Fibroblast‐derived neuropilin 1 alleviates renal fibrosis progression
International audienceAbstract Chronic kidney disease (CKD) is a major global health challenge affecting over 10% of the adult population. A hallmark of CKD progression is the transdifferentiation of kidney fibroblasts into extracellular matrix‐producing myofibroblasts, a key mechanism involved in the decline of kidney function and the development of kidney failure. Fibroblasts maintain the structural integrity of the kidney and support epithelial survival, repair, and regeneration after acute kidney injury. Maladaptive repair is a failure to resolve fibroblast activation, which ultimately progresses to chronic injury and CKD. In this study, we showed that the membrane‐bound coreceptor neuropilin 1 (NRP1) was essential to maintain fibroblast function and prevent their transdifferentiation into myofibroblasts. We used the myelin protein zero‐Cre ( P0‐Cre ) to specifically abrogate Nrp1 in kidney resident fibroblasts during fibrosis progression. We employed kidney‐induced interstitial fibrosis models combined with a lineage‐tracing strategy, single‐cell RNA sequencing analysis, and ex vivo explant cultures to reveal a cell autonomous protective role for NRP1 in limiting fibrosis. Furthermore, we extended the analysis by showing that Nrp1 conditional mutants were more prone to develop cardiac fibrosis in a mouse model of heart failure. Collectively, these findings provide new insights into the signalling pathways controlling the transition from acute to chronic kidney disease conversion and identify NRP1 as a novel regulator of fibroblast supportive function. © 2025 The Author(s). La forme scolaire d’un dispositif d’accompagnement aux habiletés sociales pour les adolescents autistes
International audienceSocial skills groups (SSGs), led by psychology or education professionals (psychologists, childpsychiatrists, psychometricians, educators, job coaches, etc.), are now a well-establishedpractice in various settings (private practices, non-profit organizations, public sector) for autisticadolescents. In this article, based on research conducted through interviews and observations,we look at the relationship between these SSGs and the school form. In particular, we show thatthey are part of the renewed school form, but also an extension of the school form outside theschool, since these groups generally have no link with the school itself. The aim is therefore tounderstand the type of educational relationship that develops in a specific extra-curricular space,and to explore the expectations of professionals, families and the young people themselveswith regard to the implementation of these groupsLes Groupes d’entraînement aux habiletés sociales (GEHS), animés par des professionnels du psychisme ou de l’éducation (psychologues, pédopsychiatres, psychomotriciens, éducateurs spécialisés, job coaches , etc.), constituent une modalité désormais bien ancrée dans les divers espaces (libéraux, associatifs, publics) accompagnant des adolescents et adolescentes autistes. Dans cet article, nous nous penchons, à l’issue d’une recherche menée par entretiens et observations, sur le rapport qui relie ces GEHS à la forme scolaire. Nous montrons notamment qu’ils relèvent de la forme scolaire dans sa forme renouvelée, mais également d’une extension de la forme scolaire hors de l’école, car ces groupes n’entretiennent généralement pas de lien avec l’école elle-même. Il s’agit donc de comprendre le type de relation pédagogique qui se noue dans un espace extra-scolaire spécifique, et d’explorer en creux les attentes des professionnels, des familles et des jeunes eux-mêmes autour de la mise en œuvre de ces groupes
Variation in caesarean section rates according to the Robson classification in four low- and middle-income countries: a meta-analysis of groups 1 to 4
International audienceBackground Measuring person-centered maternity care outcomes typically consists of two types of measures: experiences of care and satisfaction with care. There are limited validated measurement tools for these measures, particularly in low- and middle-income countries (LMICs). The QUALI-DEC study aims to improve decision-making around caesarean section. We describe development of the QUALI-DEC Study Birth Experience and Satisfaction (QD-BES) scale, and scale validation in Argentina, Burkina Faso, Thailand, and Viet Nam. Methods We used a three-phase scale development and validation approach: 1) item development, 2) scale development, and 3) scale evaluation. We systematically identified existing tools, and assessed them using the QUALI-DEC theory of change, study context, and psychometric qualities. We proposed the 10-item QD-BES scale to balance feasibility, theoretical coverage, and comprehensiveness. We conducted a baseline exit survey with post-partum women in 32 hospitals in 4 countries. We conducted exploratory factor analysis (EFA), and confirmatory factor analysis (CFA). Results 3127 women participated, most were multiparous (61.0%), without previous caesarean section (77.2%), and preferred vaginal birth (72.8%) despite high rates of caesarean section (39.4%). EFA identified three dimensions: emotional satisfaction (3-items), support and respect by providers (4-items), and communication with providers (3-items), with high loading coefficients (0.5–0.97). CFA confirmed the three-dimension scale, with good model fit (CFI and IFI: 0.95, Cronbach’s alpha: 0.70–0.90). Criterion validity was assessed by exploring characteristics of women, obstetric histories, and birth experiences. Conclusions We present psychometric validation of a scale measuring women’s satisfaction with care and experiences of childbirth care, using a systematic approach to development and validation in four LMICs. The 10-item QD-BES-scale is short, easily-administered, valid, and reliable. The QD-BES-scale is useful to contribute to the generation of new knowledge about quality of maternity care in LMICs, as well as help to meet the major challenge of implementing and measuring respectful care at scale
A High-order Multi-scale Finite Element Method (MsFEM) for solving Oseen problems in heterogeneous media
An enriched non-conforming Multi-scale Finite Element Method (MsFEM) to solve viscous incompressible flow problems in genuine heterogeneous or porous media was proposed in [Q. Feng, G. Allaire, and P. Omnes, Multiscale Model. Simul., 20(1):462–492, 2022] and further studied in [L. Balazi, G. Allaire, and P. Omnes, preprint \url{https://hal.science/hal-05198860}, 2025]. The main feature of this MsFEM is the consideration of high-order sets of weighting functions: for the velocity, they are polynomials of order on the faces and of order in the volume of the elements; for the pressure they are polynomials of order in the element volume. The present paper proposes to extend this method to the Oseen problem in heterogeneous porous media. Through numerical simulations, specially for the case , we show that the developed MsFEM is able to deal with high Reynolds numbers
Targeting the activated allosteric conformation of the endothelin receptor B in melanoma with an antibody-drug conjugate: mechanisms and therapeutic efficacy
International audienceAbstract Background Endothelin 1 receptors are one of the drivers of tumor progression in many cancers. Inhibition of their signaling pathways with antagonist drugs has been the subject of numerous clinical trials, but the results have not met expectations probably due to the high endothelin concentrations in the tumor microenvironment and their unusually high affinity for their receptors. Methods We previously reported the rendomab B49 antibody (RB49) exhibiting a preferential affinity for the activated conformation of human endothelin B receptor (ET B ), not displaced by high endothelin levels, and without any pharmacological properties that could inhibit the division of melanoma cells. In this context, we have developed xiRB49-MMAE, a chimeric antibody-drug conjugated (ADC) to monomethyl auristatin E. We have characterized its physicochemical properties, studied its binding mechanisms, and evaluated its therapeutic potential in a preclinical model. Immunohistochemical analysis of metastatic melanoma lymph nodes evaluated RB49 as a diagnostic tool for patient stratification. Results xiRB49-MMAE showed high efficacy against melanoma cells and ET B + xenograft tumor models. IHC studies indicated that 100% of melanoma patient lymph node biopsies were RB49-positive. Conclusions xiRB49-MMAE is a promising drug candidate for clinical trials in ET B + tumors. RB49 could be used as a diagnostic tool for patient stratification
