    Physiological responses to environmental drivers in dispersive larval stages of marine crustaceans: multi-population studies to understand range expansions

    Global climate change and biological invasions are reshaping biodiversity worldwide, particularly in semi-enclosed seas like the North Sea, Baltic Sea, and the Arctic Ocean, where rapid warming drives poleward species range shifts. These shifts depend on complex ecological and evolutionary processes, with multiple environmental drivers often interacting unpredictably. This underscores the use of multi-driver studies to unravel the interactive effects of co-occurring environmental drivers and ecologically relevant intraspecific variability. Because sympatric species might react differently to such change, interspecific comparisons are important in predicting potential changes in community composition. In benthic marine invertebrates with complex life cycles, larvae are the main dispersive life stages that can reach and colonise new habitats. Typically, there is a substantial variation in the magnitude of environmental effects between life stages, with larvae forming an important bottleneck in a population's ability to persist in a habitat. Coastal crabs are key species that play critical roles in food webs, functioning as both predators and prey, as well as ecosystem engineers that shape their habitats. They are also among the most successful marine invasive species. Using larvae of ecologically relevant coastal decapod crustaceans as models, this thesis investigates inter- and intraspecific variability across environmental gradients and elucidates its role in climate-driven range shifts. Using two model systems, (1) the interactive effect of salinity and temperature on larval development in invasive populations of two Hemigrapsus species, H. takanoi and H. sanguineus, along a salinity gradient from the North Sea to the Baltic Sea, and (2) the variability of larval traits like temperature tolerance in C. maenas across a temperature gradient in its native range along the European Atlantic coast were investigated. Based on individual studies on intraspecific variation, comparisons could be drawn among the three species, the native C. maenas and the two non-native species. Larval rearing experiments in factorial designs assessed survival, duration of development, and growth across environmental treatments, with larvae sourced from multiple populations and females. In the first part (Manuscript 1 & 2), the thesis demonstrates that both Hemigrapsus spp. found in Europe exhibit increased survival, developmental and growth rates compared to the native species at combinations of high temperatures and low salinities. Comparing populations of H. takanoi, a surprising lack of adaptations to low salinities was found in populations from the Baltic Sea. Larvae from the higher salinity areas in the North Sea showed higher survival and faster development than the populations from the Baltic Sea, with the population from the range edge showing poor performance across treatments. Accompanying population genetics analysis revealed no significant genetic separation among populations, indicating persistent connectivity among populations. In combination with the very low survival in the population from the range edge and the low salinity of the area, a source-sink dynamic among populations or complex ontogenetic migration patterns appear likely. In the second part (Manuscript 3 & 4), variability in larval traits, such as temperature tolerance was studied along a large latitudinal gradient in C. maenas. a strong increase in the body mass of freshly hatched larvae with latitude was found across 35° latitude of the native distribution range. Further, considerable variations in thermal tolerance across the range of this species were detected. Some variations met expectations based on the biogeography of the species, like the slightly elevated tolerance to chronically high temperatures in populations from southern Spain and the strong increase in tolerance to cold exposure (6 °C) with latitude. However, unexpected features like high survival rates in populations from the northern edge of the distribution range under chronically high temperatures were also found. In temperature ramp experiments, a correlation between the performance in cold ramps and heat ramps was found, indicating that there is no trade-off between these two traits but that they could be a measure of larval quality. The performance in the acute exposure experiments did not correlate with performance under chronic exposure, indicating that each method assesses different aspects of thermal tolerance. This thesis emphasises the importance of recognising intraspecific variation in response to environmental drivers, as it shapes both the success of invasive species and the adaptability of native species in changing environments. By integrating macrophysiological and multi-driver approaches, this thesis advances the understanding of species responses to environmental change and informs predictions about range shifts under future climates

    Retrospektive Analyse der ureterorenoskopischen Steintherapie im Zeitraum von Januar 2010 bis März 2015 in der Klinik und Poliklinik für Urologie der Universitätsmedizin Greifswald

    Die ureterorenoskopische Harnsteintherapie stellt heutzutage in der Urologie ein etabliertes Verfahren dar. Angesichts einer steigenden Inzidenz und Prävalenz sind die neuesten Erkenntnisse über Therapiemöglichkeiten der Harnsteine sicherlich relevant. Gegenstand der vorliegenden Untersuchung waren die epidemiologischen Aspekte, das prä-, peri- und postoperative Management sowie das postoperative Outcome der ureterorenoskopisch behandelten Steinpatienten in der Klinik und Poliklinik für Urologie des Universitätsklinikums Greifswald im Zeitraum von Januar 2010 bis einschließlich März 2015. Die Untersuchungsergebnisse wurden mit bereits veröffentlichten Studien verglichen, diskutiert und gewertet. Von 574 Patienten waren 379 männlich (66,0%) und 195 weiblich (34,0%). Das durchschnittliche Patientenalter betrug 55,6 Jahre. Die Mehrheit der Harnsteine trat in der Altersgruppe der 51 bis 60-jährigen Patienten auf (23,3%). Der mittlere BMI lag bei 28,3 kg/m2. Die Mehrheit der Patienten war übergewichtig (39,3%). Im distalen Ureter waren die meisten Harnsteine lokalisiert (26,0%). Die durchschnittliche Harnsteingröße betrug 7,4 mm. Die mittlere stationäre Aufenthaltsdauer lag bei 2,5 Tagen. 72,3% aller Patienten hatten keine Voroperationen. Von den Voroperationen war die ESWL die häufigste Voroperation (15,7%). 558 Patienten (97,2%) bekamen präoperativ einen Stent eingelegt. Als präoperative Bildgebung wurde eine NLA (70,4%), gefolgt von der CT (29,1%) und der intravenösen Pyelographie (0,5%) durchgeführt. Die Kombination aus einem flexiblen und einem starren Ureterorenoskop wurde am häufigsten verwendet (39,9%). Die starren Ureterorenoskope wurden bei 35,0% der Patienten eingesetzt, die flexiblen Ureterorenoskope bei 25,1%. Die Harnsteine wurden mithilfe von Steinfangkörbchen (55,9%), der Kombination aus Ho:YAG-Laser und Steinfangkörbchen (20,9%) und der Steinfasszange (11,2%) therapiert. Die Steinfreiheitsrate betrug nach dem ersten ureterorenoskopischen Eingriff 90,5%. Bei Steinen, die mit einem flexiblen Ureterorenoskop therapiert worden sind, lag die Steinfreiheitsrate bei 84,0%; diejenigen, die ausschließlich mit einem starren Ureterorenoskop therapiert worden sind, bei 94,0%. 82,7% der Patienten hatten keine Komplikationen. Die Antibiotika Ampicillin/Sulbactam und Ciprofloxacin wurden prä-, peri- und postoperativ am häufigsten gegeben. Von 172 Patienten, die weder präoperativ, perioperativ noch postoperativ ein Antibiotikum erhielten, traten bei 166 dieser Patienten keine Komplikationen auf. Bei 88,5% der Patienten war kein Folgeeingriff notwendig. Zusammenfassend lässt sich feststellen, dass die Ureterorenoskopie im Rahmen der Harnsteintherapie in der Klinik und Poliklinik für Urologie des Universitätsklinikums Greifswald eine komplikationsarme und effektive Methode zur Therapie von Harnsteinen im gesamten oberen Harntrakt ist. In wesentlichen Punkten stimmten die Ergebnisse dieser Untersuchung mit den Ergebnissen von internationalen Studien überein.Ureterorenoscopic urinary stone therapy is an established procedure in urology today. In view of the increasing incidence and prevalence, the latest findings on treatment options for urinary calculi are certainly relevant. The subject of the present study was the epidemiological aspects, pre-, peri- and postoperative management and postoperative outcome of ureterorenoscopically treated stone patients at the Department of Urology at Greifswald University Hospital in the period from January 2010 to March 2015 inclusive. The study results were compared with previously published studies, discussed and evaluated. Of 574 patients, 379 were male (66.0%) and 195 female (34.0%). The average patient age was 55.6 years. The majority of urinary stones occurred in the 51 to 60 age group (23.3%). The mean BMI was 28.3 kg/m2. The majority of patients were overweight (39.3%). Most urinary stones were localised in the distal ureter (26.0%). The average urinary stone size was 7.4 mm. The mean length of hospitalisation was 2.5 days. 72.3% of all patients had no previous operations. Of the previous operations, ESWL was the most common (15.7%). 558 patients (97.2%) had a stent inserted preoperatively. Preoperative imaging was NLA (70.4%), followed by CT (29.1%) and intravenous pyelography (0.5%). The combination of a flexible and a rigid ureterorenoscope was used most frequently (39.9%). Rigid ureterorenoscopes were used in 35.0% of patients and flexible ureterorenoscopes in 25.1%. Urinary stones were treated using stone basketing (55.9%), the combination of Ho:YAG laser and stone basketing (20.9%) and stone grasping forceps (11.2%). The stone-free rate after the first ureterorenoscopic procedure was 90.5%. For stones treated with a flexible ureterorenoscope, the stone-free rate was 84.0%; for those treated exclusively with a rigid ureterorenoscope, the rate was 94.0%. 82.7% of patients had no complications. The antibiotics ampicillin/sulbactam and ciprofloxacin were most frequently administered pre-, peri- and postoperatively. Of 172 patients who received no antibiotics preoperatively, perioperatively or postoperatively, 166 of these patients had no complications. No follow-up intervention was necessary in 88.5% of patients. In summary, it can be stated that ureterorenoscopy as part of urinary stone therapy at the Department of Urology at Greifswald University Hospital is an effective method with low complications for treating urinary stones in the entire upper urinary tract.     Investigation of the sodium nightglow in the Earth's atmosphere

    The Earth’s atmosphere is constantly hit by Interplanetary Dust Particles (IDPs) of various sizes. The majority of them are very small, with typical sizes of only a few nanometers in diameter. Most of them never hit the Earth’s surface but ablate, due to heating by friction with the surrounding air molecules, in the upper atmosphere. Those IDPs, also called meteoroides, consist of a multitude of different chemical species. One of those species is sodium. After atomic sodium ablates, it forms a layer in the MLT (Mesosphere lower- Thermosphere) region between 90 and 100 km altitude. Because sodium has a relatively long atmospheric lifetime in the MLT and a large resonance fluorescence cross-section, leading to a strong emission of radiation, it is an excellent tracer to investigate wave propagation effects and winds in the upper atmosphere. It can also specifically be used to determine the amount of IDPs reaching the Earth’s atmosphere in the form of meteoroides. An in-depth knowledge of the sodium concentration in the MLT region is therefore the key to further investigations. There are a few good ways to measure the amount of sodium in the MLT region. In this thesis, two of those are utilized to contribute to a better understanding of the amount of sodium in the upper atmosphere, as well as its chemical properties. The first is to detect the electromagnetic radiation that is emitted as a consequence of the excitation caused by the reaction of sodium with ozone and the second excites sodium with a laser and detects the amount of electromagnetic radiation that reaches the detector after the sodium atoms relax to ground-state. In the first part of this thesis, a retrieval method is developed that is robustly able to use satellite measurements of the sodium D-line nightglow to retrieve sodium concentration profiles. One focus hereby is the effect of the regularization parameter on the resulting sodium profiles. It is found that has to be chosen in a way that it smoothes the result but does not distort the shape of the resulting sodium profiles. Then, a new effective branching ratio f , that determines the amount of excited sodium after the reaction NaO + O -> Na + O2, is estimated by comparing satellite measurements from the OSIRIS instrument on Odin to ground-based Lidar measurements. The result is a branching ratio f of 0.061 +- 0.026. This value fits well into the literature, where values between 0.05 and 0.67 have been proposed. In the last chapter, monthly averaged sodium D-line LER (Limb emission rate), VER (Volume emission rate) and sodium profiles, as well as sodium VCDs (Vertical column densities), from three different satellite instruments are compared. The first is again the OSIRIS instrument on Odin, the second the SCIAMACHY instrument on Envisat and last the GOMOS instrument that was also located on Envisat. 34 months that are suitable for comparison over the period 2006 to 2011 are found. Then one sodium concentration profile is retrieved for each month with uncertainties between 2 and 15 %. While SCIAMACHY detects slightly larger LERs and VERs compared to those detected by OSIRIS, the overall seasonal variation of those two parameters agrees very well. The sodium concentration profiles, however show a greater variation in shape as well as in values. It was concluded that the reason for this high variability primarily lies in the high variability of the ozone profiles and the high sensitivity of the sodium retrieval to changes of the ozone concentration. To sum up, it is shown that we have a very good understanding of the sodium layer in the MLT region and the excitation mechanism of sodium by ozone. Most of the challenges that are encountered in this thesis, can be ascribed to a too small amount of measurements, low SNRs (Signal-to-noise ratio), or the lack of co-located measurements in both, the time and space dimension. Therefore, to further the research on sodium, new instruments are needed, whose specific goal is the detection of sodium, as well as a better coordination between the instruments to yield a greater sample size.Die Erdatmosphäre wird ununterbrochen von interplanetaren Staubpartikeln (Engl.: Interplanetary Dust Particles, IDPs) unterschiedlicher Größe getroffen. Der Großteil dieser Partikel ist sehr klein, mit typischen Durchmessern von nur wenigen Nanometern. Die meisten von ihnen erreichen nie die Erdoberfläche, sondern verglühen in der oberen Atmosphäre durch Erwärmung durch Reibung beim Kontakt mit den umgebenden Luftmolekülen. Diese IDPs, auch Meteoroide genannt, bestehen aus einer Vielzahl unterschiedlicher chemischer Elemente. Eines dieser Elemente ist Natrium. Wenn atomares Natrium verdampft, bildet es eine Schicht in der MLT-Region (Mesosphäre – untere Thermosphäre) in einer Höhe zwischen 90 und 100 km. Da Natrium in der MLT eine relativ lange atmosphärische Lebensdauer sowie einen großen Wirkungsquerschnitt für Resonanzfluoreszenz hat – was zu einer starken Strahlungsemission führt – eignet es sich hervorragend als Tracer zur Untersuchung von Wellenfortpflanzungseffekten und Winden in der oberen Atmosphäre. Es kann außerdem gezielt verwendet werden, um die Menge an IDPs zu bestimmen, die die Erdatmosphäre in Form von Meteoroiden erreichen. Ein tiefgehendes Verständnis der Natriumkonzentration in der MLT-Region ist daher entscheidend für weiterführende Untersuchungen. Es gibt einige bewährte Methoden, um die Menge an Natrium in der MLT-Region zu messen. In dieser Arbeit werden zwei dieser Methoden genutzt, um zu einem besseren Verständnis sowohl der Natriummenge in der oberen Atmosphäre als auch seiner chemischen Eigenschaften beizutragen. Die erste Methode basiert auf der Detektion elektromagnetischer Strahlung, die als Folge der Anregung durch die Reaktion von Natrium mit Ozon entsteht. Die zweite Methode nutzt einen Laser zur Anregung von Natrium und detektiert die Menge an elektromagnetischer Strahlung, die den Detektor erreicht, nachdem die Natriumatome in den Grundzustand zurückgekehrt sind. Im ersten Teil dieser Arbeit wird ein Retrieval-Algorithmus entwickelt, die es ermöglicht, Satellitenmessungen des Natrium-D-Linien-Nachthimmelleuchtens zuverlässig zur Bestimmung von Natriumkonzentrationsprofilen zu verwenden. Ein Schwerpunkt liegt dabei auf dem Einfluss des Regularisierungsparameters auf die bestimmten Natriumprofile. Es zeigt sich, dass dieser Parameter so gewählt werden muss, dass er die Ergebnisse glättet, ohne die Form der resultierenden Profile zu verfälschen. Anschließend wird eine neue effektive branching ratio f bestimmt, die angibt, wie viel angeregtes Natrium nach der Reaktion NaO + O → Na + O₂ entsteht. Dazu werden Satellitenmessungen des OSIRIS-Instruments auf Odin mit bodengestützten Lidar-Messungen verglichen. Das Ergebnis ist eine branching ratio f von 0,061 ± 0,026. Dieser Wert liegt gut im Bereich der in anderen Studien gefundenen Werte, die zwischen 0,05 und 0,67 liegen. Im letzten Kapitel werden monatlich gemittelte Natrium-D-Linien-Emissionsraten (LER – Limb Emission Rate), Volumenemissionsraten (VER – Volume Emission Rate) sowie Natriumprofile und vertikale Säulendichten (VCDs – Vertical Column Densities) von drei verschiedenen Satelliteninstrumenten verglichen. Das erste Instrument ist wieder OSIRIS auf Odin, das zweite SCIAMACHY auf Envisat und das dritte das GOMOS-Instrument, ebenfalls auf Envisat. Es konnten 34 Monate im Zeitraum von 2006 bis 2011 identifiziert werden, die sich für einen Vergleich eignen. Für jeden dieser Monate wird ein Natriumkonzentrationsprofil mit Unsicherheiten zwischen 2 und 15 % bestimmt. Während SCIAMACHY etwas höhere LER- und VER-Werte detektiert als OSIRIS, stimmen die saisonalen Schwankungen dieser beiden Parameter sehr gut überein. Die Natriumkonzentrationsprofile hingegen zeigen größere Unterschiede in Form und Betrag. Es wird vermutet, dass diese hohe Variabilität vor allem auf die große Variabilität der Ozonprofile sowie auf die hohe Sensitivität des Natriumretrievals gegenüber Änderungen der Ozonkonzentration zurückzuführen ist. Zusammenfassend zeigt sich, dass ein sehr gutes Verständnis der Natrium-Schicht in der MLT-Region sowie des Anregungsmechanismus von Natrium durch Ozon vorliegt. Die meisten Herausforderungen, die in dieser Arbeit auftreten, lassen sich auf eine zu geringe Anzahl an Messungen, niedrige Signal-Rausch-Verhältnisse (SNR) oder das Fehlen zeitlich und räumlich übereinstimmender Messungen zurückführen. Um die Forschung zu Natrium weiter voranzutreiben, werden daher neue Instrumente benötigt, die speziell auf die Detektion von Natrium ausgelegt sind, sowie eine bessere Koordination zwischen den Instrumenten, um eine größere Datenbasis zu schaffen

    Mechanisms and effects of nutrient co-limitation in freshwater plankton communities

    In recent years, there has been growing evidence of frequent Nitrogen (N) and Phosphorus (P) co-limitation of autotrophs across terrestrial and aquatic ecosystems. Despite being a relatively new concept, nutrient (or resource) (co-)limitation has been extensively studied and characterized in phytoplankton and, to a lesser extent, in herbivorous zooplankton. Nevertheless, there is not a complete understanding of I) the underlying mechanisms leading to (co-)limitation patterns at the community level, and II) how N and P (co-)limitation translates from primary producers to consumers in relation to changes in the nutritional quality of autotrophs and species composition. Through full-factorial microcosm experiments and freshwater plankton communities, this work aimed to address a research gap on mechanisms and drivers of N and P co-limitation in autotroph communities and across trophic levels. Firstly, we investigated mechanisms leading to (co-)limitation at the community level. Three phytoplankton communities were grown at different N and P limiting conditions and then factorially supplied nutrients. We compared the biomass responses of single species and the whole community to nutrient additions to assess what leads to the observed community (co)-limitation patterns. The phytoplankton species in the communities responded differently to factorial nutrient addition, i.e. they showed contrasting (co-)limitation outcomes. In contrast, patterns at the community level were mostly driven by the dominant species. These results indicate that one of the mechanisms leading to biomass patterns of nutrient (co-)limitation at the community level is the dominance of species or groups with similar traits in nutrient requirements. Furthermore, we showed that whole communities can also be limited by a single nutrient. The second objective was to explore the consequences of N and P (co-)limitation of phytoplankton for herbivores and to hypothesize which phytoplankton traits related to nutritional quality and quantity can explain the herbivore’s response. We grew a community of phytoplankton species differing in food quality traits at different N and P limiting conditions, and then factorially supplied with nutrients. The community was then offered to the rotifer Brachionus calyciflorus in a population growth assay. The conditions of limitation of phytoplankton led to changes in biomass availability for rotifers, and the response to nutrient supply did not translate one-to-one from primary producers to herbivores. Because rotifer growth patterns differed from phytoplankton depending on nutrient supplies, not only food quantity, but additional food quality traits such as stoichiometry and biochemical composition might play a substantial role in explaining the response of herbivores. Finally, we aimed to verify previous hypotheses regarding drivers related to nutritional value and community composition of N and P (co)-limited phytoplankton communities affecting consumer growth. Three communities of species of varying nutritional traits were grown at different N and P limiting conditions and supplied factorially with nutrients. Then, we offered the communities to the cladoceran Daphnia pulex in a growth assay. Apart from community composition, we measured several nutritional traits of phytoplankton, from stoichiometry, to essential biochemical compounds such as fatty acid content, to cell edibility. In particular, when phytoplankton communities were N-limited, stoichiometry showed the strongest effect, followed by traits related to community composition. At N and P co-limiting conditions, only phytoplankton community traits affected daphniid growth, whereas phytoplankton stoichiometry played a negligible role. Our findings suggest that, at varying nutrient (co-)limiting conditions, stoichiometry and community traits of phytoplankton differently influence the growth of herbivorous zooplankton. With these experiments, we aimed to provide contribution to an existing research gap regarding N and P (co-)limitation at the community level and how nutrient (co-)limitation translates across trophic levels. Although it might only be one among several mechanisms, the dominance of species or groups with similar traits in nutrient requirements can often explain (co-)limitation patterns at the community level. Furthermore, the response of different zooplankton species to N and P (co-)limited communities changes from that of their food and can be explained by an interplay between food nutritional traits, in particular stoichiometry, and traits related to community composition. As nutrient manipulations and loading are currently altering global biogeochemical cycles, it is highly relevant to understand and predict the response of organisms in aquatic communities to changes in nutrient availability.In den letzten Jahren mehren sich die Hinweise auf eine häufige Stickstoff- (N) und Phosphor- (P) Co-Limitierung von Autotrophen in terrestrischen und aquatischen Ökosystemen. Obwohl es sich um ein relativ neues Konzept handelt, wurde die (Co-)Limitierung von Nährstoffen (oder Ressourcen) im Phytoplankton und, in geringerem Maße, im pflanzenfressenden Zooplankton umfassend untersucht und charakterisiert. Dennoch gibt es kein vollständiges Verständnis I) der zugrundeliegenden Mechanismen, die zu (Co-)Limitierungsmustern auf Gemeinschaftsebene führen, und II) wie N- und P-(Co-)Limitierung von Primärproduzenten auf Konsumenten in Bezug auf Veränderungen in der Nährstoffqualität von Autotrophen und der Artenzusammensetzung übertragen wird. Mit Hilfe von vollfaktoriellen Experimenten und Süßwasserplankton-Gemeinschaften sollte in dieser Arbeit eine Forschungslücke zu Mechanismen und Triebkräften der N- und P-(Co-)Limitierung in autotrophen Gemeinschaften und über trophische Ebenen hinweg geschlossen werden. Zunächst untersuchten wir die Mechanismen, die zu einer (Co-)Limitierung auf der Ebene der Gemeinschaften führen. Drei Phytoplankton-Gemeinschaften wurden unter verschiedenen N- und P-limitierenden Bedingungen gezüchtet und dann faktoriell mit Nährstoffen versorgt. Wir verglichen die Reaktionen der Biomasse einzelner Arten und der gesamten Gemeinschaft auf die Nährstoffzufuhr, um zu ermitteln, was zu den beobachteten (Co-)Limitierungsmustern der Gemeinschaft führt. Die Phytoplanktonarten in den Gemeinschaften reagierten unterschiedlich auf die faktorielle Nährstoffzufuhr, d. h. sie zeigten kontrastierende (Co-)Limitierungsergebnisse. Im Gegensatz dazu wurden die Muster auf Gemeinschaftsebene hauptsächlich von der dominanten Art bestimmt. Diese Ergebnisse deuten darauf hin, dass einer der Mechanismen, der zu Biomassemustern der Nährstoff(co-)limitierung auf Gemeinschaftsebene führt, die Dominanz von Arten oder Gruppen mit ähnlichen Merkmalen im Nährstoffbedarf ist. Außerdem haben wir gezeigt, dass auch ganze Gemeinschaften durch einen einzigen Nährstoff limitiert sein können. Das zweite Ziel bestand darin, die Folgen der N- und P-( Co-)Limitierung von Phytoplankton für Herbivoren zu erforschen und Hypothesen darüber aufzustellen, welche Phytoplanktonmerkmale in Bezug auf Nahrungsqualität und -quantität die Reaktion der Herbivoren erklären können. Wir züchteten eine Gemeinschaft von Phytoplanktonarten mit unterschiedlichen Merkmalen der Nahrungsqualität unter verschiedenen N- und P-limitierenden Bedingungen und versorgten sie dann faktoriell mit Nährstoffen. Die Gemeinschaft wurde dann dem Rädertierchen Brachionus calyciflorus in einem Populationswachstumsversuch angeboten. Die Bedingungen der Limitierung des Phytoplanktons führten zu Veränderungen in der Verfügbarkeit von Biomasse für Rädertierchen, und die Reaktion auf das Nährstoffangebot ließ sich nicht eins zu eins von Primärproduzenten auf Herbivoren übertragen. Da sich die Wachstumsmuster von Rädertierchen je nach Nährstoffangebot von denen des Phytoplanktons unterschieden, könnten nicht nur die Nahrungsmenge, sondern auch zusätzliche Merkmale der Nahrungsqualität wie Stöchiometrie und biochemische Zusammensetzung eine wesentliche Rolle bei der Erklärung der Reaktion von Pflanzenfressern spielen. Schließlich zielten wir darauf ab, frühere Hypothesen zu überprüfen, die sich auf den Nährwert und die Zusammensetzung der Gemeinschaft von N- und P-(Co)-limitierten Phytoplankton-Gemeinschaften beziehen und das Wachstum der Konsumenten beeinflussen. Drei Gemeinschaften von Arten mit unterschiedlichen Nährwertmerkmalen wurden unter verschiedenen N- und P-limitierenden Bedingungen gezüchtet und faktoriell mit Nährstoffen versorgt. Anschließend haben wir die Gemeinschaften dem Wasserfloh Daphnia pulex in einem Wachstumstest angeboten. Neben der Zusammensetzung der Gemeinschaften haben wir verschiedene Nährstoffmerkmale des Phytoplanktons gemessen, von der Stöchiometrie über essenzielle biochemische Verbindungen wie den Fettsäuregehalt bis hin zur Essbarkeit der Zellen. Wenn die Phytoplanktongemeinschaften N-limitiert waren, zeigte die Stöchiometrie die stärkste Wirkung, gefolgt von Merkmalen, die mit der Zusammensetzung der Gemeinschaft zusammenhängen. Unter N- und P-limitierenden Bedingungen wirkten sich nur Merkmale der Phytoplanktongemeinschaft auf das Daphnienwachstum aus, während die Phytoplanktonstöchiometrie eine vernachlässigbare Rolle spielte. Unsere Ergebnisse deuten darauf hin, dass die Stöchiometrie und die Eigenschaften der Phytoplanktongemeinschaft unter verschiedenen Nährstoff (co-)limitierenden Bedingungen das Wachstum von herbivorem Zooplankton unterschiedlich beeinflussen. d P auf Gemeinschaftsebene leisten und zeigen, wie sich die (Co-)Limitierung von Nährstoffen über trophische Ebenen hinweg auswirkt. Auch wenn es sich dabei nur um einen von mehreren Mechanismen handelt, kann die Dominanz von Arten oder Gruppen mit ähnlichen Merkmalen in Bezug auf den Nährstoffbedarf häufig (Co-)Limitierungsmuster auf Gemeinschaftsebene erklären. Darüber hinaus unterscheidet sich die Reaktion verschiedener Zooplanktonarten auf N- und P-(co-)limitierte Gemeinschaften von der ihrer Nahrung und lässt sich durch ein Zusammenspiel von Nahrungsmerkmalen, insbesondere der Stöchiometrie, und Merkmalen im Zusammenhang mit der Zusammensetzung der Gemeinschaft erklären. Da Nährstoffmanipulationen und -belastungen derzeit die globalen biogeochemischen Kreisläufe verändern, ist es äußerst wichtig, die Reaktion von Organismen in aquatischen Gemeinschaften auf Veränderungen der Nährstoffverfügbarkeit zu verstehen und vorherzusagen

    Structural Elucidation of a Metagenomic Urethanase and Its Engineering Towards Enhanced Hydrolysis Profiles

    While plastics like polyethylene terephthalate can already be degraded efficiently by the activity of hydrolases, other synthetic polymers like polyurethanes (PUs) and polyamides (PAs) largely resist biodegradation. In this study, we solved the first crystal structure of the metagenomic urethanase UMG‐SP‐1, identified highly flexible loop regions to comprise active site residues, and targeted a total of 20 potential hot spots by site‐saturation mutagenesis. Engineering campaigns yielded variants with single mutations, exhibiting almost 3‐ and 8‐fold improved activity against highly stable N ‐aryl urethane and amide bonds, respectively. Furthermore, we demonstrated the release of the corresponding monomers from a thermoplastic polyester‐PU and a PA (nylon 6) by the activity of a single, metagenome‐derived urethanase after short incubation times. Thereby, we expanded the hydrolysis profile of UMG‐SP‐1 beyond the reported low‐molecular weight carbamates. Together, these findings promise advanced strategies for the bio‐based degradation and recycling of plastic materials and waste, aiding efforts to establish a circular economy for synthetic polymers

    Effect of Discontinuation of Supportive Periodontal Therapy on Periodontal Status—A Retrospective Study

    Aim To assess the impact of active (APT) and supportive periodontal therapy (SPT) on the change in probing depth (PD) and annual tooth loss in partially and fully compliant and drop‐out patients. Material and Methods Data of 280 periodontally treated partially and fully compliant (regular supportive visits, SPT duration 5.5 ± 4.5 years) and 55 drop‐out patients (SPT and drop‐out duration 8.3 ± 3.8 years, only drop‐out duration 5.3 ± 3.7 years) were recorded. PD data and the number of teeth present at the start of APT (T1) and at the start of SPT (T2) were taken from the patient files and evaluated at the time of the final examination (T3). Results Annual tooth loss during SPT was significantly higher (p < 0.001) in drop‐out patients than in partially and fully compliant patients (0.31 ± 0.50 vs. 0.19 ± 0.55, respectively). In partially and fully compliant and drop‐out patients, the mean PD (all available site data) decreased significantly between T1 (3.61 ± 0.82 vs. 3.70 ± 0.73 mm) and T2 (2.68 ± 0.40 vs. 2.76 ± 0.42 mm), while the values increased again slightly up to T3 (2.74 ± 0.41 vs. 2.99 ± 0.75 mm). Conclusions In partially and fully compliant patients, SPT had a positive impact on PD stability and medium‐term tooth preservation. In contrary to expectations, drop‐out patients, PD did not return to baseline values, although PD stability was not achieved

    Pushing at the Boundaries of Pterin Chemistry

    Pterins are molecules of substantial interest as they occur in nature in a number of forms with quite distinct and often indispensable roles. Chemically, the synthesis of the principle pterin scaffold is comparably simple, while the insolubility of the pterin building block renders synthetic derivatization extremely difficult. When aiming at modeling naturally occurring pterins of extended chemical structure, this is a considerable problem. A notable set of strategies was developed in the course of the present study, which are able to overcome the lack of reactivity of the pterin backbone. These include a strategic choice regarding protection groups, uncommon chemical transformation, ball milling and combinations thereof. Some novel pterins with quite distinct substitution motifs were successfully synthesized and characterized by spectroscopic and spectrometric analyses as well as single-crystal structural analyses for three of them

    Computer-Assisted Proof Verification for Higher-Order Automated Reasoning within the Dedukti Framework

    Automated Theorem Proving is a core area of artificial intelligence research, focusing on the development of software that autonomously proves complex statements from given premises across various application fields. However, the diversity of output encodings of specialized systems hinders the verification of derived proofs and cooperation between provers. The Dedukti framework addresses this issue by implementing the λΠ-calculus modulo, enabling proofs originating from different frameworks to be expressed, combined, and checked automatically. Formal verification of fully automated higher-order logic provers remains an unmet challenge. This thesis establishes a theoretical foundation for such verification by identifying common challenges and developing general encoding strategies. Based on these strategies, a modular encoding of selected inference rules of the versatile prover Leo-III is presented. The effectiveness of this approach is empirically evaluated through the partial automation of generating proof certificates verifiable within the Dedukti framework

    Electrochemical solid phase formation and dissolution; a non-equilibrium thermodynamic view

    Herein, solid-state electrochemical reactions are collectively viewed and studied as the formation and dissolution of a solid phase under non-equilibrium. We first consider an ideal reaction pathway where a solid phase grows and shrinks along the same reaction path. Next, the realistic situation is studied based on the voltammetry of solids to reveal how a solid phase is formed and dissolved in contact with either a liquid or another solid phase. Finally, the reaction asymmetry between phase formation and dissolution, hysteresis, and the memory effect are studied and discussed in detail
