Investigating the effect of peptide-functionalized gold nanoparticles on colon cancer cells
Colon cancer like any other cancer is a condition in which cells grow uncontrollably and may even spread to other regions of the body through metastasis. Colon cancer was ranked the second leading cause of cancer related deaths worldwide in 2018. Research to advance treatment of cancer keeps advancing daily, However, a big challenge is drug-induced side effects due to toxicity against normal body cells. Therefore, the development of controlled release technologies in conjunction with targeted drug delivery may provide a more efficient and less dangerous approach to overcome the limits of traditional chemotherapy. Including the creation of nanoscale delivery vehicles capable of directing the release of chemotherapeutic drugs into cancer cells only. This study aims to investigate p14 peptide that would specifically target colorectal cancer cells and not normal body cells to develop a targeted drug delivery system using gold nanoparticles. This study serves as a pilot study of the primary aim. To achieve this, the effect of the peptide p14 and peptide functionalized gold nanoparticles (p14-AuNP) on colon cancer cells (HT-29) and normal epithelial cells (KMST-6) was determined. Firstly, gold nanoparticles were chemically synthesised and then functionalized with p14 peptide through Polyethylene glycol. Then assessment of their effect through in vitro cytotoxicity assay (MTT) and gene expression analysis (RT-qPCR) was conducted. Nanoparticles’ synthesis and functionalization was performed and confirmed: In vitro cytotoxicity through MTT assay was successfully conducted and p14-AuNP showed toxicity against colon cancer cells and lesser toxicity towards normal cells as compared to 5-Flourouracil (commercially approved drug for colon cancer treatment). Gene expression analysis revealed that apoptosis was induced in both cell lines by p14-AuNP either through upregulation of caspase 3, 7 and/or BCL2. A cell survival gene, AKT1, also had significant effect on this. Statistical analysis of hydrogeochemical data from fractured rock aquifers around Beaufort West, South Africa
Knowledge surrounding the procedures and protocols related to groundwater sampling are currently limited. In particular, the sampling of radioactive elements from fractured rock aquifers is poorly understood and in need of further research. The aim of this study is to provide a comparative insight into groundwater sampling methods used for the sampling of major and minor elements as well as trace elements within fractured rock aquifers in the immediate vicinity of Beaufort West. The influence of sampling source, seasonality, and vertical flows on sample results are observed. Furthermore, the importance of historical datasets is raised. Aims of this study were achieved by making use of historical data from numerous studies that were conducted near the Karoo town of Beaufort West. Potential profitable uranium deposits saw a great influx of research into the geology, topography, climate and hydrogeology. Synthesising data from these historical research papers has provided valuable information regarding the influences of sampling procedures on the results. A total of 695 well data samples were analysed from four studies collected from farms around Beaufort West. The importance of adequately purging a well prior to sampling cannot be stressed enough. There are minimal chemical differences between sampling boreholes and wind pumps, however there was a noticeable difference in the inter-relationships between major chemical elements. Groundwater samplers were found to be contaminated with Zinc and Copper attributed to wind pump fittings. Boreholes were also analysed to observe any changes in depth. Sodium, temperature and uranium had little to no changes with depth due to the homogenous nature of the borehole water. Principal component analysis was conducted on 159 groundwater samples comprised of 13 variables. These samples were reduced to four principal components (PC) which explained a total of 91.09% of the total variance. Factor one explained 61.99% of the total variance and was strongly associated with water-type and lithology while factor two (13.57%) shows the potential relationship between molybdenum and uranium as well as possible copper contamination as a result of wind pump and borehole fittings. Further research should be conducted by carrying out multiple sampling procedures on a selection of controlled wells to determine the best applicable sampling procedures.Thesis (MSc) -- Faculty of Science, School of Environmental Sciences, 202
Dietary assessment of Cape Gannets (Morus capensis) and African Penguins (Spheniscus demersus) in Algoa Bay using molecular scatology
Seabirds are important marine sentinels, providing valuable information about ecosystem health and condition. Dietary studies of seabirds can inform on ecosystem functioning through identifying the condition and changes occurring in their surrounding environment. Dietary studies are also crucial as they can identify threats that seabirds face. Until recently, the diets of southern African endemic and Endangered Cape Gannets Morus capensis and African Penguins Spheniscus demersus were solely investigated through stomach content analyses, which allows for the identification and quantification of prey remains. To overcome some of the limitations associated with stomach content analysis, stable isotope and fatty acid analyses were proposed. However, these methods still require bird handling and thus impose some stress on the birds. DNA metabarcoding of faecal matter shows promise as a completely noninvasive method for seabird dietary studies. This thesis first examined whether DNA metabarcoding could be used to determine the diet of Cape Gannet and African Penguin breeding in Algoa Bay, South Africa. Then, this method was used to investigate the influence of intrinsic (both seabird species) and extrinsic (African Penguins) factors on the diet of the two seabird species. For the first data chapter, stomach content samples and faecal matter were collected concurrently from breeding Cape Gannets and African Penguins on Bird Island. DNA was successfully extracted from most faecal matter samples and the primers (18S_SSU, 16S_Fish) used for group, taxa and fish prey identification allowed amplification of prey DNA for both study species. A similar piscivorous diet was determined by both dietary methods for Cape Gannets with Sardine Sardinops sagax and Atlantic Saury Scomberesox saurus dominating the diet. Sardine also dominated the African Penguin diet in both dietary methods. In addition, DNA metabarcoding in African Penguins revealed a more diverse diet compared to the one determined by stomach content analysis. This was likely due to the limitations on the allowed number of stomach content samples to be collected for that Endangered species. Non-invasive DNA metabarcoding of faecal matter was then used to investigate whether the diet varied with breeding stages (incubation, chick-rearing) and age classes (chick-rearing adults, chicks) in both seabird species, but also breeding colonies (Algoa Bay: Bird Island, St Croix Island) in African Penguins. DNA metabarcoding revealed that breeding stages influenced the diet of Cape Gannets and African Penguins on Bird Island where prey species diversity was greater for incubating adults. On the contrary, the diet of incubating and chick-rearing penguins on St Croix Island was similar. When comparing the diet of African Penguins between Bird Island and St Croix Island, there was an increased overlap in diet composition between incubating and chick-rearing adults on St Croix Island when compared to Bird Island. Importantly, DNA metabarcoding indicated an increase in fishery interactions in Algoa Bay, particularly for Cape Gannets and the hake Merluccius spp. fishery, where the contribution of hake to chick-rearing adults and chick diet was considerably greater than incubating adult diet. In this study, I showed that DNA metabarcoding of faecal matter provides an opportunity for non-invasive dietary studies in two southern African endemic and Endangered seabird species. An assessment of return to work strategies within the South African financial services industry
The purpose of this study was to conduct research into the return to work strategies that were used in the South African Financial Services Industry following the COVID-19 pandemic lockdown. The goal was to understand the motivation for the slow return to the office and to explore what strategies organisational leadership could employ to overcome this resistance. The South African Financial Services Industry represents multiple subsectors that are highly regulated in an age where the digital transformation of industries is common, as are the complexities and risks in integration. The significance of the study is to ascertain the appropriate strategies to encourage employees the return to the office since COVID-19. This study used a quantitative approach. This was achieved in the form of a survey sent to a sample of managers in South African Financial Services Industry using an electronic survey instrument created in Google Forms. One hundred and nineteen financial services managers fitting the criteria responded to the survey. The sampling methodology was both convenient and snowball sampling, non-probability techniques. The findings indicated that there is a relationship between extrinsic benefits received by employees and the return to work. The findings from the study also revealed that a hybrid working arrangement is preferred by the South African Financial Services Industry managers compared to working from home permanently From the study it is recommended that financial services organisations employ a hybrid working model where possible. The influence of Corporate Social Responsibility (CSR) and Corporate Citizenship (CC) on the business success of wineries in the Cape Winelands
The wineries of the Cape winelands are increasingly engaging in corporate social responsibility (CSR) and corporate citizenship (CC). Both these practices are widely viewed as having the potential to increase business success of these wineries. There is however a lack of research on the state of these practices in these wineries especially against the background of reports of exploitation of workers on winelands. The aim of this study is therefore to investigate the state of CSR and CC in the Cape winelands wineries and the impact that these practices have on the business success of these wineries. The study is located in the positivistic research tradition. The data was collected using a closed-ended questionnaire which was distributed to 100 employees (managers and general workers) in the wineries. The questionnaire items were anchored on a Likert-style (disagree to agree) answering scale. Pearson correlations and multiple regression coefficients, as well as descriptive statistics were used. Eventually only 42 respondents participated in the study. The empirical findings indicated that CSR (focusing on its economic, environmental, and social impacts) and CC were significantly positively related to the business success of the wineries in the Cape winelands. CC emerged as the strongest determinant of the business success of these wineries. The empirical results also revealed reasonable to high levels of CSR and CC prevalence in these wineries. Although limitations were experienced in the study, useful managerial recommendations emerged from it empirical results. Challenges and opportunities of coastal and marine tourism in Swartkops river estuary
This study's goal was to pinpoint the challenges and untapped opportunities associated with Coastal and Marine Tourism (CMT) in the Swartkops River Estuary (SRE). CMT has become the biggest segment of the travel industry. Moreover, this form of alternative tourism is the most significant and fastest developing economic activity in the ocean. The objectives of this study were to identify the challenges that hinder the development of CMT in SRE, to determine opportunities for CMT in SRE, to evaluate NMBM’s efforts in creating and sustaining CMT in SRE and to evaluate the status of CMT in SRE in terms of CMT related development. This study fit within the interprevist paradigm and made use of a qualitative research approach and design as well as inductive reasoning. The qualitative research design consisted of data being collected through document and content analysis. Data was analysed using data analysis process of Miles and Herman (1994) as well as the coding and developing of specific themes that emerged from the data. The data in this phase was presented both in narrative format and tables. The document analysis used in this study was established based on the objectives of this study. The findings of this study revealed that key challenges that hinder the growth of CMT in Swartkops River Estuary are environmental impacts that include pollution, over-enrichment, inappropriate development, overfishing, destruction of mudflats, limited freshwater inflow, destruction of vegetation and climate change. The findings also revealed that there are opportunities for CMT in SRE such as promoting SRE as a popular ecotourism destination by developing ecotourism nodes. These include environmental education at the Aloes Nature Reserve's and the Swartkops Estuary south-east section, sustainable ecolodges and conference centres. The study recommends that the Nelson Mandela Bay Municipality should organize educational xv programmes to educate local people about the significance of conserving and safeguarding the environment for future generations. These findings also posed certain challenges linked to those identified in the analysis. It is recommended that local communities are urged to take an active role in CMT and should regard CMT as a vital development sector that produces jobs. Efficacy of recapitalisation and development programme: A comparative study of the citrus farms in Amathole district
The majority of the land reform beneficiaries have remained poor, with some being worse off than before. Despite the various government initiatives to address issues of food insecurity and poverty alleviation, there has been slow progress in terms of producing enough food to ensure local and national food security. In South Africa, the Recapitalisation and Development Programme (RECAP) was initiated to alleviate the challenges faced by land redistribution and restitution recipient farmers such as inadequate post-settlement support to increase farms production output in order for them to become successful commercial farms. Several farmers have benefitted from this programme and the have been able to increase production output while others are still struggling, or worse showing zero output. These mixed outcomes call into question the efficacy of RECAP and the need to understand why some farmers are successful while others are not. This study adopted a qualitative comparative case study approach of six RECAP beneficiary citrus farms located in the Amathole District of the Eastern Cape as well as their respective strategic partner and mentors. The findings of the study were analysed with the employ of reflexive thematic analysis. The study found that RECAP recipient citrus producers in Amathole have experienced varying degrees of success with access to finance being the key obstacle to success. Significantly, the study found that the challenges around finance do not relate primarily to the purchase of inputs to production, as was the previous understanding, but rather relate to the securing of title deeds and having a history of receiving finance which combined allow farmers to access additional finance when needed. Personal branding as a market tool
Personal Branding is deemed as an unavoidable aspect of entrepreneurship and social acceptance. Individuals should establish, develop, maintain and take advantage of a Personal Brand. The benefits of a Personal Brand should be embraced and used to market themselves and highlight their uniqueness. The aim is to investigate the relationship between Professional Life, Social Media Presence, Values, Competencies, Communication as the independent variables and Personal Branding as the dependent variable. It was concluded that there are important factors that positively influence Interpersonal Personal Brand as well as Intellectual Personal Brand, which are the two components that make up Personal Brand. There is limited research on the factors that influence Personal Branding in South Africa. Through literature review, factors were identified and used to form a conceptual model. This model was tested to measure their relationship with Personal Branding. An online questionnaire was distributed to MBA students, family and friends who made up the study’s respondents. The results obtained after conducting correlation and ChiSquare tests were used to determine the relationship between the identified factors with Personal Branding. This study explores how individuals can use Personal Branding as a marketing tool. These individuals include those who want to create or further expand their current Personal Brand and use it as a marketing tool. This treatise explored the factors needed to be considered for Personal Branding a marketing tool for individuals. Personal Branding can be used at the beginning of a career, during a transition of a career or even when wanting to explore a new career. Personal Brand was initially considered the only factor. However, this study concludes that Personal Branding is made of up of two components: Interpersonal Personal Branding as well as Intellectual Personal Branding. The findings concluded that independent factors, Professional Life, Values, Competencies and Communication all have a positive significant influence on Personal Branding as a whole, as well as the components it is made up of, Interpersonal Personal Branding and Intellectual Personal Branding. The independent factor Social Media Presence positively influences Interpersonal Personal Branding only. v This study makes theoretical contributions by broadening the definition of Personal Branding. The study defines the research methodologies for Personal Branding which could be used for future studies. The Associative Network Theory in relevance to Personal Branding is explored. The practical contribution of this research includes a hypothesised model which was tested. This study should be repeated to include a bigger sample size that is more equally distributed within South Africa. Additionally, for future studies, it would be recommended to incorporate the development of Personal Branding as an independent factor to measure its effect on Personal Branding. Assessing the impact of COVID-19 on South African organisation’s way of working: A Black Management Forum Perspective
The aim of this study was to examine impact of Covid-19 on organisation’s work environment in South Africa, based on Black Management Forum’s views. The outbreak of Covid-19 led to disruption on organisation’s ways of working, this was due to lockdown rules and regulations, restrictions imposed by Government to curb the spread of the virus. The study used quantitative method approach; online survey questionnaire was distributed to recruited Black Management Forum (BMF) members who were willing to voluntarily participate via an email and WhatsApp platform. These members were sampled using convenience and snowball non-probability sampling techniques. The concepts of the study were scientifically hypothesised to determine whether such associations exist within the context of the work environment and productivity within the different corporate sector of industries in South Africa. The study findings confirmed that most organisations were impacted, and they changed their ways of work by implementing mostly work partly from home (WPFH) hybrid work model. The study findings also revealed that, workload was more during the pandemic than before, as results of changed work environment. Other finding from the study indicated that most employees adapted well to changes, understood the need for change and they performed well. Most employees also felt more productive and well supported and respected by managers. Service sectors were more impacted than other sectors and they showed more productivity as well. Most respondents felt that the adopted ways of working would remain post-pandemic and that they prefer this hybrid model of working because it gave them an opportunity to manage their own time. vii The research is significant as it critically examined how a pandemic can create organisational changes and the extent to which organisations must adjust.
