    Method Development and Validation of Rifampicin by Using RP-HPLC Method in Pharmaceutical Formulations

    A rapid and sensitive RP-HPLC method with UV detection (219nm) for routine analysis of Rifampicin in a pharmaceutical formulation was developed. Chromatography was performed with a mobile phase containing a methanol of assay (99.8%) with flow rate of 0.9ml/min. Quantitation was accomplished with an internal standard method. The procedure was validated for linearity (correlation coefficient = 0.9957), accuracy and limit of detection (LOD) intraday precision. To test validation of the Rifampicin three factors were considered as linearity, precision, LOD where mobile phase, flowrate and pressure are respectively selected as methanol, 0.9ml/min, 1200 pascals. To ensure precise intraday measurements, the variables taken into account were the analyst and the equipment. The RSD value (0.20%) indicated a good precision of the analytical method. The proposed method was simple; highly sensitive, precise, accurate and retention time less than 3 min indicating that the method is useful for routine quality control

    Method Development and Validation of Lorazepam by Using RP-HPLC in Pharmaceutical Formulation

    High pressure liquid chromatography (RP-HPLC) is a column chromatographic technique employing high pressure pump to pass both mobile phase and sample mixture through stationary phase column and perform efficient separation. A C-18 column with mobile phase containing Acetonitrile: methanol (80: 20) was used. The flow rate was 1.0ml/min. and were monitored at 210 nm. The retention time for Lorazepam was 1.3 min. The method was validated for linearity, accuracy, precision, limit of detection. The Mean percent recovery of Lorazepam from tablet formulation was found to be 94.80 %

    Prakriti is an integral part of Ayurveda, which is an eternal science based on absolute principles. It is essential for the selection and formation of all factors for which a person will interact from conception to death, such as lifestyle, diet, etc. It represents the character of the body in relation to the Dosha and is determined at the moment of conception based on the predominant Dosha. This character does not change throughout a person\u27s lifetime and is responsible for their physical and mental attributes. Tridosha says there are seven types of Prakriti. People with certain Prakriti have different bodies in terms of how they work, how they act, how they react to things in their environment, how likely they are to get diseases, and so on. Prakriti identifies how an individual reacts to environmental conditions, lifestyle changes, and drugs, and how susceptible they are to different diseases. It\u27s one of the first ideas in preventive and personalized (or genomic) medicine. To figure out how susceptible an individual is to different diseases, what drugs to use, how much to take, how the drug reacts to the body, how the disease will progress, and how to prevent it, first Prakriti is done, then Vikriti is done.     A Controlled Evaluation of Effect of Madhumeha Nashini Gutika in Sthula Prameha with Reference to Type II Diabetes Mellitus

    Ayurveda the science of life is eternal which deals with the healthy as well as diseased individual. Prameha described in Ayurvedic literature is very much similar to the diabetes mellitus of modern medicine with the abrupt change in the modern life style the prevalence of all types of diabetes is on the rise in the world’s population. Charak has mentioned that Sthula Pramehi where Kleda Meda and Kapha are vitiated Apatarpan should be given. Properties of Madhumeha Nashini Gutika prompted us to select the drug in the management of Sthula Prameha. Methodology- The present study is open labelled randomized control trail. The study duration was 12 weeks from the day of recruitment of patients. Group A (trail group) comprised of 20 patients to whom Madhumeha Nashini Gutika 500mg BID was given while in Group B (control group) also comprised of 20 patients treated with tablet metformin in a dose of 500mg BID after food along with drinking water. All the patients were strictly examined according to the criteria of diagnosis having BMI more than 25 were included in the study randomly. Analysis- The outcome measures of this study was to evaluate the effect of therapy on the symptoms of Prameha in terms of % of relief, on physical characters of body such as wt in kg and BMI and biochemical markers such as blood sugar level, HbA1c before and after treatment in both the groups. Results and Conclusion- Overall % of relief in treated group was 73.61% which was more than control group where it was found to be 58.59%. Reduction in weight of patients of treated group was highly significant as t was 8.09 (p<0.001) as compared to control group where t was 0.2525 (p>0.1). Highly significant reduction in BMI values was also noted in treated group as t was 8.83 (p<0.001) as compared to control group where t=0.1625 (p>0.1) so we came at the conclusion that Madhumeha Nashini Gutika might be working on Bahudrava Shleshma and Bahu Abadhdha Meda dhatu

    An Overview of Patients Visited at Department of Stri-Roga and Prasuti Tantra at District Ayurveda Health Center Kathmandu

    District Ayurveda Health Center, located in Sinamangal, deals with different branches such as Stri Prasuti, Kayachikitsa, Panchakarma, Physiotherapy, Acupuncture, and Acupressure. The majority of the patients who visited DAHC were females. In this retrospective analysis, Gynec patients who underwent treatment from Shrawan 2079 to Ashad 2080 were studied. Objective: The major objective of this study was to determine the treated disease pattern and the frequency of gynec patients who benefited from treatments and services provided in the OPD of DAHC. Material and methods: The study was analytically retrospective in nature, where an analysis of registered patient case records of Stri roga and Prasuti roga from the OPD of DAHC during the period 079-080 B.S. was conducted. Result: Despite a good number of patients visiting the OPD of DAHC and patients with different gynecological diseases being treated with Ayurvedic medications, Sthanika chikitsa (local in situ procedures), Beejasanskara (pre-conceptional care), Garbhasansakara (antenatal care), and Sutika (post-natal care), there is a lack of good publicity and awareness in the general population about the wide range of treatments available for various diseases at DAHC. The follow-up cases were also good. Conclusion: The study showed that DAHC provided services for various gynecological diseases, Sthanika chikitsa (local in situ procedures), Beejasanskara (pre-conceptional care), Garbhasansakara (antenatal care), and Sutika (post-natal care) facilities, benefiting females. Hence, improving the services and conducting awareness programs can provide better Ayurvedic health services to many needy people at the district level

    Stability Indicating RP-HPLC Method Development and Validation for the Estimation of Ondansetron in Bulk and their Pharmaceutical Dosage Form

    A simple, rapid, precise and accurate stability indicating RP-HPLC method was developed and validated for the estimation of Ondansetron in bulk drug and pharmaceutical dosage form. A Phenomenex C18 (150 mm × 4.6 mm I.D., 5 µm particle size) column was used as stationary phase with mobile phase consisting of 0.1% Formic acid (pH 4.25):Acetonitrile in the ratio of 50:50 V/V. The flow rate was maintained at 0.6 mL/min and effluents was monitored at 250 nm. The retention time was 2.91 min. The linearity of the method was observed in the concentration range of 5-25 µg/mL with correlation coefficient of 0.999. The method developed was validated for linearity, precision, accuracy, system suitability and forced degradation studies like acidic, alkaline, oxidative and hydrolytic stress conditions were performed as per ICH guidelines. The results obtained in the study were within the acceptable limits and hence this method can be used for the estimation of Ondansetron in pure drug and pharmaceutical dosage form

    Prediction of Linearity and Non-Linearity in Pharmaceutical Optimization Studies with Python

    Novel simple user-friendly python programme was developed to predict linearity and non-linearity in pharmaceutical optimization. Optimization is the process of obtaining optimum formulation. There are independent and dependent variables in optimization techniques regarding pharmaceutical formulations. The number of levels of independent factor is usually selected based on the linear/ non-linear relationship existing between the dependent and independent variable. The programme is run after entering the independent and dependent variables. The program is used to detect the best fitted model based on the observed correlation between dependent and independent factors, to predict the outcome against the input (independent variables). The program output is the regression coefficients, regression equations, predicted dependent variable and standard error of point estimate. The model offering the low error of point estimate is assumed to be the best fitted model for the given data. The model is applied successfully for both linear and non-linear data

    A Clinical Study to Assess the Effectiveness of Belladonna in Various Potencies in Primary Dysmenorrhoea of Adolescent Girls

    Primary Dysmenorrhea is literally termed as Painful menstruation. Most of the women do suffer with painful menses which intervenes in their day to day activities. cause of pain due to excessive vasopressin, leukotrienes, prostaglandin, myometrial ischemia, uterine malformation. Dysmenorrhea may present with pain in abdomen before or during menses, associated with nausea, tachycardia, irritability, fatigue, and headache. In this study 30 cases presenting with complaints of primary dysmenorrhoea is selected with the symptoms of Belladonna is randomly selected. Potency is selected according to susceptibility and improved was accessed by intensity of symptoms before and after the treatment. Out of 30 cases 22 had shown marked improvement and 7 cases had moderate improvement & 1case had no improvement. This study determines the effectiveness of Belladonna in the treatment of Primary dysmenorrhoea in adolescent girls
