Size and density control of skyrmions with picometer CoFeB thickness variations—observation of zero-field skyrmions and skyrmion merging
The controlled formation and adjustment of size and density of magnetic skyrmions in Ta/CoFeB/MgO trilayers with low Dzyaloshinskii–Moriya interaction is demonstrated. Close to the out-of-plane to in-plane magnetic spin reorientation transition, we find that small energy contributions enable skyrmion formation in a narrow window of 20 pm in CoFeB thickness. Zero-field stable skyrmions are established with proper magnetic field initialization within a 10 pm CoFeB thickness range. Using magneto-optical imaging with quantitative image processing, variations in skyrmion distribution and diameter are analyzed quantitatively, the latter for sizes well below the optical resolution limit. We demonstrate the controlled merging of individual skyrmions. The overall demonstrated degree of comprehension of skyrmion control aids to the development of envisioned skyrmion based magnetic memory devices
Welche Eigenschaften bestimmen die Funktion des mitochondrialen Glutaredoxin 2 im kontrollierten Zelltod - seine Redoxaktivität oder eisenabhängige Prozesse?
Das humane Glutaredoxin 2a (Grx2a) gehört zur Proteinfamilie der Thioredoxine. Es ist
eine Oxidoreduktase mit der Fähigkeit zur Katalyse von Redoxreaktionen sowie zur Komplexierung eines [2Fe-2S]-Clusters. Grx2a besitzt ein Cys-Ser-Tyr-Cys-Motiv im aktiven Zentrum, wobei insbesondere das N-terminale Cystein essentiell für die enzymatische Aktivität
im Mono- und Dithiolreaktionsmechanismus ist. Der [2Fe-2S]-Cluster wird im enzymatisch
inaktiven Grx2a-Holoproteinkomplex durch die N-terminalen Cysteine des aktiven Zentrums
von zwei Grx2-Monomeren und zwei nicht-kovalent gebundene GSH-Moleküle komplexiert.
Unter oxidierenden Bedingungen und bei Abnahme der Menge an reduziertem GSH kann der
[2Fe-2S]-Cluster nicht mehr komplexiert werden. Der Grx2a-Holoproteinkomplex dissoziiert
in die enzymatisch aktive Apoproteinform, bei der das N-terminale Cystein des aktiven Zentrums für Reaktionen zur Verfügung steht. Auf diese Weise fungiert das Grx2a als Redoxsensor.
Mutationen innerhalb und außerhalb des aktiven Zentrums des Grx2a sorgen für eine Veränderung der Proteinstruktur, der Redoxaktivität sowie der Fähigkeit zur Bindung des [2Fe-2S]-
Clusters. Eine Überexpression von Grx2 in Tumorzellen führte zu einer Resistenz dieser Zellen gegenüber doxorubicinvermittelter Zelltoxizität. Die hierfür verantwortlichen Mechanismen sind bisher nicht sicher geklärt. Ziel dieser Arbeit war daher die Schaffung von Voraussetzungen, um den Einfluss der Eigenschaften des Grx2a auf dessen Funktion im kontrollierten Zelltod bzw. in der Zellprotektion zu untersuchen. Das Hauptaugenmerk lag hierbei auf
der Oxidoreduktaseaktivität des Grx2a, der Fähigkeit zur Katalyse von Redoxreaktionen im
Mono- und Dithiolreaktionsmechanismus und der Fähigkeit zur Komplexierung des [2Fe-2S]-
Clusters. Hierfür wurden im Rahmen dieser Arbeit mittels Mutagenese-PCR Plasmide hergestellt, welche für Grx2a-Mutanten mit unterschiedlicher enzymatischer Aktivität und Fähigkeit zur Komplexierung des [2Fe-2S]-Clusters kodieren.
Die Mutation des N-terminalen Cysteins des aktiven Zentrums zu Serin (C77S) führt zu einem enzymatisch inaktiven Grx2a, welches zudem die Fähigkeit zur Bindung des [2Fe-2S]-
Clusters verliert. Die Veränderung des Serins des aktiven Zentrums zu Prolin (S78P) verursacht den Verlust der Fähigkeit zur [2Fe-2S]-Clusterbindung und somit auch der Bildung des
enzymatisch inaktiven Grx2a-Holoproteinkomplexes. Hieraus resultiert eine gesteigerte Oxidoreduktaseaktivität. Bei Mutation des C-terminalen Cysteins des aktiven Zentrums zu Serin
(C80S) sind nur noch Reaktionen des Monothiolmechanismus möglich, sodass sich eine reduzierte enzymatische Gesamtaktivität des Grx2a zeigt. Die Fähigkeit zur [2Fe-2S]-Clusterkomplexierung bleibt dabei unbeeinträchtigt. Die Mutation eines Threonins außerhalb des aktiven
Zentrums zu Arginin (T135R) führt zu einer verminderten Fähigkeit zur Bindung des
[2Fe-2S]-Clusters und zu einer verringerten Stabilität des Grx2a-Holoproteinkomplexes. Hieraus resultiert wiederum eine erhöhte enzymatische Aktivität. Die Plasmide pEGFP-N1-
Grx2aC77S, pEGFP-N1-Grx2aS78P, pEGFP-N1-Grx2aC80S und pEGFP-N1-Grx2aT135R
konnten erfolgreich etabliert werden. Es erfolgte die Transfektion der Plasmide mittels Elektroporation in Panc-1-Zellen, welche kein endogenes Grx2a exprimieren. Nach Behandlung
mit Doxorubicin und anschließenden Zellviabilitätstests ergab sich der Verdacht, dass der
Verlust der Fähigkeit zur [2Fe-2S]-Clusterkomplexierung bei erhaltener Oxidoreduktaseaktivität mit der zellprotektiven Funktion des Grx2a assoziiert ist. Die Ergebnisse ließen sich jedoch nicht reproduzieren.
Um die Plasmide auch in Zellen mit endogener Grx2a-Expression sinnvoll verwenden zu
können, musste zunächst die Herunterregulierung dieses zelleigenen Grx2a mittels siRNA erfolgen (Knockdown). Damit die durch die Plasmide kodierten Grx2a-Mutanten nicht ebenfalls von dieser Herunterregulierung betroffen sind, war die Schaffung siRNA-resistenter
Plasmide notwendig. Durch Einbringen von stillen Punktmutationen im Bereich der siRNA-Bindungssequenz des Grx2a wurde dies erreicht. Die siRNA-resistenten Plasmide pEGFP-N1-
Grx2a-siRNAres, pEGFP-N1-Grx2aC77S-siRNAres, pEGFP-N1-Grx2aS78P-siRNAres und
pEGFP-N1-Grx2aC80S-siRNAres konnten erfolgreich etabliert werden. Bei dem Plasmid
pEGFP-N1-Grx2aT135R befindet sich die eingebrachte Mutation (T135R) im Bereich der
siRNA-Bindungssequenz, sodass hierdurch bereits eine siRNA-Resistenz erzielt wird. Mittels
spezifischer siRNA erfolgte die Herunterregulierung des endogenen Grx2 in HeLa-Zellen sowie die Transfektion der siRNA-resistenten Plasmide mittels Elektroporation. Erste Ergebnisse nach Behandlung mit Doxorubicin und anschließenden Zellviabilitätstests deuten ebenfalls
darauf hin, dass für die zellprotektive Funktion des Grx2a der Dithiolreaktionsmechanismus
von Bedeutung ist. Zudem scheint der Verlust der Fähigkeit zur [2Fe-2S]-Clusterkomplexierung bei erhaltener oder sogar erhöhter Oxidoreduktaseaktivität eine Rolle in der zellprotektiven Funktion des Grx2a zu spielen. Die genaue Evaluation bedarf jedoch weiterführender Untersuchungen.Human glutaredoxin 2a (Grx2a) is a member of the thioredoxin family of proteins. It is an
oxidoreductase with the ability to catalyze redox reactions and to complex an [2Fe-2S] cluster. Grx2a contains an active site with a Cys-Ser-Tyr-Cys motif in which especially the N-terminal cysteinyl residue is essential for the enzymatic activity in the mono- and dithiol reaction mechanism. In the enzymatic inactive holo Grx2a the [2Fe-2S] cluster is complexed by the
N-terminal active site cysteines of two Grx2a monomers and two non-covalently bound molecules of GSH. Under oxidizing conditions and decreasing amounts of reduced GSH, the
[2Fe-2S] cluster complexation becomes insufficient. This leads to a dissociation of the holo
Grx2a into its active apo form, where the N-terminal cyteinyl residue is available for enzymatic reactions. This allows Grx2a to function as a redox sensor.
Mutations in- and outside the active site cause changes in the Grx2a protein structure, its
oxidoreductase activity and the ability to complex the [2Fe-2S] cluster. Overexpression of
Grx2 in tumor cells led to an increased resistance of these cells towards doxorubicin-induced
cytotoxicity. The underlying mechanisms are still not fully understood. Aim of this thesis was
to establish the requirements for further investigastions on the influence of the characteristics of Grx2a and its function in controlled cell death and cell protection. The main focus was the Grx2a oxidoreductase activity, the catalysis of redox reactions in the mono- and dithiol reaction mechanism and the ability to complex the [2Fe-2S] cluster. Therefore mutagenesis PCRs were performed to produce plasmids which encode for Grx2a mutants with differences in their enzymatic activity and their ability to complex the iron-sulfur cluster.
The mutation of the N-terminal active site cysteine to serine (C77S) leads to an enzymatically inactive Grx2a without the ability to complex the [2Fe-2S] cluster. With the change of
the acitve site serine to proline (S78P) the Grx2a loses its ability to coordinate the [2Fe-2S]
cluster as well as the ability to form the enzymatic inactive holo Grx2a. This results in an increased oxidoreductase activity. The mutation of the C-terminal active site cysteinyl residue
to serine (C80S) leads to a Grx2a protein with the ability to only catalyze reactions in the monothiol reaction mechanism and therefore results in a decreased overall enzymatic activity.
The ability to complex the [2Fe-2S] cluster remains unaffected. Changing a threonine outside
the active site to arginine (T135R) leads to a decreased [2Fe-2S] cluster binding and a less
stable holo Grx2a. This again results in an increased enzymatic activity. The plasmids
pEGFP-N1-Grx2aC77S, pEGFP-N1-Grx2aS78P, pEGFP-N1-Grx2aC80S and pEGFP-N1-Grx2aT135R were established successfully. Via electroporation, these plasmids were transfected into Panc-1 cells which lack endogenous Grx2a. After treatment with doxorubicin and
consecutive cell viability assays, we suspected that the loss of the ability to complex the
[2Fe-2S] cluster but preserved oxidoreductase activity could be associated with the cell protective function of Grx2a. However, the results could not be reproduced.
To use the established plasmids in cells with endogenous Grx2a expression, a siRNA-mediated knockdown of this endogenous Grx2a was necessary. To protect the introduced Grx2a
mutants from being affected by the siRNA-mediated knockdown we needed to create a resistant form towards siRNA. This was achieved by the insertion of silent point mutations into
the siRNA-binding sequence of Grx2a. The siRNA-resistant plasmids pEGFP-N1-Grx2a-siRNAres, pEGFP-N1-Grx2aC77S-siRNAres, pEGFP-N1-Grx2aS78P-siRNAres and pEGFP-N1-Grx2aC80S-siRNAres were established successfully. The inserted point mutation (T135R)
of the plasmid pEGFP-N1-Grx2aT135R is located in the siRNA-binding sequence of Grx2
and thereby a resistance towards siRNA was already achieved. In HeLa cells a siRNA-mediated knockdown of the endogenous Grx2 and the transfection of the siGrx2-resistant plasmids
via electroporation was performed. First results after doxorubicin treatment and consecutive
cell viability assays suggest, that Grx2a's ability to catalyze the dithiol reaction mechanism
and an increased oxidoreductase activity are favorable for the cell protective function. However, especially the loss of the ability to complex the [2Fe-2S] cluster but preserved oxidoreductase activity seems to be important for the cell protective function of Grx2a. Nevertheless,
further investigations are needed for confirmation of these findings
Definition von altersabhängigen Referenzwerten für den Durchmesser des Ductus hepatocholedochus und des Ductus pancreaticus in der Magnetresonanz-Cholangiopankreatikographie aus einer populationsbasierten Querschnitts-Kohortenstudie
Bis dato gibt es keine eindeutige Literatur zur Definition der physiologischen Ganggrößen und
deren Einflussfaktoren im pankreatikobiliären System bei asymptomatischen Personen.
Trotzdem wird der Gangdurchmesser des PD und DHC im klinischen Alltag ständig für
diagnostische Zwecke analysiert, was zu einer Herausforderung bei asymptomatischen
Patienten mit erweiterten Gängen führt. Ziel dieser populationsbasierten Studie war es, neue
Referenzwerte der Durchmesser der Gänge bei asymptomatischen Probanden aus der
Normalbevölkerung mit Hilfe der MRCP zu definieren. Mögliche Einflussfaktoren auf das
Gangsystem, wie z.B. das Alter, CCE, BMI, das Geschlecht, Laborwerte oder Vorerkrankungen
sollten mit dieser Studie identifiziert und berücksichtigt werden. Es wurden ausschließlich
gesunde Probanden aus der Normalbevölkerung untersucht. Die Studienkohorte bestand aus
1.018 Probanden im Alter von 21–89 Jahren, die im Rahmen der SHIP-Studie rekrutiert wurden,
bei denen Durchmesser von Gallen- und Pankreasgang analysiert wurden. Weiterhin wurde
das Verhalten der Gänge nach Sekretingabe untersucht. Ein Großteil der Probanden wurden
bei einem fünf Jahres Follow-up noch einmal untersucht. Der PD wurde im Kopfbereich und
der DHC nach der Einmündung des Ductus cysticus vermessen. Es wurde festgestellt, dass sich
beide Gänge im Laufe des Lebens zunehmend erweitern. Zudem überschritten viele
Studienteilnehmer die aktuell geltenden Grenzwerte der Durchmesser des DHC bzw. PD,
insbesondere nach einer CCE. Der DHC-Durchmesser lag bei asymptomatischen Probanden
vor Sekretin-Injektion im Median bei 5,30 mm (maximal bis zu 19,2 mm), nach Sekretingabe
bei 5,21 mm (maximal bis zu 16,9 mm). Der PD-Durchmesser lag vor Sekretingabe bei 1,84 mm
(maximal bis zu 6,65 mm) und nach Sekretingabe bei 2,04 mm (maximal bis zu 6,50 mm).
Aufgrund der zahlreichen Referenzwertüberschreitungen wurden unter Berücksichtigung des
Alters mit der quantilen Regression neue Grenzwerte der Durchmesser für beide Gänge
abgeleitet. Daher sollte bei asymptomatischen Patienten ≤ 65 Jahren ein DHC-Durchmessern
65 Jahren ein DHC-
Durchmesser < 11 mm und PD-Durchmesser von < 4 mm als physiologisch betrachtet werden
und diese Personen, die diese Grenzwerte einhalten, sollten somit keine weitere Diagnostik
erhalten.
Durch die hohe Anzahl an Studienteilnehmern, das besonders ausgewogene Verhältnis
zwischen männlichen und weiblichen Studienteilnehmern, die sehr breite Altersstreuung, die
eindeutig standardisierte Untersuchung durch verifizierte Untersucher an einem MRT und die
ausführlichen weiteren Informationen über die Probanden, die vorlagen, erscheinen diese
Ergebnisse als besonders gut gesichert. Die vorliegenden Ergebnisse leisten sowohl einen
wichtigen Beitrag zur Definition der physiologischen Gangweiten als auch zur Klärung der
Einflussfaktoren auf die Durchmesser im pankreatikobiliären Gangsystem.
Diese Ergebnisse werden helfen, übermäßige Diagnostik zu vermeiden und damit direkt
Kosten im Gesundheitswesen zu sparen
Psychische Belastung und Geburt
Zusammenfassung
43,6 % der Schwangeren in Deutschland leiden unter mindestens einer psychischen Erkrankung, am häufigsten sind Depressionen und Ängste. Das Risiko für eine verlängerte Geburtsdauer, einen pathologischen Geburtsverlauf und Geburtsmodus ist erhöht. Durch eine individualisierte psychotherapeutische Intervention soll ein positiver Einfluss auf den Verlauf der Geburt erreicht werden.
In dem Präventionsnetzwerk Vorpommern: Psychosoziale und neurobiologische Gesundheit von Schwangeren und jungen Müttern (PriVileG-M), einer randomisierten kontrollierten Studie wurden die gesunden Schwangeren der gesunden Gruppe (GG) und die psychisch belasteten Schwangeren einer Interventions- (PBI) oder einer Kontrollgruppe (PBK) zugeteilt. Die Interventionsgruppe erhielt eine individualisierte Psychotherapie, die pränatal, ab ca. der 25. Schwangerschaftswoche begann. Bis zum Zeitpunkt der Geburt erfolgten ca. 7-10 Interventionen mit einer Frequenz von ein bis zwei Wochen zwischen jeder psychotherapeutischen Intervention. Im Geburtsfollow-up wurden die Geburtsparameter dokumentiert.
Die Ergebnisse zeigten hypothesenkonform, dass die gestressten bzw. psychisch belasteten Frauen eine längere Geburtsdauer und mehr pathologische Geburtsausgänge (Sectio, Forceps, Vakuumextraktion) haben. Insbesondere wurde der Verlauf der Geburt durch Angstsymptome und erlebte Kindheitstraumata, sogar unabhängig von der Gruppenzuteilung beeinflusst. Die individualisierte psychotherapeutische Intervention mit ihren 7- 10 Sitzungen bewirkte keine messbaren, signifikanten Veränderungen in der Geburtsdauer und Geburtsmodus in der Interventionsgruppe gegenüber der psychisch belasten Kontrollgruppe oder der gesunden Gruppe (GG).
Die möglichen Effekte der Psychotherapie könnten aufgrund der noch zu kleinen Gruppengröße statistisch nicht deutlich geworden sein. Denkbar ist auch, dass der Beginn in der 25. Schwangerschaftswoche zu spät gewählt war oder die Anzahl der Interventionen in der Schwangerschaft. So bleibt weiterhin eine interdisziplinäre Forschung notwendig, welche Art der Behandlung, wie oft und in welcher Schwangerschaftswoche beginnend eine psychische Stabilisierung erreichen könnte, um eine signifikante Verringerung der pathologischen Geburtsverläufe, insbesondere der Kaiserschnittrate, eine durchschnittliche Geburtsdauer und eine positive Geburtserfahrung zu erreichen.
Dies erfordert ein frühzeitiges standardisiertes Screening psychischer Belastungen, Kindheitstraumata und vorausgegangene schwierige Geburten in der gynäkologischen oder hebammengeleiteten Schwangerenvorsorge, wie es in einigen Ländern bereits zur Routine gehört.

Zerebrale Repräsentation psychopathologischer Aspekte der kraniomandibulären Dysfunktion
In einer klinischen Intervention mittels Schienentherapie mit einer Unterkieferschiene bei Patientinnen mit kraniomandibulärer Dysfunktion zeigte sich eine Reduktion des Bewegungsschmerzen und eine Veränderung der fMRT-Aktivität assoziiert mit der Trait-Angst-Reduktion.
Eine Verminderung der fMRT-Aktivität bei Okklusion in der bilateralen anterioren und
rechten posterioren Insel ging, insbesondere im ersten Zeitintervall der Intervention,
einher mit einer Verminderung der Trait-Angst. Hier zeigen sich unmittelbare Effekte
der Behandlung auf eine pathologisch erhöhte Monitoringaktivität, die mit der erhöhten
Angstmodulation einhergeht. Im späteren Verlauf scheint eher die präfrontale
Aufmerksamkeit auf die Bewegungsdurchführung vermindert zu sein.
Die zeitliche Abfolge mit zunächst einer Minderung in emotional unmittelbar
gekoppelten Prozessen des Bewegungsmonitorings gegenüber der später
einsetzenden Minderung der Aktivität bewusster präfrontaler Prozesse und den damit
einhergehenden Modulationen von Trait-Angst und Schmerz, weist auf grundsätzliche
Mechanismen der Intervention hin
In Vivo Evaluation of a Gastro-Resistant Enprotect® Capsule under Postprandial Conditions
Ready-to-fill enteric hard capsule shells are an evolving field of oral drug and nutraceutical products. Lonza Capsugel® Enprotect® capsules were recently proven to provide reliable release in the small intestine after fasted intake, but robustness against postprandial intake needed to be proven. In this study, the capsules were administered to 16 healthy young subjects after intake of a light meal. The Enprotect® capsules were labelled with 5 mg black iron oxide and 25 mg 13C3-caffeine. Magnetic Resonance Imaging was used to identify the localization and visual dispersion of the capsule filling. The salivary appearance of caffeine was considered a second independent and sensitive marker for the initial release. Whereas the fasted gastric residence time of the capsules amounted to 43 ± 32 min, it was increased to 158 ± 36 min after postprandial intake. Therefore, the mean dispersion time according to MRI and the mean caffeine appearance time were increased to 196 ± 37 min and 189 ± 37 min, respectively. But, similar to fasted administration, no capsule disintegration or leakage was observed in the stomach and 38% of the capsules disintegrated in the jejunum and 62% in the ileum. The mean dispersion time after gastric emptying and the mean caffeine appearance time after gastric emptying amounted to 38 ± 21 min and 31 ± 17 min, respectively. Both did not relevantly change compared to the fasted intake. Only the absolute dispersion time and caffeine appearance were prolonged due to the increased gastric residence and no relevant influence of the light meal was observed on the disintegration or release behavior of Enprotect® capsules after gastric emptying. The capsules also showed robust enteric properties after postprandial administration
Behavior-related health risk factors, mental disorders and mortality after 20 years in a working aged general population sample
Mortality is predicted by the sum of behavior-related health risk factors (BRFs: tobacco smoking, alcohol drinking, body overweight, and physical inactivity). We analyzed degrees and combinations of BRFs in their relation to mortality and adjusted for mental disorders. In a random sample of the general population in northern Germany aged 18–64, BRFs and mental disorders had been assessed in 1996–1997 by the Munich Composite International Diagnostic Interview. A sum score including eight ranks of the behavior-related health risk factors was built. Death and its causes were ascertained 2017–2018 using residents’ registration files and death certificates. Relations of the sum score and combinations of the BRFs at baseline with all-cause, cancer, and cardiovascular mortality 20 years later were analyzed. The sum score and combinations predicted all-cause, cardiovascular and cancer mortality. The odds ratio of the sum score was 1.38 (95% confidence interval 1.31–1.46) after adjustment for age, gender, and mental disorder. In addition to the BRFs, mood, anxiety or somatoform disorders were not related to mortality. We concluded that the sum score and combinations of behavior-related health risk factors predicted mortality, even after adjustment for mental disorders
The Greifswald Post COVID Rehabilitation Study and Research (PoCoRe)–Study Design, Characteristics and Evaluation Tools
(1) Background: COVID-19 is often associated with significant long-term symptoms and disability, i.e., the long/post-COVID syndrome (PCS). Even after presumably mild COVID-19 infections, an increasing number of patients seek medical help for these long-term sequelae, which can affect various organ systems. The pathogenesis of PCS is not yet understood. Therapy has so far been limited to symptomatic treatment. The Greifswald Post COVID Rehabilitation Study (PoCoRe) aims to follow and deeply phenotype outpatients with PCS in the long term, taking a holistic and comprehensive approach to the analysis of their symptoms, signs and biomarkers. (2) Methods: Post-COVID outpatients are screened for symptoms in different organ systems with a standardized medical history, clinical examination, various questionnaires as well as physical and cardiopulmonary function tests. In addition, biomaterials are collected for the analysis of immunomodulators, cytokines, chemokines, proteome patterns as well as specific (auto)antibodies. Patients are treated according to their individual needs, adhering to the current standard of care. PoCoRe’s overall aim is to optimize diagnostics and therapy in PCS patients
Development and Validation of a Scale to Assess Moral Disengagement in High-Carbon Behavior
The reduction of individual carbon consumption could make an important contribution to the worldwide effort to limit global warming. Based on Bandura’s theory of moral disengagement, we hypothesized that the propensity to morally disengage concerning high-carbon behaviors (e.g., eating meat or traveling by plane) is one important factor that prevents individuals from reducing their carbon footprint. To measure the propensity to morally disengage in high-carbon-related behavior contexts, a questionnaire (MD-HCB) was developed and psychometrically validated in an online study with a German sample (N = 220). Confirmatory factor analyses revealed that the final nine-item scale had a one-dimensional structure, as intended. The internal consistency of the scale was excellent (Cronbach’s α = 0.94) and the scale interpretation had predictive validity for both past low-carbon consumption behavior and the intention to engage in such behavior in the future. Correlational analyses with relevant existing instruments confirmed the construct validity of the interpretations that can be drawn from the MD-HCB, as its resulting score is related to, yet separable from, the general tendency to morally disengage and is meaningfully connected to related constructs. A pre-study with a student sample (N = 89) not only helped to identify limitations in the study design but also showed a weak predictive ability of moral competence concerning high-carbon consumption behavior and intention to change. Based on our findings, future media campaigns designed to increase people’s intention to reduce high-carbon behavior could focus on the modification of common cognitive disengagement strategies
An accessible and versatile deep learning-based sleep stage classifier
Manual sleep scoring for research purposes and for the diagnosis of sleep disorders is labor-intensive and often varies significantly between scorers, which has motivated many attempts to design automatic sleep stage classifiers. With the recent introduction of large, publicly available hand-scored polysomnographic data, and concomitant advances in machine learning methods to solve complex classification problems with supervised learning, the problem has received new attention, and a number of new classifiers that provide excellent accuracy. Most of these however have non-trivial barriers to use. We introduce the Greifswald Sleep Stage Classifier (GSSC), which is free, open source, and can be relatively easily installed and used on any moderately powered computer. In addition, the GSSC has been trained to perform well on a large variety of electrode set-ups, allowing high performance sleep staging with portable systems. The GSSC can also be readily integrated into brain-computer interfaces for real-time inference. These innovations were achieved while simultaneously reaching a level of accuracy equal to, or exceeding, recent state of the art classifiers and human experts, making the GSSC an excellent choice for researchers in need of reliable, automatic sleep staging
