Nitrogen deposition mitigates long-term phosphorus input-induced stimulative effects on soil respiration in a tropical forest

Atmospheric nitrogen (N) deposition and anthropogenic phosphorus (P) input simultaneously affect soil respiration (RS), a crucial process that mediates soil carbon (C) cycling. However, the interaction of N deposition and anthropogenic P input on RS, as well as its components—autotrophic respiration (RA) and heterotrophic respiration (RH)—remain largely unexplored. Herein, we conducted an 8-year field experiment with N and P additions in a tropical secondary forest, integrating the vegetation traits, soil physicochemical properties, organic C fractions, and microbial properties, to explore the effects of nutrient inputs and their interactions on RS, RA, and RH. Over eight years, along P input significantly increased RS by 19.2% and RH by 42.1%. These increases were partially mitigated (by 33.2% annually for RS and 58.3% annually for RH) with the addition of N. In contrast, the co-addition of N and P enhanced RA compared to alone N or P addition, suggesting that N deposition mitigated the stimulative effect of P input on RS by reducing RH rather than RA. The structural equation model further revealed that N deposition reduced RH primarily by increasing soil N:P ratio and decreasing both the labile C fraction and fungi biomass. Our findings suggest that prevalent N deposition across low latitudes could have substantially mitigate C emissions from forest soils under anthropogenic P input

With increasing age, functional limitations and chronic diseases occur more frequently. During this progress, the skin undergoes a natural ageing process, which increases the risk of adverse skin conditions. The aim of this dissertation was to investigate the effects of an evidence-based preventive skincare programme and adequate loading, to reduce adverse skin conditions in older nursing home residents. One the one hand, an explorative, controlled, cluster-randomised, pragmatic trial was conducted in institutional long-term care in Berlin, Germany. The epidemiology of care related skin problems and the effects of implementing an evidence-based skincare programme were analysed. The analysis of 314 residents over 65 years in long-term care facilities showed a high prevalence of adverse skin conditions e.g., xerosis cutis, incontinence-associated dermatitis, intertrigo, skin tears and pressure ulcers. Skin tears were in particular associated with cognitive and physical impairment, and pressure ulcers were associated with reduced mobility and increased care dependency. By implementing a structured and evidence-based skincare programme for a period of six months, the cumulative incidence of the most common care-related skin conditions was, except incontinence-associated dermatitis, lower in the intervention group compared to the control group. Besides preventive effects regarding skin health in aged nursing home residents, it could have been shown that adequate skincare with prior skin assessment was possible to implement in care practice. On the other hand, a secondary data analysis of a randomised, controlled, exploratory trial with crossover design investigated in vivo changes of skin properties after 120 minutes loading at sacral and heel skin, as well as possible associations. The measurement of skin physiological parameters after 120 minutes of loading on a basic foam mattress showed an increase of skin moisture, skin redness and skin temperature. Further analysis displayed that loading and pressure during mechanical deformation increased the erythematous response. Independently, an increase of skin temperature and humidity, as part of occlusion and a change of the microclimate, occurred

Discovering novel pharmaceutical drugs is a lengthy, complex, and resource-intensive process. Traditional approaches in chemoinformatics have provided valuable tools for predicting molecular properties, but these methods can fall short in terms of efficiency and predictive accuracy. When used in molecular generation, these limitations become more pronounced, as inaccuracies in property prediction can lead to the design of suboptimal candidates. This dissertation explores innovative approaches to drug discovery through the application of deep graph representation learning techniques, focusing on two areas: molecular property prediction and molecular generation. We propose novel graph neural network architectures designed to enhance expressiveness in 2D and 3D molecular graphs. Our approach incorporates feature transformations inspired by hypercomplex algebras or integrates group equivariance into the models, facilitating data-efficient training. For 3D molecular data, we demonstrate that rotation-equivariant networks can be scaled to process larger biomolecules and outperform invariant networks while remaining computationally efficient. Additionally, we introduce generative models based on diffusion probabilistic models that sample new 3D molecular structures with targeted properties, either for ligands in isolation or within protein-ligand complexes, using rotation-equivariant graph networks as denoisers. This aspect of our research aims to enhance the drug discovery pipeline by improving the efficiency of identifying promising drug candidates that meet multiple criteria. The results of this thesis suggest that deep graph representation learning has the potential to advance drug discovery by providing more accurate predictive tools and enhancing the ability to generate novel molecular candidates. This work contributes to the development of computational methods in drug discovery and may pave the way for further research into applying graph representation learning to complex chemical problems

Antimicrobial Resistance in Pasteurella multocida Isolates from Bovine Mastitis Can Be Associated with Multidrug-Resistance-Mediating Integrative and Conjugative Elements (ICEs)

Background/Objectives: Pasteurella multocida commonly colonizes the bovine respiratory tract and can occasionally cause intramammary infections. Here, eight P. multocida isolates from clinical cases of bovine mastitis were investigated for their molecular characteristics as well as phenotypic and genotypic antimicrobial resistance (AMR) properties. Methods: The isolates originated from quarter milk samples obtained in Germany for diagnostic purposes. Antimicrobial susceptibility testing (AST) by broth microdilution was performed according to the Clinical and Laboratory Standards Institute. Closed whole-genome sequences were generated by hybrid assembly of Illumina MiSeq short-reads and Oxford Nanopore MinION long-reads, followed by consecutive sequence analysis. Results: The P. multocida isolates belonged either to capsular:lipopolysaccharide type A:3 (n = 7) or A:6 (n = 1), and multi-locus sequence types 1 (n = 7) or 7 (n = 1). Seven isolates carried AMR genes, such as mef(C), mph(G), strA, strB, aphA1, aadA31, tet(H), tet(Y), floR, catA3, and sul2, as part of an integrative and conjugative element (ICE). These mobile genetic elements, 58,382–78,401 bp in size, were highly similar to the ICEs Tn7406 or Tn7407 that have been previously described in bovine Mannheimia haemolytica and P. multocida, respectively. Moreover, the isolates showed elevated minimal inhibitory concentrations corresponding to the identified AMR determinants. Conclusions: Molecular typing and ICE organization suggest the bovine respiratory tract as reservoir of the investigated mastitis-associated P. multocida. Horizontal cross-genus transfer of multidrug-resistance-mediating ICEs seems to occur under in vivo conditions among different pathogens from cattle in Germany, which underlines the importance of pathogen identification followed by AST for successful bovine mastitis therapy

Orthopedic implants are of high clinical relevance for the repair of musculoskeletal injuries and disorders. Conventional implants, like titanium and stainless-steel configurations, commonly require removal upon success of the bone healing process. Implant removal constitutes an extra burden for patients and adds to the healthcare costs. Bio-degradable implants present an alternative to conventional implants by making the implant removal surgery obsolete. This merit arises from the degrading nature inherent to the new implant materials such as Magnesium based alloys. Due to the material properties of these alloys, degradable implants offer temporary weight-bearing support essential for bone healing and are subsequently absorbed by the surrounding tissue. Post implantation, non-invasive tracking of the implantation site and of the healing process is part of the clinical workflow aided by imaging. The use of diagnostic X-ray or CT imaging for longitudinal monitoring of implantation sites may suffer from image artefacts caused by metallic implants. Both diagnostic imaging approaches contribute to the accumulation of ionizing radiation dosage in patients, which presents an elevated risk of developing malignancies. Hence leading to the clinical need for alternative implant monitoring methods free of ionizing radiation. Magnetic resonance imaging (MRI) is a viable alternative for monitoring of implantation sites. MRI is free of ionizing radia-tion, ensuring a safer diagnostic imaging approach. Even though a large body of literature has highlighted that conventional permanent metal-based implants potentially constrain the diagnostic quality of MRI and elevate the level of deposited radiofrequency (RF) energy in implantation sites. Yet, the impact of biodegradable implants with a dynamic structure (i.e., the degrading behavior alters the structure of the implant over time) on the quality of MRI is underexplored. Recognizing this opportunity, this thesis focuses on three main goals. First, the impact of biodegradable implants on MRI distortion artefacts at the implantation site is carefully examined. The main finding from this study is that Mg-based implants generate lower metallic distortion in MRI when compared to conventional Ti-based implants. Second, radiofrequency induced heating of biodegradable orthopedic screw implants due to the interference between the passively conducting implants and the RF power deposited in the body during an MRI examine is investigated. The main conclusion from this study is that the highest risk of RF induced implant heating is most pronounced for Mg-based screws prior to degradation. Following the careful assessment of the MRI characteristics of biodegradable implants a radiofrequency transmission field shim-ming method is developed, evaluated, validated and applied with the goal to compensate or even eliminate image distortion or RF heating in the presence of biodegradable implants to facilitate MRI-aided monitoring of implantation sites. The approach devel-oped in this thesis project makes use of RF antenna arrays customized for parallel RF transmission to generate dedicated electromagnetic field (EMF) patterns which are derived from RF excitation vector optimization tailored for MRI of implantation sites. The proposed method is validated in numerical simulations and benchmarked against con-ventional approaches in phantom experiments using a 7.0 T whole body MRI scanner. For this purpose, MRI-aided transmission field mapping, MR thermometry, and MR imaging of implantation sites were performed. In conclusion, parallel RF transmission using optimized RF excitation vectors in conjunction with dedicated RF antenna arrays provides a technical foundation en route to safe and MRI of implantation sites with diagnostic image quality free of transmission field distortions, which is the main achievement of this thesis project.

Correcting Emotional Deviance: Affective Inclusion Amid Social Inequality and Power Hierarchies

This article uses empirical data from a case study in Germany to introduce the concept of “affective inclusion.” Affective inclusion is understood as both an institutional and programmatic response to emotional deviance that arises in the wake of power hierarchies and social inequalities. The aim of these efforts is to incorporate individuals or groups into a dominant emotion culture, especially after their emotions have been labeled as deviant. The empirical study uses a unique combination of qualitative data and methods, including ethnographic observations during an anti-violence program for amateur soccer players and an analysis of a television report on the same workshop. The analysis addresses the social dynamics that determine the processes of affective closure and openness and examines the strategies, practices and techniques that institutional actors employ to address this phenomenon, including teaching emotional control and nudging participants into the prevailing emotion culture. In addition, the study shows how affective inclusion is intertwined with social inequality and discrimination

Was bedeutet Recht für uns Juristen? Auflag

Water and ions binding to extracellular matrix drives stress relaxation, aiding MRI detection of swelling-associated pathology

    Get PDF
Swelling-associated changes in extracellular matrix (ECM) occur in many pathological conditions involving inflammation or oedema. Here we show that alterations in the proportion of loosely bound water in ECM correlate with changes in ECM elasticity and stress relaxation, owing to the strength of water binding to ECM being primarily governed by osmolality and the electrostatic properties of proteoglycans. By using mechanical testing and small-angle X-ray scattering, as well as magnetic resonance imaging (MRI) to detect changes in loosely bound water, we observed that enhanced water binding manifests as greater resistance to compression (mechanical or osmotic), resulting from increased electrostatic repulsion between negatively charged proteoglycans rather than axial contraction in collagen fibrils. This indicates that electrostatic contributions of proteoglycans regulate elasticity and stress relaxation independently of hydration. Our ex vivo experiments in osmotically modulated tendon elucidate physical causes of MRI signal alterations, in agreement with pilot in vivo MRI of inflammatory Achilles tendinopathy. We suggest that the strength of water binding to ECM regulates cellular niches and can be exploited to enhance MRI-informed diagnostics of swelling-associated tissue pathology

Extraction Strategies for Profiling the Molecular Composition of Particulate Organic Matter on Glacier Surfaces

Pigmented microalgae thrive on supraglacial surfaces, producing “sticky” extracellular polymeric substances that combine into a mineral–organic matrix. Together, they enhance snow and ice melting by lowering the albedo. Understanding the chemical nature of particulate organic matter (POM) in this matrix is crucial in assessing its role in supraglacial carbon dynamics. We evaluated POM complexity in alga-rich snow and ice samples containing 0.3–6.4 wt % organic carbon (OC) via extractions with solvents of varying polarity, pH, and OM selectivity. Extraction yields were evaluated by OC analysis of the extracts, and the composition of extracted OM was analyzed using ultrahigh-resolution mass spectrometry. Individual hot water (HW), hydrochloric acid (HCl), and sodium hydroxide (NaOH) extractions achieved up to 87% efficiency, outperforming sequential, organic solvent-based extractions (<11%). OM extracted by HW, HCl, and NaOH combined had more molecular formulas (2827) than OM extracted with organic solvents (1926 formulas). Combined HW, NaOH, and HCl extractions yielded an OM composition with unsaturated, highly unsaturated, aromatic, and N-containing compounds, while unsaturated aliphatics and black carbon-derived polycyclic aromatics were enriched in the organic solvent extracts. This molecular profiling provides the first comprehensive insights into supraglacial POM composition, opening the window for understanding its role in the cryospheric carbon cycle

Evolution of the Swiss pork production systems and logistics: the impact on infectious disease resilience

    Get PDF
Livestock production systems are complex and evolve over time, affecting their adaptability to economic, political, and disease-related changes. In Europe, disease resilience is crucial due to threats like the African swine fever virus, which jeopardizes pork production stability. The European Union identifies farm production type as a key risk factor for disease spread, making it important to track changes in farm production types to assess disease risk. However, detailed production type data is often lacking in national databases. For Swiss pig farms, we used prediction and clustering algorithms to classify 9’687 − 11’247 trading farms between 2014 and 2019 by one of eleven production types. We then analyzed the pig trade network and stratified farm centrality measures (ICC and OCC) by production type. We found that 145 farms belonging to three production types have substantially higher ICC and OCC than other farms, suggesting that they could be the target of disease surveillance programs. Our predictions until 2025 show an increase both in overall pig trade network connectivity and in proportion of production types with high ICC and OCC, indicating that the structural changes in the Swiss pig production system may increase infectious disease exposure over time
