parallel to (i.e., contain) the direction of the intermediate stress under plane strain, whereas polymodal fault patterns forming under triaxial deformation cannot be explained with fault orientations oblique to the direction of the intermediate principal stress (σ2). Aiming to understand the evolution, geometry, and kinematics of polymodal faults, I use 3D scaled analogue and numerical modelling to generate triaxial strain fields under variable initial and boundary conditions to decode the mechanisms and mechanics of triaxial deformation.
In the first step, I primarily investigate how rheology and strain rate control deformation localization, faulting regime, and pattern in brittle‐viscous crustal‐scale models under constriction strain. I perform triaxial analog experiments by varying strain rates (or extension velocity), where distributed longitudinal extension resulting in crustal thinning is accompanied by lateral shortening. I found the structural style of faults and the degree of localization as a function of strain rate. As strain rate (ė) decreases, (1) fault patterns change from conjugate sets of strike‐slip faults (ė > 3 × 10⁻⁴ s⁻¹) to sets of parallel oblique normal faults (ė = 0.3–3 × 10⁻⁴ s⁻¹) to horst‐and‐ graben system (ė < 0.3 × 10⁻⁴ s⁻¹); (2) The strain localization increases systematically and gradually. The former change in fault pattern is interpreted to be affected by the strain rate dependency of vertical coupling between the model upper crust and upper mantle, which controls spontaneous switching of principal stress axes. The latter change in strain localization is controlled by mechanical coupling between the upper and lower crust.
Furthermore, many extensional systems often experience multiple phases of deformation; however, the spatial and temporal evolution of fault networks during triaxial and biaxial strain is still poorly understood. Benefiting from the first part of the work, which identifies the change in strain conditions from constriction to plane strain over strain rate, I also use a scaled analogue model to investigate fault geometry and activity across multiple phases of triaxial (constrictional) and biaxial (plane) strain by changing extension velocity to obtain time-dependent kinematic strain conditions. Our modelling results show that (1) when shifting from plane to constrictional strain, earlier developed normal faults are completely reactivated and new conjugate sets of oblique-slip faults form during the constrictional phase; (2) when shifting from constrictional to plane strain, conjugate sets of oblique-slip faults forming during constrictional strain are randomly abandoned or reactivated. New normal faults developed during plane strain cut across and link up earlier-phase faults. Kinematic interactions of fault networks between multiphase strains are identified by observing how perturbations in stress/strain domains control the geometry of new faults and mechanical obstacles hinder fault propagation.
I further explore the relationship between principal stresses and polymodal fault orientation by means of 3D scaled numerical modelling. Our models demonstrate that triaxial deformation can be accommodated by the simultaneous development of faults with different trends and partitioned into one more faulting regime. Additionally, our models can explain fault distribution with respect to the principal stress.
The modelling results show good similarity with the natural prototypes at various backgrounds. In regions under a constrictional deformation field, for example, Tibet, Anatolia, and the Friulian-Venetian basin, as well as in regions under a flattening strain, for example, northern Tibet and the Barents Sea, our modelling results provide new implications for fault geometry, fault kinematics, and stress distribution. Moreover, our models with multiphase triaxial and biaxial strain can re-interpret tectonic evolution in the Aegean and Barents Seas.Polymodale Verwerfung, die aus drei oder mehr gleichzeitig aktiven Verwerfungssätzen besteht, ist in der Natur auf Skalen von Zentimetern bis Kilometer dokumentiert worden. Das klassische und weit akzeptierte Mohr-Coulomb-Bruchkriterium kann die Orientierung konjugierter Verwerfungsflächen vorhersagen, die parallel zur Richtung der intermediären Spannung (σ2) liegen. Polymodale Verwerfungsmuster, die unter triaxialer Deformation entstehen, können jedoch nicht mit Verwerfungsorientierungen erklärt werden, die schräg zur intermediären Hauptspannung liegen.
Mit dem Ziel, die Entwicklung, Geometrie und Kinematik polymodaler Verwerfungen zu verstehen, verwende ich 3D-analoge und numerische Modellierungen im Labormaßstab, um triaxiale Deformationsfelder unter variablen Anfangs- und Randbedingungen zu erzeugen und die Mechanismen sowie die Mechanik triaxialer Deformation zu entschlüsseln.
Im ersten Schritt untersuche ich hauptsächlich, wie Rheologie und Dehnungsrate die Lokalisierung der Deformation, das Verwerfungsregime und -muster in spröde-viskosen, krustalen Modellen unter Streckungsdeformation kontrollieren. Ich führe triaxiale Analogexperimente mit variierenden Dehnungsraten (bzw. Extensionsgeschwindigkeit) durch, wobei eine verteilte longitudinale Dehnung zur Krustenverdünnung gleichzeitig mit lateraler Verkürzung auftritt. Ich fand heraus, dass der strukturelle Stil der Verwerfungen und das Maß der Lokalisierung von der Dehnungsrate abhängen. Mit abnehmender Dehnungsrate (ė) ändern sich (1) die Verwerfungsmuster von konjugierten Sätzen von Seitenverschiebungen (ė > 3 × 10⁻⁴ s⁻¹) zu parallelen Sätzen schräger Abschiebungen (ė = 0,3–3 × 10⁻⁴ s⁻¹) bis hin zu Horst-und-Graben-Systemen (ė < 0,3 × 10⁻⁴ s⁻¹); (2) Die Deformationslokalisierung nimmt systematisch und allmählich zu. Die erste Veränderung des Verwerfungsmusters wird als Folge der von der Dehnungsrate abhängigen vertikalen Kopplung zwischen oberer Kruste und oberem Mantel interpretiert, welche die spontane Umorientierung der Hauptspannungsachsen steuert. Die zweite Veränderung der Lokalisierung wird durch die mechanische Kopplung zwischen oberer und unterer Kruste beeinflusst.
Da viele extensionale Systeme mehrere Deformationsphasen durchlaufen, ist die räumlich-zeitliche Entwicklung von Verwerfungsnetzwerken während triaxialer und biaxialer Dehnung noch wenig verstanden. Aufbauend auf den Ergebnissen des ersten Teils, die eine Veränderung der Dehnungsbedingungen von Streckung zu ebenen Dehnungen in Abhängigkeit der Dehnungsrate aufzeigen, nutze ich ein skaliertes Analogmodell zur Untersuchung der Geometrie und Aktivität von Verwerfungen über mehrere Phasen triaxialer (streckender) und biaxialer (ebener) Deformation, wobei die Extensionsgeschwindigkeit geändert wird, um zeitlich variierende kinematische Dehnungsbedingungen zu simulieren. Unsere Modellergebnisse zeigen, dass (1) beim Übergang von ebener zu streckender Dehnung zuvor entwickelte Abschiebungen vollständig reaktiviert und neue konjugierte Sätze von Schrägverschiebungen gebildet werden; (2) beim Übergang von Streckung zu ebener Dehnung konjugierte Sätze schräger Verwerfungen entweder zufällig aufgegeben oder reaktiviert werden. Neue Abschiebungen, die während der ebenen Dehnung entstehen, durchschneiden und verbinden ältere Verwerfungen. Kinematische Wechselwirkungen zwischen Verwerfungsnetzwerken über mehrere Deformationsphasen hinweg werden durch Beobachtung von Spannungs-/Dehnungsstörungen erkannt, die die Geometrie neuer Verwerfungen kontrollieren, sowie durch mechanische Hindernisse, die die Ausbreitung von Verwerfungen hemmen.
Ich untersuche auch die Beziehung zwischen Hauptspannungen und der Orientierung polymodaler Verwerfungen mittels 3D-skalierten numerischen Modellen. Unsere Modelle zeigen, dass triaxiale Deformation durch die gleichzeitige Entwicklung von Verwerfungen mit unterschiedlichen Orientierungen aufgenommen werden kann, die sich auf mehrere Verwerfungsregime aufteilen lassen. Darüber hinaus können unsere Modelle die Verteilung der Verwerfungen in Bezug auf die Hauptspannungen erklären.
Die Modellergebnisse zeigen eine gute Übereinstimmung mit natürlichen Vorbildern in verschiedenen geologischen Umgebungen. Liquid Ammonia—Revisiting an Old Friend with New Purpose
Ammonia was first discovered as a chemical solvent in the 19th century. Extensive investigations during the 20th century toward its physical and chemical properties have yielded substantial knowledge regarding the compound. Historically, its merit was mainly seen in chemical synthesis and its ability to accommodate syntheses that could not be pursued in aqueous solvents. With a growing demand for low-emission energy systems, the scientific focus on ammonia shifted toward its potential for efficient hydrogen storage and transportation, as well as for direct combustion. Herein, a brief overview of the development of the role of ammonia, with the goal of highlighting its unique properties as a solvent, is provided, in particular, in the chemical syntheses of unprecedented species and reactive compounds

Online monitoring activism: civic surveillance practices as a reaction to the rise of the far-right in the COVID-19 pandemic
The rise of far-right movements during the COVID-19 pandemic has prompted a new form of individual digital activism: online monitoring activism (OMA). During the pandemic, online monitoring activists systematically collected, processed and published information from far-right Telegram channels and groups. Against the theoretical background of concepts like the monitory democracy and surveillance culture, we conducted eight semi-structured interviews with German online monitoring activists, investigating their motivations, roles, and the intricacies of their monitoring practices. Our findings reveal that online monitoring activists are driven by a sense of duty to counteract anti-democratic tendencies, which they perceive as inadequately addressed by institutional power. They gain localized and thematically specialized expertise that they share within loose networks of like-minded others. We highlight the activists’ liminal identity oscillating between virtuous citizenship and vigilantism, as well as the broader societal implications of their actions. On the one hand, they fulfill the role of active citizenship in monitory democracy; on the other, they also reinforce the transparency imperative and the wish for far-reaching security and control inherent to surveillance culture. The transparency potential afforded by their adversaries’ online connective action and mobilizing efforts legitimizes their surveillance and demands surveillance. Further normative work is needed to critically examine the extent and desirability of increased social control introduced to liberal democracy by online monitoring activism
