    TFEB erhöht die metabolische Flexibilität von Kardiomyozyten und verbessert die Herzfunktion bei druckinduzierter Herzinsuffizienz

    In den Industrieländern zählen kardiovaskuläre Erkrankungen zu den häufigsten Todesursachen. Pathologische linksventrikuläre Hypertrophie (LVH) und Herzinsuffizienz mit reduzierter Ejektionsfraktion (HFrEF) sind mit einer erhöhten Morbidität und Mortalität der betroffenen Patienten assoziiert. Bei der pathologischen LVH kommt es zu einer inadäquaten Reaktion des Herzens (maladaptives Remodeling) auf verschiedene Stressreize, wie z.B. Drucküberlastung durch eine Aortenklappenstenose oder eine arterielle Hypertonie. Das kardiale Remodeling wird von Kardiomyozyten-Hypertrophie, interstitieller und perivaskulärer Fibrose begleitet. Damit einhergehend kommt es zu Veränderungen des kardialen Proteoms und zu einem Anstieg kardialer Stressmarker (ANF, BNP, α-Skelett-Aktin) sowie zu einer Verschiebung des Verhältnisses von α-MyHC zu β-MyHC. Unter physiologischen Bedingungen ist der Stoffwechsel des Herzens überwiegend auf die Verwertung freier Fettsäuren durch die Fettsäureoxidation (FAO) und nur in geringem Maße auf die Verwertung von Glukose durch die Glukoseoxidation (GO), Laktat, Ketonkörpern und Aminosäuren zur ATP-Generierung ausgerichtet. Unter pathologischen Bedingungen, wie bei LVH und HFrEF, kommt es zu einer metabolischen Verschiebung, die sich in einer verminderten FAO und einer erhöhten GO äußert und als metabolisches Remodeling bezeichnet wird. Untersuchungen zum Zusammenhang zwischen metabolischem und kardialem Remodeling haben gezeigt, dass das metabolische Remodeling den strukturellen Störungen (kardialem Remodeling) in den Kardiomyozyten vorausgeht. Ein Regulator metabolischer Prozesse ist der nährstoffsensitive Transkriptionsfaktor EB (TFEB). TFEB fördert die Expression von Genen, die an der FAO (PGC-1α/PPARGC1A, PPARα/PPARA), der GO (HKII/HK2, GLUT1/SLC2A1, GLUT4/SLC2A4, und IRS2/IRS2) und der Energiegewinnung durch oxidative Phosphorylierung (OXPHOS, Komplex I-V) beteiligt sind. TFEB könnte daher eine Rolle beim stressinduzierten metabolischen Remodeling des Herzens spielen. In bereits publizierten Arbeiten führte die Überexpression von TFEB in murinen Herzen unter physiologischen und pathologischen Bedingungen zu einem erhöhten kardialen Remodeling und einer reduzierten Herzfunktion. Aus diesen Erkenntnissen wurde folgende Hypothese formuliert: "Die Deletion von Tfeb im Herzen verhindert das kardiale Remodeling durch Unterdrückung des metabolischen Remodelings und führt so zu einer verbesserten Herzfunktion unter chronischer Drucküberlastung". Zur Bestätigung dieser Hypothese wurde im ersten Teil dieser Arbeit untersucht, ob eine Kardiomyozyten-spezifische Deletion von Tfeb das metabolische und kardiale Remodeling in männlichen Mäuseherzen während einer durch TAC-induzierten LVH und HFrEF beeinflusst und ob die Deletion von Tfeb zu einer Verbesserung der Herzfunktion führt. Die Untersuchungen zeigten, dass unter physiologischen Bedingungen die Kardiomyozyten-spezifische Deletion von Tfeb (TfebloxP/loxP; αMHC-CRE; cKO) im Vergleich zu Kontrollen (TfebloxP/loxP; WT) zu einer leicht verminderten Herzfunktion und zu einem kardialen und metabolischen Remodeling führt. Um den Einfluss der Tfeb-Deletion unter pathologischen Bedingungen zu untersuchen, wurde in WT- und cKO-Mäusen eine LVH bzw. HFrEF durch 21- bzw. 56tägige transversale Konstriktion der Aorta thorakalis (TAC, 27G) induziert. Schein-operierte Geschwistertiere, die derselben Prozedur unterzogen wurden, allerdings ohne Aortenkonstriktion, wurden als Kontrollen verwendet. Die 21-tägige TAC-Behandlung führte zu LVH und die 56-tägige TAC-Behandlung führte zu HFrEF sowohl bei WT- als auch bei cKO-Mäusen. Morphologische, histologische und qRT-PCR-Analysen zeigten ein erhöhtes Herzgewicht, interstitielle Fibrose und eine Zunahme der Expression kardialer Stress- und Remodeling-Gene bei LVH und HFrEF in cKO- und WT-Mäusen, respektive. Proteomanalysen der Herzen zeigten eine Hemmung der FAO und der OXPHOS in HFrEF, was auf eine Verschiebung des Metabolismus von FAO zu GO hinweist. Im LVH-Modell wurden keine Veränderungen der Herzfunktion, wenige Unterschiede im metabolischen Remodeling, jedoch ein erhöhtes kardiales Remodeling in cKO- im Vergleich zu WT-Mäusen beobachtet. Im Modell der HFrEF zeigte der cKO eine verbesserte linksventrikuläre Ejektionsfraktion, Verkürzungsfraktion und Herzzeitvolumen. Damit einhergehend zeigten die cKO Tiere eine weniger starke Zunahme der linksventrikulären Masse, kleinere Myozyten-Querschnittsflächen und ein geringeres Herzgewicht als die WT-Tiere nach TAC. Im Modell der LVH zeigten die cKO Tiere ein erhöhtes kardiales Remodeling wohingegen im Modell der HFrEF ein geringerer Anstieg von Myh7 und β-MyHC zwischen cKO- und WT-Mäusen gefunden wurde. Diese Daten zeigen, dass die cKO-Mäuse eine bessere Herzfunktion in der TAC-induzierten Drucküberlastung haben und dass eine Verminderung von TFEB im Modell der HFrEF von Vorteil sein könnte. Hinsichtlich des metabolischen Remodelings wurden keine Unterschiede zwischen cKO- und WT-Mäusen gefunden. Aufgrund der Ergebnisse des ersten Teils dieser Arbeit stellte sich die Frage, ob die Überexpression von TFEB das metabolische Remodeling bzw. die Energiehomöostase in Kardiomyozyten stärker beeinflusst als die Deletion von Tfeb. Daher wurden im zweiten Teil dieser Arbeit die Veränderungen der zellulären Atmung, der ATP-Produktionsraten und die metabolischen Veränderungen im Proteom nach Überexpression von TFEB in Kardiomyozyten untersucht. Die Überexpression von TFEB in Kardiomyozyten führte zu einer Verschiebung der metabolischen Aktivität von einer primär OXPHOS-orientierten Atmung hin zu einer stärker glykolytisch-orientierten Atmung und insgesamt zu einer erhöhten (energiereicheren) metabolischen Aktivität bzw. Atmung. Dies war mit einer höheren ATP-Produktionsrate und einer Zunahme der maximalen Atmung und der Reserve Atmungskapazität assoziiert. Die erhöhte Reserve Atmungskapazität ermöglicht es den Kardiomyozyten, besser auf einen erhöhten Energiebedarf zu reagieren und erhöht die Flexibilität der Kardiomyozyten in Stresssituationen. Das Proteom zeigte, dass die Überexpression von TFEB die Abundanz mitochondrialer Proteine erhöht und die OXPHOS aktiviert. Diese Ergebnisse zeigen den positiven Einfluss von TFEB auf den Energiemetabolismus und seinen Einfluss auf das metabolische Remodeling in Kardiomyozyten. Zusammenfassend zeigen die Ergebnisse dieser Arbeit, dass die Überexpression und Deletion von Tfeb in Kardiomyozyten in Abhängigkeit von den vorherrschenden physiologischen und pathologischen Bedingungen (LVH und HFrEF) sowohl positive als auch negative Auswirkungen auf die Energiehomöostase und die Herzfunktion haben kann. Pathological left ventricular hypertrophy (LVH) and heart failure with reduced ejection fraction (HFrEF) are associated with increased morbidity and mortality in affected patients. In pathological LVH, there is an inadequate response of the heart (maladaptive remodelling) to various stress stimuli, such as pressure overload due to aortic valve stenosis or arterial hypertension. Cardiac remodelling is accompanied by cardiomyocyte hypertrophy, interstitial and perivascular fibrosis. This is accompanied by changes in the cardiac proteome and an increase in cardiac stress markers (ANF, BNP, α-skeletal actin) as well as a shift in the ratio of α-MyHC to β-MyHC. Under physiological conditions, cardiac metabolism is predominantly directed towards the utilization of free fatty acids through fatty acid oxidation (FAO) and only to a small extent towards the utilization of glucose through glucose oxidation (GO), lactate, ketone bodies and amino acids for ATP generation. Under pathological conditions, such as in LVH and HFrEF, there is a metabolic shift that manifests as decreased FAO and increased GO and is referred to as metabolic remodelling. Studies on the relationship between metabolic and cardiac remodelling have shown that metabolic remodelling precedes structural disturbances (cardiac remodelling) in cardiomyocytes. One regulator of metabolic processes is the nutrient-sensitive transcription factor EB (TFEB). TFEB promotes the expression of genes involved in FAO (PGC-1α/PPARGC1A, PPARα/PPARA), GO (HKII/HK2, GLUT1/SLC2A1, GLUT4/SLC2A4, and IRS2/IRS2) and energy production by oxidative phosphorylation (OXPHOS, complex I-V). TFEB may therefore play a role in stress-induced metabolic remodelling of the heart. In previously published work, overexpression of TFEB in murine hearts, under physiological and pathological conditions, resulted in increased cardiac remodelling and reduced cardiac function. From these findings, the following hypothesis was formulated: "Deletion of Tfeb in the heart prevents cardiac remodelling by suppressing metabolic remodelling, leading to improved cardiac function under chronic pressure overload". To confirm this hypothesis, the first part of this work investigated whether cardiomyocyte-specific deletion of Tfeb affects metabolic and cardiac remodelling in male mouse hearts during TAC-induced LVH and HFrEF, and whether deletion of Tfeb leads to improved cardiac function. The studies showed that under physiological conditions, cardiomyocyte-specific deletion of Tfeb (TfebloxP/loxP; αMHC-CRE; cKO) leads to slightly reduced cardiac function and cardiac and metabolic remodelling compared to controls (TfebloxP/loxP; WT). To investigate the influence of Tfeb deletion under pathological conditions, LVH and HFrEF were induced in WT and cKO mice by 21- and 56-day transverse constriction of the thoracic aorta (TAC, 27G), respectively. Sham-operated siblings subjected to the same procedure but without aortic constriction were used as controls. The 21-day TAC treatment induced LVH and the 56-day TAC treatment induced HFrEF in both WT and cKO mice. Morphological, histological and qRT-PCR analyses showed increased heart weight, interstitial fibrosis and increased expression of cardiac stress and remodelling genes in LVH and HFrEF in cKO and WT mice, respectively. Proteomic analyses of the hearts showed inhibition of FAO and OXPHOS in HFrEF, indicating a shift in metabolism from FAO to GO. In the LVH model, no changes in cardiac function, few differences in metabolic remodelling, but increased cardiac remodelling were observed in cKO compared to WT mice. In the HFrEF model, the cKO showed improved left ventricular ejection fraction, fractional shortening and cardiac output. Concomitantly, the cKO animals showed less increase in left ventricular mass, smaller myocyte cross-sectional areas and lower heart weight than the WT animals after TAC. In the LVH model, the cKO animals showed increased cardiac remodelling whereas in the HFrEF model, a smaller increase in Myh7 and β-MyHC was found between cKO and WT mice. These data indicate that the cKO mice have better cardiac function in TAC-induced pressure overload and that a reduction of TFEB may be beneficial in the HFrEF model. No differences were found between cKO and WT mice with regard to metabolic remodelling. Based on the results of the first part of this work, the question arose whether overexpression of TFEB affects metabolic remodelling or energy homeostasis in cardiomyocytes more strongly than deletion of Tfeb. Therefore, in the second part of this work, the changes in cellular respiration, ATP production rates and metabolic changes in the proteome after overexpression of TFEB in cardiomyocytes were investigated. Overexpression of TFEB in cardiomyocytes resulted in a shift in metabolic activity from primarily OXPHOS-oriented respiration to more glycolytic-oriented respiration and overall increased (higher energy) metabolic activity or respiration. This was associated with a higher ATP production rate and an increase in maximal respiration and reserve respiratory capacity. The increased reserve respiratory capacity allows cardiomyocytes to respond better to increased energy demands and increases cardiomyocyte flexibility in stressful situations. The proteome showed that overexpression of TFEB increased the abundance of mitochondrial proteins and activated OXPHOS. These results demonstrate the positive influence of TFEB on energy metabolism and its impact on metabolic remodelling in cardiomyocytes. In summary, the results of this work show that overexpression and deletion of Tfeb in cardiomyocytes can have both positive and negative effects on energy homeostasis and cardiac function depending on the prevailing physiological and pathological conditions (LVH and HFrEF).     Microorganisms as drivers of methane emissions in rewetted coastal peatlands

    Peatland rewetting is a recognized nature-based solution to reduce CO2 emissions and has the potential to create carbon sinks. The necessity to reduce greenhouse gases (GHG) from both natural and from anthropogenic sources is one of the most urgent challenges of our time. Rewetting artificially drained peatlands with freshwater can induce initially high methane (CH4) emissions. Using sulfate-containing brackish water to rewet coastal peatlands may prevent these CH4 peaks. This is due to substrate usage by thermodynamically more favorable microbial metabolic processes, such as sulfate reduction, instead of methanogenesis. Together with colleagues, I investigated two peatlands with different histories of rewetting and management, located on the Baltic Sea coast in north-east Germany: A coastal fen that was rewetted with freshwater and another rewetted with brackish water. In the freshwater rewetted fen, which experienced a drought shortly before a storm surge inundated the area with brackish water, a reduction of its previously high CH4 emissions was observed. While abundances of methanogenic archaea decreased during the drought, we did not see a further reduction after the brackish water inflow. Although a large part of the CH4 emission reduction is certainly a legacy effect of the drought, the increasing abundances of sulfate-reducing bacteria (SRB) following the inflow may have caused competition with methanogens for substrate. In addition or alternatively, SRB might be involved in the anaerobic oxidation of CH4. However, we did not observe increases in CH4 oxidation or in abundances of anaerobic methanotrophs in the peat soil. This suggests that methanotrophy may have taken place in the water column above the investigated peat soil. In contrast, the brackish water rewetted fen showed relatively low CH4 emissions immediately after rewetting compared to freshwater rewetted fens. Relative to the fen’s drained state, abundances of SRB and methanogens increased after rewetting, but substrate competition and CH4 oxidation most likely limited excess CH4 emissions. However, the high CO2 emissions did not decrease as expected despite rewetting, while ecosystem respiration and thus peat decomposition was significantly reduced. We observed a severe die-back of both grassland plants and near-ditch fen vegetation following the intense inundation with brackish water. The reason for the persistent CO2 emissions after rewetting could be ecosystem respiration, which was fueled by large amounts of available labile substrate, irrespectively of peat decay. Additionally, high CO2 emissions could not be prevented due to the lack of CO2 uptake by photosynthesis. This thesis contributes to the discussion on coastal peatland rewetting and provides new ideas about the interplay between a fen’s microbiology and biogeochemistry among different spatial compartments. When aiming for GHG emission reduction after rewetting this thesis highlights that not only the water type might be important to consider, but also the frequency of brackish water input as well as the flooding intensity. In order to define and evaluate rewetting approaches for future projects, comparing rewetted peatlands according to their GHG emission development is necessary. Further, long-term monitoring and multidisciplinary research are needed to provide insight on the influence of brackish water on coastal fens beyond the first year after rewetting.Die Wiedervernässung von Mooren ist eine anerkannte naturbasierte Lösung zur Verringerung von CO2 Emissionen und hat das Potenzial, Kohlenstoffsenken zu schaffen. Die Notwendigkeit, Treibhausgase (THG) aus natürlichen, aber auch aus anthropogenen Quellen zu reduzieren, ist eine der dringendsten Herausforderungen unserer Zeit. Die Wiedervernässung künstlich entwässerter Moore mit Süßwasser kann in der Anfangsphase zu hohen Methanemissionen (CH4) führen. Die Verwendung von sulfathaltigem Brackwasser zur Wiedervernässung küstennaher Moore könnte diese anfänglich hohen CH4 Emissionen verhindern. Dies ist auf die Substratnutzung durch thermodynamisch günstigere mikrobielle Stoffwechselprozesse wie die Sulfatreduktion anstelle der Methanogenese zurückzuführen. Zusammen mit Kolleg*innen untersuchte ich zwei Niedermoore an der Ostseeküste in Nordostdeutschland mit unterschiedlicher Wiedervernässungsgeschichte und -umsetzung: Ein mit Süßwasser und ein mit Brackwasser wiedervernässtes Küstenmoor. Im wiedervernässten Süßwassermoor, das eine Dürre erlebte, bevor eine Sturmflut das Gebiet mit Brackwasser überschwemmte, wurde ein Rückgang seiner zuvor hohen CH4 Emissionen beobachtet. Während die absoluten Abundanzen der methanogenen Archaeen während der Dürre abnahmen, konnten wir nach dem Einstrom des Brackwassers keine weitere Reduktion feststellen. Obwohl ein großer Teil des Rückgangs der CH4 Emissionen sicherlich auf die Dürre zurückzuführen ist, könnte die zunehmenden Abundanzen von sulfatreduzierenden Bakterien (SRB) nach dem Einstrom des Brackwassers zu einem Wettbewerb mit Methanogenen um Substrat geführt haben. Zusätzlich oder alternativ dazu könnten SRB an der anaeroben Oxidation von CH4 beteiligt sein. Wir konnten jedoch weder eine Zunahme von Methanoxidation noch steigende Abundanzen von anaeroben Methanotrophen im Boden beobachten. Dies deutet darauf hin, dass die Methanotrophie in der Wassersäule oberhalb des untersuchten Moorbodens stattgefunden haben könnte. Im Gegensatz dazu wies das mit Brackwasser wiedervernässte Niedermoor unmittelbar nach der Wiedervernässung relativ geringe CH4 Emissionen auf als mit Süßwasser wiedervernässte Niedermoore. Im Vergleich zum entwässerten Zustand des Niedermoors nahmen SRB und Methanogene nach der Wiedervernässung zu, aber die Konkurrenz um das organische Substrat und stattfindende Methanoxidation begrenzten höchstwahrscheinlich übermäßige CH4 Emissionen. Die hohen CO2 Emissionen gingen jedoch trotz Wiedervernässung nicht wie erwartet zurück, während die Ökosystematmung und damit der Torfabbau stark reduziert wurden. Wir beobachteten ein starkes Absterben von sowohl Grünlandpflanzen als auch der grabennahen Moorvegetation nach der intensiven Überflutung mit Brackwasser. Unabhängig von der Torfzersetzung könnte der Grund für die anhaltenden CO2 Emissionen an den großen Mengen an organischem Substrat liegen, welches für die Ökosystematmung nach der Widervernässung zur Verfügung stand. Außerdem konnten die hohen CO2 Emissionen nicht verhindert werden, weil die CO2 Aufnahme durch Photosynthese fehlte. Diese Dissertation leistet einen Beitrag zur Diskussion über die Wiedervernässung von Küstenmooren und liefert neue Erkenntnisse über das Zusammenspiel von Mikrobiologie und Biogeochemie in verschiedenen räumlichen Kompartimenten eines Moores. Es wird gezeigt, dass nicht nur der Wassertyp, sondern auch die Häufigkeit des Brackwassereintrags und die Intensität der Überflutung für die Reduzierung von Treibhausgasemissionen von Bedeutung sein können. Um Wiedervernässungsansätze für künftige Projekte zu definieren und zu evaluieren, ist ein Vergleich wiedervernässter Moore anhand ihrer Entwicklung bezüglich der THG erforderlich. Darüber hinaus werden langfristiges Monitoring und multidisziplinäre Forschung benötigt, um Erkenntnisse zu gewinnen, die über das erste Jahr des Brackwassereinflusses in Küstenmooren hinaus gehen

    Emergence of phenotypic plasticity through epigenetic mechanisms

    Plasticity is found in all domains of life and is particularly relevant when populations experience variable environmental conditions. Traditionally, evolutionary models of plasticity are non-mechanistic: they typically view reactions norms as the target of selection, without considering the underlying genetics explicitly. Consequently, there have been difficulties in understanding the emergence of plasticity, and in explaining its limits and costs. In this paper, we offer a novel mechanistic approximation for the emergence and evolution of plasticity. We simulate random “epigenetic mutations” in the genotype–phenotype mapping, of the kind enabled by DNA-methylations/demethylations. The frequency of epigenetic mutations at loci affecting the phenotype is sensitive to organism stress (trait–environment mismatch), but is also genetically determined and evolvable. Thus, the “random motion” of epigenetic markers enables developmental learning-like behaviors that can improve adaptation within the limits imposed by the genotypes. However, with random motion being “goal-less,” this mechanism is also vulnerable to developmental noise leading to maladaptation. Our individual-based simulations show that epigenetic mutations can hide alleles that are temporarily unfavorable, thus enabling cryptic genetic variation. These alleles can be advantageous at later times, under regimes of environmental change, in spite of the accumulation of genetic loads. Simulations also demonstrate that plasticity is favored by natural selection in constant environments, but more under periodic environmental change. Plasticity also evolves under directional environmental change as long as the pace of change is not too fast and costs are low

    Increased CX3CL1 in cerebrospinal fluid and ictal serum t-tau elevations in migraine: results from a cross-sectional exploratory case-control study

    Background To date, migraine is diagnosed exclusively based on clinical criteria, but fluid biomarkers are desirable to gain insight into pathophysiological processes and inform clinical management. We investigated the state-dependent profile of fluid biomarkers for neuroaxonal damage and microglial activation as two potentially relevant aspects in human migraine pathophysiology. Methods This exploratory study included serum and cerebrospinal fluid (CSF) samples of patients with migraine during the headache phase (ictally) (n = 23), between attacks (interictally) (n = 16), and age/sex-matched controls (n = 19). Total Tau (t-Tau) protein, glial fibrillary acidic protein (GFAP), ubiquitin carboxy-terminal hydrolase L1 (UCH-L1), and neurofilament light chain (NfL) were measured with the Neurology 4-plex kit on a Single Molecule Array SR-X Analyzer (Simoa® SR-X, Quanterix Corp., Lexington, MA). Markers of microglial activation, C-X3-C motif chemokine ligand 1 (CX3CL1) and soluble triggering receptor expressed on myeloid cells 2 (sTREM2), were assessed using an immunoassay. Results Concentrations of CX3CL1 but not sTREM2 were significantly increased both ictally and interictally in CSF but not in serum in comparison to the control cohort (p = 0.039). ROC curve analysis provided an AUC of 0.699 (95% CI 0.563 to 0.813, p = 0.007). T-Tau in serum but not in CSF was significantly increased in samples from patients taken during the headache phase, but not interictally (effect size: η2 = 0.121, p = 0.038). ROC analysis of t-Tau protein in serum between ictal and interictal collected samples provided an AUC of 0.729 (95% CI 0.558 to 0.861, p = 0.006). The other determined biomarkers for axonal damage were not significantly different between the cohorts in either serum or CSF. Discussion CX3CL1 in CSF is a novel potential fluid biomarker of migraine that is unrelated to the headache status. Serum t-Tau is linked to the headache phase but not interictal migraine. These data need to be confirmed in a larger hypothesis-driven prospective study

    The excitability of ipsilateral motor evoked potentials is not task-specific and spatially distinct from the contralateral motor hotspot

    Objective The role of ipsilateral descending motor pathways in voluntary movement of humans is still a matter of debate, with partly contradictory results. The aim of our study therefore was to examine the excitability of ipsilateral motor evoked potentials (iMEPs) regarding site and the specificity for unilateral and bilateral elbow flexion extension tasks. Methods MR-navigated transcranial magnetic stimulation mapping of the dominant hemisphere was performed in twenty healthy participants during tonic unilateral (iBB), bilateral homologous (bBB) or bilateral antagonistic elbow flexion-extension (iBB-cAE), the map center of gravity (CoG) and iMEP area from BB were obtained. Results The map CoG of the ipsilateral BB was located more anterior-laterally than the hotspot of the contralateral BB within the primary motor cortex, with a significant difference in CoG in iBB and iBB-cAE, but not bBB compared to the hotspot for the contralateral BB (each p < 0.05). However, different tasks had no effect on the size of the iMEPs. Conclusion Our data demonstrated that excitability of ipsilateral and contralateral MEP differ spatially in a task-specific manner suggesting the involvement of different motor networks within the motor cortex

    The human antigen-specific immune response to Staphylococcus aureus T cell superantigens

    Staphylococcus (S.) aureus is a pathobiont, colonizing 20% of healthy adults persistently, while the remainder is colonized transiently, resulting in a general colonization rate of approximately 37%. Beside symptom-free colonization, S. aureus is the cause of different diseases, including self-limiting food poisoning, skin and soft tissue infections, but also life-threatening pneumonia, endocarditis and sepsis. Additionally, numerous studies suggest an involvement of S. aureus in allergic diseases, in which S. aureus colonizes up to 90% of patients. The high abundance of S. aureus in healthy individuals and patients with allergic diseases is enabled by the broad spectrum of its virulence factors. Among them are the T cell superantigens (SAgs). SAgs act on the immune system in multiple ways. On the one hand, they are superantigenic, i.e. they directly crosslink receptors on antigen-presenting cells (APCs) and T cells. This causes their toxicity, which is due to excessive proliferation of immune cells and cytokine release that can culminate in toxic shock syndrome and death. On the other hand, there are indications, that SAgs are recognized by the adaptive immune system in a conventional way. Most adults have high affinity SAg-specific antibodies that can neutralize the toxins. These antibodies are produced by B cells, which need SAg-specific T cell help for differentiation, presumably provided in a conventional cognate interaction. High levels of SAg-specific IgE were detected in various allergic diseases and linked to the pathology, strongly suggesting an allergenic role of SAgs. However, specific T cells that recognize SAgs as conventional antigens have never been demonstrated and characterized. In this study, we investigated the recognition of SAgs as conventional antigens by human T cells. This was made possible through the generation of SAg toxoids, which lack superantigenicity but retain the ability to be taken up by APCs, processed and presented to specific T cells in a conventional manner. The lack of residual superantigenicity was validated by analyzing the potential of the toxoids to oligoclonally expand T cells of specific TCR Vβ families and by comparing the immune cell transcriptome after stimulation with wild-type (WT) SAgs, toxoids and conventional S. aureus antigens. We demonstrated, that of the four tested toxoids (inactivated (i) SEA, iSEB, iTSST and iTSSTw) two lack superantigenicity (iSEA and iSEB). Using iSEA and iSEB as recall antigens for the restimulation of human T cells ex vivo, we discovered a robust SAg-specific T cell memory response. In particular, we found that T cells from healthy, non-symptomatic individuals exhibited a Type 2-polarized cytokine profile when stimulated with iSEA or iSEB. In contrast, patients with chronic rhinosinusitis with nasal polyps (CRSwNP) did not exhibit SEA- or SEB-specific Th2 cells. Additionally, we studied the humoral response and observed increased anti-SAg-IgE levels in the healthy individuals, but not in CRSwNP patients. While Type 1 and Type 3 immune responses are associated with a protection against S. aureus, the polarization towards Type 2 responses have been discussed to be non-protective and an immune evasion mechanism, which facilitates colonization. Insights into the involvement of SAgs in the polarization of a specific immune memory and in allergic diseases should be taken into consideration when designing vaccines against S. aureus for individuals with or without allergic diseases. This is important, because the pre-existing immune response to S. aureus antigens might be boosted by vaccination, which would be unwanted in case of a non-protective response profile

    Massenanfall von Erkrankten im Hafen: Analyse des Notfallmanagements am Beispiel des Hamburger Hafen als Grundlage für die Entwicklung einer Einsatzstrategie

    Das Projekt Adaptives Resilienz Management im Hafen (ARMIHN) konzentrierte sich auf die Stärkung der Handlungsfähigkeit bei einem Massenanfall von Erkrankten aufgrund eines Ausbruchs von Infektionskrankheiten. Während der COVID-19-Pandemie kam es zu Ausbrüchen auf Kreuzfahrtschiffen, die mediale Präsenz erlangten. Jedoch gab es schon frühere Massenanfälle von Erkrankten auf Kreuzfahrtschiffen, die insbesondere durch Krankheitserreger wie das Influenzavirus oder das Norovirus verursacht wurden. Der erste Schritt war, sich am Beispiel des Hamburger Hafens einen Überblick über Prozesse und Ressourcen zu verschaffen und das damit verbundene Zusammenspiel der Beteiligten aufzuzeigen. Dies dient als essenzielle Grundlage für die Entwicklung einer Einsatzstrategie und bietet die Möglichkeit, vorhandene Arbeitsabläufe zu optimieren. In einer selektiven Literaturrecherche wurde anhand vorgegebener Schlüsselwörter viele Informationen gesammelt sowie bestehende Konzepte zur Großschadenslage von lokalen Behörden wie der Berufsfeuerwehr Hamburg oder des Hamburg Port Health Centers eingeholt. Identifizierte Strukturen und Prozesse wurden in Workshops mit beteiligten Organisationen und anderen Experten analysiert, zusammengeführt und diskutiert. Zusätzlich wurden die Entfernungen zwischen den nächstgelegenen Rettungswachen und den ausgewählten Krankenhäusern zum Hamburger Hafen erfasst. Die derzeit verfügbaren Konzepte haben sich bewährt, aber eine Anpassung an einen Massenanfall von Erkrankten zeigt die Chance für eine organisationsübergreifende Weiterentwicklung auf. Beschrieben wurde die Organisationsstruktur eines Massenanfalls von Erkrankten im Hamburger Hafen mit einer Vielzahl beteiligter Akteure (n = 18). Es gibt 17 beteiligte Feuer- und Rettungswachen; 14 der 31 gelisteten Kliniken wurden aufgrund ihrer fachlichen Expertise ausgewählt und deren Entfernung zum Hafen bestimmt. Darauf aufbauend konnten drei Schadensszenarien mit den am häufigsten zum Ausbruch führenden Infektionserkrankungen auf Schiffen erstellt werden. Daraus gelang wiederum die Entwicklung eines Scores zur Sichtung von Erkrankten bei einem Massenanfall. Nachfolgende Forschungsarbeiten erwähnten diese publizierte Arbeit als Beispiel dafür, wie wichtig eine Ressourcen- und Prozesseanalyse ist, um sich besser auf Großschadenslagen in einer speziellen Infrastruktur wie der eines Schiffes oder Hafens vorbereiten zu können

    Health Effects of Participation in Creating Urban Green Spaces—A Systematic Review

    The participation of citizens in creating urban green spaces is a promising approach to promoting health and wellbeing while facilitating the development of equitable cities more prepared for the challenges of climate change. This systematic review provides an overview of the current state of knowledge by synthesizing and summarizing scholarly articles reporting studies on the health effects of participation in green space creation. A review protocol was preregistered on PROSPERO (registration number CRD42023454363). Following PRISMA guidelines, we searched six databases for studies published since 2003. After screening 2676 records, we found 133 studies matching our inclusion criteria and summarized the findings on health effects in qualitative and quantitative research, differentiating between study types, population types, and intervention types. In addition, we explored secondary outcomes mentioned in the included articles. The majority of studies reported positive impacts of participatory green space creation on general, mental, and physical health. Moreover, many studies reported other personal and social benefits. The results of this review indicate that the participatory creation of urban green spaces represents a promising approach to promoting health and wellbeing. Nevertheless, further high-quality, quantitative research is required to gain a more comprehensive understanding of the relationship between participatory green space activities and health, particularly with regard to the benefits of specific intervention types and subpopulations

    Distinguishing between Low- and High-Cost Pro-Environmental Behavior: Empirical Evidence from Two Complementary Studies

    To reach sustainability goals, researchers and policymakers must focus on addressing changes toward more pro-environmental behavior (PEB). Therefore, this study shows evidence-based perceptions of low- and high-cost behaviors and deduces recommendations for PEB interventions. We applied a multi-step mixed-method approach: First, a representative online survey was conducted to collect a general quantitative overview and classify low- and high-cost behavior based on past behavior and behavioral intentions. Second, a gamification intervention that aimed to gain quantitative data about certain behaviors and perceived low- and high-costs was conducted with university students. The results of the two studies showed that PEB can easily be categorized into high-, rather-high-, rather-low-, and low-cost behavior. However, this classification is not based on emission sectors, e.g., mobility, but on the specific behavior. Interventions can be recommended according to subjective costs: For example, low-cost behavior does not need additional interventions in most cases but must be maintained as is. According to the empirical findings, high-cost behavior needs top-down interventions, while rather-high- and rather-low-cost behavior requires bottom-up interventions to achieve behavior changes. In summary, managing interventions using this classification and focusing on high-impact behavior can lead to successful behavior changes and emission reductions

    Inhibition der Fettsäuresynthase während hormonell-induzierter hepatozellulärer Karzinogenese

    Auch wenn für das fortgeschrittene HCC mittlerweile mehrere systemische Therapieansätze zur Verfügung stehen, kommt es bei vielen Patienten dennoch zu einem Progress der Erkrankung. Ein Problem stellt die Heterogenität des HCC dar. Es existieren verschiedene Subtypen des HCC, welche sich in ihren molekularen und chromosomalen Aberrationen, epigenetischen Modifikationen sowie auch ihrem Tumor-Mikromilieu unterscheiden. Gleichzeitig gibt es für die häufigsten Treibermutationen des HCC bisher keine geeigneten Inhibitoren. Dadurch erklärt sich das zum Teil nur geringe oder fehlende Ansprechen auf systemische Therapien, was die Suche nach weiteren Therapieansätzen nötig macht. FASN-Inhibitoren stellen dabei vielversprechende Kandidaten einer HCC-Therapie dar. Zum einen konnte eine Überexpression von FASN in mehreren Tumoren wie auch dem HCC nachgewiesen werden, die Expression im Normalgewebe und die damit zu vermuteten Nebenwirkungen sind sehr gering. Zum anderen sprechen mehrere präklinische Studien dafür, dass FASN eine wichtige Rolle in der Hepatokarzinogenese spielt. An Zellkulturen fand sich eine Apoptoseinduktion sowie verminderte Proliferation von hepatozellulären Tumorzellen durch FASN-Inhibitoren. Außerdem konnte in Tiermodellen unter Einsatz von FASN-Inhibitoren die Entstehung von HCC verhindert oder verzögert werden. In dem vorliegenden Modell der hormonell-induzierten Hepatokarzinogenese kommt es zur Entwicklung klarzelliger Herde, welche Präneoplasien darstellen und sich über Adenome in HCC entwickeln. Die in diesen klarzelligen Herden beschriebenen metabolischen Veränderungen sind der Hepatokarzinogenese auf dem Boden einer NASH sehr ähnlich und repräsentieren damit relativ gut einen Subtyp des HCC. Die Gabe von hochdosiertem Orlistat über eine Woche führte zu einer reduzierten Proliferationsaktivität der präneoplastischen Herde, eine erniedrigte Proliferation ließ sich jedoch auch in der zugehörigen Kontrollgruppe nachweisen, welche lediglich hochdosiert DMSO erhielt. Somit scheint die proliferationshemmende Wirkung in erster Linie über DMSO bedingt, eine Wirkung des FASN-Inhibitors Orlistat ist nicht sicher zu belegen. In der Literatur wird in Zellkulturen eine Wachstumshemmung bei hochdosierter DMSO-Gabe beschrieben. Des Weiteren führt DMSO bei einigen HCC-Zelllinien zu einer Ausreifung bzw. einem Verlust des malignen Phänotyps mit Herabregulierung u.a. der Fettsäuresynthese, was möglicherweise im vorliegenden Fall ebenfalls eine Rolle spielt. Mittlerweile stehen neue, im Gegensatz zu Orlistat oral verfügbare FASN-Inhibitoren zur Verfügung, außerdem wird daran gearbeitet, die orale Bioverfügbarkeit von Orlistat zu erhöhen. Die neuen FASN-Inhibitoren haben in Tiermodellen des HCC bereits vielversprechende Ergebnisse gezeigt. Dabei fand sich häufig ein synergistischer Effekt in Kombination mit anderen Chemotherapeutika. Letztendlich scheint die Gabe eines FASN-Inhibitors in Kombination mit einem anderen Therapeutikum sinnvoll, da präklinische Studien darauf hinweisen, dass es zu einer Resistenz gegenüber einer FASN-Inhibition durch eine Aktivierung anderer Signalwege kommen kann
