National audienceDans le contexte de la diffusion d’And Just Like That, spin-off de la série Sex and the City, cet ouvrage propose d’explorer les héritages contemporains de la série culte pour ses téléspectatrices américaines et françaises, qu’elles soient désormais trentenaires, quarantenaires ou quinquagénaires. Il s’interroge sur la manière dont Sex and the City a influencé notre rapport au sexe, à l’amour, à l’amitié, à la mode et au féminisme, et sur la trace indélébile que la série a laissé dans la culture populaire et dans le paysage audiovisuel contemporain. Le livre revient sur le contexte de création de la série et sur les raisons de son succès, avant de s’intéresser à la manière dont le texte peut être reçu et relu aujourd’hui, aux Etats-Unis et en France, à l’aune des évolutions socio-politiques de ces vingt dernières années. This thesis explores the info-communicational dynamics of food influencers on online social networks (SNs) through a critical constructivist approach. SNs are analyzed as spaces where power, identity per-formativity, and normative production are intertwined within a complex sociotechnical framework. Food influencers are not limited to a prescriptive function; they contribute to the construction of a collective imaginary around food.The study is structured around three main axes: 1) Analysis of influencers practices and discourse: fo-cusing on their identity strategies and the tensions between authenticity and economic stakes. 2) Explo-ration of values conveyed by food: examining food as a vehicle for social, identity-based, and ethical norms, using critical theories. 3) Study of young audiences’ perceptions and practices: analyzing their reception of food-related content, which reveals active engagement, ranging from adherence to critical distancing.The results show that platforms play a structuring role in the activity of influencers. These platforms tend to steer their content through algorithmic and economic mechanisms. Moreover, young audiences negotiate circulating food and identity norms, demonstrating an ability to integrate, critique, or reinterpret these discourses. This research highlights the processes of co-construction of meanings on SNs and questions the societal and ethical implications of these new forms of food mediation.Cette thèse explore les dynamiques info-communicationnelles des influenceurs alimentaires sur les réseaux socionumériques (RSN) à travers une approche constructiviste critique. Les RSN sont analysés comme des espaces où s’articulent pouvoir, performativité identitaire et production normative, dans un cadre sociotechnique complexe. Les influenceurs alimentaires ne se limitent pas à une fonction prescriptive, ils participent à la construction d’un imaginaire collectif autour de l’alimentation.L’étude s’articule autour de trois axes : (1) l’analyse des pratiques et des discours des influenceurs, se penche sur leurs stratégies identitaires et les tensions entre authenticité et enjeux économiques ; (2) l’exploration des valeurs véhiculées par l’alimentation comme vecteur de normes sociales, identitaires et éthiques, mobilisant les théories critiques ; et (3) l’étude des perceptions et pratiques des jeunes publics face à ces contenus alimentaires qui révèle une réception active, entre adhésion et distance cri-tique.Les résultats montrent que les plateformes jouent un rôle structurant dans l’activité des influenceurs. Ces dernières tendent à orienter leurs contenus par des mécanismes algorithmiques et économiques. Par ailleurs, les jeunes publics négocient les normes alimentaires et identitaires circulantes. Ils expriment une capacité à intégrer, critiquer ou réinterpréter ces discours. Cette recherche met en évidence les processus de co-construction des significations sur les RSN et elle interroge les implications sociétales et éthiques de ces nouvelles formes de médiation alimentaire

Role of the lingual GLP-1 system in sweet taste detection
International audienceSweet taste perception is mediated in the gustatory system by the TAS1R2/TAS1R3 receptor, that responds to a wide range of sweet compounds. Recently, glucose sensors known in other parts of the body to be involved in glucose detection and glucose metabolism have been located in the taste system, but their role is still not clear. The aim of this study was thus to investigate the role of one of them, glucagon-like peptide 1 (GLP-1), in modulating sweet taste sensitivity in mice. Virogenetic silencing approaches were employed to specifically reduce the expression of the GLP-1 receptor (GLP-1R) in the tongue of adult C57BL/6J mice. Behavioral responses were then assessed using brief-access taste tests in a gustometer. A series of different sweet solutions was presented to the mouse in 10-second trials in a randomized order within a block across a 20-minute session. Three tests were performed: glucose vs fructose (GvF) solutions at three concentrations (0.316 M, 0.56 M, 1.1 M); six concentrations of glucose alone; six concentrations of fructose alone. In the GvF test, a significant difference in licking responses between glucose and fructose was observed at the intermediate 0.56M concentration in GLP-1R knockdown (KD) mice. In the fructose-only test, GLP-1R KD mice exhibited increased licking responses to 0.56 M fructose compared to control mice, while a tendency to a decrease in licking behavior was observed in the glucose-only test at 0.56 M. Virogenetic silencing of GLP-1R in the taste system appears to modulate the ability of mice to perceive sugars at an intermediate concentration, suggesting a role for the GLP-1/GLP-1R system in modulating sweet taste sensitivity. Further studies will assess the impact of GLP-1/GLP-1R signaling in TAS1R2/TAS1R3 knockout models to better understand its role in taste detection without the canonical sweet information. In addition, the potential involvement of lingual GLP-1 in systemic glucose metabolism will also be investigated
Heterozygous CELF4 variants in the N-term region crucial for the RNA-binding activity lead to neurodevelopmental disorder and obesity
International audienceRNA-binding proteins play a key role in post-transcriptional events, such as mRNA splicing, transport, stability, translation and decay. Dysregulation of RNA life can have dramatic consequences. CELF RNA-binding proteins appear to be essential during embryo development. In this study, we identified 15 patients with heterozygous missense or loss-of-function variants in the CELF4 gene by exome or genome sequencing. All variants affecting the N-terminus of the protein are essential and sufficient for the RNA-binding and splicing activity or RRM domains. Most patients presented with neurodevelopmental disorders including global developmental delay/intellectual disability (11/14), seizures (9/15) and overweight/obesity (10/14) that began in childhood. Clinical features are similar to the reported celf4-mouse mutant phenotype. This study highlights the essential role of CELF4 in development and its involvement as a novel etiology of neurodevelopmental disorders with obesity
